Description
本出願は、合衆国法典第35巻119条(e)に基づき、2012年7月11日付けで出願された米国仮出願第61/670345号の利益を主張し、その内容は参照によりその全体が本明細書に組み入れられる。
本発明は、国立衛生研究所(National Institutes of Health)によって付与されたR21 CA127099のもとでの政府支援によりなされた。政府は本発明において一定の権利を有する。
本発明は、部分的に、代謝機能を調節するのに有用な方法および化合物に関する。
配列番号1は、GENBANK(登録商標)登録番号AAD38506.1と表記されたヒトDNAJドメインを含有するタンパク質MCJのアミノ酸配列である。
配列番号3は、マウスDNAJドメインを含有するタンパク質のアミノ酸配列である。

いくつかの態様において、本発明は、細胞、組織、および/または対象においてMCJポリペプチド活性を調整する方法に関する。用語「調整」は、本明細書で使用される場合、細胞中のMCJポリペプチド活性のレベル(例えば、MCJポリペプチドのレベルおよび/または機能)を変化させることを意味する。本発明のいくつかの実施形態において、MCJポリペプチド活性を変化させることは、細胞または組織中のMCJポリペプチドのレベルを変化させることを含む。したがって、細胞中のMCJポリペプチドの活性を増加させることは、細胞中のMCJポリペプチドのレベル(例えば量)を増加させることを含み得る。同様に、細胞中のMCJポリペプチドの活性を減少させることは、細胞中のMCJポリペプチドのレベル(例えば量)を減少させることを含み得る。本発明のいくつかの実施形態において、MCJポリペプチドのレベルは、MCJポリペプチドの発現を増加させることにより変化させたりまたは調整させたりできる。したがって、本発明の方法のいくつかの実施形態は、細胞、組織、または対象中のMCJポリペプチドをコードする核酸のレベルを増加または減少させることを含んでいてもよく、それにより、細胞、組織、または対象中のMCJポリペプチドの活性を増加させることができる。本発明の方法の所定の実施形態は、異常なMCJポリペプチド活性を特徴とする代謝性疾患または状態を処置するために、例えば細胞、組織または対象にMCJポリペプチドを送達することによって、細胞、組織、または対象中のMCJポリペプチドのレベルを直接的に増加または減少させることを含み得る。本明細書の他所に記載されるように、本発明のいくつかの実施形態において、本発明の方法で処置することによりMCJポリペプチドまたはその活性のレベルを増加または減少させることができる細胞としては、これらに限定されないが、肝細胞(liver cell)、筋細胞、心臓細胞、循環器系の細胞、ニューロン性細胞、グリア細胞、脂肪細胞、皮膚細胞、造血細胞、上皮細胞、精液、卵母細胞、筋細胞、脂肪細胞、腎細胞、肝実質細胞(hepatocytes)、または膵臓細胞などが挙げられる。本発明のいくつかの実施形態において、細胞は癌細胞であり、その例は、腫瘍細胞であり得る。
本発明のMCJ調節化合物(例えば、抗MCJ抗体もしくはその機能的なフラグメント、MCJポリペプチドをコードする核酸、MCJポリペプチド、または小分子MCJエンハンサーもしくは阻害剤など)は、代謝性疾患または状態を処置するのに有効な量で対象に投与される。「代謝性疾患または状態を処置するのに有効な量」は、望ましい生物学的な作用を達成するのに必要な量または十分な量である。例えば、本発明の化合物の有効な量は、(i)疾患または状態の進行を遅延または停止させる;または(ii)代謝性疾患または状態の1つまたは複数の症状を逆行させるのに必要な量であり得る。本発明のいくつかの態様によれば、有効な量は、代謝性疾患または状態において、代謝性疾患または状態の予防または処置のいずれかにおいて、組み合わせてもしくは共に投与されると、または単独で投与されると治療的応答が生じる本発明の化合物の単独の量、または他の医薬品もしくは処置と組み合わせた本発明の化合物の量である。生物学的作用は、代謝性疾患または状態に起因する症状の改善および/または完全な除去であり得る。他の実施形態において、生物学的作用は、例えば対象が疾患または状態を有さないことを示す診断検査によって証明されるような代謝性疾患または状態の完全な排除である。
MCJ調節化合物の様々な投与経路が利用可能である。選択される特定の送達様式は、当然ながら処置される特定の状態と治療効果をもたらすのに必要な投薬量とによって決まるであろう。一般的に言えば、本発明の方法は、医学上許容できるあらゆる投与様式(これは、臨床的に許容できない有害作用を引き起こすことなく有効なレベルの保護をもたらすあらゆる様式を意味する)を使用して実施が可能である。本発明のいくつかの実施形態において、本発明の化合物は、経口、経腸、経粘膜、経皮、および/または非経口経路で投与が可能である。用語「非経口」は、皮下、静脈内、筋肉内、腹膜内、および胸骨内注射または点滴技術を含む。他の経路としては、これらに限定されないが、鼻(例えば経鼻胃管を介して)、皮膚、膣、直腸、および舌下が挙げられる。本発明の送達経路は、髄腔内、心室内、または頭蓋内を含み得る。本発明のいくつかの実施形態において、本発明の化合物を遅延放出マトリックス内に入れて、このマトリックスを対象中に留置することにより投与してもよい。本発明のいくつかの態様において、化合物(例えば抗MCJ抗体もしくはその機能的なフラグメント、MCJポリペプチドをコードする核酸、MCJポリペプチド、または小分子MCJエンハンサーもしくは阻害剤など)は、特定の細胞または細胞器官を標的化する送達物質でコーティングされたナノ粒子を使用して対象細胞に送達されてもよく、このような細胞または細胞器官の非限定的な例は、ミトコンドリアである。
