本発明は、タイプカルチャーコレクション中国センター(CCTCC、ウーハン大学、ウーハン、中国)に寄託された以下の生物学的材料に関する:
ハイブリドーマ細胞株12G6は、CCTCC番号:C201527の寄託番号で、2015年4月10日に寄託された;
ハイブリドーマ細胞系7G6は、CCTCC番号:C201435の寄託番号で、2014年3月26日に寄託された;そして
ハイブリドーマ細胞株11B10は、CCTCC番号:C201432の寄託番号で、2014年3月26日に寄託された。
1.ウィルス抗原の調製
MDCK細胞に、B/アモイ/891/2006(ヤマガタ)、B/アモイ/1346/2008(ビクトリア)、B/アモイ/N697/2012(ヤマガタ)及びB/アモイ/3043/2006(ビクトリア)の4株のインフルエンザBウィルスを接種した。細胞を37℃で2日間インキュベートした後、上清を回収して増幅されたウィルスを得た。生ウィルスを収集し、4℃で0.03%ホルマリンで不活性化した。不活性化されたウィルスは、HA滴定を行い、不活化ウィルスの力価を決定した(注:HA滴定の特定の手順については、WHO運用ガイドラインを参照)。B/アモイ/891/2006(ヤマガタ)、B/アモイ/1346/2008(ビクトリア)、B/アモイ/N697/2012(ヤマガタ)、B/アモイ/3043/2006(ビクトリア)は、発明者の研究室で分離されたインフルエンザB型ウィルスの株であった。
6週齢、SPFグレードのメスのBALB/cマウスは、実験動物センター、アモイ大学によって提供された。
モノクローナル抗体を分泌するハイブリドーマ細胞は、標準的なインビボ免疫法及びPEG融合法によって得られた。詳細については、Harlowら編、「抗体実験マニュアル」、Cold Spring Harbor Laboratory 1988を参照されたい。簡単なプロセスは以下のとおりである。
不活性化されたウィルスの力価を128HAに調整し、次いでマウスを逐次免疫により免疫化した。要約すると、まず、ウィルスB/アモイ/891/2006(ヤマガタ)をフロイント完全アジュバント(CFA)と等量混合して乳化させた後、各マウスについて400μlの量で、一次免疫用多点筋肉注射によりマウスの肢に投与した。B/アモイ/1346/2008(ビクトリア)、B/アモイ/N697/2012(ヤマガタ)、B/アモイ/3043/2006(ビクトリア)を別々に混合し、不完全フロイントアジュバント(IFA)で乳化させた後、それぞれ、一次免疫後14日、28日及び42日に追加免疫のためにマウスに投与した。最後に、一次免疫化後56日で、追加免疫がマウス脾臓に行われた(ここで、免疫原はそれぞれのマウスに対して、上記ウィルスの100μlずつの混合物であった)。免疫3日後、マウスの脾臓を融合実験のために採取した。
脾臓を粉砕して脾臓細胞懸濁液を得た後、マウスミエローマ細胞SP2/0と指数増殖期で混合した。細胞融合は、PEG1500の存在下で行った。融合細胞を400ml融合培地に再懸濁し、次いで、培養のために20枚の96ウェルプレートに播種した。融合培養培地は、HATおよび20%FBSを含有するRPMI1640完全スクリーニング培地であった。
融合した細胞を96ウェルプレートで10日間培養した後、細胞の上清を赤血球凝集阻害(HI)アッセイおよびELISAのために採取した。検出のためのウィルスは、B/アモイ/891/2006(ヤマガタ)およびB/アモイ/1346/2008(ビクトリア)であった。HIアッセイでは、陽性ウェルに分泌される抗体は、インフルエンザB型ウィルスおよび赤血球の凝集を阻害することができなければならない;ELISAでは、陽性ウェル中に分泌された抗体は、ポリスチレンプレート上にコーティングされたインフルエンザB型ウィルスと特異的に反応することができなければならない。陽性クローンは、3回のクローニングに供し、安定的に抗体を分泌することができるハイブリドーマ細胞株を得た。最終的に、12G6,7G6および11B10を含む、インフルエンザB型ウィルスのHAタンパク質に対する41のハイブリドーマ細胞株が得られた。
41の安定なハイブリドーマ細胞株を、CO2インキュベーター中で増幅培養に供した後、24ウェルプレートに96ウェルプレートから移し、その後50mLの細胞ボトルにさらに培養しました。次いで、細胞ボトルから回収した細胞は、マウスの腹腔に注射した。7〜10日後、モノクローナル抗体を含む腹水をマウスの腹腔から採取した。
モノクローナル抗体を含む腹水を50%硫酸アンモニウム溶液で沈殿させた。得られた沈殿物をPBSに溶解し、AKTAシステムのプロテインAカラムで精製し、精製したモノクローナル抗体を得た。精製したモノクローナル抗体の純度をSDS−PAGEにより同定した。
