Description
一酸化窒素
一酸化窒素(NO)はいくつかの生体系において重要な分子であり、肺において連続的に生成され、呼気においてppb(10億分の1)で測定されうる。呼気における内因性NOの発見、および炎症の診断マーカーとしてのその使用は、1990年代初期にさかのぼる(公開された国際特許出願WO 93/05709およびWO 95/02181を参照せよ)。今日、内因性NOの重要性は広く認識され、数年前から、臨床分析機器が市場で入手可能である(NIOX(登録商標)、喘息患者による定期的な臨床用途のための最初のテーラーメイドNO分析計、エアロクラインAB(AEROCRINE AB)、ソルナ、スウェーデン)。
本発明者は、明細書、実施例および特許請求の範囲(本明細書によって引用される)で開示されるように、有機亜硝酸エステル類を合成および操作する新たな方法、並びに通常合成された有機亜硝酸エステル類を操作する新たな方法を開発した。
本発明の具体的な特徴はいくつかの態様で言及され、他の態様では言及されないが、これは便宜のためだけのものであり、各特徴は、本発明に従ったいずれの他の、またはすべての他の特徴とも組み合わされてもよい。
本明細書で開示された特定の形状(configuration)、工程段階、および物質は多少変化しうるので、本発明が開示および記載される前に、本発明が、このような形状、工程段階、および物質に限らないことは理解されるべきである。本発明の範囲は、添付の特許請求の範囲およびそれと同等のものによってのみ限定されるので、本明細書で用いられる用語法が特定の態様のみを記載するために用いられ、これらに限るものと意図されないこともまた理解されるべきである。
局所循環の増大が望まれる場合、注射のための添加剤として;
血管拡張効果が治療の抗腫瘍効果を増大させることができる場合、抗腫瘍薬のための補助剤として、および/または放射線療法と併用しても用いられる。抗菌剤、抗生物質など、および有機亜硝酸エステルの組合せ運搬(combined delivery)が意図される場合、別の適用は創傷治癒である。
経鼻または眼球投与のための液滴添加用またはエアロゾル形成用溶液;
経口または腸内投与のための、溶液、乳濁液、液滴、カプセル剤または錠剤;
静脈内、動脈内、気管内、筋肉内もしくは皮下投与のための注射液もしくは乳濁液、または吸入用溶液のうちの一つである。
本発明の一つの重要な臨床的利点は、本発明に従った組成物における患者への投与の場合、低酸素組織において該有機亜硝酸エステルの効果が最も有意であることである。NO形成化合物が静脈内注入として与えられる場合、いわゆるプロキシマルスティール(proximal steal)の危険(すなわち、周辺の健康な血管で血流が増大すること)が回避される。これは血管拡張物質の共通の副作用であり、血圧の低下および関連する全身症状をもたらす。
1.液体溶媒に溶解したNOの静脈内注入
該実験は、動物実験における地方倫理委員会(local ethics committee)に認可された。オスのニュージーランド白ウサギを、実施例2で与えられるプロトコルのように、麻酔し、準備し、試験した。ウサギをL−NAME(30mg kg-1)で前処理し、担体を流さずに頚静脈の中にカテーテルを通して、標準生理食塩水に溶解したNOガスを1回注入し、脂肪乳剤(イントラリピッド(登録商標)、フレゼニウス カービ)に溶解したNOガスを1回注入した。注入速度は、両方の液体について0.5mL kg-1 分-1であった。2回の注入間に、約200分間の回復期があった。
ヘパリンはカービ(ヴィトルム(Vitrum)、ストックホルム、スウェーデン)から購入し、臭化パンクロニウム(パブロン(PAVULON)(登録商標))はオルガノン(オッス、オランダ)から購入し、ペントバルビタールナトリウムはアポテークスボラーゲット(Apoteksbolaget)(ストックホルム、スウェーデン)から購入し、およびデキストラン70(マクロデックス(登録商標))はファーマリンク(Pharmalink)(スポンガ、スウェーデン)から購入した。L−NAME(NG−ニトロ−L−アルギニンメチルエステル)および通常の化学薬品はシグマケミカル社(セントルイス、ミズーリ、米国)から購入した。