本発明の所定の態様は、異常なMCJポリペプチド活性を特徴とする代謝性疾患または状態の状況を判定する方法を含む。このような方法は、MCJポリペプチドのレベルまたは機能を決定すること、および決定されたレベルまたは機能を対照のレベルまたは機能と比較することを含み得る。本発明のいくつかの実施形態において、方法は、細胞および/または対象中のMCJポリペプチド、またはMCJポリペプチドをコードする核酸のレベルを検出するためのアッセイにおけるMCJポリペプチドと特異的に結合する抗体または抗原結合フラグメントの使用を含む。このようなアッセイは、対象から生体サンプルを得ること、生体サンプル中のMCJ分子のレベルを決定すること、およびそのレベルを対照レベルと比較することを含み得る。生体サンプルは、本明細書で使用される場合、インビトロでの生体サンプルであってもよいし、またはインビボで検出される(例えば得られる)サンプルであってもよい。生体サンプルは、本明細書で使用される場合、細胞サンプル、組織サンプル、血液サンプル、体液サンプル、細胞内サンプルなどであってもよい。生体サンプルとしては、細胞、組織、または細胞器官が挙げられ、さらに、これらに限定されないが:肝細胞、筋細胞、心臓細胞、循環器系の細胞、ニューロン性細胞、グリア細胞、脂肪細胞、皮膚細胞、造血細胞、上皮細胞、精液、卵母細胞、筋細胞、脂肪細胞、腎細胞、肝実質細胞、膵臓細胞などの細胞型も挙げられる。本発明のいくつかの実施形態において、生体サンプルは、1種または複数の癌細胞を含んでいてもよい。本発明の所定の実施形態において、生体サンプルは、癌細胞を含まない。
本発明の組成物および使用説明書を含むキットも本発明の範囲内である。本発明のキットは、例えば抗MCJ抗体もしくはその機能的なフラグメント、MCJポリペプチドをコードする核酸、MCJポリペプチド、または小分子MCJエンハンサーもしくは阻害剤などの1種または複数の化合物を含んでもよく、このようなキットは、代謝性疾患または状態を処置するのに使用でき、または本発明のいくつかの態様では代謝性疾患または状態を診断またはモニターするのに使用できる。例えば抗MCJ抗体もしくはその機能的なフラグメント、MCJポリペプチドをコードする核酸、MCJポリペプチド、または小分子MCJエンハンサーもしくは阻害剤などの化合物を含有するキットは、あらゆる好適な組織学的方法、細胞学的方法、血清学的方法または他の方法を使用した、細胞、組織、ならびに/または対象中のMCJポリペプチド活性の処置方法、インビトロでの診断、予後予測、および/もしくはそのレベルのモニターのために調製が可能である。本発明のキットの構成要素は、水性培地中に、または凍結乾燥形態でのいずれかでパッケージ化されてもよい。本発明のキットは、例えば試験管、バイアル、フラスコ、ボトル、注射器、または同種のものなどの1つもしくは複数の容器手段もしくは一連の容器手段を厳重な管理下で受け入れるための区画に分けられた担体を含んでいてもよい。第一の容器手段または一連の容器手段は、1種または複数の化合物、例えば抗MCJ抗体もしくはその機能的なフラグメント、MCJポリペプチドをコードする核酸、MCJポリペプチド、または小分子MCJエンハンサーもしくは阻害剤などの化合物を含有してもよい。第二の容器手段または一連の容器手段は、化合物を細胞または組織に送達することが可能な標的化用標識またはリンカー−標識中間体を含有してもよい。
本発明の所定の態様は、細胞、組織、および/または対象においてMCJポリペプチド活性を調整する候補物質を同定および/またはスクリーニングする方法を含む。方法は、候補物質を細胞または組織と、または対象中で混合することを含み、候補物質との接触の前およびその後のMCJポリペプチド活性を決定できる。好適な対照と比較してMCJポリペプチド活性の量が高いことは、物質がMCJレベルを増加させることができることの指標である。好適な対照と比較してMCJポリペプチド活性の量が低いことは、物質がMCJレベルを減少させることができることの指標である。
方法
マウス
C57Bl/6JマウスをJackson Laboratories(Bar Harbor、ME)から購入した。MCJ標的化胚幹細胞(RRN226)をBaygenomics[カリフォルニア大学サンフランシスコ校(UCSF:University of California San Francisco)、San Francisco、CA]から得た。University of Vermont Transgenic Mouse Facilityで、遺伝子を捕獲したES細胞をC57Bl/6Jの胚盤胞に注射し、偽妊娠した雌に移植した。6匹の雄キメラを得て、そのうち5匹が95%より高いキメリズムを示した。キメラをC57Bl/6J雌と交雑したところ、それらの全てが生殖細胞系伝達を起こした(100%)。マウスをさらにC57Bl/6と少なくとも7世代戻し交配した。MCJホモ接合体ノックアウト作製のためにヘテロ接合体の雄および雌を交雑した。MCJ KOマウスをさらに、これまでに説明されたOT−I TCRトランスジェニックマウス(Hogquistら、1994 Cell 76、17〜27)およびMMTV−PyMTマウス(Guyら、1992 Mol Cell Biol 12、954〜961)とも交雑した。マウスを10から14週齢の間で使用した。飢餓の研究のために、マウスを、食物を与えずに水で36時間維持した。血液、肝臓、腎臓、および褐色脂肪を採取した。肝臓のコレステロール蓄積の研究のために、説明されているようにして(2012年10月17日に電子出版された、Plavinskaya,T.ら、Pulm Pharmacol Ther.Effects of acute and chronic low density lipoprotein exposure on neutrophil functionを参照)、マウスを高コレステロールの餌(Harlan Keklad TD.902221)で4週間維持した。University of Vermontの動物保護施設で、全てのマウスを滅菌条件下で飼育した。この手法は、University of Vermont Institutional Animal Care and Use Committeeで承認された。
