染色体DNAを異なるグループB連鎖球菌株から前述のように分離した〔Jayarao(ジャヤラオ), BM等、1991. J. Clin. この例はグループB連鎖球菌BVH-A5遺伝子のクローニングを例示する。
グループB連鎖球菌BVH-A5遺伝子(配列番号:1)の、リーダペプチドをコードする領域を含まないコード領域を、PCR (DNA Thermal Cycler GeneAmp PCR system 2400 Perkin Elmer)によって、血清型IIIのグループB連鎖球菌株NCS954から、追加の制限部位NcoI(CCATGG) 及びXhoI(CTCGAG)のための塩基伸長(base extension)を含むオリゴヌクレオチドプライマを用いて増幅させた。オリゴヌクレオチドプライマ(表1)のDMAR577及びDMAR747を用いてBVH-A5遺伝子を増幅した。PCR生成物をアガロースゲルから、QIAgenからのQIAクイックゲル抽出キットを製造者の指示に従い用いて精製し、及びNcoI 及びXhoI(Amersham Pharmacia Biotech)を用いて消化した。pET-21d(+) ベクター〔Novagen(ノバゲン社), Madison(マディソン), WI(ウィスコンシン州)〕をNcoI及びXhoIで消化し、及びアガロースゲルから、QIAgenからのQIAクイックゲル抽出キットを用いて精製した。NcoI-XhoI PCR生成物をNcoI-XhoI pET-21d(+)発現ベクターにリゲートした。リゲート生成物を、E. coli株STBL2 [F- mcrA Δ(mcrBC-hsdRMS-mrr) recA1 endA1 lon gyrA96 thi-1 supE44 relA1 λ-Δ(lac-proAB)] 〔Gibco BRL(ギブコBRL社), Gaithersburg(ゲイサーズバーグ), MD(メリーランド州)〕に製造者の推奨事項に従って転換させた。BVH-A5遺伝子を含む組換えpET-21d(+)プラスミド(rpET21d(+))を、QIAgenプラスミドキットを用いて精製し、及びDNA挿入断片を配列決定した〔Taq Dye Deoxy Terminator Cycle Sequencing kit, ABI(アプライドバイオシステムズ社), Foster City, CA〕。
この例は他のグループB株からのグループB連鎖球菌BVH-A5遺伝子のPCR増幅を説明する。
BVH-A5(配列番号:1)遺伝子のPCR増幅による存在の確認のために、次の11種の血清学的に異なるグループB連鎖球菌株を用いた: C388/90 (血清型Ia/c), ATCC12401 (血清型Ib), ATCC27591 (血清型Ic), NCS246 (血清型II/R), NCS954 (血清型III/R), NCS97SR331 (血清型IV), NCS535 (血清型V), NCS9842 (血清型VI), NCS7271 (血清型VII), NCS970886 (血清型VIII), 及びATCC27956〔ウシから単離 (bovine isolate)〕。これらの株はThe American Type Culture Collection (アメリカンタイプカルチャーコレクション)〔Rockville(ロックビル),MD, 米国〕及びNational Center for Streptococcus, Provincial Laboratory of Public Health for Northern Alberta (Edmonton, Alberta, カナダ国)から得た。E. coli株XL1-Blue MRF'をこれらの実験にネガティブコントロールとして用いた。染色体DNAを、前述したように(Jayarao, BM 等の1991年J. Clin. Microbiol. 29:2774-2778)、各グループB連鎖球菌株から単離した。BVH-A5(配列番号:1)遺伝子を、11種のグループB連鎖球菌株、及びコントロールのE. coli株から表1に示す次のオリゴヌクレオチド: DMAR577及びDMAR747を用いて精製したゲノムDNAからのPCR (DNA Thermal Cycler GeneAmp PCR system 2400 Perkin Elmer)によって増幅した。PCRを、94℃で30秒、55℃で30秒及び68℃で210秒の35回のサイクル及び68℃で10分の最後の伸長期間(elongation period)で実行した。PCR生成物を1%アガロースゲルにおいて大きさで分画し、及び臭化エチジウム染色により視覚化した。これらのPCR増幅の結果を表2に表す。増幅生成物は、BVH-A5(配列番号:1)遺伝子が試験したすべての11種のグループB連鎖球菌株のゲノム中に存在することを明らかにした。コントロールE. coliのDNAを、これらのオリゴヌクレオチドプライマを用いて同じPCR増幅にかけた場合、かかる生成物は検出されなかった。
