Description
(a)プラスミド、ベクター、又は天然/合成/突然変異ウイルス、様々は種類の改変オリゴヌクレオチド(例、PTO、LNA、2’F−ANA、タンパク質−ヌクレオチド複合体、RNAi、siRNAもしくはmicromiRNA、メチルメトキシ−、ホスホロアミデート、PNA、モルホリノ、ホスホルアミデート、シクロヘキセン(CeNA)、ギャップマー、リボザイム、アプタマー、CpG−オリゴ、DNA−ザイム、リボスイッチ、又は脂質もしくは脂質含有分子)、
(b)全種類のリンカーを含む、ペプチド類、ペプチド複合体、
(c)小分子、
(d)抗体及びそれらの誘導体、特にキメラ、Fab−フラグメント、Fc−フラグメント、又は
(e)担体、リポソーム、ナノ粒子、複合体、又は上記に挙げた構築物を含有する任意の他の送達系、
(f)酸化剤もしくはスルフヒドリル(SH基)修飾剤が含まれる。
(a)候補化合物をADPGKもしくはGDP2を含む試験系又はADPGKもしくはGPD2をコードする遺伝子とともにインキュベートする工程、及び
(b)ADPGKもしくはGPD2の生物活性をアッセイする工程を含み、
このとき好ましくは前記試験化合物の非存在下での試験系と比較したADPGKもしくはGPD2の生物活性の増加もしくは減少は、該所望の特性を有する候補化合物の存在の指標である方法に関する。
(a)前記患者からサンプルを得る工程、及び
(b)ADPGKもしくはGPD2のレベル及び/又は活性を決定する工程を含み、それにより該治療様式がADPGKもしくはGPD2の該レベル及び/又は活性に左右される方法に関する。
(実施例1)
材料及び方法
(A)化学薬品
異なることを記載しない場合、使用した全ての試薬及び酵素は、Sigma-Aldrich社(独国ミュンヘン)によって供給されたものである。ジクロロジヒドロフルオレセインジアセテート(H2DCF−DA)は、Invitrogen社(米国カリフォルニア州カールズバッド)から入手した。Ionoは、Merck社(独国ダルムシュタット)から購入した。ウエスタンブロット(WB)のための一次抗体は、Sigma-Aldrich社(独国ミュンヘン)から−マウスモノクローナル抗FLAG(M2)、ウサギポリクローナル抗FLAG、マウスモノクローナル抗γチューブリン、ウサギポリクローナル抗GPD2(Human Protein Atlas Antibodies)、Cell Signaling社(米国マサチューセッツ州デンバース)から−ウサギポリクローナル抗HK1及びウサギポリクローナル抗HK2、Abcam社(英国ケンブリッジ)から−マウスモノクローナル抗ヒトADPGK(1E4)、Santa Cruz Biotechnology社(米国カリフォルニア州サンタクルーズ)から−ヤギポリクローナル抗SOD1、Milipore社(独国ダルムシュタット)から−ウサギポリクローナル抗SOD2、ABR社(米国コロラド州ゴールデン)から−ウサギポリクローナル抗カルレチキュリン、GeneTex社(米国カリフォルニア州アーヴィン)から−ウサギポリクローナル抗β−アクチン、及びThermo Scientific社(独国ボン)から−マウスモノクローナル抗PHB1抗体であった(II−14−10)。ミトコンドリア呼吸複合体の含量は、Mito Sciences社(米国オレゴン州ユージーン)からのTotal OXPHOS WB抗体カクテルを使用して分析した。FITCコンジュゲート抗−CD3及び抗−CD20抗体は、Becton Dickinson社(独国ハイデルベルク)から購入した。架橋結合ポリクローナルヤギ抗マウス抗体(GAM)は、Southern Biotech社(米国アラバマ州バーミンガム)から入手した。ヒトCD3に対するモノクローナルマウス抗体(OKT3)は、記載されたとおりに調製した(1)。
一般に、ヒト末梢血Tリンパ球は、以前に記載されたとおりに精製した(1)。ヒト末梢血Bリンパ球は、ネガティブMACSソーティング(「B細胞単離キットII」、Miltenyi社、独国ベルギッシュグラートバハ)によってフィコール勾配相間環(Ficoll gradient inter-phase ring)から調製した。調製したT及びB細胞の均質性は、FITC結合抗−CD3もしくは抗−CD20抗体を用いた染色、及びその後のFACS分析によって検証し、>93%(B細胞)及び>90%(T細胞)であると推定された。