インフルエンザB型ウィルスの代表的な株は、異なる領域から異なる時点で単離され、異なる変異型を示すものが選択され、対照としてインフルエンザA型の代表的な株が選択された。HIアッセイを使用して、インフルエンザB型ウィルス変異株の異なる系統と、前記41モノクローナル抗体の交差反応性を決定した。HIアッセイについては、WHOオペレーションガイドラインを参照してください。インフルエンザウィルス株とモノクローナル抗体の反応性により、3つの広域スペクトルモノクローナル抗体12G6,7G6および11B10が同定され、インフルエンザB型ウィルスのヤマガタとビクトリア系統を同時に認識する(すなわち、インフルエンザB型ウィルスの異なるHA系統のHAタンパク質を認識する)ことができた(表2)。
中和活性/力価は、モノクローナル抗体が防止や病気の治療の可能性を持っているかどうかを評価するための重要な指標である。この実施例では、マイクロウェル中和アッセイを使用して、モノクローナル抗体12G6、7G6及び11B10のインフルエンザB型ウィルスの異なる系統の代表的な株に対する中和活性を測定した(方法はHulse−PostらPNAS. 2005, 102: 10682−7を参照)。実験結果を表3に示す.3つのモノクローナル抗体(12G6、7G6および11B10)は、早期に発見された未系統インフルエンザB型ウィルス、ヤマガタ系統のインフルエンザB型ウィルス及びビクトリア系統のインフルエンザB型ウィルスに対して広域交差中和活性を有していた。モノクローナル抗体7G6の中和活性は、HI活性と実質的一致していた。モノクローナル抗体7G6は、2つのウィルス、すなわち、B/ハルビン/7/1994(ヤマガタ系統)および/B/五大湖/1739/1954を除くすべての試験されたインフルエンザB型ウィルスに中和活性を有し、比較的広い活性スペクトルと非常に強い反応性示した。モノクローナル抗体11B10の中和活性スペクトルはHI活性スペクトルよりもわずかに広かった。特に、モノクローナル抗体11B10は、1962年から2012年の間のほとんどすべてのインフルエンザB型ウィルスを中和することができた(B/ハルビン/7/1994以外)。モノクローナル抗体12G6の中和活性スペクトルは、HI活性スペクトルとは全く異なっており、モノクローナル抗体12G6は、1940年から2012年の間の、すべてのインフルエンザB型ウィルスを中和することができ、非常に強く、非常に広いスペクトル中和活性を示した。
この実施例では、モノクローナル抗体12G6,7G6および11B10を使用して、エスケープ突然変異を有するインフルエンザBウィルス株を誘導およびスクリーニングした。スクリーニングされたエスケープウィルスは、プラーク精製、増幅培養、遺伝子検索、シークエンシング及びエスケープ変異部位の構造的局在化に供し、モノクローナル抗体12G6、7G6及び11B10によって認識されるエピトープが存在する領域、並びに認識されたキーエピトープアミノ酸が決定された。
(1)親ウィルス:B型インフルエンザウィルス、B/シンガポール/3/1964(ビクトリア系統)及びB/アモイ/1346/2008をエスケープ突然変異スクリーニングのための親ウィルスとして選択した。
(2)モノクローナル抗体:精製12G6,7G6,11B10
(3)エスケープスクリーニング方法:105TCID50親ウィルスを均一、単一のモノクローナル抗体と混合した。モノクローナル抗体の最終濃度は1mg/mlであり、総体積が1mlであった。前記ウィルスモノクローナル抗体複合体を室温で4時間インキュベートし、次いで96ウェルプレートにプレコートしたMDCK細胞に感染させるために使用した。2時間ウィルスモノクローナル抗体複合体で細胞をインキュベートした後、複合体は除去され、50μg/mlのモノクローナル抗体を含む維持培地を添加し、そして細胞を48時間37℃でさらに培養した。培養上清を血球凝集アッセイに行いた。陽性であると判断されたウェルのウィルスが、エスケープウィルスであった。3サイクルのプラーククローニングの後、安定したエスケープウィルス株が得られた。
(4)エスケープウィルス株のエスケープ変異部位の局在:得られたエスケープウィルス株は、RT−PCRを行い、ウィルスの構造遺伝子を得た、得られた構造遺伝子を配列決定し、親ウィルスの構造遺伝子と比較し、エスケープ突然変異および突然変異部位を同定した。
