Description
癌は、最も一般的な疾患の一つであり、西欧諸国において主要な死因である。一般に、罹患率は、ほとんどの形態の癌について、年齢と共に増加する。全般的な健康状態の増加により、人類の寿命は伸び続けているので、癌は、増加している個体数に影響を及ぼしうる。最も一般的な癌型の原因は、まだほとんど分かっていないが、環境因子(食事、タバコの煙、UV照射など)ならびに遺伝的因子(p53、APC、BRCA1、XPなどの「癌遺伝子」における生殖細胞変異)と癌発症の危険性との間の関連性を提供する知見が増えてきている。
疑いのある腫瘍由来の生検材料の顕微鏡評価が依然として癌診断の代表的な基準である。確固とした診断を得るために、腫瘍組織は、ホルマリン中で固定され、組織化学的に処理され、パラフィン包埋される。得られるパラフィンブロックから、組織切片を調製し、組織化学的方法、すなわち、ヘマトキシリン−エオシン染色、および免疫組織化学的(IHC)方法の両方を用いて染色することができる。次いで、全体的および顕微鏡分析を包含する病理学的技術を用いて、該外科的試料を評価する。該分析は、しばしば、特定の診断を指定するための基礎、すなわち、腫瘍の腫瘍型を分類し、悪性度合を等級付けするための基礎を形成する。
白金に基づく化学療法は、精巣癌、卵巣癌、子宮頸癌、肺癌、膀胱癌、結腸直腸癌、および頭頸部癌などの癌の治療に用いられる。現在、3つのFDAに承認された白金に基づく化合物(シスプラチン、カルボプラチンおよびオキサリプラチン)があるが、開発中または臨床試験中の新規な誘導体(例えば、サトラプラチン(satraplatin)およびピコプラチン(picoplatin))がある。新規な白金に基づく化合物の開発における1つの目標は、毒性を最小限にすることであり、現在使用される化合物では、毒性が強いことがある。一般的な副作用には、腎毒性、神経毒性および毛髪の喪失がある。
白金に基づく化学療法剤は、DNAに結合し、それにより、DNA付加物を誘導し、DNA構造を崩壊させる架橋を導く。通常、これらの損傷は、最終的にアポトーシスに至る。しかしながら、薬物耐性の問題がある。腫瘍のサブセットは、従来の療法に応答せず、該耐性は、内因性または獲得性のいずれかである。
癌のアジュバント治療を用いる試験の終点分析は、ある特定の療法に対して患者がどのように応答するかについて、重要な情報を与える。全生存(OS)は、長い間、標準的な主要終点と考えられてきた。OSは、原因に関係なく、例えば、死が癌によるものか、またはそうでなくても、死までの時間を考慮に入れる。追跡不能者(loss to follow-up)を検閲し、局所的再発、遠隔転移、二次原発卵巣癌および二次的な他の原発性癌を無視する。
本開示のいくつかの態様のうちの1つの目的は、白金に基づく治療に対する応答性のレベルの予測について提供することである。
本開示の第一の態様として、癌を有する哺乳動物対象が第1の群に属するか、または第2の群に属するかを決定する方法であって、第1の群の対象が第2の群の対象よりも白金に基づく治療によく応答し、
a)該対象から事前に得られた試料の少なくとも一部に存在するRBM3タンパク質またはRBM3mRNAの量を評価し、該量に対応する試料値を決定する工程、
b)工程a)で得られた試料値を参照値と比較する工程、
該試料値が参照値よりも高い場合は、c1)該対象が第1の群に属すると結論づけ、

1.抗原の生成
a)材料および方法
EnsEMBL Gnene ID ENSG00000102317によってコードされる標的タンパク質の適当なフラグメントは、ヒトゲノム配列を鋳型として用いるバイオインフォマティックツールを用いて選択された(Lindskog Mら(2005) Biotechniques 38: 723-727, EnsEMBL, www.ensembl.org.)。該フラグメントは、RBM3タンパク質(配列番号2;EnsEMBLエントリー番号ENSP00000365946)のアミノ酸18〜151(配列番号1)に対応する134アミノ酸長フラグメントの製造のための鋳型として使用した。
