Description
本発明の、他の観点、特徴及び利点並びに更なる観点、特徴及び利点は、本発明の現時点での好適な実施形態についての以下の説明から明らかになる。これらの実施形態は開示の目的で提供される。
用語「及び/又は」は、本明細書中で使用される場合、「及び」、「又は」及び「当該用語により繋げられた要素の全て又は任意の他の組合せ」の意味を包含する。
用語「アミノ酸」及び「残基」は同義語であり、天然アミノ酸及びアミノ酸アナログ(例えば、非天然アミノ酸、合成アミノ酸及び改変アミノ酸、D又はL光学異性体を含む)を包含する。
本明細書中で使用される場合、用語「NSポリペプチド」は、当該分野で認識されている非構造タンパク質をいい、好ましくは、NS2、NS3、NS4A、NS4B及びNS5Bポリペプチドからなる群から選択される非構造タンパク質をいう。にもかかわらず、融合タンパク質はNS5Aポリペプチドを含有しないことが好ましい。本発明に関して、本発明の融合タンパク質に含まれるNSポリペプチドは、独立して、現時点で同定されている任意のHCV系統又は単離株(例えば、用語「HCV」に関連して上記したもの)に由来してもよい。用語「由来する」は、単離されていること、クローン化されていること、誘導されたこと又は関連することを意味する。よって、本発明により、融合タンパク質に含まれる各々のNSポリペプチドは、下記に更に定義するように、天然型NSポリペプチドに由来してもよいし又は改変NSポリペプチドに由来してもよい。
・NS3ポリペプチドは、常に、NS4Aポリペプチド又はNS4Bポリペプチド又はNS5Aポリペプチド又はNS5Bポリペプチドの前にあり;
・NS4Aポリペプチドは、常に、NS4Bポリペプチド又はNS5Aポリペプチド又はNS5Bポリペプチドの前にあり;

更により好ましくは、任意選択肢のNS4Bポリペプチドは、配列番号7で示されるアミノ酸配列から本質的になるか或いは該アミノ酸配列からなる。
・154位のArg残基〜182位のLeu残基に伸びる部分を含んでなる;
・220位のAsp残基のAspとは異なるアミノ酸残基(例えばAsn残基)による置換を含んでなる;
・318位のAsp残基のAspとは異なるアミノ酸残基(例えばAsn残基)による置換を含んでなる;及び
・571位のTrp残基〜591位のArg残基に伸びる部分を含んで成らない。
代表的には、そのようなウイルス粒子は次の工程を含んでなる方法により産生される:
(a)本発明のウイルスベクターを適切な細胞株に導入し;
(b)感染性ウイルス粒子の産生を可能にするように、細胞株を適切な条件下で培養し;
(c)産生された感染性ウイルス粒子を細胞株の培養物から回収し;
(d)任意に、回収した感染性ウイルス粒子を精製する。
・1Mショ糖、150mM NaCl、1mM MgCl2、54mg/l Tween 80、10mMトリス(pH8.5)(特に活性物質がアデノウイルスベクターのとき)、及び
・10mg/mlマンニトール、1mg/ml HSA、20mMトリス(pH7.2)及び150mM NaCl
・生理食塩水。
本発明はまた、ヒト又は動物生物に、治療有効量の本発明の融合タンパク質、核酸分子、ベクター、感染性ウイルス粒子、宿主細胞又は組成物を投与することを含んでなる、ヒト又は動物生物の治療又は予防の方法を提供する。
しかしながら、他のプライムブーストの組合せも本発明に関して可能である。

材料及び方法
以下に記載する構築物を、Sambrookら(2001, Molecular Cloning ;A Laboratory Manual, Cold Spring Harbor Laboratory Press, Cold Spring Harbor NY)に詳細に記載されているような遺伝子工学及び分子クローニングの一般的技術に従って又は市販キットを用いる場合には製造者の推奨に従って製造する。PCR増幅技術は当業者に公知である(例えば、PCR Protocols-A guide to Methods and applications, 1990;Innis, Gelfand, Sninsky and White編, Academic Press Incを参照)。ヒトHu-7肝ガン細胞株を使用して、以下の実施例に記載された構築物からの融合タンパク質の正確な発現を評価した。培養条件は当該分野で慣用されている。例示を目的として、細胞を、10%ウシ胎仔血清、2mM L-グルタミン及び100IU/mlペニシリン/ストレプトマイシン(Sigma)を補充したダルベッコ改変イーグル培地を含有する完全DMEM培地において、37℃にて5%CO2雰囲気中で増殖させる。細胞を、Lipofectamine Plus試薬(Invitrogen)を用い、製造者の指示に従ってトランスフェクトする。NS3及びNS5Bを指向するネズミモノクローナル抗体は、市販品として購入することができるか又は当該分野で慣用の技術に従って製造することができる。
