Description
本明細書で使用される際、用語「STIMタンパク質」は、STIM1タンパク質、STIM2タンパク質、又はSTIM1とSTIM2タンパク質の両方を意味する。STIMタンパク質の例は、全て哺乳類のSTIMタンパク質を含み、例えば、ヒトSTIM1タンパク質(Genbankタンパク質受入番号Q13586、NP-003147、AAC51627)、ヒトSTIM2(Genbankタンパク質受入番号Q9P246、NP-065911、AAK82337)、マウスSTIM1(Genbankタンパク質受入番号NP-033313)、及びマウスSTIM2タンパク質(Genbankタンパク質受入番号P83093、NP-001074572、AAK82339、CAN36430)が挙げられる。
STIMタンパク質は、細胞質ドメインから細胞外部分を分離する単一膜貫通領域を備えるI型膜貫通型リンタンパク質である。脊椎動物は2種のSTIMタンパク質であるSTIM1及びSTIM2を有する一方で、無脊椎動物は、1つの遺伝型のみ、例えばキイロショウジョウバエ(Drosophila melanogaster)ではD−STIMを有する。脊椎動物と無脊椎動物のSTIMタンパク質を比較すると、保存されたゲノム構築が示され、脊椎動物で発見される2つのSTIM遺伝子は、1つの先祖遺伝子から発生したと推定されることが示されている(Williams et al. (2001) Biochem. J. 357: 673-685)。

Ca2+放出活性化カルシウム(CRAC:Ca2+ release-activated calcium)チャネルを経由したストア作動性Ca2+流入は、刺激を受けた免疫細胞内における細胞内Ca2+増加の主要な機構である(Parekh, A.B. & Putney, J.W., Physiol. Rev. 85, 757-810 (2005))。CRACチャネルは、小胞体(ER;endoplasmic reticulum)Ca2+ストアがIP3受容体に結合するイノシトール三リン酸(IP3)によって枯渇した後、開く。持続Ca2+流入は、T細胞分化及びサイトカイン発現を含む免疫細胞の多様な機能を引き起こす(Lewis, R.S., Annu. Rev. Immunol. 19, 497-521 (2001); Feske, S.,et. al., Biochem. Biophys. Res. Commun. 311, 1117-1132 (2003); Gallo, E.M., et. al., Nature Immunol. 7, 25-32 (2006))。ショウジョウバエ(Drosophila)におけるゲノム全体のRNAiスクリーニングによって、CRACチャネル活性を制御する2つのキー分子が同定された。これら2つのキー分子とは、ERのCa2+センサーであるStim(Roos, J. et al., J. U S A 103, 9357-9362 (2006))である。ショウジョウバエStim及びその哺乳類相同体Stim1とStim2(Roos, J. et al., J. Cell Biol. 169, 435-445 (2005); Liou, J. et al., Curr. Biol. 15, 1235-1241 (2005))は、ER内腔内に配されたCa2+結合EFハンドを通ってERのCa2+レベルを感知すると考えられている1回膜貫通タンパク質である(Taylor, C.W., Trends Biochem. Sci. 31, 597-601 (2006); Lewis, R.S., Nature 446, 284-287 (2007); Putney, J.W., Jr., Cell Calcium (2007))。Stim1は、ストア作動性Ca2+流入における確立した正の調節因子である(Zhang, S.L. et al., Nature 437, 902-905 (2005); Spassova, M.A. et al., Proc. Natl Acad. Sci. U S A 103, 4040-4045 (2006); Luik, R.M., et. al., J. Cell Biol. 174, 815-825 (2006); Wu, M.M., et. al., J. Cell Biol. 174, 803-813 (2006); Baba, Y. et al. Proc. Natl Acad. Sci. U S A 103, 16704-16709 (2006))が、Stim2の機能は議論中である(Roos, J. et al., J. Cell Biol. 169, 435-445 (2005); Liou, J. et al., Curr. Biol. 15, 1235-1241 (2005); Soboloff, J. et al., Curr. Biol. 16, 1465-1470 (2006); Soboloff, J. et al., J. Biol. Chem. 281, 20661-20665 (2006))。