Description
本発明は、上記の先行技術を考慮し、またヒトおよび動物用の医薬における新規の抗細菌性の予防的作用剤の需要の増加を考慮してなされた。基質特異性拡張型ベータラクタマーゼ(enhanced−spectrum beta−lactamase)(ESBL)株の療法はますます困難である。本発明の目的は、従来の抗生物質、したがって、ESBL産生体である病原性微生物の制御の代替として使用することができる新規の抗微生物ミクロシンポリペプチドを提供することである。この関連で、本発明の目的は、ESBL産生大腸菌、さらに、ドイツにおけるごく最近の大規模な大発生の原因病原体であった毒性が高い腸管出血性大腸菌(EHEC)株も含めた病原性グラム陰性細菌の処置において使用することができる新規の抗微生物ミクロシンポリペプチドを提供することである(Frank,C.ら(2011)N Engl J Med 10.1056/NEJMoa1106483;Mellmann,A.ら(2011)PLoS ONE 6(7):e22751.doi:10.1371)。EHEC細胞は、特定の抗生物質を用いて処置することによってより大量の志賀毒素を放出する。したがって、患者を抗生物質で処置することは禁忌であると考えられている。新規のミクロシンポリペプチドはEHECおよび腸管病原性大腸菌(EPEC)感染症の治療および予防において有効である。本発明のさらなる目的は、成人および小児における機能性胃腸障害、特に過敏性腸症候群の治療において使用することができる新規の抗微生物ミクロシンポリペプチドを提供することである。本発明のさらなる目的は、前記医学的使用のための、新規のミクロシンポリペプチドを産生する細胞の新規のミクロシンポリペプチドを含む組成物を提供することである。癌の治療において使用することができる新規のミクロシンポリペプチド、食料品の保存において使用することができる新規のミクロシンポリペプチドおよび包帯材のコーティングにおいて使用することができる新規のミクロシンポリペプチドを提供することも本発明の目的である。

すなわち、本発明者らは、新規の、従前のミクロシンではない、ヌクレオチド配列およびアミノ酸配列が独特であるポリペプチドミクロシンSを同定した。ミクロシンSの効果に関与する遺伝子が同定されており、それらの効果は機能的に特徴付けられている。本発明者らは、驚いたことに、古典的なベータ−ラクタム系抗生物質の大部分とは対照的に、ミクロシンSはESBL産生O104:H4アウトブレイク株ならびにO128:H2およびO157:H7 EHEC分離株を含むEHECの阻害効果を示すことを見いだした(図5)。したがって、さらに、ミクロシンSが腸管出血性大腸菌(EHEC)および腸管病原性大腸菌(EPEC)の感染症の治療において有効であることが見いだされた。したがって、ミクロシンSは、従来の抗生物質、したがって、病原性微生物の制御の代替として使用することができる。さらに、食料品保存のための使用が可能であり、それにより、本発明は完了した。さらに、プラスミドpSYM1(図1)によりコードされる大腸菌G3/10のミクロシンS(MccS)遺伝子クラスター(配列番号1)が、4種のクラスター化された遺伝子、mcsS(配列番号2)、mcsI(配列番号3)、mcsA(配列番号4)およびmcsB(配列番号5)からなることも見いだされた。大腸菌G3/10は、例えば胃腸障害を治療するために使用される十分に証明されたプロバイオティクスであるので、必ずしもミクロシンSを該株から精製しなければならないとは限らない。MccSはin vivoにおいて有利に有効であるはずである。
配列番号1は、大腸菌G3/10のミクロシンSオペロンのヌクレオチド配列である。
特に定義されていなければ、本明細書において使用される全ての技術用語および科学用語は、本発明が属する当業者に一般に理解されるものと同じ意味を有する。矛盾する場合は、定義を含めた本明細書が優先される。
本発明のミクロシンSポリペプチドは、そのアミノ酸残基配列および新規の機能的性質を特徴とする。ポリペプチドは、当技術分野で公知の方法を用いて単離することができる。すなわち、天然に存在するまたは組換えによって作製されたミクロシンSポリペプチドを、標準のタンパク質の精製技法を用いた適切な精製スキームによって細胞または組織供給源から単離することができる。しかし、ミクロシンSポリペプチドは細胞内に天然に存在してもよく、または組換えDNA技法によって作製することもでき、あるいは組換え発現の代わりに、標準のペプチド合成技法を用いてミクロシンSポリペプチドを化学的に合成することができる。本明細書では、ミクロシンSポリペプチドは、表現型により特徴付けられ、ミクロシンSと名付けられた大腸菌ミクロシンに分類される。図2では、上のパネルにミクロシンS前駆体のアミノ酸配列が示されている。2重のグリシンリーダーペプチドに下線が引かれている。アスタリスクは、クラスIIミクロシンに典型的なジスルフィド結合の形成に関与する可能性があるシステインを示す。アミノ酸配列は2重のグリシン切断部位を有するグリシンリッチペプチドを示す。配列番号6または配列番号7のアミノ酸配列を有するポリペプチドまたは配列番号6または配列番号7のアミノ酸配列を有するポリペプチドの生物活性変異体を本明細書に記載の方法において使用することができる。