試験に参加する前に全対象体からインフォームドコンセントを入手した。患者10例中9例は、本療法の前に治療を受けていなかった。患者データ(年齢、性別及びCLLサンプルの遺伝分析)は、表1に見いだすことができる。CLL患者からのB細胞は、Ficoll-Paque遠心分離(GE Healthcare社、英国チャルフォントセントジャイルズ)によって単離した。純度は>90%と推定された。本試験は、ドイツ癌研究センター(DKFZ、ハイデルベルク)の倫理指針及びヘルシンキ宣言に従って実施し、ハイデルベルクのルプレヒト・カールス大学(独国)の治験審査委員会によって承認された。
T細胞系のジャーカットJ16−145細胞、J16サブクローン(2)は、RPMI 1640(+L−グルタミン)、10%ウシ胎仔血清(FCS)中で培養した。GaussiaルシフェラーゼNF−κBレポーターJ16−145細胞は、IMDM、10%のFCS中で培養した。新鮮単離(「静止」)もしくはフィトヘムアグルチニン(PHA)活性化及び6〜8日間イン ヴィトロ拡張(「予め活性化された」)末梢ヒトT細胞は、RPMI 1640(+L−グルタミン)、10%のFCS中において2×106細胞/mlの初期濃度で培養した。予め活性化された誘導性拡張のために、「静止」T細胞は1μg/mlのPHAを用いて16時間にわたり処理し、洗浄し、引き続いて25U/mlのIL−2の存在下で6〜8日間にわたり培養した。これらの細胞は、本文書では、異なることが明示されていない場合は「T細胞」と呼ぶ。
細胞は、H2DCF−DA(5μM)を用いて30分間にわたり染色した。次に、細胞を分割し、プレート結合CD3抗体(30μg/ml)もしくはPMA(10ng/ml)を用いて1時間刺激した。処理は氷温PBSによって停止させ、ROS生成はFACS分析によって決定した(Canto II、Becton Dickinson社)。ROS生成は、平均蛍光強度(MFI)の増加として定量し、以下の式、MFI(%)の増加=[(MFI刺激−MFI未刺激)/MFI未刺激]×100(3)にしたがって計算した。
イン ヴィトロ拡張T細胞(少なくとも0.5×108)は液体窒素中で瞬間冷凍し、引き続いてETC緩衝液(20mMのTris−HCl(pH7.4)、250mMのスクロース、50mMのKC1、5mMのMgCl2)中の氷上で、わずかな改変を施したChretien他(4)に従って、30分間にわたり0.015%ジギトニンとともにインキュベートした。その後、細胞は氷温ETC緩衝液を用いて2回洗浄し、アッセイにかけた。
細胞は、27×1/2’’ニードルを使用して氷温ETC緩衝液中で破壊させ、ホモジネートを600×g、4℃で10分間にわたり遠心した。良好なサイトゾル(上清)分画としての「高g」ミトコンドリア(ペレット)を調製するために、600×gの上清を11,000×g、4℃で20分間にわたり遠心した。「ミトコンドリア濃縮」分画を調製するために、600×gの上清を10分間にわたり4℃、3,500×gで遠心した(ペレット)。引き続いて、「ER濃縮分画」を調製するために、3,500×gの上清を11,000×g、4℃で20分間遠心した。マウス心筋ミトコンドリアは、記載されたように(5)、「ミトコンドリア濃縮」分画の手順にしたがって調製した。
ETC酵素複合体の定常状態活性は、記載したように(5、6)、二波長モードで作動するコンピュータ調節可能な分光光度計(Spectramax Plus Microplate Reader、Molecular Devices社、米国カリフォルニア州サニーベール)を使用して決定した。サンプルは最終容量300μLの温度調節96ウエルプレート内で分析した。標準呼吸鎖阻害剤を添加して、酵素アッセイの特異性を確証した。全酵素活性は、タンパク質濃度に正規化した。刺激T細胞については、ETC酵素活性を瞬間冷凍及びジギトニン透過化(以前に記載したように)後に測定した。複合体Iもしくは複合体IIから複合体IIIへの電子流を試験するために、ジギトニン透過性細胞もしくはミトコンドリア分画をNADHもしくはコハク酸とともにインキュベートし、NaCNの存在下でのシトクロムcの還元を測定した。
適用した全酵素は、Sigma-Aldrich社から購入し、ウサギ筋肉から調製した。
−ヘキソキナーゼ(HK)活性は、1mMのATP、1mMのグルコース、0.5mMのNADP、0.05U/mlのグルコース6−リン酸デヒドロゲナーゼを含有するETC緩衝液内でのNADP還元としてアッセイした。
−グリセルアルデヒド−リン酸デヒドロゲナーゼ(GAPDH)活性は、10mMのKPi、1mMのNAD、3.3mMのシステイン、0.2mMのEDTA、0.2mMのMgCl2、1mMのADPを含有する緩衝液中でのNAD還元として検出された。
