(1)組換え体プラスミドpCox4DNAの調製
上述のオキシダーゼ遺伝子(配列番号1に記載のアミノ酸配列をコードする遺伝子)の組換え体プラスミドを有する大腸菌(E.coli)DH5α(pCox4)(FERM P−18002)を、LB培地(1%バクト−トリプトン、0.5%酵母エキス、0.25%NaCl)20mlに接種して、37℃で20時間振とう培養し培養物を得た。この培養物を7000rpmで5分間遠心分離することにより集菌して菌体を得た。この菌体よりQIAGEN tip−100(キアゲン社製)を用いて組換え体プラスミドpCox4DNAを抽出して精製し、組換え体プラスミドpCox4DNAを100μg得た。
(2)アミノ末端側塩基配列を改変した組換え体プラスミドpNCOPDNAの調製
配列番号6の部分配列である配列番号7〜14に記載の塩基配列をもつオリゴヌクレオチドをシグマジェノシス社の受託合成サービスにより入手した。配列番号7〜14のそれぞれをT4 Polynucleotide kinase(宝酒造社製)を用いてリン酸化する。一方、配列番号1を持つpCox4DNAを鋳型とし、配列番号15、16をプライマーとしてEx Taq polymerase(宝酒造社製)を用いてPCR反応を行い、C末端側をコードする遺伝子断片を調製した。さらに、本断片をNdeI(第一化学薬品社製)、NspI(第一化学薬品社製)で処理した。さらにpKF19kDNAをNdeIで処理した後、BAP処理(宝酒造社製試薬を使用)により脱リン酸化を行なった。こうして得られたリン酸化オリゴヌクレオチドのうち、8本のリン酸化オリゴヌクレオチド、pCox4DNA由来遺伝子断片、pKF19k断片を適量混合し、Ligation Kit ver.2(宝酒造社製)を利用しライゲーションし、得られたプラスミドをpNCOPと命名した。続いて、(1)に記載の方法によりpNCOP DNAを100μg調製した。

上記組換え体プラスミド、pNCOPDNAを鋳型としてプライマーとして配列番号17、25を用いてPCR法を実施した。この際、反応系に10%のDMSOを添加することで増幅のミスを誘発させ、変異を導入した。このようにして得たDNA断片を制限酵素NdeIで処理した後、プラスミドpKF19kを同じくNdeIで処理した後、BAP処理(宝酒造社製試薬を使用)により脱リン酸化を行なったものと混合し、Ligation Kit ver.2(宝酒造社製)を利用しライゲーションし、D.M.Morrisonの方法(Method in Enzymology,68,326〜331,1979)に従って大腸菌JM109(東洋紡社製)を形質転換し、約2500株の改変を受けたプラスミドを保有する形質転換株を得た。
(2)本発明オキシダーゼ生産株の選抜
まず、得られた上記形質転換体の全てを1mM IPTG、50μg/mlカナマイシンを含んだ滅菌済みLB培地を100μlずつ各セルに含んだ96穴プレートに植ぐ。各プレートのうち1セルは、pNCOPを保持したJM109を植え、ネガティブコントロールとする。25℃で24時間培養した。こうして得られた培養液を−80℃のフリーザーにプレートごと1時間入れて溶菌させる。その後、100μlの10%エマルゲン913を含んだ0.2%牛血清アルブミン入り100mM MES−NaOH緩衝液(pH7.0)とプレート上で混合し、37℃、5時間静置する。その後、各処理液50μlをとって下記の組成の反応試薬50μlと混ぜ合わせ反応させ、ネガティブコントロールと比較して発色の増加しているものを選択する。

