本発明者らは、これらの知見にもとづいてさらに検討を重ねた結果、本発明を完成するに至った。
(1) 非成体NOD/SCID/IL2rgnullマウスにヒト急性骨髄性白血病患者由来の白血病幹細胞含有物を移植し、該マウスを生育させることを含む、ヒト白血病細胞が選択的に増幅したマウスの製造方法;
(2) 上記(1)記載の方法により得られるマウス由来の白血病幹細胞含有物を、別の非成体NOD/SCID/IL2rgnullマウスに移植し、該マウスを生育させる工程を1回以上含む、ヒト白血病細胞が選択的に増幅したマウスの製造方法;
(3) 白血病幹細胞含有物移植後の生育期間が4週以上である、上記(1)または(2)記載の方法;
(4) ヒト白血病細胞の増幅がマウス末梢血中で生じていることを特徴とする、上記(1)〜(3)のいずれかに記載の方法;
(5) 白血病幹細胞含有物の由来する個々の患者における白血病の病態を再現することを特徴とする、上記(1)〜(4)のいずれかに記載の方法;
(6) 上記(1)〜(4)のいずれかに記載の方法により得られる、ヒト白血病細胞が選択的に増幅したマウス;
(7) a)上記(6)記載のマウスに被験物質を投与する工程、およびb)該マウスにおける白血病の改善を評価する工程、を含む、ヒト急性骨髄性白血病治療剤のスクリーニング方法;

好ましい被験物質の一例として、既知もしくは新規に見出されたAML治療ターゲット分子に対する抗体、あるいは治療ターゲット遺伝子に対するsiRNA、アンチセンス核酸(DNA、RNA)などが挙げられる。
非経口投与のための組成物としては、例えば、注射剤、坐剤などが用いられ、注射剤は静脈注射剤、皮下注射剤、皮内注射剤、筋肉注射剤、点滴注射剤、関節内注射剤などの剤形を包含する。かかる注射剤は、自体公知の方法に従って、例えば、上記活性成分を通常注射剤に用いられる無菌の水性もしくは油性液に溶解、懸濁または乳化することによって調製する。注射用の水性液としては、例えば、生理食塩水、ブドウ糖やその他の補助薬を含む等張液などが用いられ、適当な溶解補助剤、例えば、アルコール(例、エタノール)、ポリアルコール(例、プロピレングリコール、ポリエチレングリコール)、非イオン界面活性剤〔例、ポリソルベート80、HCO−50(polyoxyethylene(50mol)adduct of hydrogenated castor oil)〕などと併用してもよい。油性液としては、例えば、ゴマ油、大豆油などが用いられ、溶解補助剤として安息香酸ベンジル、ベンジルアルコールなどを併用してもよい。調製された注射液は、通常、適当なアンプルに充填される。直腸投与に用いられる坐剤は、上記物質を通常の坐薬用基剤に混合することによって調製される。
マウス
NOD.Cg-PrkdcscidIl2rgtmlWjl/Sz(NOD/SCID/IL2rg null)マウスは、Il2rg遺伝子座の完全ヌル変異(Shultz, L.D. et al. Multiple defects in innate and adaptive immunologic function in NOD/LtSz-scid mice. J Immunol 154, 180-191 (1995))をNOD.Cg-Prkdcscid(NOD/SCID)系統と戻し交雑することによって、The Jackson Laboratoryで開発された。NOD.Cg-Prkdcscidb2mtm1Unc(NOD/SCID/β2mnull)マウスは、The Jackson Laboratoryで維持された研究用コロニーから得た。マウスを、理研およびThe Jackson Laboratoryの動物施設で、各施設でInstitutional Animal Committeesによって確立されたガイドラインに従って、照射した食物および酸性化した水を用いて飼育し、規定された細菌叢のもとで維持した。
理研RCAIのInstitutional Review Board for Human Researchからの以前の承認により、全ての実験を実施した。AML患者由来の白血病細胞は、書面によるインフォームドコンセントにより収集した。サンプルは、French-American-British(FAB)分類システムのサブタイプM1(前骨髄球性を超える成熟を伴わない;症例1、2および11)、M2(骨髄芽球性、成熟を伴う;症例3、4、10)、M3(前骨髄球性;症例5、6)、M4(骨髄単球性;症例7、8および12)およびM7(巨核芽球性;症例9)を有するAML患者に由来した。BMMNC(骨髄単核球細胞)は、密度勾配遠心分離を使用して単離した。