−グリセロール−3−リン酸デヒドロゲナーゼ(GPD2)活性は、5mMのグリセロール3−リン酸、2mMのNaCN、及び60μMのシトクロムcを含有するETC緩衝液中でのシトクロムc還元として測定した。
−トリオソ−リン酸イソメラーゼ(TPI)活性は、0.2mMのNADH、4.9mMのDL−グリセルアルデヒド3−リン酸、0.4U/mlのα−グリセロリン酸デヒドロゲナーゼ(GPD1)を含有するETC緩衝液中でのNADH酸化としてアッセイした。
−ホスホフルクトキナーゼ(PFK)活性は、4mMのフルクトース6−リン酸、2mMのATP、0.5mMのNADH、0.2U/mlのアルドラーゼ、0.8U/mlのTPI、0.1U/mlのGPD1を含有するETC緩衝液中でのNADH酸化として検出した。
−ピルビン酸キナーゼ(PK)活性は、1mMのホスホエノールピルビン酸、1mMのADP、0.5mMのNADH、2U/mlのLDHを含有するETC緩衝液中でのNADH酸化として検出した。
−乳酸デヒドロゲナーゼ(LDH)活性は、1mMのピルビン酸、0.5mMのNADHを含有するETC緩衝液中でのNADH酸化として記録した。
−エノラーゼ(ENO)正反応は、1mMの2−ホスホグリセリン酸、1mMのADP、0.5mMのNADH、2U/mlのLDH、2U/mlのPKを含有するETC緩衝液中でのNADH酸化として測定した。逆反応は、1mMのホスホエノールピルビン酸、1mMのATP、0.5mMのNADH、2U/mlのLDH、2U/mlのPKを含有するETC緩衝液中でアッセイした。
−ホスホグリセリン酸ムターゼ(PGM)活性は、1mMの2−ホスホグリセリン酸、1mMのATP、1mMのホスホエノールピルビン酸、0.5mMのNADH、2U/mlのLDH、2U/mlのPKを含有するETC緩衝液中でのNADH酸化として記録した。逆反応は、1mMの3−ホスホグリセリン酸、1mMのADP、0.5mMのNADH、2U/mlのLDH、2U/mlのPKを含有するETC緩衝液中でアッセイした。
−ホスホグリセリン酸キナーゼ(PGK)逆反応は、1mMの3−ホスホグリセリン酸、1mMのATP、0.5mMのNADH、2U/mlのLDH、2U/mlのPKを含有するETC緩衝液中でアッセイした。
GPD経路を介してのフルクトース−6−リン酸流動は、1mMのフルクトース6−リン酸、1mMのATP、0.5mMのNADH、0.1U/mlのGPD1を含有するETC緩衝液中でのNADH酸化として測定した。GAPDHブランチを通しての流動は、1mMのKPi、1mMのフルクトース6−リン酸、1mMのATP、0.5mMのNAD、0.1U/mlのGAPDHを含有するETC緩衝液中で記録した。
処理後、細胞(1〜2×107細胞)を洗浄し、クレブス−リンガー緩衝液中に懸濁させた。引き続いて、細胞は、1μCi/mlのD−[3−3H]グルコースとともに37℃で10分間インキュベートした。その後、細胞は氷温クレブス−リンガー緩衝液を用いて2回洗浄し、細胞内D−[3−3H]グルコースはシンチレーションカウンタ(LS 6500液体シンチレーションカウンタ、Beckman Coulter社、米国カリフォルニア州ブレア)を使用して検出した。
ミトコンドリア呼吸速度は、以前に記載されたプロトコール(7)にしたがってコンピュータ支援高分解能Oroboros 1オキシグラフシステム(Paar社、オーストリア国グラーツ)を使用して測定した。イン ヴィトロ拡張T細胞(5〜10×107細胞)を各電極チャンバ内に配置し、可溶性抗−CD3抗体(10μg/m、GAM(2μg/ml)と架橋結合)もしくはPMA(10ng/ml)を用いて活性化した。実験構成は、以前に記載されたとおりであった(8)。「ミトコンドリア呼吸速度」は、「バックグラウンド酸素消費速度」(5μMのオリゴマイシンの存在下で参照電極を用いて記録した)を「総酸素消費速度」から減じることによって計算した((8)に記載したとおり)。
少なくとも2×107個の拡張T細胞を新鮮RPMI 1640培地(+補給物及びIL−2)中に懸濁させ、次にGAM(2μg/ml)と架橋結合させた可溶性抗−CD3抗体(10μg/ml)を用いて1時間にわたり活性化させた。培地中の乳酸濃度は、Olympus AU400システム(Olympus社、日本国東京)を用いて検出し、各細胞溶解液のタンパク質濃度に標準化した。
