Description
また、本発明は、標的化シグナルと融合された単一鎖可変断片(ScFv)とのタンパク質相互作用を用いる植物作成医薬品の免疫標的化の分野にも関する。
また、本発明は、植物作成医薬品の翻訳後修飾に関与する異種酵素の再配向の分野にも関する。
事実、治療タンパク質の大部分は、遺伝子からの遺伝的情報が、機能的遺伝子産物の形成を指令する最終工程であるいくつかのPTMを受ける。
真核生物細胞はそれらの修飾のほとんどを実現することができる。しかしながら、これらの成熟はより一般的には宿主系に特異的である。さらに、翻訳後修飾は植物細胞に対して哺乳動物細胞とは異なる。植物では、他の真核生物細胞におけるように、N−グリコシル化はERにおいて開始し、可能なN−グリコシル化配列であるAsn−X−Ser/Thrを構成する特異的アスパラギン残基へオリゴ糖前駆体(Glc3Man9GlcNAc2)が同時翻訳付加される。一旦生じて間もないタンパク質上に移動すれば、分泌された糖タンパク質が分泌経路に沿って輸送される間に、オリゴ糖前駆体は、添付の図2に模式的に示されたようにERおよびGAにおける糖残基の除去または付加に関連するいくつかの成熟を受ける。植物および哺乳動物N−グリカン成熟が異なるのは後期GAにおいてのみであり、この結果、PMPのN−グリカンにおけるα−(1,6)−結合フコース、β(1,4)−結合ガラクトースおよびシアル酸の不存在、および分岐β(1,2)−キシロースおよびコアα−(1,3)−フコースの存在を生じさせる(添付の図2参照)。
植物ER−存在タンパク質の構造分析は、かかるタンパク質が主として高マンノース型のN−グリカンを備えていることを示している(Navazio et al.,1997(参考文献41),1998(参考文献42);Pagny et al.,2000(参考文献49))。これらのオリゴ糖構造は植物および哺乳動物に共通しており、従って、免疫原性ではない。この観察は、β−(1,2)−キシロース、またはα−(1,3)−フコースのような免疫原生残基の植物作成医薬品(PMP)N−グリカンへの会合を妨げる戦略を示唆している。この戦略は、ER内での、すなわち、免疫原生グリコエピトープが付加されて植物N−グリカンを複合体化するゴルジ嚢の上流での組換えタンパク質の貯蔵にある。まず、組換え可溶性タンパク質のC末端におけるH/KDEL配列の付加が植物ERにおけるその保持で十分であることが示された(Gomord et al.,1997(参考文献24),1999(参考文献22),Saint−Jore−Dupas et al.,et 2004(参考文献57))。
事実、少数の研究のみによってC末端ジリシンモチーフの役割(Barrieu and Chrispeels,1999(参考文献3);Benghezal et al,2000(参考文献4);McCartney et al.,2004(参考文献35);Reyes et al,2006(参考文献53))、膜貫通ドメイン(TMD)の長さ(Brandizzi et al.,2002a(参考文献9))または芳香族アミノ酸豊富化ER検索シグナル(McCartney et al.,2004(参考文献35))が提供されている。α−グルコシダーゼIは、生じたばかりの糖タンパク質上のオリゴ糖前駆体の移動「エンブロック」の直後に該オリゴ糖前駆体から末端側α−
(1,2)−結合グルコース残基を特異的に除去することによってN−結合オリゴ糖前駆体の成熟に関与する、最初の酵素である(図2参照)。機能、および、結果として、ERにおけるこのII型膜タンパク質の位置は、必須である。事実、哺乳動物細胞では、かかるタンパク質が新生児で欠損していると、重度の全般性低血圧症および形成異常特徴を誘導し、74日齢において致死に至る(De Praeter et al.,2000(参考文献14))。

