同様に、DEPDC1は膀胱癌、乳癌、子宮頸癌、胆管細胞癌、CML、NSCLC、リンパ腫、骨肉腫、前立腺癌、SCLC、軟組織腫瘍において過剰発現することが、確認された。
しかしながら、PCT/JP2006/302684の中に開示されているように、DEPDC1のsiRNAは、癌細胞の増殖を抑制することができる。これらの結果は、DEPDC1が多くの癌細胞の増殖において重要な役割を果たすことを証明する。

上記の如く、MPHOSPH1(M期リン蛋白1;GenBankアクセッション番号NM_016195;配列番号:1、2)およびDEPDC1(DEP領域含有1;GenBankアクセッション番号:BM683578)、特に、その2つのバリアント(DEPDC1V1(配列番号:3、4)およびDEPDC1V2(配列番号:5、6)は、以前、cDNAマイクロアレイ技術を用いて、さまざまな癌において過剰発現するものとして同定された。MPHOSPH1は、以前G2/M移行で特異的にリン酸化されるタンパク質のうちの1つとして同定され、プラス方向誘導キネシン関連のタンパク質として特徴づけられた(Abaza A et al. J Biol Chem 2003、278:27844-52.)。特に、MPHOSPH1は、以前、細胞分裂において重要な役割を果たすプラス方向誘導分子モーターであり、HeLa細胞の終期までの後期の間に、紡錘体の中心部に集積すると実証された(Abaza A et al. J Biol Chem 2003、278:27844-52; KamimotoT et al. J Biol Chem 2001、276:37520-8.)。
MPHOSPH1のcDNAは、3つの領域、:NH2―kinasinモータードメイン(NH2-kinasin motor domain)、中心高次コイル―軸ドメイン(central coiled coil-stalk domain)およびC球状尾部ドメイン(C-globular tail domain)、から成る1780アミノ酸のタンパク質をコードする。これらのデータは、MPHOSPH1がNH2タイプキネシン関連のタンパク質であることを示唆する。
本発明の文脈において、アミノ酸改変は、結果として、本来のアミノ酸側鎖の性質を保存することが好ましい(保存的アミノ酸置換として公知の方法)。アミノ酸側鎖の性質の例は、疎水性アミノ酸(A、I、L、M、F、P、W、Y、V)、親水性アミノ酸(R、D、N、C、E、Q、G、H、K、S、T)、および共通して、以下の官能基または特性を有する側鎖である:脂肪族側鎖(G、A、V、L、I、P);ヒドロキシ基含有側鎖(S、T、Y);イオウ原子含有側鎖(C、M);カルボン酸およびアミド含有側鎖(D、N、E、Q);塩基含有側鎖(R、K、H);ならびに芳香族含有側鎖(H、F、Y、W)。ただし、括弧でくくられた文字は、アミノ酸の一文字記号を示す。
意図される癌としては、限定はされないが、膀胱癌、乳癌、子宮頸癌、胆管細胞癌、CML、結腸直腸癌、胃癌、NSCLC、リンパ腫、骨肉腫、前立腺癌、腎癌、SCLC、軟組織腫瘍が挙げられる。適切な製剤には顆粒も含まれる。適切なアジュバントは、文献(Johnson AG.(1994)C1in.Microbio1.Rev.7:277-89.)に記載されている。例示的なアジュバントは、リン酸アルミニウム、水酸化アルミニウム、およびミョウバンを含むが、これに限定されない。さらに、リポソーム製剤、薬物が直径数μmのビーズへ結合している顆粒製剤、および脂質が前記ペプチドに結合している製剤が、便利に用いられうる。投与の方法は、経口投与、皮内注射、皮下注射、静脈内注射などであってもよく、全身投与または標的腫瘍の付近への局所的投与を含みうる。本発明のペプチドの用量は、処置すべき疾患、患者の年齢、体重、投与の方法などにより適切に調整することができる。用量は、普通、0.001mg〜1000mg、好ましくは0.01mg〜100mg、より好ましくは0.1mg〜10mgであり、好ましくは数日に1回から数ヶ月に1回投与されるが、当業者は適切な用量および投与の方法を容易に選択することができ、これらのパラメーターの選択および最適化は、十分に通常の技術の範囲内である。
DNAに基づいた送達技術(delivery technologies)の例は、「ネイキッドDNA(naked DNA)」、促進化ブピビカイン(bupivicaine)、ポリマー、ペプチド媒介性)送達、カチオン性脂質複合体、および粒子媒介性(「ジーンガン(gene gun)」)または圧力媒介性送達を含む(例えば、米国特許第5,922,687号参照)。
治療的投与または免疫化に有用な幅広い種類の他のベクター、例えば、アデノおよびアデノ随伴ウイルスベクター、レトロウイルスベクター、チフス菌(Salmonella typhi)ベクター、解毒化炭疽毒素ベクターなどが、当技術分野において公知である。例えばShata MT、et al. (2000)Mol. Med. Today 6:66-71; Shedlock DJ and Weiner DB. et al. (2000)J Leukoc.Biol.68:793-806;そして、Hipp JD、et al.、In Vivo 14(2000):571-85を参照。