L−NAME(30mg kg-1)を10分間注入することによって、呼気の一酸化窒素が減少し(19ppbから<1ppb、図1)、全身の平均動脈圧(MAP、103cmH2Oから128cmH2O(データは図示していない))が増加し、心拍数(HR、274回 分-1から258回 分-1(データは図示していない))が低下した。呼気終末CO2および関連する血液ガスパラメータは標準であった。
標準生理食塩水に溶解したNOガスをすばやく注入(30分間、0.5ml kg-1 分-1)することによって、MAPが減少し(128cmH2Oから75cmH2O(データは図示していない))、HRが増加し(258回 分-1から295回 分-1(データは図示していない))、呼気の一酸化窒素がわずかに増加した(0ppbから2.5ppb、図1)。メトヘモグロビン(metHb)画分が劇的に増加した(0.1%から20%、図2)。注入後約200分、該動物はほとんど完全に回復しており、呼気のNO、MAP、HRおよびmetHbは、それぞれ0.8ppb、123cmH2O、313回 分-1および2.5%であった。
脂肪乳剤に溶解したNOガスを注入(30分間、0.5mL kg-1 分-1)すると、呼気のNOが増加し(0.8ppbから32.5ppb、図1)、MAPが106cmH2Oから55cmH2Oに低下したが、HRおよびmetHb画分(図2)はほとんど影響を受けなかった。
標準生理食塩水と比べて、NOを脂肪乳剤に溶解した場合、呼気のNOをモニターして、肺への血液循環による液体媒体に溶解したNOの投与が大量に増加(約15倍)していることが、該結果から明らかに示される。metHbが非常に増加しているという点で、脂肪乳剤と比べて、標準生理食塩水にNOを溶解することが著しく不利であることが本発明によって指摘される。動脈血酸素の飽和が低下している場合、例えば肺塞栓症のような肺高血圧症の条件下で、metHbの生成は深刻でありうる。この実験において、本発明者は非常に速い注入速度を用い、それゆえMAPは著しく減少したが、ずっと遅い注入速度でも、肺高血圧症または血栓塞栓症の条件下の肺で有利な効果を生み出すのに十分であり、全身動脈圧の大きな低下を引き起こすことなく、これを遂行しうると考えられる。呼気のNOが<1ppbから32.5ppbに増加したこと、および混合呼気におけるNOの正常濃度(normal level)が約20ppbであることに留意せよ。さらに、有利な効果はおそらく脈管構造にあるので、呼気ガスにおいてこれらの濃度を発生させることは必要でないかもしれない。MAPにおける低下の大きさはまた、内因性NO生成の阻害にも部分的に起因しうる。
異なる化合物のNO運搬能を評価するために、一連の動物実験を行った。該実験は、地方動物倫理委員会に認可された。オスのニュージーランド白ウサギ(n=12)を、異なる用量で、NO置換溶液の異なる用量の静脈内注入に付した。いくつかの生理学的パラメータ、例えば混合呼気ガスにおけるNO濃度(FENO)を実験中に測定した。
ペントバルビタールナトリウムの生理食塩水溶液(6mg ml-1)を用いて、耳静脈を通して該動物を麻酔した(40〜60mg kg-1)。サーモスタットにつないだ加温パッドによって、体温を38〜38.5℃に維持した。該動物を背臥位で置き、気管切開して(tracheotomise)、一定容量の人工呼吸器(モデル683、ハーバード アパレイタス(Harvard Apparatus)、南ナティック、MA、USA)を用いて人工呼吸させた。チャコールフィルター(charcoal filter)(110×11cm)を用いてNOを含まない空気を人工呼吸器に供給した。呼吸数は40分-1であり、ガス分析器(オスカーオキシ(Oscar-Oxy)、ダテックス(Datex)、ヘルシンキ、フィンランド)で決定されるように、呼気終末CO2(ETCO2)が4.5〜5.3%に保たれるように一回呼吸量を最初に調整し、それによって、ナフィオン(登録商標)サンプリングカテーテルを用いて、気管カニューレに連結した2つのサイドアーム(side-arm)のうちの一つからガスを試料採取した(150ml 分-1、分時換気量の15〜20%)。