Stim1とStim2の生理的役割を調査するために、変異マウスが、Stim1とStim遺伝子のコンディショナルな欠失により作製された。ここで、本発明者らは、Stim1が、ナイーブT細胞及びマウス胎児線維芽細胞(MEFs;mouse embryonic fibroblasts)においてストア作動性Ca2+流入の主要エフェクターであり、その欠損は、T細胞サイトカインの発現を著しく損なうことになることを示す。対照的に、Stim2は、ナイーブT細胞におけるストア作動性Ca2+流入に対して影響が殆どないものの、MEFsにおけるストア作動性Ca2+流入、及び分化したT細胞による、もしくは部分的にNFAT核局在化の後期を持続することによるサイトカイン発現に対して大きな要因となる。このようにして、Stim1及びStim2は、いずれも、分化したT細胞においてCa2+依存性サイトカイン発現の正の調節因子である。より大量なStim1が反応開始に必須であるが、適量のStim2がStim1の機能を増強するのに重大な役割を有する。
(コンディショナルな遺伝子ターゲティング)
Stim1及びStim2遺伝子の遺伝子ターゲティングが、既述の如く(Muljo, S.A. et al. J. Exp. Med. 202, 261-269 (2005))、C57BL/6マウス由来のBruce−4ES細胞における相同的組換えによって実施された。ターゲティングされたStim対立遺伝子を有するキメラマウスは、ヘテロ接合体Stim1neo/+又はStim2neo/+ES細胞クローンの胚盤胞注入(blastcyst injection)によって作製された(図9参照、ネオ=ネオマイシン耐性遺伝子)。Stim1−/−又はStim2−/−マウスは、CMV−Cre(Creディレーター(deleter))トランスジェニックマウスと交配させた後、ファウンダーStim1neo/+マウスの後代を交雑させることによって作製された(Schwenk, F., et. al. Nucleic Acids Res 23, 5080-5081 (1995))。Creトランスジーンを含まずにStim1+/−又はStim2+/−マウスを構築するために、Stim+/−CMV−Cre+マウスをC57BL/6マウスと交配させた。コンディショナルなStim1fl/+又はStim2fl/+対立遺伝子を作成するために、ファウンダーStimneo/+キメラマウスをFlpディレータートランスジェニックマウスと交配させ(Rodriguez, C.I. et al., Nat Genet 25, 139-140 (2000))、これによりターゲティングされたStim対立遺伝子からネオマイシン耐性カセットを取り除いた。Rag1−/−及びB6.Cg(Igha,Thy1.1、Gpi1a)マウスがJackson laboratoryから購入された。Stim遺伝子のT細胞特異的破壊を有するマウスを作製するために、CD4−Creトランスジェニックマウス(Lee, P.P. et al. Immunity 15, 763-774 (2001))を各ファウンダーStimfl/+マウスと交配させ、後代を交雑させた。全てのマウスは、ハーバード・メディカル・スクールにおいて特異的無菌バリア設備内に維持され、ハーバード・メディカル・スクールにおける動物資源及び比較医学センターによって承認された手続きに基づき、使用された。