(5)HAの立体構造におけるキーエピトープアミノ酸の局在:A/フロリダ/4/2006(ヤマガタ)ウィルスのHA蛋白質(PDB番号:4FQJ)の3D構造のファイルは、PDBデータベースからダウンロードした。3DプロッティングソフトウェアPymolを使用して、HAの受容体結合部位および12G6,7G6,11B10によって認識される重要なエピトープアミノ酸部位をHAの3D構造に局在化させた。
エスケープ突然変異の結果は、すべてのエスケープ突然変異部位が、インフルエンザB型ウィルスHAタンパク質のHA1ドメインをコードする遺伝子に存在することを示した。
約107個のハイブリドーマ細胞を半付着により培養した。接着した細胞を吹き込みにより懸濁させ、新しい4ml遠心管に移した。1500rpmで3分間遠心分離した後、細胞沈殿物を集め、100μlの滅菌PBS(pH=7.45)に再懸濁し、次いで新しい1.5ml遠心チューブに移した。Trizol(Roche、Germany)800μlを添加し、逆混合により穏やかに混合し、次いで10分間静置した。クロロホルム200μlを添加し、15分間激しく振とうし、10分間放置した後、4℃、12000rpmで15分間遠心分離した。上清を新しい1.5ml遠心チューブに移し、等量のイソプロパノールを添加し、混合し、10分間放置した後、4℃、12000rpmで10分間遠心分離した。上清を除去し、75%エタノール600μlを添加して洗浄した後、4℃、12000rpmで5分間遠心分離した。上清を除去し、沈殿物を真空下、60℃で5分間乾燥させた。透明な沈殿物を70μlのDEPC H2Oに溶解し、そして2本のチューブに回収した。1つのチューブに、1μLの逆転写プライマー(MVJkR(5’−CCGTTTGKATYTCCAGCTTGGTSCC−3’)(配列番号:31))を添加し、軽鎖可変領域をコードする遺伝子の増幅を行い、もう1つのチューブに、転写リバースプライマー(MVDJhR(5’−CGGTGACCGWGGTBCCTTGRCCCCA−3’)(SEQIDNO:32))を添加し、重鎖可変領域をコードする遺伝子の増幅を行った。各チューブに、1μlのdNTP(SangonBiotech(Shanghai)Co.、Ltd.)を添加し、チューブを72℃の水浴に10分間入れ、直ちに氷浴中で5分間置いた。10μlの5x逆転写バッファー、1μlのAMV(10U/μl、プロメガ)、および1μLのRnasin(40U/μL、プロメガ)を添加した。混合物をよく混合した後、RNAを42℃でcDNAに逆転写した。
1.材料と方法
(1)インフルエンザウィルスの調製:一定量の、以下のインフルエンザウィルス株:B/フロリダ/04/2006(ヤマガタ)、B/マレーシア/2506/2004(ビクトリア)、A/ブリスベン/20/2007(H3N2)、A/ニューカレドニア/20/1999(H1N1)を、0.03%ホルマリンで4℃で不活性化した。不活化ウィルスを、超遠心分離にショ糖密度勾配遠心分離にかけ、そして4℃で、3時間25200rpmで遠心分離した。ウィルス沈殿物を4℃で一晩、1×PBS溶解した。超遠心分離したウィルスをHA滴定によって決定し、ウィルス溶液の力価を測定した。
(2)モノクローナル抗体:上記で調製したモノクローナル抗体7G6および11B10;モノクローナル抗体の濃度は、1mg/mlであった。
(3)ELISA実験:
超遠心分離したウィルスの力価を128HAに適合させ、そして96ウェルポリスチレンELISAプレートにウェルあたり200μlプレコートした。次いで、96ウェルプレートをブロッキング液でブロッキングした。試験されるモノクローナル抗体は、初期濃度として0.1mg/mlに希釈し、そして2倍勾配希釈を15回行った。希釈したモノクローナル抗体をELISAプレートウェル当たり100μlの体積で、30分間37℃でインキュベートした。ELISAプレートを、ELISA洗浄溶液(PBST)で5回洗浄し、それから100μlのHRP標識二次抗体を加え、そして得られた混合物を30分間37℃でインキュベートした。ELISAプレートをPBSTで5回洗浄した後、発色剤を添加し、20分間発色させた。吸収値A450はその後、ELISAリーダーで読み取った。
ELISAの結果は図2に示す。図2に示すように、モノクローナル抗体7G6及び11B10は、インフルエンザB型ウィルスの2つのHA系統(すなわち、ヤマガタ及びビクトリア)との強い結合反応性を有したが(B/フロリダ/04/2006(ヤマガタ)及びB/マレーシア/2506/2004(ビクトリア)、インフルエンザA型ウィルスの代表的な株(例えば、A/ブリスベン/20/2007(H3N2)およびA/ニューカレドニア/20/1999(H1N1))と特異的な反応性を有さなかった。
結果は、広域スペクトルモノクローナル抗体7G6および11B10を使用して、インフルエンザB型ウィルスの少なくとも2つのHA系統を特異的に検出することができることを示した。