他方のサイドアームに圧力トランスデューサー(ステイサム(Statham)、ハトレイ(Hato Rey)、プエルトリコ)を連結して、吸入圧(insufflation pressure)(IP)をモニターした。人工呼吸器の出口からのガスを、1〜2cmH2Oまたは4〜5cmH2Oの呼吸終末陽圧(PEEP)を生成する2つのビーカーのいずれかに、切り替え弁を通して送った。人工呼吸を最適化し、無気肺の形成を防ぐために、実験の間、全体で10分の間隔で、より低いPEEP(9分)とより高いPEEP(1分)の間でガス流量を変えた。テルモ STC−521シリンジポンプ(テルモ、東京、日本)を用いて、グルコース(24.3g l-1)、デキストラン70(マクロデックス(登録商標) 26.5g l-1)、NaHCO3(6.2g l-1)、ペントバルビタールナトリウム(4.1g l-1)および臭化パンクロニウム(98mg l-1)を含む持続注入を、同じ耳静脈を通して5mL kg-1 時-1の速度で投与した。平均血圧(MAP)および心拍数(HR)の記録(ステイサム(Statham) 圧力トランスデューサー)、並びに動脈血サンプリングのために、左の総頚動脈にヘパリン化カテーテルを挿入した。注入の投与のために、右の頚静脈に別のカテーテルを挿入した。該動物に30〜60分のインターベンションを行わない時間(intervention-free period)を与えて、安定な循環条件および安定なFENO値を得た。
排気ベンチレーターに連結した混合室(mixing chamber)の終端で、100mL 分-1で化学発光ベースのシステム(NIOX(登録商標)、エアロクラインAB、ソルナ、スウェーデン)のサンプリングを用いて、FENOを連続的に測定した。呼気の混合の完了を、同じ容器内のCO2濃度をモニターすることによって断続的にチェックした。窒素中の認定(certified)NO標準ガス(AGA スペシャルガス(AGA Specialgas)、リディンゴ(Lidingoe)、スウェーデン)を用いてキャリブレーションを行った。
異なる出発物質または担体媒体溶液(carrier media solution)(表1)を溶解し、生理食塩水または水で希釈して、異なる濃度の溶液を得た。次いで溶液を気密容器中に置き、10分間ヘリウムを通気することによって脱酸素化した。次いで該ガラス容器に純粋な一酸化窒素ガスを3〜4分間供給した。
安定期(stabilisation period)の後、該動物に対し、頚静脈カテーテルを通して、100μl kg-1 分-1の生理食塩水担体流(saline carrier flow)(864シリンジポンプ、ユニベンター社、ゼイトゥーン、マルタ)の中に、異なる注入速度で異なる溶液を静脈内注入(CMA/100、カーネギーメディシンAB、ストックホルム、スウェーデン)した。血液サンプルを採取し、血液ガスおよび酸塩基状態(ABL300、ラジオメーターA/S、コペンハーゲン、デンマーク)を断続的に分析した。実験中、FENO、ETCO2、HR、MAPおよびIPを、グラスポリグラフ(Grass Polygraph)(グラスインスツルメント社(Grass Instruments Co)、クインシー、マサチューセッツ、米国)において連続的にモニターした。
ヘパリン(カービ ヴィトルム、ストックホルム、スウェーデン)、臭化パンクロニウム(パブロン(登録商標)、オルガノン、オッス、オランダ)、デキストラン70(マクロデックス(登録商標)、ファーマリンク、スポンガ、スウェーデン)およびペントバルビタールナトリウム(アポテークスボラーゲット)を、アポテークスボラーゲット、ストックホルム、スウェーデンから購入した。他の化学薬品は、シグマケミカル社(セントルイス、ミズーリ、米国)から購入した。
データは、平均±SEMとして与えられる。

