Description
代謝と細胞内シグナル伝達がリンクしており、これら2つのプロセスが相互にお互いを調節し、細胞の生存、増殖、および幹細胞機能の調節などの細胞活性を調節していることが示されている(非特許文献1:Wellen and Thompson, 2012) (非特許文献2:Takubo et al., 2013)。
(1)哺乳動物由来の多能性幹細胞を分化誘導する方法であって、
(a)該多能性幹細胞を、メチオニンを含まない多能性幹細胞の未分化維持培地で培養する工程、および
(b)該未分化維持培地で培養した該多能性幹細胞を、分化誘導培地で培養して分化誘導する工程、
を含む分化誘導方法。
(2)前記哺乳動物由来の多能性幹細胞が、ヒト又はマウス由来のES細胞、又はヒト又はマウス由来のiPS細胞である、前記(1)に記載の分化誘導方法。
(3)前記細胞を前記未分化維持培地で少なくとも5時間培養することを含む前記(1)または(2)に記載の分化誘導方法。
(4)前記細胞を前記未分化維持培地で少なくとも10時間培養することを含む前記(3)に記載の分化誘導方法。
(5)前記細胞を前記未分化維持培地で24時間を超えない期間で培養することを含む前記(1)〜(4)のいずれか一つに記載の分化誘導方法。
(6)メチオニンを含まない多能性幹細胞の未分化維持培地で培養された哺乳動物由来の多能性幹細胞を、さらに、内胚葉、中胚葉または外胚葉のいずれかに分化誘導する、前記(1)〜(5)のいずれかひとつに記載の分化誘導方法。
(7)前記多能性幹細胞が内胚葉への分化に耐性である幹細胞であって、かつ、前記分化誘導培地が内胚葉への分化誘導培地である、前記(1)〜(6)のいずれかひとつに記載の方法。
(8)前記多能性幹細胞が中胚葉への分化に耐性である幹細胞であって、かつ、前記分化誘導培地が中胚葉への分化誘導培地である、前記(1)〜(6)のいずれかひとつに記載の方法。

(10)メチオニンを含まない多能性幹細胞の未分化維持培地で培養されたヒトiPS細胞を、さらに、膵臓分化誘導培地で培養してインスリン産生能をもつ膵臓細胞へと分化誘導する、前記(2)〜(5)のいずれか一つに記載の分化誘導方法。
(11)メチオニンを含まない多能性幹細胞の未分化維持培地で培養されたヒトiPS細胞を、さらに、肝臓分化誘導培地で培養してアルブミン分泌能をもつ肝臓細胞へと分化誘導する、前記(2)〜(5)のいずれか一つに記載の分化誘導方法。
(13)前記哺乳動物由来の多能性幹細胞が、ヒト又はマウス由来のES細胞、又はヒト又はマウス由来のiPS細胞である、前記(12)に記載の細胞。
(14)前記細胞が前記未分化維持培地で少なくとも5時間培養された細胞である、前記(12)または(13)に記載の細胞。
(15)メチオニンを含まない多能性幹細胞の未分化維持培地で培養され、次いで膵臓分化誘導培地で培養された経歴を有する、ヒトiPS細胞から分化誘導されたインスリン産生能をもつ細胞。
(16)メチオニンを含まない多能性幹細胞の未分化維持培地で培養され、次いで肝臓分化誘導培地で培養された経歴を有する、ヒトiPS細胞から分化誘導されたアルブミン分泌能を有する細胞。
(17)前記未分化維持培地での培養が少なくとも5時間である前記(15)または(16)に記載の細胞。
(18)培地中にメチオニンを含まない多能性幹細胞の未分化維持培地。
(19)多能性幹細胞が、ヒト又はマウス由来のES細胞、又はヒト又はマウスのiPS細胞である、前記(18)に記載の未分化維持培地。
(20)前記(18)または(19)に記載の未分化維持培地と多能性幹細胞の分化誘導培地からなる、多能性幹細胞分化誘導培地キット。
ES細胞は、一般的には、胚盤胞期の受精卵をフィーダー細胞と一緒に培養し、増殖した内部細胞塊由来の細胞をばらばらにして、さらに、植え継ぐ操作を繰り返し、最終的に細胞株として樹立することができる。このように、ES細胞は、受精卵から取得することが多いが、受精卵以外、例えば、脂肪組織、胎盤、精巣細胞から取得することもでき、いずれのES細胞も本発明の対象である。
