一態様において、本発明は、活性成分を含んだポリマーナノ粒子を提供する。図1は、典型的なナノ粒子−活性成分組成物を説明する。ポリマーナノ粒子活性成分複合材料は、成分単独では見られない改善した物理的及び化学的特徴を有し得る。例えば、ポリマーナノ粒子は、活性成分の有効性を変化させることなく、活性成分の水溶性を改善することができる。いくつかの実施形態において、ポリマーナノ粒子は、活性成分単独と比較して、又は、典型的な活性成分配合物中と比較して、活性成分の土壌移動度を増加させるか若しくは低下させることができる。いくつかの実施形態において、ポリマーナノ粒子は、土壌の標的領域への土壌移動度を制御するために使用することができる。いくつかの活性成分は、示された使用には概して効果的であるが、低い水溶性を原因とする使用効率の悪さ、葉での広がり(又は葉表面の湿潤性)の効率の悪さ、クチクラ浸透性の効率の悪さ、又は、植物全体に亘る総じて不十分な転流に悩まされている。これは、配合における追加の化合物の使用と、より高濃度の活性成分を必要とする。活性成分配合物は典型的には、これらの問題を解決するために界面活性剤(例えば、アミンエトキシレート)及び有機溶媒を使用するが、これらの界面活性剤と有機溶媒とは、毒性、環境又は他の負の影響を有することもある。本発明における活性成分を含んだポリマーナノ粒子は、有効性を同様のレベルに保ちつつも、界面活性剤、有機溶媒の必要性を低減するか又は排除することさえ可能であり、且つ、活性成分の濃度要求を低下させることも可能である。いくつかの実施形態において、ポリマーナノ粒子は、活性成分に対して一般的には親和性を持たない表面又は塗膜に対する、活性成分の親和性を制御するために使用することができる。
溶液中でのポリマーのコンフォメーションは、溶媒との相互作用、濃度、及び、存在する可能性のある他の化学種の濃度を含む種々の溶液条件によって決定される。ポリマーは、pH、イオン強度、架橋剤、温度及び濃度に応じて、コンフォメーション変化を生じてもよい。高分子電解質は、高い電荷密度で、例えば、ポリマーの「モノマー」単位が完全に帯電する場合、類似する帯電モノマー単位間の静電反発力によって拡張したコンフォメーションを取る。塩の添加又はpH変化によるポリマーの電荷密度の減少は、拡張したポリマー鎖の、より密に充填した球形への、即ち、折り畳みコンフォメーションへの転移をもたらすことができる。折り畳み転移は、十分に小さい電荷密度で静電反発力を覆す、ポリマーセグメント間の引力相互作用によって促進される。同様の転移が、ポリマーの溶媒環境の変化によって誘発され得る。この折り畳まれたポリマーは、それ自体、ナノスケールの寸法であり、それ自体、ナノ粒子である。非帯電ポリマー、例えば、低い臨界溶液温度を有するポリマー(例えばポリ−(n−イソプロピルアクリルアミド)(「NIPAM」)等)に関しては、溶媒条件の変化を使用して同様の折り畳み転移を促進することができる。また、非帯電ポリマーの折り畳みは、適当な条件下、非溶媒の添加によって生じさせることができる。本明細書及び特許請求の範囲において、「折り畳まれたポリマー」の語は、ほぼ球状の形状、一般的には回転楕円体、及び、細長い又は多葉性コンフォメーションの、ナノメートル寸法の折り畳まれたポリマーをいう。この折り畳みコンフォメーションは、粒子内架橋によって永久的なものとされてもよい。架橋化学には、水素結合形成、新規な結合を形成する化学反応、又は、多価イオンとの配位が含まれる。架橋剤は、ポリマーが折り畳まれる前又は後に添加されてもよい。
高分子電解質等のポリマーの官能基の一部は、接合に使われるか、又は、他の官能基に変換されてもよい。これらの官能基は、他の用途の中でも、例えば、架橋、連結部位、重合、粒子内及び粒子間安定化のために使用することができる。例えば、二官能性分子である、アルコール基と潜在的なメタクリレート基とを含んだ2−ヒドロキシエチルメタクリレート(HEMA)は、エステル結合形成によって、ポリ(アクリル酸)(PAA)のカルボン酸基と反応し、カルボン酸基をメタクリレート基へと変換することができる。メタクリレート基は、UV放射又は高温に曝露した場合に架橋することができる。結果として、PAA鎖に沿って連結した多くのメタクリレート基が設計され、故に、架橋の程度が制御され得る。別の例では、酸クロリドと潜在的なメタクリレート基とを含んだメタクリロイルクロリドを、アミド結合の形成によってキトサンのアミン基と反応させ、アミン基をメタクリレート基に変換することができる。メタクリレート基は、UV放射又は高温に曝露した場合に、架橋剤であってもよい。結果として、キトサン骨格に沿って連結した多くのメタクリレート基を設計することができ、故に、架橋の程度も制御することができる。