プロバイオティクス大腸菌株G3/10のゲノムは、「パイロシークエンス」技術を用いて完全に配列決定され、その後、自動的にアノテートされ、さらに手動で編集された。
ミクロシンS遺伝子をクローニングすることで決定されるヌクレオチド配列により、本発明のミクロシンS遺伝子の増幅および/または他の種におけるミクロシンS遺伝子の同定において使用するために設計されたプローブおよびプライマーを生成することが可能になる。
プラスミドはベクターの1種類であり、追加的なDNAセグメントをライゲーションすることができる環状二本鎖DNAループを指す。特定のプラスミドは、それらが発生するまたはそれらが導入される宿主細胞において自律複製することができる(例えば、細菌の複製開始点を有する細菌プラスミドおよびエピソーム哺乳動物ベクター)。
細胞は、任意の原核細胞または真核細胞であり得る宿主細胞であってよい。例えば、ミクロシンSポリペプチドは、本明細書において好ましい大腸菌などの細菌の細胞、または昆虫細胞、酵母もしくは哺乳動物の細胞(例えば、チャイニーズハムスター卵巣細胞(CHO)またはCOS細胞など)において発現させることができる。他の適切な宿主細胞は当業者に公知である。
ミクロシンS免疫原は、一般には、適切な対象(例えば、ウサギ、ヤギ、マウスまたは他の哺乳動物)を免疫原で免疫化することによって抗体を調製するために使用される。適切な免疫原性調製物は、例えば、天然に存在するもしくは組換えによって発現させたミクロシンSポリペプチドまたは化学的に合成したミクロシンSペプチドを含有してよい。調製物は、フロイントの完全アジュバントもしくは不完全アジュバントなどのアジュバント、または同様の免疫賦活剤をさらに含んでよい。免疫原性ミクロシンS調製物を用いて適切な対象を免疫化することにより、ポリクローナル抗ミクロシンS抗体応答を誘導する。そのような方法は当技術分野で公知である。
ミクロシンSポリペプチド、またはさらには、本発明の細胞は、投与するために適した組成物または医薬組成物に組み入れることができる。したがって、本発明は、本発明によるポリペプチドまたは本発明による細胞を含む組成物を意図している。
1つまたは複数の容器内に、治療的または予防的に有効な量の本発明の組成物を含むキットも提供される。キットは、場合によって、本明細書に開示されている意図された医学的使用を実施するためまたは提供される方法を実施するための説明書を含んでよい。
ポリペプチド;細胞および組成物は抗微生物活性、抗細菌活性または抗腫瘍活性を示すので、本発明は、さらに、療法または予防におけるそれらの医学的使用に関する。
本発明のポリペプチドは、天然に精製された産物であってもよく、または化学的合成手順の産物であってもよく、または組換え技法によって原核生物の宿主または真核生物の宿主に産生させることもできる(例えば、培養物中の細菌細胞、酵母細胞、高等植物細胞、昆虫細胞および哺乳動物細胞によって)。

細菌株および成長条件
本明細書において使用した細菌株が表3及び表4に列挙されている。
大腸菌G3/10におけるミクロシンをコードする遺伝子クラスターの同定
6種の大腸菌:大腸菌G1/2、G3/10、G4/9、G5、G6/7およびG8のゲノムについて配列決定した。アノテーションにより、大腸菌G3/10のゲノム内に既知のミクロシンは示されなかった。大腸菌G3/10は、サイズが50.6kbの大きな接合性プラスミドpSYM1(図1)を含有する。このプラスミドは尿路病原性大腸菌分離株のプラスミドpMAS2027と99%同一である。さらに、このプラスミドは、10kbの挿入断片を含有するが、BLAST分析では、特徴付けられていない無名の遺伝子のみが示された。殺菌作用の起源を同定するために、大腸菌G3/10株をそのメガプラスミドpSYM1からキュアリング(cure)することを試みた。一般的なキュアリング手順の多く、例えば、マイトマイシンCまたは加熱処理を実施したにもかかわらず、この株をキュアリングすることはできなかった。したがって、プラスミドpSYM1をコンジュゲーションによって大腸菌G4/9に移入した。