CD8T細胞およびCD4T細胞を、これまで説明されたようにして脾臓およびリンパ節から負の選択により(Auphanら、1998 J Immunol 160、4810〜4821;Conzeら、2002 J Exp Med 195、811〜823)、さらにMACSシステムを製造元(Miltenyi Biotec、Cambridge、MA)が推奨した通りに使用して正の選択により精製した。精製されたT細胞を、プレートに結合させた抗CD3(2C11)(5μg/ml)および可溶性抗CD28(1μg/ml)(BD Biosciences、Sparks、MD)mAbで刺激した。これまで説明されたようにして(Conzeら、2002 J Exp Med 195、811〜823)抗CD3および抗CD28で活性化され、[3H]−チミジンで標識された精製CD8T細胞(105個の細胞/ウェル)を使用して増殖アッセイを行った。MCF7、MCF7/siMCJ、および293T細胞株を、これまで説明されたようにして(Hatleら、2007 Mol Cell Biol 27、2952〜2966)維持した。ロテノン(10μMで使用)をSigma Aldrich(St.Louis、MO)から購入した。リン酸カルシウムによる293T細胞トランスフェクションをこれまで説明されたようにして(Hatleら、2007 Mol Cell Biol 27、2952〜2966)行った。
ヒトおよびマウスの複数の組織のノーザンブロット(MTN)をClontech Laboratories,Inc.(Mountain View、CA)から購入し、異なる組織からのレベルを標準化したポリAのRNAを入れた。マウスおよびヒトMCJプローブ両方の放射標識を、説明されているようにして(Rinconら、1988 Eur J Immunol Nov;18(11):1791〜6)行い、ノーザンブロット分析を製造元の説明書の通りに行った。
NcoIで消化した10μgの尾部のゲノムDNAを使用した。DNAフラグメントをアガロースゲルで分離し、Hybond(商標)ナイロンメンブレンに移し、MCJのイントロン1からPCRで増幅した領域[5’−gtggg ggtgtctgtgaagtagttt−3’(配列番号6)および5’−ctgggatttaaggagttcacaa−3’(配列番号7)]を用いて放射標識したプローブを付けた。
Qiagenのmini RNeasy(登録商標)キットを製造元(Qiagen、Valencia、CA)が推奨する通りに使用してトータルRNAを単離した。上述したように(Hatleら、2007 Mol Cell Biol 27、2952〜2966)逆転写することによって第一の鎖のcDNAを得た。cDNAを使用して、従来のPCRまたはリアルタイムRT−PCRによりマウスHPRT、MCJ、ベータ2ミクログロブリンを検出した。リアルタイムRT−PCR分析(Applied Biosystems(商標)、Carlsbad、CA)のために、以下のプライマー/プローブの設計されたセットをマウスMCJに使用した[フォワード5’−ccg aat acc tgc ctc ctt ctg−3’(配列番号8)、リバース5’−aca cag cgg gga gaa ggt t−3’(配列番号9)、プローブ5’−cca aag gtc gga cgc cga cat c−3’(配列番号10)]。ハウスキーピング遺伝子としてヒポキサンチンホスホリボシルトランスフェラーゼ(HPRT)またはβ2−ミクログロブリンを使用した比較用CT分析法によって、相対値を決定した。以下のプライマーを使用してMCJの従来のRT−PCR増幅を行った[5’−aag taa tca cgg caa cag caa gg−3’(配列番号11)および5’−aat aaa agc ctg gca gcc ttg c−3’(配列番号12)]。
全細胞抽出物を、これまで説明されたようにして(Hatleら、2007 Mol Cell Biol 27、2952〜2966)トリトン溶解緩衝液中で調製した。ミトコンドリア抽出物および細胞質抽出物を、CD8T細胞およびMCF7細胞にはCell Fractionation Kit Standard(MitoSciences(登録商標)、Eugene、OR)または心臓、腫瘍、および他の組織にはMitochondrial Fractionation Kit(Active Motif(登録商標)、Carlsbad、CA)を使用して精製した。特に指定される場合、ラウリルマルトシド(1%)またはジギトニン(2%)のいずれかを用いてミトコンドリア抽出物を可溶化した。これまで説明されたようにして(Da Cruz、S.ら、2003 J.Biol.Chem.278:41566〜41571を参照)、心臓からの精製ミトコンドリア抽出物を使用してミトコンドリア内膜画分の単離を行った。これまで説明されたようにして(Hatleら、2007 Mol Cell Biol 27、2952〜2966)、ウェスタンブロット分析を行った。