本発明において、iPS細胞の培養も定法により行うことができる。例えば、フィーダー細胞としてMEF細胞を用いて、bFGF、KSR(ノックアウト血清代替物)、非必須アミノ酸、L−グルタミン、ペニシリン、ストレプトマイシン、β−メルカプトエタノールを加えた培地、例えばDMEM/F12培地を用いて維持することができる。
維持培地は、多能性幹細胞を、未分化の状態で、維持、培養するための培地であり、単に細胞を保存するために維持、培養する場合、および分化誘導に先立って細胞を維持、培養する場合のいずれにも用いられるが、本発明において用いる維持培地は、多能性幹細胞の分化誘導に先だって、多能性幹細胞を維持、培養するための培地を意味する。かかる培地は、これに限定されないが、ES/iPS細胞の維持培養に用いられる白血病阻害因子(LIF)やbFGF等の未分化維持に寄与する因子を含むことができる。本発明における多能性幹細胞、例えばES細胞又はiPS細胞の未分化維持培養は、フィーダー細胞を含む培養系、フィーダーフリーの培養系のいずれも含む。フィーダー細胞としては、例えば、mmcMEF細胞をあげることができるが、これに限定されるものではない。また、フィーダーフリーの培養系としては、例えば、マトリゲルをあげることができるが、これに限定されるものではない。
分化培地は、多能性幹細胞を、内胚葉、中胚葉又は外胚葉のいずれかに分化誘導させるための培地であり、目的の分化指向に応じて、異なった培地が用いられるのが一般的である。
本発明で用いられる培地は、血清含有培地、無血清培地であり得るが、異種成分の排除による細胞移植の安全性の確保という点、および/またはMet除去を十分に制御するという点からは、無血清培地が好ましい。ここで、無血清培地とは、無調整又は未精製の血清を含まない培地を意味し、精製された血液由来成分や動物組織由来成分(例えば、増殖因子)が混入している培地は無血清培地に該当するものとする。かかる無血清培地としては、例えば、市販のKSRを適量(例えば、1−20%)添加した無血清培地、インスリンおよびトランスフェリンを添加した無血清培地、細胞由来の因子を添加した培地等をあげることができるが、これらに限定されない。
本発明で用いられる培地はまた、脂肪酸又は脂質、アミノ酸(例えば、非必須アミノ酸)、ビタミン、増殖因子、サイトカイン、抗酸化剤、2−メルカプトエタノール、ピルビン酸、緩衝剤、無機塩類等、任意の成分を含有できる。ただし、本発明に従って、メチオニンを培地から除く場合は、培地はメチオニンを含有しない。
本発明における多能性幹細胞、例えばES細胞又はiPS細胞の分化誘導は、フィーダー細胞を含む培養系、フィーダーフリーの培養系のいずれも含む。フィーダー細胞としては、例えば、mmcM15細胞をあげることができるが、これに限定されるものではない。また、フィーダーフリーの培養系としては、例えば、擬似基底膜sBM(synthesized Basement Membrane substratum)をあげることができるが、これに限定されるものではない。
ここで、多能性幹細胞をΔMet培地で培養(または維持)とは、多能性幹細胞をΔMet培地中に置くことを意味し、細胞が増殖する場合および増殖せず単に培地中に一定期間置かれる場合のいずれも含む意味である。
本発明のメチオニン除去培地と組み合わせることができる分化培地は、特に限定されず、一般の分化培地の他、特許文献1に記載の本発明者らが報告した特定のアミノ酸が除去された分化培地も用いることができる。
本発明の方法に従い、上記の期間、多能性幹細胞をメチオニン除去培地で培養した後、培地を分化誘導培地に交換して多能性幹細胞の分化誘導を行うことにより、分化誘導効率が改善できる。
(1)セルライン