特定の実施形態では、本発明のポリマー粒子を架橋することが望ましい。架橋は、光、温度、化学量論的試薬によって、又は、塩若しくは触媒の存在下、誘発することができる。架橋は、折り畳まれたナノ粒子内の表面層若しくは特定の位置、又は、粒子全体に亘って生じてもよい。光誘発性架橋は、種々の波長のUV及び可視光によって、空気中又は不活性環境で、光開始剤を使用するか又は使用せずに誘発することができる。UV波長領域において活性化する光開始剤の例としては、限定するものではないが、フェニルビス(2,4,6−トリメチルベンゾイル)−ホスフィンオキシド(イルガキュア819、チバビジョンコーポレーション)、アセトフェノン、及び、ベンゾフェノン(例えば2−ヒドロキシ−2−メチルプロピオフェン等)が挙げられる。可視光波長領域で活性化する光開始剤の例としては、限定するものではないが、例えば、ベンジル及びベンゾイン化合物、並びに、カンファキノンが挙げられる。架橋反応は、触媒の存在下又は非存在下、外部架橋剤の添加によって誘発されてもよい。PAAを架橋するために使用される外部架橋剤の例としては、限定するものではないが、例えば、二官能性又は多官能性アルコール(例えば、エチレングリコール、エチレンジオキシ−ビス(エチルアミン)、グリセロール、ポリエチレングリコール)、二官能性又は多官能性アミン(例えば、エチレンジアミン、1,6−ジアミノヘキサン、1,8−ジアミノオクタン、JEFFAMINE(登録商標)ポリエーテルアミン(ハンツマン)、ポリ(エチレンイミン))が挙げられる。これらの多官能性アミンは、アルカリ性の性質のため、折り畳み剤として使用することができ、ポリマーに更なる官能基を付与することを促進することができ、蛍光プローブとしてピレンを使用して特徴付けられる修飾された疎水性又は極性を含む。この反応には往々にして触媒が必要とされる。かかる触媒としては、限定するものではないが、カルボジイミド化合物、例えば、N−(3−ジメチルアミノプロピル)−N’−エチルカルボジイミドヒドロクロリド)(「EDC」)が挙げられる。化学架橋剤の他の例としては、限定するものではないが、二官能性若しくは多官能性アジリジン(例えば、XAMA−7、バイエルマテリアルサイエンス有限責任会社)、二官能性若しくは多官能性エポキシ、又は、2価若しくは多価イオンが挙げられる。
種々の態様において、本発明は、活性成分を含んだポリマーナノ粒子の製造方法を説明する。一態様において、ポリマーは、高分子電解質を含み、ナノ粒子は、高分子電解質ナノ粒子と称される。活性成分を含んだ高分子電解質ナノ粒子は、種々の方法で製造することができる。例えば、高分子電解質は、例えば、ミセル、液滴形成、又は、活性成分を含んだ高分子電解質ナノ粒子を生成するための類似する他の配合技術を使用して活性成分へと吸着させることができる。
いくつかの実施形態において、ポリマーナノ粒子は、会合した活性成分を含まずに形成される。活性成分は、ナノ粒子が完全に形成された後にナノ粒子と会合する。会合工程は、いくつかの異なる方法で達成されてよく、各々は、いくつかの異なる工程を含む。
修飾高分子電解質は、上述の通り、2種類以上の官能基、例えば、重合可能な基(HEMA)及び活性成分分子を同じポリマー骨格に沿って、又は、反応性対(クリック反応のためのアルキンとアジド)の2つの官能基を含んでいてもよい。更に、各々が反応性対のうちの1つの反応基を含んだ2つの高分子電解質の混合物が、ポリマー粒子、例えば、アルキン修飾PAA及びアジド修飾PAAを生成してもよい。一実施形態において、修飾高分子電解質は、ポリマー粒子を生成してもよい。図3は、これらの粒子を製造する工程を説明する。これらの工程は、(a)先に説明した手順に従ってPPAを、例えばHEMAによって修飾し、pH感受性ポリマーを生成することと、(b)HEMA修飾PPAをpH>6の水中に溶解させることと、(c)溶解のpHを低下させる(pH<6)ことと、(d)架橋することとを含む。この方法によって製造されるポリマー粒子の平均径は、50〜1000nmの範囲である。いくつかの実施形態において、粒径は、pH値によって制御されてもよい。大きな径は、pH値が約5〜約6の範囲にある場合に生じ、小さな径は、pH値が約3〜約5の範囲にある場合に生じた。
本明細書で記載するポリマー粒子は、活性成分を運ぶために使用することができる。ポリマー粒子に活性成分を装填するのに使用される方法のいくつかは、適当な溶媒に粒子を溶解させることを含む。ポリマーナノ粒子が溶解する場合(例えば、溶媒中で別々の個々の粒子として見出される)、該ナノ粒子を装填することが可能であるのに加えて、それらが凝集するか又は分散した形態にある場合にもポリマーナノ粒子を装填することが可能である。一実施形態において、活性成分をポリマーと会合させる方法は、(a)活性成分を溶解し、適当な溶媒に高分子電解質粒子を溶解又は分散させる工程と、(b)溶媒を除去する工程とを含む。会合した活性成分を含んだ得られるポリマー粒子は、(c)適当な条件で望ましい溶媒に粒子を再懸濁する工程と、任意に(d)活性成分を含んだ乾燥粒子を溶媒から回収する工程とを含んだ方法によって更に処理することができる。いくつかの実施形態において、活性成分とナノ粒子との間の会合を促進することのできる化学物質を添加してもよい。この化学物質は、架橋剤、配位剤、又は、活性成分若しくはナノ粒子の化学官能基を変更する化学物質であってもよく、例えば、活性成分若しくはナノ粒子の電荷又はプロトン化状態を変化させるpHの変化を含む。
種々の態様において、本発明はまた、活性成分の界面活性剤(例えば、界面活性な活性成分)の製造方法を提供する。これらの界面活性な活性成分は、種々の手法により製造することができる。一実施形態において、このことは、(a)官能基を含んだ水に不溶な活性成分と、相補的な反応基を含んだ水溶性化学物質とを混合する工程と、(b)反応を進行させ、室温又は高温で、必要であれば副産物の除去によって完了させる工程と、任意に、(c)使用する場合には有機溶媒を除去する工程とを含んでいてもよい。必要であれば、反応のために触媒を使用することもできる。特定の条件下、活性成分の界面活性剤は、製造時に活性を有する。他の条件では、活性成分の界面活性剤は、活性成分の界面活性剤からの活性成分の解放を生じ得るような溶液条件、例えばpH等で活性化されるに過ぎない。
