Description
数十年間にわたって、臨床医は、特定の疾患での不溶性タンパク質凝集物の形成に注目してきた。アルツハイマー病(Selkoe、1997、2002)では、CNSでのβ−アミロイド含有プラークの存在が、神経変性及び認知症に関係している。同様に、他の神経変性疾患も、脳内にタンパク質凝集を伴うことが最近になって発見された。例えば、クールー、クロイツフェルト・ヤコブ病及びウシ海綿状脳障害などのプリオン疾患は、プリオンタンパク質(PrP)のアミロイド沈着と関連している。ハンチントン病などのポリグルタミンリピート疾患も、ニューロンサイトゾル及び核内封入体と関連している(DiFigliaら、1997)。これらの封入体は、アミロイドと同様に着色する原線維からなる(Scherzingerら、1997)。最後に、パーキンソン病では、基底核の細胞の細胞質中に存在するレビ小体として知られている封入体は、タンパク質α−シヌクレインのアミロイド様凝集体を含有する。(Conwayら、2000;Serpellら、2000)
壊滅的な慢性神経変性障害を有する患者でのニューロンの消失は、多数の因子に起因し、その大部分(例えば、酸化ストレス、イオンのアンバランス、代謝の欠損、神経伝達物質のアンバランス、神経毒性)はこのような疾患の全てに共通する(Doble,1999)。明らかに特定の疾患に独特な因子でも、構造変化及び他の変化をもたらす自己化合物(self−compound)の細胞外沈着、さらにしばしばプラーク形成に達する凝集の点での変化を含む一定の共通する特徴を共有する(Hardy及びSelkoe,2002)。
Cop1とも称されるコポリマー1は、非病原性の合成ランダムコポリマーであり、4種のアミノ酸:L−グルタミン酸(E)、L−アラニン(A)、L−チロシン(Y)及びL−リシン(K)をおよそ1.5:4.8:1:3.6の比で、不規則な順序で含有するポリペプチドの不均一な混合物である。その作用様式は論争の的のままであるが、Cop1は明らかに、ヒト多発性硬化症(MS)、マウスで研究された関連自己免疫状態の実験的自己免疫脳脊髄炎(EAE)の進行を遅らせる。酢酸グラチラマーとしても知られているCop1の一形態は、いくつかの国で、Copaxone(登録商標)(Teva Pharmaceutical Industries Ltd.,Petach Tikva,Israel)の商品名で多発性硬化症の治療に関して承認されている。
CAG突然変異により生じる初期の病理学的、分子及び細胞異常の研究を可能にする、ハンチントン病の様々なマウスモデルが確立されている。HD R6/2トランスジェニックマウスモデルを、本発明でのin vivo試験系として選択した。これらのマウスは、ハンチンチンをコードするCAGリピート長の長いヒトハンチントン病遺伝子のエキソン1を過剰発現する(Mangiariniら、1996)。HD R6/2トランスジェニックマウスは、5〜6週齢という早期に行動運動欠失(behavioral−motor deficits)を示す。行動異常は、8週齢までは現れず、その後、低体重、握り締め(clasping)、振戦及び痙攣を伴う進行性の重度な神経表現型、さらに10〜14週齢での早期の死亡が生じる(Carterら、1999)。
(i)動物
8〜13週齢のC57BL/6J系のマウスは、Animal Breeding Center of The Weizmann Institute of Science(Rehovot,Israel)により供給された。実験で使用する前に、ケタミン80mg/kg及びキシラジン16mg/kgを腹腔内投与することによりマウスに麻酔をかけた。ハンチンチンをコードするヒト遺伝子を過剰発現するトランスジェニックR6/2マウスは、Jackson Laboratoryから得た。Institutional Animal Care及びUse Committee(IACUC)により確立された規則に従い全ての動物を取り扱った。
コポリマー1(中央MW:7200ダルトン)は、Teva Pharmaceutical Industries Ltd.(Petach Tikva,Israel)により供給された。
免疫のために、マウスの側腹部の1カ所に、リン酸緩衝生理食塩水(PBS:100μl)に溶かしたCop1を皮下注射した。対照マウスには、ビヒクルのみを注射した。