親和性免疫精製(Cocalico Biologicals,Inc.(商標)、Reamstown、PA)後にマウスMCJのN末端ペプチド(35aa)で免疫化することにより抗マウスMCJウサギポリクローナルを作製した。抗ヒトMCJマウスモノクローナルAbはすでに説明されている(Hatleら、2007 Mol Cell Biol 27、2952〜2966)。使用される他の抗体は:抗アクチン、抗グリセルアルデヒド−3−リン酸デヒドロゲナーゼ(抗GAPDH)、抗ウサギIgG、抗ヤギIgG、抗マウスIgG(Santa Cruz Biotechnology、Santa Cruz、CA)、抗CoxIV(Cell Signaling Technology(登録商標)、Danvers、MA)、抗NDUFA9および抗NDUFS3(MitoSciences(登録商標)、Eugene、OR)、ならびに複合体IIIコア1タンパク質(MitoSciences);抗グリコーゲン合成酵素(Cell Signaling Technology(登録商標));抗ホスホエノールピルビン酸カルボキシキナーゼ(抗PEPCK)(Santa Cruz Biotechnology、Dallas、TX);カルレチクリン(Enzo Life Science、Farmingdale、NY);ならびにシンタキシン(sintaxin)11(BD Bioscience、San Jose、CA)であった。LumiGLO(登録商標)化学発光基質システム(KPL、Gaithersburg、MD)を使用して、タンパク質を可視化した。上記で説明したようにして作製されたミトコンドリア抽出物と複合体I免疫捕獲モノクローナル抗体(MitoSciences(登録商標)、Eugene、OR)とを製造元(MitoSciences(登録商標)、Eugene、OR)が推奨する通りに使用して複合体Iの免疫沈降を行った。上記で説明したようにして作製されたミトコンドリア抽出物を使用して複合体Iおよび複合体IIIの免疫沈降を行い、洗浄剤としてマルトシド(1%)と複合体Iまたは複合体III免疫捕獲モノクローナル抗体(MitoSciences)とを製造元が推奨する通りに使用して可溶化した。次いでMCJの存在については免疫沈降物を試験するか、または複合体I、III、もしくはIVサブユニットについては特異的なサブユニットをウェスタンブロッティングで試験した。次いでMCJならびに複合体IおよびIIIサブユニットの存在について免疫沈降物をウェスタンブロット分析で試験した。
CD4、CD8、Thy1.1、Thy1.2、Vα2に対する直接標識された抗体[BD Biosciences(Sparks、MD)およびBioLegend(登録商標)(San Diego、CA)]を使用して細胞表面の染色を行った。UV−ブルー色素[Molecular Probes(登録商標),Inc、Life Technologies(Beverly、MA)]を製造元が推奨する通りに使用して細胞生存率に関する染色を行った。テトラメチルローダミン、エチルエステル、過塩素酸塩(TMRE)[Molecular Probes(登録商標)、Inc、Life Technologies、(Beverly、MA)](10μM)を用いて37℃で20分、製造元が推奨する通りに染色することによりミトコンドリア膜電位分析を行った。Mito−SOX(商標)−レッド(Invitrogen(登録商標)、Life Technologies、Beverly、MA)(2.5μM)を用いて37℃で10分、製造元が推奨する通りに染色することによりミトコンドリアROS分析を行った。LSRIIフローサイトメーター(BD Biosciences、Sparks、MD)およびFLOWJO(登録商標)ソフトウェア(Flowjo、Ashland、OR)を使用したフローサイトメトリー分析で全てのサンプルを試験した。
本発明者らがこれまでに述べたようにして(Hatleら、2007 Mol Cell Biol 27、2952〜2966)、共焦点顕微鏡分析のための細胞の調製およびトランスフェクトした293T細胞の免疫染色を行った。ミトコンドリアおよび核それぞれのマーカーとして、Mito Tracker(登録商標)およびTO−PRO(登録商標)[Molecular Probes(登録商標)、Life Technologies(Beverly、MA)]を使用した。HAでタグ付けされたMCJ、それに続いて抗ウサギ二次Ab[Molecular Probes(登録商標)、Life Technologies、(Beverly、MA)])を検出するために、抗HAタグAb(Cell Signaling Technology(登録商標)、Danvers、MA)を使用した。Zeiss LSM510 META共焦点レーザー走査イメージングシステム(Carl Zeiss Microimaging、Thornwood、NY)を使用した共焦点顕微鏡法によりサンプルを試験した。
本発明者らがこれまでに述べたようにして(Hatleら、2007 Mol Cell Biol 27、2952〜2966)、MCJでトランスフェクトした293T細胞、新たに単離されたCD8T細胞または心臓組織の固定された埋め込み型調製物を使用してMCJに関して免疫電子顕微鏡分析を行った。MCJの検出には、抗マウスMCJウサギポリクローナルAbを使用した。