種々の態様において、界面活性な活性成分と、活性成分を含んだポリマーナノ粒子とを一緒に使用して、活性成分の装填量を増加させたナノ粒子を製造することができ、それは分散液としてより安定性が高い。界面活性な活性成分は、ナノ粒子に吸着される可能性がある。種々の実施形態において、これは、(a)界面活性な活性成分を合成する工程と、(b)本発明に従う活性成分を含んだポリマーナノ粒子を調製する工程と、(c)界面活性成分と、活性成分を含んだポリマーナノ粒子の分散液とを混合する工程とを含んでいてもよい。工程(c)は、種々の方法で行われてよい。界面活性成分は、ナノ粒子分散液に直接的に添加してもよい。種々の実施形態において、界面活性成分がまず、pHがナノ粒子分散液と同じ水中に溶解され、次いで、ナノ粒子分散液に添加される。いくつかの実施形態において、逆の順序で添加が行われてもよい。いくつかの実施形態において、分散液及び活性成分溶液のpHは、5〜9であってもよい。添加される界面活性成分の量は、ナノ粒子に結合しない界面活性成分の別々のミセルを形成するのに十分な濃度よりも低くてもよい。種々の実施形態において、界面活性成分が、薄めずにナノ粒子分散液に添加されてもよい。いくつかの実施形態において、界面活性成分が、活性成分を含んだポリマーナノ粒子の調製の間に添加されてもよい。
種々の態様において、本発明は、活性成分を含んだナノ構造を含むポリマー水溶液の製造方法を提供する。活性成分を含んだナノ構造を含むポリマー水溶液は、種々の方法で製造されてよい。例えば、限定するものではないが、活性成分を既存の水溶性モノマーにグラフトすること、活性成分を含んだモノマーと水溶性部分を含んだモノマーとをランダムに又は制御可能に共重合することが挙げられる。一実施形態において、活性成分を既存のポリマーにグラフトすることは、(a)活性成分を既存の水溶性ポリマーにグラフトする工程と、(b)グラフトしたポリマーを溶媒中に溶解させる工程とを含むであろう。いくつかの実施形態において、これは、(a)活性成分を官能化する工程と、(b)活性成分を既存の水溶性ポリマーにグラフトする工程と、(c)グラフトしたポリマーを溶媒に溶解させる工程とを含むであろう。特定の実施形態において、ポリマーは、折り畳むことのできる又はできない高分子電解質である。
種々の態様において、活性成分の界面活性剤を使用して、活性成分のナノ構造を含んだポリマー溶液への活性成分の装填を増加させてもよい。また、活性成分の界面活性剤を使用して、ポリマー粒子の調製の間、ポリマー平均径を減少させてもよい。最終的には、活性成分の界面活性剤を使用して、ポリマー溶液の粘度を減少させてもよい。
例1:NaClを使用したポリ(アクリル酸)(PAA)溶液の処理によるポリマーナノ粒子の製造:
磁気攪拌子を具えた250mLビーカー中、固体PAA(0.100g、Mw=450,000ダルトン)及び脱イオン化水(100g)の重量を量った。PAAが完全に溶解するまで溶液を磁気攪拌し、次いで、1N NaOH水溶液を使用して、pHを9.63に調節した。
HEMA修飾PAAの合成(低度のHEMAグラフト):250mlの丸底フラスコに、固体PAA(3.0g、Mw=450,000ダルトン)及び液体DMSO(100g)を移動した。PAAが完全に溶解するまで、フラスコを磁気攪拌した。固体4−(ジメチルアミノピリジン)(DMAP,0.34g)及び液体2−ヒドロキシエチルメタクリレート(HEMA,10.8g)を反応フラスコに移した。DMAPが完全に溶解するまで反応混合物を攪拌し、その後、固体N−(3−ジメチルアミノプロピル)−N’−エチルカルボジイミドヒドロクロリド(EDC,0.53g)を移動させた。反応混合物を室温で16時間攪拌した。16時間後、混合物を700mLの2−プロパノールを含んだ1Lビーカーに滴下し、沈殿を得た。上清を廃棄し、沈殿物を2−プロパノールで2回洗浄した(各々100ml)。真空下で一晩、残余の2−プロパノールを除去することにより、固体HEMA修飾PAAを得た。
例3:外部塩の非存在下における、ポリ(アクリル酸)と架橋剤との混合物からのポリマーナノ粒子の製造:ポリ(アクリル酸)の水溶液(500mL)(Mw=450,000D)2mg/mLを、2Lビーカー中で調製した。NaOH水溶液(10N)を使用して、混合物のpHを6.8に調節した。別のビーカー(1L)に、固体1,8−ジアミノヘキサン(0.4031g)及び逆浸透(RO)水(500mL)を添加した。ジアミノヘキサンは完全には溶解しなかった。混合物のpHをpHメータでモニターし、HCl(2N)水溶液を使用して3.70まで低下させ、室温で30分間攪拌した。溶液は、まだほとんど沈殿を含まなかったが、2層のキムワイプを介して濾過することにより除去した。濾過したジアミノヘキサン混合物は透明であり、溶液を激しく攪拌しながらポリ(アクリル酸)溶液を含んだビーカーに注いだ。得られた混合物を1時間攪拌し続け、pHを5.80と測定した。次いで、混合物(300mL)を、冷却器に連結した500mLの1口反応フラスコに移動した。その後、反応混合物を24時間還流し、架橋反応を生じさせた。24時間後、反応フラスコを室温まで冷却し、pHを5.96と測定した。図8:24時間の還流前後でのPAA/1,8−ジアミノオクタン混合物のTEM画像。
