癌細胞に感染するコクサッキーウイルスA11型に由来する核酸又は癌細胞に感染するエコーウイルス4型に由来する核酸を含む。
本発明の実施の形態1について詳細に説明する。本実施の形態に係る医薬組成物は、癌細胞に感染するCVA11型又は癌細胞に感染するEV4型を含む。CVA11型及びEV4型は、細胞表面のウイルス受容体に結合することによって、当該細胞に感染することができる。ウイルス受容体として、例えば、崩壊促進因子(DAF又はCD55)、細胞間接着分子‐1(ICAM‐1又はCD54)、インテグリンα2β1(CD49b)が知られている。CVA11型及びEV4型がウイルス受容体と相互作用することで、CVA11型及びEV4型のカプシドが脱安定化される。これにより、CVA11型及びEV4型の脱外皮が誘導される。
本発明の実施の形態2について詳細に説明する。本実施の形態に係る医薬組成物は、癌細胞に感染するCVA11型に由来する核酸又は癌細胞に感染するEV4型に由来する核酸を含む。
EV4型(Du toit)の調製
EV4型は、RD細胞を用いて増殖させた。Dulbecco’s modified Eagle medium(DMEM)を用いて継代培養したRD細胞にEV4型を1時間孵置した後、培地をDMEMに置換し、細胞変性効果が始まるまで静置した。培地を除去後、培養用シャーレにOPTI-MEM Iを添加し、セルスクレーパーを用いて細胞を剥離回収した。なお、EV4型及びRD細胞は、インキュベーター内で37℃、5%CO2下で培養した。液体窒素を用いて、回収したRD細胞の凍結、融解を3回繰り返した後、4℃、3000rpmで15分間遠心を行い、上清を回収した。回収した上清(ウイルス溶液)は、−80℃で保存した。
以下で用いるMOIは、以下の方法で算出した。
細胞を96穴のプレートに5×103cells/100μl/ウェルで播種し、37℃、5%Co2下で5時間維持した。ウイルスは、OPTI-MEM Iで100倍又は1000倍希釈して、これをMOI測定用のウイルス原液とした(ここでの希釈倍率の常用対数をLとする)。ウイルス原液を10倍ずつ段階希釈し(ここでの希釈倍率の常用対数をdとする)、希釈系列液を調製した。次に、各ウェルに希釈系列液を0.05mlずつ添加した(添加した希釈系列液の体積をvとする)。120時間後に50%以上の細胞変性効果が認められたウェル数の合計を8で除算した値Sを算出し、MOIを以下の式で算出した。

クリスタルバイオレット法により、EV4型による殺癌細胞効果としての細胞傷害性を評価した。試験対象の各細胞を72時間後にコンフルエントになる密度(3×104cells/ウェル)で24穴のプレートに播種した。適切な感染力価(MOI=0.001、0.01、0.1)になるように、OPTI-MEM IでEV4型を希釈し、EV4型の希釈液を調製した。約6時間後、プレートから培地を除去し、EV4型の希釈液を各ウェルに200μl添加し、37℃、5%CO2下にプレートを1時間維持した。次に、EV4型の希釈液を除去し、各細胞用培地を各ウェルに1ml加え、72時間培養した。72時間後、リン酸緩衝食塩水(PBS)で緩やかに洗浄し、0.5%グルタルアルデヒド含有PBSを各ウェルに300μl添加した後、15分間室温に静置することで生存接着細胞を固定した。その後、グルタルアルデヒド含有PBSを除去し、PBSで洗浄後、2%エタノール及び0.1%クリスタルバイオレットを含有する滅菌水を、各ウェルに300μl添加し、室温で10分間静置することで生細胞を染色した。
