SlyD融合モジュールを含む希少試薬を用いる市販のイムノアッセイにおいては、タンデムSlyD(グルタルジアルデヒドにより重合した、短いペプチド配列を介して連結した2分子のSlyD)等の通常化学的に架橋したシャペロン分子が、干渉を低減するために加えられる。背景技術の項で言及したように、製造工程のために、これらの化学的に架橋した添加剤は多少不均一であり、十分な収量で厳密に再現可能な方法で供給することができない。
Skp/SlyDシャペロン融合ポリペプチドのクローニング及び精製
発現カセットのクローニング
適切な発現構築物を生成するために、EcSkp−EcSlyD−EcSlyDをコードする発現カセットを、発現プラスミドpQE80L(Qiagen,Hilden,Germany)に2段階クローニング法により結合した。

好ましい融合ポリペプチドの完全アミノ酸配列を配列番号1に示す。
推測上の干渉防止ポリペプチドを十分量で得るために、EcSkp−EcSlyD−EcSlyDを大腸菌内で組み換え発現させた。この目的のため、大腸菌BL21 Codon+(Merck(Novagen(登録商標),Darmstadt,Germany)を、生成された発現構築物pQE80Skp−diSlyDで形質転換した。
Skp/SlyDポリペプチド融合物と、SlyD及びSlpAの融合タンパク質は、実質的に同じ手順を用いて精製した。細胞溶解について、凍結したペレットを、冷却した50mMリン酸ナトリウム(pH8.0)、7.0M GdmCl、5mMイミダゾールに再懸濁し、懸濁液を氷上で少なくとも2時間撹拌し、細胞溶解を完了させた。遠心分離及びろ過(0.45μm/0.2μm)後、未精製の溶解物を、5.0mM TCEPを含む溶解バッファーで平衡化したNi−NTAに供した。それに続く洗浄工程は、それぞれの標的タンパク質に合わせて、50mMリン酸ナトリウム、(pH8.0)、7.0M GdmCl及び5.0mM TCEP溶液にイミダゾールを5〜15mMの範囲で調整した。少なくとも10〜15倍の洗浄バッファーを使用した。次いで、GdmCl溶液を50mMリン酸カリウム(pH8.0)、100mM KCl、10mMイミダゾール、5.0mM TCEPと置換し、マトリクス結合タンパク質の立体配座再フォールディングを誘導した。同時精製したプロテアーゼの再活性化を防止するために、プロテアーゼ阻害剤カクテル(Complete(登録商標)EDTA−free,Roche)を再フォールディング緩衝液に加えた。一晩の反応で、合計15〜20倍のカラム容積の再フォールディング緩衝液を使用した。次いで、3〜5倍のカラム容積の50mMリン酸カリウム(pH8.0)、100mM KCl、10mMイミダゾールで洗浄することにより、TCEP及びComplete(登録商標)EDTA−free阻害剤カクテルの両者を除去した。次いで、非特異的に結合しているタンパク質混入物を除去するために、イミダゾール濃度(50mMリン酸ナトリウム(pH8.0)と100mM KCL溶液)を、EcSkp−EcSlyD融合タンパク質について60〜70mMに、EcSkp−EcSlpA−EcSlpAについて50mMに、SlyD融合ポリペプチドについて30mMに上昇させた。次いで、野生型のタンパク質を、同じ緩衝液中の500mMイミダゾールで溶出させた。タンパク質を含む画分は、トリシン−SDS−PAGEにより純度について評価し、貯蔵した。最後に、タンパク質をサイズ排除クロマトグラフィー(Superdex HiLoad、Amersham Pharmacia)に供し、タンパク質含有画分を貯蔵し、Amiconセル(YM10)中で10〜20mg/mLに濃縮した。
分光測定
タンパク質濃度の測定は、Uvikon XL二光線束分光光度計を用いて実施した。モル吸光係数(εM280)は、Pace (1995), Protein Sci. 4, 2411-2423に記載された手順を用いて決定した。EcSkp−EcSlyD−EcSlyDについては、13410M−1cm−1のモル吸光係数(εM280)を用い、EcSkp−EcSlyDについては、7450M−1cm−1のモル吸光係数(εM280)を用いた。EcSkp−EcSlpA−EcSlpAについては、4470M−1cm−1のモル吸光係数(εM280)を用いた。大腸菌の反復SlyD構築物、SlyD、SlyD−SlyD、SlyD−SlyD−SlyD、SlyD−SlyD−SlyD−SlyD及びSlyD−SlyD−SlyD−SlyD−SlyDについては、5960M−1cm−1、11920M−1cm−1、17880M−1cm−1、23840 M−1cm−1及び29800M−1cm−1のモル吸光係数を用いた。