ADPGK活性は、pH6.0及び37℃もしくは42℃で1mMのADP、1mMのグルコース、0.5mMのNADP、5μMのジアデノシンペンタリン酸、及び0.05U/mlのグルコース6−リン酸デヒドロゲナーゼを含有するETC緩衝液中のNADPの還元としてアッセイした。アッセイは、本質的に組み換えマウスADPGKタンパク質について以前に記載された方法に基づいていた(9)。
AK活性は、pH7.5及び37℃で1mMのADP、1mMのグルコース、0.5mMのNADP、1U/mlのHK及び0.05U/mlのグルコース6−リン酸デヒドロゲナーゼを含有するETC緩衝液中のNADPの還元としてアッセイした。
ATP濃度は、製造業者の取扱説明書にしたがって「CellTiter Glo」アッセイ(Promega社)及び96ウエル照度計(Orion L、Berthold社、独国バード・ヴィルトバード)を使用して決定した。結果は、タンパク質濃度に正規化した。
少なくとも5×107イン ヴィトロ拡張ヒトT細胞は、1時間にわたりプレート結合抗−CD3抗体(30μg/ml)もしくはPMA(10ng/ml)を用いて処理した。細胞を洗浄し、ペレット化し、液体窒素中で瞬間冷凍し、ドライアイス上に静置した。ユビキノール/ユビキノン抽出は、氷温200μLヘキサン:イソプロパノール(2:1)の添加及びその後の0℃での5分間にわたる遠心(15,000g)によって実施した。次に、150μLの上清を直ちに注入し、RP−HPLCによって分離した。HPLCの分離及び同定条件は、以前に公表された方法に基づいていた(10、11)。適用したクロマトグラフィーシステム、プロポーショニングバルブを備えるLachrom LC−7100低圧勾配ポンプ、L-7350/L-7351 Peltier冷却モジュールを備えるL-7350カラムサーモスタット、L−7450A UV/VIS−DAD−ダイオードアレイ検出器(Merck−Hitachi社、独国ダルムシュタット)、パルス式アンペロメトリック検出器(PAD)817 Bioscan(Metrohm社、スイス国ヘリザウ)、カラムLiChrospher RP18e 125×4mm(Merck社、独国ダルムシュタット)、4−チャネル脱ガス装置(Knauer社、独国ベルリン)、メタノール:ヘキサン:イソプロパノール:酢酸(83:27.5:1.5:1.5 v/v+酢酸ナトリウム4.2g/1;pH=6)を用いた均一濃度溶出、流量−1ml/分、温度25℃。同定は、UV/VIS−DAD吸収スペクトルに基づき、電圧アンペロメトリック検出器を用いて検出された酸化還元反応によって促進された。定量は、波長275nm(ユビキノン)及び289nm(ユビキノール)でのUV−VISクロマトグラムに基づいて実施した。UV−VIS/DAD分析の選択された定性的及び定量的結果は、HPLC−MS/ESI(Agilent社、米国カリフォルニア州サンタクララ)によって確証した。
GaussiaルシフェラーゼをベースとするNF−κBレポーターを用いて安定的にトランスフェクトしたJ16−145ジャーカットT細胞は、Marcus Brechman博士及びRudiger Arnold博士の厚意により提供された。手短に言えば、培地を交換した後に、細胞は、各実験条件に対して3回ずつ6時間にわたりIono(10μM)及び/又はPMA(10ng/ml)を用いて誘導した。20μLの収集培地は、「Gaussia-Juice」(P.J.K.社、独国クラインブリッタースドルフ)及び96ウエルプレートルミノメータ(Orion L、Berthold社、独国バード・ヴィルバード)を使用して発光の読み取りに適用した。発光シグナルは、トリパンブルー除去法によって評価した生細胞の数に正規化した。
少なくとも8×107個のF−ADPGKもしくはEVジャーカットT細胞は、150mMのNaCl、5mMのEDTA、10mMのTris−HCl、0.5%のTriton X−100を含有する緩衝液中において45分間にわたり4℃で溶解させた。溶解液は11,000g、4℃で10分間にわたり遠心した。上清は、抗FLAG M2抗体(ANTI-FLAG(登録商標)M2親和性ゲル、Sigma社)と共有的に架橋結合させたアガロースビーズ上に適用した。洗浄工程及びFLAGペプチドを用いた溶出は、製造業者のプロトコールにしたがって実施した。溶出液を直接的にADPGK活性アッセイにかけた、又は冷凍してWBにより分析した(3)。