オーバーラップするオリゴヌクレオチドを使用して、それぞれNS4A-NS3-NS5B(3666ヌクレオチド)及びNS4A-NS3-NS4B-NS5B(3747ヌクレオチド)融合タンパク質をコードする合成遺伝子を作製した(GeneART, Regensburg, Germany)。融合体の設計を図9に示す。ヌクレオチド配列を、ホモサピエンス(配列番号11及び12並びに図3及び4)、Pichia(配列番号13及び14並びに図5及び6)、及びE. coli(配列番号15及び16並びに図7及び8)における高度発現に最適化した。得られた融合体のアミノ酸配列を、NS4A-NS3-NS5Bについては配列番号9に、NS4A-NS3-NS4B-NS5Bについては配列番号10に示す。合成ヌクレオチド配列の各々を、pPCR-Scriptプラスミド(Stratagene)のSacI及びKpnI制限部位にクローニングした。
ウェスタンブロット分析
トランスフェクションの1日前に、Huh-7(5×105/ウェル)を6ウェルプレート中に播種した。細胞を、gWiz NS4A-3-5B Hs、gWiz NS4A-3-4B-5B Hsベクター又はネガティブコントロールとしてのgWizプラスミドでトランスフェクトした。トランスフェクション後48時間で、細胞を50mMトリス-HCl、150mM NaCl、0.1% SDS、1% NP40及び0.5%デオキシコール酸Na中で溶菌した。溶菌サンプルを95℃で5分間加熱し、タンパク質をSDS-8%ポリアクリルアミド上での電気泳動により分離した。免疫ブロッティングを、NS3特異的マウスモノクローナル抗体(例えば、8G4C3、bioMerieux)又はNS5B特異的マウスモノクローナル抗体(例えば、11F6C8、bioMerieux)を一次抗体として、ヤギ抗マウスIgG-ペルオキシダーゼ抗体(Sigma)を二次抗体として使用して実施した。市販のECLキット(Amersham Biosciences)を使用してシグナルを明らかにした。
トランスフェクションの1日前に、ガラスカバースリップを6ウェルプレート中に配置し、0.2%ゼラチンで10分間処理し、次いで各ウェル中に5×105のHuh-7細胞を播種した。細胞単層を、gWiz NS4A-3-5B Hs、gWiz NS4A-3-4B-5B Hsベクター又はネガティブコントロールとしてのgWizプラスミドでトランスフェクトした。トランスフェクション後48時間で、カバースリップをPBS中で2回洗浄し、細胞を4%PFAで10分間固定し、次いで洗浄し、PBS中0.1% Triton X-100で10分間透過化処理した。カバースリップを洗浄し、一次抗体としてのNS3特異的マウスモノクローナル抗体(例えば、8D8E1、bioMerieux)又はNS5B特異的マウスモノクローナル抗体(例えば、5B-12B7、D. Moradpourによる提供)で室温にて1時間処理した。洗浄後、TRITC抗マウスIgG(Dako)を30分間加えた。洗浄後、カバースリップを80%グリセロール中にマウントした。ラメ(lame)をCarl Zeiss Axioplan顕微鏡で観察し、AxioCamカラーデジタルカメラで撮影した。
Pascoloら(1997, J. of Experimental Medicine 185, 2043-2051)により記載されたように作製したHLA-A2.1トランスジェニックマウスを飼育した。これらマウスは、H-2Db及びマウスβ2-ミクログロブリン遺伝子がノックアウトされており、ヒトβ2mのC末端がキメラ重鎖(HLA-A2.1 α1-α2、H-2Dbα3膜貫通ドメイン及び細胞質内ドメイン)のN末端に共有結合したトランスジェニック単鎖組織適合性クラスI分子を発現する。
プラスミド注射の5日前に、HLA-A2トランスジェニックマウスに10μM心臓毒(Latoxan)での前処置をした。gWiz NS4A-3-5B、gWIZNS4A-3-4B-5B及びgWiz(ネガティブコントロール)プラスミドを、100μg(100μlの1× PBS中)の用量にて2週間間隔で2回筋肉内に注射した。2回目の免疫の2週間後に、例えばHimoudiら(2002, J. Virol. 76, 12735-46)に記載されたように、HLA-A2制限CD8+T細胞応答を調べた。