例5:Al(NO 3 ) 3 を使用したポリ(アクリル酸)溶液の処理による、中空構造及び空洞を有するポリマーナノ粒子の製造(図1)
水酸化アルミニウムカプセル化PAAナノ粒子の調製:Al(NO3)3水溶液(25mM,300mL)を、磁気攪拌子を具えた1Lビーカー(A)に装填し、NaOH水溶液(100mM,145mL)を、供給ポンプによってビーカーにゆっくりと添加した。別の1Lビーカー(B)に、ポリアクリル酸水溶液(Mw=450KD,pH 7.4mg/mL,300mL)を装填し、磁気攪拌子によって攪拌した。ビーカー(A)の溶液を、供給ポンプによって3時間に亘って、ビーカー(B)にゆっくりと添加し、その間、NaOH水溶液(100mM)を継続的に添加することによってビーカー(B)の溶液のpHを7に維持した。得られた溶液を、攪拌条件下、UVランプ(252nm)の下、2時間UV照射した。VirSonic超音波装置を使用して(50%の出力で)溶液を超音波分解に供し、その後、NaOH水溶液(100mM)を添加することによってpH8.5に調節した。上記溶液を、ロータリーエバポレータ(「rotovap」)で10倍濃縮した。NaCl/エタノール溶液を添加することによって、形成されたPAAカプセル化Al(OH)3粒子を沈殿させた。沈殿物を遠心分離し、70%エタノールで3回すすいだ。沈殿物をDI水に再懸濁し、凍結乾燥して乾燥粉末を得た。DLSによってPAAカプセル化Al(OH)3粒子の特性決定を行い、平均径を20nmと決定した。
例6:酸を使用したHEMA修飾ポリ(アクリル酸)の処理によるポリマー粒子の製造:
HEMA修飾PAAの合成(高度のHEMAグラフト):250mLの丸底フラスコに、固体PAA(2.0g,Mw=450,000ダルトン)及び液体DMSO(100g)を添加した。フラスコをPAAが完全に溶解するまで磁気攪拌した。固体4−(ジメチルアミノ)ピリジン(DMAP,0.34g)と液体2−ヒドロキシエチルメタクリレート(HEMA,21.7g)とを反応フラスコに添加した。反応混合物を、DMAPが完全に溶解するまで攪拌し、その後、固体N−(3−ジメチルアミノプロピル)−N’−エチルカルボジイミドヒドロクロリド(EDC,2.67g)を添加した。反応混合物を16時間室温で攪拌した。16時間後、混合物を、900mLの脱イオン水を含んだ1Lビーカーに滴下し、沈殿を生成した。上清を廃棄し、沈殿を脱イオン水で2回(各回500mL)洗浄した。沈殿を、標準0.100N NaOH(118mL)を用いて脱イオン水(400mL)中に再溶解し、固体含有率0.73%、pH9.75の透明溶液を得た。これらの結果から、HEMAグラフトのレベルを算出し、27モル%を得た。
3−アジドプロパノールの合成:100mLの丸底フラスコ中で、液体3−クロロプロパノール(10.0g,1.0等量)、固体アジドナトリウム(17.19g,2.5等量)をDMF中、100℃で40時間反応させた。反応混合物を室温まで冷却し、分液漏斗に注ぎ、ジエチルエーテル(300mL)と塩水溶液(500mL)とで抽出した。有機層を分離して、MgSO4上で乾燥した。回転蒸発によって室温でジエチルエーテル溶媒を除去し、粗製3−アジドプロパノール(12.5g)を生成した。1H−NMR(δ,ppm)CDCl3:3.76〜3.73(t,2H,HOCH 2CH2CH2N3),3.46〜3.43(t,2H,HOCH2CH2CH 2N3),2.09(br−s,1H,OH),1.86〜1.80(m,2H,HOCH2CH 2CH2N3)。IR neat(cm−1):2100(N=N=N)。
攪拌子を具えた250mLビーカーに、N3修飾PAA水溶液(0.78重量%の12.85g)、アルキン修飾PAA水溶液(0.50重量%の20.04g)、及び脱イオン水(167.11g)の重量を量った。得られた混合物は、0.1重量%のポリマーを含み、pH値は8.03、粘度は359秒であった。50mLの混合物を、攪拌子を具えた100mLビーカーに移した。攪拌及びpHメータによるpHのモニターの間、HCl水溶液(1N)をビーカーに滴下した。透明溶液は、pH約6.2で半透明となり、その後、約5.7で不透明となった。酸性化を止め、分散液の粘度と粒子径とを測定した。DLS測定により、平均粒径は128nm(100%体積強度)、粘度は22℃で68秒と決定した。
例8:ポリマー粒子を使用したピクロラムの装填
2.5mLメタノール、例3に従って調製した8.9mgのポリマー粒子、及び、20.64mgのピクロラム(4−アミノ−3,5,6−トリクロロ−2−ピリジンカルボン酸)を、10mLのガラスバイアル中で混合した。2N HCl溶液を添加することによって溶液のpHを2に維持した。上記溶液を、透明になるまでボルテックスに供した。メタノールを蒸発によって除去した。2mLのDI水を乾燥混合物に添加し、0.5N NaOH溶液を添加することにより、溶液のpHを8に維持した。溶液を、透明になるまでボルテックスに供した。この溶液を凍結乾燥し、ピクロラムを装填したポリマー粒子の乾燥粉末を得た。各工程で保持されるピクロラムの量を、UV−Vis分光法を使用して測定した。
