本発明は、自己免疫疾患の診断において有用な方法および組成物を提供する。特に、本発明は、グレーブス病の診断および管理における使用のための方法および組成物を提供する。例えば、1つの組成物は、循環する甲状腺刺激免疫グロブリンに対する改良された感度および特異性を有するキメラ甲状腺刺激ホルモン受容体を含む。そのようなキメラ受容体を用いるアッセイを、糖質コルチコイドの存在下で最適化することができる。
典型的には、若い成人におけるグレーブス病の臨床像は、非常に容易に認識される。患者は一般的には男性より女性に多く、限定されるものではないが、発汗、動悸、緊張、興奮、不眠、震え、頻繁な排便、および良好な食欲にも拘わらない体重減少などの症候が報告されている。理学的検査は通常、軽度の眼球突出、凝視、眼瞼遅滞、平滑性、広汎性、非圧痛性の甲状腺腫、大きい心音を伴う頻脈(特に、運動後)、ならびに頻繁には収縮期雑音もしくは胸骨左縁擦過傷、震え、爪甲離床症、および手掌紅斑を示す;頻繁には、甲状腺上で雑音が聞こえ、頸部のブンブン音がほとんど常に存在する。これらの症候を有する患者においては、グレーブス病が容易に認識され、研究室試験により確認することができる(Federman, Thyroid, DaleおよびFederman(編)、Scientific American Medicine, 3:1-6, Scientific American, New York, NY (1997)を参照)。
グレーブス病は、抗体を介する自己免疫反応により引き起こされる甲状腺障害である。グレーブス病患者においては、主に甲状腺刺激抗体(TSAb)および甲状腺阻害抗体(TBAb)などの、TSHRを認識する自己抗体(TRAb)は異種性である。TSAbは甲状腺機能亢進症を引き起こすTSHアゴニストとして作用するが、TBAbは甲状腺機能低下症を引き起こすTSHアンタゴニストとして機能する。TSAbおよびTBAbはTSHR上の異なるエピトープに結合するが、TBAb結合はTSAbの刺激効果を「中和」することができる。TSAbがTSHRに結合する時、それはcAMPシグナリング経路を誘導するが、TBAbはこの効果を有さない。
TSAbを測定する1つの方法は、「FRTL-5」として知られるラット甲状腺細胞系を用いる。Interthyroid Research Foundation (Baltimore, MD)から入手可能なこの細胞系は、ヒトTSAbと交叉反応する受容体を発現する。TSAbの存在下では(すなわち、例えば、これらの抗体を含有するグレーブス病患者に由来する血清への前記細胞の曝露の際に)、FRTL-5細胞はcAMPを産生するように刺激される。次いで、このcAMPを、ラジオイムノアッセイ法を用いて、溶解させた細胞の一部または細胞を入れた培地中で測定する。FRTL-5細胞は、グレーブス病の特徴である自己抗体に関するバイオアッセイを初めて成功させるための基礎を形成した。米国特許第4,609,622号(参照により本明細書に組み入れられるものとする)を参照されたい。

Metabol., 76:499(1993)により記載されたように実施されるFRTL-5細胞を用いる典型的なアッセイは、細胞培養培地中で用いられる通常の増殖構成要素に加えて、6種のホルモンを含む特殊な完全培地(すなわち、例えば、6H培地)中、96穴プレート中にFRTL-5細胞(30,000細胞/ウェル)を播種することを含む。5%CO2中、加湿された37℃インキュベーター中で2〜3日間インキュベートした後(すなわち、細胞が集密になった時)、TSHが6H培地中の6種のホルモンの1つである場合、TSHが欠損した(それにより5H培地をもたらす)「飢餓培地」に培地を交換する。次いで、細胞を、2〜3日毎に培地交換をしながら、インキュベーター中で4〜5日間維持する。この時間の間に、細胞は増殖または複製しない。続いて、細胞を診断アッセイにおいて用いることができる。
