Claims (11)
- 流体中の細胞を分離する装置であって、
サンプル入口(20)に連通される第1の駆動ポンプ(1)と、分離カラム(3)と、検出カラム(4)と、分離カラムで分離された細胞を検出カラムに入れ込むように駆動するための第2の駆動ポンプ(2)と、複数の三方弁およびサブ駆動ポンプと、を備え、
前記分離カラム(3)は、一端が第1の三方弁(5)を介して第1の駆動ポンプ(1)に接続され、他端が第2の三方弁(6)を介して第2の駆動ポンプ(2)に接続され、前記検出カラムは、第1の三方弁(5)の3番目のチャンネルポートに接続され、前記第2の三方弁(6)の3番目のチャンネルポートは、サンプル出口(21)に接続され、
細胞液体容器(18)は、前記装置とともに循環通路を形成できるように、前記サンプル入口(20)とサンプル出口(21)との間に着脱可能に接続され、
前記装置は、サンプル入口(20)と、第1の駆動ポンプ(1)と、第1の三方弁(5)と、分離カラム(3)と、サンプル出口(21)と、細胞液体容器(18)と、サンプル入口(20)と、を順次に連通させる主循環管路(1a)をさらに備え、
主循環管路に抗凝固生理食塩水を注入するための第3の三方弁(9)は、主循環管路に接続されるとともに、サンプル入口(20)と分離カラム(3)との間に位置し、主循環通路に抗凝固拮抗剤を注入するための第4の三方弁(10)は、主循環管路に接続されるとともに、サンプル出口(21)と分離カラム(3)との間に位置し、前記サブ駆動ポンプは、第3の三方弁(9)の3番目のチャンネルポートおよび/または第4の三方弁(10)の3番目のチャンネルポートに接続され、
前記分離カラム(3)は、並列する少なくとも5本のサブ濾過カラム(19)を含み、前記サブ濾過カラムは、固定ブラケット(13)と、固定ブラケットのボトム及び側面周りに付けられる、ポリカーボネートまたはポリエステル材で孔径が5−25μmであるトラックエッチ膜(7)とを含み、
前記検出カラム(4)は、活動可能に接続される2部分を備え、2部分の自由端外側にはいずれも、管路に適応する液体チャンネルポート(8)が設けられ、前記2部分の間には密閉中空キャビティが形成され、かつ、中空キャビティには、ポリカーボネートまたはポリエステル材で孔径が5−25μmであるトラックエッチ膜が一つまたは複数の層設けられている装置。 - 前記サブ駆動ポンプは第1のサブポンプ(11)と第2のサブポンプ(12)とを含み、第1のサブポンプ(11)は、前記第3の三方弁(9)の3番目のチャンネルポートに接続され、第2のサブポンプ(12)は、前記第4の三方弁(10)の3番目のチャンネルポートに接続される、ことを特徴とする請求項1に記載の装置。
- 前記第3の三方弁(9)は、サンプル入口(20)と第1の駆動ポンプ(1)との間、または、第1の駆動ポンプ(1)と分離カラム(3)との間に位置し、前記第4の三方弁(10)は、サンプル出口(21)と第2の三方弁(6)との間、または、第2の三方弁(6)と分離カラム(3)との間に位置する、ことを特徴とする請求項1に記載の装置。
- 前記検出カラム(4)におけるトラックエッチ膜(7)の孔径は、前記分離カラム(3)におけるトラックエッチ膜(7)の孔径以下である、ことを特徴とする請求項1から3のいずれか一つに記載の装置。
- 前記分離カラム(3)は、サンプル入口(20)に近接する端に位置する液体分散エリアと、中央に位置するサブ濾過カラム(19)とを含む、ことを特徴とする請求項1から3のいずれか一つに記載の装置。
- 前記液体分散エリアは、一端面が、液体チャンネルポート(8)が開口しているポートカバープレート(15)であり、他端面が、すべての前記サブ濾過カラムの端面に対応するスルーホールが開口している支持プレート(14)である、ことを特徴とする請求項5に記載の装置。
- 前記第1の駆動ポンプと前記第2の駆動ポンプは、蠕動ポンプである、ことを特徴とする請求項1から3のいずれか一つに記載の装置。
- 前記第1のサブポンプまたは第2のサブポンプは、蠕動ポンプまたはプッシュ式輸液ポンプである、ことを特徴とする請求項2に記載の装置。
- 前記主循環管路に設けられるとともに第1の駆動ポンプ(1)と分離カラム(3)との間に位置する圧力センサと、管路ヒータとをさらに備える、ことを特徴とする請求項1から3のいずれか一つに記載の装置。
- 前記分離カラム(3)および検出カラム(4)におけるトラックエッチ膜は、いずれも孔径が8‐10μmである、ことを特徴とする請求項1から3のいずれか一つに記載の装置。
- 前記検出カラム(4)は、可塑化ポリ塩化ビニルからなる、ことを特徴とする請求項1から3のいずれか一つに記載の装置。