組織学的分析のために、肝臓、腎臓、褐色脂肪、および乳癌を採取し、ホルマリン中で固定し、パラフィンに埋め込んだ。パラフィン包埋ブロックからの組織切片を慣例的な手法に従ってH&Eで染色した。Optronics(登録商標)(Goleta、CA)製のMagnafire(登録商標)デジタルカメラを備えた、またはEVOS(登録商標)XL Core顕微鏡(AMG、Life Technologies、Grand Island、NY)を備えたOlympus(登録商標)BX50光学顕微鏡によって画像を得た。肝臓における脂質蓄積を分析するために、新たに採取した肝臓をOCT中で凍結させ、凍結断片をオイルレッドOで染色した。グリコーゲンの組織学的分析のために、パラフィン包埋肝臓および腎臓断片で過ヨウ素酸シッフ(PAS:periodic acid−Schiff)染色を行った。グルコース(HK)アッセイキットを製造元(Sigma Aldrich、Saint Louis、MO)が推奨する通りに使用して、肝臓抽出物中のグリコーゲンレベル(1mgに相当)を決定した。トリグリセリド比色キット(BioVision、Milpitas、CA)およびケトン体キット(Cayman Chemicals、Ann Arbor、MI)を製造元が推奨する通りに使用して、血清中のトリグリセリドレベルを決定した。遊離脂肪酸定量化キット(Cayman Chemical(登録商標)、Ann Arbor、MI)および/または遊離脂肪酸定量化キット(BioVision、Milpitas、California)を製造元が推奨する通りに使用して、血清中および肝臓抽出物中の遊離脂肪酸(FFA:free fatty acid)レベルを決定した。Abcamキット(Abcam、Cambridge、MA)を使用して、肝臓抽出物中のコレステロールレベル(1mgに相当)を決定した。グルコース−モニタリングシステム(LifeScan、Milpitas、California)を使用して、血液中のグルコースレベルを決定した。
複合体Iの精製のためのプロトコールに従って作製されたミトコンドリア抽出物(MitoSciences(登録商標)、Eugene、OR)を使用して、複合体I活性の分析を行った。活性アッセイは、MitoSciences(登録商標)(Eugene、OR)製の複合体I酵素活性マイクロプレートアッセイキットおよび製造元が推奨するプロトコールを使用した。ミトコンドリア抽出物をサンプル間で標準化し、合計で1μg(心臓)、5〜20μg(MCF7およびMCF7/siMCJ細胞)、または1.8μg(T細胞)をアッセイに使用した。
サンプル1つあたり104個の細胞(CD8T細胞、MCF7細胞、293T細胞)、および製造元からの推奨に従ってATPlite(登録商標)発光ATP検出アッセイシステム(PerkinElmer(登録商標)、Waltham、MA)、およびTD−20/20ルミノメーター(Turner Biosystems、Promega(登録商標)、Madison、WI)を使用して、細胞内ATPレベルを決定した。乳癌組織中のATPレベルを分析するために、細胞質およびミトコンドリア抽出物を上記で説明したようにして作製し、ATPlite(登録商標)キット(PerkinElmer(登録商標)、Waltham、MA)を使用してタンパク質10μgに等しい量をATP含量に関してアッセイした。

これまでにMCJ/DnaJC15の発現は、ヒト卵巣および乳癌細胞で試験されてきた(Hatleら、2007 Mol Cell Biol 27、2952〜2966;Lindseyら、2005 Int J.Cancer Jan 15;118(2):346〜52.;Shridharら、2001 Cancer Res 61、4258〜4265;Strathdeeら、2004 Carcinogenesis 25、693〜701)。これまでに、MCJは脊椎動物に由来しており、脊椎動物においてMCJは高度に保存されることが報告されたが(Hatleら、2007 Mol Cell Biol 27、2952〜2966)、マウスにおいてそれに相当するものを報告する研究はなかった。ヒトおよびマウスMCJタンパク質配列の比較分析から、ほぼ同一な膜貫通およびC末端DnaJドメイン領域、同様に高度に保存された(57%)N末端領域(1〜35アミノ酸)と共に全体的に75%の同一性(図1A)が示された。マウスのMCJ遺伝子の組織発現パターンを同定するために、上記で概説した方法を使用してノーザンブロット分析を行った。MCJのmRNAは心臓で極めて豊富であり、それに続いて肝臓および腎臓で豊富であった(図1B)。MCJはいくつかのヒト癌細胞で発現されるが、正常なヒト組織における特異的なMCJ発現分布はいまだ不明である。良性のヒト組織のノーザンブロット分析から、ヒトのmcj遺伝子発現の分布はマウスのmcj遺伝子に類似することが示された(図1C)。CD8T細胞を使用して行われた以前のマイクロアレイ分析から、MCJはこの免疫細胞型にも存在することが示された(データ示さず)。免疫系の異なる集団におけるMCJ発現をさらに調査するために、マウス脾臓およびリンパ節からCD4およびCD8T細胞、加えてB細胞を単離し、リアルタイム逆転写酵素(RT)PCRを行った。興味深いことに、CD8T細胞ではmcj遺伝子発現は極めて高かったが、CD4T細胞およびB細胞ではほとんど検出不可能であった(図1D)。