グレーブス病のための初期診断方法を、飢餓培地を除去し、ホスホジエステラーゼ阻害剤(例えば、0.5 mMメチルイソブチルキサンチン;IBMX)を含む特殊な低塩化ナトリウム、高スクロースバッファー(HBSS NaCl + 222 mMスクロース;このバッファーの調合法は、0.0608 g/L KH2PO4、0.144 g/L CaCl3、0.373 g/L KCl、0.048 g/L MgSO4、0.097 g/L Na2PHO4、1.0 g/L D-グルコース、76 g/L(すなわち、222 mM)スクロース、4.77 g/L HEPES、および10 g/L BSA;pH 7.2 - 7.4である)を含む刺激培地を添加して、この酵素がcAMPを破壊するのを防止することにより実施した。患者の免疫グロブリン(IgG)の特殊調製されたサンプル、対照、および標準物を、通常は3回、好適なウェルに添加し、プレートを5%CO2中、加湿された37℃インキュベーター中で2時間インキュベートする。このインキュベーション後、5〜10μlの培地を各ウェルから取り出し、ラジオイムノアッセイ系において使用して、cAMPの存在を検出する。典型的には、このアッセイを、患者について2回、約6個の標準物と共に実行し、対照も2回実行する。通常、このアッセイには、一晩のインキュベーションが必要であり、抗体に結合した放射標識されたcAMPから遊離の放射標識されたcAMPを分離するために次の日にも約1時間のインキュベーションが必要である。
リポーター遺伝子(すなわち、例えば、ルシフェラーゼ)の使用を介してTSIにより引き起こされるcAMP量の増加を容易に評価するために設計されたCHO-R細胞の使用により、さらなる改良が提供された(Evansら、J. Clin. Endocrinol. Metabol., 84:374 (1999))。かくして、この遺伝子操作された細胞系(すなわち、CHO-Rluc)の導入を用いる場合、cAMPの検出および定量のための以前に開発されたラジオイムノアッセイにおいて用いられる放射性化合物の使用に付いて回る複雑性および危険性が排除される。これらの細胞を用いる場合、単に細胞から培地を除去し、溶解バッファーを添加し、20〜30分間溶解を起こさせ、溶解物のサンプルを除去し、ルシフェラーゼ基質を添加し、ルミノメーターを用いて15秒間隔に渡って光出力を測定することにより、ルシフェラーゼを測定する。しかしながら、以下の実験の節に示されるように、この方法は、曖昧な結果および必要とされるさらなる改善を提供する。
一実施形態においては、本発明は、蛍ルシフェラーゼ遺伝子と共にTSH/LH/TSHキメラ受容体(すなわち、例えば、RMc4)を発現する組換え細胞系(すなわち、例えば、CHOおよびRD)を意図する。一実施形態においては、この発現は、ヒト糖タンパク質αサブユニットプロモーターにより駆動される。本発明の機序を理解する必要はないが、キメラ受容体を用いることにより、遮断抗体(すなわち、例えば、TBAb)の結合は排除され、および/または減少すると考えられる。一実施形態においては、キメラ受容体は、TSAb結合領域のみが発現されるように、少なくとも1個の遺伝子改変を含む。CHO-Luc細胞またはKRONUS(登録商標)アッセイと比較した場合、組換え細胞系は高い特異性を有すると考えられる。
上記のように、本発明は、免疫応答の発生をモニタリングするための方法および組成物も提供する。特に、本発明は、ワクチン接種に対する個体の応答をモニタリングするのに好適な方法および組成物を提供する。
一実施形態においては、本発明は、グレーブス病を診断する、および/またはグレーブス病の療法をモニターするための遺伝子操作されたチャイニーズハムスター卵巣(CHO)および/またはヒト横紋筋肉腫細胞(RD)の使用を意図する。
1. この系は、より高いシグナル:ノイズ(S:N)またはシグナル:バックグラウンド(S:B)比をもたらすより低いルシフェラーゼ活性バックグラウンドをもたらす。
一実施形態においては、本発明は、高い検出感度および特異性で甲状腺刺激ホルモン受容体(TSH-R)自己抗体(すなわち、例えば、甲状腺刺激免疫グロブリン;TSI)を検出する新規診断細胞系を意図する。一実施形態においては、細胞系は、組換えチャイニーズハムスター卵巣細胞(すなわち、例えば、CHO-K1細胞)を含む。一実施形態においては、細胞系は、ヒト横紋筋肉腫(RD)細胞を含む。
