Description
用いた抽出手順を上述したが、先に概略された多くの工程に用いることができる別の方法が多数存在するが、これらの方法は、論理的拡張である。テープストリップからミエロペルオキシダーゼ及び酸化脂質を分離させるために用いる抽出溶媒は、好適な回収をもたらす、任意の適切な水性、有機、又は有機/水性混合物であってよい。LC/MS/MS及び抗体系イムノアッセイは概して、それらの高選択性及び感度のために生物学的マトリックスでの有機分子の定量分析のための最先端の手法として認識されている。しかし、要求される感度及び選択性を提供する任意の分析法及び/又は他の方法を用いてよい。例えば、キャピラリー電気泳動、超臨界流体及び他のクロマトグラフィー法、並びに/又はこれらの組み合わせを含む、生体分子を評価するための他の方法が使用されてきた。同様に、また、核磁気共鳴分光法、質量分析法、電気化学及び蛍光アッセイを含む、分離法を伴わない器具手法も使用されてきた。更に、競合及び非競合酵素結合免疫吸着アッセイ(ELISA)及びラジオイムノアッセイ(RIA)などリガンド結合法又は他の標識化スキームも用いられてきた。酵素系アッセイには、タンパク質分析において長い使用歴がある。細胞系又は組織系の手法のいずれかを用いるバイオアッセイも、検出手段として用いられてきた。本発明の一実施形態では、毛髪プラックからの酸化的ストレス及び酸化的損傷のバイオマーカーの定量化は、テープストリップサンプルで使用されるのと同じ基本的な抽出及び分析法で行うことができる。
上述の好適な方法を用いて測定した皮膚のテープストリップサンプル上のミエロペルオキシダーゼのレベルは、テープストリップ抽出物に見られるタンパク質の量を用いて正規化することができる。正規化は、テープストリップ抽出物中のタンパク質の量でミエロペルオキシダーゼの量を除して行う。
Hsp27の細胞保護特性は、その活性酸素種を調節する能力及びグルタチオンレベルを上昇させる能力からもたらされる。
生物系におけるタンパク質の酸化は、システイニルチオールの自発的自動酸化、タンパク質の活性酸素種(ROS)との相互作用、及びオキシダーゼにより触媒される意図的かつ制御された反応により起こる。タンパク質のROSとの反応は、システイン、メチオニン、チロシン、フェニルアラニン、及びトリプトファン残基の酸化をもたらし得る。メチオニン酸化はメチオニンスルホキシドの測定により、チロシンの酸化は形成されるジチロシン量により、フェニルアラニンの酸化はo−チロシン及びm−チロシンの形成によりモニターされ、トリプトファン残基の酸化は、N−ホルミルクヌレニン、キヌレニン、及び/又はキノリン酸をモニターすることにより追跡される。また、アルブミンcys34残基の共有結合的及び酸化的修飾は、軽い酸化的ストレスの特異的バイオマーカーであることが示唆されている。通常、これらのタンパク質修飾付加残基は、完全な酵素加水分解又は化学消化後に測定される。
DNA損傷は概して、酸化的ストレスの1つの物差しであり、主な理由は、内因性活性酸素種(ROS)により引き起こされるフリーラジカル損傷のためである。無傷のDNAへの酸化的損傷は、COMETアッセイを用いて測定することができる。酸化的損傷は、8−オキソデオキシグアノシン(8−oxodG)とも呼ばれる8−ヒドロキシデオキシグアノシン(8-hydroxydexoygaunosine)(8OHdG)、4,6−ジアミノ−5−ホルム−アミドピリミジン(FapyAde)、及び2,6−ジアミノ−4−ヒドロキシ−5−ホルムアミドピリミジン(FapyGua)を含む、種々のヒドロキシル化ヌクレオチドをモニターすることにより測定することができる。
イソプロスタンは、アラキドン酸(AA)及びω−3−エイコサペンタエン酸(EPA)などの多価不飽和脂肪酸(PUFA)のフリーラジカル触媒酸化から形成される一連のプロスタグランジン様アイソマーであり、典型的にPUFAの酸化はリン脂質形態に起こる。AA酸化に由来するイソプロスタンは、F型プロスタン環(F2−イソプロスタンと呼ばれる)をもたらし、5−F2−シリーズIsoP、8−F2−シリーズIsoP、12−F2−シリーズIsoP、及び15−F2−シリーズIsoPs8−シリーズを生じさせる。同様に、D/E−環及びA/J環イソプロスタンも、AAから形成される。F3−ファミリーのイソプロスタン(Isoprosanes)はEPAから同様に形成され、5−F3−シリーズ、8−F3−シリーズ、11−F3−シリーズ、12−F3−シリーズ、15−F3−シリーズ、及び18−F3−シリーズイソプロスタンを生じさせる。