1mLのメタノール、例3に従って調製した6.8mgのポリマー粒子、及び、10mgのイマゼタピル(2−[4,5−ジヒドロ−4−メチル−(1−メチルエチル)−5−オキソ−1H−イミダゾール−2−イル]−5−エチル−3−ピリジンカルボン酸)を、5mLのガラスバイアル中で混合した。2N HCl溶液を添加することによって、溶液のpHを2に維持した。上記溶液を、透明になるまでボルテックスに供した。メタノールを蒸発によって除去した。1mLのDI水を乾燥混合物に添加し、0.5N NaOH溶液を添加することにより、溶液のpHを8に維持した。溶液を透明になるまでボルテックスに供した。この溶液を凍結乾燥して、イマゼタピルを装填したポリマー粒子の乾燥粉末を得た。各工程で保持されるイマゼタピルの量を、UV−Vis分光法を使用して測定した。
8mLのメタノール、例3に従って調製した2.1mgのポリマー粒子、及び、18.2mgのチフェンスルフロン−メチル(メチル3−[[[[(4−メトキシ−6−メチル−1,3,5−トリアジン−2−イル)アミノ]カルボニル]アミノ]スルホニル]−2−チオフェンカルボキシレート)を、10mLのガラスバイアル中で混合した。2N HCl溶液を添加することによって溶液のpHを2に維持した。上記溶液を透明になるまでボルテックスに供した。メタノールを蒸発によって除去した。1mLのDI水を乾燥混合物に添加し、0.5N NaOH溶液を添加することによって溶液のpHを8に維持した。溶液を透明になるまでボルテックスに供した。この溶液を凍結乾燥し、チフェンスルフロン−メチルを装填したポリマー粒子の乾燥粉末を得た。各工程で保持されるチフェンスルフロン−メチルの量を、UV−Vis分光法を使用して測定した。
4mLのメタノール、例3に従って調製した3.1mgのポリマー粒子、及び、28.5mgのチアメトキサムを、10mLのガラスバイアル中で混合した。2N HCl溶液を添加することによって溶液のpHを2に維持した。上記溶液を透明になるまでボルテックスに供した。メタノールは蒸発によって除去した。1mLのDI水を乾燥混合物に添加し、0.5N NaOH溶液を添加することによって溶液のpHを8に維持した。溶液を透明になるまでボルテックスに供した。この溶液を凍結乾燥し、チアメトキサムを装填したポリマー粒子の乾燥粉末を得た。各工程で保持されるチアメトキサムの量は、UV−Vis分光法を使用して測定した。
4mLのメタノール、例1に従って調製した3.1mgのポリマー粒子、及び、28.5mgのチアメトキサム(3−[(2−クロロ−5−チアゾリル)メチル]テトラヒドロ−5−メチル−N−ニトロ−4H−1,3,5−オキサジアジン−4−イミン)を、10mLのガラスバイアル中で混合した。2N HCl溶液を添加することによって溶液のpHを2に維持した。上記溶液を透明になるまでボルテックスに供した。メタノールは蒸発によって除去した。1mLのDI水を乾燥混合物に添加し、0.5N NaOH溶液を添加することによって溶液のpHを8に維持した。溶液を透明になるまでボルテックスに供した。この溶液を凍結乾燥し、チアメトキサムを装填したポリマー粒子の乾燥粉末を得た。各工程で保持されるチアメトキサムの量は、UV−Vis分光法を使用して測定した。
4mLのメタノール、例4に従って調製した3.2mgのHEMA修飾PAA、及び、28.4mgのチアメトキサム(3−[(2−クロロ−5−チアゾリル)メチル]テトラヒドロ−5−メチル−N−ニトロ−4H−1,3,5−オキサジアジン−4−イミン)を、10mLのガラスバイアル中で混合した。HEMA修飾PAAをNaClの存在下、UV放射によって処理した。2N HCl溶液を添加することによって溶液のpHを2に維持した。上記溶液を透明になるまでボルテックスに供した。メタノールは蒸発によって除去した。2mLのDI水を乾燥混合物に添加し、0.5N NaOH溶液を添加することによって溶液のpHを8に維持した。溶液を透明になるまでボルテックスに供した。この溶液を凍結乾燥し、チアメトキサムを装填したHEMA修飾PAAの乾燥粉末を得た。各工程で保持されるチアメトキサムの量は、UV−Vis分光法を使用して測定した。
例9に従って調製した10mgの固体ナノカプセル配合物及び20mLのDI水を、(封止用ケープを有する)50mlガラスバイアルに添加した。DI水の添加による持続放出試験の時間を計った。そして、ミニメイト接線流動濾過カプセル(Minimate Tangential Flow Filtration capsule)(TFF,3K,オメガ膜,PALL)によって上記溶液を継続的に汲み出した。試験装置を以下の図10Aに示す。放出媒質からの試料を、浸透液から必要な時間間隔で0.2ml回収し、残りの浸透液を迅速にガラスバイアルに戻した。
50μlのエチルアセテート、例2に従って調製した1.2mgのポリマー粒子、及び、1mLのDI水を5mLのガラスバイアル中で混合した。溶液のpHは3と測定された。上記溶液を、油相が消失するまで攪拌した。次いで、アトラジン(6−クロロ−N−エチル−N’−(1−メチルエチル)−1,3,5−トリアジン−2,4−ジアミンの120μLエチルアセテート溶液(溶液中のアトラジン濃度:22mg/mL)を添加し、油相が消失するまで攪拌した。蒸発によってエチルアセテートを除去し、懸濁液を形成した。この溶液を凍結乾燥して、アトラジンを装填したポリマー粒子の乾燥粉末を得た。各工程で保持されたアトラジンの量は、UV−Vis分光法を使用して測定した。