T細胞枯渇(TCD)BMMNC移植のために、全BMMNCを、マウス抗hCD3、抗hCD4および抗hCD8モノクローナル抗体(BD Immunocytometry, San Jose, CA)と共にインキュベートし、その後抗マウスIgG抗体コンジュゲート化免疫磁気ビーズ(Dynal, Norway)を用いてT細胞を除去した。ソートしたhCD34+hCD38-AML細胞の移植のために、AML患者のBMMNCを、蛍光色素コンジュゲート化マウス抗hCD34モノクローナル抗体および抗hCD38モノクローナル抗体(BD Immunocytometry, San Jose, CA)で標識し、その後FACSAria(Becton Dickinson, San Jose, CA)を使用して蛍光標識細胞分取(FACS)を行なった。混入を回避するために、FSC/SSC高さおよびFSC/SSC幅の分析によって、ダブレットを排除した。hCD34+hCD38-細胞の純度は、ソート後には98%よりも高かった。新生仔(誕生後2日以内)および成体(10〜12週齢)のマウスに、137Cs源照射器を使用して、100cGy/分で、それぞれ100cGyおよび300cGyの全身照射を与え、その後2時間以内にAML細胞の静脈内注射を行なった。
BMは大腿骨および脛骨から収集し、PBは移植レシピエントの眼窩静脈叢から収集した。BM細胞およびPB細胞の表面表現型を、マウス抗hCD33、hCD13、hCD34、hCD38、hCD45、hGPA、hCD41aおよびラット抗mTer119、ならびにmCD41aモノクローナル抗体(BD Immunocytometry)を使用するフローサイトメトリーによって分析した。生着したhCD45+hCD33+細胞をソートし、May-Grunwald Giemsa染色によって形態学的に試験した。ソートしたhCD45+hCD33+レシピエントBMMNCは全てMay-Grunwald-Giemsa染色によって確認されるとおり白血病芽球であったので、ヒトAML生着をレシピエントBMにおける%hCD45+hCD33+として定義した。
パラホルムアルデヒドで固定し、脱灰し、パラフィン包埋した切片を、ソートしたhCD34+hCD38-AML BM細胞を移植したレシピエントの大腿骨から調製した。各切片をヘマトキシリンおよびエオシン(HE)染色に供した。TUNEL染色を、Biopathology Institute(Oita, Japan)によるApopTagペルオキシダーゼin situアポトーシス検出キット(Intergene, Purchase, NY)を使用して、標準的な手順に従って実施した。CD34+細胞集団の局在を決定するために、切片をマウス抗hCD34抗体(Immunotech, France)で免疫染色し、ラット抗マウスオステオポンチン抗体(Immuno-Biological Laboratories, Tokyo, Japan)を使用した。LSCの定量的局在について、骨内膜領域および中心領域を、以前に記載されたように(Nilsson, S.K., Johnston, H.M. & Coverdale, J.A. Spatial localization of transplanted hemopoietic stem cells: inferences for the localization of stem cell niches. Blood 97, 2293-2299 (2001))、それぞれ骨内膜から12細胞内側または外側の領域として定義した。各領域内の細胞計数を、Zeiss Axiovert 200(Carl Zeiss, Germany)を用いて光学顕微鏡下で実施した。ホーミング分析のために、症例4、7および10由来の4×103〜4×104のBM hCD34+hCD38-細胞を、6〜8Gyを照射した後に新生仔レシピエントに注射した。3日目にレシピエントを屠殺し、大腿骨および脛骨を組織学的分析のために得た。合計10,065の細胞が、大腿骨および脛骨の切片から計数された。生着後局在分析について、合計24,973の細胞が、症例4、10および11由来のBM hCD34+hCD38-細胞のレシピエントから調製した大腿骨および脛骨の切片を使用して計数された。免疫蛍光標識を、モノクローナル抗ヒトCD45抗体(Dako, Denmark)およびモノクローナル抗ヒトCD34抗体(Immunotech)を使用して実施した。白血病細胞の表面上のhCD34およびhCD45の同時局在を決定するために、ポリクローナルヤギ抗ヒトCD45抗体(Santa Cruz Biotechnology, CA)を使用した。Alexa 488コンジュゲート化ロバ抗マウスIgGおよびAlexa 568コンジュゲート化ロバ抗ヤギIgG(Invitrogen, CA)を二次抗体として使用した。共焦点レーザー走査画像化を、Zeiss LSM 510(Carl Zeiss, Germany)を使用して実施した。