抗体による感染症の受動免疫療法は、抗ウィルス治療の潜在的に有効な経路である。これは、本発明のモノクローナル抗体は、異なる時間に異なる領域から単離されたインフルエンザB型ウィルス少なくとも2つのHA系統(例えば、ヤマガタ、ビクトリア)に対して高い中和活性を有することが、先の実施例におけるマイクロウェルでインビトロ中和アッセイによって実証された。本発明によるモノクローナル抗体は、インフルエンザB型ウィルスの少なくとも2つのHA系統(例えば、ヤマガタ及びビクトリア)に対する広い中和反応性スペクトラム及び高い中和力価によって特徴付けられた。さらに、インビボで本発明のモノクローナル抗体の抗ウィルス効果を実証するために、本発明のモノクローナル抗体は、インフルエンザB型ウィルスのヤマガタ系統およびビクトリア系統による感染に対する抗ウィルス治療効果を、ヤマガタ系統及びビクトリア系統のインフルエンザB型ウィルスに感染したBALB/cマウスに基づき、動物バイオセーフティ研究所で検証した。実験は以下に従った。
動物:Balb/Cマウス、SPF、6−8週齢、メス、重さ約20g。
モノクローナル抗体:12G6,7G6,11B10
インフルエンザB型ウィルスのマウス適応系統:
インフルエンザB型ウィルスのヤマガタ系統のマウス適応株:B/フロリダ/04/2006、略してFL04−MAと呼ばれる。
インフルエンザB型ウィルスのビクトリア系統のマウス適応株:B/ブリスベン/60/2008、略してBR60−MAと呼ばれる。
麻酔薬:イソフルラン
動物群:マウスを1日前に動物バイオセーフティ研究所に送り、G1、G2、.....等と指定されたケージあたり5匹のマウスを有する群に分けた;各マウスの体重を記録し、レジメンを表9に示す。
ウィルス感染:ヤマガタ系統ウィルスB/フロリダ/04/2006を105 TCID50/μlに事前に希釈し、ビクトリア系統ウィルスB/ブリスベン/60/2008を106TCID50/μlに予め希釈し、そして50μl/マウスの用量でマウスに接種した。接種前に、マウスをイソフルランで麻酔し、次いで鼻腔を介してウィルスを接種することにより感染させた。
MAbの介入:ウィルス感染後24時間(dpi.1)は、抗体処置群のマウスは、一定の投与量、すなわち10mg/kgで抗体を注入された。注入量は100μlであった。
観察:ウィルス感染後1−14日、体重、生存、および対応する行動症状を毎日記録した。
マウスはヤマガタ系統のインフルエンザB型ウィルスFL04−MAとビクトリア系統のインフルエンザB型ウィルスBR60−MAに別々に1週間の致死量で感染させた。感染後1日目に、抗体を治療群のマウスの尾静脈に注射した。モノクローナル抗体の治療効果は、体重を測定し、各群のマウスの生存率を計算することによって決定した。実験結果を図3−5に示した。
図3−5の実験結果は、ネガティブコントロール群のマウスでは、実験全体で有意な体重変動は観察されなかった;有意な体重減少は、二つのウィルスに感染した対照群で観察された。FL04−MAウィルス対照群の全てのマウスは8日、感染後に死亡し;BR60−MAウィルス対照群の全てのマウスは感染5〜10日後に死亡した。前記2種のインフルエンザB型ウィルスの場合、10mg/kgの用量での12G6、7G6、11B10抗体注入による介入は、感染マウスの体重回復につながった(図3B、3D、図4B、図4D、図5B、及び図5D)。さらに、ウィルス感染の1〜14日後にマウスの生存が観察された。その結果、10mg/kgの用量のモノクローナル抗体12G6,7G6,11B10は、ウィルスに感染したマウスを通常14日間生存させることができ、治療有効性は100%に達した(図3A、3C、4A、4C、5Aおよび5C)。この結果は、本発明による広域スペクトルモノクローナル抗体が、インフルエンザB型ウィルスの2つのHA系統による感染およびそれによって引き起こされる疾患を効果的に予防および治療することができることを示した。
1.モノクローナル抗体12G6は、宿主細胞へのB型インフルエンザウィルスの侵入を阻害することができる。
(1)材料および方法
ウィルス:B/フロリダ/4/2006(ヤマガタ)、B/ブリスベン/60/2008(ビクトリア);
HA特異的抗体:モノクローナル抗体12G6、B/フロリダ/4/2006ウィルスに対するポリクローナル抗血清、およびB/マレーシア/2506/2004ウィルスに対するポリクローナル抗血清;
インフルエンザB型ウィルスNPタンパク質に対して特異的なウサギポリクローナル抗血清;
GAM−FITC(FITC標識ヤギ抗マウス抗体、緑色蛍光);
細胞:MDCK細胞;