上記試験対象の細胞表面におけるCD49b、CD55の発現量を定量するため、蛍光標識した抗体と反応させた各細胞を、フローサイトメトリー法で解析した。培養された細胞を回収し、2×106cells/mlになるようにPBSに懸濁した。96穴のプレートの各ウェルに、得られた細胞懸濁液を100μlずつ分注した(2×105cells/ウェル)。4℃、2000rpmで5分間遠心後、ペレットにフルオレセイン・イソチオシアネート(FITC)標識抗ヒトCD49b抗体、又はフィコエリシン(PE)標識抗DAF(CD55)抗体を含む1%ウシ血清アルブミン(BSA)含有PBSを100μlずつ添加した。暗所条件下、プレートを氷中に1時間静置し、抗体を反応させた。コントロールの細胞は、アイソタイプIgG抗体で標識した。
表1は、EV4型の癌細胞に対する細胞傷害性、CD49b及びCD55の発現量を示す。癌細胞に対する細胞傷害性は、クリスタルバイオレット法で評価した細胞傷害性が66%以上の場合を「3+」、33%以上66%未満の場合を「2+」、0%より高く33%未満の場合を「+」、0%の場合を「−」で示した。この結果、小細胞肺癌、非小細胞肺癌、大腸癌、膵癌のいくつかの細胞株で高い細胞傷害性を認めた。また、食道癌扁平上皮癌における全ての細胞株で、極めて低いMOI=0.001でも高い細胞傷害性を認めた。食道癌扁平上皮癌における全ての細胞株は、いずれもCD49b及びCD55の90%以上の高い発現を認めた。このことから、EV4型は、CD49b及びCD55をウイルス受容体として癌細胞に感染することが示唆された。
ヒト正常皮膚角質細胞(HaCaT)に対しては、いずれのMOIにおいても細胞傷害性が0%であった。
EV4型の細胞傷害性に対する各種阻害剤の影響(1)
EV4型の細胞傷害性に対する影響を検討した阻害剤は、汎カスパーゼ阻害剤としてのZ−VADfmk(R&D Systems社)、PI3K阻害剤としてのLY294002(Santa Cruz Biotechnology社)、MEK阻害剤としてのPD0325901(Wako社)、PTEN阻害剤としてのbpV(Merck社)である。
図2は、T.Tnに対するEV4型(DMSO、MOI=0.1)のみ、EV4型とアポトーシス阻害剤としての汎カスパーゼ阻害剤(Z−VADfmk)の併用、EV4型とPI3K阻害剤(LY294002)の併用、EV4型とMEK阻害剤(PD0325901)の併用、EV4型とPI3Kを阻害することが知られるPTEN阻害剤(bpV)の併用において、感染4日後の細胞傷害性に対する影響を顕微鏡像により示した図である。EV4型による細胞傷害性は、アポトーシス阻害剤及びPI3Kを阻害することが知られるPTEN阻害剤により減弱された。また、EV4型による細胞傷害性は、PI3K阻害剤及びMEK阻害剤により増強された。
したがって、カスパーゼ依存性アポトーシス及びPI3K/Akt、MEK/ERK細胞増殖シグナル伝達系はEV4型による細胞傷害性(EV4の食道癌細胞内増殖機構)に関与することが示唆された。これにより、EV4型を含む医薬組成物は、近年固形癌治療の臨床試験で使用されている新規分子標的薬PI3K(Akt)あるいはMEK阻害剤(MAPキナーゼキナーゼ阻害剤)との併用によって、その抗腫瘍効果が増強されることが強く示唆された。
EV4型の細胞傷害性に対する各種阻害剤の影響(2)
EV4型の細胞傷害性に対するMEK阻害剤PD0325901の影響をより定量的に検討した。
図3は、対照及びEV4型を感染させたTE8の細胞生存率を示す。PD0325901を作用させることで、EV4型によるTE8の殺細胞効果が有意に増強された。
このことからも、EV4型を含む医薬組成物は、MEK阻害剤との併用によって、その抗腫瘍効果が増強されることが強く示唆された。
EV4型のin vivo抗腫瘍効果の評価(1)