麻酔をかけたC57B BL/6Jマウスの右眼に、強膜の上部で27ゲージ針で穴を開け、30ゲージ針を有する10μlハミルトンシリンジを硝子体からなるべく遠くに挿入した。マウスに、生理食塩水に溶かしたL−グルタミン酸塩を全体積で1μl(200nmol)眼内注射した。
実験の終了72時間前に、RGCを標識した。マウスに麻酔をかけ、定位装置に置いた。頭蓋を暴露し、乾燥及び清浄に保った。ブレグマを同定し、マーキングした。注射の所定の点は、脳表面から2mmの深さ、前後軸においてブレグマの2.92mm後、正中線に対して0.5mm側方であった。左右の半球の所定の座標上の頭皮を開口した。次いで、ハミルトンシリンジを使用して、神経トレーサー(neurotracer)染料FluoroGold(生理食塩水中5%の溶液;Fluorochrome,Denver,CO)を適用し(1μl、各半球で0.5μl/分の速度)、創傷の上の皮膚を縫合した。染料の逆行取り込みは、生存細胞のマーカーを提供する。
マウスに致死用量のペントバルビトン(170mg/kg)を与えた。その眼を摘出し、網膜を切り離し、パラホルムアルデヒド(PBS中4%)中の平坦な全体マウントとして調製した。同一のサイズ(0.5mm2)の4〜6個の選択領域からの標識細胞数を測定した。選択した領域は、視神経円板からほぼ同じ距離(0.3mm)に位置させて、視神経円板からの距離の関数としてのRGC密度における変動を克服した。マウスに与えられた治療を知らない観察者が、蛍光顕微鏡(倍率×900)で、領域を計測した。各網膜での1領域当たりのRGCの平均数を算出した。ニューロンを保護する際の様々なワクチン製剤の有効性を、生存RGCを計測することにより測定する。

グルタミン酸は、CNS中に低濃度で通常は存在するアミノ酸であり、主要な興奮性神経伝達物質として役立っている。しかしながら、多くの神経変性疾患では、グルタミン酸レベルが毒性レベルまで上昇して、細胞損傷をもたらしている。したがって、このモデルを選択し、Cop1神経保護ワクチン接種を確立し、治療レジメンを最適化した。グルタミン酸毒性を、C57Bl/6Jマウスの眼にグルタミン酸を眼内注射し、次いで、視覚信号を脳に伝達するニューロンであるRGCのその後の死を測定することにより、評価する。
グルタミン酸誘発RGC死に対するCop1ワクチン接種の用量の影響を研究するために、完全フロイントアジュバント中で乳化されたCop1(CFA;Cop1 25、75又は225μg、全体積100μl)をC57BL/6Jマウスの側腹部の1カ所に皮下注射し、7日後に、グルタミン酸(200nmol)をマウスの硝子体に注射した。7日後に、生存RGCを計測した。予め免疫しなかった場合のグルタミン酸毒性後のRGC死の量を保護可能な細胞に100%と見なした。図1に示されている結果により、Cop1を25μg又は75μg投与することにより、有効なワクチン接種が得られた。
Cop1免疫の神経保護作用を維持するために、Cop1の反復注射を評価した。この目的は、長期RGC生存効果を最適化しうる反復Cop1注射の最適なレジメンを決定することであった。
Cop1ワクチン接種に対する細胞免疫応答と神経保護作用との相間関係
2種のex vivoマーカー、T細胞刺激指数及びインターフェロン−γ(IFN−γ)は、効果と相関している。刺激指数は、Cop1応答性T細胞がリンパ球集団内に存在する程度を示している。IFN−γ分泌は、「Th1」サブタイプのT細胞に特徴的である。したがって、これらのマーカーは、細胞免疫応答をプロファイリングする手段をもたらす。
ハンチントン病のin vivo動物実験系
Cop1ワクチン接種の有益な効果を、HD R6/2トランスジェニックマウス試験系を使用して、神経保護効果に関して実験した。R6/2トランスジェニックマウスは、複数のCAGリピートの挿入を含む突然変異ヒトハンチンチン遺伝子を過剰発現する(Mangiariniら、1996)。これらのマウスは、5〜6週齢の早期から始まる進行性の行動−運動欠損を示し、これは、10〜13週目で早発死をもたらした。症状には、低体重、抱擁、振戦及び痙攣が含まれた(Carterら、1999)。
ハンチントン病でのヒト臨床試験