100μlのエチルアセテート、例2に従って調製した1.2mgのポリマー粒子、及び、1mLのDI水を5mLのガラスバイアル中で混合した。溶液のpHは3と測定された。上記溶液を、油相が消失するまで攪拌した。次いで、TMXが消失するまで、6.5mgのチアメトキサム(TMX,95%)を添加して攪拌した。蒸発によってエチルアセテートを除去し、懸濁液を形成した。この溶液を凍結乾燥して、TMXを装填したポリマー粒子の乾燥粉末を得た。各工程で保持されたTMXの量は、UV−Vis分光法を使用して測定した。
例2に従って調製した11.32mgのポリマー粒子、5.9mgのアゾキシストロビン(メチル(αE)−2−[[6−(2−シアノフェノキシ)−4−ピリミジニル]オキシ]−α−(メトキシメチレン)ベンゼンアセテート)、及び、4mLのメタノールを10mLのガラスバイアル中で混合した。溶液のpHは3と測定された。攪拌条件下、8.15gの水をゆっくりと(0.119mL/分)添加した。蒸発によってメタノールを除去し、懸濁液を形成した。この溶液を凍結乾燥して、アゾキシストロビンを装填したポリマー粒子の乾燥粉末を得た。各工程で保持されたアゾキシストロビンの量は、UV−Vis分光法を使用して測定した。
3mLのメタノール、例3に従って調製した11.0mgのポリマー粒子、及び5.3mgのアゾキシストロビンを10mLのガラスバイアル中で混合した。上記溶液を、透明になるまでボルテックスに供した。メタノールは蒸発によって除去した。5mLのDI水を乾燥混合物に添加し、0.5N NaOH溶液を添加することによって溶液のpHを7に調節した。溶液を攪拌して懸濁液を形成した。この溶液を凍結乾燥して、アゾキシストロビンを装填したポリマー粒子の乾燥粉末を得た。各工程で保持されたアゾキシストロビンの量は、UV−Vis分光法を使用して測定した。
例3に従って調製した12.8mgのポリマー粒子、6.0mgのアゾキシストロビン及び4mLのメタノールを、10mLのガラスバイアル中で混合した。溶液のpHを3と測定した。攪拌条件下、6.0gの水をゆっくりと(0.119mL/分)添加した。メタノールを蒸発によって除去し、懸濁液を形成した。この溶液を凍結乾燥して、アゾキシストロビンを装填したポリマー粒子の乾燥粉末を得た。各工程で保持されたアゾキシストロビンの量は、UV−Vis分光法を使用して測定した。
例20:カチオン性ポリ(アリルアミン)により被覆した2,4−ジクロロフェノキシ酢酸(2,4−D)のナノ粒子の製造
固体2,4−ジクロロフェノキシ酢酸(2,4−D)(0.158g,0.72mmol)及び作製したばかりの脱イオン化水(50mL)を、攪拌子と共に100mLのガラスビーカーに添加した。媒質をpHメータに連結し、測定値は2.76であった。攪拌した分散液に、NaOH(10N)水溶液を滴下した。pHが増加すると、より多くの固体2,4−Dを溶解した分散液は、より透明になった。最終的には、固体2,4−Dの全てが完全に溶解し、溶液は透明であった。溶液のpH及び粘度は、10.76及び25.4℃で0.93cPと測定された。参照のため、同じ機器を使用して純水の粘度を測定し、26.4℃で0.92cPの値を示した。
固体2,4−ジクロロフェノキシ酢酸(2,4−D)(16.0g,72.4mmol)を、2Lのガラスビーカーに移す前に微粉末に粉砕した。作製したばかりの脱イオン水(1L)を、1Lメスシリンダーで計量し、攪拌子と共にビーカーに移した。媒質をpHメータに接続し、測定値は2.60を示した。攪拌した分散液に、10NのNaOH水溶液を滴下した。pHが増加すると、より多くの2,4−Dが溶解し、分散液はより透明になった。最終的には、固体2,4−Dの全てが完全に溶解し(約7mLの10N NaOHを添加した)、溶液は透明であった。溶液のpHは、7.44であった。
固体2,4−ジクロロフェノキシ酢酸(2,4−D)(18.0g,81.4mmol)を、2Lのガラスビーカーに移す前に微粉末に粉砕した。作製したばかりの脱イオン水(1062mL)を、1Lメスシリンダーで計量し、攪拌子と共にビーカーに移した。媒質をpHメータに接続し、pHは2.56であった。攪拌した分散液に、10NのNaOH水溶液を滴下した。pHが増加すると、より多くの2,4−Dが溶解し、分散液はより透明になった。最終的には、固体2,4−Dの全てが完全に溶解し(約8mLの10N NaOHが添加された)、溶液は透明であった。溶液のpHは、7.60であった。
例20で調製した2,4−Dを含んだ水性ポリマーナノ粒子を、植物の処理に直接に使用した。この配合物の2,4−D濃度は、8g/Lである。2つの活性濃度(8g/L及び4g/L)を、植物の試験に使用した。植物は、処理までにトレーの中で2週間育てられ、処理の間にランダム化ブロック構造に組織化された。1つのトレーには6つの植物(オオムギ、イヌエビ、シロザ、アオビユ、ヒメチチコグサ(low cudweed)及びハッカ)が存在し、これらは、種々の作物種及び雑草種を代表する。噴霧ボトルを使用して植物に噴霧することによって処理を適用し、1ヘクタール当たり200リットルと同等の割合で噴霧液を適用することによって較正した。目視による植物毒性(%植物損傷)の評価は、処理の4、8、12及び15日後に行われた。評価は、統計的ソフトウェアプログラムに入力され、分散分析をデータに基づいて行った。分散分析が、処理間で著しい差を示唆した場合には、平均分離(mean separation)が行われた。