ヒトES細胞は、熊本大学の治験審査委員会によって承認され、日本政府のhES細胞ガイドラインに従って使用した。未分化ヒトES細胞(khES1、khES3)(Suemori et al.,Biochem Biophys Res Commun 345, 926-932. 2006参照)とiPS細胞(201B7、253G1)(Takahashi et al., Cell 131, 861-872. 2007参照) (Toe) (国立成育医療研究センター(東京、日本)の豊田らによって樹立されたものを、NIBIO(日本)のセルバンクから得た。)は、非特許文献4(Shiraki et al., Genes Cells, 2008)の記載に従って維持した。Met除去試験においては、完全維持培地で培養した未分化細胞である201B7細胞を、完全CSTI-7培地(細胞化学研究所(CSTI)、仙台、日本)またはMet除去CSTI-7培地にて、マトリゲルコートプレート上で培養した。
フィーダーフリー内胚葉分化は、iPS細胞を、1.6×105cells/cm2にて、フィブロネクチンコートしたプレートに播種し、そして翌日分化培地に切り替えた。分化培地は、100 ng/mLアクチビン、2% B27を補充したRPMI 1640培地を用いた。メチオニン除去による処理は、培地を分化培地に切り替える前に、細胞をMet除去した維持培地を用いて一定時間(例えば、5時間、または10時間)培養した。
ES/iPS細胞を、1.6×105cells/cm2にて、フィブロネクチンコートしたプレート上に播種し、翌日分化培地に切り替えた。中胚葉への分化は、50 ng/mLのBMP4(Peprotech)とITS(Invitrogen)を補充したStemline II 無血清培地(Sigma)を用いた。外胚葉への分化は、10μM SB431542(Merck)、0.2μM Dorsomorphin(Sigma)、1% N2(Invitrogen)、2% B27(Invitrogen)を補充した、RPMI 1640培地(Invitrogen)を用いた。メチオニン除去による処理は、培地を分化培地に切り替える前に、細胞をMet除去した維持培地(CSTI)で一定時間(例えば、5時間または10時間)培養した。
リアルタイムPCR、免疫細胞化学、フローサイトメトリー分析及びウェスタンブロットは、非特許文献5(Shiraki et al., PLoS One, 2011)に記載された方法に従って行った。プライマー配列を表1に示す。フォワードプライマーを上から配列番号1〜10、リバースプライマーを上から配列番号11〜20とする。また、抗体情報を表2に示す。免疫細胞化学的分析では、全細胞(DAPI陽性細胞)に対する陽性細胞を、ImageXpress Micro 細胞イメージングシステム (Molecular Devices Sunnyvale、CA)を用いて定量した。
固定した細胞をDAPIおよび抗リン酸化ヒストンH3 ser10抗体(pH3、millipore)を用いて染色した。画像は、ImageXpress Microで取得し、細胞周期のアプリケーションモジュール(モレキュラーデバイス)を用いて分析した。
アポトーシスは、インサイテュー細胞死検出キット(In Situ Cell Detection Kit:Roche、Penzberg、ドイツ)を用いて、ターミナルデオキシヌクレオチジルトランスフェラーゼ媒介d-UTPニックラベリング(TUNEL)法によって検出した。
細胞増殖は、Click-iTtm EdU Kit (Invitrogen)を用いて、細胞周期のS期の間のゲノムDNA中へのEdUの取り込みによって測定した。
Hcyの測定は、Jiangら(Talanta 77, 1279-1284., 2009)の記載に基づいて、タンデム質量分析、TQD(UPLC-MS/MS、Waters Corporation、Milford、MA、USA)を備えた超高性能液体クロマトグラフィーを用いて行った。分離は、Acquity UPLC BEH C18カラムを使用した。培養した培地を回収し、10mM DTTにて37℃で10分間インキュベートし、細胞から分泌された全Hcyを含む培地を得た。培地を50%のアセトニトリルを用いて脱タンパクし、ろ過し、次いで、等量の50mMトリス-HCl、pH8.8で希釈した。各試料を注入し、各代謝物の段階的に希釈した標準液から得られた標準曲線に基づいて、目的物のそれぞれの量を算出した。