RBM3の全長転写産物(配列番号3)のヌクレオチド281−682に対応する遺伝子フラグメントは、特異的プライマーを用いて、ヒトRNAプールからRT−PCRによって首尾良く単離された。該フラグメントは、標的タンパク質RBM3(配列番号2)のアミノ酸18〜151をコードする。膜貫通領域およびシグナルペプチドは成熟タンパク質では切断されているので、該標的タンパク質(配列番号2)の該134アミノ酸フラグメント(配列番号1)は、イー・コリでの有効な発現を確実にするために膜貫通領域を欠き、かつ、いずれかのシグナルペプチドを欠くように設計された。さらに、該タンパク質フラグメントは、他のヒトタンパク質に対して低い相同性を有する独特の配列からなるように、生じたアフィニティー試薬の交差反応性を最小限にするように、かつ、立体構造エピトープの形成を可能にし、かつ、いまだ細菌系における効率の良いクローニングおよび発現を可能にするのに適当なサイズであるように設計された。
a)材料および方法
上記で得られた精製したRBM3フラグメントを抗原として用いて、ナショナルガイドライン(スウェーデン許可番号第A84−02号)にしたがって、ウサギを免疫化した。一次免疫として、フロイントの完全アジュバント中200μgの抗原で該ウサギを筋内に免疫化し、4週間隔で3回、フロイントの不完全アジュバント中100μgの抗原で追加免疫した。
ポリクローナル抗体調製の質は、抗体精製におけるストリンジェンシーの度合いに依存することが証明されており、以前に、標的タンパク質に由来しないエピトープに対する抗体の涸渇が他のタンパク質に対する交差反応性およびバックグラウンド結合を回避するために必要であることが示された(Agaton Cら(2004) J. Chromatogr. A 1043: 33-40)。かくして、高い特異性の単一特異性ポリクローナル抗体が、His6−タグに対する抗体ならびにABP−タグに対する抗体の涸渇によって生じることを保証するために、タンパク質マイクロアレイ分析を行った。
3.モノクローナル抗体の生成
a)材料および方法
セクション1で得られた精製フラグメント(配列番号1)をモノクローナル抗体の産生のための抗原として用いた。抗原は、AbSea Biotechnology Ltd(中国、北京)に送られ、簡単に言うと、該抗原をBALB/cマウス(4−6週齢、雌)中に3週間隔で皮下注射した。第1の注射のために、該抗原をフロイントの完全アジュバントと混合し、続く注射のために、フロイントの不完全アジュバントと混合した。融合の3日前、最後に抗原をマウスに静脈内注射した。マウス脾臓細胞をSp2/0ミエローマ細胞系統と融合することによって、ハイブリドーマを生成した。ELISAを用いていくつかの細胞系統をスクリーニングすることにより、該抗原(配列番号1)に特異的な抗体を分泌する細胞を同定し、さらなる特徴付けのために、Atlas Antibodies ABに送った。ELISA、ウェスタンブロット(WB)および免疫組織化学(IHC)において陽性の結果を示した細胞系統を選択し、AbSea Biotechnology Ltdによってサブクローニングを行った。
細胞系統をELISAによってスクリーンして(AbSeaにて)、抗原(配列番号1)を認識するが、アフィニティータグHis−ABPを認識しないモノクローナル抗体(mAbs)を産生する系統を同定した。8個の細胞系統がELISAにおいて、抗原配列番号1に対する特異的結合を示し、さらなる試験のために選択された。選択された8個のクローンの各々について、150−300μl上清を収集し、アジドを加え、該上清を氷上で、Atlas Antibodies ABに送った。AbSeaの指示にしたがって、到着まで該上清を+4℃で保管した。細胞系統のさらなる試験は、3つの興味深い細胞系統、ウェスタンブロットおよびIHC分析において陽性結果を示したクローン1B5、6F11および7G3の同定をもたらした。これらのクローンは、AbSea Biotechnology Ltd.によって実施されるサブクローニングおよび拡大のために選択された。
