Description
以下の物質および方法が、本実施例において示される結果を得るのに使用された。
SR-A欠損マウス(非特許文献7)はC57BL/6Jマウス(非特許文献11)と戻し交配され、M.フリーマン(ハーバード・メディカル・スクール)およびB.ベルヴィン(ダートマス・メディカル・スクール)によって提供された(非特許文献5および11)。野生型(WT)C57BL/6マウスはジャクソン研究所(メイン州、バール・ハーバー)から購入した。マウスはロズウェル・パーク癌研究所の特定病原体除去施設内で維持された。動物のケアと実験は、施設および国立衛生研究所(NIH)のガイドラインに従って行われ、施設の実験動物委員会によって承認された。B16(F10)細胞(H-2b)、ATCCおよびD121細胞株(H-2b)由来の自然発症マウスメラノーマ、我々の研究所のS.フェルローネによって提供されたルイス肺癌の亜系統は、DMEM中で維持され、10%の熱失活ウシ胎児血清(ニューヨーク州、グランドアイランド、ライフテクノロジー社)、2mMのL-グルタミン、100U/mlのペニシリン、および100μg/mlのストレプトマイシンが追加された。
腫瘍細胞は、137CS-照射器中で100Gyでイオン化照射(IR)によって処理されるか、UV光(カリフォルニア州、ラ・ホーヤ、ストラタジーン社、ストラタリンカー1800)に5分間曝された。細胞はその後洗浄され、PBS中に1×107細胞/mlにて再懸濁された。細胞溶解物の調製のために、腫瘍細胞はPBS中で懸濁され、4サイクルの急速な凍結/融解曝露に供され、細胞片(デブリ)を除去するために4℃にて10分間12000rpmで回転された。
腫瘍チャレンジ(tumor challenge)の研究のために、マウス(1グループ当たり5匹のマウス)は、1×106の照射腫瘍細胞による左側腹部の皮下注射で免疫された。いくつかのケースでは、一週間後に2度目の追加免疫が行われた。免疫化の7日後、マウスは、右側腹部への生B16(マウス当たり2×105細胞)またはD121腫瘍細胞(マウス当たり4×105細胞)の皮下注射によってチャレンジされた。治療の研究のために、マウスは、0日目に2×105のD121腫瘍細胞またはB16腫瘍細胞を接種され、その後、2,4,6および8日目に照射腫瘍細胞を用いて治療された。腫瘍の増殖は一日おきにモニターされた。腫瘍体積(腫瘍量)は式[V=(最短径2×最長径)/2]を使用して計算された。
すでに記述されているようにELISPOTアッセイを用いて、腫瘍特異的もしくは抗原特異的IFN-γを分泌するT細胞を測定するために、免疫されたマウスまたは腫瘍のないマウスから脾細胞を単離した(非特許文献12)。手短に説明すると、ろ過プレート(マサチューセッツ州、ベッドフォード、ミリポア社)は4℃にて一晩、10μg/mlのラット抗マウスINF-γ抗体(カリフォルニア州、サンディエゴ、ファーミンゲン社、クローンR4-6A2)でコートされた。プレートはその後洗浄され、10%のFBSを含有する培養液でブロックされた。脾細胞(1×106/ウェル)は、5μg/mlのH-2Kb制限CTLエピトープ TRP2180-188(SVYDFFVWL)(非特許文献13)もしくは H-2Db制限CTLエピトープ gp10025-32(EGSRNQDWL)(非特許文献14)とともに、10U/mlのIL-2の存在下で37℃にて24時間インキュベートされた。いくつかのケースで、照射B16もしくはD121細胞(脾細胞:腫瘍細胞=20:1)が刺激剤として使用された。プレートはその後十分に洗浄され、5μg/mlのビオチン化INF-γ抗体(カリフォルニア州、サンディエゴ、ファーミンゲン社、クローンXMG1.2)とともに4℃にて一晩インキュベートされた。洗浄後、0.2U/mlのアビジン-アルカリホスファターゼD(カリフォルニア州、バーリンゲーム、ベクターラボラトリー社)が添加され、室温にて2時間インキュベートされた。スポットは、5-ブロモ-4-クロロ-3-インドリルホスファターゼ/ニトロ ブルー テトラゾリウム(インディアナ州、インディアナポリス、ベーリンガー・マンハイム社)を添加することによって展開され、室温にて20分間インキュベートされた。スポットはELISPOTカウンター(ドイツ、カールツァイス社)を用いてカウントされた。