接合体のスクリーニングを可能にするために、まずアンピシリン耐性カセットをpSYM1に組み込み、その結果pSYM1−ST76Anが生じた。生じた大腸菌G4/9pSYM1−ST76AnはEPEC接着を有意に阻害することができた(図3)。プラスミドpSYM1が示されている効果に関与する遺伝子を有することが結論づけられた。プラスミドpSYM1の4.7bpの断片をpBR322にクローニングし、その結果、プラスミドpAZ6が生じ、次いでこれを大腸菌G4/9に入れて形質転換した。pAZ6により、大腸菌G4/9がEPEC接着効率を有意に阻害することができるようになり(図3)、これは、pSYM1の4.7kbの断片が、大腸菌G3/10のEPEC接着阻害効果に関与することを示している(図2)。自動的にアノテートされたオープンリーディングフレーム(ORF)のBLAST分析により、特徴付けられたタンパク質またはタンパク質ファミリーとの相同性が小さいことが明らかになり、これにより、ミクロシンをコードするオペロンとの関係が示されている。それにもかかわらず、ミクロシン自体は、未検出のままであった。mcsI、mcsAおよびmcsBと称される3種の遺伝子をpBR322にクローニングし、その結果、pAZ8が生じた。ミクロシンをコードする遺伝子である可能性のあるこのオペロンの上流の小さなORF(図2)をpACYC184にクローニングし、大腸菌G4/9pAZ8にサブクローニングした。このステップにより、推定ミクロシンとミクロシン−ヘルパータンパク質を別々のプラスミドにコードさせることが容易になった。プラスミドpAZ8およびpAZ9を含有する大腸菌G4/9のみがEPEC接着を有意に阻害したが、G4/9pAZ8はG4/9 野生型と同様にEPEC接着に影響を及ぼす(図3)。mcsSと名付けられた小さな遺伝子は、ミクロシンSと名付けられたミクロシンをコードする。
ミクロシンSの機能的な特徴付けおよびその自己免疫の解明
細菌の接着は、多くの感染症の極めて重要な最初のステップである。したがって、ヒト腸上皮細胞への接着の阻害を数量化する試験系は、宿主に対する有益な効果を実証するために適している。第1の実験において、CACO−2細胞またはLOVO細胞の集密の単層を、細菌試験株EcN、大腸菌G3/10、大腸菌G4/9または大腸菌G4/9pAZ6と一緒に、大腸菌:宿主細胞100:1のMOIでプレインキュベートした。2時間インキュベートした後、細胞を洗浄し、EPEC:宿主細胞100:1のMOIを用いてEPEC E2348/69を感染させた。大腸菌G3/10および大腸菌G4/9pAZ6はEcNと同様にEPEC接着を阻害することができた。EcNによる接着阻害は、株のミクロシン活性に左右されることが示された。%単位の接着効率は、いかなるプレインキュベーションもしていない接着(陰性対照)を100%に設定し、それと比較したEPECの接着として表されている。データは、2連のウェルでの少なくとも3回の別々の実験の平均±SDである。図4は、種々のミクロシン発現株(大腸菌G3/10、G4/9 pAZ6およびEcN)ならびに非発現株(大腸菌G4/9)と一緒にプレインキュベートした後の、EPEC E2348/69野生型およびmcsIを有するプラスミド相補突然変異体のヒト腸上皮細胞への接着効率を示す。結果として、mcsIをミクロシンS自己免疫遺伝子であると確実に同定することができた。
腸管出血性大腸菌に対する影響
腸管出血性大腸菌は腸上皮細胞に接着する。第2の実験において、LOVO細胞の集密の単層を細菌試験株と一緒に、大腸菌:宿主細胞100:1の感染多重度(MOI)でプレインキュベートした。EcNおよび大腸菌G3/10の影響を試験した。2時間インキュベートした後、細胞を洗浄し、EHEC:宿主細胞100:1のMOIを用いてEHECを感染させた。in vitro接着アッセイを用いて3つのEHEC分離株を指標株として試験した。HUS関連EHEC86−24血清型O157:H7およびEHEC O104:H4アウトブレイク株の2つの異なる分離株ならびにEHEC O128:H2患者分離株を使用した。EHEC O104:H4分離株はESBL産生体であるので、医学的処置は限定される。%単位の接着効率は、いかなるプレインキュベーションもしていない接着(陰性対照)を100%に設定し、それと比較したEHECの接着として表されている。データは、2連のウェルでの3回の別々の実験の平均±SDである。EHEC接着は、大腸菌G3/10によってのみ阻害されたが、これは、EcN由来のMccM/H47と大腸菌G3/10由来のMccSの作用機構が異なることを示す。図5は、EcNまたは大腸菌G3/10と一緒にプレインキュベートした後の、EHEC株86−24血清型O157:H7(A)、ドレスデンにおいて単離されたEHEC O104:H4(B)ならびにフランクフルト(オーデル)において単離されたEHEC O104:H4(C)およびO128:H2(D)のヒト腸上皮細胞内への接着効率を示す。