4.細菌性ディスプレーIを用いるエピトープマッピング
配列番号1に対応するRBM3 DNA(すなわち、ENSP00000365946のアミノ酸18−151またはENST00000376755の塩基対261−682)を鋳型としてベクターpAff8cを用いるPCRによって増幅した。増幅したDNAを超音波処理によって、種々の長さ(約50−150bp)に断片化し、次いで、ブドウ状球菌性ディスプレーベクター(pSCEM2)中にライゲートし、エス・カルノサス(S.Carnosus)中に形質転換して、約100000個の形質転換体を得た。フレーム内DNAフラグメントは、ブドウ状球菌の表面にペプチドとして展示された。抗体(上記セクション2で得られた配列番号1に対して選択的)および蛍光標識した二次試薬と共にインキュベーション後、フローサイトメトリーを用いて陽性および陰性細胞を別々に分類して、エピトープおよび非エピトープ提示細胞を単離した。単離した細胞をパイロシークエンシングによって配列決定し、エピトープの同定のために、配列を最終的にRBM3抗原に対してアラインした。
a)合成ペプチド調製
RBM3上のPrEST HPRR232631(配列番号1)に対応する25個のビオチン化ペプチドからなるPEPスクリーンライブラリーをSigma−Genosys(Sigma-Aldrich)によって合成した。該ペプチドは、全PrEST−配列を含むと共に、10アミノ酸オーバーラップを有する15アミノ酸長であった。該ペプチドを80%DMSO中、最終濃度10mg/mlに溶解させた。
ニュートラアビジン(Neutravidin)(Pierce, Rockford, IL)をカルボキシル化ビーズ(COOHミクロスフェア、Luminex-Corp. Austin, TX)上に、製造者のプロトコールにしたがって固定化した。フィルター膜底ミクロタイタープレート(MultiScreen-HTS, Millipore, Billerica, MA)を用いて、以前に記載されていたように(Larssonら(2009) J Immunol Methods 15; 34(1-2): 20-32, Schwenkら(2007) Mol Cell Proteomics 6(1) 125:32)、106ビーズのカップリングを行った。異なる色彩コードIDを有する25個の別個のビーズ群を、1−エチル−3−(3−ジメチルアミノ−プロピル)カルボジイミドおよびN−ヒドロキシスクシンイミドを用いて活性化した。ニュートラアビジン(MES中100μg/ml)をビーズに加え、振盪機上で120分間インキュベートした。最後に、ビーズを洗浄し、再懸濁し、NaN3で補足したタンパク質含有バッファー(BRE、ELISAのブロッキング試薬、Roche, Basel, Switzerland)中、4℃で保管するために、ミクロ遠心チューブに移した。全てのカップリングしたビーズ集団を超音波洗浄器(Branson Ultrasonic Corporation, Danbury, CT)中で5分間超音波処理した。該ビオチン化ペプチドをBRE中で20μMの濃度まで希釈し、各ペプチド100μlをカップリング反応に用い、該反応は、RTにて振盪しながら60分間行った。最後に、ビーズを3x100μlのBREバッファーで洗浄し、さらなる使用まで4℃で保管した。
