Description
このマイクロバブル前駆体は、気体形成性化学物質のエマルジョンが液体状態で安定であり得る。そして、超音波エネルギを与えられたときに微小気泡を生成する造影剤であって、血管より組織に移行可能なサイズの造影剤を提案している。
本発明の一態様は、超音波造影機能を有する基材の表面に、少なくとも1つのターゲッティング用の分子プローブと、少なくとも1つの光学プローブとを備える造影剤を提供する。
上記態様においては、コンジュゲート用の第1の分子プローブと、第2の分子プローブに標識された光学プローブと、疾患特異的に細胞または組織に結合するターゲッティング用の第3の分子プローブとを備えていてもよい。
また、上記態様においては、前記第2の分子プローブと前記第3の分子プローブとがモル比1:1の割合で含有されていてもよい。
また、上記態様においては、前記造影物質が、超音波照射により気化する水不溶性物質であってもよい。
また、上記態様においては、前記造影物質が気体であってもよい。
本実施形態に係る造影剤1は、図1に示されるように、超音波造影可能な基材2と、該基材2の表面に修飾された少なくとも1つの分子プローブ3〜5および少なくとも1つの光学プローブ6とを備えている。
第1の分子プローブ3としては、例えば、ビオチンが用いられる。
第2の分子プローブ4としては、例えば、ストレプトアビジンが用いられる。
第3の分子プローブ5としては、例えば、CCA1抗体が用いられる。
光学プローブ6としては、例えば、Alexa Fluor(AF)647が用いられる。
本実施形態の造影剤1の製造方法は、図2に示されるように、第1の分子プローブ3を基材2の表面に標識して第1の溶液を生成する第1の工程S1と、光学プローブ6を第2の分子プローブ4に標識して第2の溶液を生成する第2の工程S2と、第3の分子プローブ5に第1の分子プローブ3を標識して第3の溶液を生成する第3の工程S3と、第2の工程で生成された第2の溶液と第3の工程で生成された第3の溶液とを混合して第4の溶液を生成する第4の工程S4と、第1の工程S1で生成された第1の溶液に、第4の工程S4で生成された第4の溶液を混合して反応させる第5の工程S5と、該第5の工程S5において生成された混合液を遠心分離する第6の工程S6とを含んでいる。
さらに、第6の工程S6では、第5の工程S5において生成された混合液を、例えば、3000Gで5min間遠心分離することにより、本実施形態に係る造影剤1が製造される。本工程では、造影剤1上に修飾されなかった遊離の複合体を造影剤と分離し、取り除く。
第2の溶液の濃度は、例えば、吸光測定法により概算するこができる。また、第3の溶液の濃度は、例えば、吸光測定法およびBradford法を用いて精度よく測定することができる。
本実施形態に係る造影剤1を用いて、シャーレ上に培養した大腸がんの細胞(DLD−1)に対する特異的結合特性を調べた。
このとき、第6の工程S6で遠心分離した上清を除去し、沈殿物をPBS(例えば100μL)に再懸濁させた溶液を細胞に振りかけ、30分静置後に培養液で表面を洗浄した。
共焦点蛍光顕微鏡画像を図3(a)、(d)、図4(a)、(d)に、顕微鏡明視野像を図3(b)、(e)および図4(b)、(e)に、また、これらの重畳画像を、図3(c)、(f)、図4(c)、(f)にそれぞれ示す。また、フローサイトメータの解析結果を図5(a)〜(d)に示す。
そして、8%アクリルアミドゲル中にパーフルオロペンタン換算濃度0.02mg/mLになるように本実施形態に係る造影剤を含有させたファントム(試料A)を水槽7内に設置し、これに向かって集束超音波トランスデューサ8を用いて3MHzのパルス超音波(デューティ比0.01、1000サイクル)を1秒間照射する。
また、このように、光学プローブ6とCCA1抗体等の分子プローブ5が基材2の表面に搭載されていても、気相化が可能であることが確認された。