実施例1で確認したEV4型の癌細胞に対する細胞傷害性による腫瘍退縮能を、ヒト食道癌扁平上皮癌の細胞株TE8による担癌ヌードマウスを用いて検討した。TE8をPBSで洗浄し、1.0×107cells/mlになるようにOPTI−MEM Iに懸濁した。 6−8週齢のBALB/cヌードマウスの右側腹部にTE8を含む懸濁液を、100μlずつ27G針を用いて皮下注射した。マウスは非投与群と3つのEV4型投与群に無作為に分配した。腫瘍の長径及び短径は、ノギスを用いて測定した。腫瘍長径が4mm前後になって腫瘍がマウスに定着したのを確認して、50μlのOPTI−MEM Iに懸濁したEV4型溶液を腫瘍内に1回投与した。各EV4型投与群に対して投与したEV4型の感染価は、1.0×105、1.0×106、1.0×107TCID50とした。TCID50は、ヒト横紋筋肉腫の細胞であるRD細胞を用いて、ウイルス感染の5日後に評価した。非投与群に対しては、右側腹部にEV4型を含まないOPTI−MEM Iを、EV4型投与群と同量投与した。EV4型投与4日後、各EV4型投与群の腫瘍体積及び体重を測定した。なお、腫瘍体積は、長径×短径×短径×0.5で算出した。
図4は、非投与群の腫瘍体積に対する各EV4型投与群の腫瘍体積の相対値を示す。EV4型を投与したマウスでは、非投与群と比較して、EV4型の用量依存的に腫瘍体積の増加が有意に抑制された。また、図5は、非投与群及び各EV4型投与群の体重を示す。この結果、EV4型投与群において有意な体重減少を認めなかった。この時点での体重減少は、有害事象を示唆するため、体重減少を認めないことは、EV4型投与による明らかな有害事象を認めなかったといえる。この点は、致死性の重篤な有害事象が認められたエンテロウイルスCVA21型と異なっており、EV4型の安全性を裏付けるものである。
EV4型のin vivo抗腫瘍効果の評価(2)
EV4型の癌細胞に対する細胞傷害性による腫瘍退縮能を、TE8による担癌ヌードマウスを用いてさらに検討した。TE8をPBSで洗浄し、OPTI−MEM Iに懸濁した。ヌードマウスの右側腹部に、2.0×106cells/投与になるようにTE8を、27G針を用いて皮下接種した。ノギスで測定した腫瘍長径が4mm以上になったのを確認して(0日目)、50μlのOPTI−MEM Iに懸濁したEV4型溶液を、1×107TCID50/投与となるように1日1回、計10回腫瘍内に投与した。なお、対照には、同量のOPTI−MEM Iを投与した。腫瘍退縮能の感染価に対する依存性を検討するため、同様の実験に5×107TCID50/投与群を加えて行った。評価項目は、上記実施例4と同様に腫瘍体積と体重とした。また、有害事象の有無を確認した。
図6は、腫瘍体積の経時変化を示す。対照と比較して、EV4型を投与したマウスでは、腫瘍体積の増大の抑制が見られた。EV4型を投与したマウスの腫瘍体積は、最初のEV4型投与後18日目において対照の50%程度にしかならず、有意な腫瘍退縮能を示した。この実験におけるマウスの体重の経時変化を図7に示す。対照と同じように体重が維持され、重篤な有害事象は見られなかった。
2種類の感染価のEV4型を投与したときの腫瘍体積の経時変化を図8に示す。EV4型投与群は、EV4型による腫瘍体積の増大の抑制には、感染価依存性が見られた。この実験におけるマウスの体重の経時変化を図9に示す。感染価を高めてEV4型を投与しても、対照と同じように体重が維持され、重篤な有害事象は見られなかった。
以上のことから、EV4型は、複数回投与しても比較的長期間に渡って安全性が高く、抗腫瘍効果を発揮することが示された。
CDDP抵抗性腫瘍に対するEV4型のin vivo抗腫瘍効果の評価
食道癌の化学療法で用いられるCDDP(シスプラチンともいう)に抵抗性を示すTE8担癌ヌードマウスに対するEV4型の腫瘍退縮能を検討した。TE8を実施例5と同様にヌードマウスに接種し(0日目)、腫瘍長径が4mm以上になったのを確認して、生理食塩水に溶解したCDDPを125μg/投与で腹腔内に投与した(2日目)。なお、未治療群には、同量の生理食塩水を投与した。TE8の接種後8日目から、さらに50μlのOPTI−MEM Iに懸濁したEV4型溶液を1×107TCID50/投与又は5×107TCID50/投与で1日1回、計5回腫瘍内に投与した。なお、未治療群及びCDDP投与群には、同量のOPTI−MEM Iを投与した。評価項目は、上記実施例4と同様に腫瘍体積と体重とした。