全25種類のビーズIDを含有するビーズ混合物を調製し、PBS中50ng/mlに希釈した各抗体45μlを5μlのビーズ混合物と混合し、RTにて60分間インキュベートした。フィルター底マイクロタイタープレート(Millipore)を洗浄に用い、各インキュベーション後、全てのウェルを3x100μlのPBSTで洗浄した。50μlのR−フィコエリトリン標識抗−ウサギIgG抗体(0.5μg/ml、Jackson ImmunoResearch)または50μlのAlexa Fluor555ヤギ抗−マウスIgGを添加し(0.4μg/ml)、RTにて60分間最終インキュベートした。
単一特異性ポリクローナル抗体(抗−RBM3)およびモノクローナル抗体6F11の特異性は、合成ビオチン化ペプチドと結合したビーズを用いるアッセイにおいて試験された。抗−RBM3は、該ペプチドのうち8個、すなわち、6、7、8、14、15、16、24および25(PrEST配列上の3つの明確な領域に対応する)(コンセンサス配列 配列番号6、7、8および9)に対して強い結合を示した。特に、配列番号9に対応するペプチド24および25は、強力なシグナルを生じた。モノクローナル抗体6F11は、2つのペプチド:15および16(PrEST配列上の1つの明確な領域に対応する)(コンセンサス配列 配列番号8)と反応した。抗−RBM3と6F11の両方がペプチド15および16に結合したので、これらの抗体が該領域内に1以上のエピトープを共有することが示される。配列番号6は配列番号4内にあり、配列番号8は有る程度まで配列番号5とオーバーラップすることに注目すべきである。

7.ディスカバリーTMA
a)材料および方法
20の異なる癌型由来の新生物組織を含有するパラフィンコアを、実施例のセクション3で得られたモノクローナル抗体1B5を用いて分析した。組織マイクロアレイ(TMA)生産のためのドナーブロックとして用いられた全ての組織は、地域倫理委員会の承認にしたがって、ウプサラの大学病院の病理学部(Department of Pathology, University Hospital)での保管物から選択された。TMA分析に用いられた組織切片は、診断および分類を決定するために調べられた。診断目的で、全組織をホルマリン固定し、パラフィン包埋し、切片化した。
表2は、種々の癌型由来の組織試料におけるRBM3タンパク質発現のレベルを示す。RBM3タンパク質発現は、甲状腺癌を除き、試験された全ての癌型において見出される。しかしながら、甲状腺癌については、たった3人の異なる対象由来の試料しか調べることができなかった。例えば、乳癌対象では、染色スコアは0〜2の範囲であり、子宮頸癌対象では0〜2の範囲であり、結腸直腸癌対象では0〜3の範囲であり、頭頸部癌対象では0〜2の範囲であり、肺癌対象では0〜2の範囲であり、卵巣癌対象では0〜2の範囲であり、精巣癌対象では0〜2の範囲であり、尿路上皮癌対象では0〜2の範囲であることが示される。かくして、RBM3タンパク質は、一般的に、癌組織において差次的に発現されていると結論づけることができ、種々の癌型における治療予測マーカーとしてのその役割を支持する。
a)材料および方法
原発性侵襲性上皮卵巣癌について外科的処置がされた154人の患者由来の保管されたホルマリン固定パラフィン包埋組織をスウェーデンのLund and Malmoe大学病院の病理学部から収集した。患者は、1982年〜2007年に診断された。倫理的許可は、ルンドの地域倫理委員会から得た。
コホートの最初の分析は、各々、図1Aおよび図1Bに示されるように、全患者についてのOSが約38%であり、OCSSが約39%であることを明らかにした。
a)材料および方法
データは、1992年〜2006年に上皮卵巣癌と診断された285人の患者コホートに基づいて、以前に行ったマイクロアレイ研究から収集した。患者の同意についての施設内倫理委員会(Institutional Review Board)の承認は、全患者について得られた。
RBM3発現のアレイ分析は、285個の腫瘍試料のうち210個について分析された。二分化された変数が統計学的分析のために構築された。図19に示されるように、高RBM3発現レベルを伴う腫瘍を有したシスプラチン処置患者は、低RBM3発現レベルを伴う腫瘍を有した患者よりも、再発のない著しく良好な生存を示した。