ここに、本発明の適用は、ERおよび/またはGAに対する不活化抗体を特異的に用いて、コンパートメントに局在化された特異的酵素を不活化する。この目的では、本発明によるERおよび/またはGA保留ペプチドシグナルは、究極的な目標でERまたはGA酵素の触媒ドメインに対する特異的親和性を有する抗体に融合させて、内因性酵素を不活化して、組換えタンパク質の成熟を制御し、または調節している。組換えタンパク質の役割は、ここでは組換えポリペプチドの成熟に関与する酵素を捕獲することである。本発明は、従って、宿主細胞内の内因性酵素を「免疫調節」するのを可能とする。

本発明によると、ポリペプチドは、特に、植物細胞または全植物において、遺伝子組換えによって産生されるのに必要な任意のポリペプチドであってよい。本発明で産生することができるポリペプチドは、例えば、本発明の組換えポリペプチドの開示において先に引用したものである。

実施例1
標的化シグナルの同定
1.初期ゴルジII型膜タンパク質の局在化
初期ゴルジコンパートメントにおけるN−グリカンプロセッシング酵素の選択的保留を可能とするメカニズムを良好に理解するために、N−グリカンプロセッシング機構の4つの異なるメンバー(A−グルコシダーゼI、マンノシダーゼI、N−アセチルグルコサミニルトランスフェラーゼIおよびb−1,2−キシロシルトランスフェラーゼ、添付の図4)に対する一連のGFP融合の局在化を、タバコBY−2細胞における安定した発現後に実験した。

2.ルミナールドメインは、ManIおよびGNTIのゴルジおよびER標的化に必要でないことを示す実験
3つの植物グリコシル化酵素の特異的ゴルジ保留に関して、GNTI,XylTおよびArabidopsis ManI(添付の図2)は、それらの特異的標的化が、GNTIについての細胞質テイル、TMD、およびステムを含めたそれらのN末端部分に含有されたシグナルによって媒介することを示す(Essl et al.,1999(参考文献16),Dirnberger et al.,2002(参考文献15);Pagny et al.,2003(参考文献50);Strasser et al.,2006(参考文献61))。本実施例は、ManIおよびGNTIの標的化においてルミナールドメインの役割を調べる。

and Jackson,1996(参考文献62);Zerangue et al.,1999(参考文献66))、またはERからゴルジまでのそれらの輸出(Giraudo and Maccioni,2003(参考文献20))のいずれかを容易とするシグナルを含有する。植物においては、細胞質テイルの長さは、異なるグリコシダーゼおよびグリコシルトランスフェラーゼの間で広く変化できる(添付の図8)。例えば、GCSIおよびManIは長い細胞質テイルを含有する(各々、51/29個のアミノ酸)。比較として、GNTIおよびXylTのCTは11個のアミノ酸のみから構成される。
4.膜貫通ドメイン(TDM)の長さがゴルジ標的化、およびManIのサブコンパートメント化において重要な役割を演じることを示す実験
哺乳動物細胞では、どのようにしてII型膜タンパク質がゴルジ内で異なるレベルで保持されるかを説明するために、いくつかのモデルが提案されてきた。

6.TMD長さモデルが全てのII型膜タンパク質に適用されないことを示す実験
TMD長さが、植物分泌経路に沿った全てのグルコシダーゼおよびグリコシルトランスフェラーゼのサブコンパートメント化を可能とする唯一のゴルジソーティング決定基であり得るか否かを決定するために、特徴付けられたグリコシル化酵素のN末端配列を比較した(添付の図14)。

7.シロイヌナズナ GCSIII型膜タンパク質の局在化
GCSIはタバコ小胞体に厳格に蓄積される。
