Description
しかし、これまでの徐放研究は薬物を長時間かけて徐々に放出するものであって、薬物の放出開始時期も含めて生体内にてin situに徐放制御できるものは報告されていない。
中でも、中枢神経疾患の一つであるパーキンソン病は、薬剤などで病態の進行を食い止めることは可能と言われているが、完治は、神経幹/前駆細胞の移植による治療でしか不可能であろうと言われており、細胞移植医療の実現に大きな期待が寄せられている。
しかしながら、この幹細胞移植による治療において、大きな問題として挙げられるのが、幹細胞を移植しても、損傷した組織には、幹細胞を制御できるだけの能力がなく、移植細胞が制御されないため、損傷組織の再生には至らないという問題である。
そこで、様々な方法が模索されているものの、有効な手法の確立には至っていない。
一方で、幹細胞の分化制御において、複数の因子が段階的に作用することで効率良く分化誘導されることが、分子生物学的知見から周知の事実となっている。
つまり、ある分化状態において、Aという因子が作用し、その次にBという因子が作用するといった具合に、段階的に且つ時機を規定して複数の因子を作用させることが、幹細胞の効率的な分化誘導には必要不可欠である。
しかし、従来のタンパク質DDSは、機能因子の作用時機を規定できるようなシステムの構築には至っていない。
この理由として、これまで、内包したタンパク質を目的の場所に運搬して目的の細胞の近くで徐放させることのみが目的事項とされ、作用させる時機を規定しようという考え方自体が要求されていなかったことも理由として挙げられる。
本発明者らは、移植した幹細胞を効率良く分化誘導させ組織を再生できる材料を創製することにより、パーキンソン病治療のための神経幹/前駆細胞移植医療が実現に向けて大きく前進すると考え、in situで分化誘導制御できるタンパク質運搬材料の創製を課題として研究を行い、本発明に係る組成物創製に至った。
本発明において徐放制御とは、生体内において生理活性物質の放出を開始するタイミングと放出速度が制御されていることをいう。
また、神経分化に特化しない場合においては、塩基性線維芽細胞増殖因子(bFGF)、血管新生関連因子の1つである血管内皮細胞増殖因子(VEGF)、毛様体神経栄養因子(CNTF)、神経細胞を取り巻く外環境を構築する細胞に効果的な血小板由来増殖因子(PDGF)、炎症反応の鎮静化を担う因子であるインターロイキン10(IL10)などのタンパク質も細胞制御のための効果的な因子である。
生理活性物質を内層の生分解性物質に担持させる際には、生理活性物質が不活性になることがなく且つ安定的な結合を用いる。
ここでいう安定的な結合とは、108〜1010M−1の会合定数を有する生理活性物質と生分解性物質との間の特異的な分子間相互作用による結合である。
生理活性物質としてタンパク質を選び、内層の生分解性物質としてコラーゲンを選んだ際に、安定的な担持を達成する手段の一つとして、遺伝子工学技術を応用したキメラタンパク質合成法を利用することが挙げられる。
例えば、タンパク質のC末端にコラーゲン結合性ペプチド(CBP)を融合したキメラタンパク質を作製すると、タンパク質はペプチド端とコラーゲンとの間に作用する分子間相互作用でコラーゲンに担持される。
また、この担持方法であれば、タンパク質が不活性になることもない。
例えば、神経幹/前駆細胞は脳由来神経栄養因子(BDNF)の作用をうけて初期神経に分化誘導されるが、その間はグリア細胞由来神経栄養因子(GDNF)は不要とされている。
これは、GDNF受容体が、初期分化が誘導されることで大量に発現するものであり、GDNFが必要となるのは受容体が発現した後だからである。
従って、GDNFを作用させるタイミングを初期神経から中後期神経、ドーパミン神経に分化する段階に合せるのが有効である。
そこで例えば、GDNFのC末端にコラーゲン結合性ペプチド(CBP)を融合させたGDNFキメラタンパク質を合成し、内層として用いたコラーゲンに担持させると内層(コラーゲン)に酵素的又は加水分解的な分解が起こらない限り、GDNFが放出されないことになるため、生理活性物質の放出開始のタイミングを制御することができる。
よって、内層の生分解が開始されると生理活性物質が外部に透過する外層を用いるのが好ましい。
本発明に用いる内層としてはコラーゲン,ゼラチン,ヒアルロン酸等の生体由来の天然高分子が好ましく、外層としては非晶質性の生分解性高分子が好ましい。