モノクローナル抗体12G6(10μg/ml)、B/フロリダ/4/2006のウィルスに対するポリクローナル抗血清、B/マレーシア/2506/2004のウィルスに対するポリクローナル抗血清、及びPBS(抗体を含まない対照)を別々にインフルエンザB型ウィルスの二系統とともに1時間37℃でインキュベートした。次いで、インキュベートした混合物を、96ウェルプレートで培養した単層MDCK細胞に加え、細胞をさらに16〜18時間培養した。培養後、細胞を培地で3回洗浄した。次に、市販のキットを用いて細胞をDAPI染色(青色蛍光)に付した。細胞を、インフルエンザB型ウィルスのNPタンパク質に対するウサギポリクローナル抗血清(第一抗体)及びGAM−FITC(二次抗体として、緑色蛍光)を用いて免疫蛍光分析を行った。
結果は図6に示された。結果として、PBSをインキュベーションに使用した場合、試験したインフルエンザB型ウィルスの2系統の両方がMDCK細胞に入る可能性があることが示した(細胞は、緑色蛍光を示した)。B/フロリダ/4/2006ウィルスに対するポリクローナル抗血清をインキュベーションに使用した場合、B/フロリダ/4/2006(ヤマガタ)はMDCK細胞に進入できなかった(細胞は青色の蛍光を示した);一方、B/ブリスベン/60/2008(ビクトリア)はMDCK細胞に入ることができた(細胞は、緑色蛍光を示した)。B/マレーシア/2506/2004ウィルスに対するポリクローナル抗血清をインキュベーションに使用した場合、B/ブリスベン/60/2008(ビクトリア)はMDCK細胞に侵入できなかった(細胞は青色蛍光を示した)、B/フロリダ/4/2006はMDCK細胞に入る可能性がある(細胞は緑色の蛍光を示した)。モノクローナル抗体12G6をインキュベーションに使用した場合、インフルエンザB型ウィルスの2系統のいずれもMDCK細胞に入り得なかった(細胞は青色蛍光を示した)。結果は、モノクローナル抗体12G6が、インフルエンザB型ウィルスの2系統の宿主細胞への侵入を阻害し得ることを示した。
(1)材料および方法
ウィルス:B/フロリダ/4/2006(ヤマガタ)、B/ブリスベン/60/2008(ビクトリア);
HA特異的抗体:モノクローナル抗体12G6;
実験用試薬:ギムザ染色、10mM MES(Sigma、カタログ番号:M3671−250G)および10mM HEPES(Sigma、カタログ番号:H3375、pH5.5)およびTPCKトリプシン(Sigma、カタログ番号:T1426);
細胞:MDCK細胞;
感染多重度MOI=0.3で、MDCK細胞を、それぞれ、B/フロリダ/4/2006(ヤマガタ)およびB/ブリスベン/60/2008(ビクトリア)によって感染させた。24時間後、細胞を培地で3回洗浄し、残余のウィルスを除去した。2.5mg/ml TPCKトリプシンを、その後、細胞に添加した。15分間37℃でのインキュベーション後、細胞を培養培地で3回洗浄して、残余のトリプシンを除去した。後に、所定の濃度(0μg/ml、5μg/ml、20μg/mlまたは100μg/ml)の抗体12G6を細胞に添加し、そして30分間37℃でインキュベートした。抗体溶液をそれから除去し、細胞を2分間37℃で、10mM MESおよび10mM HEPES(pH5.5)中でインキュベートした(アロステリズムは酸性条件下で容易にHAタンパク質に起こり、それによって、ウィルスエンベロープの細胞膜へ融合を促進する。3回の培養培地で洗浄した後(洗浄後、ウィルス/細胞培養環境は、酸性から中性になった)、ウィルスに感染した細胞は、3時間37℃でさらに培養した。その後、細胞を固定し、ギムザ染色で染色し、細胞内での膜融合の有無を観察した。抗体が細胞膜へのウィルスエンベロープの融合を阻害することができる場合は、膜融合の結果である合胞体を染色後に観察することができない。逆に、抗体がウィルス膜の細胞膜への融合を阻害できない場合、膜融合の結果である合胞体を、染色後に観察することができる。
実験結果を図7に示した。結果として、抗体12G6細胞をインキュベーションに使用しなかった場合(例えば、0μg/ml抗体)、試験したインフルエンザB型ウィルスの2つの系統の両方が、MDCK細胞の膜融合をもたらし得た(すなわち、多くの合胞体が出現した)。5μg/mlまたは20μg/mlの抗体12G6を場合、MDCK細胞の膜融合は有意に阻害された(すなわち、合胞体の数は有意に減少した)、20μg/mlの抗体は5μg/mlの抗体よりも強い阻害を示した。さらに、100μg/mlの抗体12G6をインキュベーションに使用した場合、MDCK細胞の膜融合は完全に阻害された(すなわち合胞体が出現しなかった)。結果は、モノクローナル抗体12G6は、インフルエンザB型ウィルスの2系統の細胞への膜融合を阻害することができ;そしてモノクローナル抗体12G6の阻害活性は用量依存的であることを示した。