WT−ADPGKの一過性発現のために、pCMV6−ACプラスミド(Origene社から提供、米国メリーランド州ロックビル)又は空コントロールpCMV6−ACベクター(Origene社)内にクローニングしたヒトADPGK転写産バリアント1(UniProtKB正準配列Q9BRR6−1)のcDNA配列を使用した。J16−145細胞もしくはJ16−145 NF−κB Gaussiaルシフェラーゼレポータークローンは、製造業者の取扱説明書にしたがってAMAXAヌクレオフェクション(nucleofection)テクノロジー(「Cell Line Nucelofector(登録商標)Vキット」、Lonza社、独国ケルン)を使用して2μgのプラスミドDNA/トランスフェクションを用いてトランスフェクトした。トランスフェクション24時間後、細胞に該実験方法を適用した。FLAG−ADPGKタンパク質を安定性で発現するJ16−145細胞(F−ADPGKジャーカット細胞)のプール又は空ベクター(コントロール細胞)は、Addgene社(米国マサチューセッツ州ケンブリッジ)によって供給されたレトロウイルス発現ベクターを使用して生成した。各々、pWZL−Neo−Myr−Flag−ADPGK(Addgene社製プラスミド20417、タンパク質骨格上のN末端FLAG−タグ、UniProtKB Q9BRR6−1)もしくはpWZL−Neo−Myr−Flag−DEST(Addgene社製プラスミド15300)(12)。レトロウイルスベクターは、リン酸カルシウム法によってPhenix−Ampfo産生細胞(Allele Biotechnology社;米国カリフォルニア州サンディエゴ)内へトランスフェクトした。ウイルス形質導入は、製造業者のプロトコールにしたがってポリブレン及びスピン感染法(spin-infection)を使用して実施した(2時間/2,000g)。細胞は、選択圧下で、RPMI 1640(+L−グルタミン)、1mg/mlのG418硫酸塩(Roth社、独国カールスルーエ)が補給された10%のFCS中で培養した。
RNAは、TRIzol試薬(Invitrogen社)(CLL細胞及びコントロールB細胞)又は「Rneasy Mini」キット(QIAGEN社、独国ヒルデン)を製造業者の取扱説明書にしたがって用いて単離した。全RNA(1μg)は、「逆転写(RT)−PCRキット」(Applied Biosystems社、米国カリフォルニア州カールスバッド)を用いて逆転写させた。定量的リアルタイムPCRは、「Power SYBRグリーンPCRマスターミックス」(Applied Biosystems社)を使用して実施した。遺伝子発現は、7500リアルタイムPCRシステム及び配列検出ソフトウエア(Applied Biosystems社製、v.2.0.2)を使用して分析した。遺伝子発現レベルは、内因性標準物質としてβアクチン発現を使用して正規化した。誘導率(X)は、式X=2-ΔΔCt、ここで、Ctはサイクル閾値を表し、及びΔCt=Ct対象遺伝子−Ct参照遺伝子を使用して計算した。ΔΔCtは、「誘導」サンプルのΔCt値と対応する「非誘導」サンプルのΔCtとの差である。平均誘導率を計算した。CLL患者及び健常ドナー由来のB細胞サンプル中の相対ADPGK基礎発現レベルは、係数Y=2(Ct対象遺伝子−Ct GAPDH)×1,000を使用して比較した(1)。実験間変動を補正するために、2つの別個の実験(3例ずつの5CLLサンプル及び各4もしくは5例の健常ドナーサンプルを含む2枚の別個の96ウエルプレート)から得た値にz-トランスフォーメーションに供した。以下のプライマーを遺伝子発現分析に使用した。βアクチン、センス5−ACCGTGAGAAGATGACCCAGA−3’、アンチセンス5’−TCACCGGAGTCCATCACGAT−3’。IL−2、センス5’−CAACTGGAGCATTTACTGCTG−3’、アンチセンス5’−TCAGTTCTGTGGCCTTCTTGG−3’。IL−8、センス5’−GAATGGGTTTGCTAGAATGTGATA−3’、アンチセンス5’−CAGACTAGGGTTGCCAGATTTAAC−3’。ADPGK、センス5’−TCATTGCAGGAAGTGGATGA−3’、アンチセンス5’−GCATGGGGAGCTTTTAACTG−3’。IκBα、センス5’−GTCAAGGAGCTGCAGGAGAT−3’、アンチセンス5’−GATGGCCAAGTGCAGGAA−3’。GPD2、センス5’−ACCCTGGCTGGAGGAACT−3’、アンチセンス、5’−CCCTTTCACTGCCTTTTGAA3’及び、いずれかの場所(1)で記載されたIL−4プライマー。