この例は、グループB連鎖球菌BVH-A5遺伝子のCMVプラスミドpCMV-GHにおけるクローニングを例示する。
リーダペプチドを有しない、グループB連鎖球菌BVH-A5(配列番号:1)のDNAコード領域を、プラスミドベクターpCMV-GH〔Tang(タン)等, Nature, 1992, 356:152〕においてサイトメガロウイルス(CMV)プロモータの転写調節下にあるヒト成長ホルモン(hGH)遺伝子の下流の相(phase downstream)に挿入した。CMVプロモータは、E. coli細胞中で非機能性のプラスミドであるが、真核細胞中へのプラスミドの投与により活性になる。また、ベクターはアンピシリン耐性遺伝子を取り込む。
この例は、グループB連鎖球菌BVH-A5ポリペプチド抗原に対する免疫反応を誘発するためのDNAの使用を例示する。
8匹のメスのBALB/cマウス〔Charles River(チャールズリバー), St-Constant(セントコンスタント), Quebec, カナダ国〕の群を、50μgの顆粒球−マクロファージコロニー刺激因子(GM-CSF)−発現プラスミドpCMV-GH-GM-CSFの存在下のBVH-A5(配列番号:1)遺伝子をコード化する50μgの組換えpCMV-GH (Laboratory of Dr. Stephen A. Johnston, Department of Biochemistry, The University of Texas, Dallas, Texas)と共に2又は3週の間隔で100μLの3回の筋肉内注射によって免疫化した。コントロールとして、マウスの群に、50μgのpCMV-GH-GM-CSFの存在下の50μgのpCMV-GHを注入した。血液試料を、眼窩の洞(orbital sinus)から各免疫化に先立って及び3回目の注射後の7日に収集し、及び血清抗体応答を、精製したBVH-A5-His・Tag組換えポリペプチドを被覆抗原として用いるELISAによって決定した。
この例は、組換えグループB連鎖球菌BVH-A5ポリペプチドの生産及び精製を例示する。
配列番号:1に相当するBVH-A5遺伝子を有する組換えpET-21d(+)プラスミドを用い、E. coli株BL21(DE3)(F-ompT hsdSB (r- Bm- B) gal dcm (DE3)) (Novagen, Madison, WI)をエレクトロポレーション(Gene Pulser II apparatus, BIO-RAD Labs, Mississauga, Ontario, カナダ国)によって形質転換した。このE. coli株において、組換えポリペプチドの発現を制御するT7プロモータはT7 RNAポリメラーゼ(lDE3プロファージ上に存在)によって特異的に認識され、その遺伝子はIPTGによって誘導可能なlacプロモータの制御下にある。形質転換体BL21(DE3)/rpET21を、37℃において250rpmで撹拌しながら、1mL当たり100 μgのカルベニシリン〔Sigma-Aldrich(シグマ−アルドリッチ) Canada Ltd., Oakville, Ontario, カナダ国〕を含有するLBブロス(ペプトン10g/L, 酵母抽出物5g/L, NaCl 10g/L)中で、A600が0.6の値に達するまで増殖させた。グループB連鎖球菌BVH-A5-His・Tag組換えポリペプチドの生産を誘導するため、細胞を追加の3時間、1mMの終濃度のIPTGの存在下にインキュベートした。500mLの培養物から誘導された細胞を遠心分離によってペレット化し、及び-70℃で凍らせた。
この例は、BVH-A5のHis-標識(tagged)GBS組換えポリペプチドのヒト血清及びGBS抗原性調製物(antigenic preparation)で免疫化した後のマウスから収集した血清との反応性を例示する。
表3に示すように、BVH-A5 His-標識組換えポリペプチドは正常ヒト血清のプール中に存在する抗体によって免疫ブロットにおいて認識された。これは、普通にGBSに接触したヒトがそのポリペプチドに特異的な抗体を発生させることを明確に示すもので重要な結果である。これらの特定のヒト抗体は、GBS感染に対する保護に関与している。加えて、免疫ブロットは、また、マウスモデルにおいて有意な保護を誘導する外面ポリペプチドの豊富なGBS抗原性調製物で免疫化されたマウスから収集した血清も、また、BVH-A5 His-標識組換えポリペプチドを認識する抗体を発生させることを明らかにした。これらの結果は、このポリペプチドが、マウスを感染に対して保護し、及びそれが相当するBVH-A5 His-標識組換えポリペプチドと反応する抗体を誘導するGBS抗原性調製物中に存在することを示す。