ヒト試験の第1の目的は、ハンチントン病患者での、プラセボに対するCop1(Copaxone(登録商標)又は他のCop1製剤)20mg又は2×20mg用量の連続投与の忍容性、安全性及び免疫学的応答を評価することである。この試験の第2の目的は、次の神経学的臨床パラメータ、統一ハンチントン病評定スケール(Unified Huntington’s Disease Rating Scale(UHDRS))及び全運動スケール(Total Motor Scale(TMS))を測定することによる、Cop1投与後のHD疾患患者の神経学的経過を評価することである。
神経変性疾患は、原因は異なるが、同様に広がる。このセクションでの実験では、凝集β−アミロイドの眼内注射により生じたニューロン消失は、正常なマウスより、免疫欠損マウスにおいて著しく多かったことを示す。天然のCD4+CD25+調節T細胞(Treg)を除去又は添加することにより、神経変性は減弱又は増大した。β−アミロイドを伴わない網膜由来抗原又はコポリマー1のワクチン接種は、眼ニューロン消失を低減させた。マウス海馬切片では、活性化T細胞に遭遇した小膠細胞は、凝集β−アミロイドの細胞毒性を克服した。これらの知見により、「防御的自己免疫」の概念は支持され、所与のT細胞ベースのワクチン接種は、毒性の種類に関わりなく、特定の部位で防御的であることを示しており、局所で活性化されたT細胞は、ニューロンの損傷への抵抗に役立つ小膠細胞表現型を誘発することを示唆している。Cop1又は関連化合物のワクチン接種により、又はTreg抑制を低減する化合物での処置により、アルツハイマー及び他の神経変性疾患を阻止するか、遅らせることができる。
(vii)動物
Association for Research in Vision及びOphthalmology(ARVO)の研究における動物使用に関する決議に従い、マウスを処理した。全て特定病原不在の、8から13週齢のオスのC57BL/6J野生型、BALB/c/OLA野生型及びヌードマウスが無菌条件下に、Animal Breeding Center of The Weizmann Institute of Science(Rehovot,Israel)により供給された。マウスを光及び温度調節室で飼育し、各実験で年齢を合わせた。マウスに、ケタミン(80mg/kg;Ketaset,Fort Dodge,IA)及びキシラジン(16mg/kg;Vitamed,Ramat−Gan,Israel)のi.p.投与により麻酔をかけた。組織を切除する前に、マウスを致死量のペントバルビトン(170mg/kg;C.T.S.,Kiryat Malachi,Israel)で殺した。
記載されているように(Pepperbergら、1991)、Con Aで親和性クロマトグラフィー処理することにより、ウシ光受容体内レチノイド結合タンパク質(IRBP)を網膜抽出物から精製した。Puig et al.(1995)により改変された、Buczylko及びPalczewski(Palczewskiら、1994)の方法で、Con Aカラム流のフロースルーから、ウシS抗原(アレスチン)を調製した。PBS中にホモジェナイズした同系網膜から、全網膜ホモジネート(WRH)を調製した。Sigma−Aldrich,St.Louis,MO.から、オボアルブミン(OVA)、Con A及びβ−アミロイドペプチド1〜40(Aβ1〜40)を購入した。Aβ(1〜40)ペプチドを内毒素不含の水に溶かし、記載されているように(Ishiiら、2000)、Aβ(1〜40)をインキュベーションすることにより、β−アミロイド凝集物を形成させた。酢酸グラチラマー(Copaxone(登録商標);Cop−1)はTeva Pharmaceuticals Ltd.(Petach Tikva,Israel)から購入した。
マイコバクテリウム・ツベルクローシス(Mycobacterium tuberculosis)(5mg/ml;Difco)を含有する等体積のCFA(Difco,Detroit,MI)中でそれぞれ乳化されているIRBP(50μg)、S−抗原(50μg)、Aβ1〜40(50μg)、WRH(600μg)又はCop−1(75μg)で、成体マウスを免疫した。エマルション(全体積0.15ml)を側腹部の1カ所に皮下注射した。対照マウスには、CFA中のPBS、又はPBSのみを注射した。