分子プローブ3〜5や光学プローブ6も、本実施形態に用いたものに限定されない。
このように構成された造影剤1をマウスの尻尾に注射すると、ナノサイズによるEPR効果と抗体によるアクティブ結合とによって、図10(a)、(b)に示されるように、腫瘍組織にのみ選択的に集積させることができる。これにより、内包物質を気相化させなくても超音波イメージングを行うことができるという利点がある。
本実施形態に係る造影剤は、内包物質として、2種類の気体、パーフルオロヘキサンとパーフルオロペンタンとを混合比1:1で混合した気体を採用している。
このように構成された本実施形態に係る造影剤の大きさと他の製造方法による造影剤の大きさとの比較を表1に示す。表1は2つの実験例である。
具体的には、HCT116細胞およびAGS細胞の培養皿に添加し、例えば1時間静置した後、培地で表面を洗浄し、例えば、3時間後に共焦点レーザ顕微鏡で特異的な細胞内取り込みを観察するとともに、12時間の培養後にPBSで洗浄し、トリプシン/EDTAで細胞を培養皿から剥離して、フローサイトメータ解析を行った。
このようにすることで、本実施形態に係る造影剤は、標識した抗体に対応する抗原にのみ特異的に結合させることができ、他の細胞と明瞭に区別することができるという利点がある。
この製造キット21は、第1〜第3の溶液を無菌的に収容する収容部22,23,24と、これら収容部22〜24を接続する流路25,26とを備えている。各流路25,26にはバルブ27,28が設けられており、バルブ27,28の操作によって流路25,26を開閉することができるようになっている。

Claims (9)
- 超音波造影機能を有する基材の表面に、ビオチンと、ストレプトアビジンに標識されたAlexa Fluor(AF)647と、前記ビオチンによって標識されている疾患特異的に細胞または組織に結合するCCA1抗体とを備え、
該CCA1抗体は細胞膜透過性を有し、生体投与時にはナノサイズである造影剤。 - 前記ストレプトアビジンと前記CCA1抗体とがモル比1:1の割合で含有されている請求項1に記載の造影剤。
- 前記基材が、内部に造影機能を有する造影物質を内包した脂質膜からなる外殻を有するカプセル状造影剤である請求項1または2に記載の造影剤。
- 前記造影物質が、生体投与時に液体状であって、生体投与後の超音波照射により気化する水不溶性物質である請求項3に記載の造影剤。
- 前記造影物質が気体である請求項3に記載の造影剤。
- 請求項1に記載の造影剤の製造方法であって、
前記ビオチンを前記基材の表面に標識した第1の溶液を生成する第1の工程と、
前記ストレプトアビジンに前記Alexa Fluor(AF)647を標識した第2の溶液を生成する第2の工程と、
前記CCA1抗体に前記ビオチンを標識した第3の溶液を生成する第3の工程と、
前記第2の工程で生成された第2の溶液と前記第3の工程で生成された前記第3の溶液とを混合して第4の溶液を生成する第4の工程と、
前記第1の工程で生成された第1の溶液に前記第4の工程で生成された第4の溶液を混合して反応させる第5の工程と、
該第5の工程で生成された混合液を遠心分離する第6の工程とを含む製造方法。 - 前記第4の工程において混合される前記第2の溶液中の前記ストレプトアビジンと、前記第3の溶液中の前記CCA1抗体とがモル比1:1の割合である請求項6に記載の製造方法。
- 前記第5の工程において、前記第1の工程において生成された前記第1の溶液を10倍以上に希釈した後に混合する請求項6または請求項7に記載の製造方法。
- 請求項1に記載の造影剤の製造キットであって、
前記ビオチンを前記基材の表面に標識した第1の溶液、前記ストレプトアビジンに前記Alexa Fluor(AF)647を標識した第2の溶液、および前記CCA1抗体に前記ビオチンを標識した第3の溶液を個別に分離して収容する複数の収容部と、これら収容部を接続する開閉可能な流路とを備える製造キット。