3外面タンパク質の豊富なGBS抗原性調製物での免疫化後に収集したマウス血清をプールし及び1/500に希釈して免疫ブロッティングを行った。これらのマウスは致命的なGBS負荷(challenge)に対して保護された。
この例は、切断された(truncated)BVH-A5遺伝子生成物のポリメラーゼ連鎖反応(PCR)によるクローニング及び切断されたBVH-A5分子の発現を説明する。
遺伝子断片を、PCR(DNA Thermal Cycler GeneAmp PCR system 2400 Perkin Elmer)によって、血清型IIIグループB連鎖球菌株NCS 954のゲノムDNAから、表1に示すオリゴヌクレオチドプライマを用いて増幅させた。切断されたBVH-A5遺伝子生成物を発現ベクター中にクローニングし、及び配列決定する方法は、例2に説明した方法と同様である。組換えポリペプチドを、超音波処理したIPTG-誘導E. coli培養物の遠心分離後に得た上清画分から、例6で説明したようにHis-結合金属キレート樹脂(QIAgen)を用いて精製した。生じた遺伝子生成物を表4に記載する。
この例は、マウスの組換え切断BVH-A5ポリペプチドを用いる免疫化によって誘導された、致命的なグループB連鎖球菌感染に対する保護を例示する。
メスのCD-1マウス(Charles River)の群を、10μgのQuilAアジュバント〔Cedarlane Laboratories Ltd(セダーレーンラボラトリーズ社), Hornby, Ontario, カナダ国〕の存在下の切断されたBVH-A5-1-His・Tagポリペプチドの20μgを用い、2週の間隔で3回の皮下での免疫化を行った。コントロールマウスには、PBS中のQuilAアジュバントのみを用いて注入した。血液試料を、眼窩の洞から、各免疫化に先立って1日、14日、及び28日及び3回目の注入後の14日(42日)に収集した。1週間後、マウスに致死的な投与量のGBS株を負荷した。グループB連鎖球菌の負荷接種の試料を、血液寒天培地プレート上で平板培養し、CFUを決定し及び負荷投与量を確かめた。死亡を7日の時期に記録した。生存のデータを表5に示す。BVH-A5-1及びBVH-A5-2組変えポリペプチドのいずれかで免疫化したマウスの74%より多くがGBS株C388/90 (Ia/c)での負荷に対して保護された。同様の保護は、NCS 251 (II)及びNCS 535 (V)での致死的な負荷に対して得られた。これに対し、BVH-A5-3でのマウスの免疫化はGBS株C388/90 (Ia/c)での負荷に対してかかる保護を与えなかった。BVH-A5-1及びBVH-A5-2で免疫化した群について決定された生存率は、フィッシャーの直接確率検定によってコントロール群と統計的に異なることが示された。
Claims (6)
- 配列番号2で表されるアミノ酸配列の少なくとも15個の隣接するアミノ酸からなる抗原性ポリペプチド断片を含み、前記抗原性ポリペプチド断片は、配列番号2のアミノ酸配列からなるポリペプチドに特異的に結合する抗体を導き出すことが可能である、分離されたポリペプチド。
- ポリペプチドは担体タンパク質と接合されている、請求項1に記載の分離されたポリペプチド。
- 請求項1または2に記載のポリペプチドおよび薬学上許容可能な担体、希釈剤、または補助剤を含む、組成物。
- 配列番号2で表されるアミノ酸配列からなるポリペプチドに特異的に結合する抗体を導き出すための薬の製造における、請求項3に記載の組成物の使用。
- 1種またはそれよりも多くの抗原性ポリペプチド断片を含み、前記1種またはそれよりも多くの抗原性ポリペプチド断片は配列番号2で表されるアミノ酸配列の少なくとも15個の隣接するアミノ酸からなり、前記1種またはそれよりも多くの抗原性ポリペプチド断片は、配列番号2のアミノ酸配列からなるポリペプチドに特異的に結合する抗体を導き出すことが可能である、キメラポリペプチド。
- 2またはそれよりも多くの抗原性ポリペプチド断片を含み、前記2またはそれよりも多くの抗原性ポリペプチド断片は各々配列番号2で表されるアミノ酸配列の少なくとも15個の隣接するアミノ酸からなり、および前記2またはそれよりも多くの抗原性ポリペプチド断片はキメラポリペプチドを形成するために連結されており、前記2またはそれよりも多くの抗原性ポリペプチド断片は、配列番号2のアミノ酸配列からなるポリペプチドに特異的に結合する抗体を導き出すことが可能である、キメラポリペプチド。