(1)材料および方法
ウィルス:B/フロリダ/4/2006(ヤマガタ)、B/ブリスベン/60/2008(ビクトリア);
HA特異的抗体:モノクローナル抗体12G6、および陰性対照抗体(抗HIV mAb 5G6);
インフルエンザBウィルスのNPタンパク質に対して特異的なウサギポリクローナル抗血清;
GAM−HRP;
細胞:MDCK細胞;
MDCK細胞を、40000細胞/ウェルの密度で96ウェルプレートに接種した。4時間後、過剰量のインフルエンザB型ウィルスを細胞に添加して細胞を感染させた。感染の3時間後、ウィルス溶液を除去し、細胞をPBSで3回洗浄して遊離ウィルスを除去した。細胞培養プレートには、培養液(抗体を含まない対照)、または所定濃度のモノクローナル抗体12G6(2μg/ml、0.2μg/mlまたは0.02μg/ml;培地に希釈)または所定の濃度の陰性対照抗体(20μg/ml又は2μg/ml;培地に希釈)を加えた。37℃でさらに16〜18時間インキュベートした後、細胞上清および細胞溶解物をそれぞれ回収し、インフルエンザB型ウィルスのNPタンパク質に対するウサギポリクローナル抗血清(一次抗体)及びGAM−HRP(二次抗体)を用いて、イムノブロットアッセイを行った。
実験結果を図8に示した。結果として、インフルエンザB型ウィルスに感染したMDCK細胞を、抗体とインキュベートしないか又は陰性対照抗体とインキュベートした場合、有意な量のNPタンパク質が培養上清および細胞溶解物に検出することができた。これは、インフルエンザB型ウィルスの2つの系統がMDCK細胞で増殖することができ、宿主細胞からインフルエンザB型ウィルスが放出され、そして陰性対照抗体は、宿主細胞からインフルエンザB型ウィルスの放出を阻害することができなかったことを示した。対照的に、モノクローナル抗体12G6は、インフルエンザB型ウィルスに感染したMDCK細胞をインキュベートするために使用した場合、NPタンパク質は細胞溶解物中で検出することができたが、モノクローナル抗体12G6の濃度の増加とともに培養上清中のNPタンパク質の量が減少した。モノクローナル抗体12G6の濃度が2μg/mlに達したとき、NPタンパク質は培養上清中に全く検出されず、インフルエンザB型ウィルスの放出が完全に阻害されたことを示した。これらの結果は、モノクローナル抗体12G6は、宿主細胞からインフルエンザB型ウィルスの系統の放出を阻害でき、モノクローナル抗体12G6の阻害活性は用量依存的であることを示した。
Srivastava V. et al., J Virol, 2013 May, 87(10):5831−40に記載された方法によって、モノクローナル抗体12G6のADCC及びCDC活性を測定した。
(1)材料および方法
ウィルス:B/マサチューセッツ/02/2012様(ヤマガタ)、およびB/ブリスベン/60/2008(ビクトリア);
HA特異的抗体:12G6、及び陰性対照抗体(抗HIV mAb 5G6);
細胞:MDCK細胞、およびマウスNK細胞;
細胞染色用試薬:PKH−67(SIGMA−ALDRICH、カタログ番号:PKH67GL;一般に使われている細胞膜色素として)、及び7−AAD(eBioscience、カタログ番号:00−6993−50;核酸色素として、死んだ細胞を同定するため);
マウスNK細胞単離キット(NK Cell Isolation Kit II mouse、製造元:MACS、カタログ番号:130−096−892)を用いて、マウスの脾臓(すなわち、エフェクター細胞)からNK細胞を単離し、使用した。感染の多重度MOI=10で、MDCK細胞(すなわち、標的細胞)を、インフルエンザB型ウィルスに感染させた。3時間後、1x105細胞/mLの細胞濃度で、100μlの細胞を96ウェルプレートに播種した。1時間培養した後、MDCK細胞の膜を、PKH−67色素で染色した。染色後、試験される抗体を所定の濃度に希釈し(20μg/ml、2μg/ml、または0.5μg/ml)、そして50μl/ウェルの容量で培養プレート中の細胞に添加し、次いで、37℃で15分間インキュベートした。その後、ADCCアッセイのために、エフェクター細胞(100μl)を培養プレート中の細胞に、エフェクター細胞と標的細胞の比が、50:1になるように、添加し;CDCアッセイのために、100倍希釈したモルモット血清(100μl)を補体として、培養プレート中の細胞に添加した。培養プレートを37℃で2時間インキュベートし、次いで、色素7−AADを、1μl/ウェルの量で添加し、そして得られた混合物を5分間インキュベートした。インキュベーション後、細胞をフローサイトメーターで分析し、そして死んだ標的細胞の割合を算出した。加えて、バックグラウンドコントロールとして、抗体を使用しなかった条件の下で実験を繰り返し、エフェクター細胞とインキュベートした、感染した標的細胞の自発的溶解率を表した。加えて、1%TritonX−100(抗体の代わりに試験される)を陽性対照として、実験を繰り返し、エフェクター細胞とインキュベートした、感染した標的細胞の最大溶解率を表した。