図10は、腫瘍体積の経時変化を示す。CDDP投与群は、8日間、腫瘍体積の増大を抑制したものの、その後腫瘍体積の増大が見られた。このようにCDDPに抵抗性を示すTE8担癌ヌードマウスに対して、1×107TCID50及び5×107TCID50のEV4型を投与したマウスは、対照及びCDDP投与群それぞれと比較して、どちらも有意に腫瘍体積の増加を抑制し、CDDP抵抗性腫瘍に対して抗腫瘍効果を示した。
図11は、TE8担癌ヌードマウスの体重の経時変化を示す。EV4を投与しても体重は維持され、重篤な有害事象は見られなかった。
以上のことから、EV4型は、抗癌剤抵抗性の腫瘍に対して、高い安全性のもとで、抗腫瘍効果を発揮することが示された。また、EV4型は、CDDPなどの抗癌剤と併用することで抗癌剤抵抗性の腫瘍に対して強力な抗腫瘍効果を発揮することが示された。
CVA11型のクリスタルバイオレット法を用いたスクリーニング(1)
実施例1と同様にCVA11型による殺癌細胞効果としての細胞傷害性を評価した。試験対象の細胞は、小細胞肺癌(SBC−5、SBC−3)、非小細胞肺癌(H1299、H460、LK87、A549)、肺扁平上皮癌(QG95)、大腸癌(DLD−1、SW620、Lovo、Caco−2)、悪性中皮腫(MSTO、H2052、H2452、H28)、食道癌扁平上皮癌(T.Tn、TE6、TE8)、舌扁平上皮癌(HSC3、HSC4)、下咽頭癌(FaDu)、喉頭癌(Hep2)、ヒトBリンパ腫(Daudi)である。
上記試験対象とした細胞表面におけるCD54、CD55の発現量を定量するため、蛍光標識した抗体と反応させた各細胞を、上記実施例1と同様にフローサイトメトリー法で解析した。CD54の検出には、アロフィコシアニン(APC)標識抗ヒトICAM−1(CD54)抗体を用いた。なお、Daudiに関しては、ウイルス受容体の定量の対象外とした。
表2は、CVA11型の細胞傷害性、CD54及びCD55の発現量を示す。非小細胞肺癌の細胞株H1299、H460に対してMOI=0.001以上で強い細胞傷害性を認めた。A549に対しては、MOI=0.1で強い細胞傷害性を認めた。一方、大腸癌の全ての細胞株に対してMOI=0.1において強い細胞傷害性を認め、SW620に対しては、MOI=0.001でも強い細胞傷害性を認めた。また、LoVoに対しては、MOI=0.01でも強い細胞傷害性を認めた。食道癌扁平上皮癌の細胞株TE6、TE8に対しては、MOI=0.01で強い細胞傷害性を認めた。その他、小細胞肺癌の細胞株SBC−3、悪性中皮腫の細胞株MSTO及びH2052、下咽頭癌の細胞株FaDu、ヒトBリンパ腫の細胞株Daudiに対しても細胞傷害性を認めた。
また、CD54の発現の低い細胞(SBC−5、LK87、QG95、H28、Hep2)では細胞傷害性を認めず、ICAM−1がCVA11型のウイルス受容体のひとつであることが示唆された。
CVA11型の感染機序の検討
(大腸癌の細胞株に対するCVA11型による細胞傷害性への抗ICAM−1中和抗体の影響)
大腸癌細胞株(DLD−1及びSW620)を3×104cells/ウェルで48穴のプレートに播種した。プレートから培地を除去後、各細胞用の培地で希釈した抗ヒトICAM−1抗体(R&D Systems社)溶液10μg/mlを添加した。1時間後、培地を除去後、MOI=0.001となるようにOPTI-MEM Iで希釈したCVA11型希釈液を、各ウェルに100μl添加し、37℃、5%Co2下で1時間維持した。感染後、CVA11型希釈液を除去し、各細胞用の培地を対応するウェルに1ml添加し、37℃、5%CO2下で12時間培養した。培養後、光学顕微鏡にて各ウェルを撮影した。なお、DLD−1及びSW620に対する培地は、それぞれRPMI+10%FBS、DMEM+10%FBSとした。