150mg/kg のAra-C(Biogenesis, Poole, UK)の腹腔内(i.p.)注射を、移植の12〜16週間後に、症例4(n=2)および7(n=4)由来の104の精製BM hCD34+hCD38-細胞を移植したNOD/SCID/IL2rg nullレシピエントにおいて実施した。PBを、注射の時点(0日目)およびその後4日毎に得、MNCをマウス抗hCD45および抗hCD33モノクローナル抗体で標識し、フローサイトメトリーを用いてhCD45+hCD33+細胞の割合を決定した。PB 1ml当たりのAML細胞数を、各レシピエントにおいて、PB hCD45+hCD33+細胞の割合と総PB白血球計数/mlとを乗算することによって決定した。ヘモグロビン濃度、血小板計数および血漿アスパラギン酸アミノトランスフェラーゼ(AST)レベルを、Ara-C投与の前および投与の8日後に分析した。
移植の12〜20週間後に、症例4、7および11由来のソートしたBM hCD34+hCD38-細胞104〜105を移植したNOD/SCID/IL2rg nullレシピエントに、150mg/kgのAra-Cをi.p.注射した。注射の3日後、BMを、hCD45+ゲート内のhCD34+hCD38-画分、hCD34+hCD38+画分およびhCD34-画分を最初に分割し、Annexin Vおよび7-アミノ-アクチノマイシンD(7AAD)染色(BD Immunocytometry, San Jose, CA)によって各画分中の生存集団を定量することによって、アポトーシス細胞の存在について分析した。Annexin V-7AAD-hCD45+hCD34+hCD38-細胞をソートし、103の細胞を、照射した新生仔NOD/SCID/IL2rg nullレシピエントに静脈内注射した。
in vivoのBrdU取り込みについて、ヒトAML生着レシピエントを、2mgのBrdUでパルスし(i.p.)、BMを、FITC BrdU Flow Kit(BD Biosciences, San Diego, CA)ならびに上記のhCD34抗体およびhCD38抗体を使用して、注射の24時間後に分析した。細胞周期のG0期にある細胞の定量のために、ヒトAML生着レシピエントのBM細胞を、5μg/mlのHoechst 33342および30μg/mlのPyronin Yで標識し、その後抗hCD34抗体および抗hCD38抗体を使用する表面染色を実施した。
マイクロアレイ分析のために、AML患者および対応する三次レシピエントのBMから、2〜5×104のhCD34+hCD38-細胞およびhCD34+hCD38+細胞を、FACSAriaを用いて精製した。総RNAを、ISOGEN-LS試薬(Nippon Gene, Toyama, Japan)を使用して抽出し、RNAの完全性をBioanalyzer(Agilent, Santa Clara, CA, USA)を用いて評価した。cDNA合成、aRNA増幅、ビオチン化および断片化を、One-Cycle Target Labeling Kit(Affymetrix, Santa Clara, CA, USA)を用いて実施した。15μgの標識サンプルをハイブリダイゼーションカクテルに添加し、製造者の指示書に記載されたとおりに、45℃で16時間、Human Genome U133 Plus 2.0 GeneChips(Affymetrix)とハイブリダイズさせた。洗浄およびストレプトアビジン-フィコエリトリン染色を、GeneChip Fluidics Station(Affymetrix)を使用して実施した。続いて、チップを、GeneChip Scanner 3000(Affymetrix)を使用してスキャンした。各プローブセットについて正規化したハイブリダイゼーション強度を、GeneSpringソフトウェアパッケージ(Agilent)のGC-RMA法(Subramanian, A. et al. Gene set enrichment analysis: a knowledge-based approach for interpreting genome-wide expression profiles. Proc Natl Acad Sci U S A 102, 15545-15550 (2005))をデフォルトの設定で用いて計算した。