SlyDポリペプチド融合タンパク質の干渉防止活性
SlyDポリペプチド融合タンパク質の干渉防止作用は、自動化Elecsys(登録商標)2010分析装置(Roche Diagnostics GmbH)で評価した。Elecsys(登録商標)は、Rocheグループの登録商標である。測定は、二重抗原サンドイッチ形式で実施した。
EcSkp−EcSlpA−EcSlpAポリペプチド融合タンパク質の干渉防止活性
EcSkp−EcSlpA−EcSlpAの干渉防止活性を、自動化Elecsys(登録商標)2010分析装置(Roche Diagnostics GmbH)で評価した。Elecsys(登録商標)は、Rocheグループの登録商標である。測定は、二重抗原サンドイッチ形式で実施した。
組み換え型EcSkp−EcSlpA−EcSlpAポリペプチドは、二重抗原サンドイッチ(DAGS)イムノアッセイ形式で評価した。この目的を達成するため、欧州特許第2127678号明細書に開示されたPmSlyD−mgG2及びEcSlpA−mgG2を、ヒト血清中の抗HSV−2抗体を特異的に検出するために、それぞれビオチン及びルテニウム複合体として用いた。成熟型糖タンパク質G2(mgG2)は、単純ヘルペスウイルス2型(HSV−2)の免疫優勢抗原であり、欧州特許第2127678号明細書に開示され、HSV−2感染を検出するのに非常に貴重なツールである。PmSlyD−mgG2−ビオチン及びEcSlpA−mgG2−ルテニウム複合体を、R1(試薬緩衝液1)及びR2(試薬緩衝液2)中でそれぞれ300ng/mLの濃度で使用した。
円偏光二色性分光法により検出される熱的に誘導されるSkp−SlyD−SlyDの展開
近紫外二色性(UV CD)スペクトルは、温度自動調節セルホルダーを用い、Jasco−720分光偏光計により測定し、平均残余楕円率に変換した。緩衝液は、50mMリン酸カリウム、250mM KCl、0.5mM EDTA(pH7.5)であった。経路の長さは0.2cmであり、タンパク質濃度は8.2mg/mLであった(147μMの単量体は49μMの三量体に相当する)。範囲は250〜330nmであり、バンド幅は1.0nmであり、走査速度は20nm/分であり、解像度は0.5nmであり、反応は1秒であった。ノイズ比を改善するために、スペクトルを9回測定し平均化した。
Claims (10)
- 1個の多量体化ドメインと、SlyD又はSlpAの少なくとも1分子とを含む数分子のフォールディングヘルパーポリペプチドを含有する融合ポリペプチドであって、前記多量体化ドメインがSkpであり、更なる標的ポリペプチド抗原配列が前記融合ポリペプチドの配列の一部に含まれない、前記融合ポリペプチド。
- 1分子のSkpが、2個の隣接するSlyD又はSlpA分子と融合している、請求項1に記載の融合ポリペプチド。
- 配列番号1(Skp−タンデム−SlyD)を含む融合ポリペプチド。
- 配列番号9(Skp−タンデム−SlpA)を含む融合ポリペプチド。
- 干渉を低減し、又は偽陽性の結果を最小にするための、イムノアッセイにおける、請求項1〜4のいずれか1項に記載の融合ポリペプチドの使用。
- アッセイ試薬のタンパク性成分の溶解度を上昇させるための、請求項1〜4のいずれか1項に記載の融合ポリペプチドの使用。
- 請求項1〜4のいずれか1項に記載の融合ポリペプチドを含む、イムノアッセイにより分離試料中の検体を検出するための試薬キット。
- 干渉を低減し、又は偽陽性の結果を最小にするための試薬として、請求項1〜4のいずれか1項に記載の融合ポリペプチドを使用する、分離試料中の検体を検出する方法。
- 下記工程を含む、分離試料中の検体を検出する方法;
a)体液試料を、前記試料中に存在する前記検体と特異的に結合し得る、特異的結合パートナーと混合することにより免疫反応混合物を生成する工程、
b)前記特異的結合パートナーを前記試料に加える前、同時又は後のいずれかに、請求項1〜4のいずれか1項に記載の融合ポリペプチドを前記免疫反応混合物に加える工程、
c)前記体液試料中に存在する検体が、前記特異的結合パートナーと免疫反応し、免疫反応生成物が形成されるのに十分な時間、前記免疫反応混合物を維持する工程、及び、
d)前記免疫反応生成物のいずれかの存在及び/又は濃度を検出する工程。 - 分離試料中に存在する検体が抗体である、請求項8又は9に記載の方法。