グレーブス病に関連するTSHおよび甲状腺刺激自己抗体のための結合部位の同一性を、ヒト/ラットキメラTSH-R構築物を構築することにより最初に試験した。対応するラット配列を含むヒトTSH-Rの部分的置換は、以下のキメラ受容体をもたらした:i)アミノ酸残基8-165を置換するMc1+2;ii)アミノ酸90-165を置換するMc2;およびiii)アミノ酸残基261-370を置換するMc4。このデータは、アミノ酸残基8-165が、TSHにより必要とされるものと同じではない甲状腺刺激自己抗体にとって特異的なエピトープを含むことを示唆していた。特発性粘液水腫甲状腺刺激抗体、グレーブス病甲状腺刺激抗体、およびTSHの間の結合部位における有意な異種性が観察された。Taharaら、「Immunoglobulins From Graves' Disease Patients Interact With Different Sites On TSH Receptor/LH/CG Receptor Chimeras Than Either TSH Or Immunogloblins From Idiopathic Myxedema Patients」、Biochem Biophys Res Comm 179:70-77 (1991)を参照されたい。
次いで、陰性および陽性TSI血清に対するCHO-RMc4luc、RD-RMc4lucおよびCHO-Rluc細胞系の応答を比較した。細胞を、TSI陰性および陽性血清と共に3時間インキュベートした。次いで、細胞を溶解させ、ルシフェラーゼ活性をVeritas Microplat Luminometerにより測定した。この結果は、CHO-RMc4lucおよびRD-RMc4luc細胞系の両方が、CHO-Rluc細胞と比較した場合、非常により高い検出感度を有することを示していた。図10A、10B、10C、10Dおよび10Eを参照されたい。TSI陽性血清に由来するルシフェラーゼRLUと、陰性血清との比率(S/N比)の比較は、CHO-RMc4lucおよびRD-RMc4luc細胞が、CHO-Rluc細胞系よりも6〜8倍および2.1〜4倍感度が高かったことを示している。それぞれ、図10Bおよび10Dを参照されたい。CHO-RMc4luc細胞は、RD-RMc4luc細胞よりも約1.3〜3.5倍感度が高かった。図10Eを参照されたい。さらに、CHO-RMc4lucは、より低いバックグラウンドおよび高いシグナル/陰性(S/N)比をもたらすTSI陰性血清を用いて試験した場合、CHO-Rlucよりも低いレベルのルシフェラーゼ活性を誘導した。図10Aを参照されたい。さらに、CHO-RMc4lucおよびRD-RMc4luc細胞系は両方とも、非常に低い標準偏差値を示した。それぞれ、図10Aおよび10Cを参照されたい。19番と命名されたTSI陽性血清は、CHO-RMc4luc、RD-RMc4lucおよびCHO-Rluc細胞系に対して高いルシフェラーゼ誘導レベルを示した。この血清を希釈し、これらの細胞系上で試験して、感度を比較した。図10Eを参照されたい。
上記で考察されたように、CHO-RMc4luc細胞系は、現在用いられているCHO-Rluc細胞系を超えて、感度および特異性における無限の利点を提供する。例えば、グレーブス病抗体による刺激に際して、CHO-RMc4luc細胞は、相対光単位(RLU)出力により測定されるようにルシフェラーゼ応答の増加を提供する。この改善により、プロトコルから1日の飢餓工程を排除することができる。
上記データは、飢餓期間はCHO-RMc4luc細胞の正確性および感度の改善をももたらすことを示すため、飢餓期間を用いずに優れたCHO-RMc4luc細胞アッセイを開発するためのさらなる調査を行った。
