Description
独国特許出願番号4041703、W. N−ハロアミノ酸およびN,N−ジハロアミノ酸、並びにそれらの誘導体は、アミノ酸またはその誘導体(このものから、該ハロゲン化アミノ酸を製造する)を、アミノ酸のアミノ基上の1または2個の水素原子の、1または2個のハロゲン原子(すなわち、フルオロ、クロロ、ブロモ、ヨード原子である)による置換を生じる反応条件下で、ハロゲン供給源と反応させることによって製造する。これらの製造方法は、当該分野の化学者にとって知られる。

本発明の酸性溶液を用いて、患部をすすぐことによって、口内炎(口腔の潰瘍)または口唇ヘルペス(cold sores)を処置し得る。例えば、該溶液を、口唇ヘルペスに1日3〜4回、毎回2〜3回の適用で浸漬し、そして該溶液を該口唇ヘルペスと20〜30秒間接触させることによって、使用し得る。該溶液はまた、歯科口腔衛生における口腔のすすぎ液として、および感染症を制御するために使用し得る。この場合に、該溶液は、咽頭感染症と闘うための含嗽用液として使用し得る。該溶液は、より特定の領域については綿棒の補助で適用し得る。該溶液は、患者の要求および症状に従って、1日に1回〜数回使用し得る。

本発明の組成物は、婦人科の感染症(例えば、泌尿器感染症など)の処置のために使用し得る。例えば、様々な微生物、酵母菌(例えば、モニリア症、カンジダアルビカンスなど)、細菌感染症、HSV−2、HIV、または他の病原体は、本発明の溶液を用いて有効に処置し得る。場合により、本発明の溶液の適用は、婦人科の感染症の処置のための他の薬物療法と一緒に使用し得る。例えば、性病と予想される妊娠中の女性患者における産道の洗浄液としての使用、および潜在的には、病院の分娩室内での誕生後の赤ん坊の表面(right)における入浴および洗浄の溶液としての使用、または透析室内でのカテーテルおよびシャントの消毒剤としての使用。
本発明の化合物は、それらを局所的な感染症において使用するためのクリーム剤、軟膏剤、またはローション剤中に含有させることによって、該疾患を処置するために使用し得る。該クリーム剤、軟膏剤、またはローション剤は、広範囲な皮膚疾患に使用し得て、そして本発明の化合物の抗微生物性活性を皮膚の外側(表皮)の真下に存在する微生物にまで運搬するために、浸透エンハンサーを含有し得る。
本発明の等張性溶液は、外科手術部位の感染症の発症(これは、しばしば長期の入院、時には死亡を生じる)を予防するために、外科手術の間に灌注液として使用し得る。生理食塩水の代わりの本発明の溶液の使用は、特に、感染症の割合が10%程と高いこともあり得る胃の外科手術および長期間の手術の場合に、該感染症の危険を実質的に低下し得る。
本発明の溶液は、医学的なデバイスおよび外科手術の道具の表面上の病原体を減少して、該道具およびデバイスを使用するかまたはそれらをインプラントされる患者の感染症を予防するのに使用し得る。該溶液はまた、カテーテルおよびシャントのエントリーポート上で起こる感染症(これは、該感染症に特に起こり易い)の減少または排除のためにも使用し得る。
本発明の溶液は、室内の表面、ビヒクルの内部、または他のそれら大きく制限された空間に、直接的に、または霧を作るデバイス(エアゾール適用)からの運搬によって適用して、存在すると予想され得る感染性病原体を減少しまたは排除し得る。それらの適用の場合には、そのものは、感染性病原体が検出された手術室、部屋、ビヒクル、および生物戦薬剤が撒布された他の表面を除染するのに使用し得る。
本発明の溶液は、食物(例えば、肉、果物、および野菜を含むが、これらに限定されない)上の病原体を減少するのに使用し得る。該溶液は食物への洗浄液または霧として適用し得て、あるいは該食物を該溶液中に液浸し得る。タウリンは該適用方法の主な残留物であり、そしてタウリンはヒトの食物において安全であると考えられている必須栄養素である。