ここで選択された発色した10株について2mlのLB培地(50μgカナマイシン添加)で液体培養し、プラスミドに含まれる改変されたコレステロールオキシダーゼを生産させた。培養後、得られた培養物を超音波破砕し、8000rpmで5分間遠心して得たその粗酵素抽出液0.5mlを0.5mlの10%エマルゲン913を含有する0.2%牛血清アルブミン入り100mM MES−NaOH緩衝液(pH7.0)と混ぜ、37℃で一晩反応させた。この反応液と未処理の粗酵素液の2倍希釈液の活性を測定し、活性残存率(反応液の活性/未処理の粗酵素抽出液の活性)を算出した。同様にして培養、抽出、熱処理をし、活性測定をした改変前の性質を異にするオキシダーゼと活性残存率の比較を行い、活性残存率が40%から80%に向上した改変されたコレステロールオキシダーゼとその生産大腸菌3株を得た。そのうちの1種(R1)遺伝子をコードするプラスミドpNCP1を独立行政法人産業技術総合研究所 特許生物寄託センターにFERM BP−10343として寄託した。
(3)改変の起こった部位の同定
改変されたコレステロールオキシダーゼ遺伝子を保持するプラスミドを回収し、CEQ2000XL DNA解析システム(ベックマンコールター社製)を用いて塩基配列を決定し、それを元に改変されたアミノ酸残基の同定を行った。
これにより、本発明オキシダーゼ(R1)は、配列番号1に表されるアミノ酸配列において配列番号2に示すように、175番目のアスパラギン酸がアスパラギンに置換されていることが分かった。これは、配列番号5においては133番目に相当する。同様に本発明オキシダーゼ(R7)では、配列番号1に表されるアミノ酸配列において、173番目のプロリンがロイシンに、220番目のアスパラギン酸がグルタミン酸に置換されていることが分かった。また、これらは配列番号5においてはそれぞれ131番目、178番目に相当する部位の変異である。
(4)変異点の分離と組み合わせ
これらの変異点をそれぞれ単独で導入してみた。即ち、pNCOP DNAに配列番号17,18(配列番号5の133番目をアスパラギン酸がアスパラギンに置換するようにデザインされている)の配列をプライマーとしてKOD plus polymerase(東洋紡社製)を用いて、PCR反応を行った後、アガロースゲル電気泳動にてプラスミドの全長に相当するDNA断片の増幅を確認し、制限酵素DpnI(メチル化したDNAに作用する;第一化学薬品製)で処理した後、大腸菌JM109に形質転換し、カナマイシンを添加したLB寒天培地で選択した。生育してきたコロニーをカナマイシン入りの液体培地で培養し、コレステロールオキシダーゼ遺伝子を保持するプラスミドを回収し、塩基配列を確認し目的どおりの変異が導入されていることを確認する。配列番号5の131番目(プロリンをロイシンに置換)、178番目(アスパラギン酸をグルタミン酸に置換)についても131番目については配列番号19,20を、178番目については配列番号21,22を代わりに用いて同様の方法で、部位特異的変異コレステロールオキシダーゼ遺伝子を得た。さらに、ここで得られた部位特異的変異コレステロールオキシダーゼ遺伝子を保持するプラスミドを鋳型に2重変異体コレステロールオキシダーゼ遺伝子を得た。この際、131番目と133番目の変異点は近接しているため、配列番号23,24を用いて行った。さらに3重変異体コレステロールオキシダーゼ遺伝子を保持するプラスミドを得た。
(5)変異点と界面活性剤耐性
上記で得た変異点の異なる変異型コレステロールオキシダーゼ遺伝子をもつプラスミドを保持した大腸菌JM109をそれぞれ1.0mM IPTG添加LB−カナマイシン培地10mlで30℃、24時間攪拌培養し、得られた培養液を8000rpm、5分間遠心分離して集菌し、20mM リン酸カリウム緩衝液(pH7.0)1ml懸濁した。その後、超音波により菌体を破砕し、10,000rpm、10分の遠心により上清を回収した。これらの粗酵素抽出液200μlを200μlの2%エマルゲン913入り0.2%牛血清アルブミン含有100mM MES−NaOH緩衝液(pH7.0)と混合し、30℃、7日間静置し、界面活性剤未処方で30℃、7日間静置したものと酵素活性の残存率を比較した(表1)。

