a)ビタミンE誘導体、アネキシンなどの陰イオン性リン脂質結合タンパク質、ホスファチジルセリンなどの陰イオン性リン脂質、およびステロールからリポソームを形成すること、ならびに
b)リポソーム組成物中に輸送物をカプセル化すること
を含む方法を提供する。
角膜上皮ならびに血液脳関門および血液網膜関門の内皮などの物理的障壁は、密に接着した細胞のシートで構成されている。細胞間の接合部(角膜またはBBBのいずれでも)は、化合物の移動を制限する、タイトジャンクションを形成するタンパク質複合体によって制御される。これらの接合部の選択性は、上皮組織の特定の型に依存するが、一般的には、低分子量(100〜200Da)の親水性薬物またはイオンのみが傍細胞経路を利用することができる。親油性薬物は、BBBの内皮を透過することができるが、それらの通過は、やはり、400Da未満の分子に制限される。角膜において、角膜実質は、親油性薬物にさらなる障壁を与え、それらの後眼部への透過を阻止する。
脳薬物の世界市場は、主に、薬物の大部分がインビボでBBBを通過せず、通過するものは、高脂溶性および400〜500ダルトン(Da)未満の低分子量を有する小分子に限定されるという事実により低開発分野である。これらの化学的性質を有する小分子の数は、全薬物の2%未満である。他の薬物はBBBを通過しない。BBBの解決がなければ、脳を治療する可能性があり得る全薬物の98%より多くは、開発されない。これは、神経保護薬を含む新薬が不足している、アルツハイマー病、筋萎縮性側索硬化症、ハンチントン病、パーキンソン病、脳卒中、脳脊髄損傷、脳癌、および多発性硬化症などの重篤な疾患における障害となっている。大分子を輸送する能力により、組換えタンパク質、抗体、およびRNA/DNA分子のCNS内の標的部位への送達も可能となるであろう。
リポソーム調製
リポソームを、脂質フィルム再水和方法によって調製した。クロロホルム:メタノール(5:1 v/v)中に溶解した関心対象の脂質(ホスファチジルコリン(PC)、ホスファチジルセリン(PS)、コレステロール、およびα−トコフェロール)を、適切な量で混合し、溶媒を定常窒素流下で除去した。使用範囲は、15〜30%のコレステロール(好ましくは15%)、5〜20%のPS(好ましくは10%)、および0.1〜20%のビタミンE誘導体(好ましくは1%)であることが示され、残りはPCで構成された。その結果生じた脂質フィルムを、適切な容量の緩衝液(PBS±フルオロフォア/輸送物)中で再水和した。52mMの貯蔵濃度のリポソームを調製し、その後、それをUDDSに使用するのに6.5mMに希釈した。
Lucentis(ラニビズマブ)は、血管内皮増殖因子A(VEGF−A)を標的にするヒト化モノクローナル抗体断片である。その抗体のタンパク質性質により、蛍光色素の特定のアミノ酸へのコンジュゲーションが可能になる。Lucentis(10mg/ml)を0.1Mの重炭酸ナトリウム緩衝液、pH8.3中で洗浄し、約2mg/mlに濃縮した。標識反応については、タンパク質を任意の一級アミン部位(リシン残基およびN末端)で標識する100μgのalexafluor−488(DMSO中に溶解したスクシンイミジルエステル)を使用して室温で行った。その薬物はまだ特許権下であり、そのタンパク質のアミノ酸配列はパブリックドメインで入手できないため、標識に利用できる一級アミン基の数はわからない。コンジュゲートしていない色素をゲル濾過で除去し、標識されたLucentisを1×PBS pH7.4へ溶出した。分光法によるそのタンパク質の分析は以下の結果を生じた。
薬物が組成物を使用して眼底へ送達され得るかどうかを決定するために、過剰のLucentis−488を、エレクトロポレーションのプロセスを使用して組成物によってカプセル化した。カプセル化の通常の方法である、凍結/融解は、これまで、タンパク質の変性を引き起こしていた。15μlのLucentisを50μlの組成物に加え、細菌細胞および哺乳類細胞の形質転換に通常使用する、0.1cmギャップのGene Pulserキュベットに移した。キュベットに、以下の条件:2.5kV、25μF、および200Ω下でBioRad Gene Pulserを使用して電気パルスをかけた。通常の使用において、電気パルスは、小分子(一般的にはプラスミドDNA)が細胞のリン脂質膜を通過するのを可能にする。リポソームは細胞膜と構造的類似性を有するので、その方法は、凍結/融解に代わる良好なカプセル化方法である可能性が高いと判断された。
凍結/融解によるカプセル化を、液体窒素中、輸送物の存在下で担体を凍結し、その後、迅速に、温水の定常流下で試料を融解することによって行った。そのプロセスを10回繰り返して、妥当な時間スケール内で最適なカプセル化がもたらされた。そのプロセスは、単純な分子についてはうまくいくが、タンパク質などの複雑な分子には変性をもたらし得る。
