<尿検査>
1.2-trial Y-maze
[Aging parameters]
The degree of aging was evaluated by the following parameters (survival rate, aging score, urinalysis, tumor and ulcer incidence, brain function (behavior) evaluation).
<Survival>
Assessed by creating Kaplan-Meier survival curve Measurement N = 13 (breeding period: 60 weeks)
<Aging score>
Scores were obtained by scoring 11 items such as exploratory behavior, hair and skin roughness, degree of hair loss, corneal turbidity, and spinal flexion. In detail, the score was calculated | required by the following method generally used as one of the criteria of an anti-aging effect.
That is, the degree of aging that appears in the appearance of mice based on the grading score system (Toshio Takeda, development of aging-promoting model mice, intractable disease models and animal experiments, 180, (1984), Soft Science) As evaluation items for evaluating the above, behavior immediately after taking out the mouse from the gauge (reactivity), reaction when attempting to capture by hand from above (glossiness), glossiness, and roughness of hair and skin (coarness) ), Loss of hair, ulcer of the skin, periphalmic eruption, corneal opacity, corneal ulcer 11 items of turbidity (cataract) and spinal flexion are set, and the age of aging is determined for each evaluation item according to the following criteria (see Table 1) for the 6-week-old mouse group. The obtained 11 item scores were totaled to give a score. The degree of aging was measured every 3 weeks.
<Urinalysis>
Tests for glucose, protein, bilirubin, urobilinogen, pH, occult blood, ketone bodies, E nitrate, leukocytes, creatinine, etc. Measurement N = 13 (breeding period: 60 weeks)
FIG. 3 shows the evaluation results of occult blood and leukocytes, which are indicators of inflammation.
<Tumor and ulcer incidence>
The tumor (lymph tumor) was determined by observation of its appearance. Judgment was made when the tumor was found. Other visceral tumors were judged by necropsy at the time of death.
The ulcer (skin ulcer) was judged by observation of the appearance.
In the measurement, each group N = 13 and the breeding period was 60 weeks.
<Evaluation of brain function (behavior)>
1. 2-trial Y-maze test Specifically, in order to make the mouse recognize the spatial arrangement, first place several tall objects as marks on the outside of the arm of the Y-shaped maze, and fix this object. Prepared.
Next, the mouse was trained by placing the mouse in the device with one of the arms randomly closed and allowing the mouse to search freely for 10 minutes.
Then, one hour after the training, the blocked arms were opened and the mouse was searched for the three arms so that the three arms (start arm, familiar arm, novel arm) could freely move back and forth. A video of 5 minutes of action was recorded.
Finally, analysis was performed. That is, the time when all the limbs of the mouse entered the arm was regarded as entering, and the stay time of each arm was measured.
In the evaluation after 6 months, N = 7 in the control group, N = 10 in the group of Example 1, and N = 8 in the group of Example 2. In the evaluation after 12 months, N = 6 in the control group, N = 10 in the group of Example 1, and N = 8 in the group of Example 2.
2. Novel Object Recognition Test First, mice were acclimated for 3 days (minimum 2 days) by leaving them in their cage for 10 minutes.
Next, two objects (Lego) were affixed to the cage bottom with gummed tape or the like. The mice were placed in the center of the cage and trained. The video of the search was recorded for 10 minutes from directly above. Lego as a known object was collected and the surface was cleaned with ethanol.
Subsequently, after 1 hour from the training, a known object and a novel object (golf ball) at the time of training were attached to the bottom of the cage with gummed tape or the like, the mouse was placed in the cage, and the mouse was searched.
The video was recorded and the object search time was measured visually. Even if the mouse was on the object, it was not counted unless it was searching for the object, such as when the face of the mouse was facing up. Mouse passages were not counted.
Then, analysis was performed by the following formula using “search time in training”, “search time in test”, and “new object recognition rate (%)” as indices.
Novel object recognition rate (%) = (Time to search for novel object) / (Sum of time to search for both objects)
In the evaluation after 6 months, N = 9 in the control group, N = 10 in the group of Example 1, and N = 8 in the group of Example 2. In the evaluation after 12 months, N = 6 in the control group, N = 10 in the group of Example 1, and N = 8 in the group of Example 2.