固体2,4−ジクロロフェノキシ酢酸(2,4−D)(8.0g,36.2mmol)を、2Lのガラスビーカーに移す前に微粉末に粉砕した。作製したばかりの脱イオン水(1L)を1Lのメスシリンダーによって計量し、攪拌子とともにビーカーに移した。媒質をpHメータに接続し、測定値は2.76であった。攪拌している分散液に、10NのNaOH水溶液を滴下した。pHが増加すると、より多くの固体2,4−Dが溶解し、分散液はより透明になった。最終的には、固体2,4−Dの全てが完全に溶解し、溶液は透明であった。溶液のpHは、8.50であった。
固体グリホサート(N−(ホスホノメチル)グリシン)(8.0g,94.6mmol)及び作製したばかりの脱イオン水(1L)を、攪拌子と共に2Lビーカーに添加した。媒質をpHメータに連結し、測定値は2.20であった。攪拌している分散液に、NaOH(50重量%)水溶液を滴下した。pHが3まで増加すると、固体グリホサートの全てが完全に溶解し、分散液はより透明になった。NaOH水溶液(50重量%)を、媒質のpHが7.2に達するまで添加した。
例26:溶媒としてトルエンを、触媒として濃縮H 2 SO 4 を使用した、カルボワックスMPEG350(ダウから供給、メトキシ終端ポリ(エチレングリコール)、Mn=350)を使用した2,4−Dのエステル化
固体2,4−D(3.0g,13.6mmol)、液体カルボワックスMPEG350(5.0g,14.3mmol)、トルエン(150mL)を、250mLの丸底フラスコに攪拌子と共に添加した。反応フラスコをディーン−スタークトラップ及び冷却器に連結した。反応混合物を24時間還流し、その後、室温まで冷却した。エチルアセテートとトルエンとの混合物(50/50,v/v)を移動溶媒として用いた薄層クロマトグラフィーを使用して、反応の完了を確認した。トルエンをロータリーエバポレータで除去し、2,4−D活性成分の界面活性剤の薄黄色液体を得た。残余のトルエンをさらに、真空ポンプで除去した。1H−NMR(300MHz,D2O):δ3.38(s,3H,CH 3 (OCH2CH2)n−OCH2CH2−O(O)C−),3.36〜3.73(m,PEG,−(CH3−(OCH 2 CH 2 )n−OCH 2 CH2−O(O)C−),4.36(t,2H,CH3−(OCH2CH2)n−OCH2CH 2 −O(O)C−),6.81(d,1H,芳香性のH),7.18(dd,1H,芳香族のH),7.38(d,1H,芳香族のH)。
固体2,4−D(20.0g,90.5mmol)、液体カルボワックスMPEG750(68.4g,90.5mmol)、シリカゲル60A(200〜400メッシュ)(1.0g)を250mLの丸底フラスコに攪拌子とともに添加した。反応フラスコをディーンスタークトラップと冷却器とに連結した。反応混合物を、窒素ガスの緩流の下、150℃に加熱した。反応の間、エステル化反応の副産物と水とをディーンスタークトラップ中で濃縮し、回収した。薄層クロマトグラフィーを使用して、反応の完了をモニターした。反応は3時間で完了した。粗製生成物を濾過してシリカゲルを除去し、2,4−D活性成分からなる界面活性剤の薄黄色液体を得た。1H−NMR(300MHz,D20): δ3.38(s,3H,CH 3 −(OCH2CH2)n−OCH2CH2−O(O)C−),3.36〜3.73(m,PEG,−(CH3−(OCH 2 CH 2 )n−OCH 2 CH2−O(O)C−),4.36(t,2H,CH3−(OCH2CH2)n−OCH2CH 2 −O(O)C−),6.81(d,1H,芳香族のH),7.18(dd,1H,芳香族のH),7.38(d,1H,芳香族のH)。
例24に従って製造した液体2,4−D界面活性剤(2,4−D自体の4.0gに相当する34.72g)及び2L脱イオン化水を、攪拌子と共に3Lのプラスチックビーカーに移した。2,4−D界面活性剤を完全に溶解させ、溶液は、薄黄色であったが、pH値2.76で透明であった。NaOH(10N)水溶液2、3滴を溶液に添加し、pHを6.65まで増加させた。このpHで、溶液の粘度は、24.0℃で1.08cPaであり、体積分布分析により得られた動的光散乱の結果は、平均径252nmの単一分布を示した。
例29:2,4−Dの増加した装填を含んだナノ粒子の製造
固体2,4−ジクロロフェノキシ酢酸(2,4−D)(4.0g,18.1mmol)を、2Lのガラスビーカーに移す前に微粉末に粉砕した。作製したばかりの脱イオン水(1l)を1Lのメスシリンダーで計量し、攪拌子と共にビーカーに移した。媒質をpHメータに接続した。攪拌している分散液に、NaOH(10N)水溶液を滴下した。pHが増加すると、より多くの固体2,4−Dが溶解し、分散液はより透明になった。最終的には、全ての2,4−Dが完全に溶解し、溶液は透明であった。溶液のpHは、9.20と測定された。
この例は、PAAカプセルが、活性成分を装填し、オタワ砂を介して移動することができることを示した。疎水性蛍光染料(修飾したホスタソールイエロー)が、モデル活性成分として使用された。
標準的なオタワ砂(VWR,CAS#14808−60−7)を脱イオン水で2回洗浄し、使用前に空気中で乾燥させた。乾燥した砂を、カラム中で固相として使用し、PAAカプセルを含んだ及び含まない染料をカラムに装填した。
この結果は、修飾ホスタソールイエローが、カラムからカプセルへ移動しなかったことを示す。