The 10 colored strains selected here were liquid-cultured in 2 ml of LB medium (added with 50 μg kanamycin) to produce modified cholesterol oxidase contained in the plasmid. (3) Identification of the site where the modification occurred The plasmid carrying the modified cholesterol oxidase gene was recovered, the nucleotide sequence was determined using the CEQ2000XL DNA analysis system (manufactured by Beckman Coulter, Inc.), and the modification was performed based on it. (4) Isolation and combination of mutation points These mutation points were introduced individually. That is, PCR was performed using KOD plus polymerase (manufactured by Toyobo Co., Ltd.) using pNCOP DNA as a primer with the sequence of SEQ ID NOs: 17 and 18 (designed so that aspartic acid was substituted with aspartic acid at position 133 of SEQ ID NO: 5). (5) Mutation point and surfactant resistance The Escherichia coli JM109 retaining the plasmids having the mutant cholesterol oxidase genes with different mutation points obtained above was stirred at 30 ° C. for 24 hours in 10 ml of LB-kanamycin medium supplemented with 1.0 mM IPTG. After culturing, the resulting culture was collected by centrifugation at 8000 rpm for 5 minutes, and suspended in 1 ml of 20 mM potassium phosphate buffer (pH 7.0). 上記のようにして得られた本発明オキシダーゼ(R1)を生産する形質転換体、大腸菌(E.coli)JM109(pNCP1)を1mM IPTG入りLB−カナマイシン培地10Lに植菌し、ジャーファーメンターを用いて、通気量1L/min、攪拌速度600rpmの条件で、30℃、24時間攪拌培養した。得られた培養液10Lを7000rpm、10分間遠心分離して集菌し、20mM リン酸カリウム緩衝液(pH7.0)1Lに懸濁した。その後、超音波により菌体を破砕し、10,000rpm、10分の遠心により上清を回収し、粗酵素液とした。この粗酵素液を60℃、30分熱処理を行い、20mM リン酸カリウム緩衝液(pH7.0)で10倍希釈後、10,000rpm、10分の遠心により上清を回収する。これを限外膜AIP−2013(旭化成製)で5mM EDTA、20mM Tris−HCl(pH8.8)に透析し、500mlに濃縮する。その後、5mM EDTA、20mM Tris−HCl(pH8.8)で平衡化した200mlのQAE−Sephadex A−50に懸濁し、ろ液と200mlの5mM EDTA、20mM Tris−HCl(pH8.8)で洗いこんだものを活性画分として回収する。回収した本発明オキシダーゼ(R1)の比活性は4.0U/A280であった。

得られた本発明オキシダーゼ(R1)の理化学的性質は、下記の通りであった。

前述した酵素活性の測定方法により、基質としてコレステロールに対する反応を確認した。
(2)示適pH
緩衝液として、100mM酢酸緩衝液(pH5.5〜6.0)、100mMリン酸カリウム緩衝液(pH6.0〜8.22)、100mMTris−HCl緩衝液(pH7.58〜9.5)、100mM NaHCO3−NaOH緩衝液(pH9.43〜11.0)を用い、夫々のpHにおいて、温度37℃にて酵素反応を行った結果、その相対活性は、図1に示す通りであった。図1より、本発明オキシダーゼの示適pHは6.5〜8.0であることが判かった。
(3)作用適温の範囲
前述の活性測定法における反応液と同一組成よりなる反応液を用い、種々の温度にて本酵素の活性測定を行った結果、図2に示す通りであった。図2より、本発明オキシダーゼの作用適温の範囲は、55〜65℃であった。
(4)安定pH範囲
緩衝液として、100mM酢酸緩衝液(pH3.5〜6.0)、100mMリン酸カリウム緩衝液(pH6.0〜8.22)、100mMTris−HCl緩衝液(pH7.58〜9.5)、100mM NaHCO3−NaOH緩衝液(pH9.43〜11.0)を用い、夫々のpHにおいて、25℃で20時間処理した後、本発明オキシダーゼの残存活性を測定した結果、図3に示す通りであった。図3より、安定pH範囲は、pH3.5〜8.5であった。
(5)熱安定性
100mMリン酸カリウム緩衝液(pH7.0)を用いて、各温度で15分間処理した場合の熱安定性の結果は、図4に示す通りであり、本発明オキシダーゼは、70℃付近まで安定であった。
(6)分子量
常法により、マルチゲル10/20(第一科学薬品社製)を用いるSDS−PAGE法により分子量を求めた。約60,000であった。
(7)界面活性剤耐性
もとのバークホルデリア セパシア(Burkholderia cepacia)菌株ST−200由来のコレステロールオキシダーゼ(CHO(WT))と界面活性剤に対する安定性を比較した。それぞれ2%の界面活性剤を含む0.2%牛血清アルブミン入り100mM MES−NaOH緩衝液(pH7.0)と2U/mlのコレステロールオキシダーゼを等量混合し、37℃で24時間保存したものである(表3)。