材料及び方法、セクションI(v)に記載されているように標識を実施した。
麻酔をかけたC57BL/6J又はBALB/c/OLAマウスの右眼に、強膜の上部で27ゲージ針で穴を開け、30ゲージ針を有するハミルトンシリンジを硝子体からなるべく遠くで挿入した。各マウスに、L−グルタミン酸塩(400nmol;Sigma−Aldrich)又は凝集Aβ1〜40(50μM;Sigma−Aldrich)を含有するPBSを全体積1μlで注射した。
実験期間の終了時に、マウスに、致死用量のペントバルビトン(170mg/kg)を与えた。その眼を摘出し、網膜を切り離し、PBS中4%のパラホルムアルデヒド中の平坦な全体マウントとして調製し、同一のサイズ(0.076mm2)の4〜6個の領域からの標識細胞を計測した(Schoriら、2001b)。各網膜で、1領域当たりのRGCの平均数を算出した。対側(損傷なし)眼でのRGCの数も計測して、内部対照として役立てた。
眼内グルタミン酸注射の48時間後に、マウスを屠殺し、その眼を除去し、凍結切片化(cryosectioning)のために処理した。凍結切片を3.7%ホルマリン中、室温で10分間固定し、PBSで2回洗浄した。切片を100%メタノールに−20℃で15分間移し、エタノール100%、95%及び70%中で2回、5分間順次洗浄し、次いで、PBSと共に10分間インキュベーションした。浸透化のために、プロテイナーゼKを用いて、プロテアーゼを室温で20分間消化した。製造者指示に従い、その場でのアポトーシス検出キット(R&D Systems,Minneapolis,MS)を使用して、DNA断片の開放端を標識した。ストレプトアビジン−フルオレセイン結合体と共に供給されたフルオレセイン検出キットを使用して、標識された末端を検出した。蛍光顕微鏡を使用して、フルオレセイン染色された細胞を可視化した。
慣用の手順により調製された脾細胞をラット抗マウスフィコエリトリン(PE)結合CD25抗体と共にインキュベーションし、これに、抗PEビーズ(Becton−Dickinson,Bactlab Diagnostic,Haifa,イスラエル)とのインキュベーションを続けた。洗浄した後に、脾臓細胞を「欠失感受性」プログラムを伴うAutoMacs(Miltenyi Biotec,Bergisch Gladbach,ドイツ)にかけた。回収された集団を、FACSsort(Becton Dickinson,Franklin Lakes,NJ)(Kipnisら、2002a)により分析した。
リンパ節(腋下、鼠径部、表在、頚部、下顎及び腸間膜)及び脾臓を採取し、すりつぶした。T細胞をT細胞カラム(R&D Systems)で精製した(負の選択により富化)。富化されたT細胞を抗CD8マイクロビーズ(Miltenyi Biotec)と共にインキュベーションし、負に選択されたCD4+T細胞を、PBS/2%ウシ胎児血清中のPE−結合抗CD25抗体(30μg/108細胞)と共にインキュベーションした。次いで、これらを洗浄し、抗−PEマイクロビーズ(Miltenyi Biotec)と共にインキュベーションし、AutoMACSを用いて磁気分離にかけた。残った細胞をカラムから精製CD4+CD25+細胞(Treg)として溶離した。5×105細胞/mlを脾臓由来APC(3000radで照射)及び抗CD3抗体0.5μg/mlと共に含有し、マウス組換えIL−2 100単位(mrIL;R&D Systems)を補足した培地中で、CD4+CD25−T細胞からなる負の画分(エフェクターT細胞、Teff)をさらに4日間活性化させた。
免疫の10日後に、マウスを殺し、その流入領域リンパ節を切除し、微細な金網に通した。L−グルタミン(2mM)、2−メルカプトエタノール(5×10−5M)、ペニシリン(100IU/ml)、ストレプトマイシン(100IU/ml)及び自系マウス血清1%(vol/vol)を補足したRPMIを含有する刺激培地中で、洗浄したリンパ球(2×106細胞/ml)を関連抗原(IRBP1〜20又は凝集Aβ1〜40、それぞれ10μg/mlで)で活性化した。37℃、相対湿度90%、CO27%で48時間のインキュベーションした後、リンパ球をPBSで洗浄し、計測し、毒性用量(50μM)の凝集Aβ1〜40を硝子体内注射した後1時間以内に、自系マウスに腹腔内注射した。