あるいは、本発明のペプチドを抗原提示細胞へ固定化した後、細胞をワクチンとして対象に投与することができる。例えば、エクスビボ投与は以下の段階を含みうる:
a:対象から抗原提示細胞を収集する段階、および
b:段階aの抗原提示細胞を本発明のペプチドと接触させる段階。
例えば、本方法は以下の段階を含みうる:
a:対象から抗原提示細胞を収集する段階、
b:段階aの抗原提示細胞を本発明のペプチドと接触させる段階、

段階dにより得られた細胞傷害活性をもつCD8陽性T細胞は、ワクチンとして対象に投与することができる。

b:T細胞を以下のペプチドと接触させる段階。

本発明の文脈において、「活性された細胞障害性T細胞」は、IFN―ガンマ産生、IFN―ガンマ放出および腫瘍細胞の細胞死を誘導する。
核酸またはそれらを含むベクターは有効にT細胞に導入することができる、ここで、T細胞は好ましくは患者由来である。都合のよいことに、本発明は、迅速にまた容易に強い癌細胞殺傷特性を持つ改変T細胞を供給するため患者自身のT細胞(または他の哺乳類のそれら)を迅速に改変することができるoff−the−shelf組成物を提供する。
一般に、抗腫瘍免疫は、以下のような免疫応答を含む:
MPHOSPH1および/またはDEPDC1を発現している細胞を含む腫瘍に対する細胞障害性リンパ球の誘導、
MPHOSPH1および/またはDEPDC1を発現している細胞を含む腫瘍を認識する抗体の誘導、そして、
抗腫瘍サイトカイン産生の誘導。
CD4+ T細胞はまた抗腫瘍免疫において重要であるため、ペプチドの抗腫瘍免疫誘導作用は、これらの細胞の活性化効果を指標として用いて評価することができる。
本発明は以下の実施例により例証されるが、それらに限定されない。しかしながら、本願明細書において記載されている材料、方法およびそのようなものは、本発明の態様を例示しているが、決して本発明の範囲を制限することを意図しない。よって、そのときに記載されている材料、方法およびそれらと同様または同等なものは、本発明の実施をまたは試験に用いうる。
材料および方法
細胞株
A24LCL細胞(HLA―A24/24)、ヒトB―リンパ芽球腫細胞株、T2細胞およびCOS7は、ATCCから購入された。
HLA―A*2402およびHLA―A*0201分子に結合するMPHOSOH1またはDEPDC1由来の9―merおよび10―merペプチドは結合予測ソフトウェア「BIMAS」(bimas.dcrt.nih.gov/cgi-bin/molbio/ken_parker_comboform)を用いて予測された(Parker KC et al. J Immunol. (1994);152no(1):163-75. Kuzushima K et al. Blood. (2001); 98 no(6):1872-81.)。これらのペプチドは、Sigma(札幌、日本)によって、標準的な固相合成方法に従って合成されて、逆相HPLCによって精製された。
純度(>90%)およびペプチドの同一性はそれぞれ、HPLC分析およびマススペクトル分析によって決定された。ペプチドは、20mg/mlでジメチルスルホキシド(DMSO)に溶解されて―80度Cで保存された。
単球由来の樹状細胞(DC)は、HLAに提示されるペプチドに対するCTL反応を誘導する抗原提示細胞(APC)として用いられた。
DCは他で報告されているように、インビトロで生成された。(Nukaya I et al.、Int. J. Cancer (1999)80、92-7.Tsai V et al. J.Immunol (1997)158:1796-802.)簡潔には、Ficoll―Paque(Pharmacia)溶液によって正常なボランティア(HLA―A*2402および/またはHLA―A*0201)から分離された末梢血単核細胞(PBMCs)は、それらを単球画分について濃縮するために、プラスチック組織培養フラスコ(Becton Dickinson)への粘着性により分離した。単球の濃縮集団を、2%加熱不活性化自己血清(AS)を含むAIM−V(Invitrogen)中、1000U/mlのGM−CFS(Kirin Brewery Companyより提供)および1000U/mlのIL−4(Genzyme)の存在下で培養した。
