「VCAM-1」または「血管細胞接着分子1」という用語は、その転写が内皮細胞において誘導されるが、他の細胞型によっても発現される、細胞接着のためのタンパク質を意味する。VCAM-1は、1989年にOsbornら(Osbornら(1989年)、Cell. 59巻、1203〜1211頁)によって発見され、クローニングされた。VCAM-1は、特にリンパ球および単球により構成的に発現されるVLA-4(最晩期抗原4)とも呼ばれるα4β1インテグリンと相互作用する。ICAM-1、2および3などの他の接着分子と同様に、VCAM-1は、アテローム動脈硬化症時の内皮への単球の接着に関与する。VCAM-1はまた、腸にリンパ球を動員するためにα4β7インテグリンと相互作用する。
本発明に関して、「抗体」および「免疫グロブリン」という用語は、同じ意味を有し、無差別に用いられる。通常の抗体において、2つの重鎖は、ジスルフィド架橋により互いに結合しており、各重鎖は、ジスルフィド架橋により軽鎖に結合している。2種類の軽鎖、すなわち、ラムダ(λ)およびカッパ(κ)軽鎖が存在する。抗体の機能活性を決定する重鎖の5つの主なクラス(またはアイソタイプ)、すなわち、IgM、IgD、IgG、IgAおよびIgEが存在する。各鎖は、異なる配列ドメインを含む。軽鎖は、2つのドメイン、すなわち、可変ドメイン(VL)および定常ドメイン(CL)を含む。重鎖は、4つのドメイン、すなわち、可変ドメイン(VH)および3つの定常ドメイン(CH1、CH2およびCH3、CHと総称する)を含む。重鎖(VH)および軽鎖(VL)の可変領域は、結合の認識および抗原に対する特異性を決定する。軽(CL)および重(CH)鎖の定常領域のドメインは、抗体の鎖の会合、分泌、経胎盤移動度、補体への結合およびFc受容体への結合などの重要な生物学的特性を与える。断片Fvは、軽鎖および重鎖の可変部からなる免疫グロブリンのFab断片のN末端部分である。抗体の特異性は、抗体の結合部位と抗原決定基との間の構造的相補性にある。抗体の結合部位は、超可変領域または相補性を決定する領域(CDR)に主として由来する残基で構成されている。時折、非超可変領域または「フレームワーク」領域(FR)の残基がドメインの全体的な構造およびしたがって結合部位に影響を与えることがある。

本発明によるナノボディは、医用イメージングに特に有用である。これに関連して、ナノボディを検出可能なマーカーと結合させることは、特に興味深い。本発明者らは、ナノボディcAbVCAM1-5、cAbVCAM1-3、cAbVCAM1-8、およびcAbVCAM1-9が、検出可能なマーカー、特にテクネチウム99mなどの放射性核種と結合させた場合にそれらの特性を保持していたことを示した。
本発明者らは、ナノボディcAbVCAM1-5、cAbVCAM1-3、cAbVCAM1-8、およびcAbVCAM1-9がアテロームプラーク、特に大動脈プラークの特異的トレーサーを形成し、イメージングによるその検出を可能にすることを示した。
アテロームプラークの出現は、それ自体が心血管疾患であり、様々な心血管合併症を発生させ得るアテローム動脈硬化症の徴候である。したがって、アテロームプラークの検出は、心血管疾患の特徴付けである。他方で、イメージングによる発達、すなわち、事前に同定されたアテロームプラークの進行または退縮を追跡調査する可能性は、心血管疾患が診断された患者における治療処置の有効性を評価する方法である。
a)アテロームプラークが検出された対象に上で定義したナノボディを投与するステップと、
b)前記アテロームプラークにおけるナノボディとの結合を検出するステップと、
c)前記対象に治療を施す前と後にステップa)およびb)を反復するステップと
からなるステップを含むことができ、
前記プラークにおけるナノボディの結合の欠如または減少が治療効果を有する治療の指標である。
本発明はさらに、薬物、特に、心血管疾患を治療することを目的とする薬物を製造するための上で定義したナノボディの使用に関する。それを必要とする患者への治療上有効量の上で定義したナノボディの投与を含む治療方法も本発明の一部である。
本発明はまた、試料中のVCAM-1のインビトロでの検出のための上で定義したナノボディの使用に関する。