ADCC活性またはCDC活性=(テストグループにおける死んだ標的細胞の百分率−バックグラウンド対照群における死んだ標的細胞の百分率)/(陽性対照群における死んだ標的細胞の百分率−バックグラウンド対照群における死んだ標的細胞の百分率)*100%
実験結果は図9に示された。結果として、陰性対照抗体をインキュベーションのために使用した場合、NK細胞やモルモット血清の存在下で、ウィルスに感染したMDCK細胞で明らか溶解が発生しなかった。これは、陰性対照抗体が、2つの系統のインフルエンザB型ウィルスに対してADCC及びCDCを誘発しないことを示した。対照的に、抗体12G6は、インキュベーションのために使用した場合、NK細胞またはモルモット血清の存在下でのウィルス感染MDCK細胞で、明らかな溶解が発生した。そして、用いた抗体の濃度の増加と共に、MDCK細胞の溶解が促進された。これらの結果は、モノクローナル抗体12G6は、試験した2つの系統のインフルエンザB型ウィルスに対してADCC及びCDCを誘発でき;モノクローナル抗体12G6の活性は用量依存的であることを示した。
Claims (23)
- モノクローナル抗体またはその抗原結合断片であって、
(1)配列番号1〜3に記載のアミノ酸配列をそれぞれ有するVH CDR1〜3、および配列番号4〜6に記載のアミノ酸配列をそれぞれ有するVL CDR1〜3、
(2)配列番号7〜9に記載のアミノ酸配列をそれぞれ有するVH CDR1〜3、および配列番号10〜12に記載のアミノ酸配列をそれぞれ有するVL CDR1〜3、または
(3)配列番号13〜15に記載のアミノ酸配列をそれぞれ有するVH CDR1〜3、および配列番号16〜18に記載のアミノ酸配列をそれぞれ有するVL CDR1〜3;
を含み、
インフルエンザB型のHAタンパク質のHA1ドメインに特異的に結合することができる、上記モノクローナル抗体またはその抗原結合断片。 - (1)配列番号19に記載のVHおよび配列番号20に記載のVL;
(2)配列番号21に記載のVHおよび配列番号22に記載のVL;または
(3)配列番号23に記載のVHおよび配列番号24に記載のVL;
を含む、請求項1に記載のモノクローナル抗体またはその抗原結合断片。 - 以下の1以上の特徴を有する、請求項1又は2に記載のモノクローナル抗体またはその抗原結合断片:
(1)モノクローナル抗体またはその抗原結合断片は、Fab、Fab’、F(ab’)2、Fd、Fv、dAb、相補性決定領域断片、単鎖抗体、マウス抗体、ヒト化抗体、キメラ抗体、または二重特異性または多特異的抗体から選択される;
(2)モノクローナル抗体またはその抗原結合断片は、非CDR領域を含み、非CDR領域は、ネズミ以外の種由来のものである;
(3)モノクローナル抗体は、タイプ・カルチャー・コレクション中国センター(CCTCC)に、寄託番号CCTCC NO:C201527、CCTCC NO:C201435;及びCCTCC NO:C201432としてそれぞれ寄託されているハイブリドーマ細胞株12G6、7G6及び11B10によって産生される抗体である;
(4)モノクローナル抗体またはその抗原結合断片は、インフルエンザB型ウイルスのヤマガタ系統及びビクトリア系統のHAタンパク質のHA1ドメインに特異的に結合することができる;
(5)モノクローナル抗体またはその抗原結合断片は、インフルエンザB型ウイルスのヤマガタ系統およびインフルエンザB型ウイルスのビクトリア系統に対して赤血球凝集抑制活性を有する;
(6)モノクローナル抗体またはその抗原結合断片は中和活性を有し、インフルエンザB型ウイルスのヤマガタ系統及びインフルエンザB型ウイルスのビクトリア系統を中和することができる;
(7)モノクローナル抗体またはその抗原結合断片は、インフルエンザB型ウイルスのヤマガタ系統およびビクトリア系統の宿主細胞への侵入を阻害する;
(8)モノクローナル抗体またはその抗原結合断片は、宿主細胞からインフルエンザB型ウイルスのヤマガタ系統およびビクトリア系統の放出を阻害する;