培養7日後、サイトカイン産生DCを、AIM−V中、3mcg/mlのβ2-ミクログロブリンの存在下に、20 mcg/mlの合成ペプチドを用いて20℃、4時間パルスした。これらのペプチドパルスされたDCは、MMC(30分間の30mcg/ml)によって不活性化されて、Dynabeads M―450 CD8(Dynal)およびDETACHa BEADTM(Dynal)による陽性選択によって得られた自己由来CD8+T細胞と1:20の比率で混合された。これらの培養物を48ウェルプレート(Corning)に配置した;各ウェルには、AIM-V/2%AS0.5ml中、1.5×104個のペプチドパルスしたDC、3×105個のCD8+ T細胞、および10ng/mlのIL-7(Genzyme)を含む。3日後、これらの培養物に、最終濃度20IU/mlのIL-2(CHIRON)を追加した。7日目および14日目に、T細胞を、自己ペプチドパルスしたDCでさらに再刺激した。DCは、上記と同じ方法により毎回調製した。CTLは、21日目における3回目のペプチド刺激後に、ペプチドパルスしたA24LCL細胞またはT2細胞に対して試験した。
CTLは、Riddell SR et al.(Walter EA et al. (1995)N Engl J Med 333:1038-44.; Riddell SR、et al. (1996)Nature Med. 2:216-23.)に記載された方法と類似の方法を用いた培養で拡張させた。合計5×104個のCTLを、40ng/mlの抗CD3モノクローナル抗体(Pharmingen)の存在下で、二種のヒトB―リンパ芽球腫細胞株を含む25mlのAIM-V/5%AS中に再懸濁した。培養の開始から1日後、120IU/mlのIL-2を培養物に加えた。培養物に、5日目、8日目、および11日目に30IU/mlのIL-2を含む新鮮なAIM-V/5%ASを供給した。
希釈は、96ウェルの丸底マイクロタイタープレート(Nalge Nunc International)において0.3個、1個、および3個のCTL/ウェルになるように行なわれた。CTLは、7×104細胞/ウェルの二種のヒトB―リンパ芽球腫細胞株、30ng/mlの抗CD3抗体、および125U/mlのIL-2と共に、5%AS含む合計150 mcl/ウェルのAIM-V中で培養された。10日後、50 mcl/ウェルのIL-2を、IL-2が最終濃度において125U/mlになるように培地へ加えた。CTLのCTL活性は14日目に試験され、CTLクローンは上記と同じ方法を用いて拡張された。
特異的なCTL活性を検討するために、IFN―ガンマELISPOTアッセイおよびIFN―ガンマELISAが行われた。簡潔には、ペプチドパルスされたA24―LCLまたはT2細胞(1x104/ウェル)は、スティミュレーター細胞として調製された。
48ウェルの培養細胞または限界希釈後のCTLクローンは、レスポンダー細胞として用いられた。IFN―ガンマELISPOTアッセイおよびELISAは、製造手順に基づき行われた。
イプシュタインバーウイルス(Epstein Bar virus)が変換されたHLA―A24 B―LCLs(A24LCL)は、樹立された。Jiyoye、EB―3、COS7、HT1376、RT―4およびJ82は、アメリカンタイプカルシャーコレクション(Rockville、MD)から購入された。A24LCL、JiyoyeおよびEB―3は、10%のウシ胎児血清(GEMINI Bio製)および1%の抗生剤溶液(シグマ)を含むRPMI1640において維持された。COS7、HT1376、RT―4およびJ82は、適切な培地および抗生物質において維持された。COS7およびHEKのトランスフェクションは、FUGENE6(ロシュ)を用いて行われた。HLA―A*0201分子を発現している安定的クローン、HEK―A2細胞は、pcDNA6.2―HLA―A2プラスミドのトランスフェクションによって樹立され、5mcg/mlのBlastcidinSの存在下で、限界希釈法によって分離された。
ペプチド特異的なCTLの誘導のために、免疫処置は、マウスごとに、50mcl(100mcg)のHLA―A24拘束性ペプチドおよび50mclのIFAを含む、100mlのワクチンの混合物を用いてされた。ワクチンは、0日目に最初の免疫を右側腹部において、そして、二回目を7日目に左の横腹において皮下注射された。14日目、ワクチン接種されたマウスの脾細胞は、いかなるインビトロ刺激もしないで、レスポンダー細胞に用いられ、ペプチドの有無にかかわらずパルスされたRLmale1細胞はIFN―ガンマELISPOTアッセイのスティミュレーター細胞として用いられた。
癌におけるMPHOSPH1およびDEPDC1発現上昇
cDNA―マイクロアレイを用いてさまざまな癌から得られた網羅的遺伝子発現プロプロファイルデータは、MPHOSPH1(GenBankアクセッション番号:NM_016195;配列番号:1)、そして、2つの異型を有したDEPDC1(GenBank Accession番号:BM683578);DEPDC1 V1(配列番号:3)およびDEPDC1 V2(配列番号:5)の発現が上昇することを示した。MPHOSPH1発現は、31の膀胱癌中30、36の乳癌中8、18の子宮頸癌中18、17の胆管細胞癌中5、31のCML中25、11の結腸直腸癌中6、14の胃癌中6、5つのNSCLC中5、7つのリンパ腫中7、10の骨肉腫中6、22の前立腺癌中7、18の腎癌中10および21の軟組織腫瘍中15において、対応する正常組織と比較して有意に上昇した。DEPDC1発現は、25の膀胱癌中23、13の乳癌中6、12の子宮頸癌中12、6つの胆管細胞癌中6、4のCML中3、4の結腸直腸癌中2、6のNSCLC(非小細胞肺癌)中6、7のリンパ腫中7、14の骨肉腫中10、24の前立腺癌中11、14のSCLC(小細胞肺癌)中14および31の軟組織腫瘍中22において、対応する正常組織と比較して有意に上昇した(表1)。