CD4+、CD8+T細胞サブセットの除去(欠乏)は、免疫化前の6日間に一日おきにそれぞれ投与された200μgのGK1.5および2.43mAb腹腔内注射によって達成された。細胞サブセットの有効な除去は、ワクチン接種の1日前の脾細胞のFACS解析によって確認され、実験期間中、1週間に2度の抗体注射によって維持された。アイソタイプ-マッチド(isotype-matched)抗体はコントロールとして使用された。食細胞の機能阻害のために、200μlのPBS中の1mgのカラゲナン(シグマ社、typeII)が、記述されているように腹腔内注射によって投与された(非特許文献15)。
マウスは3%のチオグリコール酸ブロスを腹腔内に注射され、誘発されたマクロファージは4日後に腹膜灌流によって収集された。マクロファージは10%ウシ胎児血清を含有するダルベッコ変法イーグル培地中で一晩培養され、非付着性細胞は洗浄により除去された。このように調製されたマクロファージはフローサイトメトリーによりCD11b(>96%)に対しルーチン的に陽染した。UV処理された腫瘍細胞は、PBS中で37℃にて5分間、2nMの5(および6)-カルボキシフルオセイン二酢酸サクシニミジルエステル(オレゴン州、ユージーン、モレキュラー・プローブ社、CFSE)で標識された。非結合色素は、等容積のウシ胎児血清とともに37℃にて30分間インキュベートすることによって消光された。細胞は完全培地で洗浄され、チオグリコール酸-誘発マクロファージとともに、2:1の比で4時間共培養された。浮遊細胞は洗い流され、付着マクロファージは収集され、CD11b-PE抗体(カリフォルニア州、サンディエゴ、ファーミンゲン社)で染色された。マクロファージによる貪食は、二重陽性染色細胞のパーセンテージとしてB-D FACScaliber(ベクトン・ディッキンソン社)を用いたFACSによって数量化された。
腫瘍の増殖はスチューデントt検定を用いて解析された。腫瘍のないマウスは、ログランク統計解析によって比較された。p<0.05の値が有意と見なされた。
前述の物質と方法を使用することによって、以下の結果が得られた。
SR-A欠乏が腫瘍免疫原性に影響を及ぼすかどうかを判定するために、低免疫原性且つ高転移性の腫瘍である、D121ルイス肺癌細胞(非特許文献16および17)が使用された。野生型(WT)C57BL/6およびSR-A-/-マウスはどちらも、イオン化照射(IR)処理D121腫瘍細胞で免疫され、一週間後に生存腫瘍細胞を用いたチャレンジを続けた。予想通り、IR-D121腫瘍細胞で免疫されたまたは免疫されないWTマウスは、腫瘍チャレンジ(誘発)により急激に増殖する腫瘍を発生した(図1の上のパネル)。非免疫SR-A-/-マウスに接種されたD121腫瘍は野生型マウスにおけるのと同様に増殖するのに対し(図1の下のパネル)、単一用量の照射D121腫瘍細胞によるワクチン接種は、その後の腫瘍チャレンジに対して、SR-A-/-マウスを完全に保護することができた。腫瘍のないSR-A-/-マウスは、8ヶ月後でさえ、第二の腫瘍チャレンジに抵抗性であり、長期の免疫記憶の存在を示唆した(データは示さない)。増強された腫瘍-防御免疫の普遍性は、他の弱免疫原性腫瘍であり、異なる組織学的な起源に由来する、B16(F10)メラノーマにおいて確認された(データは示さない)。さらに、UV-処理B16腫瘍細胞による単一用量のワクチン接種は、SR-A-/-でのこの予防的な環境において腫瘍拒絶をもたらし、WTマウスではもたらさなかった(図2)。これは、放射線源が処理腫瘍細胞の免疫原性に影響しないことを示唆している。腫瘍溶解物によるSR-A-/-マウスの免疫化も、SR-A-/-マウスにおける腫瘍の増殖を有意に減少させたが、全ての動物は最終的に腫瘍が発達した(図2)。
D121腫瘍細胞の拒絶における免疫エフェクター細胞の関与が、インビボにおける抗体欠乏研究によって調査された。マウスは、免疫化の前に、抗-CD4 Ab GK1.5もしくは抗-CD8 Ab 2.43による処理によって、CD4+もしくはCD8+T細胞サブセットが枯渇した。枯渇(欠乏)の完全性は98%を超えることが、脾臓およびリンパ節集団のFACS解析によって評価された(データは示さない)。マウスはその後、4×105のD121腫瘍細胞でチャレンジされた(図3)。CD8+T細胞の欠乏は、腫瘍防御免疫を完全に抑制し(p=0.002、vs IgG処置グループ)、他方、CD4+T細胞の欠乏は、D121腫瘍の拒絶に影響を与えなかった(p>0.05、vs IgG処置グループ)。カラゲナン(非特許文献15)もプライミング期間中に食細胞を枯渇させるために使用された。食細胞の欠乏も腫瘍防御効果を減少することが観察された(p=0.002、vs IgG処置グループ)。