非晶質性の生分解高分子としては、ポリ乳酸(PLA),ポリグリコール酸(PGA),ポリカプロラクトン(PCL),ポリヒドロキシ酪酸等のポリヒドロキシアルカン酸(PHA)及び乳酸−グリコール酸共重合体(PLGA)等が例として挙げられる。
最終的には、外層そのものの消失が要求されるが、PLGAは乳酸とグリコール酸の配合比率を変えることで、生体内での分解速度をコントロールすることができる。
そこで、体内で免疫担当細胞をはじめとする様々な細胞による貪食を受けない大きさの微粒子にするのが好ましい。
また、生理活性物質は微粒子から徐放されない限り、生体内の様々な細胞による消費を避けなければならない。
従って、そのような場合は、内層は生理活性物質を担持した略球状のコアであり、前記外層がコアの被覆層である粒子形状になっているのが好ましく、さらに粒子の大きさが平均で5〜60μmの範囲が好ましい。
粒子は細胞に貧食されない大きさであれば、問題はない。
この場合も、被覆層を形成した粒子の外層が生理活性物質の放出後に消失する。
また、外層の厚さを調整することによっても内層の生分解を遅らせることが可能であり、放出させたいタイミングを厳密に制御することができる。
以下、具体例を挙げて説明する。
本発明者の過去の研究から、コラーゲンは、生体内での半減期が約10日から2週間であることが分かっている。
神経幹/前駆細胞が神経前駆細胞や初期神経細胞に分化するのに約1週間程度必要とし、その後、GDNFのシグナルを受けてドーパミン神経に分化する。
そのタイムスケールから考えて、神経へ分化している途中にちょうどコラーゲンの分解に伴うGDNFの放出が行われ、効率良く分化を誘導できることになる。
従来から存在するタンパク質DDS技術を用いた場合では、神経幹/前駆細胞と微粒子の共移植直後からGDNFが少しずつ徐放される。
しかしながら、神経幹/前駆細胞にとって、移植直後にGDNFは必要なく、分化が進行した初期神経細胞において必要となる。
不必要なときにGDNFが放出されることは、無駄になるばかりか、GDNFが移植細胞以外のホスト由来の細胞に作用することに伴う副作用も考えられる。
また、本当に必要な時にGDNFが枯渇している可能性が高い。
また、PLGAをはじめとする外層を形成する生分解性高分子の膜厚を制御することによりコラーゲンの分解速度を調節でき、コラーゲンの生分解によりGDNFが放出後、数か月で微粒子自体が消失する。
<実験1:タンパク質内包微粒子の作製>
Water/Oil/Waterダブルエマルション法を用いて微粒子状の組成物を作製した。
4%PLGAおよび0.1%Pluronic−F68(SIGMA ALDRICH社製:ポリエチレングリコール−ポリプロピレングリコール−ポリエチレングリコールブロック共重合体)を含むクロロホルム溶液をOil相とし、0.5%コラーゲンおよび0.1%Pluronic−F68を含む水溶液をWater相とした。
また、Water相にはコラーゲン結合性ペプチド融合グリア細胞由来神経栄養因子(GDNF−CBP)を20μg/mLになるように添加した。
この2種の溶液を混合し、マイクロ超音波処理を行うことにより微粒子を作製した。
得られた微粒子懸濁液を0.5%Pluronic−F127(SIGMA ALDRICH社製)水溶液に滴下し、界面活性剤によるコーティング処理を行った後、遠心分離により回収した。
微粒子のコーティング処理においては、親水性生分解性高分子(例えばポリビニルアルコールなど)によるコーティングも可能である。
回収した微粒子は、限外濾過法などを利用して、5〜40μm径の微粒子のみにサイズ分画した。
図1(A)は、限外濾過によるサイズ分画精製後に得られた微粒子の走査型電子顕微鏡像である。
また、図2に上記微粒子の粒径分布をグラフに示す。
微粒子にGDNF−CBPが取り込まれていることを調査するため、赤色蛍光(Alexa594)でラベル化したGDNF−CBPを用いて、同様に微粒子を作製した。
一般的なタンパク質の蛍光ラベル化方法を利用して、Alexa594ラベル化GDNF−CBPを作製し、微粒子に内包させた。
図1(B)は、赤色蛍光ラベル化GDNF−CBPを内包した微粒子の蛍光顕微鏡像を示す。
なお、図1(B)はグレースケールになっているので分かりにくいが、実際の画像は微粒子が赤色蛍光を発していたことから、GDNF−CBPは目的通り、微粒子に内包されていることが分かった。
内包したGDNF−CBPの放出時機規定が可能であることを調査するため、リン酸緩衝液(PBS)およびコラーゲン分解酵素含有PBS中に浸漬した微粒子からのGDNF−CBPの放出について評価した。