MPHOSHP1(SEQ ID No:2)由来のそれらのペプチドに対するCTLは、上記の「材料および方法」の項に記載されるプロトコールに従って生成された。
結果として生じるIFN―ガンマELISPOTアッセイによって評価されるように、検出可能な特異的なCTL活性を有するCTLは、図1Aおよび図2Aに示される。
図1Aにおいて、MPHOSPH1―A24―9―278(配列番号:7)によって刺激されたウェルナンバー#4の細胞は、コントロールと比較して強力なIFN―ガンマ生産を示した。
図2Aにおいて、MPHOSPH1―A24―10―278(配列番号:8)によって刺激されたウェルナンバー#8の細胞は、コントロールと比較して強力なIFN―ガンマ生産を示した。次に、陽性ウェルのこれらの細胞は拡張され、限界希釈された。
図1B(MPHOSPH1―A24―9―278(配列番号:7))および図2B(MPHOSPH1―A24―10―278(配列番号:8))に示すように、ペプチドパルスのない標的に対する活性と比較して、ペプチドパルスされた標的に対する高い特異的なCTL活性を有するCTLクローンが樹立された。
これらのペプチドに対して高められた樹立したCTLクローンは、MPHOSPH1およびHLA―A*2402を内因的に発現している標的細胞を認識する能力について検討された。
MPHOSPH1およびHLA―A*2402を内因的に発現する標的細胞の特異的モデルであるところの、全長MPHOSPH1遺伝子およびHLA―A*2402分子によってトランスフェクトされたCOS7に対する特異的CTL活性を、MPHOSPH1-A24-9-278 (SEQ ID NO: 7)で高められたCTLクローンをエフェクター細胞として用いて試験した。
全長MPHOSPH1によってトランスフェクトしたがHLA―A*2402はしないCOS7およびHLA―A*2402でトランスフェクトしたが全長MPHOSPH1はしないCOS7をコントロールとして調製した。
COS7に対する最も高い特異的なCTL活性を有するCTLクローンは、MPHOSPH1およびHLA―A24の両者によってトランスフェクトしたものであった。しかしながら、MPHOSPH1もHLA―A24によってもトランスフェクトしないCOS7に対しては有意な特異的CTL活性を示さなかった(図4)。
MPHOSPH1―A24―9―278(配列番号:7)ペプチドに対して高められた樹立したCTLクローンは、MPHOSPH1を内因的に発現している腫瘍細胞を認識するための能力を検討された。MPHOSPH1およびHLA―A24を内因的に発現するHT1376細胞に対するCTL活性は、MPHOSPH1―A24―9―278(配列番号:7)により高められたCTLクローンをエフェクター細胞として用いて試験された。
内因的にMPHOSPH1を発現するが、HLA―A24を発現しないJ82細胞およびRT―4細胞は標的細胞として用いられた。樹立したCTLクローンは、MPHOSPH1およびHLA―A24を両者とも発現するHT1376細胞に対して、高いIFN―ガンマ産生を示した。一方、このCTLは、MPHOSPH1を発現するがHLA―A24を発現しないJ82およびRT―4細胞に対しては有意なCTL活性を示さなかった(図5)。それは、MPHOSPH1―A24―9―278(配列番号:7)ペプチドがHLA―A24分子とともに腫瘍細胞表面に自然に加工され、CTLによって認識されることを明確に証明した。
H―2Kd分子(マウスMHCクラスIの1つ)がHLA―A24分子に対するペプチド・アンカーのモチーフに似ており、HLA―A24拘束性ペプチドと一部交差反応することは公知である。本発明者らは、次に、MPHOSPH1―A24―9―278ペプチドがインビボでCTLを誘導するかどうかを、このペプチドをBALB/cマウス(H―2kd)にワクチン接種することによって検討した。IFA接合ペプチドは、0および7日目にBALB/cマウスに皮下注射された。14日目に、脾細胞は収集され、ELISPOTアッセイのためのレスポンダー細胞として用いられた。ペプチドを注入されたすべてのマウス(Ani1〜5)の脾細胞は、IFA単独を注入されたコントロールマウス(nega1〜3)と比較して強力なIFN―ガンマの産生を示した。(図6)。このデータは、MPHOSPH1―A24―9―278ペプチドがインビボでもCTL反応を誘導できることを示した。