Rで実行されるpvclust(Suzuki, R. & Shimodaira, H. Pvclust: an R package for assessing the uncertainty in hierarchical clustering. Bioinformatics (Oxford, England) 22, 1540-1542 (2006))(http://www.r-project.org/)および2007年2月に構築された「Curated」遺伝子セットコレクションを使用するGene Set Enrichment Analysis(Subramanian, A. et al. Gene set enrichment analysis: a knowledge-based approach for interpreting genome-wide expression profiles. Proc Natl Acad Sci U S A 102, 15545-15550 (2005))を、パラメータをデフォルトに設定して使用する階層的クラスタリング分析に、このマイクロアレイデータを供した。同じmRNAプロフィールを使用する対応する分析もまた実施した(Culhane, A.C., Thioulouse, J., Perriere, G. & Higgins, D.G. MADE4: an R package for multivariate analysis of gene expression data. Bioinformatics (Oxford, England) 21, 2789-2790 (2005))。CD34+CD38-細胞とCD34+CD38+細胞との間で有意に示差的な発現を示す遺伝子を規定するために、マイクロアレイデータを、p値<0.01で統計的t検定によって分析した。特に、RefSeqコレクション(http://www.ncbi.nlm.nih.gov/RefSeq/)中のヒトタンパク質コード遺伝子をこの分析において考慮した。全てのマイクロアレイデータを、アクセッションID CBX21でCIBEX(日本の公開の遺伝子発現データベース(http://cibex.nig.ac.jp/))に登録した。
平均%生着における差異を、BMDP Statistical Software(SPSS, Chicago, IL)を使用して、ノンパラメトリックFriedman二元配置分散分析によって分析した。ソートした集団における一次および二次生着の平均%の差異を、Kruskal-Wallis検定(GraphPad Prism, GraphPad, San Diego, CA)によって分析した。両側t検定を、ホーミングおよび生着局在、アポトーシスならびに細胞周期分析のために使用した(GraphPad Prism, GraphPad, San Diego, CA)。
初代ヒトAML細胞は、新生仔NOD/SCID/IL2rg nullモデルにおいて高い効率で生着する。80〜90%のAML芽球を含む9人の患者のBMから、4×106のT細胞枯渇BM単核細胞(BMMNC)を、致死未満で照射したレシピエントに静脈内注射した。ヒトAMLの生着は、成体NOD/SCID/IL2rg null(11.9%)レシピエントおよび新生仔NOD/SCID/β2mnull(12.9%)レシピエントと比較して、新生仔NOD/SCID/IL2rg nullレシピエント(37.8%)において有意に効率が高かった(図1a)。さらに、全てのAML患者のBMMNCは、新生仔NOD/SCID/IL2rg nullレシピエントにおいて生着したが、3/9(2人のM3および1人のM7)のBMMNCは、成体NOD/SCID/IL2rg nullレシピエントおよび新生仔NOD/SCID/β2mnullレシピエントにおいて生着できなかった。
CB17/SCIDマウスおよびNOD/SCIDマウスにおいて、BM中のヒトAML hCD34+hCD38-の生着は以前に実証されているが、末梢血(PB)生着の検出は限定的であった(Lapidot, T. et al. A cell initiating human acute myeloid leukaemia after transplantation into SCID mice. Nature367, 645-648 (1994)およびLumkul, R. et al. Human AML cells in NOD/SCID mice: engraftment potential and gene expression. Leukemia16, 1818-1826 (2002))。新生仔NOD/SCID/IL2rg nullレシピエントにおいて、外因性サイトカインも他の操作もなしに、ソートした初代AML hCD34+hCD38-細胞を103の少量静脈内注射した後に、用量依存的なPB AMLの生着が検出されたが、5×104〜106のhCD34+hCD38+細胞または106のhCD34-細胞の注射は、検出可能な生着を生じなかった(図1b-f, 表1)。