(9)モノクローナル抗体またはその抗原結合断片は、宿主細胞とインフルエンザB型ウイルスのヤマガタ系統およびビクトリア系統の膜融合を阻害する;
(10)モノクローナル抗体またはその抗原結合断片は、インフルエンザB型ウイルスのヤマガタ系統およびビクトリア系統に対して、ADCCを引き起こす;及び
(11)モノクローナル抗体またはその抗原結合断片は、インフルエンザB型ウイルスのヤマガタ系統およびビクトリア系統に対して、CDCを引き起こす。 - 単鎖抗体は、scFvであり、且つ/又はキメラ抗体は、ヒトマウスキメラ抗体である、請求項3に記載のモノクローナル抗体またはその抗原結合断片。
- 非CDR領域は、ヒト抗体由来のものである、請求項3に記載のモノクローナル抗体またはその抗原結合断片。
- 請求項1〜5のいずれか一項に記載のモノクローナル抗体またはその抗原結合断片をコードする、単離された核酸分子。
- 請求項6に記載の単離された核酸分子を含むベクター。
- 請求項6に記載の単離された核酸分子または請求項7に記載のベクターを含む、宿主細胞。
- 請求項8に記載の宿主細胞を適切な条件下で培養し、該細胞培養物からモノクローナル抗体またはその抗原結合断片を回収することを含む、請求項1〜5のいずれか一項に記載のモノクローナル抗体またはその抗原結合断片の製造方法。
- 以下からなる群から選択されるハイブリドーマ細胞系:
1)タイプ・カルチャー・コレクション中国センター(CCTCC)に、寄託番号CCTCC NO:C201527で寄託されたハイブリドーマ細胞株12G6;
2)タイプ・カルチャー・コレクション中国センター(CCTCC)に、寄託番号CCTCC NO:C201435で寄託されたハイブリドーマ細胞株7G6;
及び
3)タイプ・カルチャー・コレクション中国センター(CCTCC)に、寄託番号CCTCC NO:C201432で寄託されたハイブリドーマ細胞株ハイブリドーマ細胞系11B10。 - 請求項1〜5のいずれか一項に記載のモノクローナル抗体またはその抗原結合断片、請求項6に記載の単離された核酸分子、請求項7に記載のベクター、又は請求項8に記載の宿主細胞を含む組成物。
- 検出可能なマーカーのついた又はつかない、請求項1〜5のいずれか一項に記載のモノクローナル抗体またはその抗原結合断片;及び
任意選択で
前記モノクローナル抗体またはその抗原結合断片を特異的に認識し、任意選択で、検出可能なマーカーで標識されている二次抗体
を含む、キット。 - 請求項1〜5のいずれか一項に記載のモノクローナル抗体またはその抗原結合断片を使用することを含む、試料中のインフルエンザB型ウイルスまたはそのHAタンパク質の存在またはレベルをインビトロで検出するための方法。
- 試料中のインフルエンザB型ウイルスまたはそのHAタンパク質の存在またはレベルを検出するための、または
被験体がインフルエンザB型ウイルスに感染しているかどうかを診断するためのキットの製造における、請求項1〜5のいずれか一項に記載のモノクローナル抗体またはその抗原結合断片の使用。 - インフルエンザB型ウイルスは、インフルエンザB型ウイルスのヤマガタ系統およびビクトリア系統から選択され、且つ/又は
試料は、対象からの排泄物、口腔または鼻分泌物である、
請求項14に記載の使用。 - 対象は、ヒトである、請求項14または15に記載の使用。
- 請求項1〜5のいずれか一項に記載のモノクローナル抗体またはその抗原結合断片並びに薬学的に許容される担体および/または賦形剤を含む、医薬組成物。
- さらに、追加の医薬活性剤を含む、請求項17に記載の医薬組成物。
- 追加の医薬活性剤は、抗インフルエンザ剤である、請求項18に記載の医薬組成物。
- インフルエンザB型ウイルスを含む試料を、請求項1〜5のいずれか一項に記載のモノクローナル抗体またはその抗原結合断片と接触させることを含む、試料中のインフルエンザB型ウイルスの毒性をインビトロで中和する方法。
- インフルエンザB型ウイルスはインフルエンザB型ウイルスのヤマガタ系統およびビクトリア系統から選択される、請求項13または20に記載の方法。
- 対象におけるインフルエンザB型ウイルスの感染または感染症に関連する疾患を予防または治療における使用のための、請求項1〜5のいずれか一項に記載のモノクローナル抗体またはその抗原結合断片を含む医薬組成物。
- 以下の1以上の特徴を有する、請求項22に記載の医薬組成物:
(1)対象はヒトであり;
(2)インフルエンザB型ウイルスはインフルエンザB型ウイルスのヤマガタ系統およびビクトリア系統から選択され;
(3)薬剤は、単独で使用されるか、または追加の薬学的に活性な薬剤と組み合わせて使用され、
(4)感染症に関連する疾患はインフルエンザである。