結果として、IFN―ガンマELISPOTアッセイによって評価された検出可能な特異的CTL活性を有するCTLは、図7に示される。図7Aに示すように、MPHOSPH1−A2−9−282(YIYDLFVPV(配列番号:9))で刺激されたウェルナンバー#1および#5の細胞は、コントロールと比較して、強力なIFN―ガンマ生産を示した。図7Bに示すように、MPHOSPH1―A2―9―638(RLAIFKDLV(配列番号:10))によって刺激されたウェルナンバー#8の細胞は、コントロールと比較して強力なIFN―ガンマ生産を示した。図7Cに示すように、MPHOSPH1―A2―10―1714(TMSSsKLSNV(配列番号:11))によって刺激されたウェルナンバー#4の細胞は、コントロールと比較して強力なIFN―ガンマ生産を示した。
DEPDC1由来のそれらのペプチドのためのCTLは、上記の「材料および方法」の項目に記載されているプロトコールに従って生成された。結果としてIFN―ガンマELISPOTアッセイによって評価されるように、検出可能な特異的CTL活性を有するCTLは図10に示される。図10Aに示すように、DEPDC1―A24―9―294(EYYELFVNI(配列番号:12))によって刺激されたウェルナンバー#10の細胞は、コントロールと比較して強力なIFN―ガンマ生産を示した。したがって、陽性ウェルのこれらの細胞は拡張され、限界希釈された。図10B(DEPDC1―A24―9―294(配列番号:12))に示すように、ペプチドをパルスしない標的に対する活性と比較してペプチドパルスされた標的に対して高い特異的CTL活性を有するCTLクローンが樹立された。DEPDC1―A24―9―294(EYYELFVNI(配列番号:12))(図11)によって刺激されたこのCTLクローンは、いずれのペプチドもパルスしない標的に対しては、少しも有意な特異的CTL活性を示さず、ペプチドパルスされた標的に対して強力な特異的なCTL活性を示した。この結果は、このCTLクローンがペプチド特異的な細胞障害性を有することを示唆する。
これらのペプチドに対して高められた樹立したCTLクローンは、DEPDC1およびHLA―A*2402を内因的に発現している標的細胞を認識するための能力が検討された。標的細胞が内因的にDEPDC1およびHLA―A*2402発現する特異的なモデルである、全長DEPDC1遺伝子およびHLA―A*2402分子によってトランスフェクトするCOS7、に対する特異的なCTL活性は、エフェクター細胞としてDEPDC1―A24―9―294(EYYELFVNI(配列番号:12))により増えたCTLクローンを用いて、試験された。全長DEPDC1によってトランスフェクトするがHLA―A*2402はしないCOS7およびHLA―A*2402をトランスフェクトさせるが全長DEPDC1はしないCOS7はコントロールとして調製された。CTLクローンは、DEPDC1およびHLA―A24の両者をトランスフェクトさせたCOS7に対して高い特異的なCTL活性を示した。しかしながら、DEPDC1もHLA―A24によってもトランスフェクトしないCOS7に対しては有意な特異的CTL活性を示さなかった(図12)。
DEPDC1―A24―9―294ペプチドに対して高められた樹立したCTLクローンは、DEPDC1を内因的に発現している腫瘍細胞を認識するための能力が検討された。DEPDC1およびHLA―A24を内因的に発現するHT1376細胞に対するCTL活性は、DEPDC1―A24―9―294によって高められたCTLクローンをエフェクター細胞として用いて試験された。J82細胞およびRT―4細胞は、内因的にDEPDC1を発現するがHLA―A24を発現しない標的細胞として用いられた。樹立したCTLクローンは、DEPDC1およびHLA―A24を発現するHT1376細胞に対して、高いIFN―ガンマ産生を示した。一方、DEPDC1を発現するがHLA―A24は発現しないJ82およびRT―4細胞に対しては、有意なCTL活性を示さなかった(図13)。それは、DEPDC1―A24―9―294がHLA―A24分子を有する腫瘍細胞表面に自然に加工され、CTLによって認識されることを明確に証明した。

IFN―ガンマELISPOTアッセイによる選択時に、検出可能な特異的CTL活性を有しているCTLは、図15および表6に示される。DEPDC1―A02―10―644((SLMIHTFSRC配列番号:240))によって刺激されたウェルナンバー#4および#7の細胞は、コントロールと比較して強力なIFN―ガンマの産生を示した。DEPDC1―A02―10―575(SLLPASSMLT(配列番号:241))によって刺激されたウェルナンバー#2の細胞は、コントロールと比較して強力なIFN―ガンマに産生を示した。DEPDC1―A02―10―506(QLCRSQSLLL(配列番号:243))によって刺激されたウェルナンバー#7の細胞は、コントロールと比較して強力なIFN―ガンマに産生を示した。DEPDC1―A02―10―765(KQFQKEYPLI(配列番号:244))によって刺激されたウェルナンバー#1の細胞は、コントロールと比較して強力なIFN―ガンマに産生を示した。DEPDC1―A02―10―395(IMGGSCHNLI(配列番号:249))によって刺激されたウェルナンバー#1の細胞は、コントロールと比較して強力なIFN―ガンマに産生を示した。