GCSIのERにおける選択的保留を可能とするメカニズムを理解するために、GCSI標的化におけるルーメンドメインの役割をまず調べた。ERルーメンに位置するGCSIの部分がこのグリコシダーゼをERに標的化するにおける役割を演じるかを決定するために、GCSIの最初の150アミノ酸(CT+TMD+stem81アミノ酸)または最初の90アミノ酸(CT+TMD+stem21アミノ酸)をGFPに融合し、対応するキメラタンパク質を、各々、GCS150およびGCS90と命名した(図15に添付)。GCS150およびGCS90をAgro浸潤によって、BY−2浮遊培養細胞において安定に発現させるか、またはタバコ葉表皮細胞において一過的に発現させ、共に、正確に(図16A、Bに添付された)全長構築物について従前に観察されているように、(各々、図17A、C、Eおよび17B、D、Fに添付された)双方の発現系におけるERで観察した。これらの切断融合がタバコ葉表皮細胞において一過的に発現される場合、全発現レベルが既に強く減衰している場合には形質転換から5日後にER標識が依然として観察され、一方、同一条件で分析したゴルジ融合が専らゴルジに位置することに注意するのは重要である(Saint−Jore−Dupas et al.,2006;データは示さず)。このデータはグルコシダーゼIの触媒ルーメンドメインが酵素のER位置で必要でなく、GCSIの最初の90アミノ酸がGFPをERに保持するのに十分であること示している。
哺乳動物および酵母ERに存在する多くの膜タンパク質のN末端サイトゾル領域は、厳格な保留(Nilsson et al.,1994(参考文献46);Opat et
al.,2000(参考文献48),Hardt et al.,2003(参考文献28);Ciczora et al.,2005(参考文献12))、またはゴルジからERへ戻るそれらの検索(Teasdale and Jackson,1996(参考文献62);Zerangue et al.,1999(参考文献66))を容易とするシグナルを含有し、一方、いくつかの他のものはERからゴルジへの輸出を促進する(Giraudo and Macioni,2003(参考文献20))。植物においては、少数の研究のみが、膜タンパク質のCTにおけるER輸出配列の存在を示し(Contreras et al.,2004(参考文献4613);Yuasa et al.,2005(参考文献65);Hanton et al.,2005b(参考文献27))、および膜タンパク質ER保留を担うサイトゾルモチーフの特徴付けをいう(Benghezal et al.,2000(参考文献4);McCartney et
al.,2004(参考文献35))。
ク質を明るいスポットに再度位置させた(図18C〜18D、図19A)。加えて、D13GCS90はERマーカーmRFP−HDELと同時局在化されなかった(図19B)が、タバコは表皮細胞における一過的発現の後にはST−mRFTゴルジマーカーと完全に同時局在化された(図19C)。興味深いことには、GCSIの13のN末端アミノ酸のXYLT35ゴルジマーカーへの融合はERにおけるレポータータンパク質をブロックした(図18G〜18H;図19G)。GCS13−XYLT35融合タンパク質はGCS90構築物のようにERを標識し(図18A〜18D)、mRFP−HDEL ERマーカーと同時局在化された(図19H)。強力なER標識に加えて、GCS13−XYLT35をタバコ葉表皮細胞において発現した場合に、少数の明るいスポットがやはり観察された。これらのスポットは移動し、ST−mRFPゴルジマーカーと同時局在化される(図19I)。
アルギニン残基がAtGCSIの13のN末端アミノ酸におけるER保留に必須であるか否かをさらに調べるために、AtGCSIのヒトホモログのN末端に位置するアルギニンリッチなドメイン、またはI型植物ER存在膜タンパク質A.thalianaカルネキシンのサイトゾルC末端に位置するアルギニンリッチなドメインいずれかのためのペプチドを変化させた。
11.GCSIのサイトゾルテイルにおけるアルギニン残基はER局在化情報を含有することを示す実験