脾臓及びリンパ節からのCD4+T細胞の精製、TH分化の導入、10nMのPMA及び1μMイオノマイシン、又はプレート結合抗CD3と抗CD28を用いた刺激、及び細胞内染色及びフローサイトメトリー分析によるサイトカイン産生の評価が、既述の如く実施された(Ansel, K.M. et al., Nature Immunol. 5, 1251-1259 (2004))。Foxp3発現は、抗Foxp3(eBioscience)を用いて、製造業者のプロトコルに基づき細胞内染色で評価され、フローサイトメトリーで分析された。レトロウイルス形質導入が、既述の如く(Wu, Y. et al., Cell 126, 375-387 (2006))、KMVレトロウイルス発現プラスミドを用い、空の又は含有Stim1又はStim2cDNAのいずれか、その後にGFPのcDNAで、内部リボソーム侵入部位(IRES;internal ribosome entry site)の支配下で実施された。これらのCa2+流入欠損にもかかわらず、STIM1欠損T細胞は、通常通り、CD25(IL−2R−α鎖)を上方制御した。したがって、分化培養物がIL−2で維持されていたために、略同等数の細胞が回復した(7日間、分化したSTIM欠損細胞の全細胞数は野生型の細胞数の60−70%であった)。そして、STIM欠損細胞は、野生型細胞と同じ効率性で、レトロウイルス形質導入されることが可能であった。
Stim1−/−及びStim2−/−マウス胎児線維芽細胞(MEFs)は、Stim1+/−又はStim2+/−マウスのいずれかを交雑させることによって得られるE14.5胚から、標準プロトコルを用いて構築された。MEFsは、ハイグロマイシン耐性遺伝子を運ぶプラスミド内にSV40ラージT抗原でレトロウイルス形質導入されることによって不死化され、続いてハイグロマイシン選択された。
細胞をPBS内で再懸濁することによって、細胞抽出物が調製された。その後、それらを、50mMのNaCl、50mMのTris−HCl(pH6.8)、2%SDS、10%グリセロール(最終濃度)を含む緩衝液で溶解した。タンパク質濃度が、BCAタンパク質試薬キット(Pierce)を用いて決定され、その後、2−メルカプトエタノールが最終濃度100μMへと追加され、サンプルは煮沸された。ウエスタンブロットが標準プロトコルに基づき実施された。STIM1ポリクローナル抗体は、ヒトSTIM1(CDNGSIGEETDSSPGRKKFPLKIFKKPLKK-COOH, (SEQ. ID. No.1、N末端のシステインが、ペプチドを担体タンパク質と結合させることを目的として導入されている箇所)のC末端ペプチドに対して生成され(Open Biosciences)、1:2000で使用された。アフィニティー精製ポリクローナル抗体は、ヒトSTIM2(CKPSKIKSLFKKKSK, (SEQ. ID. No. 2)、N末端のシステインが、ペプチドを担体タンパク質と結合させることを目的として導入されている箇所)のC末端ペプチドに対して生成され、2μg/mlで使用された。アクチンに対するポリクローナル抗体(I-19; SC-1616; Santa Cruz)は、1:500で使用された。mycエピトープ標識に対するモノクローナル抗体は、9E10ハイブリドーマ細胞株の上澄みから精製された。FACS分析に用いられる全ての下記の蛍光共役抗体は、eBioscience又はBD Pharmingenから購入した。Pacific Blue-CD4 (RM4-5), FITC-CD8 (53-6.7), FITC-Thy1.1 (HIS51), FITC-IgE (R35-72), FITC-CD11c (M1/70), PE-IL-2 (JES6-5HA), PE-IL-4 (11B11), PE-Foxp3 (FJK-16s), PE-CD19 (1D3), PE-CD125 (T21.2), PE-CD44 (IM7), PE-CD95 (Jo2), PerCP-B220 (RA3-6B2), PsrCPCy5.5-CD4 (RM4-5), PECy7-Thy1.2 (53-2.1), APC-IFNγ (XMG1.2), APC-IL-10 (JES5-16E3), APC-TNFα (MP6-XT22), APC-CD25 (PC61.5), APC-TCRb (H57-597), APC-CD38 (90), APC-CD62L (MEL-14), bio-CD5 (53-7.3), bio-CD69 (H1.2F3) 及び PE-ストレプトアビジン。