a)材料および方法
腫瘍材料は、1995年〜2008年にUMASの病理学部で精巣胚細胞(testicular germ-cell)腫瘍(TGCT)と診断された30人の患者から収集された。該試料のうち5個は純粋なセミノーマであり、25個は、非セミノーマ性TGCTであった。非セミノーマ性TGCTは、International Germ Cell Cancer Collaborative Group(IGCCC)にしたがって分類された。25個の試料のうち4個は、IGCCC中程度予後として分類されたケースであり、5個の試料は、IGCCC予後不良として分類された。残りの16個の非セミノーマ性TGCTケースは、IGCCC予後良好として分類された。該研究の許可は、ルンド大学の倫理委員会から得られた(ref nr 447-07および493-09)。5人の患者は疾患により死に、そのうち4人は、IGCCC不良で、1人は、IGCCC中程度であった。全ての患者は、シスプラチン処置を受けた。
RBM3発現に、0、1、2または3のアノテーションを付加し、ここに、3は、最も高い染色スコア(SS)を示した。図20に示されるように、RBM3発現強度は、確立された予後カテゴリーとよく相関する。疾患によって死亡した患者は、全て、比較的低いレベルのRBM3発現を示す。これは、また、予後不良カテゴリーに属するほとんどの患者に当てはまるが、真に強いRBM3発現(SS=3)を有する全ての患者および強いRBM3発現(SS=2)を有するほとんどの患者は、予後良好カテゴリーに見出すことができる。予後良好であることがよく知られているセミノーマ患者の全ては、真に強いRBM3発現(SS=3)を有した。
11.シスプラチン感受性および耐性細胞系統におけるRBM3発現
a)材料および方法
シスプラチン感受性細胞系統(A2780)およびシスプラチン耐性細胞系統(A2780−cp70)におけるRBM3タンパク質の発現をウェスタンブロットによって分析した。ウェスタンブロットは、還元条件下、17%SDS−PAGEゲル上で選択されたヒト細胞系統由来の全タンパク質抽出物を分離し、次いで、製造者の推奨にしたがって、RVDF膜(Bio-Rad Laboratories)へ電気転写することによって実施した。該膜は、室温にて1時間ブロックされ(0.1%Tween20を含有する1xPBS中5%BSA)、一次アフィニティー精製抗体(ブロッキングバッファー中で1:1000希釈)と共にインキュベートし、PBST中で洗浄した。二次HRP−コンジュゲート抗体(ヒツジ抗−マウス免疫グロブリン/HRP、GE)をブロッキングバッファー中で1:1000希釈し、ChemidocTMCCDカメラ(Bio-Rad Laboratories)およびウェスタンブロッッティングルミノール試薬(Santa Cruz Biotechnologies, Inc)を用いて、製造者のプロトコールにしたがって、化学発光検出を行った。
ウェスタンブロット分析の結果を図22に示す。RBM3タンパク質の発現は、シスプラチン感受性細胞(A2780)におけるよりも、シスプラチン耐性細胞(A2780−cp70)において低い。使用された全抗体(抗RBM3、6F11、1B5)は同様の結果を示す。
a)材料および方法
細胞生存率に対するシスプラチンの効果は、製造者の推奨にしたがい、WST−1アッセイ(Roche Applied Science)によって決定された。シスプラチンの添加1日前に、A2780およびA2780−Cp70細胞を96ウェルプレートにおいて、100μlの適当な培地中2500細胞/ウェルの密度で播種した。該細胞をシスプラチン(1−100μM)で48時間処理した。試料は3連で作成した。1ウェルにつき10μlのWST−1溶液を加え、細胞を37℃で4時間インキュベートした。450nmの波長および690nmの参照波長にて、スキャニングマルチウェル分光光度計、ELISAリーダーを用いて、各ウェルの吸光度を測定した。