脾細胞(2×105)を、抗マウスIFNγモノクローナル抗体(Pharmingen;10μg/ml)で被覆したMultiscreenプレート(Millipore)において三つ組ウェルで、完全αMEM培養培地(Invitrogen)中、ネガティブコントロールとしての10ユニット/mlの組換えIL-2(PeproTech Inc, England)単独又はポジティブコントロールとしての5μg/mlのコンカバリン(Concavalin)A又は10μMのペプチド(DLM無関係ペプチド、Martinら, 2004, J. Med. Virol. 74, 397-405及びHimoudiら, 2002, J. Virol. 76, 12735-46により記載されたNS3タンパク質中に位置するGLLペプチド)の存在下で40時間培養した。IFNγ産生細胞を、以前に記載されたようなサイトカイン特異的酵素結合免疫スポットアッセイ(ELISPOT)(Himoudiら, 2002, J. Virol. 76, 12735-46)により定量した。ネガティブコントロールウェル中のスポット数(個々のIFNγ産生細胞を表す)を、ペプチドを含有する試験ウェル中の数から減算した。結果を三つ組ウェルについて得た平均値として示す。
融合タンパク質NS4A-3-5B及びNS4A-3-4B-5Bをコードする配列を、図9に概説したように構築した。配列を、以下に列挙するように、3つの異なる宿主(GeneART, Regensburg, Germany)ヒト(Hs)、Pichia pastoris(Pp)及びE. coli(Ec)のそれぞれにおける発現に最適化した:
Pichiaにおける発現に最適化したNS4A-3-5B Pp(配列番号13及び図5)
E. coliにおける発現に最適化したNS4A-3-5B Ec(配列番号15及び図7)
ホモサピエンスにおける発現に最適化したNS4A-3-4B-5B Hs(配列番号12及び図4)
Pichiaにおける発現に最適化したNS4A-3-4B-5B Pp(配列番号14及び図6)
E. coliにおける発現に最適化したNS4A-3-4B-5B Ec(配列番号16及び図8)。
プラスミドgWiz NS4A-3-5B Hs及びNS4A-3-4B-5B HsがHLA-A2トランスジェニックマウスにおいてCD8+T細胞応答を誘導する能力を評価するために、免疫原性研究を、「材料及び方法」に記載したように従って実施した。2回目の免疫の2週間後に、IFNγELISPOTアッセイによりHLA-A2制限CD8+T細胞応答を調べた。図12に示されるように、pgWiz NS4A-3-5Bは、HLA-A2制限性であり周知の免疫優性GLLペプチド(NS3タンパク質、Martinら, 2004, J. Med. Virol. 74, 397-405)に特異的なIFNγ産生T細胞を誘導することができた。このことは、このエピトープの正確な提示を示唆する。
NS4A-NS3-NS5B(3666ヌクレオチド)及びNS4A-NS3-NS4B-NS5B(3747ヌクレオチド)に対応する、ホモサピエンスにおける発現に最適化された融合体をコードする合成遺伝子(配列番号11〜12)を、次のプライマーOTG18033(GGGGGGCTAGCGCCACCATGGGCAGCGTGGTGATTG;配列番号19)及びOTG18036(GGGGGGGTACCCTCATCATCTAGGCCTGGCCCTG;配列番号20)を用いて、対応するpPCR-ScriptプラスミドからPCRにより増幅した。両フラグメントを、アデノウイルス配列(それぞれ、アデノウイルスヌクレオチド1〜458及びヌクレオチド3328〜5788)に囲まれたCMV駆動発現カセットを含有するアデノウイルスシャトルプラスミドのNheI及びKpnI制限部位に挿入して、相同組換えによるベクターゲノムの作製を可能にした(Chartierら, 1996, J. Virol. 70, 4805-4810)。得られたアデノウイルスベクターは、E3(ヌクレオチド28592〜30470)及びE1(ヌクレオチド459〜3511)を欠失しており、E1領域が、CMV前初期エンハンサー/プロモーターと、キメラヒトβ-グロビン/IgGイントロン(Promegaから入手可能なpCIベクター中に見出されるようなもの)と、2つのポリタンパク質形態のいずれかをコードする配列と、SV40後期ポリアデニル化シグナルとを(5'から3'に)含有する発現カセットにより置換されている。得られたアデノウイルスベクターを、pTG17476(NS4A-3-4B-5B)及びpTG17477(NS4A-3-5B)と命名した。慣用のE1補完細胞株にPacI線状化ウイルスゲノムをトランスフェクトすることにより、組換えアデノウイルスAdTG17476及びAdTG17477を作製した。ウイルスの増殖、精製及び力価決定を以前に記載されているように行った(Erbs, 2000, Cancer Res. 60, 3813-3822)。