DEPDC1―A02―10―296ペプチド(YELFVNILGL(配列番号:254))およびDEPDC1―A02―9―653(CVLCCAEEV(配列番号:225))に対して高められた樹立したCTL株は、DEPDC1およびHLA―A2を内因的に発現している標的細胞を認識する能力について検討された。最初に、我々は、効率的に特異的なCTL反応を測定するためにHLA―A*0201(HEK―A2)を構成的に発現するHEK293細胞株を樹立した。DEPDC1およびHLA―A2を発現する標的細胞の特異的なモデルである、全長DEPDC1遺伝子をトランスフェクトさせたHEK―A2細胞に対する特異的なCTL活性は、エフェクター細胞としてDEPDC1―A02―10―296(YELFVNILGL(配列番号:254))またはDEPDC1―A02―9―653(CVLCCAEEV(配列番号:225))によって高めれた樹立したCTL株を用いて試験された。Mock発現ベクターをトランスフェクトしたHEK―A2、そして、DEPDC1由来のものに対応しないペプチドでパルスしたHEK―A2はネガティブコントロールとして調製された。樹立したCTL株は、DEPDC1をトランスフェクトしたHEK―A2に対して特異的なCTL活性を示した。一方で、当該CTL株は、トランスフェクトしたMock発現ベクターおよびDEPDC1―A02―9―674ペプチドまたはDEPDC1―A02―9―462ペプチドがパルスされたHEK―A2に対して、有意な特異的CTL活性を示さなかった(図17)。それは、DEPDC1―A02―10―296DEPDC1―A02―9―653ペプチドが、HLA―A2分子とともに標的細胞表面に自然に加工され、CTLによって認識されることを明確に証明した。
本発明のペプチドに対して樹立されるCTLは、強力な特異的CTL活性を示した。
これはMPHOSPH1―A24―9―278(配列番号:7)、MPHOSPH1―A24―10―278(配列番号:8)、MPHOSPH1―A2―9―282(配列番号:9)、MPHOSPH1―A2―9―638(配列番号:10)、MPHOSPH1―A2―10―1714(配列番号:11)、DEPDC1―A24―9―294(配列番号:12)、DEPDC1―A2―9―589(配列番号:192)、DEPDC1―A2―9―619(配列番号:195)、DEPDC1―A2―9―290(配列番号:197)、DEPDC1―A2―9―563(配列番号:209)、DEPDC1―A2―9―653(配列番号:225)、DEPDC1―A2―10―674(配列番号:228)、DEPDC1―A2―10―302(配列番号:230)DEPDC1―A02―10―644(配列番号:240)、DEPDC1―A02―10―575(配列番号:241)、DEPDC1―A02―10―506(配列番号:243)、DEPDC1―A02―10―765(配列番号:244)、DEPDC1―A02―10―395(配列番号:249)、DEPDC1―A02―10―224(配列番号:253)、DEPDC1―A02―9―297(配列番号:226)、DEPDC1―A02―10―296(配列番号:254)およびDEPDC1―A02―10―301(配列番号:255)のシーケンスがヒト免疫系を感作することが知られている他の分子由来のペプチドに相同であることを示唆する。この可能性を排除するため、BLASTアルゴリズム(http://www.ncbi.nlm.nih.gov/blast/blast.cgi)を用いて、ペプチド配列をクエリーとして相同性解析が行われた。有意な配列相同性は、示されなかった。

材料および方法
ペプチドおよびアジュバント
合成されたGMPグレードのペプチドは、ネオマルチペプタイドシステム(Neo Multi Peptide System)(MPS)(サンディエゴ、CA)より購入された。アジュバントとして、不完全フロイントアジュバント(IFA)(MONTANIDE*ISA51)が、用いられた。1mgの適切なペプチドは、1mgのIFAによって乳化された。
本発明者らは、免疫組織化学的解析を行った。手術または生検から得られた膀胱癌由来の腫瘍細胞または腫瘍組織は、それぞれのMPHOSPH1およびDEPDC1特異的ポリクローナル抗体によって染色された。染色のプロトコールは、以前に述べられたように(Kanehira M et al. Cancer Res.;67(7):3276-3285, 2007., Kanehira M et al. Oncogene. 2007 Apr 23; [Epub ahead of print])東京大学医科学研究所ヒトゲノムセンターにおいて確立された。HLA―A*2402発現は、SRL(立川、日本)で、行われ試験された