本発明の化合物は、適当な量の該化合物を製造時の該溶液中に含有させることによって、微生物が、注射および注入での使用、並びに眼での使用を意図する溶液中で生存することができないことを確認するための手法として使用し得る。
本発明の溶液は、外科医および看護士の手を安全に且つすばやく消毒して、手術中に感染性病原体を運ぶという危険を低下する手法として使用し得る。加えて、本発明の溶液は、外科的な切開の領域における患者の皮膚(手術の前および後)から感染性病原体を排除するのに使用し得る。
長期間の非治癒性創傷を患っている患者は、本発明の酸性組成物を用いて、1日基準で、典型的には1日に約1または2回処置する。
本発明の別の態様において、本発明の溶液は、個々の1回使用容器中に該溶液を含むようにパッケージングし得る。該1回使用容器は、例えば包帯の1回の交換における適用またはその等価物のために使用し得る。本発明の該1回使用容器は、通常使用する絆創膏と組み合わせて使用し得る。本発明の別の態様において、創傷治癒キットは、本発明の溶液の1回使用容器を、様々な使用方法のために特殊化された絆創膏と一緒に含み得る。

(製造方法)
試薬:全ての溶液は、脱イオン水またはミリポア(Millipore)水を用いて製造した。NaOCl(6%)溶液は、VWR社から購入した。タウリンは、シグマ社から購入した。NaClおよびHClは、試薬−グレードとした。
この研究において、NNDCTは、タウリン粉末をHOCl溶液(pH 3.5)中に、HOCl/タウリンの比率を2で溶解することによって製造した。
(抗微生物性活性)
(殺菌性活性)
殺菌性活性を測定するために、我々は大腸菌(ATCC 11229)を使用した。細菌培養物を滅菌生理食塩水中で希釈して、接種菌液を調製した。様々な被験物品を、既に1.0×105〜2.0×105コロニー形成単位(CFU)/細菌のmLを含有する個々のチューブに移し、そして静かに渦巻くことによって混合し、次いで37℃で1または24時間インキュベートした。被験物品を防腐剤として使用する場合にはインビボで産生することができるという条件をできる限り模倣しようとする試みに際しては、ペトリ皿での細菌プレーティングを、中和剤を加えることなく、設計した曝露時の直後に行ない、そして別個に中和剤(コントロールとして)を加えた。従って、曝露時の1〜24時間後に、0.1mLを取り出し、そしてプレートした。プレートを37℃でインキュベートし、そして細菌の数を直接的なコロニー計数によって計数して、生存している細菌をCFU/mL単位で数えた。正の増殖コントロールは、滅菌0.9% 生理食塩水を用いて調製した。全ての被験物品を、3回試験した。該結果を表にして、様々なpHレベルでのHOCl、OCl−、NNDCT、および0.9% 生理食塩水の抗微生物性の有効性の範囲の比較を示した。pH 3.5で、NNDCTは、60分間の場合では有効な抗微生物性の濃度の範囲は0.0149〜1.49mMの間を、24時間の場合では有効な抗微生物性の濃度の範囲は0.000149〜1.49mMの間を示し、一方で、HOClの場合の有効な抗微生物性の濃度は60分間の場合では0.016mMで、24時間の場合では0.0016mMで開始した。pH 3.5の場合には、NNDCTは、大腸菌に対してHOClより良いか、あるいは同程度に有効であった。

細胞毒性は、比色定量アッセイシステム(これは、Scudieroらによって最初に記載され、3'−(フェニルアミノ−カルボニル)−3,4−テトラゾリウム−ビス(4−メトキシ−6−ニトロ)ベンゼンスルホン酸水和物(XTT)を使用する)、プロチェック(ProCheck)(登録商標)細胞生存率アッセイ(これは、ヒトおよび他の腫瘍セルラインを用いる、培養物中の細胞増殖および薬物感受性についての可溶性テトラゾリウム/ホルマザンのアッセイの評価であり、Scudiero DA, Shoemaker RAH, Paul KD, Monks A, Tierney S, Nofziger TH, Currens MJ, Seniff D, Boyd MRによる, Cancer Res. 1988 Sep 1; 48(17): 4827-33によって記載)によって評価した。