トランスフェクションのために使用したsiRNAオリゴヌクレオチドは次の通りであった。ノンサイレンシングコントロール(未標識の「AllStars」ノンサイレンシング、確証されたsiRNA、QIAGEN社)、ヒトADPGK(Hs_ADPGK_5−8 FlexiTube siRNA、QIAGEN社)、若しくはヒトGPD2(Hs_GPD2_1、4−6 FlexiTube siRNA、QIAGEN社)に対して特異的。ジャーカットT細胞もしくは末梢「静止」ヒトT細胞は、製造業者の取扱説明書にしたがって800nMのsiRNAオリゴヌクレオチドを用いて実施したヌクレオフェクション(「Cell Line Nucelofector(登録商標)Vキット」もしくは「T Cell Nulcleofector(登録商標)キット、Lonza社)によってトランスフェクトした。
ミトコンドリア濃縮分画のBN−PAGE分析は、改変を施したHaeger他(13)にしたがって実施した。ミトコンドリア膜のタンパク質複合体は、1.5Mの6−アミノカプロン酸、50mMのBis−Tris及びジギトニン(6g/g(タンパク質))の存在下、4℃で30分間可溶化した。引き続いて、サンプルを11,000×g及び4℃で30分間遠心した。この上清にサンプル緩衝液(1.5Mの6−アミノカプロン酸、50%グリセロール、1%(w/v)クーマシーブルーG250)を補給し、直ちにBNゲル(3〜13%)上に塗布した。ゲルは16時間にわたり35Vの一定電流でランさせた。タンパク質は2時間にわたり、0.8mA/cm2(膜)へ限定した電流を用いてブロッティングした。ウシ心臓精製複合体III(BC1複合体)は、Ulrich Brand教授の厚意により寄贈された。
TCR誘因は予め活性化されたT細胞においてワールブルク(Warburg)効果様代謝変化を誘導する
ミトコンドリア呼吸の減少及び解糖の増加がワールブルク効果の中心を成す(11〜14)。酸素電極を適用して、無傷ヒトT細胞のミトコンドリア呼吸における活性化誘導性変化を測定した。ワールブルク表現型と一致して、作動性抗−CD3抗体もしくはPMAを用いた処理はミトコンドリア酸素消費の有意な阻害を生じさせたが(図1A及び図7A)、他方、放射標識グルコースの細胞取り込みは、刺激後に急速に上昇した(図1B)。細胞内ATPレベルにおける連続的上昇(図1C)は、酸素正常状態にもかかわらずイン ヴィトロ拡張ヒト末梢T細胞のTCR誘因はいっそうより解糖性の表現型に向かう急速な変化を引き起こすことを強調した。
酸化シグナルの生成には、ミトコンドリア内の主要なバイオエネルギー及び超微細構造的変化が付随する
TCR誘因性PKCθは、ミトコンドリア呼吸複合体Iによる酸化シグナル生成を誘導する(3)。活性化誘導性ワールブルク代謝変化とミトコンドリアROS遊離の機序との関係についてのより明確な洞察を得るために、呼吸鎖複合体のバイオエネルギー状態について調査した。このために、瞬間冷凍及びジギトニン透過化イン ヴィトロ拡張ヒトT細胞を適用した。図1Gに示したように(上方パネル)、TCR誘因は複合体I及びIIの酵素活性の有意な減少を生じさせたが、複合体IIIの活性は増加した。PMAを用いて処理した細胞において同様のパターンを観察することができた(図1G、下方パネル)。データは、これらの現象がTCR誘因ミトコンドリアCa2+取り込みから独立していることを証明している。さらにこれらのデータは、DAG/PKCθ依存性経路が関係していることを示唆している。この前提と一致して、PKC及びTCR誘導性ROS生成の阻害剤であるビス−インドリル−マレイミデートI(BIM)を用いたT細胞の前処理は、観察された変化を有意に遮断した(図1H及びI)。
偏向された解糖流はGPD2媒介性ミトコンドリアROS遊離をもたらす