移植されたhCD34+hCD38- LSCは、hCD45+hCD34+hCD38-細胞ならびにhCD45+hCD34+hCD38+およびhCD45+hCD34-のAML非幹細胞を生じ、初代LSCのin vivoでの自己複製能および分化能の両方が実証されている。原発性AML疾患をin vivoでよりよく再現することに加えて、循環PB AML細胞の存在は、単一のレシピエントにおける複数の時点にわたる個々の患者のAMLの試験を可能にし、AMLの発症、増殖、増悪、治療の効果、再発のすべてをマウスに再現することができるため、これは、新生仔NOD/SCID/IL2rg nullモデルの顕著な利点である。
連続移植により、AML BM hCD34+hCD38-細胞の機能的自己複製能および分化能(LSCの決定的な特性)が実証された。二次および三次のレシピエントにおいて、104および103の少量の精製AML BM hCD34+hCD38-細胞の注射はそれぞれ、充分高率な長期生着を生じ、hCD34+hCD38- LSCおよびhCD34+hCD38+/hCD34-芽球を生じた(図1b, g-j, 表1)。一次レシピエント同様、ソートされたhCD34+hCD38-細胞の二次および三次のレシピエントは全て、AMLの生着を示した(図1b, 表1)。対照的に、5×104〜106のhCD34+hCD38+の精製BM細胞も105〜2×106のhCD34-の精製BM細胞も、二次レシピエントおよび三次レシピエントのPBおよびMBにおいて検出可能な白血病を開始させなかった(図1b, i, j)。これらの知見は、LSCがhCD34+hCD38-のままであり、その間異種BM微小環境が、連続移植過程の間のヒトLSCの自己複製および分化を支持することを実証している。ヒトAMLは、患者から三次レシピエントへと1年を超えてin vivoで累積的に維持され、このことは、LSCの長期生着能および自己複製能を実証している。
正常なヒト造血は、連続移植過程において観察されず、レシピエントPBの赤血球および血小板は全てマウス起源であった(図2a)。わずか数十個の正常造血幹細胞からヒト血小板が産生されることを考慮に入れると、これは、選択的に白血病幹細胞が生着し、マウス体内において白血病を形成していることの証拠である。さらに、レシピエントPBのヘモグロビン濃度および血小板計数は、白血病負荷(leukemic burden)に反比例し、これは、AML疾患の進行に特徴的な正常な造血の抑制に類似している(図2b)。レシピエントの大腿骨および脾臓の巨視的外観により、AML疾患と一致する赤血球形成の抑制が確認され(図2c)、これは、ソートされたhCD34+hCD38+およびhCD34-AML細胞または正常BM hCD34+hCD38-細胞が移植された際には観察されなかった知見である。レシピエントの脾臓サイズは症例間で変動したが、腫大した脾臓は、ヒトAML細胞を一貫して含んだ(データ示さず)。これらの知見は、NOD/SCID/IL2rg nullマウスにおけるLSC生着およびAML増殖(expansion)が、ヒト白血病誘発を再現するという、さらなる機能的証拠を提供する。
ヒト初代(ここで初代とは、患者より直接得られた細胞であることを意味するため、培養などにより患者細胞の性質が人為的影響を受けていない)LSCのための微小環境ニッチはin vivoで規定されていないが、ヒトLSCの自己複製および分化した白血病性子孫の増殖によって特徴付けられる首尾よい長期生着は、NOD/SCID/IL2rg nullマウスのBMがヒトLSCの生存および増殖のための支持的微小環境を提供する能力を実証している。すなわち、環境はヒトでなくマウスであるものの、ヒト白血病幹細胞の機能的、細胞学的特徴を維持することができる環境を、本移植システムは有している。LSCのための微小環境ニッチを直接位置決めするために、レシピエントの大腿骨切片を、静脈内LSC注射の3日後に試験した。