Description
50kgバッチのWPM粉末を生成した。このバッチは、タンパク質が4wt%になるように乳清タンパク質分離物WPI(プロラクタ(Prolacta)90、Lactalis、Retiers、フランス)をpH5.9±0.05(天然のpH6.48を1MのHClで調節)の軟水(160mg.L−1Na+)に分散させた分散液を熱処理することで得た。WPI分散液を60℃に予熱し、次いで1000L.h−1の流量で作動するSojaプレートプレート熱交換器(PHE)を用いて85℃に加熱し、続いてチューブ状熱交換器での15分の保持時間後に4℃に冷却した。こうした操作条件下、レイノルズ数Reは約1,500であり、PHEでの層流が確保された。初期タンパク質の85%超がWPMに変換された(26,900gで20分間の遠心分離によりWPMを除去した後に278nmの吸光測定によって判定)。WPM分散液は136±7nmの流体力学半径と、0.1の多分散性指数(動的光散乱DLSによって判定)を示した。その後、WPM分散液を温度10℃、流量180L.h−1で全表面が6.8m2(Novasep Process、Miribel、フランス)の二つのカルボセップ(Carbosep)0.14膜を用いた精密ろ過によって22wt%まで濃縮した。次いで、GEA Niro SD6.3N噴霧乾燥機(Sφborg、デンマーク)を用いて(供給速度:WPM濃縮物25kg.h−1;入口空気温度:145〜150℃;出口空気温度:75〜77℃;噴霧ノズルφ:0.5mm;噴霧圧力40bar)液体濃縮物を噴霧乾燥し、2kgアルミニウム密封バッグに入れて10℃で保存した。WPM粉末は、ミクロゲルの形態のタンパク質を97%含有していた。その組成(g/100gの湿式粉末)は:タンパク質(Nx6.38、Kjeldhal)91;水分3.6;ラクトース3;脂肪0.4、及び灰分2であった。タンパク質サンプルのHNO3/H2O2無機化及びVista MPX同時ICP−AES分光計(Varian Inc.Palo Alto、CA、米国)を用いた分析に基づいて判定した粉末のミネラル組成(g/100gの湿式粉末)は:Ca2+0.320;K+0.409;Na+0.468;Mg2+0.060;Cl−0.178であった。
実施例1で調製した乳清タンパク質ミセル(WPM)粉末を4wt%になるように、10mLのミリQ(Milli−Q)水に20分間の超音波プローブ(直径6mm、振幅30%、パルスオン1秒、パルスオフ0.5秒)を用いた超音波処理によって分散させた。超音波処理時、分散液は氷浴に浸して温度を50℃未満に維持した。プロセスの最後に測定した分散液温度は、40〜45℃であった。20分後、ほぼ全ての顆粒が脱凝集しており(数個の顆粒がまだ分散液中に観察可能であったが、無視できる数であった)、約215nmの粒子径及び0.07未満の多分散性指数(動的光散乱により判定)が得られた。
油相としてヘキサデカン(Sigma、>99%)を使用した以外は実施例2の場合と同じ方法でエマルションを調製した。すぐにクリーミングが起こる直接エマルションが形成された。
油の体積分率がエマルションに対する影響を調べるため、ミグリオール(登録商標)/水体積比は様々であるが、WPM質量/油質量比を60mg/gに固定した様々なサンプルをpH4.7、0.06MのNaN3で調製した。油の体積分率が高いほどクリーミングに対する安定性が向上する。80%v/v未満のエマルションは安定であるが、80%v/vを超えるエマルションは相分離することがわかった(図9)。
WPMによって安定化させたエマルションを室温で8カ月保存した後に観察した。図10及び11は、pH値の異なる水中ミグリオール(登録商標)エマルション(50vol%)の肉眼及び顕微鏡での観察結果を示す。乳化前の水相は0.4wt%のNaN3防腐剤を含むWPMの0.11wt%分散液であった。油滴のサイズ分布は、8カ月にわたりほとんど変化せず、乳清タンパク質ミセルは、エマルションを合一に対して長期にわたって安定化する効果があることを示している。
Claims (11)
- 乳清タンパク質ミセル、水及び分散油滴を含み、前記乳清タンパク質ミセルによって安定化されており、前記油滴の平均直径が40〜1000μmであり、前記油の体積分率が40%〜80%である、エマルション。
- 前記乳清タンパク質ミセルがエマルションの0.01〜5wt%の濃度で存在する、請求項1に記載のエマルション。
- 前記油滴が、油溶性の生体活性化合物を含有する、請求項1又は2に記載のエマルション。
- 前記油が、精油;ヒマワリ油;オリーブ油;パーム油;ヤシ油、ラッカセイ油;パーム核油;コーン油;ヘーゼルナッツ油;ゴマ油、及びこれらの混合物からなる群から選択される、請求項1〜3のいずれか一項に記載のエマルション。
- pHが2〜8である、請求項1〜4のいずれか一項に記載のエマルション。
- 前記油滴が、合一に対して少なくとも6カ月間安定している、請求項1〜5のいずれか一項に記載のエマルション。
- 水相と油滴とが対比色を有する、請求項1〜6のいずれか一項に記載のエマルション。
- 請求項1〜7のいずれか一項に記載のエマルションを含み、食品組成物;飲料;飲料増強剤、例えばコーヒークリーマー;化粧品組成物;医薬組成物;栄養製剤;又は経管栄養製剤である組成物。
- 前記食品組成物が、乳製品;アイスクリーム;ソース;スープ;デザート;菓子製品;ベーカリー製品;サラダドレッシング;又はペットフードである、請求項8に記載の組成物。
- 請求項1〜7のいずれか一項に記載のエマルションを乾燥することにより得られた組成物。
- 水に分散されるための、請求項10に記載の組成物。