登録基準は、以下の通り;
1. 標準的な化学療法によって過去に治療をうけて失敗となった手術不能の再発性膀胱癌患者。
2. 日本の判定基準において一般状態が0または1である患者。
3. 20歳から80歳の患者
4. RECISTガイドラインは考慮しないが、治療前に画像検査(CT/MRI)によって原発腫瘍または転移と認められた患者
5. 前治療(手術、化学療法、放射線療法、温熱療法、他の免疫療法その他)後4週以上経過した患者
6. 3ヵ月以上の予後が予測される患者
7. 骨髄機能(白血球数2000以上、15000以下、血小板50000以上を)、肝機能(GOT150以下、GPT150以下、T―bil3.0以下)、腎機能(Cr3.0以下)の患者
8. HLA―A*2402を有する患者
9. MPHOSPH1および/またはDEPDC1の発現を有する患者の腫瘍
1. 妊娠中の患者
2. 授乳中の患者
3. 妊娠の意思のある患者
4. 制御困難な感染症を有する患者
5. 臨床試験期間中に以下の薬剤を必要とする患者
ステロイドの全身投与
免疫抑制薬の全身投与
6. 医師または治験責任医師によってこの試験に登録するべきと思われない患者
腫瘍がM phase phosphoprotein 1(MPHOSPH1)および/またはDEP domain containing 1 (DEPDC1)を発現する、 HLA―A*2402を有する登録された膀胱癌患者は、HLA―A*2402―拘束性エピトープペプチドMPHOSPH1―9―278(IYNEYIYDL(配列番号:7))および/またはDEPDC1―9―294(EYYELFVNI(配列番号:12))によって免疫された。それぞれのペプチドは、1mLの不完全フロイントアジュバント(IFA、MONTANIDE*ISA51)と組み合わせられて、週に一度腋窩あるいは鼠径部の病変に、皮下注射された。1コース4回注射と規定され、続いて1コース後、免疫学的および臨床的評価のため、採血並びにCT/MRIが行われた。
副作用の評価は、国立癌研究所―共通毒性判定基準第3版(NCI―CTC ver.3)に従って行われた。
これはこの研究のセカンダリーエンドポイントの1つで、我々はペプチド特異的なCTL反応が発生したかどうかを確認する。特異的なCTL反応の計測は、以下の通り;
末梢血単核細胞は回収され、適切なペプチドによって再刺激された。CTL反応は、IFN―ガンマELISPOTアッセイによって、14日目に試験された。
臨床反応の評価は、RECIST基準に従って行われた。
表7は、本臨床研究の概要を示す。症例3のグレード2の発疹を除き、重篤な副作用はなかった。マイナーレスポンス(症例3)およびミックスレスポンス(症例4)が得られた。MPHOSPH1の発現は5症例中4症例であったが、DEPDC1のそれは5症例中5症例であった。
症例2において、標準化学療法の失敗でかなり進行した膀胱癌をもつ72歳の女性が本臨床試験に登録された。図18において、彼女の腫瘍の抗原発現は、MPHOSPH1およびDEPDC1が強く発現することを示した。従って、我々はMPHOSPH1およびDEPDC1由来のエピトープペプチド2種をワクチン接種した。症例2は、膀胱癌の局所再発を有した。それは、RECIST基準基づき安定(SD)と評価された(図19)。
症例3において、標準化学療法の失敗でかなり進行した膀胱癌をもつ49歳の男性が、本臨床試験に登録された。DEPDC1のみが強く発現された(図20)。従って、我々は、DEPDC1由来のエピトープペプチドを単独でワクチン接種した。症例3は、膀胱癌の複数の肺転移を有した。右(図21)および左(図22)の肺葉転移において、進行率がワクチン接種後に低下した。特に、腫瘍のサイズが3コース後に減少した。図23は、RECIST基準に基づいた抗腫瘍効果を示す。転移性腫瘍の進行率がワクチン接種後に低下したことが明らかにされた。
それは、DEPDC1由来のエピトープペプチドを用いているワクチン接種によってマイナーレスポンスが得られることを示した。症例3の免疫学的評価に関して、特異的CTL反応は、ワクチン接種の前後で測定された。特異的CTL反応はワクチン接種後強く示された(図24)。それは、DEPDC1由来のエピトープペプチドによって誘導されるCTLが抗腫瘍効果を示しうることを明確に示した。
症例4において、標準的化学療法の失敗でかなり進行した膀胱癌をもつ74歳の男性が本臨床試験に登録された。MPHOSPH1およびDEPDC1は、彼の腫瘍で発現した(図25)。従って、我々は、MPHOSPH1およびDEPDC1由来の2種類のエピトープペプチドをワクチン接種した。症例4は、膀胱癌の局所再発を有していた。1コースのワクチン接種後、腫瘍のサイズは、RECIST基準に基づき、20%減少した(図26)。しかしながら、肺の新しい転移性病変が発生した。それは、MPHOSPH1およびDEPDC1由来の2種類のエピトープペプチドを用いるワクチン接種によってまちまちの反応が得られることを示した。