記載されているように(Butovskyら、2001)、新生(0日齢)BALB/c/OLAマウスの大脳皮質から、小膠細胞を精製した。IFN−γ(20ng/ml;R&D Systems)、β−アミロイド(Sigma−Aldrich;凝集Aβ1〜4025μM)又は活性化T細胞(1ウェル1.5×105)を培地に12時間加えた。処理の後に、小膠細胞をPBSで3回洗浄し、海馬切片で適用するために調製した。
8〜10日齢のBALB/c/OLAマウスを断頭し、その脳を迅速に無菌条件下に取り出し、1%のL−グルタミン(Gibco)を補足した最小必須培地(MEM;Gibco、Carlsbad,CA)からなる氷冷調製培地にpH7.35で入れた。大脳前頭極を取り出し、ビブラトーム(Pelco,Redding,ドイツ)の上で、前方から開始して、脳を350μm水平切片に切断した。海馬を含む切片を、6ウェルプレート中、空孔サイズ0.4μmのFalcon細胞培養インサート上で培養した。培地は、50%のMEM、25%のハンクス平衡塩類溶液(Gibco)、25%の正常ウマ血清、2%のグルタミン、インスリン−トランスフェリン−亜セレン酸ナトリウム補足10μg/ml(Boehringer Mannheim,Mannheim,ドイツ)、グルコース2.64mg/ml(Braun,Melsungen、ドイツ)、ストレプトマイシン0.1mg/ml、ペニシリン100U/ml及びビタミンC0.8μg/ml(全て、Sigma−Aldrichから)を含有していた。器官型海馬切片培養物(OHSC)を35℃、5%のCO2を含む加湿雰囲気下で、24時間インキュベーションしたが、その間、切片を未処置で放置するか、1ウェル当たり4×105小膠細胞で処置した。インキュベーション期間終了時に、培地にヨウ化プロピジウム(PI)(5μg/ml;Sigma)を30分間加えることにより、組織損失を評価した。次いで、過剰のPIを培地と共に洗い流し、切片を顕微鏡のために調製し、可視化した。OHSC中での神経細胞死を定量するために、Image−Proソフトウェア(Media Cybernetics,Carlsbad,CA)を使用して、各切片でのPI強度を評価した。両側スチューデントのT検定を使用して、特定の処置でのPI染色強度を未処置の対照の強度と比較した。
網膜タンパク質は、グルタミン酸中毒に対する保護的なT細胞ベースの応答を誘発しうる
我々は、異なる遺伝的背景を有するマウスは、有害な条件に抵抗するその能力において異なることを以前に示した(Schoriら、2001b;Kipnisら、2001;Schoriら、2002)。この違いによって、少なくとも一部では、T細胞依存性保護的応答を示す能力において、系統に関連する変化が生じる(Kipnisら、2001)。ミエリンタンパク質は眼でのグルタミン酸毒性に対してマウスを保護することができないことが観察されたこと及び網膜タンパク質によってラットでのグルタミン酸毒性に対する保護が成功したことを考慮して(Schoriら、2001b;Mizrahiら、2002)、我々は、網膜タンパク質でのマウスの免疫は、グルタミン酸毒性に暴露した後のそのニューロン生存率を改善しうるのではないか、もしそうであるならば、そのワクチンが、同じ部位に注射された他の脅威的化合物(凝集β−アミロイド)に対しても有効であろうということに興味を持った。グルタミン酸(400nmol)をC57BL/6Jマウスの右眼に注射し、48時間後に、末端デオキシヌクレオチジルトランスフェラーゼビオチン−dUTPニック末端標識(TUNEL)した網膜凍結断片を調べた。アポトーシス細胞死が、RGC層では観察された(図10A)。毒性用量の眼内グルタミン酸(400nmol)を注射する6日前に、この系統のマウスに、CFA中の全網膜タンパク質のホモジネートをワクチン接種すると、1週間後の検査により、CFA中に乳化したPBSを注射した年齢及び系統が適合する対照マウスで観察された生存率よりも高いRGCの生存率が明らかになった(それぞれ2239.7±153.3及び1480.6±176.2、P<0.0001;両側スチューデントのt検定)。この発見は、網膜成分のワクチン接種は、免疫マウスのグルタミン酸毒性に抵抗する能力を高めることを示している。図10Bは、ワクチン接種が、正常対照での数に対するパーセンテージとして表されるRGC損失を著しく低下させたことを示している(平均±SEM;図10B)。