(1)実施例1:維持培地からのアミノ酸除去による影響
未分化201B7細胞の培養を維持している間、すなわち、分化培地で分化誘導する前の未分化維持期間において、2日間維持培地から単一のアミノ酸を除去し、細胞生存率への影響を検討した。201B7細胞をマトリゲル(BD)で24時間コートした96ウェルプレート(corning)に、5x104cells/wellの密度で播種した。培養液は、未分化維持培養培地Reproff(Reprocell、日本)を用いた。播種2日後に、培養液を完全培地(Complete)および各種アミノ酸除去培地(細胞科学研究所に依頼して作成)に変更し、48時間後の細胞数について解析した。結果を図2に示す。各種アミノ酸除去培地に変更後の、48時間後の細胞数を示している。その結果、Leu、Lys、トリプトファン(Trp)、MetまたはCysの除去は、細胞数の減少をもたらすことがわかった。
未分化hiPS細胞(201B7)におけるメチオニン代謝の役割を調べるために、維持培地で、Met除去、メチルドナー(MD:メチオニン、葉酸、ビタミンB12、ベタインおよびコリン)除去、またはCys除去を行い48時間培養した。また、Met除去またはMD除去に対する、各種メチオニン関連代謝物(Met、SAM、SAH、Hcy、Cys、およびMTA)の補充効果を確認するために、各除去培地に対してそれぞれを100μMとなるようにして補充した。結果を図3に示す。MetまたはMD除去は、多能性未分化201B7細胞の生存を阻害し、これは、MetまたはSAMの補充によって救出された(図3上図)。HcyまたはMTAの補充効果を、MetまたはSAMの補充と比較した(図3下図)。細胞生存の救出において、Met補充が最も効果的であり、次いでSAMであった。Hcyの補充は、Metに比べて限定的であり、MD除去条件下での細胞生存を救うことができなかった。Hcyは、MDなしでMetには変換できないので、この結果はMetのレベルが重要であることを示している。MTAの補充は、MetおよびMDが除去された状態下での細胞枯渇を救出した。このことは、MTAがサルベージサイクルを通してMetに変換されたことを示唆している。これとは対照的に、HcyまたはCys補充は、Cys除去によって引き起こされる細胞枯渇を救った。このことは、CysおよびMetの未分化細胞の生存に対する影響が、異なったメカニズムを通して起こっていることを示している。
ヒトES細胞(khES3)を実験に使用した。未分化細胞を、コントロール完全培地で培養した場合の細胞外Hcy濃度の推移を図4左図にしめす。右図には、未分化細胞(白色)および分化5日目の内胚葉細胞(灰色)を実験材料として、コントロール完全培地(C)およびメチオニン除去培地(ΔMet)で24時間培養した場合のHcyの総排出量を示している。未分化細胞は内胚葉細胞に対して、コントロールの状況でHcy細胞外排出量が顕著に高くなった。また、未分化細胞では、メチオニン除去によりHcy細胞外排出が顕著に抑制されたが、内胚葉ではそのような抑制は見られなかった。