その結果を図3及び図4に示す。
図3のグラフ(A)はコラーゲン分解酵素(コラゲナーゼ)の濃度が0.3%で図3のグラフ(B)はその濃度が0.15%のものである。
図4のグラフ(C)はPBS中のコラーゲン分解酵素の濃度が0.01%で、図4のグラフ(D)はコラーゲン分解酵素を含まないPBS中に20日間浸漬した結果を示す。
グラフ(D)では20日間にわたってGDNFの放出がほぼ認められないことから、実験1にて作成した組成物(微粒子)はコラーゲン分解酵素存在下でしかGDNFの徐放がないことが確認できた。
コラーゲン分解酵素の濃度を変えたグラフ(A),(B),(C)を比較すると、PBSのみの段階ではGDNF−CBPの放出が認められずにコラーゲン分解酵素を添加すると、GDNF−CBPの徐放が認められ、コラーゲン分解酵素の濃度が高い程、GDNF−CBPの放出速度が速い。
0.01%コラーゲン分解酵素存在下では徐放量及び放出速度が0.15%,0.3%に比べ減少するものの、充分に徐放効果があることが認められた。
また、1mgの微粒子に含まれるGDNF−CBP量は1.46±0.21μgであった。
この結果から、コラーゲンの分解に合わせて、タンパク質の放出が制御できることが分かった。
また、コラーゲン分解酵素の濃度又は、PLGAの膜厚の変更(微粒子作製時のPLGAの濃度を変更することにより外層厚を変更)に応じて、放出速度が制御できることが分かっている。
微粒子に内包したGDNF−CBPの活性について調査した。
まず、微粒子を作製する際に界面活性剤を使用している。
界面活性剤は、ポリエチレングリコール−ポリプロピレングリコール−ポリエチレングリコールの3元ブロック共重合体(SIGMA ALDRICH社製:Pluronic−F68またはPluronic−F127)を使用した。
ここで、Pluronic−F68およびF127と混合したタンパク質の活性を細胞培養試験により調査した結果、今回用いた界面活性剤はタンパク質に対して問題がないことが分かった。
また、微粒子から放出されたGDNF−CBPについて、同様に細胞培養試験によって活性を調査した。
図5は、神経幹/前駆細胞にBDNFを添加した後、7日目に本発明に係る実験にて作成した微粒子から放出したGDNF−CBPを添加した蛍光マーカー画像を示し、(1)は神経前駆細胞の蛍光画像を示し(2)は成熟神経の蛍光画像を示し(3)は神経前駆細胞、成熟神経及び核の蛍光画像を重ね合せたものを示す。
その結果、市販のGDNFと同等の活性を有することが分かった。
これらの結果から、微粒子の作製過程でもタンパク質の失活は無く、また、微粒子に内包されている状態においてもGDNFが失活することはないと考えられる。
あらかじめ一般的な初期神経分化誘導法を用いて4日間培養した初期神経細胞に、実験1にて作成した微粒子を図6に示すように、a:50μg,b:100μg,c:150μg添加した各群に、0.01%コラーゲン分解酵素を含む培地中で14日間培養を行った。
なお、0.01%コラーゲン酵素は毎日添加し、培地は2日毎に半量を交換した。
図6はグレースケールにても分かりやすいように中後期神経マーカー(MAP2)のみの蛍光画像を示す。
この結果から微粒子の添加量が多くなるに伴い、徐放されるGDNF量が多くなり、成熟神経の割合が多くなることが分かった。
なお、図示を省略したが、神経マーカーであるβ−tubulinIIIの蛍光画像を見ると微粒子の添加量が多くなるにつれて神経突起が太く成長していることも確認できた。
比較のために、微粒子を添加したがコラーゲン分解酵素を全く添加していないものも調査したところ、MAP2陽性細胞の出現が全く認められず、β−tubulinIII陽性細胞の突起も細く短いものばかりで神経の成長が殆ど認められなかった。
図7は実験1にて作成した微粒子の細胞分化誘導をMAP2陽性細胞率で評価したグラフを示す。
コラーゲン分解酵素の濃度は0.01%である。
これにて200μg/106cellsの添加量までは、添加量が多くなるに伴いMAP2陽性細胞率は高くなるものの、300μg/106cellsでは若干低下することが明らかになった。
図8は、あらかじめ培養を行った神経幹/前記細胞に、実験1にて作成した微粒子の添加をしないものと、各種量を添加したものの生細胞数を比較した結果を示す。
これにより今回調査した300μg/106cells以下の範囲であれば細胞への影響がないことが分かった。