細胞生存率を測定するための同様な方法が、他の研究者によって使用されている。3個の細胞タイプを使用した:ダルベッコ変法イーグル培地、並びに対応する増殖因子および抗生物質を有するケラチノサイト限定(defined)培地中で培養した、マウス肺上皮細胞(L929)、原発性ヒト皮膚線維芽細胞、および原発性ヒトケラチノサイト細胞。細胞をトリプシン処理し、顕微鏡下で計数し、そして平底96ウェルプレートのウェル当たり1000〜2000個の細胞を播種した。細胞を37℃で終夜増殖した。翌日に、組織培地を除去し、そして細胞を新しい培地で1回ですすぎ、次いで組織培地(50μL)中に放置した。被験物品を2倍の希釈物として調製し、そして200μLを4−ウェルの各セット中に加えた(ウェル当たりの総量=250μL)。細胞を被験物品に室温で60分間、曝露した。該曝露時の直後に、各ウェルの被験物品を除去し、そして細胞を新しい培地(250μL)を用いて餌を与えた。プレートは、37℃で18〜20時間、インキュベートした。翌日、培地を再び取り出し、そして10/100μLのXTT−試薬を含有する新しい培地(100μL/ウェル)を用いて置き代えた。細胞を、着色の発生が得られるまで、増殖条件(5%CO2、37℃での加湿したインキュベーター)下でインキュベートし、光から防止した。吸光度を、モレキュラー・デバイス・サーモマックス・プレートリーダー(Molecular Device ThermoMax Plate reader)を用いて、750nmを対照波長として450nmで読み取った(培地のみのアッセイブランクウェル上のプレートをブランクとする)。XTT試薬を与えた未処置の細胞だけが、正の細胞増殖コントロールとして機能した。
NNDCTは、米国薬局方標準細胞アッセイ(United States Pharmacopoeia's standard cell assay)(マウス肺上皮細胞、L929)、並びに原発性ヒト皮膚細胞を使用するインビトロでの安全性試験を厳密に行なった。我々は、NNDCTが、他の防腐性被験物品(HOCl、およびポビドン−ヨウ素)と比較して、両方の細胞タイプ:原発性ヒト線維芽細胞およびL929細胞において非常に低い細胞毒性を有することを発見した(以下を参照)。細胞毒性が大きな問題であるポビドンヨウ素と違って、NNDCTは非常に安全な毒性プロファイルを有して、細胞と適合し得ることを示した。実際には、該治療係数(TI)(これは、NNDCTの場合の最小殺菌濃度(MBC)に対する、アッセイ細胞によって許容される濃度の比率(インビトロ細胞毒性、またはIC50)として定義する)は、HOClおよびポビドン−ヨウ素の場合にそれぞれ約300および7であるのと比較して、約5,000であった(表2)。

(2−アミノ−2−メチルプロパンスルホン酸の製造)
2−アミノ−2−メチル−1−プロパノール(アルドリッチ社製、5.0g、1.0当量)をDCM(100mL)中に溶かし、BOC無水物(14.6g、67mmol、1.2当量)を0℃で数回にわけて加え、そして該混合物をrtで終夜撹拌した。該溶液を、氷浴を除かずに、終夜ゆっくりと昇温させた。溶媒を回転蒸発によって除去した。EtOAc(50mL)を加え、続いて水(50mL)を加えた。該有機層を分離し、そしてこのものを水およびブラインを用いて1回以上洗浄し、次いで乾燥し(Na2SO4を使用)、そして濃縮して粗生成物(12g)を得た。
例えば、N,N−ジクロロ−1,1−ジメチルエタンスルホン酸の製造方法を、以下に記載する。
工程1:1,1−ジメチルエタンスルホン酸の製造(Braghiroli, D.; Bella, M. D.による, Tetrahedron Letters, 1996, 37, 7319-7322)