動物モデル
HLA-A2.1トランスジェニックマウスを、Pascoloら(1997, J Ex Med)により記載されたように作製した。これらマウスは、H-2Db及びマウスβ2-ミクログロブリン遺伝子がノックアウトされ、ヒトβ2mのC末端がキメラ重鎖(HLA-A2.1 α1-α2、H-2Dbα3膜貫通ドメイン及び細胞質内ドメイン)のN末端に共有結合したトランスジェニック単鎖組織適合性クラスI分子を発現する。
3群のマウスがこのプロトコールに含まれ、それぞれ、AdTG17476(Ad NS4A-3-4B-5B)を注射した4匹のマウス、AdTG17477(Ad NS4A-3-5B)を注射した4匹のマウス、空のAd(Ad1514)を注射した2匹のマウスであった。アデノウイルスは、前脛骨筋に、100μlの1×PBS中109IUの用量で1回筋肉内注射した。細胞性免疫応答を注射の2週間後に調べた。
ELISPOTアッセイ
各群のマウスからの脾細胞を回収し、赤血球を溶解した。2.105細胞を、抗マウスIFNγモノクローナル抗体(Pharmingen;10μg/ml)で被覆したMultiscreenプレート(Millipore)において三つ組ウェルで、完全αMEM培養培地(GIBCO BRL)中、ネガティブコントロールとしての10ユニット/mlの組換えマウスIL-2(PeproTech Inc, England)単独又は10μMのペプチド(DLM無関係ペプチド、NS3タンパク質に位置するGLL及びKLTペプチド、NS5Bタンパク質に位置するKLQペプチド(Martinら, 2004, J Med Virol. 74, 397-405及びHimoudiら, 2002, J Virol., 76, 12735-12746)又はポジティブコントロールとしての5μg/mlのコンカバリンAの存在下で40時間培養した。IFNγ産生細胞を、以前に記載されたようなサイトカイン特異的酵素結合免疫スポットアッセイ(ELISPOT)(Himoudiら, J Virol 2002)により定量した。ネガティブコントロールウェル中のスポット数(個々のIFNγ産生細胞を表す)を、ペプチドを含有する試験ウェル中の数から減算した。結果を三つ組ウェルについて得た平均値として示す。
アデノウイルス免疫の2週間後に、HLA-A2制限T細胞応答をIFNγELISPOTアッセイにより調べた。図14に示されるように、AdTG17476及びAdTG17477の両方が、全てのワクチン接種動物において、HLA-A2制限GLLエピトープに特異的なIFNγ産生T細胞を高頻度で誘導することができた(図14A、中央値:それぞれ1210及び868スポット)。AdTG17476の方が統計学的に有意に高効率であった(Mann-Whitney検定、p=0.0209)。両ウイルスとも、より低い頻度(中央値:187及び90スポット)で、亜優性(subdominant)のKLTエピトープに特異的なIFNγ産生T細胞もまた誘導した(図14B)。加えて、AdTG17476をワクチン接種したマウスの2:4、AdTG17477をワクチン接種したマウスの1:4がNS5Bタンパク質のKLQエピトープを標的する応答を発現した(図14C)。

- HCV感染、HCV関連の疾患及び病的状態の治療又は予防を意図する薬物の製造のための、請求項1に記載の融合タンパク質、請求項2〜9のいずれか1項に記載の核酸分子、請求項10〜14のいずれか1項に記載のベクター、請求項15に記載の感染性ウイルス粒子、請求項16に記載の宿主細胞又は請求項18に記載の組成物の使用。
- 請求項18に記載の組成物が抗HCV免疫応答をプライムするか若しくはブーストするかのいずれか又はプライムしブーストするために使用される、プライムブースト治療様式に従って行われる、請求項19に記載の使用。
- 1又はそれより多いHCV薬での化学療法と組み合せて行われる請求項19又は20に記載の使用。