図9は、MAP2の蛍光画像を示し、(a)はPBS中にて1ヶ月間冷蔵保存した微粒子、(b)はPBS中にて1ヶ月間−30℃の凍結保存した微粒子、(c)はPBS中にて凍結乾燥させた状態で1ヶ月間保存した微粒子をそれぞれ用いて2週間培養による分化誘導試験を行った後のMAP2蛍光画像を示す。
図10は、Type1〜6に示す各保存条件にて1ヶ月間保存した後の2週間培養分化誘導におけるMAP2陽性細胞の割合を示す。
このことから保存方法としては、凍結保存又は凍結乾燥保存が好ましい。
ここでHBPとは、ヒアルロン酸に特異的な結合するペプチド配列のことをいう。
外層は実験1と同様にPLGAである。
図11に、上記微粒子をヒアルロン酸分解酵素の濃度0.1%及び0.01%のPBS中に浸漬した際の放出されるGDNF−HBPの濃度変化を測定したグラフを示す。
この結果から、コラーゲンの替わりにヒアルロン酸を用いても徐放制御が可能であることが分かる。
また、この微粒子を神経幹/前駆細胞と共存させ、2週間培養を行ったところ、成熟神経への分化が認められ、ヒアルロン酸分解酵素を添加しなかった培養系では成熟神経への分化が認められなかった。
このことから本発明に係る徐放制御組成物は、内層のタンパク質等を担持する生分解性物質を適宜、替えても成立させることができる。
これにより、内層に担持させる生理活性物質も適宜選択使用できることがわかる。
また、外層としてポリL−乳酸(PLLA)を用いても同様に作成できた。
これにより、外層の高分子も適宜選択使用できることが分かる。
移植は、ラット線条体に行った。
神経幹/前駆細胞(106cells)+GDNF内包PLGA/コラーゲン微粒子+Matrigel(市販品)を混合して線条体へインジェクションした。
移植後、所定期間で移植組織周辺(移植領域をメスで切り出してくる)を回収し、RNAを抽出し、cDNAに逆転写しPCRを行った(RT−PCR)。
この実験により、分化マーカータンパク質のmRNA発現について調査した。
その結果を図14に示す。
GAPDHは、ハウスキーピング遺伝子で、これにより、PCRを行う際のサンプル間の遺伝子量を揃えた(規格化)。
Nestinは神経幹細胞マーカー、β−tubulin IIIは神経マーカーであり初期神経から成熟神経まですべてのステージの神経で発現している。
MAP2は、中後期神経マーカーで成熟神経マーカーの1つ。
TH(Tyrosine hydroxylase)は、ドーパミン神経マーカーである。
移植前、組織(Striatum)にはNestinの発現が極めて少ないが、移植後Nestin発現が認められる=移植細胞由来。
その後にNestin発現が減少し、移植細胞が分化していると考えられる。
TubulinのmRNA発現が、移植後7〜21日にかけて増加している。
これは、移植細胞の神経への分化に由来するものと考える。
MAP2のmRNA発現が、移植後D14から見られD21で増大していること、また、THのmRNA発現がD21で発現が見られることから、移植後1週間くらいから、リリースされたGDNFの効果で分化が誘導されていると考えられ、そしてドーパミン神経へ誘導できていると言える。
Claims (6)
- 生理活性物質を担持した生分解性物質からなる内層と、当該内層とは異なる生分解性物質からなる外層とからなり、
前記生理活性物質はタンパク質であり、
前記内層の生分解性物質がコラーゲン又はヒアルロン酸であり、
前記担持手段は生理活性物質であるタンパク質を、内層との分子間相互作用の強いコラーゲン結合性ペプチド又はヒアルロン酸結合性ペプチドを融合させたキメラタンパク質にすることで生理活性物質と内層との会合定数が108〜1010M−1の範囲であることを特徴とする生理活性物質徐放制御組成物。 - 前記外層は内層の生分解に伴い、内層に担持された生理活性物質が外部に透過するものであることを特徴とする請求項1記載の生理活性物質徐放制御組成物。
- 前記内層は略球状のコアであり、前記外層がコアの被覆層である粒子形状になっていることを特徴とする請求項1又は2に記載の生理活性物質徐放制御組成物。
- 前記粒子の大きさが平均で5〜60μmの範囲であることを特徴とする請求項3記載の生理活性物質徐放制御組成物。
- 前記外層の生分解性物質が非晶質性の生分解性高分子であることを特徴とする請求項1〜4のいずれかに記載の生理活性物質徐放制御組成物。
- 前記非晶質性の生分解性高分子が乳酸−グリコール酸共重合体(PLGA),ポリ乳酸(PLA),ポリヒドロキシアルカン酸(PHA)のいずれかである請求項5記載の生理活性物質徐放制御組成物。
