Description
本発明において、「擬微小重力環境」とは、宇宙空間等における微小重力環境を模して人工的に作り出された微小重力(simulated microgravity)環境を意味する。こうした擬微小重力環境は、例えば、回転で生じる応力によって地球の重力を相殺することにより実現される。すなわち、回転している物体は、地球の重力と応力のベクトル和で表される力を受けるため、その大きさと方向は時間により変化する。結局、時間平均すると物体には地球の重力(1g)よりもはるかに小さな重力しか作用しないこととなり、宇宙空間によく似た「擬微小重力環境」が実現される。
本発明では、擬微小重力環境を実現するために、回転式のバイオリアクターを使用する。そのようなバイオリアクターとしては、例えば、RWV(Rotating-Wall Vessel:US 5,002,890)、RCCS(Rotary Cell Culture SystemTM:Synthecon Incorporated)、3D-clinostat、ならびに特開平8−173143号、特開平9−37767号、および特開2002−45173号に記載されているようなものを挙げることができる。これらのバイオリアクターの中には、1軸回転式のものと2軸以上の多軸回転式のものがあるが、本発明では1軸回転式のバイオリアクターを用いることが好ましい。多軸回転式(例えば、2軸式のclinostat等)では、ずれ応力(シェアストレス)を最小化することができず、またサンプル自体も回転するため、1軸回転式のようにベッセル内にふわふわと浮かんだ状態を再現することができないからである。このふわふわと浮かんだ状態が、特別な細胞足場材料なしに大きな3次元的組織塊を得るための重要な条件となる。なかでも、RWVおよびRCCSはガス交換機能を備えているという点で優れている。
本発明では軟骨組織構築の材料として骨髄細胞を用いる。本発明に用いられる骨髄細胞とは、骨髄由来の分化・増殖能力を有する未分化細胞であり、特に骨髄由来の間葉系幹細胞が好ましい。前記細胞は、樹立された培養細胞株のほか、軟骨組織の移植を必要とする対象(患者)の生体から単離された骨髄細胞を好適に用いることができる。該細胞は移植対象者から採取された後、常法に従って結合組織等を除去して調製することが好ましい。また、常法により一次培養を行い、予め増殖させてから用いてもよい。さらに移植対象者から採取した培養は、凍結保存されたものであってもよい。つまり、予め採取した骨髄細胞を凍結保存しておき、必要に応じて利用することもできる。
本発明では、トランスフォーミング成長因子(TGF-β: transforming growth factor-β)、デキサメタゾン、骨形成タンパク質(BMP: Bone Morphogenetic Proteins)、および骨誘導性転写因子遺伝子を含むウイルスベクターから選ばれるいずれか1または2以上を添加した培地を用いる。用いられる培地としては、MEM培地、α-MEM培地、DMEM培地等、骨髄細胞の培養に通常用いられる培地を、細胞の特性に合わせて適宜選んで用いることができる。
本発明で用いられるデキサメタゾンは、骨・軟骨細胞分化促進作用を有し、骨軟骨組織構築においては汎用されている分化誘導因子である。通常骨分化のために必要とされるデキサメタゾンの量は軟骨分化のために必要とされる量の10分の1程度である。すなわち、軟骨分化を目的とする場合には100nMが適切な濃度であるが、骨分化を目的とする場合には10nMが適切な濃度となる。
本発明で用いられる骨形成タンパク質(BMP: Bone Morphogenetic Proteins)は、骨形成作用を有する因子として特定されたタンパク質である。現在までに、13のBMPのcDNAがクローニングされている。BMP-1を除くBMP(たとえば、BMP-2、BMP-4、BMP-5、BMP-6、BMP-7およびBMP-9等)はすべてTGF-βスーパーファミリーに属する。BMPは未分化な間葉系細胞や、骨芽細胞、軟骨細胞に作用して骨形成を促進するだけでなく、骨以外の種々の組織にも存在し、これらの細胞に作用してその分化の方向性を制御する。本発明では、BMPのなかでも、TGF-β系のシグナル伝達経路を経て、骨芽細胞の分化に必須の転写因子Runx2(runt-related gene 2)の転写を促進する作用を有するBMP-2や米国で認可承認を受けているBMP-7(OP-1)を用いることが好ましい。
本発明で用いられる骨誘導性転写因子は、未分化の細胞を骨に分化誘導する、骨誘導性の転写因子で、例えばCbfa1、Cbfb、osterix等を挙げることができる。Cbfa1は1993年京都大学の小川らによってクローニングされ、大阪大学の小守らにより間葉系幹細胞から骨芽細胞に分化誘導するのに必要不可欠であることが確認された転写因子である(Komori, T. et al., (1997) Cell 89, 755-764)。Cbfbは造血に関与する因子として知られてきたが、近年Runx2との相互作用により骨形成に必須の役割を担っていることが確認された。また、Osterixは骨組織特異的に発現するZinc-Finger型転写因子であり、骨芽細胞の分化等、骨形成に重要な役割を担っていることが確認されている。
細胞は、二次元培養(静置培養)である程度増殖させた後、トリプシン等を用いて細胞を分散した後、TGF-βおよびデキサメタゾンを添加して擬微小重力環境下において培養して軟骨組織を形成する(軟骨組織構築工程)。次いで、上記したデキサメタゾンあるいは骨形成タンパク質を含む培養液に変えて擬微小重力環境下において培養するか、Cbfa1を導入したウイルスベクターを感染させて擬微小重力環境下において培養する(骨組織構築工程)。なお、Cbfa1感染後の培養には、当該ベクターを含まない骨形成性の培養液(例えば、前記した上記したデキサメタゾンあるいは骨形成タンパク質を含む培養液等)を用いることができる。
