材料及び方法
試験ポリペプチドの症出及び産生
修飾オステオポンチンポリペプチド配列番号1は、XhoI及びBamHI制限酵素部位の使用によりpCEP4発現ベクターへクローニングした。pCEP4発現ベクターは、細菌に発現のためにアンピシリン耐性遺伝子を、そして真核EBNA細胞に発現のためにヒグロマイシン耐性遺伝子を含有した。ベクターを含有するポリペプチドをコンピテントXL−10細菌細胞へ形質転換し、増殖させ、そしてQiagen midi prep kitで分離した。分離ベクターは次いで、製造プロトコール (Invitrogen) に従ってFugeneトランスフェクション試薬の使用により、ヒトEBNA細胞へトランスフェクトした。
産生ポリペプチドを含有するEBNA細胞培養からの培地を培養の3〜4日後に回収した。pCEP4プラスミドによって産生したポリペプチドは、Ni−セファロースゲルクロマトグラフィー(Invitrogen)による分離を容易にするのに使用される、挿入His−タグを含有した。回収培地を結合緩衝液(20mMリン酸ナトリウム、500mM塩化ナトリウム、pH7.8)で希釈し、Ni−セファロース懸濁液を4℃で一夜シェーカーにおいてインキュベーション前に加えた。Ni−セファロースゲル懸濁液は1000gで7分間遠心分離してペレット化し、そしてBioRad 使い捨てミニカラムに注入した。非結合タンパク質は結合緩衝液で除き続いて緩衝液(20mMリン酸ナトリウム、500mM塩化ナトリウム、pH6.0)で洗浄した。ポリペプチドは500mMイミダゾールでカラムから溶出した。
研究は4実験群から成り、各々5匹の雄C57BL/6Nマウスを含んだ。

毛包密度に対する修飾オステオポンチンポリペプチド(配列番号1)による処置の効果は、下記の表1(a)及び(b)にまとめた。
データは、配列番号1の例示的修飾オステオポンチンポリペプチドが、約60%毛包数/mm表皮を増加させることを示す。
材料及び方法
動物
マウス(C57BL/6)は6〜8週齢で使用した。
・発毛の休止期段階における動物の選択。
・動物の背後部のクリッピング
・試験ペプチド/媒体の皮下注射
・視覚分析:百分率成長期誘導;平均発毛スコア;視覚メラニン形成:
・組織学的解析:皮下組織における卵胞数;皮膚厚み用の形態計測。
発毛に対する本発明の例示的ポリペプチドの効果を表5に示す。
試験した変異オステオポンチンポリペプチドはすべてインビボで顕著な発毛効果を示して、より短い例示的ポリペプチドFOL−004(配列番号5)及びFOL−005(配列番号63)は特に速い作用発現を示した。
VDVPNGRGDSLAYGLR [配列番号121]
・動物に1、5及び9日目に規定の処置を投与した
・処置は背部クリップ皮膚へ皮下注射によって投与した
・処置効果は実施例Bに記載のように毎日観察しそしてスコア化した。
発毛に対するポリペプチド「FOL−001」の効果は表7及び図6に示す。
本発明の対応の変異ペプチド配列(FOL−004;配列番号5)と比べて、同等の非変異野生型配列はFOL−004で認められたものの50%未満の最大効果で発毛促進の遅延発現を呈した。
材料及び方法
動物及び処置は実施例Bで上記に記載の通りであった。
すべての動物は16日目に犠牲にした。
実施例Bでの発毛評価期間の完了後、動物を安楽死させ、そして内面の観察のために背部皮膚を剥離しそして除いた。
剥離皮膚切片を実施例Aに記載のように組織学的解析のために調製した。皮下組織における卵胞数及び次いで皮膚厚みを測定した。
結果を下記の表8及び9にまとめた。
本発明の例示的変異オステオポンチンポリペプチドは、皮下組織における毛包数の増加及び/又は皮膚厚みの増強からも明らかなように、成長期の顕著な誘導を示した。
Claims (17)
- (a)配列番号1、5及び63から成る群から選択されるアミノ酸配列から成る修飾オステオポンチンポリペプチド;及び
(b)薬学的に許容される及び/又は化粧品的に許容される添加剤、担体又は希釈剤:を含んでなる哺乳動物の発毛を促進する組成物。 - 修飾オステオポンチンポリペプチドは、配列番号1のアミノ酸配列から成る、請求項1に記載の組成物。
- 修飾オステオポンチンポリペプチドは、配列番号5のアミノ酸配列から成る、請求項1に記載の組成物。
- 修飾オステオポンチンポリペプチドは、配列番号63のアミノ酸配列から成る、請求項1に記載の組成物。
- 局所投与のための請求項1〜4のいずれか1項に記載の組成物。
- 経皮投与のための請求項1〜4のいずれか1項に記載の組成物。
- 皮内投与のための請求項1〜4のいずれか1項に記載の組成物。
- 配列番号63のアミノ酸配列から成るポリペプチド。
- 配列番号5のアミノ酸配列から成るポリペプチド。
- 配列番号1のアミノ酸配列から成るポリペプチド。
- 脱毛に関連する疾患又は病態の処置又は予防に用いる、請求項8〜10のいずれか1項に記載のポリペプチド。
- 脱毛に関連する疾患又は病態の処置又は予防に用いる、請求項1〜7のいずれか1項に記載の組成物。
- 脱毛症の処置又は予防に用いる、請求項12に記載の組成物。
- 脱毛症が成長期毛又は休止期毛の喪失に関連する、請求項13に記載の組成物。
- 脱毛症が:
(a)男性ホルモン性脱毛症;
(b)牽引性脱毛症;
(c)アナゲン脱毛;
(d)テロゲン脱毛;
(e)円形脱毛症;
(f)完全脱毛症;
(g)全身性脱毛症;
(h)脱毛性毛瘡;
(i)ムチン性脱毛症;
(j)腫瘍性脱毛症;
(k)瘢痕性脱毛症(cicatricial alopecia);及び
(l)瘢痕性脱毛症(scarring alopecia);
から成る群から選択される、請求項12〜14のいずれか1項に記載の組成物。 - 哺乳動物の発毛促進のための薬剤の調製における請求項8〜10のいずれか1項に記載の修飾オステオポンチンポリペプチドの使用。
- 薬剤は、脱毛症の処置又は予防のためである、請求項16に記載の使用。