該実験は、地方動物倫理委員会に認可された。オスのニュージーランド白ウサギ(n=2)を、実施例2のように麻酔し、準備し、呼気のNOについて試験した。テルモ STC−521シリンジポンプ(テルモ、東京、日本)を用いて、グルコース(25.9g l-1)、デキストラン70(マクロデックス(登録商標)、28.2g l-1)、NaHCO3(6.6g l-1)、ペントバルビタールナトリウム(2.1g l-1)、および臭化パンクロニウム(パブロン(登録商標)、40mg l-1)を含む持続注入を、耳静脈を通して10ml kg-1 時-1の速度で投与した。平均動脈圧および心拍数の記録(ステイサム 圧力トランスデューサー)、並びに動脈血サンプリングのため、左の総頚動脈にヘパリン化カテーテルを挿入した。薬物の投与のため、右の頚静脈に別のカテーテルを挿入した。中央開胸術(mid thoracotomy)を行い、上行大動脈の周りにフロープローブ(T201 2−チャネル超音波血流計(T201 2-channel ultrasonic blood flow meter)に連結、トランソニックシステムズ社)を置いて、心臓の出力を測定した。右室壁を通して一つのカテーテルを導入し、ユニフロー(Uniflow) 圧力トランスデューサー(バクスター)を用いて肺動脈圧を測定するために、肺動脈に入れた。左心房壁を通して左心房に別のカテーテルを挿入し、圧力トランスデューサー(ステイサム、ハトレイ、プエルトリコ)を用いて左房圧を測定した。実験の間、これらのカテーテルを通してシリンジポンプ(ペルフゾル セキュア(Perfusor Secure)、B.ブラウン メルズンゲン AG、ドイツ)を用いて生理食塩水を低流量(3mL h-1)で流して、凝固を防いだ。肺血管抵抗を、共通の式を用いて計算した。手術後、該動物に30〜60分のインターベンションを行わない時間を与えて、安定な循環条件およびFENO値に達した。
1,2−プロパンジオールを25%(v/v)水溶液に溶解し、グリセロールを25%(v/v)水溶液に溶解し、気密シリンダーに入れた。溶液を、10分間ヘリウム通気によって脱酸素化し、次いで3分間純粋な一酸化窒素ガスで処理した。気密シリンダーから、溶液を静脈内注入用シリンジに移した。
両方の動物において、トロンボキサン受容体アゴニストU46619(9,11−ジデオキシ−11α,9α−エポキシメタノプロスタグランジン F2α;ラロダンファインケミカルズAB(Larodan Fine Chemicals AB)、マルメー、スウェーデン)の静脈内注入(CMA/100、カーネギーメディシンAB、ストックホルム、スウェーデン)によって、肺高血圧症を誘導した。原液は、MH 55669 −33− 酢酸メチル中、10mg ml-1であり、注入前にそれを生理食塩水中、10μg ml-1に希釈した。頚静脈カテーテル中、U46619溶液を100ml 分-1の担体流の中に注入(30μl kg-1 分-1)して、肺動脈圧を約40cmH2Oに増加させ、この注入を実験の間ずっと継続した。肺高血圧症に対抗するNO溶液の能力を調査するため、U446619注入の間、その2つの動物に、異なる用量(10、30および100μl kg-1 分-1)でグリセロール−NO注入または1,2−プロパンジオール−NO注入を行った。頚静脈カテーテルを通して、該NO溶液を、注入ポンプ(CMA/100、カーネギーメディシンAB、ストックホルム、スウェーデン)を用いて生理食塩水担体流の中に注入した。動脈血サンプルを採取し、実験の間、血液ガスおよび酸塩基状態(ABL300、ラジオメーターA/S、コペンハーゲン、デンマーク)を断続的に分析した。実験中、呼吸パラメータおよび血行動態パラメータを、グラスポリグラフ(グラスインスツルメント社、クインシー、MA、米国)において連続的にモニターした。

グリセロール−NOおよび1,2−プロパンジオール−NO注入によって、U46619誘導肺高血圧症が用量依存的様式で軽減され(図8および9)、最高用量(100μl kg-1 分-1)によって、肺血管抵抗がほとんど正常化された。
方法
麻酔および外科的処置
該実験は、地方動物倫理委員会に認可された。オスのニュージーランド白ウサギ(n=4)を、実施例3のように麻酔し、準備し、呼気のNOについて試験した。薬物および肺塞栓症物質の投与のため、カテーテルを右の頚静脈に挿入した。中央開胸術を行い、上行大動脈の周りにフロープローブ(T201 2−チャンネル超音波血流計に連結、トランソニックシステムズ社)を置いて、心臓の出力を測定した。肺動脈圧(ステイサム 圧力トランスデューサー、ステイサム、ハトレイ、プエルトリコ)および左房圧(ステイサム、上記)の測定のため、それぞれ左の頚静脈を通して肺動脈に、および左心房にヘパリン化カテーテルを入れた。肺血管抵抗を、共通の式を用いて計算した。右下後肢の前区画(anterior compartment)からの筋肉を切除し、生理食塩水に入れた。手術後、該動物に30〜60分のインターベンションを行わない時間を与えて、安定な循環条件およびFENO値に達した。