ナノボディの発生および生産
VCAM-1を標的とするナノボディは、一般的に以前に公表された方法(Arbabi Ghahroudiら(1997年)、FEBS Lett.、414巻、521〜526頁)に従って発生させた。より具体的には、ヒトコブラクダをヒトおよびマウス組換えVCAM-1タンパク質(RnD Systems、Minneapolis、USA)の両方により免疫化した。血中リンパ球を単離し、RNAを精製した。単鎖抗体の可変ドメイン(VHHまたはナノボディ)を2段階RT-PCR法を用いて増幅し、バクテリオファージM13のタンパク質3と同時性でクローニングした。ナノボディをファージディスプレイに供し、固定化免疫原上のバイオパニングに用いた。可溶性ナノボディを含む全細菌抽出物を、TNFαで刺激したbEND5細胞におけるELISAおよびフローサイトメトリーにおける陽性シグナルに基づいて個々のVCAM-1リガンドを選択するために用いた。配列決定の後、選択した抗VCAM-1ナノボディおよび無関係の対照ナノボディcAbBcII10を大腸菌中でヘキサヒスチジンによるタグ付きタンパク質として生産させ、Saerensら(2005年)、J. Mol. Biol.、352巻、597〜607頁に記載されているように精製した。
細胞株-bEND5マウスの内皮細胞株(ECACC、Salisburg、GB)を補足DMEM中で培養した。VCAM-1の発現は、18時間にわたる10ng/mLのTNFαによる刺激により誘導した。
Gainkamら(2008年)、J. Nucl. Med.、49巻、788〜795頁に記載されているようなトリカルボニル法を用いることにより、ナノボディを99mTcで放射性標識した。放射化学的純度は、標識後直ちに、PBS中20℃で6時間後に、および注射後(p.i.)3時間目にマウス血液中で評価した。後者の場合、100μLの試料採取した全血を遠心分離し、血漿をNanosep 10K Omega膜を用いてろ過した。放射化学的純度は、ACN/TFA勾配で移動相により溶離するカラムC4を用いることによりRP-HPLCにより測定した。放射能は、放射線検出器(γ-RAM Model 4、LabLogic)を用いることにより観測した。
250×103個のbEND5細胞を24ウエルプレート(plaques)に播種し、10ng/mLのTNFαで18時間にわたり刺激した。5nMの各ナノボディ-99mTcを0.5mLのPBS+1%のヒト血清アルブンミン(human albumen serum)(HAS)中で37℃で1.5時間インキュベートした。結合特異性を測定するために、500倍過剰の非標識ナノボディを用いた競合試験を行った。洗浄後、結合99mTc -ナノボディを収集し、ガンマ線計数器(Cobra II Inspector 5003、Canberra Packard、USA)で計数した。実験は、3連で行った。ウエルへの非特異的結合を差し引き、結果をTNFα陰性条件に対して正規化した。
すべての動物実験は、「Recherche du Service de Sante des Armees de Grenoble(CRSSA)」(グレノーブルの陸軍医療保健施設の研究センター)の委員会により承認された。31±1週齢の雌対照マウスおよびApoE-/-マウスを用いた(Charles-River、France)。ApoE-/-マウス(n=37)に0.39%のコレステロールを含むWestern飼料(Safe、France)を給餌したが、C57Bl/6J対照マウス(n=9)には標準飼料を与えた。C57Bl/6J対照マウス(n=4)においてさらに評価したナノボディcAbVCAM1-5(n=6)を除いて、各抗VCAM-1ナノボディを3匹のApoE-/-マウスにおいて評価した。抗体cAbBcII10は、ApoE-/-マウス(n=4)および対照マウス(n=5)の両方において負の対照として評価した。99mTcで放射性標識したナノボディの投与(67±4MBq i.v.)の2時間後に、2%イソフルラン(Baxter)を用いてマウスを麻酔し、SPECT/CT収集を行った(nanoSPECT、Bioscan、下文参照)。