さらに他の実施形態においては、本発明は、キメラTSH受容体を用いてグレーブス病の診断アッセイを実施するためのキットを提供する。このキットは、本発明の細胞系に基づく診断方法を含む1個以上の容器を含むのが好ましい。いくつかの実施形態においては、前記容器は、限定されるものではないが、デキサメタゾン、コルチゾン、ヒドロコルチゾン、プレドニゾン、またはフルチカゾンなどの糖質コルチコイドを含んでもよい。いくつかの実施形態においては、このキットは、全ての対照、アッセイを実施するための指示書、ならびに結果の分析および提示のための任意のソフトウェアを含む、患者血清中の循環するTSH自己抗体を検出するためのグレーブス病診断アッセイを実施するのに必要な、またはそれにとって十分な全ての成分を含む。いくつかの実施形態においては、前記キットは、細胞系をトランスフェクトすることができるキメラTSH受容体をコードするベクターを含む。いくつかの実施形態においては、このキットは、単一の反応で診断アッセイを実施し、診断、予後、もしくは予測情報を提供する(例えば、研究者もしくは医師に対して)のに必要な、またはそれにとって十分な全ての材料を含む。例えば、そのようなキットは、キメラTSH受容体とルシフェラーゼ受容体系を含む細胞系を含んでもよい。いくつかの実施形態においては、前記キットは、Mc4キメラTSH受容体のための第1の核酸配列、ルシフェリン/ルシフェラーゼリポーター系のための第2の核酸配列、およびプロモーターのための第3の核酸配列を含む1個以上のベクターを含む。他の実施形態はまた、バッファー、対照試薬、検出装置、ソフトウェア、説明書、およびTSH自己抗体標準調製物を含んでもよい。
以下、本発明の実施形態を示す。

実験
以下の実施例は、本発明の特定の好ましい実施形態および態様を証明し、さらに例示するために提供されるものであり、その範囲を限定するものとして解釈されるべきではない。
これらの実験においては、CHO-Rluc細胞を、グレーブス病患者中のTSIを検出するための試験方法における使用のためにW-25 CHO-R細胞から調製した。ピューロマイシン耐性細胞のプールを取得し、ウシTSHに応答する光出力について試験した。最も高い光出力を有するクローンを、以下に記載の実験における使用のために選択した。
これらの実験においては、実施例1に記載のように調製されたCHO-Rluc細胞を、グレーブス病の診断のためのアッセイにおいて用いた。試験のための24個の単層を調製するために、最初に96穴マイクロタイタープレート中の24ウェルを、50〜100μlの0.1%ゼラチン溶液(Sigma)を添加することにより処理して、試験のために選択された24ウェルの底部への細胞の結合を増強した。室温で約1分間インキュベートした後、ゼラチン溶液を、吸引によりそれぞれのウェルから除去した。このゼラチンは1分より長い時間、ウェル上に残存し得る。細胞がウェルにより急速に付着し、より迅速に集密に達するように、ゼラチンはウェルをコラーゲンでコーティングするのに役立つ。しかしながら、細胞を植え付け、ゼラチンを用いずに集密まで増殖させ、依然としてうまく実施することができる。
これらの実験においては、患者のIgGを、本発明の方法における試験のために調製した。38人のよく知られ、特性評価された、未治療のグレーブス病患者に由来する凍結乾燥されたIgGサンプルは、Dr. B.Y. Cho (Department of Internal Medicine, Seoul National University, College of Medicine, Seoul, Korea)の厚意により提供されたものである。このサンプルの多くはCHO-RおよびFRTL-5細胞を用いる標準的な方法で以前に試験されたものであるので、これらの試験結果は、これらのサンプルのうちの35個について公知であった。
これらの実験においては、Evansら(Evansら、J. Clin. Endocrinol. Metabol., 84:374 (1999))により記載された方法を用いて、CHO-Rluc細胞の性能を評価した。実施例2に記載のように調製した96穴マイクロタイタープレート中で用いられた24穴中の細胞単層に由来する培地を吸引し、10%低ホルモンウシ血清(Sigma)を含む100μlのHam's F-12培地と交換し、5%CO2を含む加湿雰囲気中、35〜37℃で一晩インキュベートした。