グリセロールを25%(v/v)水溶液に溶解し、気密シリンダーに入れた。溶液を、10分間ヘリウム通気によって脱酸素化し、3分間純粋な一酸化窒素ガスで処理した。気密シリンダーから、溶液を静脈内注入用シリンジに移した。
筋組織の肺塞栓形成(MPE)のための物質を、先に記載した技術の変更によって調製した。したがって、切除された前脛骨骨格筋を目に見える結合組織から除去し、次いで均質化し、0.1g 筋肉 ml-1の濃度まで標準生理食塩水に溶解し、50 IE ヘパリン ml-1を混合物に加えた。該破砕物を0.5mm メッシュに通過させて、静脈カテーテルの三方活栓中での障害を防いだ。
全グループにおいて、150μl kg-1 分-1の担体流(864シリンジポンプ、ユニベンター社、ゼイトゥーン、マルタ)の中に、150μl kg-1 分-1の流量でMPE物質(300μl kg-1、すなわち30mg kg-1)を静脈内注入(CMA/100、カーネギーメディシンAB、ストックホルム、スウェーデン)することによって、肺塞栓症を誘導した。3つの動物はいずれの処置も受けていなかったが(対照群)、1つの動物は30分間グリセロール−NO(100μl kg-1 分-1)の静脈内注入を受け、それは肺塞栓形成の20分後に開始した。頚静脈カテーテルを通して、100mL 分-1の生理食塩水担体流(864シリンジポンプ、ユニベンター社、ゼイトゥーン、マルタ)の中に、NO溶液を注入ポンプ(CMA/100、カーネギーメディシンAB、ストックホルム、スウェーデン)を用いて注入した。動脈血サンプルを採取し、実験の間、血液ガスおよび酸塩基状態(ABL300、ラジオメーターA/S、コペンハーゲン、デンマーク)を断続的に分析した。実験中、呼吸パラメータおよび血行動態パラメータを、グラスポリグラフ(グラスインスツルメント社、クインシー、MA、米国)において連続的にモニターした。

肺塞栓症の誘導によって、全動物において、肺血管抵抗が類似のレベルに増大した。グリセロール−NOの注入によって、処置した動物における肺血管抵抗が明らかに減少し、それによって肺塞栓症における肺高血圧を低下させるNO溶液の潜在能力が示された。図10参照。

該実験は、地方動物倫理委員会に認可された。オスのニュージーランド白ウサギ(n=10)を、実施例3のように麻酔し、準備し、呼気のNOについて試験した。テルモ STC−521シリンジポンプ(テルモ、東京、日本)を用いて、グルコース(25.9g l-1)、デキストラン70(マクロデックス(登録商標)、28.2g l-1)、NaHCO3(6.6g l-1)、およびペントバルビタールナトリウム(3.1g l-1)を含む持続注入を、耳静脈を通して7.5mL kg-1 時-1の速度で投与した。平均動脈圧および心拍数の記録(ステイサム 圧力トランスデューサー)、並びに動脈血サンプリングのために、左の大腿動脈にヘパリン化カテーテルを挿入した。薬物の投与のために、右の頚静脈に別のカテーテルを挿入した。左室圧の測定(ステイサム 圧力トランスデューサー)および左心室dP/dtのオンライン計算のため、左の総頚動脈を通してヘパリン化カテーテルを左心室に入れた。実験中、第2誘導を示す表面心電図が表示された。第四肋間腔を通して左開胸術を行い、外科用縫合材料(5−0 ビクリル(Vicryl)(登録商標)、エチコン、製造者 ジョンソン&ジョンソン ユトランド(Jutl)、ブリュッセル、ベルギー)を用いて、心臓の基底から約1cmの左室壁における回旋動脈の大きな前下行枝の周りに冠状動脈の結紮糸(coronary ligature)を入れた。締めたりゆるめたりすることができる冠状動脈のスネア(coronary snare)を、結紮糸およびPE管を用いて作成した。手術後、該動物に30〜60分のインターベンションを行わない時間を与えて、安定な循環条件およびFENO値に達した。