GCSIの13個の最初のアミノ酸内の4個のアルギニン残基の役割をより正確に定義するために、PCR部位特異的突然変異誘発を用いてロイシンまたはアラニン残基いずれかによってこれらの残基を置換し(構築物の詳細については表1参照)、得られた融合タンパク質をタバコ細胞で発現させた。
中で示すようにゴルジ装置に対応する(図20F)。ER標識は検出されなかった(図20E)。細胞内局在化に対する同一効果は、4つのロイシン残基によるこれらの4つのアルギニン残基置換後に観察された(図20G〜20I)。これらの観察は、N末端に対して位置6〜7〜10および12に位置する4つのアルギニンがGCS90のER存在性を担う構造的情報についてコードしているようである。
12.RR、RXRまたはRXXRモチーフを保有する融合タンパク質が、ゴルジに会合して、ERに蛍光スポットで蓄積することを示す実験
次の工程は、R/L6−7GCS90、R/L10−12GCS90およびR/L6−12GSC90キメラ融合タンパク質による蛍光スポットとして標識された構造を同定することであった。R/L6/7GCS90タンパク質に関して説明したように、GFP蛍光は、ERにおいて、および蛍光構造において蓄積し、それはゴルジスタックよりも小さい(図20J〜20L、および図21A〜21B)。細胞におけるこれらの構造の局在化をより正確に規定するために、mRFP−HDEL ERマーカー、ST52−mRFPゴルジマーカー、およびR/L6−7GCS90、R/L10−12GCS90またはR/L6−12GSC90融合タンパク質のいずれかを同時に発現させた。
al.,2005に記載されたようにER出口部位(ERES)で有り得た。これを証明するために、R/L6−7GCS90、R/L10−12GCS90およびR/L6−12GSC90融合タンパク質をタバコ葉表皮細胞においてSar1p−mRFPと同時発現させた。
突然変異した形態Sar1p/GTP−mRFPの発現はERにおいてゴルジR/LGCS90の保留に導いた。
13.AtGCSIのER保留はN末端アルギニンモチーフのみに依存しないことを示す実験
本実施例においては、ゴルジレポータータンパク質に融合したGCSIアルギニンモチーフはそれをERに局在化させた。しかしながら、これらのシグナルが全長GCSIのER局在化に関与する主な保留シグナルであるという証拠はない。
先の実施例に示したように、表3にリストした各シグナルは、緑色蛍光タンパク質のようなレポータータンパク質をERおよび/またはGAへ標的化するのに十分である(添付の図25Aおよび25B参照)。

植物ER−存在タンパク質の構造分析は、それらが専ら高マンノースタイプN−グリカンを担うことを示している(Navazio et al.,1997(参考文献41),1998(参考文献42);Pagny et al.,2000(参考文献49))。これらのオリゴ糖構造は植物および哺乳動物に共通しており、したがって、免疫原性ではない。この観察は、β1,2キシロースまたはα1,3フコースのような免疫原性残基の植物が作成した医薬品(PMP)N−グリカンの会合を妨げるための戦略を示唆した。この戦略は、ER内への、すなわち、免疫原性グリコエピトープが成熟する植物N−グリカンにそこで加えられるゴルジ嚢の上流への組換えタンパク質の貯蔵にある。まず、組換え可溶性タンパク質のC末端におけるH/KDELアミノ酸配列の付加は、植物ERにおけるその保留に十分であることが示された(Gomord et al.,1997(参考文献24),1999(参考文献22))。
配列(配列番号1〜8)は、ERへ抗体の重鎖および軽鎖を標的化するのに用いられてきた。
これらの抗体は専ら非免疫原性高マンノース型のN−グリカンを表わし(Sriraman et al.,2004(参考文献59);Petrucelli et al.,2006(参考文献51))、これは、a−マンノシダーゼIのような、ERおよびシスゴルジに位置した酵素に制限されるグリカン成熟に基づく非常に有効な再循環を示す(Nebenfhur et al.,1999(参考文献43))。従って、免疫原性N