多価不飽和脂肪酸の酸化は、酸化的ストレスに応答して起こることが多く、脂肪酸ヒドロペルオキシドのラジカル駆動形成をもたらし、これは、更なる反応を経て、生物学系で広い多様性のα,β−不飽和アルケナール、例えばマロンジアルデヒド、アクロレイン、クロトンアルデヒド、4−ヒドロキシノネナール、4−ヒドロキシヘキセナール、及び4−オキソ−ノネナールを得ることができる。これらのα,β−不飽和アルケナールは、酸化的ストレスのバイオマーカーとして使用されている。
α,β−不飽和アルケナールは、反応性求電子試薬であり、ヘキサノイル−リジン、ヘキサノイル−ヒスチジン、Ne−(3−メチルピリジニウム)リジンなどの後期リポキシド化最終生成物(ALE)と呼ばれる共有結合タンパク質付加物などの数多くの求核化合物と、及びメルカプツール酸化合物(グルタチオン、システイン、及びメルカプツール酸)と生成物を形成して、チオエーテル(例えば1,4−ジヒドロノンメルカプツール酸、3−ヒドロキシプロピルメルカプツール酸、及びカルボキシエチルメルカプツール酸)を形成する。
タンパク質の糖化は、集合的にメイラード反応と呼ばれる非酵素複合体シリーズの並発及び連続反応であり、全ての組織及び体液で起こる。糖化付加物は、グルコースと、反応性α−オキソアルデヒド、例えばグリオキサール、メチルグリオキサール、及び3−デオキシグルコソン、並びにその他の糖類誘導体と、のタンパク質の反応により形成することができる。糖化の初期段階反応は、フルクトシル−リジン及びN−末端アミノ酸残基由来フルクトサミンの形成をもたらし、初期糖化付加物(EGA)と呼ばれる。後期段階反応は、後期糖化最終生成物と呼ばれる安定な最終段階付加物、例えばモノリシル付加物(カルボキシメチルリジン(CML)、カルボキシエチルリジン(CEL)、及びピラリン)、モノバリル付加物(カルボキシメチルバリン(CMV)及びカルボキシエチルバリン(CEV))、ヒドロイミダゾロン、ビス(リシル)イミダゾリウム架橋(GOLD、MOLD、DOLD)、及びリジンとアルギニンとの架橋に由来するペントシジンを形成する。EGA及びAGEは、タンパク質がタンパク質分解により分解されたとき又はタンパク質が化学溶解により分解されたとき、放出される。AGEの形成及び蓄積は、加齢関連疾患の進行に関係付けられてきた。ここ20年間にわたる研究は、加齢に関連付けられる疾患のほとんどでAGEを暗示している。CMLは、加齢、アテローム性動脈硬化症、及び糖尿病におけるタンパク質への酸化的ストレス及び長期損傷の一般的マーカーであり得る。
内因性抗酸化剤は、ラジカル機構により引き起こされる酸化的損傷の制御において鍵となる防御的役割を果たす。鍵となる内因性抗酸化剤は、アスコルビン酸(AsA)、グルタチオン(GSH)、α−トコフェロール、及びコエンザイムQ10(CoQ)を含む。これらの内因性酸化還元(酸化及び還元形態)抗酸化剤のレベルの変化は、酸化的ストレスの測定値として使用することができる。
被験者を、頭皮の状態を確立するために認定検査員により評価する。フケ被験者について、認定検査員により評価される眼に見えるフレーク化のレベルを特定する。非フケの健康な頭皮の被験者は、各バイオマーカーの正常化レベルを確立するために基準時で評価される。被験者は、2つの別個の臨床研究(調査番号1及び調査番号2)において認定検査員によって認定された。調査番号1には、基準時のMPOレベルで評価される60人の非フケ被験者が含まれる。119人のフケ患者に1% ZPT含有フケ防止シャンプーを適用し、115人のフケ患者に1%硫化セレン含有フケ防止シャンプーを適用し、60人のフケ被験者にフケ防止有効成分を含有しない非フケシャンプーを適用した。被験者を基準時、並びに製品使用の1週間後、2週間後、及び3週間後のMPOレベルについて評価する。調査番号2には、基準時の酸化脂質で評価される30人の非フケ被験者が含まれる。60人のフケ被験者にフケ防止シャンプーを適用し、60人のフケ被験者にフケ防止有効成分を含有しない非フケシャンプーを適用した。被験者を基準時、及び製品使用の3週間後に再び酸化脂質について評価した。