取り込まれなかったフルオロフォアを、60,000×gで45分間の超遠心によって除去した。上清を捨て、ペレットを新鮮なPBS中に再懸濁した。
投与前に、負荷されたリポソームを、陰イオン性リン脂質結合タンパク質アネキシンVと、5mg/mlのリポソームおよび300μg/mlのアネキシンVの最終濃度が達成されるように混合した。リポソームおよびアネキシンの濃度のバリエーションを使用して、異なる送達レベルを達成することができる。安定性問題により、最大濃度の2mg/mlのアネキシンを使用することができるが、20mg/mlを超えるリポソームの用量は塞栓形成を増加させるであろう。
麻酔された動物(ダークアグーチ系(Dark Agouti)ラット)を、共焦点レーザー走査型検眼鏡(cSLO)を使用してイメージングした。各動物の蛍光ベースライン画像をとった。簡単に述べれば、動物を、眼内がイメージングされるようにcSLOの前に位置づける。488nmのアルゴンレーザー波長を小さなスポットへと集中させ、1対のミラーによって網膜にわたってスキャンし、その波長においてフルオロフォアを励起させる。生じた蛍光を、関心対象の深さの面より上または下の面における不要な蛍光を排除する効果を生じる共焦点開口に光学的に集中させる。網膜神経線維層の面においてイメージングを行った。したがって、ベースライン画像は、95%強度に設定されたアルゴンレーザーでの固有の488自己蛍光のみを記録する。
ベースライン画像が記録された後、10μlの本発明のカプセル化Cy3標識IgGをラット眼上に局所的に投与した。Cy3は、アルゴンレーザー励起後、検出することができる、よく知られた蛍光色素である。局所処置から1時間後、ベースラインと同じ設定、すなわち、アルゴンレーザー488nmの波長、95%強度で眼を再びイメージングした。その後、得られた画像は、(ベースラインとしての)固有の488自己蛍光、加えて、網膜神経線維層の面に到達した任意のCy3標識IgGを記録する。
1.輸送物を送達するためのUDDSの改良
輸送物送達の成功のための成分は、ビタミンE誘導体、ホスファチジルコリン、ホスファチジルセリン、コレステロール、およびアネキシンを含む。異なる輸送物/構成物比を可能にするためのプロセスの改良において、精製方法およびカプセル化方法を、下記のように最適化した:
カプセル化の成功を評価することにおける主な課題は、取り込まれなかった材料の除去のための信頼でき、かつ効率的な方法を生み出すことである。下記に概要を示したいくつかの方法が選択されている。
60,000gでのリポソームの超遠心は、リポソームをペレット化し、それらと共にいかなるカプセル化された材料も沈降するであろう。しかしながら、個々の分子は、この速度ではペレット化する能力がなく、上清中のままである。上清を除去し、ペレットを新鮮な緩衝液中に再懸濁する。試料を洗浄するこのプロセスを、上清がもはや有意な量のカプセル化されなかった材料を含有しなくなるまで、繰り返す。
カラム
試料を、分子をそれらのサイズに基づいて分離するカラムに入れる。リポソームなどの巨大分子構造は、迅速にカラムを通過するが、より小さな分子の進行は遅延する。フルオレセインについて、例えば、5,000daの分子カットオフを有する使い捨てのPd−10カラムを用いた。
試料を、適切な分子量に対応するポアを含む透析チューブに入れた。リポソームなどのこのカットオフより大きい粒子は、チューブ内に保持され、一方、そのポアより小さい粒子は、より大きいバルク溶液中へ自由に拡散することができる。透析を一晩行い、2回の透析ステップ後でさえも、かなりのバックグラウンド蛍光があった。
精製のための最も適切な方法を決定したので、低分子量の化合物のカプセル化のための異なる方法も調べた。これらの方法には、以下の工程を含む:
(i)化合物の存在下でリポソームを作製する工程、
(ii)ボルテックス工程、
(iii)超音波処理、
(iv)凍結融解工程、および
(v)エレクトロポレーション。
輸送物を分子量によって類別すると、以下の3つのバンドが同定された:低分子量蛍光化合物、例えば、フルオレセインナトリウム(分子量:376.28da)、中位の分子量、例えば、aBT(8,000da)およびLucentis(48,000da)、ならびに高分子量、例えば、Avastin(149,000da)。サイズ排除カラムシステムを使用することの一つの利点は、特定の分子量のカットオフを有する異なる樹脂を使用することが可能であることである。
リン酸濃度に対応する吸光度値を提供するために、試料を5MのHClによって処理し、ローダミンBおよびヘプタモリブデン酸アンモニウムの混合物とインキュベートすることができる。溶液中のリン酸の量は、存在するリン脂質のレベルに直接反映する。