以前に記載の実験を考慮して、様々な培地製剤の効果を、ウシTSHおよびヒトTSIの測定においてCHO-Rluc細胞と共に使用するために調査した。これらの実験においては、様々な培地製剤を、bTSHおよびグレーブス病患者の血清から抽出したIgGを用いて、CHO-Rlucアッセイにおける「飢餓」および「刺激」工程について試験した。
反応速度を援助または増強するためにin vitroでの抗原/抗体反応においてPEGを用いることができるため、PEGを刺激培地中に含有させた試験を行った。この化合物は抗原/抗体複合体の解離速度または解離を減少させることができるため、本発明の方法におけるPEGの使用を調査した。
これらの実験においては、PEGを用いる代替的なプロトコルを試験した。第1に、CHO-Rluc細胞の凍結バイアルを解凍し、増殖培地中に希釈し(それぞれの細胞バイアルの内容物を2.5 mlの培地に添加した)、100μlのこの細胞懸濁液を、以前に記載のように調製された、96穴マイクロタイタープレートの24個のゼラチン被覆ウェルのそれぞれに添加した。プレートを、5%CO2を含む雰囲気を有する35〜37℃の加湿されたインキュベーター中、20〜24時間インキュベートした。これにより、緩く集密になった単層が得られた。
これらの実験においては、6%PEG-8000を含む刺激培地を用いる本発明の方法を、標準的なHBSSを含有する飢餓培地および刺激培地を用いる方法と比較して、Dr.Choから得られた35個の未治療のグレーブス病IgG標本についてルシフェラーゼ値を得た。本発明の方法において用いられるのと同じIgGサンプルを用いるFRTL-5およびCHO-R細胞についでのDr.Choにより得られたcAMPの値を、図2、3および4中のCHO-Rルシフェラーゼの結果と比較して示す。図5は、bTSHに応答するルシフェラーゼの直線性を示す。
これらの実験においては、ワクチンレシピエントの免疫応答を測定、モニタリングする。本発明はそのように限定されることを意図されないが、本実施例は、単純ヘルペスウイルス(HSV)ワクチンに対する被験者の免疫応答のモニタリングを記載する。
本実施例は、グレーブス病自己抗体を検出するためのキメラTSH-R受容体を含む細胞系の構築の一実施形態を提供する。
TSH-Rキメラ受容体をコードする第1の核酸配列とニューロマイシン耐性遺伝子をコードする第2の核酸配列とを含むプラスミドを、ルシフェラーゼ遺伝子および糖タンパク質ホルモンαサブユニットプロモーターに連結した。
QIAGEN RNA/DNAキット(QIAGENカタログ番号14123)を用いて、ヒト胚性腎細胞から染色体DNAを単離した。糖タンパク質αサブユニットプロモーター断片を、PCR鋳型としての単離された染色体DNAおよび以下に示す2対のオリゴヌクレオチドプライマー:

を用いるPCRにより増幅した。
ヒト糖タンパク質αサブユニットプロモーターを、SacIおよびBglIIを用いる制限切断によりベクターpcDNA2.1から単離した。次いで、得られる316 bpの断片を、プラスミドpGHP/Lucの構築のために、pGL2エンハンサープラスミド(Promega, Madison, WI)のSac I/Bgl II部位中にサブクローニングした。
抗生物質選択(陽性クローン選択)のためのネオマイシン耐性遺伝子を、XhoIおよびHicIIの制限酵素を用いてベクターpMC1 (Stratagene Ceder Creek, TX)から単離した。次いで、得られた断片を、SV40プロモーター(Dr. Leonard Kohnから入手)により駆動されたTSHR/LHキメラ受容体を含むプラスミドpMc4のXbaI部位中にサブクローニングした。最終プラスミドをpMc4-neoと命名した。