1,2−プロパンジオールを25%(v/v)水溶液に溶解し、D−グルコースを水(60%、w/v)に溶解し、気密シリンダーに入れた。溶液を、10分間ヘリウム通気によって脱酸素化し、次いで3分間純粋な一酸化窒素ガスで処理した。気密シリンダーから、溶液を静脈内注入用シリンジに移した。
全グループにおいて、インターベンションを行わない時間の後、冠状動脈のスネアを締めて、心電図においてST上昇によって確認される虚血を誘導した。虚血の30分後、冠状動脈のスネアを再び開いて、120分の再灌流を開始し、次いで実験を終えた。該動物を3つのグループに分割した:
(1)対照、
(2)30μl kg-1 分-1 グルコース−NOの静脈内注入による処置を虚血の15分で開始し、再灌流の60分後に停止、
(3)10μl kg-1 分-1 1,2−プロパンジオール−NOの静脈内注入による処置を虚血の15分後に開始し、再灌流の120分後に停止。頚静脈カテーテルを通して、100mL 分-1の生理食塩水担体流(864シリンジポンプ、ユニベンター社、ゼイトゥーン、マルタ)の中に、NO溶液を注入ポンプ(CMA/100、カーネギーメディシンAB、ストックホルム、スウェーデン)を用いて注入した。動脈血サンプルを採取し、実験中、血液ガスおよび酸塩基状態(ABL300、ラジオメーターA/S、コペンハーゲン、デンマーク)を断続的に分析した。実験中、呼吸パラメータおよび血行動態パラメータ、並びに心電図の第2誘導を連続してモニターし、コンピュータベースの収集システム(バイオパック(Biopac)、バイオパックシステムズ社(BIOPAC Systems Inc.)、ゴレタ、CA、米国)を用いてチャネルあたり1000Hzの速度で収集した。
実験後、再灌流の間、不整脈に関してグループを分析した。再灌流の1分ごとに不整脈の心拍動について調査し、特定の分にこのような拍動が含まれていたら、この分を不整脈分(arrhythmic minute)と定義した。再灌流中のこれらの不整脈分をまとめて、各グループにおける累積不整脈分を得、次いで各グループの標本サイズに標準化した。
購入した薬物は:ヘパリン(カービ ヴィトルム、ストックホルム、スウェーデン)、デキストラン70(マクロデックス、ファーマリンク、スポンガ、スウェーデン)およびペントバルビタールナトリウム(アポテークスボラーゲット、ストックホルム、スウェーデン)であった。通常の化学薬品は、シグマケミカル社(セントルイス、MO、米国)から購入した。
全グループにおいて、再灌流は不整脈を誘導した。しかしながら、図11に示されうるように、グルコース−NO(n=3)または1,2−プロパンジオール−NO(n=3)による処置によって、対照(n=4)と比較して不整脈の発生率が明らかに減少した。
反応物の注入およびサンプリングのためのテフロン(登録商標)でコーティングしたゴム隔膜を備えたガラスシリンダー中で、全ての反応混合物および標準液の希釈液をそれぞれ調製した。
1mL/分の一定総流量を運搬する2つのLDC コンスタメトリックポンプ(Constametric pump)を用いて、C8、C18またはCNカラム(Waters社 ノバパック(登録商標) 4μm 粒径 8×100mm ラジアルコンプレッションカラム(radial compression column))における逆相クロマトグラフィーを用いた高速液体クロマトグラフィー(HPLC)によって、反応混合物、並びに市販品の有機亜硝酸エステル類および有機硝酸エステル類の分析を行い、それは33分で0〜100% メタノール(溶離溶媒B)の直線勾配を生成するコンピュータ化グラジエント制御システムで制御され、pH 6でギ酸アンモニウム(5mM)を含む水性(aqueous)の初期溶離溶媒(溶媒A)中にサンプルを注入した1分後に開始した。溶離した化合物の分離を、210nmでLDC スペクトロモニター 可変(variable) UV モニターセットによって決定し、続いてファルマシア ウェーブスキャン 波長走査検出器によって、4秒の間隔で190と370nmの間のUVスキャンを生じ、それをコンピュータに保存し、各HPLCを実行した後に分析した。LDC スペクトロモニターからの出力を、ストリップチャートレコーダーにおいて連続して記録した。