CD4+T細胞は、既述の如く(Ansel, K.M. et al., Nature Immunol. 5, 1251-1259 (2004))、分離され、一晩ローディング培地(RPMI1640 10% FBS)で培養され、1μMのfura−2/AM(Invitrogen)を、30分間22−25℃且つ1×106細胞/ml濃度でローディングした。測定前に、T細胞を、ポリ−L−リジンがコーティングされたカバースリップに15分間付着させた。抗CD3を刺激するために、T細胞は、5μg/mlのビオチン共役抗CD3(クローン2C11、BD pharmingen)とともに、15分間22−25℃で培養された。そして、抗CD3架橋結合が、10μg/mlのストレプトアビジン(Pierce)を用いて細胞をかん流することによって実現された。長期間のCa2+イメージングのために、分化したCD4+T細胞が、1μMのfura−PE3でローディングされた後、10nMのPMA及び0.5μMのイオノマイシンで、2%ウシ胎仔血清が補充された2mMCa2+を含むリンゲル液中で刺激された。画像を獲得する間、細胞は、37℃に暖められた緩衝液で絶えずかん流された。[Ca2+]iの測定は、既述された如く(Feske, S. et al., Nature 441, 179-185 (2006))、実施及び分析された。Ca2+流入速度は、0.2乃至2mM細胞外Ca2+において、細胞内Ca2+濃度(d[Ca2+]i/dt)内の初期上昇の最大速度によって推論され、割合「d[Ca2+]i/dt」として表現される。ここで、d[Ca2+]iとは、細胞外Ca2の再追加とCa2流入反応のピークとの間の20秒の時間間隔(dt)にわたる[Ca2+]iの極大差である。各実験では、100−150の個別T細胞又は少なくとも30の個別MEFsが、Igor Pro分析ソフトウェア(Wavemetrics)を用いて、340/380の比率で分析された。
CD4+T細胞は、非分極条件下で培養され、5日後に採取された。そして、表示時間の間96ウェルプレートにおける200μl内1×105細胞/ウェルで、10nMのPMAと1μMイオノマイシンを用いて、様々な時間刺激された。そして、149×Gで3分間遠心分離することによって、384ウェルプレート(5000−8000細胞/ウェル;3ウェル/サンプル)内のポリ−L−リジン−コーティングされたウェルに付着させた。細胞は、3%(vol/vol)のパラホルムアルデヒドに固定された後、抗−NFAT1(NFAT1の67.1ペプチドに対する精製ウサギポリクローナル抗体)(Ho, A.M.,et. al., J. Biol. Chem. 269, 28181-28186 (1994))、及びインドカルボシアニン共役抗ウサギ二次抗体で染色された。そして、DNA挿入色素DAPI(4,6−ジアミジノ−2−フェニルインドール)で対比染色された。画像は、20倍対物レンズを備えるImageXpress Micro 自動イメージングシステム(Molecular Devices)を用いて取得され、MetaXpressソフトウェアバージョン6.1(Molecular Devices)の移行アプリケーションモジュールを用いて分析された。核移行への細胞質性は、抗NFAT1−Cy3染色とDAPIとの間の強度相関関係を算出することによって評価された。T細胞は、NFAT1−インドカルボシアニン染色の>90%が、DNA挿入色素DAPIからの蛍光信号と関係がある場合に、核NFAT1を有しているとして得点付けされた。各データポイントは、ウェルあたり少なくとも300の個別細胞の平均を示す。
0.6×106のHEK293細胞は、一晩、6ウェルプレート上にプレーティングされ、次の日、製造業者のプロトコルに基づき、リポフェクタミン2000トランスフェクション試薬(Invitrogen, Carlsbad, CA)を用いて、ヒトSTIM1及びSTIM2に対するsiRNAs(Dharmacon, Inc., Lafayette, CO)がトランスフェクトされた。トランスフェクションの手順は、48時間後に繰り返されることにより、ノックダウンの効率を増大させた。細胞は、72時間後に、免疫ブロット分析又は[Ca2+]i測定のために採取された。ノックダウンの程度を監視するために、STIM1の免疫ブロット法が、ヒトSTIM1に対するポリクローナル抗体(M. Dziadek から親切にも贈与されたもの(U Auckland, New Zealand))を1:1000の希釈において用いた第2のトランスフェクションの後、72時間後に実施された。STIM2の転写レベルは、スクランブルコントロールsiRNA (siC2)、siSTIM1、及びsiSTIM2がトランスフェクトされた細胞において、リアルタイムRT−PCRによって測定された。STIM2の閾値サイクル(CT)は、GAPDHハウスキーピング遺伝子発現レベル(ΔCT)に標準化され、0.5ΔCt*106(任意ユニット)としてプロットされた。ここで用いられるsiRNA配列は、STIM1: AGGUGGAGGUGCAAUAUUA (SEQ. ID. No. 3) ; STIM2#1: UAAACCUCCUGGAUCAUUA (SEQ. ID. No. 4); STIM2#2: CUUUAAGCCUCGAGAUAUA (SEQ. ID. No. 5)であった。