0.9% NaCl溶液(pH 3.5)中の1.6mM N,N−ジクロロ−1,1−ジメチルエタンスルホン酸(NNDC−DMESA)(1L)を製造するために、1000mLのメスフラスコ中にNaCl(8.6g)を加え、次いで該フラスコにミリポア水(500mL)を加えて、該塩を溶解した。該NaCl溶液中に1M HCl(2mL)を加え、続いて0.158M NaOCl(22mL)を加えた。該溶液を混合した。次いで、該フラスコ中に1,1−ジメチルエタンスルホン酸(0.355g)を加え、そして該メスフラスコをミリポア水を用いてマークまで満たした。該反応が完結するまで(これは、例えばUVまたはNMRによって示される)、該溶液を撹拌した。我々は、N,N−クロル化オルニチン、N,N−ジクロロホモタウリン、およびN,N−ジクロロアラニンを製造した。全てのこれらのジクロロ化合物は、非常に似たUVスペクトル(λ最大値=〜300nm)およびモル吸光率を有した。

(0.05M ジクロロホモタウリン溶液(1L)の製造)
工程1.0.1M HOCl(pH<5)(1L)を製造した。
工程2.工程1のHOCl溶液中にホモタウリンナトリウム(3−アミノ−1−プロパンスルホン酸ナトリウム、MW=161.13)(8.06g)を加えた。該溶液を約15分間撹拌した。
工程3.工程2の溶液の一部を取り出し、そして100倍希釈した。該生成物を同定するために、該希釈溶液のUVスペクトル(これは、303nmでλ最大値を有する)を測定した(添付する表を参照)。
工程4.工程2において得られる該溶液のpHを、NaOHまたはHClを用いて所望するpHにまで調節した。
工程5.ジクロロホモタウリンの濃度を測定するために、工程3の方法を繰り返した(モル吸光率は329.0M−1cm−1であり、添付する表を参照)。

我々の発見の結果は、0.9% 生理食塩水(pH 3.5)中でのNNDCTの抗微生物性活性を支持する。これらの抗微生物性活性は、μM単位のレンジで考慮され得るべきであると測定され、そして濃度および曝露時間を増大することによって有意に増大した。それに対して、細胞毒性は、mM単位のレンジで1000倍以上の大きいレンジで観察された。我々は、NNDCT処置細胞が該処置に対して許容であり得て、そして我々のXTTアッセイにおいて未処置コントロール細胞と比較して、正常な細胞増殖周期を受けることができることを示した。
pH 3.0、3.5、4.0、および5.0で濃度が1.49mMを有するNNDCT溶液を製造した。該溶液のスペクトルおよび濃度は、UV−可視分光光度計を用いて測定した。該結果は、NNDCT溶液のスペクトルおよび濃度がpHの範囲が3.0〜5.0で変わらないことを示した。
1000mL メスフラスコ中にNaCl(8.8g)、1.0M HCl(2mL)、およびタウリン(0.278g)を加え、続いて該フラスコ中に脱イオン水(約800mL)を加えた。該フラスコを振り混ぜて、NaClおよびタウリンの粉末を溶解した。次いで、0.15Mの該NaOCl溶液(22mL)を該フラスコ中に加えた。該フラスコを脱イオン水を用いてマークまで満たした。該溶液を磁気撹拌バーと一緒に5分間撹拌した。得られた溶液の濃度およびpHを、UV可視分光光度計および新たに較正したベックマン(Beckman)pHメータを用いて測定した。この溶液は、濃度が1.49mMおよびpH値が3.85であった。上記のNNDCT溶液(pH=3.85)(100mL)を250mL ビーカー中にピペットし、1.0M HCl溶液(0.09mL)をこの溶液に加え、そして撹拌した。この溶液の最終的なpHは、3.0であった。pH 3.85の溶液であるNNDCT(100mL)を250mL ビーカー中にピペットし、5.0M NaOH溶液(0.003mL)を該溶液中に加え、そして撹拌した。この溶液の最終的なpHは、4.85であった。