リポソーム試料を、FPLC Superdex75およびSuperdex200カラムに流し、溶出液を1.5mlの画分に収集した。超遠心された場合、いずれの画分も予想されたリポソームペレットを生じず、リン脂質アッセイは、図10に示されているように、緩衝液対照と比較して無視できる量のリン酸しか示さなかった。
重力送りPD−10カラムに関する限界は、5,000Daの分子量カットオフ(MWCO)であり、それは、5,000Daより大きい任意の化合物間の差を分割することはできないことを意味していた。したがって、30,000Daのより高いMWCOを有するG−50の粗い樹脂などの他の樹脂は、リポソーム破裂を引き起こすことなくリポソームから小タンパク質を分離することができるはずである。
フルオレセインに関して超遠心を用いようと試みたときに遭遇した問題は、主に濃度依存性であった。存在する色素の莫大な量は、プロセスの効率を制限したが、α−BTおよびLucentisなどのタンパク質ははるかに低い濃度で使用されることになっているので、必要とされる洗浄の回数はより少ないはずである。
より現実的な透過標的を作製するために、本発明者らは、カプセル化効率をさらに増加させようと試みた。これを行うための以下の3つの可能な方法がある:
→最初のα−BT濃度を増加させる
→脂質濃度を増加させる
→凍結融解およびエレクトロポレーションなどのカプセル化の代替方法を適用する
カプセル化の程度の正確な値は計算することが困難であるとはいえ、高脂質溶液の濁度による光散乱の増加が、蛍光測定値に干渉し、蛍光シグナルを低下させる(図14参照)。
カプセル化値を正確に決定するために、フルオレセインを、緩衝液およびリポソームの両方と、単独で、ならびにSDS、NP40、Tween−20、およびTriton X−100を含む様々な界面活性剤の存在下で、インキュベートした。
1.成功した網膜への送達をスクリーニングするための新規な方法
異なるUDDS組成物の効力を評価するために、本発明者らは、網膜送達を評価するための新規な標識アルファ−ブンガロトキシン(αBT)を同定した。(Sigma(T9641)から入手した)αBTは、図16に示されているように、網膜における動脈を特異的に標識し、それにより、優れたマーカーを代表している。
カプセル化材料の放出を利用するために、カプセル化αBTおよびアネキシンV−776を含有するリポソームを、硝子体内注射によって投与し、3時間後、眼をイメージングした。
本発明者らは、アネキシンについてのコドン最適化配列を入手した。図18が示しているように、新しい系は、高レベルのアネキシンV−GSTを発現し、静脈内送達のための十分な量のアネキシンの生成を可能にする。
A.血液網膜関門(BRB)
a)フルオレセインナトリウムの網膜への静脈内送達
主要な目的は、血液脳関門および血液網膜関門を通過するカプセル化材料の送達の成功を示すことであった。UDDSのために最適化されたカプセル化および放出を確立したので、UDDSのフルオレセインナトリウムを網膜および脳へ送達する能力を評価した。
眼におけるカプセル化リポソームからの輸送物の治療的放出を評価するために、眼あたり500ngの組換えヒトVEGF(rhVEGF165)を使用するVEGFモデルを開発した。図21は、血管蛇行の増加(A、反射画像)、VEGF注射から24時間後にとられたフルオレセイン血管造影後に見られる、血管の漏れおよび肥厚(B、蛍光画像)を有する眼を示す。これらの効果の逆転は、2.5μlの治療グレードのLucentisの硝子体内注射を使用して達成された(C〜D)。Lucentis処理された眼は、未処理のVEGFモデルに対して、血管サイズを正常に縮小し(C、反射画像)、漏れがない正常な血管組織(D、蛍光画像)を示す。
Lucentisの分子量が高いため、血液網膜関門を通過することができず、静脈内投与には適しないものとなっている。この研究は、カプセル化lucentis(lucentis UDDS)が、静脈内投与によって、BRBを通って網膜にうまく送達され得るかどうかを調べるために設計された。1つの眼をVEGF処理し、対側眼を未処理のままにしておいた動物を調製した。VEGF処理の時点で、ラットはまた、1mlのカプセル化Lucentis(A、B)か300μlの臨床グレードLucentis(C、D)のいずれかの静脈内注射を受けた。より多い容量のカプセル化材料を投与し、カプセル化効率によるLucentis濃度の低下を考慮した。
フルオレセインの脳への静脈内送達
300μlの5%フルオレセインナトリウムまたは1mlの0.2%カプセル化フルオレセインナトリウムを、ラットに尾静脈を通して投与し、網膜をモニターした。遊離フルオレセインを、前に概要を示したように、超遠心およびPD−10カラムを使用することによって除去した。2時間後、一般的な麻酔下で、動物は、4%パラホルムアルデヒドでの固定灌流を受けた。脳を4%パラホルムアルデヒド中に一晩、保存し、その後、800umのスライスに切断し、共焦点蛍光顕微鏡下で調べた。