CD4+T細胞は、非極性条件下で培養され、5日後に採取された。パッチクランプ記録は、ITC−18入力/出力ボード(Instrutech)及びiMacのG5コンピュータに連動するAxopatch 200増幅器(Axon Instruments)を用いて実施された。電流は、1kHzで4極ベッセルフィルタを用いてフィルターされ、5kHzでサンプリングされた。記録電極は、100μlピペットから引き出され、シルガード(Sylgard)でコーティングされ、熱研磨され、最終抵抗2−5MΩであった。刺激及びデータ取得と分析は、Igor Pro platform (Wavemetrics)において開発された社内ルーティーンを用いて実施された。全データは、バス内のリンゲル液(10mV)と比較したピペット液の液間電位、及び20mM[Ca2+]O+25μMのLa3+に集められたリーク電流について修正された。標準細胞外リンゲル液が含有したのは(mMあたり)、:130NaCl、4.5KCl、20CaCl2、1MgCl22、10D−グルコース、及び5Na−Hepes(pH7.4)であった。ある実験では、2mMのCaCl2が標準細胞外溶液に使用され、NaCl濃度は150mMまで上がった。標準二価遊離(DVF:divalent-free)リンゲル液が含有したのは(mMあたり)、150NaCl、10mM四酢酸、1mMのEDTA、及び10mMのHepes(pH 7.4)であった。25nMのカリブドトキシン(Sigma)が全ての細胞外溶液に添加されることにより、Kv1.3チャネルからの汚染を排除した。標準内部溶液が含有したのは(mMあたり)、145mMのCsアスパラギン酸、8mMのMgCl2、10BAPTA(1,2−ビス(o-アミノフェノキシ)エタン−N,N,N',N'−四酢酸) 及び10mMのCs−Hepes(pH 7.2)であった。平均結果とは、平均値±標準誤差を示す。曲線当てはめは、Igor Pro 5.0において組込関数を用いた最小二乗法を用いて実施した。Na+の浸透性に対するCs+の浸透性は、以下の関係[数1]を用いた生体工学の逆転電位から計算された。
組織は、3−4ヶ月齢のマウスから採取され、10%ホルマリンで固定された。ヘマトキシリン及びエオシン染色が、標準手順に基づき実施された。
Thy1.1+コンジェニックマウス由来のCD4+CD25+及びCD4+CD25−T細胞は、CD4+T細胞がDynabeadsマウスCD4及びDETACHaBEADマウスCD4(Invitrogen)によって精製された後、FACSVantageによって選別された。野生型Thy1.1+、CD4+CD25+、又はCD4+CD25−T細胞(3×105)が、2週齢のマウスの腹腔内に注入された。注入されたマウスは、養子移植後の8週間後に分析された。
Thy1.2+DKOマウス(3×106細胞)からのT細胞枯渇骨髄細胞は、Thy1.1+DKOコンジェニック野生型マウス(1.5×106細胞)からのT細胞枯渇骨髄細胞と混合され、後眼窩洞を介して、亜致死的放射を受けたRag1−/−マウス(450ラド)内に注入された。再構築されたマウスは、骨髄移植後10−12週間後に分析された。
CD4+CD25−及びCD4+CD25+T細胞は、CD4+T細胞増殖後に、MACS CD25 マイクロビーズ(Miltenyi Biotec)を用いて正に選択された。精製されたCD4+CD25−T細胞(2×107/ml)は、CFSE(カルボキシフルオセイン二酢酸サクシニミジルジエステル)(1.25μM)を用いて10分間37℃で培養された。細胞は、72時間、抗CD3及び抗CD28で刺激され、細胞分裂数がフローサイトメトリーで評価された。インビトロ抑制アッセイは、CFSE標識CD4+CD25−T細胞(5×104)と、表示割合のCD4+CD25+T細胞とを共培養することにより実施された。このCD4+CD25+T細胞は、コントロール同腹子又はDKOマウスから精製されたものである。この共培養は、丸底型プレートにおいて、37℃で72時間、マイトマイシンC処理されたT細胞枯渇脾細胞(5×104)と0.3μg/mlの抗CD3(2C11)存在下で実施された。