アミノ酸または該アミノ酸の塩(粉末)を、OX−:アミノ酸のモル比率が1:1であるリン酸緩衝液(例えば、pH 7.4)中の塩基性OX−(X=Cl、Br)溶液(pH>8)またはOX−(X=Cl、Br)溶液に加えた。該混合物の溶液を約15分間撹拌した。該生成物を同定し、そして該反応の完結をUV−ビス分光計によって追跡した。該溶液のpHを塩酸または水酸化ナトリウム溶液を用いて、目的のpH値にまで調節した。該溶液の濃度を、λ最大値での対応するモル吸光度を用いることによって、UV分光計を用いて測定した。より詳細な方法は以下の実施例中に記載する。
(0.05Mモノクロロホモタウリン溶液(1L)の製造)
工程1:pH>8を有する0.05M NaOCl溶液(1L)を調製した。
工程2:工程1における該NaOCl溶液中に、ホモタウリンナトリウム(3−アミノ−1−プロパンスルホン酸ナトリウム、MW=161.13)(8.06g)を加えた。該溶液を約15分間撹拌した。
工程3:工程2の溶液のアリコートを採取し、そして100倍に希釈した。該生成物を同定するために、該希釈溶液のUVスペクトルを測定した(これは、252nmでのλ最大値を有する)(以下の表を参照)。
工程4:工程2で得られた溶液のpHを、NaOHまたはHClを用いて目的のpHにまで調節した。
工程5:工程3の操作を繰り返して、該モノクロロホモタウリンの濃度を測定した(モル吸光度は368M−1cm−1である、以下の表を参照)。

等量のヒトアデノウイルスタイプ5(Ad5、McEwen菌株)を、以下の3個の試料の各々と混合した。該混合物を37℃で1時間インキュベートし、次いでこのものを組織培地(失活2%ウシ胎児血清を含有する、ダルベッコ改変イーグル培地[DMEM])中で希釈した。次いで、該混合物を上記と同じ希釈液を用いて10倍段階希釈で希釈した。各希釈混合物(0.1mL)を、12ウェルプレート(セルソース社製、ATTC)中で増殖させたA549細胞単層上に播種させ、そして1時間吸着させた。該接種物を除き、そして該単層を希釈剤を用いてすすぎ、そしてアガロース/DMEMを表面を覆うように適用した。該プレートを、5%CO2雰囲気下、37℃で6日間インキュベートした。次いで、該単層を固定化させ、染色し、そして該プラークをカウントした。

Claims (19)
- (a)式(I):
A−C(R1R0)R(CH2)n−C(YZ)−X’
[式中、
Aは、HalNH−、またはHal2N−であり;
Halは、フルオロ、クロロ、ブロモ、およびヨードからなる群から選ばれるハロゲンであり;
Rは、炭素−炭素の単結合、または炭素原子数が3〜6個の二価のシクロアルキレン基であり;
R1は、低級アルキルであって、
R0は低級アルキルであるか;あるいは、
R1およびR0はそれらが結合する炭素原子と一緒になって、(C3〜C6)シクロアルキル環を形成し;
nは、0または1〜13の整数であり;
Yは、水素、低級アルキル、−NH2、−NHHal、または−NHal2であり;
Zは、水素または低級アルキルであり;そして、
X’は、水素、−COOH、−SO3H、−P(=O)(OH)2、または−B(OH)2であり;
但し、
Rが二価のシクロアルキレン基であり、nが0または11以上の整数である場合には、該二価の基Rまたは二価の基−(CH2)n−は、適宜−NHHalまたは−NHal2で置換される]
で示されるN−ハロアミノ酸もしくはN,N−ジハロアミノ酸またはその誘導体
(ここで、該誘導体は、医薬的に許容し得る塩、低級アルカノールとのエステル、または置換基X’が結合する炭素原子と結合する−NH2基の低級アルカノイル誘導体である);および、
(b)次亜ハロゲン酸またはその塩からなる群から選ばれるハロゲン化化合物、