hCD34+AML細胞は、正常な「マウス」造血幹細胞の微小環境ニッチの部位(Zhang, J. et al. Identification of the haematopoietic stem cell niche and control of the niche size. Nature 425, 836-841 (2003); Calvi, L.M. et al. Osteoblastic cells regulate the haematopoietic stem cell niche. Nature 425, 841-846 (2003);およびArai, F. et al. Tie2/angiopoietin-1 signaling regulates hematopoietic stem cell quiescence in the bone marrow niche. Cell 118, 149-161 (2004))である骨内膜表面を裏打ちしていることが見出され、一方でBM腔の中心領域は、照射に起因してほぼ細胞が消失した空洞に近い状況であった(図2d)。骨内膜領域の細胞は、形態学的に均一ではなく、典型的に丸い細胞のみならず紡錘型の細胞および他の異型形状の細胞もCD45を発現し、丸形の細胞だけでなく紡錘形の細胞もヒト白血病細胞であって骨内膜細胞や血管内皮細胞ではないことを実証している(図5a-b)。CD45とCD34との同時局在は、共焦点顕微鏡画像によっても確認された(図5c-f)。hCD34+AML細胞は、移植の4ヶ月後(このとき、BMにはAML細胞が充満している)に、マウス骨芽細胞に隣接する骨内膜表面に優先的に残留する(図2e, g)。定量的分析により、LSCがBMの骨内膜領域にホーミングしてそこに存在し続けることが実証された(図2f)。これらの細胞はAML患者から得られたので、ソートされたhCD34+hCD38-細胞は、LSC同様正常HSCも含んでいる可能性がある。しかし、ソートされたhCD34+hCD38-細胞のレシピエントにおいて正常なヒト造血の生着が存在しないことは、これらの細胞の大多数(数的にも機能的にも)が悪性腫瘍起源であることを示している。これらの知見は、AML進行の間の正常な造血の抑制が、共用のBM微小環境ニッチについての競合の結果であり得ることを示唆している。
治療剤のin vivo試験は、AMLに対する新規治療の開発を大いに促進し、個々の患者のための治療戦略の最適化を導き得る、いわゆる個人医療(テーラーメード医療)の実現に繋がると期待される。本発明者らは、シトシンアラビノシド(Ara-C)のin vivo抗白血病細胞毒性を、AMLが生着したNOD/SCID/IL2rg nullレシピエントにおいて試験した(図3a,左)。シトシンアラビノシドは、AMLの治療のための標準的な化学療法剤である。Ara-C処置したマウスにおいて、PB AML負荷は、8日目までに顕著に減少した。細胞毒性効果には、全ての処置マウスにおいて貧血および血小板減少症が伴い、1匹の動物では軽症の肝毒性が生じ、これは、Ara-Cの既知の毒性プロフィールと一致する(図3a,右)。AMLを有する患者の場合と同様、1回のAra-C処置の効果は一過性であり、その後、PB AML細胞計数の回復によって実証されるとおり、16〜28日目までにAMLが再発した。
この薬物試験モデルを使用して、本発明者らは、ヒトAML幹細胞画分および非幹細胞画分の化学受容性を試験した。Ara-C処置の3日後のBMのフローサイトメトリー分析により、hCD34+hCD38-LSCでは、AML非幹細胞画分と比較して、統計的に有意にAra-Cに対する抵抗性が高いことが明らかになった(図3b)。化学療法抵抗性の白血病細胞の局在を同定するために、本発明者らは、Ara-C処置の3日後の大腿骨切片を使用して、ヘマトキシリン−エオシン(HE)およびTUNEL染色を実施した。組織学的研究により、骨内膜表面を裏打ちするTUNEL陰性生存細胞から、骨髄腔の中心のアポトーシス細胞が顕著に分離されていることが実証された(図3c)。免疫染色により、骨内膜に隣接する細胞はその表面上にhCD34を発現するがhCD38は発現せず、オステオポンチン+骨芽細胞に隣接することが確認された(図3d,e)。Ara-C処置マウス由来の103のAnnexinV-7AAD-hCD34+hCD38-細胞の注射は、二次レシピエントにおいてヒトAMLを発症させたが、hCD34+hCD38+細胞もhCD34-細胞もヒトAMLを発症させず、このことは、稀なLSCの化学療法抵抗性がAML再発の原因であることを示している。BrdU取り込みおよびHoechst/Pyronin Y染色によって実証されるように、hCD34+hCD38- AML細胞は、hCD34+hCD38+細胞およびhCD34-細胞と比較して、比較的細胞周期静止である(図3f)。Ara-Cを含む多数の化学療法剤は、細胞周期依存的な細胞毒性を発揮するので、LSCの細胞周期静止は、LSCの化学療法剤抵抗性の基礎をなす機構の1つである可能性がある。
