Description
2000年7月13日に出願された対応する米国特許出願第09/615102号は、「イオン移動度及び質量のハイブリッド分析器」という名称の米国特許第5909258号の部分継続出願である1999年5月17日に出願された「イオン移動度及び質量の分析器」という名称の関連米国特許出願第09/313492号の部分継続出願である。
本発明は概略的に分子を少なくともその構造と気相イオンとしての質量電荷比とに基づいて特性分析する装置に関し、より詳細には生体分子を含む有機分子及び無機分子に関する組成、配列順序及び/または構造の情報の迅速で感度の高い分析を行う装置に関する。
ほとんど全ての従来の順序規定手法の中心的部分は、クロマトグラフィーによる、あるいはポリアクリルアミドのゲルの電気泳動(PAGE)による複合した組の配列順序に関連する分子部分の分析である。PAGEに基づいた自動配列順序決定装置は現在存在し、典型的には多数の塩基に特定して終止した生体分子生成物中に含有され、その後にポリアクリルアミドゲルにより処理される多くの蛍光染料を必要とする。離散した長さの生成分子が放射源による励起に応じた放出蛍光によってゲルの底部近くで検出される。
Claims (5)
- 液体クラマトグラフィー装置を操作してイオンまたは分子の塊を保持時間の関数として時間的に分離するステップと、
上記保持時間の関数として予め時間的に分離された上記イオンまたは分子の少なくとも一部をイオン移動度の関数として順次時間的に分離するステップと、
上記イオン移動度の関数として予め時間的に分離された上記イオンの少なくとも一部を選択的にフィルタリングしそれによって所望の質量電荷比だけを有するイオンを順次生ずるステップと、
所望の質量電荷比だけを有する上記イオンの少なくとも一部を娘イオンに順次細分するステップと、
イオン移動度の関数として予め時間的に分離された前記イオンの少なくとも一部をイオン質量の関数として順次時間的に分離するステップと、
の各ステップからなることを特徴とするイオンを時間的に分離する方法。 - イオンまたは分子の塊を保持時間の関数として時間的に分離するための液体クロマトグラフィー装置と、
イオンまたは分子の塊を保持時間の関数として時間的に分離するための前記液体クロマトグラフィー装置に連結されたイオン入口と、イオン出口とを有し、イオンをイオン移動度の関数として時間的に分離するように作用するイオン移動度分析器(IMS)と、
上記IMSのイオン出口に連結されたイオン加速領域を有しイオンをイオン質量の関数として時間的に分離するように作用する質量分析器(MS)と、
上記IMSと上記MSとの間に配置されたイオンフィルタリング装置であって、該イオンフィルタリング装置が所望の質量電荷比を有するイオンだけを通過させられるように動作可能である、イオンフィルタリング装置と、
上記フィルタリング装置と上記MSとの間に配置された衝突セルであって、該衝突セルがその内に緩衝気体を受け入れるように動作可能であり、それによって該衝突セルに入るイオンが上記緩衝気体と衝突して娘イオンに細分される、衝突セルと
からなることを特徴とするイオンを時間的に分離するための装置。 - 上記MSが、検出されたイオンの関数としてイオン信号を生成するイオン検出器を含み、
上記MSの上記イオン検出器に接続される入力部を有するコンピュータであって、上記イオン信号を処理しそれに基づいてイオン質量、イオン移動度及び上記保持時間の関数として上記イオンの塊の少なくとも一部に関する情報を決定するコンピュータをさらに含む
ことを特徴とする請求項2に記載の装置。 - イオン源をさらに含むことを特徴とする請求項2に記載の装置。
- 上記イオン源が、電気スプレー式イオン化源を含む
ことを特徴とする請求項4に記載の装置。