マウスを過量のペントバルビタールナトリウム(CEVA)により安楽死させ、大動脈ならびに他の主要臓器を注意深く回収した。大動脈を10%のホルマリンの溶液中で付着組織から分離し、上行大動脈から腸骨動脈まで12セグメントに切断した。病変の拡大指標を次のように各セグメントに割り当てた:(-)病変なし(対照セグメント)、(+)動脈セグメントの長さの50%までの範囲にわたる病変、(++)動脈セグメントの長さの50%を超える範囲にわたる病変および(+++)セグメントの全長に広がる病変。大動脈セグメントおよび組織試料の重量を測定し、それらの放射能をウエルガンマ線計数器(Cobra II、Packard)により測定した。結果は、バックグラウンドノイズおよび放射性崩壊に関して補正し、1gの組織当たりの注射量の百分率(%ID/g)として表した。大動脈病変および対照の吸収は、それぞれ(+++)または(-)と分類されたすべてのセグメントにおける平均吸収と定義した。病変対対照、病変対血液および病変対心臓比も測定した。
10%のロバ血清を用いて20℃で1時間のブロッキングの後、抗VCAM-1ヤギ一次抗体(Santa-Cruz Biotechnology、0.5μg/ml)を大動脈切片に4℃で一夜適用した。抗ヤギロバビオチニル化二次抗体(Jackson ImmunoResearch)を20℃で1時間インキュベートし、DABを発色剤として用いた。切片をヘマトキシリンにより対比染色した。マーキング特異性は、対照スライド上の一次抗体を省略することによって確認した。対照マウスおよびApoE-/-マウスのサブグループにおいて、VCAM-1免疫マーキングを心臓、筋肉、唾液腺、骨髄、リンパ神経節、脾臓および胸腺について行った。
静脈内注射の2時間前に、麻酔動物を温度調節機能付きベッドにおき、全身SPECT/CT収集を行った(nanoSPECT、Bioscan)。おおよそのCT収集時間は、次の収集パラメーターを用いることにより10分であった:45kVp、240投影および1投影当たり1,000ms。らせんSPECT収集は、24投影および45分の収集を用いることにより1mmの分解能を有する小開口径(stenopeic)多孔コリメーターを装着した4ヘッド(ヘッド当たり9×1.4mm(全直径))で行った。CTおよびSPECT収集を専用ソフトウエアパッケージ(InVivoScope)を用いて再構成し、マージし、定量化した。弓対血液および弓対心臓比を測定するために、関心領域(ROI)を大動脈弓、左心室腔および左心室壁に描いた。
各動物について、12大動脈セグメントの異なるレベルで得られた20μmの厚さを有する3群のスライドの一夜暴露の後にオートラジオグラフ画像が得られた。画像を専用ソフトウエアパッケージ(Image Gauger、Fujifilm)を用いて定量化した。アテローム動脈硬化性病変および負の対照VCAM-1大動脈壁の周りにROIを描いた。結果をバックグラウンドノイズについて補正し、平均病変対対照比の形で表した。
ウサギVCAM-1をコードするプラスミドを一時的にトランスフェクトした、またはしなかった105個のCHO細胞を1μgのナノボディ、1μgの抗Hisタグモノクローナル抗体(Serotec)および200ngの抗マウスラットIgG1(BD Biosciences)とともに連続的にインキュベートした。結合をFACS Canto II分析装置(BD Biosciences、Franklin Lakes、USA)で測定し、データをFlowJoソフトウエアパッケージ(TreeStar、Ashland、USA)により解析した。負の対照は、ナノボディを省略することによって実施した。
すべての結果を平均値±平均値の標準誤差の形で表示した。対応のあるデータおよび対応のないデータを比較するためにマン-ホイットニーのUおよびウィルコクソンのノンパラメトリック検定を用いた。差は、p<0.05で有意とみなした。
抗VCAM-1ナノボディの発生