LSCはin vivoでAMLを排他的に開始し得、従来の化学療法薬物に対して抵抗性であるので、化学療法抵抗性のLSCを特異的に標的化する新規治療戦略が、ヒトAMLにおける疾患再発を予防するために必要とされる(Guzman, M.L. et al. Preferential induction of apoptosis for primary human leukemic stem cells. Proc Natl Acad Sci U S A 99, 16220-16225 (2002);Guzman, M.L. et al. The sesquiterpene lactone parthenolide induces apoptosis of human acute myelogenous leukemia stem and progenitor cells. Blood 105, 4163-4169 (2005);およびTaussig, D.C. et al. Hematopoietic stem cells express multiple myeloid markers: implications for the origin and targeted therapy of acute myeloid leukemia. Blood 106, 4086-4092 (2005))。最近、細胞表面マーカーCD44は、マウスおよびヒトのLSCのホーミングおよび生着の両方において重要であると報告された(Jin, L., Hope, K.J., Zhai, Q., Smadja-Joffe, F. & Dick, J.E. Targeting of CD44 eradicates human acute myeloid leukemic stem cells. Nat Med 12, 1167-1174 (2006)およびKrause, D.S., Lazarides, K., von Andrian, U.H. & Van Etten, R.A. Requirement for CD44 in homing and engraftment of BCR-ABL-expressing leukemic stem cells. Nat Med 12, 1175-1180 (2006))。LSCのホーミングの妨害によるAMLの開始の予防に加えて、既に生着していてAML非幹細胞を生じることができるLSCを根絶し得る治療を開発することが、緊急の課題である。原発性AML患者における非幹細胞画分と比較した、LSCの網羅的なトランスクリプトーム分析は、個々の患者のレベルで、LSCにおいて特異的に発現される分子標的を同定するための強力なツールである。LSCの機能的特性を保持するAMLモデルマウスの開発は、LSCおよび非LSCを提供することにより、包括的な遺伝子プロファイリングを促進し得る。4人の原発性AML患者および対応する4匹のレシピエントマウスのBMからの、機能的に規定された初代ヒトLSCの予期される単離の後、本発明者らは、hCD34+hCD38-細胞とhCD34+hCD38+細胞とを比較するために、包括的遺伝子発現プロファイリングを実施した。患者BM由来のhCD34+hCD38-細胞とレシピエントマウスBM由来のhCD34+hCD38-細胞との間の遺伝子発現プロフィールの高い相関は、LSCにおける遺伝子発現シグネチャが、連続移植の間マウスBM微小環境において比較的安定のままであったことを示している(図4a, 図6)。この結果は、レシピエントマウスBM由来のhCD34+hCD38-細胞の分析が、患者BM由来のhCD34+hCD38-細胞の分析を補完する情報を提供できることを示している。従って、本発明者らは、各々の元の原発性AML BMおよび対応するレシピエントマウスBM由来のhCD34+hCD38+細胞と比較して、hCD34+hCD38-細胞において一貫して濃縮された遺伝子を明確にするために、遺伝子セット濃縮分析を実施した(図4b)。LSC画分において最も顕著に濃縮された遺伝子セットは、MAPK(mitogen-activated protein kinase)、SRC(v-src sarcoma viral oncogene homolog)、FYN(FYN oncogene related to SRC, FGR, YES)およびPXN(パキシリン)などの遺伝子を含むインテグリン経路であった(Cursi, S. et al. Src kinase phosphorylates Caspase-8 on Tyr380: a novel mechanism of apoptosis suppression. Embo J 25, 1895-1905 (2006)およびJanes, S.M. & Watt, F.M. New roles for integrins in squamous-cell carcinoma. Nat Rev Cancer 6, 175-183 (2006))。