未分化hiPS細胞(201B7細胞)の細胞増殖に対する維持培地中のMet濃度の効果を調べた。細胞を完全維持培地およびΔMet培地で48時間培養し、細胞数の変化を確認した。結果を図5に示す。培地中のMet濃度は、120μMから12μMへと下げたとき、細胞増殖が減少した(図5A)。時間を追った試験により、細胞数および細胞増殖の有意な減少が、Met除去5時間後から観察され、SAMレベルの減少と一致した(図5BおよびC)。対照的に、アポトーシスは、Met除去の24時間後に有意に増加した(図5D)。G0−G1期停止、並びにSまたはG2−M期における細胞集団の減少は、24時間という長期のMet除去に次いで観察され、細胞内Met減少のタイミングと一致した(図5E)。以上のことより、未分化なヒトES/iPS細胞を、メチオニン除去培地で培養すると、細胞数が24時間では変化しないが48時間では顕著に減少し、このことは、細胞周期が止まり、G0/1 arrestが起きていることが起因していると考えられる。また、24時間後には細胞増殖が止まっていることはEdU陽性細胞の解析からも確認できた。
これらの結果は、未分化幹細胞が増殖を維持するためにMetを必要としていることを示唆している。また、SAMは、早期にセンサーとして機能することを示している。Met除去はSAMの減少と増殖の減少をもたらし、24時間という長期のMet除去は、細胞内Met濃度の減少を引き起こし、アポトーシスにつながった。
細胞生存障害を引き起こす責任シグナル伝達経路の特定を行った。完全培地またはMet除去培地で5時間培養した未分化hiPS細胞の遺伝子発現プロファイルを比較した。その結果、図7に示すように、細胞周期遺伝子、例えば、P21、GADD45A、GADD45B、MDM2、またはアポトーシスに関与するp53依存性遺伝子、例えば、DHRS2(Hep27タンパク質をコードする遺伝子)、EGR1、FAS、TNFRSF10A、TNFRSF10DおよびRIPK2、の発現の有意な増加が観察された。未分化hiPS細胞を使用し、培地から5時間に渡ってMetを除去した場合、EGR1、P21、およびDHRS2の転写レベルがアップレギュレートされたことを確認した。結果を図8に示す。
Met除去の5時間後に、細胞内のSAMとSAHが減少し、それは、p53−p38の活性化を誘発した。そこで、Met除去により、多能性マーカーの発現が影響されたかどうかを検討した。
ヒトiPS細胞(201B7)をマトリゲル(BD)で24時間コートした6 well plate (corning) に5x105 cells/wellの密度で播種した。培養液はReproff (Reprocell)を用いた。播種2日後に培養液をCompleteおよびメチオニン除去培地(ΔMet)(細胞科学研究所に依頼した特注培地)に変更し、5, 7, 9, 11, 13, 15, 24時間後のNanog発現についてウェスタンブロット法で解析した。結果を、図13に示す。Met除去の5時間後に、未分化hiPS細胞において、未分化マーカーであるNanog発現が減少した。一方、他の未分化マーカーであるOct3/4の発現には変化が見られなかった。これにより、未分化状態維持において、培地中からメチオニンを除去すると細胞は分化しやすい状態になることが分かった。
次いで、5〜10時間の短期のMet除去が、hiPS細胞の分化能を増大させるかどうかを検討した。上記(A)(2)および(3)に従って、ΔMet培地で5時間又は10時間培養後、内胚葉への分化培地又は中胚葉或いは外胚葉への分化培地に移して分化を行った。結果を図15に示す。SOX17+とFoxa2発現レベルの割合(図15A−C)は、対照条件に比べてMet除去で増加し、このことは、Met除去が胚性内胚葉への分化を増強することを示した。そこで、分化の増強は、他の胚葉においても起こるかどうかを検討した。hiPS細胞から、Metが奪われそして中胚葉分化に順応するように向けられたとき、Tタンパク質を発現する細胞の高い割合と、PDGFRα転写産物の高発現が観察された(図15D−F)。同様に、外胚葉および神経系譜へと分化するように向けられた場合、PAX6又はMAP2の転写産物の発現レベルは、完全培地で培養された場合と比較して増加した(図15G、H)。これらの結果は、細胞を5〜10時間という短期のMet除去に曝すことにより、NANOG発現を減少し、hiPS細胞の3胚葉への全体の分化能が増加することを示した。