−グリカンの、ER保留シグナルへの融合を介してのPMPへの会合への防止は可能である。
実施例B:ERおよび/またはGAにおける抗体または抗体断片の発現(添付の図15、レーンB)
ERおよび/またはGAにおける抗体または抗体断片の発現は、組換えタンパク質産生を改良するための異なる戦略を与える。例えば、それは、非修飾(突然変異した)治療タンパク質をERおよび/またはGAへ標的化するのに導くことができる。
実施例C:植物細胞のERおよび/またはGAにおける同種または異種酵素の発現、および該細胞における組換えタンパク質の発現
実施例1:植物細胞のER/GAにおける哺乳動物グリコシルトランスフェラーゼをアドレスすることによる植物細胞でのN−グリコシル化のヒト化(添付の図15、レーンC)
植物においては、他の真核生物細胞におけるように、オリゴ糖前駆体(Glc3Man9GlcNAc2)の、生成直後のポリペプチド上の特異的アスパラギン残基への同時翻訳付加と共に、N−グリコシル化はERにおいて開始する。一旦タンパク質に移動されたならば、かつ分泌された糖タンパク質が分泌経路に沿って輸送されている間に、オリゴ糖は複雑なN−グリカンをもたらすいくつかの成熟を受ける。免疫応答のトリガリング(Wright and Morrison,1994(参考文献64))のような、それらのエフェクター機能で用いられる抗体を含めた多くの医薬品は、それらのイン・ビボ活性および安定性に対してグリコシル化を必要とする。これは、可能な植物の使用が組換え抗体の産生で供給できるかの理由であり、「ヒト化」非免疫原性N−グリカンを得るためにはこれらの植物特異的成熟を阻害するのが必要になる。
hGalTおよびGNTlhGalT発現についてのプラスミドはpBLTI121から組み立てた(Pagny et al.,2003参考文献50)。CaMV 35SプロモーターはHindIII−XbaI部位においてアルファルファプラストシアニンプロモーターによって置き換えた。ヒトβ(1,4)−ガラクトシルトランスフェラーゼ
(hGalT)遺伝子(UDPガラクトース:β−N−アセチルグルコサミニド:β(1,4)−ガラクトシルトランスフェラーゼ;EC 2.4.1.22)は、EcoRI消化でpUC19−hGalTから単離した。クレノウ処理の後、1.2kb hGalT断片をSmaI部位においてpBLTI221にクローン化した。次いで、フラグタグを、FGalT順方向(5’−GACTCTAGAGCGGGAAGATGAGGCTTCGGGAGCCGCTC−3’ 配列番号92)および逆方向RGalTFlagStu(5’−AAGGCCTACGCTACTTGTCATCGTCATCTTTGTAGTCGCACGGTGTCCCGAAGTCCAC−3’配列番号93)プライマーを用いるPCRによってコーディング領域のC末端に融合させた。次いで、PlastoプロモーターおよびNosターミネーターの制御下でこのXbaI−StuI断片をバイナリベクターpBLTI121をクローニングすることによってR622を産生した。
アルファルファ(Medicago sativa L.)、エコタイプR2336は、アグロバクテリウム・ツメファシエンス(Agrobacterium tumefaciens)AGL1を用いて形質転換し、以下のように修飾した:葉柄、幹および葉外植体をアグロバクテリウムと5〜7日間共培養した。共培養工程は、SH2K培地中で0.8〜1のODおよび(1.5%スクロースの代わりの)3%スクロースにおいてアグロバクテリウムの未希釈培養で行った。応答性外植体の胚発生から得られたよく樹立された植物を温室中の土壌に移し、葉を分析した。