β−アミロイドの毒性に抵抗する能力は、T細胞に依存する
神経毒性に対してする保護のための生理学的に適した抗原は、それ自体毒性の化合物(グルタミン酸)ではなく、損傷部位に位置する自己抗原であることが判明したが、次いで、我々は、毒性が同じ部位に限られる場合、同じワクチン接種が、別の毒性自己化合物に対して有益であるかどうかを試験した。この仮定を、凝集β−アミロイドの毒性に対するワクチン接種の効果を試験することにより調べた。凝集β−アミロイド(Aβ)1〜40(5又は50μM)をC57BL/6Jマウスの右眼に注射した。推定されているこの化合物とアルツハイマー病での眼神経障害との関連のためではなく(Bakalashら、2002;Schwartz,2004)、βアミロイドがRGC死をもたらしうるので(Jenら、1998)、このモデル(β−アミロイドの眼内注射)を選択し、したがって、その使用により、抗原特異性の概念をさらに調べることができた。凝集β−アミロイドの眼注射の1又は2週間後に、生存RGCを計測した。1週間後には、生存可能なRGCは2257±77(5μM注射)及び2071±30(50μM注射)であり、2週間後には、これらは、2062±41(5μM)及び1952±21(50μM)であった。非免疫マウスでは、全部で3445±57個のニューロンが計測された。同じ実験条件で、ビヒクルのみの注射により生じた毒性は、正常な網膜ではRGCの5%より多くには影響を与えなかった。図11Aは、β−アミロイド誘発ニューロン消失を、正常野生型網膜でのRGCの平均数に対するパーセンテージとして示している。図11B及び11Cは、それぞれPBS及び凝集Aβ1〜40を眼内注射された後にマウスから切除された全マウント網膜の代表顕微鏡写真を示している。
天然の調節CD4+CD25+T細胞は、網膜でのβ−アミロイド毒性に対抗する体の能力を制限する
介入の不在下では、凝集Aβ1〜40の毒性に抵抗するマウスの能力はT細胞に依存するという前記観察(図11)によって我々は、凝集β−アミロイドの毒性に自発的に抵抗する神経組織の能力は、他のCNS損傷の場合において記載されたように(Kipnisら、2002a;Schwartz及びKipnis,2002a)、天然に生じる調節CD4+CD25+T細胞によって抑制されるかどうかを調べることを思いついた。もしそうであれば、このような制御の除去又は脆弱化は、β−アミロイド関連神経変性状態に対する保護に必要な自己免疫T細胞を利用する付加的な方法として役立つはずである。
T細胞は、小膠細胞が炎症性細胞毒性表現型を展開することを防ぐ
自己免疫T細胞が自己化合物を克服するために役立つ方法の1つは、小膠細胞の活性の制御による(Schwartzら、2003)ことを、我々は以前に提案していた。我々のグループは、器官型海馬切片培養(OHSC)を使用して、ラット小膠細胞を凝集Aβ1〜40で予備処置すると、神経組織に対して細胞毒性になり、MHC−IIを発現するその能力が抑制される(Butovskyら、未刊行の観察)ことを示した。したがって、我々は、凝集Aβ1〜40に暴露されたネズミ小膠細胞は、細胞毒性になるかどうか、もしそうであるとしたら、活性化T細胞は、毒性を克服しうるかどうかを決定するためにin vitro実験を実施した。マウス小膠細胞を凝集Aβ1〜40に暴露した後、マウスOHSCに加えると、非処置か、非免疫小膠細胞で処置されたOHSCで観察されたよりもかなり多い神経死が生じた(図16)。しかしながら、凝集Aβ1〜40に暴露する時点で、加える小膠細胞を活性化エフェクター(CD4+CD25−)T細胞にも暴露すると、この消失は著しく低下した(図16A)。様々に処置されたOHSC及び非処置対照の代表顕微鏡写真を図16B(1〜4)に示す。これらの知見は、適切に制御された量の活性化T細胞への小膠細胞の暴露は、小膠細胞が細胞毒性になることを妨げるだけでなく、それらを神経保護性にするという主張を支持する。このバイオアッセイは、抗原提示を必要としないので、in vitroでアッセイされた小膠細胞毒性は、MHC−II発現の欠失を反映しないことを特記する。
アルツハイマー病に関するin vivo動物実験系