STIM1及びSTIM2の両方がインビボで普遍的に発現する(Williams, R.T. et al., Biochem. J. 357, 673-685 (2001))ため、Stim1及びStim2のloxP-で挟まれた対立遺伝子を有するマウスが作製された(図9)。これらのマウスを、CMV−Creディレーター株と交配させる(Schwenk, F., Baron, U. & Rajewsky, K. Nucleic Acids Res 23, 5080-5081 (1995))ことにより、全組織でStim1とStim2欠損の影響を調べた。C57BL/6のバックグラウンドにおけるSTIM1欠損マウスは、E18.5において予想されるメンデル比で生存していたが、周産期致死性を示した。これは、75%が死産であり、残りの子も2日以内に死亡した。対照的に、STIM2が欠如したマウスは、産後4週間まで生存したが、僅かな発達遅延を示し、4−5週齢で死亡した(表1a及び1b)。STIM1欠損マウスの周産期致死性を「レスキュー」するために、これらのマウスを、非近交系ICRマウス株と交配させた。周産期致死性はまだ高い(38%)ものの、非近交系STIM1欠損子孫の半分は、重度の発達遅延を伴い2日生存し、次の2週間以内に原因不明で死亡した(表1c)。
STIM1欠損CD4+T細胞は、筋小胞体ATPアーゼ(SERCA)ポンプのインヒビターであるタプシガルギン(TG)を用いたERのCa2+ストアの受動枯渇後、又は抗CD3とのTCR架橋結合後、殆どCa2+流入を示さなかった(図1a)。静止コントロール及びSTIM1欠損T細胞は、抗CD3、又は抗CD3及び抗CD28での刺激を受けた際に、表面CD3及びTCRβの類似発現を示し、ERのCa2+ストアの類似枯渇と活性化マーカーCD25とCD69の発現を示した(図10a−c)。STIM1欠損CD4+T細胞は、PMAとイオマイシン(図1b)又は抗CD3及び抗CD28(図10d)による刺激後、IL2の産生に失敗した。全体として、これらの結果は、STIM1が初代マウスT細胞においてストア作動性Ca2+流入及びCa2+依存性サイトカイン産生を制御するという最初の遺伝学的証拠を提供する。
STIM2欠損T細胞で観察された相対的にかなり低いサイトカイン発現によるストア作動性Ca2+流入の少量の減少を調整するために、Ca2+流入が、細胞質内によく保持されたカルシウム指示薬であるFuraPE−3を備える細胞をローディングすることによって、より長い時間スケールにおいて検査された(Vorndran, C., et. al., Biophys J 69, 2112-2124 (1995))。STIM2欠損T細胞は、野生型T細胞と比較して減少したストア作動性Ca2+流入を示し、20分後、維持細胞内遊離Ca2+濃度([Ca2+]i)のより低い横ばい状態を維持した(図3a)。Ca2+シグナル伝達がSTIM2欠損細胞において減少していることを確認するために、Ca2+依存性転写因子NFAT18の核移行が監視された。核移行は、MetaXpressプログラム(データ示さず)を用いて定量化され、野生型、STIM1欠損及びSTIM2欠損マウスからのヘルパーT細胞を、非分極条件下、その後PMA及びイオノマイシンを用いた刺激した条件下で(サイトカインアッセイに用いられるものと同じ条件)、1週間分化させた。これにより、NFAT1核移行(3b、c)及びサイトカイン発現(図3d、e)が直接的に比較可能となった。
全細胞パッチクランプ記録法は、STIM1及びSTIM2の欠失がCRAC電流(ICRAC)に影響を与えるか否かを決定するために用いられた。タプシガルギン(TG)によるERのCa2+ストア枯渇に応答して、コントロールマウスCD4+T細胞は、ヒトT細胞におけるICRACの特性に類似した特性を有するCa2+電流を示した(図4)。これらの特性は、20mMのCa2+(図4a)存在下における非常にポジティブな逆転電位を備える内向き整流性の電流−電圧関係、20mMCa2+における高速不活性化(図12c)、二価陽イオン遊離(DVF)溶液におけるNa+電流の非増強作用(図4b)、低いCs+透過性(Cs+透過性/Na+透過性=0.2±0.04;n=14サンプル)、細胞外Ca2+のマイクロモル濃度によるNa+電流の遮断(図12b)、及び低及び高濃度の2−ホウ酸アミノエトキシジフェニルによる相乗作用と阻害(Prakriya, M. & Lewis, R.S., J. Physiol. 536, 3-19 (2001))を含んでいた(とはいっても、マウス細胞における2−ホウ酸アミノエトキシジフェニルによる相乗作用は、ジャーカット又はヒトT細胞におけるもの程は強く現れなかった;図12d、データ示さず)。さらに、10mMカルシウム特異的BAPTAをパッチピペットに含むことは、20mM[Ca2+]o内の内向き電流の発生の遅延、続いて全細胞の破壊、ストア枯渇に反応するICRACの発生の示唆を引き起こした(データ示さず)。全体として、これらの結果は、マウスT細胞が、ICRACの特性と区別がつかない特性を有するCa2+電流を有していることを示した。