タンパク質は、5mLの抽出緩衝液(0.7Mサッカロース、0.5M トリス、30mM HCl、0.1M KCl、2%β−メルカプトエタノール)中の500mgの新鮮なアルファルファの葉から4℃にて30分間抽出した。不溶性物質を、4℃における10分間の間の遠心(5,000g)によって排除した。4℃にて、30分間の間に、1容量の水飽和フェノールを加えることによって得られた上清を処理する。次いで、フェノール性画分に含有されたタンパク質および糖タンパク質を、5容量のPB(メタノールに溶解させた0.1M酢酸アンモニウム)によって−20℃にて一晩沈殿させた。5mLのPBでペレットを洗浄した後、タンパク質および糖タンパク質は、Bakker et a
l.,2001に従前に記載されているように、ペプシンおよびPNGaseAでの順次の処理によって消化した。次いで、N−グリカンを2−アミノベンズアミド(2−AB)によって蛍光標識した。
実施例D:シグナル配列と融合されたZERA(登録商標)タンパク質の発現
本実施例においては、ZERA(登録商標)および配列シグナル(配列番号8)およびマンノシダーゼIを含む融合タンパク質を作成して、膜タンパク質としてのグルコシダーゼをプロテインボディーに蓄積させた。標的化シグナル(配列番号8)を用いて、酵素を膜ERに蓄積させ、ZERAペプチドを介して凝集体を形成することによってプロテインボディーの産生を可能とする。
プラスミドの構築物:ヒトマンノシダーゼI(アクセス番号Q9UKM7)に融合したZERAをコードするADNcをPCRによって増幅し、SPeIおよびSacIエンドヌクレアーゼ部位において標的シグナル(配列番号8)を含有するpBLTI121にサブクローンした。
熱ショックによって、pVKH18En6−mRFP、PBLTI121−GFPおよびpBIN20−GFP−融合をアグロバクテリウム・ツメファシエンス(株GV3101 pMP90、Koncz and Schell,1986)に導入した。トランスジェニックアグロバクテリウムを、カナマイシン(100mg.mL−1)およびゲンタマイシン(10mg.mL−1)を含有するYEB培地(リットル当たり、ビーフ抽出物5g、酵母抽出物1g、スクロース5g、MgSO4−7H2O 0.5g)上で選択し、Gomord et al.,1998に記載されているように、これを用いて、タバコ(c.v.Bright Yellow−2)BY−2細胞を形質転換した。形質転換したタバコ細胞を、PBLTI121−GFPおよびpBIN20−GFP−融合のためのカナマイシン(100mg.mL−1)、またはpVKH18En6−mRFPのためのヒグロマイシン(40mg・mL−1)、およびセフォタキシム(250mg・mL−1)の存在下で選択した。GFPおよびmRFP融合を同時発現する二重形質転換体のために、ミクロカルスをまずカナマイシンプレート上で選択し、次いで、ヒグロマイシンプレート上に移した。スクリーニング後、GFPおよび/またはmRFP融合を発現するカルスを用いて、トランスジェニック細胞の浮遊培養を開始した。3〜4日齢BY−2浮遊培養細胞を実験で用いた。

Claims (21)
- 配列番号1、配列番号4及び配列番号8からなる群から選択されたアミノ酸配列のみからなるポリペプチドであって、前記アミノ酸配列が、小胞体の膜結合形態下にて、同アミノ酸配列と融合される組換えポリペプチドを保留させるために使用される、ポリペプチド。
- 請求項1に記載のアミノ酸配列のみからなるポリペプチドと、前記アミノ酸配列がタンパク質のC末端またはN終末端に融合されている該タンパク質と、を含む組換えタンパク質。
- 前記タンパク質が酵素、抗体またはその部分、レポータータンパク質、組換えタンパク質、および治療的に活性なタンパク質よりなる群から選択される、請求項2に記載の組換えタンパク質。
- 前記タンパク質が、グリコシダーゼ、グリコシルトランスフェラーゼ、プロテアーゼ、キナーゼ、デカルボキシラーゼ、エピメラーゼ、ヌクレオチド糖トランスポーターよりなる群から選択された酵素である、請求項2に記載の組換えタンパク質。