本発明者らは、ヒトおよびマウスVCAM-1に対する交差反応ナノボディを開発しようとした。したがって、ナノボディは、ヒトおよびマウス組換えタンパク質VCAM-1によりヒトコブラクダを免疫化し、それによってもたらされたファージディスプレイにより得られた免疫ナノボディのライブラリーをバイオパニングすることにより発生させた。個々のナノボディを含む全細菌抽出物を、VCAM-1組換えタンパク質とのそれらの結合についてELISAで、VCAM-1を発現するbEND5細胞へのそれらの結合についてフローサイトメトリーでスクリーニングした。
bEND5細胞におけるフローサイトメトリー実験により、12の選択したナノボディがVCAM-1と相互作用したことが示された(図1)。Table 4(表4)に要約したSPR分析により示されたように、すべての選択したナノボディが0.2〜45.7nMの範囲の高い親和性でVCAM-1に結合した。さらに、10種の選択したナノボディのうち、6種は、1ナノモル程度にとどまる親和性でhVCAM-1と交差反応性を示した(Table 4(表4))。これらの6種のナノボディは、cAbVCAM1-1、cAbVCAM1-3、cAbVCAM1-5、cAbVCAM1-8、cAbVCAM1-9およびcAbVCAM1-10であった。SPR競合試験に基づいて、ナノボディcAbVCAM1をエピトープターゲティングに関してcAbVCAM1-1/5、cAbVCAM1-2/3/6/7/9/10およびcAbVCAM1-4/8の3つのカテゴリーに分類した。すべてのナノボディが59.4〜87℃超の範囲の開折温度によって示されたように強い熱安定性を示した(Table 4(表4))。
ApoE-/-マウスにおける99mTcにより標識されたナノボディの体内分布をTable 5(表5)に要約する。対照cAbBcII10を含むすべてのナノボディは、97±16〜315±33%ID/gと腎臓における強い吸収および膀胱における強い活性を示した。興味深いことに、99mTc-cAbVCAM1の吸収は、リンパ組織において高く、99mTc-cAbVCAM1-3(脾臓および胸腺)、99mTc-cAbVCAM1-4/5(脾臓、胸腺および骨髄)および99mTc-cAbVCAM1-9(胸腺)については統計的差に達しすらした(p<0.05対99mTc-cAbBcII10)。肺および肝臓(それぞれ2.5±0.8および2.7±0.9%ID/gの平均吸収)を除いて、吸収は、血液および心筋を含む他の試験した組織において2%ID/g未満であった。
図3に示すように、VCAM-1の構成的発現が、対照マウスC57Bl/6Jおよび高コレステロール血症マウスApoE-/-の両方におけるリンパ組織(すなわち、骨髄、リンパ神経節、脾臓および胸腺)に認められたが、心臓、筋肉および唾液腺におけるVCAM1の発現は認められなかった。さらに、強いVCAM-1マーキングが大動脈病変、内皮ならびにアテローム動脈硬化性プラーク内にも認められたが、C57Bl/6J対照マウスの大動脈には認められなかった。
Table 4(表4)に要約した選択基準に基づいて、ナノボディcAbVCAM1-5、cAbVCAM1-3、cAbVCAM1-8およびcAbVCAM1-9は、医用イメージングにおける放射性トレーサーとして用いるのに最も有望であると本発明者らによりみなされた。ナノボディcAbVCAM1-5をさらなる評価の対象として特に選択した。
本試験は、そのような交差反応性リガンドが十分に特徴付けがなされた動物モデルにおいてバリデートされた結果の臨床診療への移転のために特に興味深い程度に、ヒトおよびマウスVCAM-1同族体の両方を認識するナノボディを発生させるために本発明者らにより開発された。6種の交差反応性ナノボディを含む、1ナノモル程度のマウスおよび/またはヒト同族体に対する親和性を有する10種の抗VCAM-1ナノボディを発生させ、生産した。さらに、2種のナノボディcAbVCAM1-1およびcAbVCAM1-5は、ウサギVCAM-1に対して交差反応性である。ナノボディ、特にcAbVCAM1-5の高い耐熱性は、高い放射化学的純度(>95%)を有する99mTcにより50℃でそれらを放射性標識する可能性をもたらした。さらに、陽性VCAM-1マウス内皮細胞におけるインビトロ結合試験により、これらの標識ナノボディがmVCAM1の特異的リガンドのままであったことが明らかにされている。