制限酵素XhoIおよびXbaIを用いてベクターpCMV/Bsd (Invitrogen, Carlsbad, CA)から単離された、抗生物質選択遺伝子ブラストシジンを、TSHR/LHキメラ受容体を含むプラスミドpMc4のXbaI部位にサブクローニングした。Taharaら、「Immunoglobulin From Graves' Disease Patients Interact With Different Sites On TSH Receptor/LH/CG Receptor Chimeras Than Either TSH Or Immunoglobulins From Idiopathic Myxedema Patients」、Biochem Biophys Res Comm 179: 70-77 (1991)を参照されたい。最終プラスミドをpMc4-Bsdと命名した。
蛍ルシフェラーゼリポーター遺伝子を含むヒト糖タンパク質αサブユニットプロモーターを、SmaIおよびAccIによる制限切断後、ベクターpMc4/Lucから単離した。次いで、単離されたDNA断片を、pMc4-neoプラスミドのPfoI部位中にサブクローニングした。最終プラスミドを、pMV4-GHP/Lucと命名した。
7種の異なる哺乳動物細胞系を試験して、最も低いサイクリックAMP基底レベルと最も高い潜在的誘導レベルを有する細胞系を選択した。この結果により、CHOおよびRD細胞が、最も低いサイクリックAMP基底活性および最も高い潜在的誘導レベルを示すことが示された。この経験的研究手法は、細胞培養型の適切な選択により、アッセイ感度を最大化する。例えば、より低いサイクリックAMP基底レベルは、ルシフェラーゼアッセイの感度を増加させる。また、サイクリックAMPの最も高く誘導された発現は、ルシフェラーゼアッセイの正確性を改善する。
本実施例は、CHO細胞の永続的トランスフェクションを記載する。
本実施例は、検出を容易にするための一連の2個のプラスミドpGHP/LucおよびpMc4-Bsdを用いるヒト横紋筋肉腫(RD)(ATCC番号:CCL-136)細胞系のトランスフェクションを記載する。
凍結バイアルに由来するCHO細胞を希釈し、増殖培地(10%(v/v)ウシ胎仔血清および9種の必須アミノ酸を含むHam's F12培地)中、37℃および5%CO2で16時間増殖させた。16時間後、培地を除去し、CHO細胞を洗浄し、100μl/ウェルの「飢餓」HBSS培地を再供給した。次いで、CHO細胞を22〜24時間インキュベートした。インキュベーション後、培地を除去し、CHO細胞を洗浄し、100μl/ウェルの反応バッファーを再供給した。次いで、CHO細胞を、BSA、PEG、スクロース、グルコース、および塩(Diagnostic Hybridsカタログ番号40-300500)を含む反応バッファー中、1:11希釈率の患者血清を用いて、37℃および5%CO2で4時間誘導した。
CHO-Rluc(041307A)およびCHO-RMc4luc(062707, P8, 3e6/ml)細胞を、5 mlのCHO増殖培地中にそれぞれ入れた。この細胞保存液から、十分なものを取り、それぞれのデキサメタゾン濃度(100、50、40、25、12.5、および0μM)に関して、CHO-Rlucアッセイについては66,666細胞/ウェルおよびCHO-RMc4lucアッセイについては50,000細胞/ウェルとなるように6個のチューブに分注した。デキサメタゾン保存液は、CHO増殖培地中で500μMであった。全てのデキサメタゾンを新鮮に作製した。これらのバイアルを遠心分離し、細胞をCHO増殖培地中の様々な濃度のデキサメタゾンに入れた。
4個のサンプルがグレーブス病について陽性であると知られ、5個のサンプルがグレーブス病について陰性であると知られる9個のヒト血清サンプルを調製した。9個のサンプルのそれぞれを、非飢餓/デキサメタゾンプロトコルおよび飢餓プロトコルの両方の各条件で試験した。