- 請求項1記載のアミノ酸配列、または請求項3もしくは4に記載の組換えタンパク質をコードする核酸。
- 請求項5に記載の核酸を含む核酸ベクター。
- 請求項1に記載のアミノ酸配列のみからなる1つのポリペプチド、
請求項3または4に記載の1つの組換えタンパク質、および
請求項6に記載の1つの核酸ベクター、
のうちの少なくとも一つを含む植物細胞。 - 前記植物細胞が請求項3または4に記載の少なくとも1つのタンパク質を含み、該タンパク質が異種タンパク質である、請求項7に記載の植物細胞。
- 請求項1に記載のアミノ酸配列のみからなる1つのポリペプチド、
請求項3または4に記載の1つの組換えタンパク質、および
請求項6に記載の1つの核酸ベクター、
のうちの少なくとも一つを含む植物。 - 前記植物が請求項3または4に記載の少なくとも1つのタンパク質を含み、該タンパク質が異種タンパク質である、請求項9に記載の植物。
- 前記植物がアルファルファ、シロイヌナズナ(Arabidospsis thaliana)、タバコ(Nicotiana tabacum)、ダイズ(Glycine max)、アカナス(Lycopersicon esculentum)、およびトマト(Solanum lycopersicum)よりなる群から選択される、請求項9または10に記載の植物。
- 翻訳後修飾されたポリペプチドの産生方法であって、
−植物細胞を請求項6に記載の少なくとも1つの核酸ベクターでトランスフェクトまたは形質転換する工程であって、該ベクターは、翻訳後修飾される前のポリペプチドである組換えタンパク質、または該ポリペプチドとは異なる組換えタンパク質をコードする工程と、
−前記トランスフェクトされた植物細胞を成長させる工程と、
−翻訳後修飾されたポリペプチドを収穫する工程と、
からなり、
前記組換えタンパク質が前記ポリペプチドとは異なる場合、当該方法は、前記植物細胞を前記ポリペプチドをコードする少なくとも1つの核酸ベクターでトランスフェクトする工程をさらに含む、方法。 - 前記組換えタンパク質が、翻訳後修飾される前のポリペプチドである、請求項12に記載の方法。
- 前記組換えタンパク質が前記ポリペプチドとは異なり、前記組換えタンパク質が、前記ポリペプチドを認識し前記ポリペプチドに特異的に結合する抗体またはその部分である、請求項12に記載の方法。
- 前記組換えタンパク質が前記ポリペプチドとは異なり、前記組換えタンパク質が前記ポリペプチドの翻訳後修飾に関与する同種または異種酵素である、請求項12に記載の方法。
- 前記組換えタンパク質が前記ポリペプチドとは異なり、前記組換えタンパク質が、前記ポリペプチドの翻訳後修飾に関与する酵素を調節する抗体またはその部分である、請求項12に記載の方法。
- ポリペプチドが貯蔵タンパク質と同時発現される、請求項12〜16のいずれか一項に記載の方法。
- 前記ペプチドの翻訳後修飾が、小胞体(ER)およびゴルジ装置(GA)の少なくとも一方のコンパートメント膜において行われる、請求項12〜17のいずれか一項に記載の方法。
- 前記翻訳後修飾されたポリペプチドが治療的に活性なタンパク質である、請求項12〜18のいずれか一項に記載の方法。
- 前記植物細胞が、アルファルファ(Medicago sativa)、シロイヌナズナ(Arabidospsis thaliana)、タバコ(Nicotiana tabacum)、ダイズ(Glycine max)、アカナス(Lycopersicon esculentum)、およびトマト(Solanum lycopersicum)よりなる群から選択される植物に関連する細胞である、請求項12〜19のいずれか一項に記載の方法。
- 前記組換えポリペプチドが膜タンパク質であり、かつ前記アミノ酸配列が前記膜タンパク質を前記小胞体に固定させる、請求項1に記載のポリペプチド。