T細胞においてインビボでStim1とStim2の欠失が組み合わされた影響を分析するために、Stim1fl/flStim2fl/flCD4−Cre+「ダブルノックアウト」(短縮形DKO:double knockout)マウスが作製された。これらのマウスは、胸腺細胞性、並びにCD4−CD8−ダブルネガティブ(DN)、CD4+CD8+ダブルポジティブ(DP)、及びCD4+及びCD8+シングルポジティブ(SP)細胞の数及び比率を評価したところ、従来の胸腺成熟においては欠損を示さなかった(データ示さず)。2つの可能性のある説明としては、STIMタンパク質が長寿命であることにより、残りのSTIM1及びSTIM2タンパク質は、遺伝子欠失がDP段階で生じた後、又は胸腺細胞が末梢T細胞によって使用されたものとは異なるSTIM独立Ca2+流入機構を用いた後に、存在可能である及び機能的に良好であることができる。T細胞成熟及び胸腺選択におけるSTIMタンパク質の役割は、Cre発現がT細胞又は造血細胞成熟の早期に開始されたマウスを用いて、別個の研究の一環として調査されている。
LAT(Y136F)変異マウスの自己反応性表現型は、自己反応性T細胞が末梢部に逃げることを可能とする低下した陰性選択(Sommers, C.L. et al. , J Exp Med 201, 1125-1134 (2005)、及びTreg細胞の数の減少(Koonpaew, S., Shen, S., Flowers, L. & Zhang, W. LAT-mediated signaling in CD4+CD25+ regulatory T cell development. J Exp Med 203, 119-129 (2006))に起因したものであった。胸腺細胞の陽性及び陰性選択におけるSTIMタンパク質の役割の直接的な検査は、Lck−Creを用いてT細胞成熟の初期段階においてStim1及びStim2が除去されたマウスを必要とする。そして、これらマウスをHY−TCRトランスジェニックマウスと交配させた。その一方で、5−6週齢のStim1fl/flStim2fl/flCD−Creマウスの胸腺、脾臓、及びリンパ節におけるTreg細胞数の明確な減少(図6c、d、データ示さず)が記録された。DKOマウスの脾臓及びリンパ節におけるTreg細胞の比率は、加齢に伴い増加したが、それにもかかわらず、コントロールマウスの比率の10−20%間にとどまった(図6c)。これらの増加は、末梢リンパ器官の大きさに年齢依存的に増加した結果であると推定された。STIM1又はSTIM2のいずれかが欠如したマウスは、正常な数のCD4+CD25+Foxp3+Treg細胞を含んでいた(データ示さず)。Treg細胞のその他のマーカーであるGITRを発現する細胞数は、DKOマウスでも減少した(データ示さず)。また、さらに留意されるべきは、より加齢(8週)のDKOマウスからのCD25−及びCD25+T細胞におけるStim1及びStim2の完全な欠失であった(図15a)。CD4+CD25−T細胞と同様に、DKOマウスからのCD4+CD25+T細胞は、タプシガルギン又は抗CD3を用いた処置に反応して、低下したCa2+流入を示した(図6e及び図15b)。

Claims (2)
- 細胞内においてCa2+媒介性サイトカイン発現を抑制し且つ該細胞内においてストア作動性Ca2+流入の大幅な減少を引き起こさない検査薬を同定する方法であって、
a.少なくとも1つの検査薬を、STIM2タンパク質又はその機能性フラグメントをコードする異種核酸を含む組換え細胞に接触させる工程を備え、
前記異種核酸によってコードされたSTIM2タンパク質又はその機能性フラグメントが、ヒトSTIM2タンパク質と少なくとも80%が同一であるアミノ酸配列を有し、
前記方法はさらに、
b.前記細胞内においてCa2+媒介性サイトカイン発現を測定する工程と、
c.細胞膜を介するイオン流動もしくは電流又は膜電位の変化を測定する工程か、
前記細胞からの蛍光信号の変化を検出する工程か、
前記細胞からの発光信号の変化を検出する工程か、
前記細胞の膜電位の変化を測定する工程のいずれかとを備えることを特徴とする方法。 - 前記細胞が、T細胞であることを特徴とする請求項1記載の方法。
