コンフルエントとなった培養2日目に、それまでの培地を、DMEM(Doulbecco's modified Eagle's Medium)培地と交換し、スターフルーツ抽出液を0.25重量%(乾燥物換算で0.0025重量%)となるように加え、さらに24時間培養した。
その後、培地に250mM濃度のシュウ酸溶液1mLを加え、細部を溶解した。
続いて、100℃で60分間、室温で放置し、遠心分離によって得た上清を細胞ライセートとした。そして、細胞ライセート中のエラスチン量をBiocolor社のFastin Elastin Assayキットを用いて定量した。