B16メラノーマは、それがgp100およびTRP-2を含む、複数のメラノーマ関連抗原を発現するため(非特許文献18)、内在性腫瘍抗原に特異的な免疫応答を評価するための関連モデルとして使用された。照射B16腫瘍細胞による免疫化後、脾細胞はWTもしくはSR-A-/-マウスから単離され、CTLエピトープ gp10025-32またはTRP2180-188で刺激された。ELISPOTアッセイは、照射B16細胞で免疫されたSR-A-/-動物の脾細胞が、非免疫マウスまたは免疫WTマウスの脾細胞と比較して、強い抗原特異的IFN-γ産生を示すことを明らかにした(図4)。さらに、免疫SR-A-/-マウスの脾細胞はまた、照射B16細胞でインビトロで刺激された際、高レベルのIFN-γを産生するが、D121細胞では産生せず、刺激を受けたCTLsの腫瘍特異性を示唆した。
アポトーシス細胞貪食の機能障害は、自己寛容の破綻をもたらす可能性があり(非特許文献19〜21)、SR-Aはアポトーシス細胞のクリアランスに関連している(非特許文献22)。我々はSR-A-/-およびWTマウス由来のマクロファージの食作用能を比較した。貪食は、CFSEも含有したCD11b+Mφを検出することによってFACscan解析で測定された。食細胞の取り込みの定量化は、両方のマウスに由来するMφが瀕死の腫瘍細胞を効率的に貪食することを示した(p>0.05)(図5)。結果はさらに、細胞を蛍光顕微鏡で可視化することによって確認され(データは示さない)、瀕死細胞のクリアランスに対する、APCs上の重複性のレセプターの存在を示唆している(非特許文献23)。
予防免疫が、SR-A-/-マウスにおいて腫瘍拒絶をもたらすという事実を考慮して、我々は、担癌マウスにおけるワクチン接種の治療効果を特定した。SR-A-/-マウスは、まず0日目にD121腫瘍細胞を樹立され、その後、2,4,6,8日目に照射D121腫瘍細胞で処理された。無処理のSR-A-/-マウスでは、D121腫瘍は著しく成長した。しかしながら、照射D121細胞の投与は、腫瘍成長速度の著しい低下をもたらし、マウスの50%が腫瘍なしのままであった(図6、p<0.05 vs 無処理群)。同様の治療効果がB16メラノーマモデルでも見られた(図6)。
重要なことに、我々はDCsにおいてSR-Aを特異的に阻害することが、所望の抗原に対する哺乳類の免疫応答を増強できることを実証した。
Claims (6)
- 個体において、所望の抗原への免疫応答を増強するための組成物であって、
特異的に阻害されたクラスAマクロファージスカベンジャー受容体(SR-A)を有する樹状細胞を含むこと、および
前記所望の抗原が腫瘍抗原であり、前記クラスAマクロファージスカベンジャー受容体がshRNAによって特異的に阻害されていること
を特徴とする組成物。 - 前記樹状細胞が前記所望の抗原に曝露されたものである、請求項1に記載の組成物。
- 前記所望の抗原を発現する腫瘍の増殖を抑制するために投与される、請求項1または2に記載の組成物。
- 前記樹状細胞が、前記個体から単離されたものであり、且つ、前記所望の抗原に曝露されたものである、請求項1〜3のいずれか1項に記載の組成物。
- 個体において、所望の抗原への免疫応答を増強するための組成物であって、
前記所望の抗原と、クラスAマクロファージスカベンジャー受容体(SR-A)を特異的に阻害する能力を有する薬剤とを含むこと、および、
前記所望の抗原が腫瘍抗原であり、前記薬剤がshRNAを含むこと
を特徴とする組成物。 - 腫瘍と診断された個体において、前記腫瘍によって発現された抗原への免疫応答を増強することによって、前記腫瘍の増殖を抑制する組成物であって、
クラスAマクロファージスカベンジャー受容体(SR-A)を阻害する能力を有する薬剤と、前記腫瘍抗原とを含むこと、および
前記薬剤がshRNAを含むこと
を特徴とする組成物。
