本発明の組成物

最初の3つは抗生物質耐性細菌である。
A.バウマンニは大部分の抗生物質に対して非常に耐性であり、そして戦闘による創傷において蔓延している。
C.ディフィシル胞子では、致死性でありうる病原性感染を引き起こす細菌胞子を殺すことが非常に困難である。

医療機器上の胞子の殺菌

材料および方法
株および増殖条件

[0087]Qiagen TissueLyser(QIAGEN、米国カリフォルニア州サンタクララ)を用いることによって、バイオフィルム試料をホモジナイズした。滅菌5mmスチールビーズおよび500μL 0.1mmガラスビーズを、500μL TE緩衝剤を含む試験管に添加し、そして30Hzで5分間処理した。次いで、QIAamp DNAミニキット(QIAGEN)を用いることによって、バイオフィルム試料由来の細菌DNAを抽出した。Nanodrop分光光度計(Nyxor Biotech、フランス・パリ)を用いて、DNA試料を定量化し、そして20ng/μlに希釈した。3つの個々の細菌由来のDNAもまた、陽性対照として、20ng/μlに希釈した。定量的リアルタイムPCRを用いて、バイオフィルム試料中の3つの細菌の比に相当する、各細菌に関する特異的遺伝子レベルをアッセイした。LightCycler480(Roche、ドイツ・マンハイム)を用いることによって、遺伝子レベルを検出した。20μlリアルタイムPCR反応のため、LightCycler480 SYBRグリーンIマスターキット(Roche)を用いた。各試料を3回アッセイした。各試料の相対的遺伝子レベルを計算し、そして分析した。簡潔には、定量的リアルタイムPCR反応後に、異なる試料中のターゲット遺伝子の閾値周期(Ct値)を得た。規準化DNA Ct値を関心対象の遺伝子のCt(ターゲット遺伝子)から減じて、試料のdCt値を生じた。標準物質(最高のdCt値を持つ試料)のdCt値をすべての他の試料から減じて、ddCt値を生じた。相対遺伝子レベルとして、すべての試料に関して、2の−ddCt乗(2−ddCt)を取った。各細菌の遺伝子発現レベルは、所定のDNA抽出試料内の各細菌の相対比に相当する。
[0088]バイオフィルム形成に対するさらなる多数の化学物質処理に関して正しい濃度を試験するために、まず、400ppm、1000ppm、4000ppm、8000ppm、および12000ppmのPPAおよびPPA−EP濃度を用いた(図2および3)。PPAおよびPPA−EPは、バイオフィルム形成に対して明らかでそして有意な阻害効果を示した。したがって、400ppmおよび4000ppmをアッセイのための最終濃度として用いた。バイオフィルム形成の主観的観察を行い、そしてバイオフィルムバイオマス乾燥重量もまた測定して、より客観的な測定値を提供した。処理のすべては、乾燥重量に基づくと、対照バイオフィルムよりもより低いバイオマス形成を示し、そしてバイオマスの減少は、バイオフィルム形成の視覚的減少と相関した(表1)。400ppmのPPAおよびPPA−EPを添加すると、バイオフィルムバイオマス乾燥重量は42.2%および52.8%減少した。4000ppmのPPAおよびPPA−EPを添加すると、バイオフィルム成長はまったく観察されなかった。細菌プレート計数によって、4000ppm PPAおよびPPA−EPは、細菌増殖を完全に阻害することがさらに確認された。

[0092]本実施例は、PKCA溶液がシミュレーションされた創傷溶液環境において細菌を殺すことを立証する。PPAを含有するPKCA溶液を、50mMリン酸緩衝液中、6.0のほぼ生理学的pHにして、そして10%ウシ胎児血清(FBS)の存在下でメチリシン耐性黄色ブドウ球菌(MRSA)を殺す能力に関して試験した。100ppm(0.85mM)のPPAを含有するPKCA溶液は、FBS溶液内で、1分間に6対数のMRSAを殺すことが示された。
[0094]タンパク質環境におけるPKCA混合物の性能を決定するため、高タンパク質環境における性能に関して、そして創傷環境の単純なシミュレーションに関して、PPA混合物をMRSAに対する殺菌有効性に関して試験した。10%ウシ胎児血清(FBS)中に懸濁されたMRSAに対して、上述のような浸漬試験によってPPA混合物を試験した。PPA混合物は、10%FBS中で200ppmのPPAに曝露された際、15秒以内で、6対数のMRSAを殺した(表5)。これは、15秒間に、水中に懸濁されたMRSAを殺すのに必要なPPA濃度の倍であった。したがって、高タンパク質環境においては、濃度の増加が必要であるが、高タンパク質環境において、密集したMDROを殺すための活性は、なお迅速で、そして有効であった。

[0096]リン酸緩衝液中に懸濁されたアシネトバクター・バウマンニに対してもまた、PPA混合物を試験して、異なるpHのPKCA混合物の緩衝溶液が、創傷治癒プロセスの異なる期に使用可能であるかどうかを決定した。この試験の結果は、PPAが50ppmでは60秒間で、そして100ppmではわずか15秒間で、6対数のアシネトバクター・バウマンニを殺すであろうことを立証した(図7)。

Claims (31)
- ペルオキシα−ケトカルボン酸、該ペルオキシα−ケトカルボン酸の対応脱カルボキシル化カルボン酸、過酸化水素、α−ケトエステル、および該ペルオキシα−ケトカルボン酸に対応する少なくとも1のカルボン酸と該ペルオキシα−ケトカルボン酸に対応する該カルボン酸のアニオンの混合物を含む組成物であって、該ペルオキシα−ケトカルボン酸が、ペルオキシα−ケトピルビン酸、ペルオキシα−ケト酪酸、ペルオキシα−ケト吉草酸、及びそれらの混合物から選択される組成物。
- 請求項1に記載の組成物であって、該α−ケトエステルがアルキルα−ケトエステルである組成物。
- 請求項2に記載の組成物であって、該アルキルα−ケトエステルがピルビン酸アルキルエステルである組成物。
- 請求項1に記載の組成物であって、該α−ケトエステル対該ペルオキシα−ケトカルボン酸のモル比が0.02:1〜10:1である組成物。
- 請求項1に記載の組成物であって、該ペルオキシα−ケトカルボン酸が、ペルオキシα−ケトピルビン酸である組成物。
- 請求項1に記載の組成物であって、クリーム、ジェル、液体、ローション、皮膚用パッチ剤、洗浄ジェル、スプレー、ドレッシング剤、フィルム、ビーズ、ディスク、織物、又は繊維としての配合物である組成物。
- 表面を組成物と接触させることによる、該表面上の微生物の量の減少に用いるための該組成物であって、該組成物が請求項1〜6のいずれか1項に記載の組成物の有効量を含む組成物。

- 被験体の創傷領域に組成物を局所投与することによる、該被験体における創傷の治療に用いるための該組成物であって、該組成物が請求項1〜6および10〜23いずれか1項に記載の組成物を含む組成物。
- 被験体の創傷領域に組成物を局所投与することによる、被験体おける創傷から生じる敗血症の防止に用いるための該組成物であって、該組成物が請求項1〜6および10〜23のいずれか1項に記載の組成物の有効量を含む組成物。
- 包帯材料中に配合された請求項1〜6および10〜23のいずれか1項に記載の組成物を含む創傷治療組成物。
- 請求項29に記載の創傷治療組成物であって、創傷に適用する際に、ペルオキシα−ケトカルボン酸が前記包帯材料から放出される創傷治療組成物。
