導入後、終濃度10%ウシ胎仔血清入りD−MEM/F−12培地中、37℃で培養し、培養24時間後に細胞を回収し、5x105個の遺伝子導入MSCsをヌードマウス左心室腔内に注入して全身投与を行った。当該ヌードマウスは、MSCs投与直前にそれらの背中にマウス9L細胞を皮下注射した担癌動物モデルである。投与24日後に、皮下腫瘍を採取し、動物組織DNA抽出用キットDNeasy Tissue Kit(QIAGEN社)を用いてDNA抽出を行った。キット標準の手順に従い,シリカゲルメンブレンへの吸着・溶出によりDNAを濃縮抽出した。抽出DNAにチミジンキナーゼ遺伝子に該当するプライマーセット(5‘−TTCTGGCTCCTCATGTCGG−3’(配列番号1)および5‘−ATTGGCAAGCAGCCCGTAA−3’(配列番号2))および対照としてラットGAPDH遺伝子に該当するプライマーセット(5‘−CAGCAATGCATCCTGCAC−3’(配列番号3)および5‘−GAGTTGCTGTTGAAGTCACAGG−3’(配列番号4))を適用して、チミジンキナーゼ遺伝子の量を定量的PCRにより測定した。上記3つすべてのプラスミドを導入したMSCsを投与した動物は、チミジンキナーゼ発現レトロウイルスベクタープラスミドのみを導入したMSCsを投与した動物や、チミジンキナーゼ発現レトロウイルスベクタープラスミドおよびVSV−G発現プラスミドを導入したMSCsを投与した動物と比較して、約122倍の遺伝子増幅効果を示した(図3)。なお、MSCsを投与していない担癌動物モデル、および遺伝子導入を行っていないMSCsを投与した担癌動物モデルにおいては、チミジンキナーゼ遺伝子は検出されなかった。
実施例6:担癌動物モデルにおける治療遺伝子導入およびGCVによる治療効果
方法