抽出したサンプルの実験:
溶離したピークを、小さな密封したサンプルバイアル中の等容積のトルエンで抽出し、簡単に振とうし、必要であれば遠心分離ですばやく分離した。トルエン相をオートサンプラーバイアルに入れ、1μl画分を、40℃の初期温度でアジレント DB−5MS(0.25mm i.d.、長さ30m、内面コーティング:95% メチルシリコンおよび5% フェニル)ガスクロマトグラフィーカラムに注入し、その後、200または250℃の温度に達するまで、カラムの温度を10℃/分ずつ上昇させた。溶離化合物の検出を、ヒューレット パッカード 5973 質量選択検出器(四重極MS)によって行った。ガスクロマトグラフは、ヒューレット パッカード エンハンスト パラメータ インジェクターシステムを有するヒューレット パッカード 6890 シリーズ GCシステムであった。データを収集し、質量スペクトルのワイリー 7n ライブラリ(Wiley 7n library)を有するエンハンスト ケムステーション G1701 BA コンピュータプログラムで解析した。
一つの実験において、約100% プロパノールおよびNOガスからなる反応混合物(1マイクロリットル)の直接注入を、気密マイクロシリンジを用いてガスクロマトグラフィーカラムに直接注入することによって行い、抽出手順およびオートサンプリングシステムを迂回した。市販品の亜硝酸プロピルをトルエンに希釈し、比較のために同様に注入した。
1,2−プロパンジオール 25%または約100%のサンプルをNOガスで処理し、30mL/分の総He流量(分割画分(split fraction) 1:20)において0.5マイクロリットルで、アジレント DB−1701カラムを現在備えているGC−MSシステムに注入した。温度グラジエントは上記のとおりであった。メタン流が供給される化学イオン化インターフェースを用いて、電子衝撃(EI)モードで、または化学イオン化モード(CI)で質量分析を遂行した。
HPLC:
すべての3つのノバパック カラムおよびサーモカラムにおいて、プロパノールとNOガスの間の反応から、単一ピークの生成物が70〜80% メタノールのグラジエント濃度で溶離した。ノバパック C18 システムにおいて、これは約28分の溶離時間に対応していた(図12、パネルC)。28分のピークは、ウェーブスキャン検出器で決定されるように、HPLC溶離液中、225nmで単一のUV最大吸収(UV absorbance max)を有していた。ガス型シリンダー(gas-type cylinder)において、1:1000で脱酸素化(10分 Heパージ)した水に希釈した亜硝酸プロピル(カール インダストリーズ社(Karl Industries Inc.)、オーロラ(Aurora)、OH、米国)の標準サンプルを注入した場合、225nmで同一のUV最大吸収を有する小さなピークが28分で得られた(図12、パネルA)。亜硝酸プロピルの希釈をNOガスで飽和させた水中で行った場合、28分のピークは5〜10倍の大きさであり(図12、パネルB)、やはり同じUVの特徴を示す。類似の結果がC8およびCNカラムにおいて得られ、C18システムと比較して多少短い保持時間であるが、単一の保持されたピークがプロパノールおよびNOからなる反応混合物から得られた。亜硝酸プロピルの市販品のサンプルもまたこれらのシステムにおいてプロパノール+NO 反応生成物の保持時間と一致するピークを生じ、より小さな大きさであるが、上記と同じUVの特徴を示した。硝酸プロピルの市販品のサンプルもまた、対照のためにCNカラムHPLCシステムにおいて調査し、亜硝酸プロピルの溶離時間と著しく異なった溶離時間を有しており、28分のピーク、またはプロパノール−NOもしくは亜硝酸プロピルクロマトグラムにおけるいずれの他のピークとも一致していなかった。