系統発生的分析から、MCJの祖先は酵母に存在するTim14であることが示された(Mokranjacら、2003 Embo J 22、4945〜4956)。Tim14はミトコンドリア内膜に局所しており、TIM23トランスロカーゼの構成要素である(Mokranjacら、2005 J Biol Chem 280、31608〜31614)。これまでに、(内因性MCJを発現しない)293T細胞でのMCJの過剰発現分析、および共焦点顕微鏡法を使用したMito Tracker(登録商標)染色(Hatleら、2007 Mol Cell Biol 27、2952〜2966)に基づいてMCJはミトコンドリア中に局在しないことが報告された。加えて、トランスフェクトした293T細胞において、免疫電子顕微鏡法(IEM)では輪郭のはっきりしたミトコンドリアでMCJの免疫染色は見出されなかったが、未同定の他の電子密度の高い小胞/細胞器官でMCJの免疫反応性が観察された(Hatleら、2007 Mol Cell Biol 27、2952〜2966)。正常組織でMCJの可能性のある機能をより厳密に調べるために、生理学的条件下で、IEMによって心臓における内因性MCJの局在化を試験した。ほとんどのMCJの免疫反応性が、輪郭のはっきりしたミトコンドリアで、内膜において優勢に見出された(図2A)。さらに精製CD8T細胞でもIEMによりMCJ発現を試験した。結果から、ほとんど例外なくMCJはCD8T細胞のミトコンドリアに局在することが示された(図2B)。24時間にわたるMCJを過剰発現する293T細胞における、IEMによるMCJのより詳細な分析から、内膜に形成されたクリスタが失われ分解を受けた、不明瞭な、膨張したミトコンドリアである可能性がある未同定の細胞器官中に、MCJの免疫反応性が存在することが解明された(図2C)。MCJでトランスフェクト(24時間)した293T細胞中のMCJおよびMito Tracker(登録商標)に関する共焦点顕微鏡分析から、非トランスフェクト細胞ではMito Tracker(登録商標)染色によりミトコンドリアが明示されたが、MCJを過剰発現する細胞ではミトコンドリア染色はほとんど検出不可能であったことが示された(図2D〜F)。したがって、MCJは、生理学的条件下ではミトコンドリアに局在するが、この細胞器官においてMCJが長期的に過剰発現されると明らかな構造的損傷をもたらすことが見出された。
ミトコンドリアの主要な機能は、酸化的リン酸化を介して細胞にエネルギー源としてATPを供給することである。電子の移動に加えて、ミトコンドリア電子移動鎖(ETC:mitochondria electron transfer chain)の複合体I、III、およびIVは、ミトコンドリアのマトリックスから膜間腔への膜を介したH+輸送を促進することによりミトコンドリア膜電位(MMP:mitochondrial membrane potential)の確立に寄与する。複合体V(ATPシンターゼ)は、ミトコンドリアのマトリックスへのH+の還流により生成したエネルギーを使用して、ATPを生成する。MCJがミトコンドリアの機能を調整できるのかどうかを決定するために、MCJを発現することが公知の(Hatleら、2007 Mol Cell Biol 27、2952〜2966)MCF7乳癌細胞中のATPレベルを、MCF7/siMCJ細胞中のレベルと比較した(ここでMCF7細胞は、MCJのshRNAによりMCJ発現がノックダウンされていた)(Hatleら、2007 Mol Cell Biol 27、2952〜2966)。興味深いことに、MCF7/siMCJ細胞中のATPレベルは、MCF7細胞よりも著しく高かった(図3A)。MCF7/siMCJ細胞におけるATP量の上昇は、主としてミトコンドリアに起因するものであり、これはなぜなら、ロテノンによるETC複合体Iの阻害によりATPレベルが低減されたためである(図3B)。これらの結果から、MCJは、ミトコンドリアのATP合成の負の調節因子であることが示唆された。これらの結果をさらに確認するために、MCJ発現後、293T細胞中のATPレベルを試験した。293T細胞をわずか16時間でトランスフェクトすることにより、24時間後に観察されるミトコンドリアの損傷を回避した(図2)。プラスミド対照細胞と比べてMCJを発現する細胞ではATPレベルが高度に低減されたことから(図3C)、MCJがATP生産を負に調節することがさらに実証された。
野生型CD8T細胞に対するMCJ KOのCD8T細胞におけるミトコンドリアの過分極がそれらの発達に影響を与える可能性があるかどうかを試験した。MCJノックアウトマウスでは、リンパ節(図5A)および脾臓(データ示さず)中のCD8T細胞のパーセンテージおよび数は影響を受けなかった。同様に、MCJ KOマウスにおいて、胸腺細胞集団の差も観察されなかった(データ示さず)。したがって、MCJは、末梢免疫系におけるCD8T細胞の発達またはホメオスタシスにとって必須ではないようである。さらにMCJの喪失がCD8T細胞の増殖応答に与える可能性がある作用も試験した。精製CD8T細胞を抗CD3および抗CD28抗体で活性化した。[3H]−チミジンの取り込みにより増殖を測定した。MCJ KOマウスと対照野生型マウスとの間でCD8T細胞の増殖応答における実質的な差は観察されなかった(図6A)。加えて、ミトコンドリア中のMCJが存在するにもかかわらず、MCJ欠損マウスからのCD8T細胞は、活性化中にそれほど速く細胞死に至らなかった(図5B)。また活性化中の野生型CD8T細胞とMCJ KOのCD8T細胞との間の活性化マーカー(例えばCD69、CD44、CD25)の発現も同等であった(データ示さず)。したがって、MCJは、CD8T細胞の活性化またはクローン増殖には必要ではないようである。抗CD3および抗CD28AbによるCD8T細胞の活性化中のMMPの調節をさらに試験した。野生型CD8T細胞では、活性化中にMMPが次第に増加した(図6B)。MCJ KOのCD8T細胞および野生型CD4T細胞でも活性化に伴いMMPはわずかに増加したが、活性化前にすでに高度に過分極化した段階であったためその作用はそれほど顕著なものではなかった(図6B)。2日後、野生型CD8T細胞中のMMPレベルは、活性化されたMCJ KOのCD8T細胞および野生型CD4T細胞のMMPレベルと同様のレベルに達した(図4B)。またmROSレベルも、活性化から2日後、野生型CD8T細胞とMCJ KOのCD8T細胞との間で同等であった(図7)。