本臨床試験の理論的根拠は、以下に述べられる;
1. MPHOSPH1およびDEPDC1は精巣を除く正常組織においては発現されないので、両抗原は非常に腫瘍特異的である。
2. 強力で特異的なCTLはこれらのエピトープペプチドによって樹立されたことから、これらのペプチドは強い免疫原性を有すると考えられる。
3. 日本人集団の60%がHLA―A*2402をもつ。
4. これらのペプチドは、臨床試験に適用するのに化学的に十分安定である。
本研究の目的は、その毒性、免疫学的応答および抗腫瘍活性の臨床的な情報を得ることである。
当該TAAは、MPHOSPH1および/またはDEPDC1に関連する疾患(例えば癌)に対するペプチドワクチンとして、更なる発展を保証する。

- 配列番号:7、8および12からなるグループから選択されるアミノ酸配列からなるペプチド。
- 細胞傷害性T細胞誘導能を有するペプチドであって、1または2個のアミノ酸が置換された配列番号:7、8および12からなるグループから選択されるアミノ酸配列からなり、該置換が、N末端からの第2位のアミノ酸の、フェニルアラニン、チロシン、メチオニンもしくはトリプトファンへの置換であるか、かつ/またはC末端アミノ酸の、フェニルアラニン、ロイシン、イソロイシン、トリプトファンもしくはメチオニンへの置換である、ペプチド。
- 配列番号:10、11、192、195、197、209、225、226、228、230、240、241、243、244、249、253、254および255からなるグループから選択されるアミノ酸配列からなるペプチド。

- 請求項1〜4のいずれか一項に記載のペプチドをコードするDNAを含むベクター。
- 配列番号:1、3および/または5の遺伝子の過剰発現と関連した疾患を治療または予防するための医薬組成物であって、請求項1〜4のいずれか一項に記載の一つ以上のペプチド、または前記ペプチドをコードするポリヌクレオチドを含む医薬組成物。
- 疾患が癌である、請求項6に記載の医薬組成物。
- 癌が、膀胱癌、乳癌、子宮頸癌、胆管細胞癌、CML、結腸直腸癌、胃癌、NSCLC、リンパ腫、骨肉腫、前立腺癌、腎癌、SCLCおよび軟組織腫瘍からなるグループから選択される、請求項7に記載の医薬組成物。
- 請求項1〜4のいずれか一項に記載のペプチドおよびHLA抗原からなる複合体を表面に提示するエキソソーム。
- HLA抗原がHLA―A24である、請求項9に記載のエキソソーム。
- HLA抗原がHLA―A2402である、請求項10に記載のエキソソーム。
- HLA抗原がHLA―A2である、請求項9に記載のエキソソーム。
- HLA抗原がHLA―A0201である、請求項12に記載のエキソソーム。
- 請求項1〜4のいずれか一項に記載の一つ以上のペプチドを抗原提示細胞とインビトロで接触させる工程を含む、細胞傷害性T細胞誘導能を有する抗原提示細胞を誘導する方法。
- 請求項1〜4のいずれか一項に記載の一つ以上のペプチドと接触させた抗原提示細胞を、CD8陽性T細胞とインビトロで共培養することにより、細胞傷害性T細胞を誘導する方法。
- 抗原提示細胞に請求項1〜4のいずれか一項に記載のペプチドをコードするポリヌクレオチドを含む遺伝子を導入する工程を含む、細胞傷害性T細胞誘導能を有する抗原提示細胞をインビトロで誘導する方法。
- 請求項1〜4のいずれか一項に記載のペプチドと接触させた抗原提示細胞を、CD8陽性T細胞と共培養することにより誘導された、または、HLA―A24またはHLA―A2との関連において請求項1〜4のいずれか一項に記載のペプチドと結合するTCRサブユニットポリペプチドをコードする核酸が導入された、単離された細胞傷害性T細胞。
- HLA抗原と請求項1〜4のいずれか一項に記載のペプチドとの間で形成された複合体を含む、抗原提示細胞。
- 請求項14に記載の方法により誘導される、抗原提示細胞。
- 配列番号:1、3および/または5の遺伝子を発現している細胞の細胞増殖を阻害するためのワクチンであって、活性成分として請求項1〜4のいずれか一項に記載の一つ以上のペプチドを含むワクチン。
- 細胞が癌細胞である、請求項20に記載のワクチン。
- 癌が、膀胱癌、乳癌、子宮頸癌、胆管細胞癌、CML、結腸直腸癌、胃癌、NSCLC、リンパ腫、骨肉腫、前立腺癌、腎癌、SCLCおよび軟組織腫瘍からなるグループから選択される、請求項21に記載のワクチン。
- HLA抗原がHLA―A24またはHLA―A2である対象への投与のために調製される、請求項20〜22のいずれか一項に記載のワクチン。
- 抗原提示細胞を誘導するための組成物であって、請求項1〜4のいずれか一項に記載の一つ以上のペプチド、または前記ペプチドをコードするポリヌクレオチドを含む、組成物。
- 細胞傷害性T細胞を誘導するための組成物であって、請求項1〜4のいずれか一項に記載の一つ以上のペプチド、または請求項18記載の抗原提示細胞を含む、組成物。