10時間の短期のMet除去が、hiPS細胞の膵臓分化を促進するかを検討した。hiPS細胞(201B7)を、Met除去培地で7時間前処理した後に膵臓分化を行った。膵臓分化に関しては、既報の方法(Shahjalal H et al., J Mol. Cell. Biol., 2014、PMID 24970864)をもとに行った。22日間の培養後に細胞を4%パラホルムアルデヒドで固定して、抗インスリン抗体で染色し、核をDAPIで染色した。結果を図16に示す。Complは完全培地、ΔMetはMet除去培地で10時間前処理した後に、それぞれ膵臓分化を行った結果を示している。インスリン陽性細胞の割合は、完全培地では6%、ΔMet群では43%まで増加した。
10時間の短期のMet除去が、hiPS細胞の肝臓分化を促進するかを検討した。hiPS細胞(Toe)を、Met除去培地で5時間前処理した後に肝臓分化を行った。肝臓分化に関しては、既報の方法(Yamazoe T et al., J Cell Sci., 2013, PMID 24101719)をもとに行った。培養上清中に分泌されるアルブミンをELISAを用いて定量した。結果を図17に示す。Complは完全培地、ΔMetはMet除去培地で10時間前処理した後に肝臓分化を行った結果を示している。培養17日目(D17)においてアルブミン分泌量はΔMet群で増加することから、ΔMet前処理が肝臓分化を促進することが示唆された。
Claims (13)
- 哺乳動物由来の多能性幹細胞を分化誘導する方法であって、
(a)該多能性幹細胞を、メチオニンを含まない多能性幹細胞の未分化維持培地で培養する工程、および
(b)該未分化維持培地で培養した該多能性幹細胞を、分化誘導培地で培養して分化誘導する工程、
を含む分化誘導方法。 - 前記哺乳動物由来の多能性幹細胞が、ヒト又はマウス由来のES細胞、又はヒト又はマウス由来のiPS細胞である、請求項1に記載の分化誘導方法。
- 前記細胞を前記未分化維持培地で少なくとも5時間培養することを含む請求項1または2に記載の分化誘導方法。
- 前記細胞を前記未分化維持培地で少なくとも10時間培養することを含む請求項3に記載の分化誘導方法。
- 前記細胞を前記未分化維持培地で24時間を超えない期間で培養することを含む請求項1〜4のいずれか一つに記載の分化誘導方法。
- メチオニンを含まない多能性幹細胞の未分化維持培地で培養された哺乳動物由来の多能性幹細胞を、さらに内胚葉、中胚葉または外胚葉のいずれかに分化誘導する、請求項1〜5のいずれかひとつに記載の分化誘導方法。
- 前記多能性幹細胞が内胚葉への分化に耐性である幹細胞であって、かつ、前記分化誘導培地が内胚葉への分化誘導培地である、請求項1〜6のいずれかひとつに記載の方法。
- 前記多能性幹細胞が中胚葉への分化に耐性である幹細胞であって、かつ、前記分化誘導培地が中胚葉への分化誘導培地である、請求項1〜6のいずれかひとつに記載の方法。
- 前記多能性幹細胞が外胚葉への分化に耐性である幹細胞であって、かつ、前記分化誘導培地が外胚葉への分化誘導培地である、請求項1〜6のいずれかひとつに記載の方法。
- メチオニンを含まない多能性幹細胞の未分化維持培地で培養されたヒトiPS細胞を、さらに、膵臓分化誘導培地で培養してインスリン産生能をもつ膵臓細胞へと分化誘導する、請求項2〜5のいずれか一つに記載の分化誘導方法。
- メチオニンを含まない多能性幹細胞の未分化維持培地で培養されたヒトiPS細胞を、さらに、肝臓分化誘導培地で培養してアルブミン分泌能をもつ肝臓細胞へと分化誘導する、請求項2〜5のいずれか一つに記載の分化誘導方法。
- 培地中にメチオニンを含まない多能性幹細胞の未分化維持培地と多能性幹細胞の分化誘導培地からなる、多能性幹細胞分化誘導培地キット。
- 多能性幹細胞が、ヒト又はマウス由来のES細胞、又はヒト又はマウスのiPS細胞である、請求項12に記載の多能性幹細胞分化誘導培地キット。