ミトコンドリア代謝を弱めるMCJの役割は、基礎(生理学的な)条件下でMCJ欠損マウスの異常な表現型がないことと相関する。しかしながら、エフェクターCD8T細胞の休止期の誘導と同様に、MCJの欠損は、代謝バランスが破壊されている状況で作用を有する可能性がある。絶食は強烈な代謝の変化を引き起こすが、これはなぜなら、十分なグルコースがないことにより、脂肪組織中に保存されたトリグリセリドが遊離脂肪酸(FFA)に加水分解し始め、そのFFAが血漿に動員されて肝臓に輸送されるためである。肝臓において、FFAはミトコンドリアのβ−酸化に取り込まれてエネルギー源として使用される。MCJは肝臓で高度に発現されるため、MCJの非存在下におけるミトコンドリア代謝の増加/維持は、FFAのβ−酸化を促進して、それらの肝臓での蓄積を最小化する可能性があるとの仮説が立てられた。絶食時代謝中のMCJの役割を試験した。正常な(給餌)条件下での肝臓の組織像的分析から、野生型マウスとMCJ欠損マウスとの間に検出可能な差がないことが示された(図8A)。36時間絶食した野生型マウスの肝臓は、小胞が豊富な細胞の存在によって決定したところ明らかな脂肪変性の徴候を示した(図8A)。それに対して、絶食したMCJ KOマウスからの肝臓では最小の徴候しか検出されなかった(図8A)。オイルレッドO染色による肝臓の凍結断片における脂質の蓄積の分析からさらに、絶食した野生型マウスにおいて大量の脂質の存在が確認されたが、MCJ KOマウスの肝臓では低いレベルでしか存在しなかった(図8B)。これらの結果から、MCJ KOマウスにおけるミトコンドリアのFFA酸化の維持は、肝臓における脂質の蓄積を最小化することが示唆された。トリグリセリドおよびFFAの血清レベルも試験した。絶食したMCJ KOマウスにおいて、トリグリセリド(図8C)およびFFA(図8D)の両方のレベルが野生型マウスと比較して低減されたことから、保存された脂質の消費が促進されたことがさらに実証される。一貫して、絶食したMCJ KOマウスでは最小の白色脂肪量が維持されることが観察された(データ示さず)。加えて、野生型マウスとMCJ KOマウスとの間に正常条件(給餌)での褐色脂肪には明白な差が見出されなかった(図9Aおよび9B)にもかかわらず、絶食後の褐色脂肪のグロス分析(図10A)および組織学的分析(図10B)では、MCJ KOマウスにおいて褐色脂肪がほぼ完全に存在しないことが解明された。したがって、結果から、MCJは、ミトコンドリアの機能の負の調節因子として、絶食中の代謝調節において役割を果たすことが示された。
絶食中のマウスで代謝の促進が確認されたが、MCJ KOマウス(nice)において絶食によって引き起こされる体重減少は、野生型マウスにおける体重減少とは、最初の体重は2つのグループで同等であったとしても(図11B)統計学的に異なっていなかった(図11A)。加えて、絶食中の血中グルコースレベルの分析から、最初のグルコース低下(12時間)(図11C)または回復(図11D)のいずれかの間、MCJ KOマウスと野生型マウスとの間に統計学的に有意差はなかったことが示された。これらの結果から、MCJの非存在下で肝臓における脂質代謝促進のバランスをとるメカニズムの存在が示唆された。肝臓でのミトコンドリアによる遊離脂肪酸(FFA)β−酸化の増加は、(β−酸化を介した)ATPレベルの増加、加えて脂質の破壊の結果生じるグリセロールレベルの増加に繋がる可能性がある。ATPおよびグリセロールの蓄積は、糖生成(多大なエネルギーを要するプロセス)を開始させて過剰なエネルギーを保存するためのシグナルとして肝臓で感知できる。絶食後の肝臓中のATPレベルの分析を行い、野生型マウスの肝臓に対するMCJ KOマウスの肝臓におけるATPレベルの増加を確認した(図11E)。肝臓中のグリコーゲンレベルをPAS染色で試験した。予想通りに、絶食した野生型マウスの肝臓においてグリコーゲンはほとんど検出不可能であった(図11F)。しかしながら、絶食したMCJ KOマウスの肝臓には高レベルのグリコーゲンが蓄積した(図11F)。肝臓に加えて、糖生成は、それほどではないが腎皮質でも起こる可能性がある。絶食したMCJ KOマウスにおいて、グリコーゲンの蓄積は、PAS染色により腎臓のいくつかの領域でも見出される場合がある。肝臓抽出物中のグリコーゲンの生化学的分析からさらに、絶食したMCJ KOマウスにおけるグリコーゲンの選択的な蓄積が実証された(図11G)。絶食していない野生型マウスの肝臓中のグリコーゲンの基礎レベルと絶食していないMCJ KOマウスとの間に差は見出されなかった(図11G)。絶食したMCJ KOマウスの肝臓において脂質の代わりにグリコーゲンが蓄積されたことは、肝臓と体重との比率が野生型マウスと比べて増加したことと相関していた(図11H)。グリコーゲンは、基質としてUDP−グルコースを使用してグリコーゲン合成酵素により合成される。絶食したMCJ KOマウスからの肝臓では、野生型マウスに比べてグリコーゲン合成酵素のレベルがアップレギュレートされた(図11I)。それに対して、ピルベートからのグルコース合成において必須の糖新生酵素であるホスホエノールピルビン酸カルボキシキナーゼ(PEPCK)のレベルは、絶食したWTおよびMCJ KOマウスで同等であり(図11I)、これは、MCJ KOマウスの肝臓に存在するグリコーゲンは、トリグリセリドの加水分解によって生じたグルコースと一緒に生成した可能性があることを示唆する。