例31:ミクロ環境感受性蛍光プローブとしてのピレンの使用
異なるポリマーナノ粒子に由来するピレンミクロ環境を、以下のナノ粒子:Na+−折り畳まれたポリアクリル酸(Na−PAA)、ZnO/ポリアクリル酸ナノ粒子(ZnO−PAA)、Zn2+−折り畳まれたナノ粒子(Zn−PAA)、Na+−折り畳まれたPMAAナノ粒子(Na−PMAA)、Na+−折り畳まれたP(MAA−co−EA)ナノ粒子(Na−P(MAA−EA)、ポリ(ビニルピロリドン)−折り畳まれたポリアクリル酸ナノ粒子(PVP/PAA)について検査した。ピレンナノ粒子水溶液を以下のようにして調製した。1.0mgのピレンを10mLのジクロロメタンに溶解し、ストックピレン溶液(0.1mg/mL)として使用した。ピレンナノ粒子水溶液を調製するために、10マイクロリットルのストックピレン溶液を20mLのシンチレーションバイアルに添加し、ドラフト中で1時間空気乾燥した。その後、80mgの固体ナノ粒子又はポリマー、10gの脱イオン水及び磁気攪拌子をバイアルに加えた。次いで、バイアルの蓋を固く閉め、アルミニウムホイルで覆い、溶液を室温で2日攪拌した。異なるナノ粒子及びポリマーの全てについて同じ手順を使用した。HCl(0.1N及び1N)並びにNaOH(0.1N及び1N)水溶液を使用して、溶液のpHを調節した。励起及び発光の双方のスリット幅が2.5nmであり、励起波長340nmを使用した発光スペクトルを、パーキンエルマーLS55発光スペクトロメータで測定した。図10は、水中のピレン及び低pHのNa−P(MAA−co−EA)ナノ粒子中でのピレンの発光スペクトルを示す。第1(I1,〜373nm)及び第3(I3,〜384nm)の振電バンドの発光強度を記録し、種々のポリマーナノ粒子系について比(I1/I3)を算出した。種々のポリマーナノ粒子のこれらの比を表4に示す。
赤色染料2番の溶解度について、いくつかのナノ粒子配合物中での溶解度と、水単独中での溶解度とを比較した。100mgのナノ粒子(Na+−折り畳まれたポリアクリル酸ナノ粒子(Na−PAA)、ZnO/ポリアクリル酸ナノ粒子(ZnO−PAA)、Zn2+−折り畳まれたナノ粒子(Zn−PAA)、Na+−折り畳まれたPMAAナノ粒子(Na−PMAA)、Na+−折り畳まれたP(MAA−co−EA)ナノ粒子(Na−P(MAA−EA)、Zn2+−折り畳まれたナノ粒子(Zn−PAA))を、0.5mgの赤色染料2番及び脱イオン水30mLと混合した。1時間激しく混合した後、赤色染料2番及びナノ粒子の種々の溶液を3500rpmで10分間遠心分離し、分散していない染料を分離した。ポリマーナノ粒子を含んだ溶液からの上清液体は、明るい赤色であったが、水のみを含んだ溶液からの上清液体は、無色であった。ポリマーナノ粒子を含んだ溶液からの上清液体の赤色は、ポリマーナノ粒子とそれらを配合することによって染料の溶解度が増加したことを示す。
100mgのバニリン及び100mgのPAA粒子を2ドラムガラスバイアルに入れた。メタノール5mLをガラスバイアルに添加した。溶液を、磁気攪拌プレートにおいて攪拌子を使用して30分間攪拌した。50mLのRO水を、別の250mlガラスビーカーに取り、磁気攪拌子を使用して攪拌した。メタノール混合物を、攪拌した水中に滴下した(1ml/分)。上記溶液を2時間攪拌した。得られた溶液は、半透明であった。ロータリーエバポレータを使用して、溶液からメタノールを除去した。得られた溶液を凍結乾燥し、乾燥粉末を得た。凍結乾燥した固体は、RO水中の200ppmのバニリン溶液として、再分散させた。

Claims (20)
- ポリマーナノ粒子、及び、該ポリマーナノ粒子と会合した少なくとも1つの中性の非イオン性有機化合物を含み、
前記ポリマーナノ粒子が、折り畳まれた(collapsed)水溶性高分子電解質を含み、
前記の折り畳まれた(collapsed)水溶性高分子電解質は、少なくとも100,000ダルトンの分子量を有する、架橋しているランダム共重合体であり、
前記ポリマーナノ粒子は、直径100nm未満であり、そして、
前記中性の非イオン性有機化合物が、染料、顔料、着色剤、油、UV光吸収分子、香料、香味分子、保存料、導電性化合物、熱可塑性化合物、接着促進剤、浸透促進剤、防錆剤及びこれらの組み合わせからなる群より選択される、組成物。 - 前記ポリマーナノ粒子が、50nm未満の直径である、請求項1に記載の組成物。
- 前記ポリマーナノ粒子が、20nm未満の直径である、請求項1に記載の組成物。
- 前記ポリマーナノ粒子が、多数の高分子電解質分子を含む、請求項1に記載の組成物。
- 前記中性の非イオン性有機化合物が、疎水性である、請求項1に記載の組成物。
- 前記高分子電解質が、電磁波放射誘発性架橋、化学誘発性架橋、又は、熱誘発性架橋のいずれか1つによって架橋される、請求項1に記載の組成物。
- 前記中性の非イオン性有機化合物が、ポリマーナノ粒子の非存在下における溶解度よりも高い濃度で分散する、請求項1に記載の組成物を含んだ分散液。
- 前記高分子電解質が、ポリ(アクリル酸)、ポリ(メタクリル酸)、ポリ(スチレンスルホン酸塩)、キトサン、ポリ(ジメチルジアリルアンモニウムクロライド)、ポリ(アリルアミンヒドロクロライド)又は、これらの組み合わせを含む、請求項1に記載の組成物。
- 前記中性の非イオン性有機化合物の少なくとも一部が、前記ポリマーナノ粒子の内部に存在する、請求項1に記載の組成物。