3個の細胞プレートを、CHO-RMc4プロトコルを用いて分析した。プレート1:CHO-Mc4luc細胞(4e6細胞/プレート)+CHO-Mc4反応バッファー中に40μMのデキサメタゾンを含む増殖培地。プレート2:CHO-RMc4luc細胞(4e6細胞/プレート)+CHO-Mc4反応バッファー中に40μMのデキサメタゾンを含まない増殖培地。プレート3:CHO-Rluc細胞(4e6細胞/プレート)+CHO-Rluc反応バッファー中に40μMのデキサメタゾンを含まない増殖培地。
3個のプレートを、CHO-Rプロトコルを用いて分析した。プレート1:CHO-RMc4細胞(4e6細胞/プレート)+CHO-Mc4反応バッファー中に40μMのデキサメタゾンを含む増殖培地。プレート2:CHO-RMc4luc細胞(4e6細胞/プレート)+CHO-Mc4反応バッファー中に40μMのデキサメタゾンを含まない増殖培地。プレート3:CHO-Rluc細胞(4e6細胞/プレート)+CHO-Rluc反応バッファー中に40μMのデキサメタゾンを含まない増殖培地。各プロトコルに関するサンプルの配列を同一に配置した。図16を参照されたい。
本実施例は、CHO-RMc4アッセイの改善された感度が、実施例16に従うデキサメタゾン(Dex)による飢餓培地期間の置換に限定されないことを示すデータを提供する。これらのデータは、4種の代用糖質コルチコイド(GC)がシグナル強度の改善において同等の効果を有することを示している。

- a)i)キメラTSH受容体をコードする配列番号3を含む安定にトランスフェクトされたベクターを含む非飢餓細胞系であって、前記配列番号3がルシフェラーゼ遺伝子に連結されている、前記細胞系;
ii)グレーブス病を有すると疑われる患者に由来する血清サンプル;及び
iii)細胞培養培地;
を提供すること;
b)該血清サンプルと、該細胞系及び該培地とを、検出可能なシグナルが放出されるような条件下で接触させること、
を含む方法。 - 前記方法が、工程c)前記サンプル中に存在する甲状腺刺激ホルモン受容体自己抗体濃度と相関するシグナル強度を測定することをさらに含む、請求項1に記載の方法。
- 前記接触が、ポリエチレングリコールをさらに含む、請求項1に記載の方法。
- 前記キメラTSH受容体が、ラット絨毛性ホルモンゴナドトロピン受容体に由来するアミノ酸配列を含む、請求項1に記載の方法。
- 前記アミノ酸配列が、ヒトTSH受容体アミノ酸配列のアミノ酸残基262-335に対応する73個のアミノ酸を含む、請求項1に記載の方法。
- キメラTSH受容体遺伝子を含むベクターを含む、血清TSH自己抗体を検出するためのキットであって、前記キメラTSH受容体遺伝子が配列番号3を含み、且つルシフェラーゼ遺伝子に連結されている、前記キット。
- 前記キットが、前記ベクターによりトランスフェクトすることができる細胞系をさらに含む、請求項6に記載のキット。
- 前記キットが、ポリエチレングリコールをさらに含む、請求項6に記載のキット。
- 前記細胞系がCHO細胞及びRD細胞から成る群より選択される、請求項7に記載のキット。
- a)i)キメラTSH受容体をコードする配列番号3を含む安定にトランスフェクトされたベクターを含む非飢餓細胞系であって、前記配列番号3がルシフェラーゼ遺伝子に連結されている、前記細胞系;
ii)グレーブス病を有すると疑われる患者に由来する血清サンプル;及び
iii)細胞培養培地;
を提供すること;
b)該血清サンプルと、該細胞系及び該培地とを、検出可能なシグナルが放出されるような条件下で接触させること;並びに
c)検出可能な甲状腺刺激抗体の存在について観察すること、
を含む方法。 - 前記培養細胞が、ルシフェラーゼ遺伝子で安定にトランスフェクトされた横紋筋肉腫細胞及びルシフェラーゼ遺伝子で安定にトランスフェクトされたチャイニーズハムスター卵巣細胞から成る群より選択される、請求項10に記載の方法。
- 前記観察を、ルミノメーターを用いて行う、請求項10に記載の方法。
- 前記検出可能なシグナルがcAMP濃度を決定する、請求項12に記載の方法。
- 前記方法が、増殖培地及び刺激培地から成る群より選択される培地をさらに含む、請求項13に記載の方法。