を含有する、殺菌剤、抗菌剤、抗感染剤、抗微生物剤、殺胞子剤、消毒剤、抗真菌剤、または抗ウイルス剤としての医薬組成物。 - R0が低級アルキルである、請求項1記載の医薬組成物。
- Rが炭素−炭素の単結合であり、nが0または1〜7の整数である、請求項1記載の医薬組成物。
- −NHHal基または−NHal2基がX’基に対してアルファ−、ベータ−、またはガンマ−位に存在する、請求項1記載の医薬組成物。
- −NHHal基または−NHal2基が二価の基Rまたは−(CH2)n−と結合する、請求項4記載の医薬組成物。
- Halがクロロであって、そして次亜ハロゲン酸が次亜塩素酸である、請求項1記載の医薬組成物。
- 誘導体が医薬的に許容し得る塩であって、そして該次亜ハロゲン酸が塩の形態で存在する、請求項1記載の医薬組成物。
- 組成物が、N−ハロアミノ酸もしくはN,N−ジハロアミノ酸またはその誘導体を0.01mM〜1Mまたは0.1〜100mMの間の濃度で有し、そしてpHの範囲は2〜7、3〜4.8、3.0〜4.5、3.5〜4.5の間、または3.5である、請求項1記載の医薬組成物。
- (a)式(IIA):
HalNH−C(R1R0)−(CH2)n−C(YZ)−X (IIA)
[式中、
Halが、フルオロ、クロロ、ブロモ、およびヨードからなる群から選ばれるハロゲンであり;
R1が、低級アルキルであって、
R0が低級アルキルであるか;あるいは、

(ここで、該誘導体が、医薬的に許容し得る塩、低級アルカノールとのエステル、または置換基Xが結合する炭素原子と結合する−NH2基の低級アルカノイル誘導体である)
を含有する、請求項1記載の医薬組成物。 - 請求項9記載の安定化医薬組成物。
- 組成物が、殺菌剤、抗菌剤、抗感染剤、抗微生物剤、殺胞子剤、消毒剤、抗真菌剤、または抗ウイルス剤としてのその使用にとって十分な長期間の安定性および有効期間を保証する容器中で保存する、請求項9記載の医薬組成物。
- 哺乳動物における細菌、微生物、胞子、真菌、またはウイルスの活性を原因とする感染症を予防または治療するための医薬の製造における、請求項1または請求項9のいずれかに記載の医薬組成物の使用。
- 医薬組成物が2〜7の間のpHの範囲を有する、請求項12記載の使用。
- 医薬組成物が、殺菌剤、抗菌剤、抗感染剤、抗微生物剤、殺胞子剤、消毒剤、抗真菌剤、または抗ウイルス剤としてのその使用にとって十分な長期間の安定性および有効期間を保証する容器中で保存する、請求項12記載の使用。
- 等張性でありそして生理学的にバランスがとれている、請求項9記載の医薬組成物。
- N−ハロアミノ酸が、N−クロロ−2,2−ジメチルタウリン;N−クロロ−1,1,2,2−テトラメチルタウリン;N−ブロモ−2,2−ジメチルタウリン;N−ブロモ−1,1,2,2−テトラメチルタウリン;N−クロロ−2,2,3,3−テトラメチル−β-アラニン;および、N−クロロ−3,3−ジメチルホモタウリン;並びに、それらの医薬的に許容し得る塩およびエステル、からなる群から選ばれる、請求項1または請求項9のいずれかに記載の医薬組成物。
- 細菌、微生物、胞子、真菌もしくはウイルスの増殖、または感染症および感染源の繁殖を制御しまたは防止するための、表面、医学的なデバイス、食物または物質の消毒における、請求項1または請求項9のいずれかに記載の医薬組成物の使用であって、但し該表面または物質はヒトまたは動物の表面ではない、該使用。
- 消毒する物質が、食物、動物の餌、外科的な装置、外科的な設備、医学的なデバイス、およびそれらの目的に使用する設備からなる群から選ばれる、請求項17記載の使用。
- 創傷治癒の促進、開口創傷における病原体の減少、創傷の汚染除去、火傷における病原体の減少、皮膚疾患の処置、組織の修復および再生、眼の消毒もしくは汚染除去、口腔消毒、抗真菌療法、肺感染症における病原体の減少、または移植用の臓器中の感染量の減少のための医薬の製造における、請求項1または請求項9のいずれかに記載の医薬組成物の使用。