過剰還元ユビキノンによって誘導されたミトコンドリアROS産生は、高血糖性もしくは低酸素性細胞における糖代謝の増加と関連することは公知である(18〜21)。TCR誘因直後の解糖代謝流をアッセイするために、全主要解糖酵素の活性を測定した(図3A及びB)。興味深いことに、TCR後に有意に変化した唯一の酵素活性は、エノラーゼ(ENO)の逆反応であった(図3B)。使用したアッセイはNADHの存在下でのピルビン酸の還元を監視することによるホスホエノールピルビン酸(PEP)特異的ATP加水分解しか検出しないので、阻害剤を使用して検出した酵素活性をさらに特徴付けた。ENO阻害剤であるNaFは、検出された逆ENO活性を遮断した。興味深いことに、ヒスチジンリン酸化の阻害剤であるNaVO3(28)は、該活性を減少させたが、遮断はしなかった。第一に、それは、その誘導性増加を無効にした(データは示していない)。注目すべきことに、PEP依存性ヒスチジンリン酸化によって媒介されたホスホグリセリン酸ムターゼ(PGM)活性化は、近年、ピルビン酸キナーゼ(PK)アイソフォームM2(PK−M2)を発現する急速に増殖する正常細胞及び癌細胞において記載されてきた(29、30)。PK−M2は活性化T細胞内で高度に発現するので(31)、逆ENO様活性において検出された上昇は、最近記載された未知のPEP利用ヒスチジンキナーゼによって媒介されるのであろう(29)。これはさらに、観察されたENO様活性が部分的にのみATP依存性である、及びその活性化誘導性増加がATP非依存性であるという所見によって支持されている。
T細胞活性化誘導性ADPGK酵素活性は解糖流動をアップレギュレートする
ユビキノン及びミトコンドリアROS遊離のグルコース誘導性過剰還元は、アップレギュレートされた解糖流動に依存すると考えられる(18、19)。解糖は、HK、PFK、GAPDH、及びPKに主要調節点を備える高度に調節された一連の反応である(図3A)。本実験の結果は、これらの酵素活性のいずれについても変化を示さなかった(図3B)。結果としてHK生成物阻害の消失を生じさせるHKとミトコンドリアVDACとの関連は、グルコース流動をアップレギュレートするまた別の機序として作用する(37)。拡張ヒトT細胞のミトコンドリア濃縮膜分画のWB分析は、高いHK1及び低いHK2濃度を明らかにした(図10A)。それでもなお、TCR誘因は、ミトコンドリアとのHK1/2関連の有意な増強、又はミトコンドリア関連性HK活性のアップレギュレーションを生じさせなかった(図10A及びB)。次に、呼吸媒介性ATP生成と共役したミトコンドリア関連性HK活性について調査した(図4A)。TCR誘因は、呼吸基質、ADP及びコハク酸の存在下でHK様活性を明確に誘導した(図4B)。この活性は、アデニル酸キナーゼ(AK)阻害剤であるジアデノシン五リン酸(Ap5A、5μM、40%までの阻害)によって軽度にのみ遮断されたが、他方Ap5はミトコンドリアAKを強力に阻害した(5μM、80〜90%の阻害)。極めて重要であることに、HK様活性における活性化誘導性増加はAp5Aによる影響を受けなかった。驚くべきことに、この強化された酵素活性はコハク酸の添加には非依存性であったが、ADPの存在には明白に依存性であった。さらに、呼吸鎖阻害剤NaCN及び脱共役CCCPに対する非感受性は、ミトコンドリアによって生成されたATPからの実際的独立性を証明した(図4B)。これらの結果に興味をそそられて、文献を検索し、グルコースをリン酸化できるタンパク質を、ADP‐ADP依存性グルコキナーゼ(ADPGK)を利用することによって同定することができた(38)。だが好熱性古細菌(thermophilic archaea)にとって典型的であるように、ADPGKは哺乳動物内に存在し、T細胞を含むヒト造血性細胞内で高度に発現する(図5E)(31、39)。さらに、グルコース−6−リン酸による阻害の報告された欠如は、ADPGKを解糖流動の特に優れた推定活性化因子にさせる(38)。
ADPGKは酸化シグナルの生成を媒介する
次に、ADPGK発現を低下させることが活性化誘導性ROS生成に及ぼす影響についてアッセイした。ADPGKの減少した発現(図5A)は、ジャーカットT細胞内でのROSのPMA誘因産生を阻害した(図5B)。同時に、ADPGKのノックダウンはPMAもしくはPMA/iono誘因性NF−κB活性化(図5C)及びIL−2、IκBα及びIL−8遺伝子のNF−κB依存性発現の阻害を生じさせた(図5D及び図12)。「静止」非拡張T細胞もミトコンドリアからの酸化シグナルを生成する(1)。同様に「静止」T細胞の活性化はGPD2及びADPGK酵素活性における上昇を引き起こした(図5F)。酵素活性の低誘導は、イン ヴィトロ拡張「予め活性化された」T細胞と比較して「静止」T細胞におけるROS生成の低い程度に対応する(1)。それでも、「静止」末梢ヒトT細胞内のADPGK発現のsiRNA媒介性ノックダウンも活性化誘導性酸化シグナルの低下を生じさせた(図5G)。
ADPGK活性はより高い炎症促進性温度で上昇する