アテローム動脈硬化性病変に吸収されることに加えて、本発明によるナノボディは、体内分布およびSPECTイメージング実験により示されたように、正常および高コレステロール血症マウスの両方におけるリンパ組織によっても吸収された。特に、mVCAM-1に対する最も強い親和性を有する3種の交差反応性ナノボディ(cAbVCAM1-5/3/9)は、脾臓および骨髄において最も強い吸収を示した。mVCAM-1の構成的発現が免疫組織化学検査により脾臓、骨髄、リンパ神経節および胸腺に認められた。したがって、リンパ組織への抗VCAM-1抗体の結合は、VCAM-1タンパク質へのそれらの特異的結合におそらく起因していた。
Table 4(表4)に要約したパラメーターに基づいて、ナノボディcAbVCAM1-5、cAbVCAM1-3、cAbVCAM1-8およびcAbVCAM1-9は、特に医用イメージングにおける使用を視野に入れると、他のナノボディと比較して特に好都合な特性を有すると本発明者らによって確認された。最も有望なナノボディは、ナノボディcAbVCAM1-5である。実際、cAbVCAM1-5は、最高の病変対対照および病変対心臓比ならびに強い病変対血液比を示す。さらに、それは、ヒトおよびマウスVCAM-1の両方に対する十分な親和性を有し、最高の耐熱性および最高の生産収量を有する。最後に、cAbVCAM1-5は、アテローム動脈硬化症の試験における参照動物モデルである、ウサギVCAM-1に対して交差反応性である。cAbVCAM1-3は、最高の病変対血液比ならびに強い病変対対照および病変対心臓比を有する。cAbVCAM1-8は、特に高い病変対心臓比およびヒトVCAM-1に対する最高の親和性を有する。最後に、cAbVCAM1-9は、アテローム動脈硬化性病変における最高の取込みならびにヒトおよびマウスVCAM-1に対する非常に十分な親和性を有する。さらに、cAbVCAM1-5、cAbVCAM1-3およびcAbVCAM1-9は、それらのCDRにおいてリシンを有さない。これは、リシンの存在が、PETイメージングを目的としてナノボディをそれらのアミン残基を介して蛍光または放射性標識により標識することを可能にするカップリング技術に対して不利な点を有し得る場合に有利である。
99mTc-cAbVCAM1-5は、HPLCによって示されたように、インビトロで放射性標識後6時間までならびにインビボでマウス血液中で安定である。これは、注射後3時間目にSPECT/CTイメージングを適用する可能性をもたらす。この時に、ApoE-/-マウスの大動脈弓に位置していたアテローム動脈硬化性病変は、心筋および血液における低いバックグラウンドノイズ活性で、SPECT/CTイメージングによって本発明者らにより成功裏に同定された。オートラジオグラフィーおよび免疫組織化学検査により、99mTc-cAbVCAM1-5の大動脈吸収が陽性VCAM-1アテローム動脈硬化性病変に集中していたことが確認された。したがって、これらの実験から、99mTc-cAbVCAM1-5は、アテローム動脈硬化性病変において起こる炎症過程の非侵襲的なインビボイメージングのための適切な放射性トレーサーであることがわかる。
抗体由来の他の放射性トレーサーがSPECTを用いて易破綻性アテロームプラークを撮像することについて最近評価された(Temmaら(2010年)、J. Nucl. Med.、51巻、1979〜1986頁、Kugeら(2010年)、Eur. J. Nucl. Med. Mol. Imaging、37巻、2093〜2104頁)。しかし、完全な抗体のクリアランスが低いため、標的対バックグラウンドノイズ比が最適に及ばないものとなり、抗体断片を用いる必要性が力説される。アテローム動脈硬化性病変のSPECTまたはPETイメージングについて以前に評価された他の放射性トレーサーのうち、18FDGは、ヒトにおける頸動脈病変のインビボでのイメージングを可能にする、マクロファージにおける高い吸収を示した(Ruddら(2002年)、Circulation、105巻、2708〜2711頁)。しかし、心筋における強いバックグラウンドノイズのため、心筋における吸収を低減することを意図した特定の食事を使用する可能性があるにもかかわらず、冠病変のイメージングは依然として非常に困難なままである(Wykrzykowskaら(2009年)、J. Nucl. Med.、50巻、563〜568頁)。同様に、アテローム動脈硬化症マウスモデルにおいて、Laitinenらは、注射後1時間目の18FDGの心筋における吸収は、アテローム動脈硬化性病変における0.41±0.16%ID/gに対して18.13±10.59%ID/gであったことを示した(Laitinenら(2006年)、Eur. J. Nucl. Med. Mol. Imaging、33巻、1461〜1467頁)。