シスプラチン処理後のA2780およびA2780−cp70細胞の生存率を分析した場合、親A2780細胞と比べて、シスプラチン耐性A2780−cp70細胞が増加した生存率を有することが明確に示される(図23)。

ヒト卵巣癌細胞系統A2780およびシスプラチン耐性細胞変種A2780Cp70を5%CO2の加湿インキュベーター中、37℃にて、グルタミン、10%仔ウシ血清および1%ペニシリン/ストレプトマイシンを補足したRPMI−1640中で維持した。シスプラチン(Sigma)を0.9%NaCl中に溶解して1mg/mlのストック溶液を得、最終濃度(1−100μM)まで細胞培養物に添加した。
RBM3に対するsiRNA(Applied Biosystems, Carlsbad, Ca)または対照siRNA(Applied Biosystems)でのトランスフェクションをリポフェクタミン2000(Invitrogen, Carlsbad, CA)を用いて、最終濃度50nM siRNAで実施した。
細胞生存率に対するシスプラチンの効果は、実施例のセクション12に記載されるように、WST−1アッセイによって決定した。
図24に示されるように、低RBM3タンパク質発現は、シスプラチン処理後、シスプラチン感受性細胞の増加した生存率を導く。非曝露細胞と比べた場合、異なるRBM3 siRMAに曝露した細胞において生存率に顕著な相違がある。これらの結果は、低RBM3タンパク質発現がシスプラチン耐性を示すこと、および高RBM3タンパク質発現がシスプラチン感受性を示すことを示す。
14.非限定的な例
患者における癌の確立後、該患者由来の腫瘍組織試料を得る。該腫瘍組織は、該腫瘍の初期の外科的除去由来の試料または腫瘍生検から得てもよい。さらに、「陰性参照」を提供するために、低RBM3タンパク質発現を有するか、または本質的にRBM3タンパク質発現を欠く組織を含む保管材料から、試料を採取する。かかる保管組織は、例えば、試験対象と同じ癌型由来の予め確立された低RBM3タンパク質発現レベルを有する組織であってもよい。さらに、「陽性参照」を提供するために、高RBM3タンパク質発現を有する組織、例えば、試験対象と同じ癌型由来の予め確立された高RBM3タンパク質発現レベルを有する悪性メラノーマ組織を含む保管材料から、試料を採取する。
免疫組織化学は、実施例のセクション8に記載されるように実施する。各試料由来の1以上の試料をガラススライド上に載せ、60℃で45分間インキュベートし、(目的の試料がパラフィン化されている場合、)キシレン中で脱パラフィン化し(2x15分)、段階的アルコールで脱水する。抗原回復のために、スライドをTRS(標的回復溶液、pH6.0、DakoCytomation)中に浸し、DecloakingチャンバーR(Biocare Medical)中、125℃で4分間沸騰させる。スライドをAutostainerR(DakoCytomation)中に置き、内在性ペルオキシダーゼを最初にH2O2(DakoCytomation)でブロックする。複数の試料切片を載せる理由は、結果の正確性を増すためである。
Claims (17)
- 癌を有する哺乳動物対象が第1の群に属するか、または第2の群に属するかを決定する方法であって、第1の群の対象が第2の群の対象よりも白金に基づく治療によく応答し、
a)該対象から事前に得られた試料の少なくとも一部に存在するRBM3タンパク質またはRBM3mRNAの量を評価し、該量に対応する試料値を決定する工程、
b)工程a)で得られた試料値を参照値と比較する工程、
該試料値が参照値よりも高い場合は、
c1)該対象が第1の群に属すると結論づけ、
該試料値が参照値と等しいか、または参照値よりも低い場合は、
c2)該対象が第2の群に属すると結論づける工程