ADPGK活性については、主として好熱性古細菌(thermophilic archaea)に関して記載された(38、40)。注目すべきことに、哺乳動物ADPGKについての二次構造予測は、低い配列相同性にもかかわらず熱安定性古細菌ADPGKの公知の二次構造との高度の構造的類似性を明らかにしている(図14)(40、41)。このため、ADPGK活性の温度依存性をヒトT細胞内で試験した。図6Dに示されたように、ADPGK触媒反応の速度は生理的温度範囲内で指数関数的に上昇し、37℃よりも42℃における方が3.5倍高かった。これとは対照的に、HK活性における温度駆動上昇は線形のままであった。さらに、42℃では、ADPGK媒介性反応はHKの速度に匹敵する速度に到達する。これらのデータは、ヒトADPGKの熱安定性を示唆しており、炎症反応及びNF−κB誘因において果たす可能性のある役割を強化する。そこで、炎症部位での上昇した温度はT細胞活性化及び炎症促進性サイトカインのNF−κB依存性発現を促進すると思われる。
ADPGK及びGPD2は悪性慢性リンパ球性白血病(CLL)細胞内で高度に発現する
多数の癌は、NF−κB経路の構成的活性化に厳密に依存する(16)。ROSもしくはワールブルク表現型の高内因性レベルは多数の悪性新生物の特徴である(11、14、42)。このため、癌細胞内のADPGK発現について調査した。T細胞を起源とする癌は、B細胞悪性新生物に比較してヒトでは希である。特に、CLLは、致死的予後を備える最も一般的な白血病の1つを構成する(43)。さらに、CLL細胞には、強化NF−κB活性、解糖及び高内因性ROSレベルが付与されている(15、17、43〜45)。そこで、悪性CLL細胞及び正常B細胞内のADPGK及びGPD2発現レベルを比較した(表1)。
T細胞活性化には、好気性解糖に向かうミトコンドリア呼吸からの代謝変化が結び付いている(12)。このいわゆる「ワールブルク表現型」は、急速に増殖する正常細胞に特徴的な特徴であるが、悪性細胞にとっても特徴的である(11、14)。以前の研究は、増殖するT細胞が高TCR誘因ミトコンドリアROS産生によって特徴付けられることを証明している(1、3)。このミトコンドリア生成酸化シグナルはNF−κB活性化に寄与するので、増殖及び炎症反応を刺激する。興味深いことに、癌細胞は好気性解糖表現型を提示することが多く、高内因性ROS産生及びNF−κB経路の構成的活性化を伴っている(14、16、18、42)。さらに、高血糖性もしくは低酸素性条件下での解糖のアップレギュレーションは、ミトコンドリアROS生成を増加させることが公知であるが(18、19、21)、他方、T細胞活性化はグルコース取り込みに依存する(9、10)。そこで、T細胞活性化誘導性ミトコンドリアROS産生の代謝分析は、シグナリングレベルでのこれらの様々な現象を結び付ける分子リンクを提供できる。

- (a)新生物を治療するための医薬組成物であって、前記医薬組成物は、
ADP依存性グルコキナーゼ(ADPGK)及び/又はグリセロール−3−リン酸デヒドロゲナーゼ(GPD2)の生物活性、又は、(b)前記ADPGK又はGPD2をコードする遺伝子の発現、を調節できる化合物を含み、
前記化合物は、
前記ADPGK又はGPD2に対するアンチセンスヌクレオチド、
前記ADPGK又はGPD2をコードする前記遺伝子の発現を減少又は阻害するsiRNA、
前記ADPGK又はGPD2に対して向けられたアンタゴニスト抗体又は同一特異性を有するそのフラグメント、
活性型の突然変異誘因によって生成される前記ADPGK又はGPD2の不活性バージョン、又は、
前記ADPGK又はGPD2の前記不活性バージョンをコードするポリ核酸、であり、
そして、
治療対象の前記新生物は、強化NF−κBレベルを示すB細胞慢性リンパ球性白血病(CLL)又は腫瘍である、医薬組成物。 - 強化NF−κBレベルを示すB細胞慢性リンパ球性白血病(CLL)又は腫瘍を治療できる化合物を同定するための方法であって、
候補化合物をADPGK若しくはGPD2、又はADPGK若しくはGPD2をコードする遺伝子を含む試験系とともにインキュベートする工程、及び、

このときADPGK又はGPD2の前記生物活性の減少は、ADPGK若しくはGPD2の生物活性、又はADPGK若しくはGPD2をコードする遺伝子の発現を調節する所望の特性を有し、強化NF−κBレベルを示すB細胞慢性リンパ球性白血病(CLL)又は腫瘍を治療できる候補化合物の存在の指標であり、
前記ADPGK又はGPD2の前記生物活性の減少は、前記試験化合物の非存在を特徴とする試験系との比較によって決定される、方法。