Cop1ワクチン接種の有利な作用は、トランスジェニックマウス試験系を使用して、神経保護作用の発揮に関して調べることができる。
前記セクションIIに記載の本発明の結果は、凝集Aβ1〜40及びグルタミン酸などの他の毒性物質により誘発される毒性を中和する治療用ワクチンを開発する際には、毒性物質が、よくあるように、同じ部位に全て位置している場合、同じワクチンを使用することができることを示唆している。前記セクションIIで使用されたマウスモデルでは、2種の神経毒性自己化合物を眼に注射し、同じ抗原、即ち、眼に存在するタンパク質に由来するペプチドをワクチン接種することにより、いずれに対しても保護が達成された。我々は、この知見を、損傷部位に構成的に存在する優性自己抗原は、この部位での脅威的条件に対する自己保護的抗原であるという原理の証拠として解釈する。さらに我々は、天然に生じるCD4+CD25+調節T細胞(Treg)の欠失は、このような抗原に対する自発応答を、したがって、凝集β−アミロイドの毒性作用に抵抗する能力を高めうることを示している。しかしながら、治療戦略として、我々は、部位特異的自己タンパク質自体を用いてではなく、コポリマー1、自己抗原の合成弱アゴニストを用いてのワクチン接種を提案する(Schoriら、2001b;Kipnisら、2000;Angelovら、2003;Ziemssenら、2002)。それというのも、後者は、自己免疫疾患、自己免疫の本質的に不十分な制御を伴って起こりうる危険を誘発するリスクなしに、保護的ワクチンとして使用することができるためである(Schwartz及びKipnis,2002)。別の戦略として、我々は、Tregの活性を弱める何らかの操作を使用することを提案する(Kipnisら、2003)。
パーキンソン病(PD)は、黒質でのドーパミン作動性ニューロンの進行性損失及び線条体での神経伝達物質ドーパミンの欠失を特徴とする神経変性運動障害である。現在、PDの最適なモデルは、1−メチル−4−フェニル−1,2,3,6−テトラヒドロピリジン(MPTP)動物モデルである。MPTPは、ヒト及び非ヒト霊長類の両方でパーキソニズムを誘発することが判明している。ドーパミン作動性ニューロンへと細胞内輸送される、MPTPの代謝産物である1−メチル−4−フェニルピリジニウムイオン(MPP+)により生じる神経毒性は、ヒトPDを模倣すると考えられ、PDにおける神経保護を研究するための良好なモデルをもたらす。MPTPマウス試験系又はパーキンソン病のための他の適切なモデルを使用して、神経保護作用の行使に関して、Cop1免疫の有利な効果を試験する。Cop1でのPDのための神経保護治療は、神経変性作用及び疾患進行の速度を減衰させることができる。
(xix)動物
Animal Breeding Center of The Weizmann Institute of Science(Rehovot,Israel)から供給された8〜13週齢のC57BL/6J系統のオスのマウスを、Institutional Animal Care及びUse Committee(IACUC)により作成された規則に従い取り扱う。
Cop1(中央MW:7200ダルトン)は、Teva Pharmaceutical Industries Ltd.(Petach Tikva,Israel)に由来する。MPTP.HCl(Sigma)を0.9%のNaClに溶かした。
免疫のために、PBS(100μl)に溶かしたCop1をマウスの側腹部の1カ所で、皮下注射した。対照マウスにはビヒクルのみを注射した。
異なる群のオスのSJLマウスを、例えば1日に2時間間隔で4回、PBS中のMPTP.HCl(Sigma)18〜20mg/kgを腹腔内注射で、又は1日2回2日間にわたってMPTP20mg/kgを皮下注射することにより様々なスケジュールのMPTPで処置して、強度及び時間経過において異なる変性プロセスを誘発させる。対照マウスには、対応する用量のビヒクルのみを投与する。
パーキンソンMPTP齧歯類モデルでのCop1の効果
オスのC57BL/6Jマウスに、2時間間隔で4回、PBS中のMPTP.HCl20mg/kgを腹腔内注射する。最後のMPTP投与の12時間後に、Cop1(PBS中のCop1 75μg又は150μg/マウス)又はPBS(対照群)をMPTP処置された動物に投与する。