これらのインテグリン関連遺伝子を介したシグナル伝達は抗アポトーシス性であり、腫瘍細胞における細胞周期停止をもたらすか、あるいは白血球付着を調節する。さらに、本発明者らは、教師なし階層的クラスタリングによって、LSCにおいて統計的信頼度で示差的に発現される遺伝子を同定した(図4c)。その遺伝子は、LSC特異的治療の潜在的な標的である、転写因子、アポトーシスのネガティブレギュレータおよび膜貫通分子をコードする(図4c)。LSCにおける、細胞周期をG0からG1に推進するサイクリンA(CCNA1)の相対的下方調節は、細胞周期分析によって明らかなように、LSCにおける%G0頻度の増加と適合するように思われる(図3f)。対照的に、BAALC(brain and acute leukemia, cytoplasmic)(Tanner, S.M. et al. BAALC, the human member of a novel mammalian neuroectoderm gene lineage, is implicated in hematopoiesis and acute leukemia. Proc Natl Acad Sci U S A 98, 13901-13906 (2001))(その発現は、AML患者の予後の悪さと相関する)は、LSCにおいて高度に発現される。さらに、BMI-1との相互作用を介してポリコーム複合体を形成し、転写リプレッサーとして作用するリングフィンガータンパク質1(RING1)もまた、LSCにおいて過剰発現される。NOD/SCID/IL2rg nullマウスにおいて機能的に規定された集団を比較する、より多数のAML患者を試験する包括的トランスクリプトーム分析が、将来LSC特異的分子標的の検出に必要とされる。
1)長期生着性で自己複製性のLSCは、AML患者およびレシピエントマウスのBMにおいて排他的に存在すること;
2)LSCが骨芽細胞リッチな骨内膜領域にホーミングしてそこに定着し、増殖すること;
3)骨芽細胞リッチな領域内の大多数のLSCがG0期にあり、Ara-C処置に対して抵抗性であってこれがAML再発に寄与すること;および
4)マウスBM中に生着したLSCが、表現型、機能および遺伝子発現パターンにおいて患者のAML LSCの特徴を保持し、これにより個々のAML患者における新規LSC特異的治療標的の同定が可能となること。
Claims (10)
- 4週齢以下の非成体NOD/SCID/IL2rgnullマウスにヒト急性骨髄性白血病患者由来の白血病幹細胞含有物を移植し、該マウスを生育させることを含む、ヒト白血病細胞が選択的に増幅し、骨髄の骨芽細胞が豊富に存在する領域に隣接する骨内膜表面にヒト白血病幹細胞が存在するマウスの製造方法。
- 請求項1記載の方法により得られるマウス由来の白血病幹細胞含有物を、別の4週齢以下の非成体NOD/SCID/IL2rgnullマウスに移植し、該マウスを生育させる工程を1回以上含む、ヒト白血病細胞が選択的に増幅し、骨髄の骨芽細胞が豊富に存在する領域に隣接する骨内膜表面にヒト白血病幹細胞が存在するマウスの製造方法。
- 白血病幹細胞含有物移植後の生育期間が4週以上である、請求項1または2記載の方法。
- ヒト白血病細胞の増幅がマウス末梢血中で生じていることを特徴とする、請求項1〜3のいずれか1項に記載の方法。
- 白血病幹細胞含有物の由来する個々の患者における白血病の病態を再現することを特徴とする、請求項1〜4のいずれか1項に記載の方法。
- 請求項1〜4のいずれか1項に記載の方法により得られる、ヒト白血病細胞が選択的に増幅したマウス。
- a)請求項6記載のマウスに被験物質を投与する工程、および
b)該マウスにおける白血病の改善を評価する工程、
を含む、ヒト急性骨髄性白血病治療剤のスクリーニング方法。 - c)該マウスにおける被験物質の副作用をモニターする工程、
をさらに含む、請求項7記載の方法。 - a)ヒト急性骨髄性白血病患者もしくは請求項6記載のマウスから単離したhCD34+hCD38+細胞およびhCD34+hCD38-細胞、ならびに正常なヒト臍帯血もしくは骨髄またはヒト化マウスより単離したhCD34+hCD38+細胞およびhCD34+hCD38-細胞における遺伝子発現を網羅的に検出する工程、ならびに
b)正常および白血病におけるhCD34+hCD38+細胞とhCD34+hCD38-細胞との間で示差的に発現される遺伝子を同定する工程、
を含む、ヒト白血病幹細胞マーカー遺伝子の同定方法。 - 該マーカー遺伝子が静止期ヒト白血病幹細胞のマーカー遺伝子である、請求項9記載の方法。