データの統計分析:データは、基準時及び治療後のフケ被験者並びに基準時の非フケ被験者について収集した。被験者間並びに被験者内(基準時→治療)での検体レベルの大幅な多様性を考慮すると、分析前にデータをlog10スケールに変換することがより効率的である。統計分析は、同等のlog10比値(即ち、log10(第3週/基準時)に基づいて実行した。次に、報告された最終結果を元のスケール(又は、基準時値からの%変化)に再変換して、解釈を容易にする。全ての統計分析は、SAS JMP(バージョン7.0.2)ソフトウェアを用いて実行した。統計的有意性の判定には、p値は≦0.20(両側)を用いた。
Claims (11)
- 被験者のフケの状態を評価するための非侵襲的方法であって、
a)前記被験者から皮膚細胞サンプル/上皮細胞サンプルを採集することと、
b)前記上皮細胞サンプル/皮膚細胞サンプルの1つ以上のバイオマーカーのレベルを検出することであって、前記1つ以上のバイオマーカーが、ミエロペルオキシダーゼ、(±)−9−ヒドロキシ−10E,12Z−オクタデカジエン酸及び(±)−13−ヒドロキシ−10E,12Z−オクタデカジエン酸(HODE)、並びにこれらの混合物からなる群から選択されることと、
c)検出されたバイオマーカーの前記レベルが、被験者が酸化的ストレス及び/又は酸化的損傷を有するか否かを示すことと、
を含む、方法。 - 皮膚サンプルの採集が、接着物品、毛髪プラック、皮膚洗浄液、及びそれらの混合物からなる群からである、請求項1に記載の方法。
- 前記非襲的方法が、
a)哺乳類の上皮に接着物品を貼付することと、
b)上皮細胞を前記接着物品に接着させることと、
c)前記接着物品を前記哺乳類の上皮から除去することと、
d)抽出のために前記接着物品を準備することと、
e)前記接着物品に接着した前記上皮細胞からのバイオマーカーを抽出することであって、前記バイオマーカーが、ミエロペルオキシダーゼ、(±)−9−ヒドロキシ−10E,12Z−オクタデカジエン酸及び(±)−13−ヒドロキシ−10E,12Z−オクタデカジエン酸(HODE)、並びにこれらの混合物からなる群から選択されることと、
f)前記接着物品に接着した前記上皮細胞からの前記バイオマーカーを測定することと、
g)治療後の基準サンプルと比較した前記上皮細胞の前記バイオマーカーの量を決定することと、
を含む、請求項1又は2に記載の方法。 - 前記検出されたバイオマーカーの前記レベルが、前記バイオマーカーを前記接着物品上のタンパク質の量で除して、又は前記(±)−9−ヒドロキシ−10E,12Z−オクタデカジエン酸及び前記(±)−13−ヒドロキシ−10E,12Z−オクタデカジエン酸については対応する非酸化親脂質の量で除して正規化される、請求項2又は3に記載の方法。
- 前記上皮が角質層を含む、請求項1〜4のいずれか一項に記載の方法。
- バイオマーカーの基準レベルと比較したとき、バイオマーカーレベルのレベル変化がある、請求項1〜5のいずれか一項に記載の方法。
- 適用前の基準レベルのバイオマーカーと比較したとき、抗真菌毛髪トリートメントの適用後に正規化バイオマーカーの基準からの変化がある、請求項4〜6のいずれか一項に記載の方法。
- 適用前の基準バイオマーカーと比較したとき、フケ防止シャンプーの適用後3週間の期間にわたり少なくとも40%の正規化バイオマーカーの減少がある、請求項4〜7のいずれか一項に記載の方法。
- 適用前のミエロペルオキシダーゼバイオマーカーの基準レベルと比較したとき、フケ防止シャンプーの適用後3週間の期間にわたりミエロペルオキシダーゼ正規化バイオマーカーの87%の減少がある、請求項4〜8のいずれか一項に記載の方法。
- 適用前のバイオマーカーの基準レベルと比較したとき、ジンクピリチオンシャンプーの適用後に正規化バイオマーカーの変化がある、請求項4〜9のいずれか一項に記載の方法。
- 適用前のバイオマーカーの基準レベルと比較したとき、硫化セレンシャンプーの適用後に正規化分子バイオマーカーの変化がある、請求項4〜9のいずれか一項に記載の方法。