を含む方法。 - 癌を有する哺乳動物対象の白金に基づく治療の強度のレベルを決定する方法であって、
a)該対象から事前に得られた試料の少なくとも一部に存在するRBM3タンパク質またはRBM3mRNAの量を評価し、該量に対応する試料値を決定する工程、
b)工程a)で得られた試料値を参照値と比較する工程、
該試料値が参照値よりも高い場合は、
c1)該対象が第1の強度の白金に基づく治療を施されるべきであると結論づけ、
該試料値が参照値と等しいか、または参照値よりも低い場合は、
c2)該対象が第2の強度の白金に基づく治療を施されるべきであると結論づけ、ここに、該第2の強度は該第1の強度よりも高い、工程
を含む方法。 - 癌が精巣癌、卵巣癌、肺癌、膀胱癌、結腸直腸癌、子宮頸癌、乳癌および頭頸部癌から選択される、請求項1または2記載の方法。
- 白金に基づく治療がカルボプラチン、パラプラチン、オキサリプラチン、サトラプラチン、ピコプラチンおよびシスプラチン治療から選択される、請求項1〜3のいずれか1項記載の方法。
- 試料が対象由来の腫瘍細胞を含む、請求項1〜4のいずれか1項記載の方法。
- 工程a)が、
aI)試料に、評価されるべきRBM3タンパク質との選択的相互作用が可能な定量化できるアフィニティーリガンドを添加し、該添加を該試料中に存在するRBM3タンパク質に対する該アフィニティーリガンドの結合を可能にする条件下で実施し、
aII)該試料に結合したアフィニティーリガンドを定量化して量を評価する
ことを含む、請求項1〜5のいずれか1項記載の方法。 - a)RBM3タンパク質との選択的相互作用が可能な定量化できるアフィニティーリガンド、および
b)該定量化できるアフィニティーリガンドの量を定量化するために必要な試薬
を含む、請求項1〜6のいずれか1項記載の方法を実施するためのキット。 - 該アフィニティーリガンドが、アミノ酸配列が配列番号5からなるペプチド、または20アミノ酸以下からなり、かつ、配列番号8、16および17から選択されるアミノ酸配列の全長を含むRBM3フラグメントと選択的相互作用が可能なアフィニティーリガンドである、請求項6記載の方法。
- 該アフィニティーリガンドが、アミノ酸配列が配列番号5からなるペプチド、または20アミノ酸以下からなり、かつ、配列番号8、16および17から選択されるアミノ酸配列の全長を含むRBM3フラグメントと選択的相互作用が可能なアフィニティーリガンドである、請求項7記載のキット。
- 癌を有する哺乳動物対象の白金に基づく治療のための治療予測マーカーとしての、RBM3タンパク質またはRBM3mRNA分子のイン・ビトロでの使用。
- アミノ酸配列が配列番号5からなるペプチド、または20アミノ酸以下からなり、かつ、配列番号8、16および17から選択されるアミノ酸配列の全長を含むRBM3フラグメントと選択的相互作用が可能な、アフィニティーリガンド。
- 癌を有する哺乳動物対象の白金に基づく治療のための治療予測剤としてのRBM3タンパク質との選択的相互作用が可能なアフィニティーリガンドのイン・ビトロでの使用。
- 癌を有する哺乳動物対象の白金に基づく治療のための治療予測剤の製造における、RBM3タンパク質との選択的相互作用が可能なアフィニティーリガンドの使用。
- 癌を有する哺乳動物対象の治療のための医薬の製造における白金に基づく治療剤の使用であって、該対象が高RBM3タンパク質または高RBM3mRNAである、使用。
- 白金に基づく治療剤がカルボプラチン、パラプラチン、オキサリプラチン、サトラプラチン、ピコプラチンまたはシスプラチンである、請求項14記載の使用。
- RBM3タンパク質との選択的相互作用が可能なアフィニティーリガンドを含む、白金に基づく治療剤での治療に対する応答を予測するための診断組成物。
- 該治療がカルボプラチン、パラプラチン、オキサリプラチン、サトラプラチン、ピコプラチンまたはシスプラチン治療である、請求項16記載の組成物。