ICMA−1を発現していないRD細胞又はプラスミドを用いた遺伝子導入法によりICAM−1遺伝子を導入した細胞RD−ICAM−1を、24穴プレートに3×104cells/ウェルで播種した。10%FBSを含むDMEM培地で6時間培養後、OPTI-MEM Iに希釈したCVA11型を各MOIで1時間、感染培養した。3日後にクリスタルバイオレット法により細胞傷害性を評価した。なお、フローサイトメトリー法を用いて、RD−ICAM−1におけるICAM−1の発現を確認した。
図15は、培養後に撮影した大腸癌の細胞株(SW620及びDLD-1)の光学顕微鏡画像を示す。CVA11型のみでは、両細胞株ともにMockと比較して、細胞数が減少しており、明らかなCPE(Cytopathic Effect)を伴う細胞傷害性を認めた。一方、抗ICAM−1中和抗体の存在下では、CVA11型による細胞傷害性が消失した。
図16は、ICAM−1強制発現実験におけるRD細胞及びRD−ICAM−1のクリスタルバイオレット染色後の顕微鏡像を示す。ICAM−1を発現していないRD細胞では、細胞傷害性を示さなかったのに対して、ICAM−1を発現させたRD−ICAM−1では、強い細胞傷害性が見られた。
以上により、ICAM−1は、CVA11型の感染に関与するウイルス受容体の1つであることが示された。
従って、ICAM−1が、CVA11型のウイルス受容体であることが証明され、ICAM−1はCVA11型による細胞傷害性に重要である。このため、CVA11型は、癌細胞の中でも特にICAM−1を発現する細胞に対して強い細胞傷害性を発揮することが示唆された。
CVA11型のクリスタルバイオレット法を用いたスクリーニング(2)
実施例7と同様にクリスタルバイオレット法を用いて、CVA11型による殺癌細胞効果としての細胞傷害性を評価した。ただし、本実施例では、試験対象の各細胞を、当該細胞の大きさ及び増殖速度を勘案して、2×104〜2×105cells/ウェルで播種した。約6時間後、プレートから培地を除去し、EV4型の希釈液を各ウェルに200μl添加し、37℃、5%CO2下にプレートを1時間維持した。次に、EV4型の希釈液を除去し、各細胞用培地を各ウェルに1ml加え、72時間培養した。EV4型の希釈液は、MOI=0.001、0.01、0.1又は1.0に調製した。
試験対象の細胞は、HSC3、HSC4及びDaudiを除く実施例7で用いた細胞に加えて、非小細胞肺癌(H1975、H2009)、悪性中皮腫(MESO1、MESO4)、食道癌扁平上皮癌(TE1、TE4、TE5、TE9、KYSE170)、大腸癌(WiDr、HT29)、トリプルネガティブ乳癌(MDA−MB−468、MDA−MB−231)、乳癌(MCF7)、子宮頸癌(HeLa)である。なお、A549、H1975及びTE4は、それぞれゲフィチニブ一次抵抗性、ゲフィチニブ二次抵抗性及びCDDP抵抗性である。
表3は、呼吸器癌の細胞に対するCVA11型の細胞傷害性及びCD54の発現量を示す。SBC−5、H28及びMESO4を除くすべての細胞で、少なくともMOI=0.1で細胞傷害性を認めた。CVA11型は、ゲフィチニブ一次抵抗性であるA549及びゲフィチニブ二次抵抗性であるH1975に対してもMOI=0.1で強い細胞傷害性を示した。また、細胞傷害性とCD54の発現量には相関性が見られ、MOI=0.1で細胞傷害性を認めなかった細胞のうち、SBC−5及びH28におけるCD54の発現量が低かった。