C18 HPLCクロマトグラフィーから28分のピークを試料採取し、トルエン中で抽出し、GC−MSシステムに注入した際、いくつかのピークが溶離した。初めに(1.5分)、予期される空気ガスのピーク(例えばCO2および酸素−18)がNOピークとともに観察された。3分に、亜硝酸プロピルについての質量スペクトルの特徴を示すピークが、MSによって同定された。GC−MSシステムにおいて3.5分でプロパノールが溶離し、同定され、続いて5.5分で少量のベンゼン(トルエンの分解産物)、次いで8分で大きなトルエンのピークが同定された。
市販品の亜硝酸プロピルの直接のオンカラム注入によって、その溶離時間(3分)が確認され、プロパノール+NOガス 反応混合物から抽出されたピークにおいて観察され、C18 HPLCにおいて28分で溶離および収集される亜硝酸プロピルの質量スペクトルの特徴が確認された。気密反応容器から得られ、脱気トルエン中で直ちに希釈され、GC−MSに注入されたプロパノール+NOガス 混合物の直接のオンカラム注入によって、溶離の最前部にNOガスの初期ピーク、3分に亜硝酸プロピル、および3.2分からその後数分溶離するプロパノールの非常に大きなピーク、続いてトルエン(図13)が含まれていた。結論として、HPLCの実行から精製されたピークの分析によって、またはプロパノール+NOガス 反応混合物をGC−MSシステムに直接注入した後に、亜硝酸プロピルは、HPLC(市販品の基準との比較によって)とGC−MSの両方によって、プロパノール+NOガス 混合物中で同定された。
アジレント DB−1701カラムにおける自動注入を用いて、プロパンジオールおよびプロパンジオール−NOを同様に分析した。図14、15aおよび15bに示される結果によって、本発明に従った方法の後に生じる反応混合物には、亜硝酸1,2−プロパンジイルおよび未反応のプロパンジオールの2つのアイソフォームが含まれるという仮説がサポートされる。図14bは、マスフラグメント30(有機亜硝酸エステル類についての共通のフラグメント)の主要な強度が、化合物1および2についての2つのクロマトグラフィーピークに見られることを示す。図15aおよびbにおける質量スペクトルによって示される構造から予期されるように、図16aおよびbに示される化学イオン化を用いた質量分析によって、化合物1および2についての質量スペクトルにおけるM−1およびM+1分子イオンが示される。
亜硝酸エチル、亜硝酸ブチル、および亜硝酸イソブチルの市販品のサンプル(シグマ−アルドリッチ)を上記のように嫌気的に希釈し、ノバパック C8またはCNカラムを用いてHPLCシステムに注入した。すべてのサンプルにおいて約225nmのUV最大吸収を有するピークが得られたが、それは化合物の親油性の鎖長から判断される予期される差異に対応する異なる保持時間で得られた。有機亜硝酸エステルの希釈が脱気NO飽和溶液中でなされた場合、すべてのこれらのピークはより大きかった。225nmでUV最大吸収を有するそれぞれのピークは、対応のアルコール(エタノール、ブタノール、イソブタノール)をNOガスで処理することによって得られる単一の類似のピークと一致している。これは、対応の有機一亜硝酸エステルが形成されている証拠と考えられた。
グルコース、グリセロールまたはショ糖の25〜100%(重量/容積)水溶液が脱気され、NOガスで処理される場合、いくつかのピークが逆相クロマトグラフィーによるHPLCクロマトグラムにおいて同定された。これらのピークの中には、有機亜硝酸エステルが形成されていることを示唆する225nm UV吸収の特徴を示すものもあった。
HPLCシステムからの溶離液のサンプルを収集し、1% ヨウ化ナトリウムの熱酢酸溶液を含む反応容器の中に注入することによって、NO/亜硝酸エステル 含量について分析した。オゾンと化学発光反応させて一酸化窒素を検出し、放射された光子を数えることで「NO+オゾン−生成された励起NO2生成物」を決定するように最適化された化学発光分析計システムの中に、放出された一酸化窒素をパージした。
グルコースおよびプロパノールの処理からの、HPLCにおいて保持されたピークとして同定された反応生成物は、すべてこの比較的穏和な還元処理によってNOを生じることが示された。ニトログリセリン(三硝酸グリセリン)はこれらの条件下でNOの放出を生じず、それによって、グリセロールとNOの間の反応混合物には、有機硝酸エステルよりもむしろ有機亜硝酸エステルが含まれていることが確認された。