本発明の骨・軟骨ハイブリッド組織は、表層周辺に骨組織が形成され内部は軟骨組織が維持された構造を有する。これは、静置培養では軟骨組織は徐々に骨化もしくは繊維化が進むが、RWVによる擬微小重力環境下での培養では内部に軟骨組織が閉じ込められ、組織にかかる重力も特殊な状態になり、培養液の浸透する表層から骨組織形成が進むからである。
1.材料
(1)ウサギ骨髄由来間葉系幹細胞の調製
ウサギ骨髄由来間葉系幹細胞は、2週齢のJW系家兎(14days, 雌)の大腿骨よりManiatopou1osらの方法(Maniatopou1os, C., Sodek, J., and Me1cher, A. H. (1988) Ce11 Tissue Res. 254, p317-330)に従って採取した。採取した細胞を、10% FBS(Sigma社製)およびAntibiotic-Antimycotic(GIBCO BRL社製)を含むDMEMで3週間にわたって培養し、増殖させた(図1)。
下表1に示す組成で、以下の各培養液を調製した。
1)Chondrogenic Medium
2)Osteogenic Medium (DEX)
3)Osteogenic Medium (BMP2)
4)Osteogenic Medium (Cbfa1感染溶液)
なお、感染後はOsteogenic Medium (DEX)と同じ組成の培養液を用いた
マウスの骨芽細胞から単離したTotal RNAからAMV reverse transcriptaseを用いてcDNAを合成し、これを鋳型としてCbfa1のcDNA(GenBank Accession No. AF010284:配列番号1)に特異的なプライマーを用いてPCRによりcbfa1 cDNAを増幅して得た。

Cbfa1 cDNAはTA cloning vector (Invitrogen, pCR II-TOPO)にクローニングして大量調製した。Cbfa1 cDNAを制限酵素のSpe IとEcoR Vで切り出し、平滑末端化した後、Adenovirus Cre/loxP kit(宝酒造, 6151)を用いてコスミドベクターpAxCALNLwのSwa Iサイトに挿入し、Kitの説明書に従って組換えアデノウイルスを作製した。作製したウイルスの力価は、約1011PFU/mlの値を示し、感染効率は非常に高いことが確認された。
(1)ウサギ骨髄由来間葉系幹細胞の培養
上記のようにして調製したウサギ骨髄由来間葉系幹細胞をRWVべッセルに播種し、2週間の静置(二次元)培養をChondrogenic mediumで行った。2週間後、擬微小重力条件下でChondrogenic mediumを用いて二週間培養したのち、Chondrogenic medium(control)、Osteogenic medium (DEX)、Osteogenic Medium (BMP2) 10ml中で、3週間にわたってRWVバイオリアクター(Synthecon社製)を用いての培養を行った。Cbfa1についてはhondrogenic mediumを用いて二週間培養した直後に、Osteogenic medium (Cbfa1感染用)で1時間培養した後、Osteogenic medium (DEX)に切り替えて上記と同様5週間にわたってRWVバイオリアクターを用いて回転培養を行った。実験のスキームを図2に示す。
比較として、同様の培養物をコニカルチューブ中で5週間静置培養した。
RWV培養軟骨組織および静置培養軟骨組織は、培養5週間後にヘマトキシリン・エオジン(HE)、サフラニンO、およびトルイジンブルーで組織染色を行い、軟骨基質産生能を評価した。
5週間培養して得られたRWV培養軟骨組織と、静置培養軟骨組織について、それぞれ抗コラーゲンタイプIモノクローナル抗体(Developmental Studies Hybridoma Bank製)、抗コラーゲンタイプIIモノクローナル抗体(第一ファインケミカル社製)、および抗オステオカルシン抗体を用いた免疫染色を行った。染色像を図8および図9に示す。
図3に示されるように、どの培養液でも静置培養(pellet culture)よりRWV培養の方が大きく、丈夫な組織ができていることが肉眼所見から分かる。3種のosteogenic mediumでの培養に比べ、chondrogenic mediumによる培養で構築した組織の方が白っぽいことが分かる。

Claims (5)
- 以下の工程を含む、骨・軟骨ハイブリッド組織の構築方法であって:
1) 骨髄細胞および/または間葉系幹細胞を二次元培養により増殖させる、
2) 細胞を分散後、TGF-βおよびデキサメタゾンを添加して擬微小重力環境下において培養する、
3) デキサメタゾンを含み、TGF-βを含まない培養液に交換して、擬微小重力環境下においてさらに培養する、
前記擬微小重力環境が、回転で生じる応力によって地球の重力を相殺することにより擬微小重力環境を地上で実現するRWV (Rotating-Wall Vessel)バイオリアクターを用いて得られるものであり、該RWVバイオリアクターの回転速度が、形成された組織塊が液中に浮いている状態になるように調整されている、上記方法。 - 骨髄細胞の播種密度が106〜107/cm3、RWVの回転速度が直径5cmベッセルに対して8.5〜25rpmの条件下で培養が行われる、請求項1に記載の方法。
- 前記骨髄細胞が軟骨組織の移植を必要とする対象から採取された細胞である、請求項1または2に記載の方法。
- 骨・軟骨ハイブリッド組織が、表層周辺に骨組織が形成され内部は軟骨組織が維持された構造を有する、請求項1〜3のいずれか1項に記載の方法。
- 請求項1〜4のいずれか1項に記載の方法によって構築された骨・軟骨ハイブリッド組織。