種々の大腸菌および他のenterobacteriaceaeにおけるミクロシンS遺伝子MccSについてのスクリーニング
ヌクレオチドおよびアミノ酸のデータベースにおいてミクロシンSの配列は未知である。MccS遺伝子クラスターについてのスクリーニングを38種の大腸菌株、2種の赤痢菌株および2種のSalmonella株において、多重PCR、および阻害対照として遺伝子recAを用いて実施した。試験した株は、異なる起源の一般的な実験室株、ヒト臨床分離株および動物分離株である。McsS遺伝子は、試験した42種のヒト分離株および動物分離株ならびに実験室株のいずれにおいても検出することができなかった。結果は図6に示されている。
Claims (15)
- a)配列番号6もしくは配列番号7のアミノ酸配列と少なくとも90%以上同一であるアミノ酸配列を含み、
b)配列番号1もしくは2のいずれか1つのヌクレオチド配列、もしくはその相補体を含むヌクレオチド配列と少なくとも90%以上同一であるヌクレオチド配列を含む核酸分子によりコードされ、
c)配列番号1もしくは2のいずれか1つのヌクレオチドの配列、もしくはその相補体を含むヌクレオチド配列と相補的なヌクレオチド配列とストリンジェントな条件下でハイブリダイズする核酸分子によりコードされ、または
d)配列番号6もしくは配列番号7のアミノ酸配列を含むポリペプチドの天然に存在する対立遺伝子変異体を含み、
抗微生物活性を有する単離されたポリペプチド。 - a)配列番号1もしくは2のいずれか1つのヌクレオチド配列、もしくはその相補体と少なくとも90%以上同一であるヌクレオチド配列を含み
b)配列番号1もしくは2のいずれか1つのヌクレオチド配列、もしくはその相補体を含むヌクレオチド配列と相補的なヌクレオチド配列とストリンジェントな条件下でハイブリダイズする核酸分子を含み、
c)配列番号6もしくは配列番号7のアミノ酸配列と少なくとも90%以上同一であるアミノ酸配列を含むポリペプチドをコードするヌクレオチド配列を含み、または
d)配列番号6もしくは配列番号7のアミノ酸配列を含むポリペプチドの天然に存在する対立遺伝子変異体をコードする核酸分子を含み、
抗微生物活性を有するミクロシンSポリペプチドをコードする単離された核酸分子。 - 請求項2に記載の核酸分子または請求項1に記載のポリペプチドをコードする核酸分子を含むプラスミド。
- 請求項1に記載のポリペプチドに選択的に結合する抗体。
- 請求項1に記載のポリペプチドを含む組成物。
- 薬学的に許容される担体をさらに含む医薬組成物である、請求項5に記載の組成物。
- 表面用の消毒薬である、請求項5に記載の組成物。
- 療法または予防において使用するための、請求項1に記載のポリペプチドまたは請求項5に記載の組成物。
- 微生物感染症の治療または予防において使用するための、請求項1に記載のポリペプチドまたは請求項5に記載の組成物。
- 微生物感染症が腸管病原性大腸菌および/または腸管出血性大腸菌(EPEC、EHEC)の感染症を含む、または溶血性尿毒症症候群(HUS)を伴う微生物感染症の治療または予防において使用するための、請求項9に記載のポリペプチド。
- 胃腸障害または機能性胃腸障害の治療または予防において使用するための、請求項1に記載のポリペプチドまたは請求項5に記載の組成物。
- 請求項1に記載のポリペプチドを作製するための方法であって、請求項2に記載の核酸分子もしくは請求項3に記載のプラスミドを使用すること、または液相もしくは固相ペプチド合成技法によって請求項1に記載のポリペプチドを合成することを含む方法。
- ミクロシンSに対して感受性の細菌を同定するためのin vitro方法であって、請求項1に記載のポリペプチドまたは請求項2に記載の核酸分子によりコードされるポリペプチドまたは請求項5に記載の組成物を細胞と接触させることを含み、細胞の腸上皮細胞への接着効率が低下することにより、細胞のミクロシンSに対する感受性が示される方法。
- 食料品を保存するための方法であって、
食料品に少なくとも1種類の天然の抗微生物性作用剤を添加することを含み、作用剤が
a)請求項1に記載のポリペプチド;および/または
b)請求項5に記載の組成物
である方法。 - 包帯材をコーティングするための方法であって、
包帯材を、
a)請求項1に記載のポリペプチド;および/または