本発明者らは、医用イメージングに使用するための重要な特性を有する、ヒトVCAM-1に対して交差反応性の4種の抗VCAM-1ナノボディを同定し、生産した。本発明者らは、特に、99mTc-cAbVCAM1-5が、SPECT/CTイメージングによりインビボでアテローム動脈硬化性病変を成功裏に同定する可能性をもたらしたことを直接的に示した。
Claims (15)
- a)アミノ酸配列(i)CDR1としてのYTNSIMYMA(配列番号1)、(ii)CDR2としてのAIRFPDDS(配列番号2)および(iii)CDR3としてのRSSPYSFAWNDPSNYNY(配列番号3)、または
b)アミノ酸配列(i)CDR1としてのFTYSSYYMS(配列番号4)、(ii)CDR2としてのGINVDGSN(配列番号5)および(iii)CDR3としてのGSGRDSYDCYSGSWCP(配列番号6)、または
c)アミノ酸配列(i)CDR1としてのFTFSNYYMT(配列番号7)、(ii)CDR2としてのRINSDGS(配列番号8)および(iii)CDR3としてのGKSSV(配列番号9)、または
d)アミノ酸配列(i)CDR1としてのFTFSSYYMS(配列番号10)、(ii)CDR2としてのGINVDGSN(配列番号11)および(iii)CDR3としてのGSGRDSYDCYSGSWCP(配列番号12)
を含むVCAM-1に対するナノボディ。 - 配列番号13、配列番号14、配列番号15および配列番号16のアミノ酸配列からなる群から選択されるアミノ酸配列を含む、請求項1に記載のナノボディ。
- 配列番号13のアミノ酸配列からなる、請求項1または2に記載のナノボディ。
- 検出可能なマーカーと結合されている、請求項1から3のいずれか一項に記載のナノボディ。
- 前記検出可能なマーカーが放射性核種である、請求項4に記載のナノボディ。
- インビボでの非侵襲的医用イメージングにおける造影剤として使用するための、請求項1から5のいずれか一項に記載のナノボディ。
- 診断または予後診断法において使用するための、請求項1から6のいずれか一項に記載のナノボディ。
- 薬物として使用するための、請求項1から3のいずれか一項に記載のナノボディ。
- 試料中のVCAM-1のインビトロでの検出のための、請求項1から5のいずれか一項に記載のナノボディの使用。
- 請求項1から3のいずれか一項に記載のナノボディを、薬学的に許容される担体とともに含む医薬組成物。
- 請求項1から3のいずれか一項に記載のナノボディをコードする核酸配列を含む核酸。
- 請求項11に記載の核酸を含むベクター。
- 請求項11に記載の核酸または請求項12に記載のベクターがトランスフェクトされた、それに感染した、またはそれで形質転換された細胞。
- 請求項1から3のいずれか一項に記載のナノボディを発現する組換え宿主細胞を生産する方法であって、
(i)請求項11に記載の核酸または請求項12に記載のベクターをインビトロまたはエクスビボでコンピテント宿主細胞に導入するステップと、
(ii)得られた組換え宿主細胞をインビトロまたはエクスビボで培養するステップと、
(iii) 場合によって、前記ナノボディを発現するかつ/または分泌する細胞を選択するステップと
を含む、前記方法。 - 請求項1から3のいずれか一項に記載のナノボディを生産する方法であって、
(i)前記ナノボディの発現を可能にするための適切な条件下で請求項13に記載のトランスフェクト細胞または感染細胞または形質転換細胞を培養するステップと、
(ii)発現ナノボディを回収するステップと
を含む、前記方法。