ミトコンドリア呼吸はATPを生成する最も効率的なメカニズムであるが、ある種の環境では、細胞は、それより効率が低いにもかかわらず解糖を選択する。したがって、癌細胞は、腫瘍増殖に有利な好気条件下でさえも、ミトコンドリアの酸化を介するよりも解糖からそれらのATPを生成する。この現象はワールブルク効果として知られており、ミトコンドリアの酸化的リン酸化が損なわれたことに起因する癌細胞の代謝的な適応のようである(Cairnsら、2011 Nat Rev Cancer 11、85〜95;Koppenolら、2011 Nat Rev Cancer 11、325〜337;Warburg、1956 J Mol Biol 272、477〜483)。MCJはMMPおよびATPの負の調節因子であることから、MCJの欠損がインビボで腫瘍の進行に影響を与えることができるかどうかを検討した。乳房上皮細胞中での特異的な発現を駆動させるMMTV(マウス乳癌ウイルス)プロモーター/エンハンサーの制御下でポリオーマウイルスの中型T(MT)抗原が発現されるMMTVPyMTトランスジェニックマウスからの腫瘍で、MCJ発現を試験した(Guyら、1992 Mol Cell Biol 12、954〜961)。正常な乳腺と乳癌の両方でMCJが発現されたが、レベルは腫瘍のほうがわずかに高いようであった(図12A)。ウェスタンブロット分析によるMMTV−PyMT腫瘍におけるMCJ細胞局在性の分析で、乳癌におけるミトコンドリアのMCJ局在化がさらに確認された(図12B)。正常な乳腺および乳癌からのミトコンドリア抽出物におけるMCJの分析からも、腫瘍からのミトコンドリアにおけるMCJ発現が、正常な乳腺のミトコンドリアと比較して増加したことが確認された(図12C)。MCJ KOマウスをMMTV−PyMTマウスと交雑し、それに続いて乳癌を発生させた。興味深いことに、MMTV−PyMT/MCJ KOマウスは腫瘍を発生させたが、MMTV−PyMT同腹子と比較してその速度は遅かった。以前の研究と一致して(Guyら、1992 Mol Cell Biol 12、954〜961)、MMTV−PyMT同腹子では10〜12週齢までに腫瘍を明確に検出できたが、MMTV−PyMT/MCJ KOマウスでは、18〜20週間よりも前に明確な腫瘍は触診で確認できなかった。腫瘍サイズが大きくなったためにマウスを安楽死させなければならなかったマウスの年齢を使用して生存曲線を確立した。生存曲線から、MMTV−PyMT/MCJ KOマウスにおいて、MMTV−PyMTマウスと比較して腫瘍増殖の有意な遅延が示された(図12D)。組織学的分析から、MMTV−PyMTマウスとMMTV−PyMT/MCJ KOマウスからの類似のサイズの腫瘍との間に明白な差は示されなかった(図12E)。腫瘍増殖の遅延が、エネルギー源としての細胞質の解糖に対するミトコンドリア呼吸のバランスの変化に関連するかどうかを検討するために、野生型およびMCJ KOの腫瘍から生成した細胞質およびミトコンドリア抽出物でATPレベルを試験した。野生型マウスからの腫瘍の比率と比較した、MCJ KOマウスからの腫瘍におけるミトコンドリア/細胞質のATPレベルの相対的な比率において有意な増加がみられた(図12F)。したがって、MCJの非存在下で見出されるミトコンドリア代謝の強化は、乳癌増殖の減少と相関していた。
MCJがミトコンドリア膜電位を調節できる分子メカニズムを調査して、可能性のある関連タンパク質を発見するために、ベイトとしてMCJのN末端領域を使用してファージディスプレイスクリーニングを行った。スクリーニングの結果から、可能性のあるMCJと相互作用するタンパク質としてミトコンドリアETC内の複合体Iの1つのサブユニット(NDUFv1)が解明された(データ示さず)。哺乳動物の複合体I(NADH−ユビキノンオキシドレダクターゼ)は、49個の同定されたサブユニット、フラボモノヌクレオチド(FMN)グループ、および8つのFe−Sクラスターを含有する(Clasonら、2009 J Struct Biol 169、81〜88)。MCJは、複合体Iの実際に存在するサブユニットとして説明されていないが、MCJは複合体Iと相互作用してその活性を調節する可能性があると考えられた。MCJが複合体Iと結合するかどうかを決定するために、野生型およびMCJ KOマウスの心臓のミトコンドリア抽出物から複合体Iを免疫沈降した。免疫沈降物中のMCJの存在をウェスタンブロット分析で試験した。野生型からの複合体Iの免疫沈降物中にMCJの分子量に相当するバンドが存在したが、MCJ KOマウスからの複合体Iの免疫沈降物中には存在しなかった(図13A)。この免疫沈降の対照として、複合体Iの2つのよく特徴付けられたサブユニットであるNDUFA9およびNDUFS3の発現(図13A)を試験した。したがって、MCJは、ミトコンドリアの複合体Iと結合する。
Claims (28)
- 対象における代謝性疾患または状態を処置するための医薬組成物であって、MCJ調節化合物を対象における代謝性疾患または状態を処置するのに有効な量で含み、代謝性疾患または状態が肝臓の糖代謝性疾患または肝臓の脂質代謝性疾患であり、癌ではなく;ここでMCJ調節化合物は、抗MCJポリペプチド抗体もしくはその機能的なフラグメントまたはMCJ siRNAを含み、MCJ調節化合物は、MCJポリペプチド活性を減少させ、MCJポリペプチド活性を減少させることは、MCJポリペプチドのレベルまたは機能を減少させることを含む、前記医薬組成物。
- MCJ調節化合物が、対象においてMCJポリペプチド活性を減少させ、対象においてミトコンドリアの代謝活性を増加させる、請求項1に記載の医薬組成物。
- 対象が、カロリー摂取レベルを低減させる処置を受ける、請求項1に記載の医薬組成物。