以上の結果より、CVA11型を含む医薬組成物は、非小細胞肺癌、肺扁平上皮癌、小細胞肺癌、悪性中皮腫、食道癌扁平上皮癌、下咽頭癌、大腸癌、乳癌、子宮頸癌、Bリンパ腫等の治療に有効であることが示唆された。しかも、当該医薬組成物は、ゲフィチニブ一次抵抗性又はゲフィチニブ二次抵抗性の非小細胞肺癌、CDDP抵抗性の食道癌扁平上皮癌、オキサリプラチン抵抗性の大腸癌及びトリプルネガティブ乳癌にも抗腫瘍効果を有することが示唆された。
癌幹細胞マーカーの検出
上記試験対象に含まれる大腸癌の細胞(DLD−1、HT29及びWiDr)の表面における癌幹細胞マーカーCD133の発現量を定量するため、PE標識抗ヒトCD46抗体で標識した各細胞を、上記実施例1と同様にフローサイトメトリー法で解析した。CD133の検出には、APC標識抗ヒトCD133抗体を用いた。
フローサイトメトリー法で解析結果を図18に示す。CD133を発現する細胞の割合は、DLD−1(1.8%)に対して、オキサリプラチン抵抗性のHT29及びWiDrでより大きかった(それぞれ30.9%及び63.6%)。
このように、大腸癌由来のオキサリプラチン抵抗性細胞株において、CD133の発現量が高いことが示された。また、実施例9に示したように、CVA11型は、オキサリプラチン抵抗性のHT29及びWiDrに強い細胞傷害性を示すため、CVA11型は、癌幹細胞に対しても細胞傷害性を有することが示唆された。
オキサリプラチン抵抗性腫瘍に対するCVA11型のin vivo抗腫瘍効果の評価
100μlのPBSに懸濁したオキサリプラチン抵抗性癌細胞WiDrを5×106をヌードマウスの右側腹部に接種した。播種から1日目より腫瘍長径が4mm以上になったのを確認して、50μlのOPTI−MEM Iに懸濁したCVA11型溶液を、2日おきに合計4回腫瘍内に投与した。1回の投与におけるCVA11型の感染価は、3×106TCID50とした。TCID50は、ヒト子宮頚癌の細胞であるHeLaを用いて、ウイルス感染の5日後に評価した。腫瘍体積を上記実施例4と同様にして評価した。対照には、50μlのOPTI−MEM Iを腫瘍内に投与した。
図19は、腫瘍体積の経時変化を示す。CVA11型を投与したマウスは、対照と比較して腫瘍体積の増加を抑制し、有害事象も見られなかった。
このように、CVA11型は、生体におけるWiDrに対して抗腫瘍効果を示したため、CVA11型を含む医薬組成物は、オキサリプラチン抵抗性腫瘍に対する抗腫瘍効果を有することが示唆された。
Claims (9)
- 癌細胞に感染するコクサッキーウイルスA11型を含む、
小細胞肺癌、非小細胞肺癌、悪性中皮腫、大腸癌、食道癌、頭頸部癌、悪性リンパ腫、乳癌及び子宮頸癌からなる群から選択される癌を治療するための医薬組成物。 - 前記コクサッキーウイルスA11型は、
カプシドが除去されたことを特徴とする請求項1に記載の医薬組成物。 - フォスフォイノシトール3キナーゼ阻害剤と併用される、
ことを特徴とする請求項1又は2に記載の医薬組成物。 - MAPキナーゼキナーゼ阻害剤と併用される、
ことを特徴とする請求項1乃至3のいずれか一項に記載の医薬組成物。 - 抗癌剤と併用される、
ことを特徴とする請求項1乃至4のいずれか一項に記載の医薬組成物。 - CDDP抵抗性の癌の治療に使用される、
ことを特徴とする請求項1乃至5のいずれか一項に記載の医薬組成物。 - ゲフィチニブ抵抗性又はオキサリプラチン抵抗性の癌の治療に使用される、
ことを特徴とする請求項1乃至6のいずれか一項に記載の医薬組成物。 - 前記癌細胞は、癌幹細胞である、
ことを特徴とする請求項1乃至7のいずれか一項に記載の医薬組成物。 - 癌細胞に感染するコクサッキーウイルスA11型に由来する核酸を含む、
小細胞肺癌、非小細胞肺癌、悪性中皮腫、大腸癌、食道癌、頭頸部癌、悪性リンパ腫、乳癌及び子宮頸癌からなる群から選択される癌を治療するための医薬組成物。
