Description
そのため、牛乳中のヨーネ菌の熱耐性機序や、有効な加熱殺菌システム開発のために実験室内で加熱実験を行う強いニーズがでてきた。
牛乳中の病原微生物に対する加熱殺菌評価実験の実施にあたり、実際に牛乳工場で使用されている加熱温度と加熱時間及び加熱冷却の温度スロープの再現を簡易装置により実施することは非常に困難であり、その改善が求められていた。
ヨーネ病は、出生後、早期に子牛が経口的にヨーネ菌に感染することで成立するが、ヨーネ病の感染から発症までのプロセスは、未だ不明の部分が多く、個体レベルでの感染経過の差異が他の疾病に類を見ないほど大きい。感染のステージにより診断可能な技術が限られており、現在でも尚、全ての感染ウシを診断するに至っていない。
ヨーネ菌がヒト難病疾患であるクローン病の原因であるとする仮説や論文が出されてきたが、2005年にランセット誌に、クローン病患者の半数の血液から本菌が分離されたという報告が出され、これに先駆けて、米国の市販牛乳の約3%がヨーネ菌生菌に汚染されていたという報告がされ、家畜衛生上、公衆衛生上もこの病気を早期診断して清浄化する社会的ニーズは非常に高まっている。
また、ヨーネ菌が加熱により活性化されるという報告もあり、家畜衛生領域のニーズとして仔牛への経口感染防止のための初乳や原乳中のヨーネ菌殺滅技術や公衆衛生(食品衛生)領域のニーズとして、市乳の加熱殺菌条件の評価をするための実験器具や方法の開発が望まれている。
そして牛乳中の病原微生物に対する加熱殺菌評価実験の過程で、これはそのための実験器具の材質に問題があることが明らかになった。
従来、実験室内で用いられる微小チューブは、プラスチック、特にポリエチレンやポリプロピレンなどのポリオレフィンでできており、これらは熱伝導性が極めて悪い素材であることが分かった。
本発明は、牛乳の加熱殺菌の評価に限らず、様々な試料の加熱殺菌評価試験並びに広く化学反応や酵素反応など、内容物(試料)に対する鋭敏な温度感作を必要とする諸実験に広範に利用することができる。
本発明の温度感作実験用金属製微量試験管は、液体試料の加熱及び冷却条件による成分の変化、例えば酵素活性、酵素反応の推移、タンパク質の変性やポリメラーゼ連鎖反応(PCR)など、様々な化学反応の実施や測定にも応用が可能である。
即ち、請求項1に係る本発明の温度感作実験用微量試験管は、材質が金属であることを除いて、その大きさなどはエッペンドルフチューブとして公知のプラスチック製の微量試験管と同様である。
熱伝導度0.708cal/s/cm2以上の金属としては、金(熱伝導度:0.708cal/s/cm2(℃/cm))、銅(熱伝導度:0.941cal/s/cm2(℃/cm))、銀(熱伝導度:0.998cal/s/cm2(℃/cm))などを挙げることができる。
プラスチックの熱伝導度は、金属に比べ一般に低く、最低のポリプロピレン(熱伝導度:1.3×10-4cal/s/cm2/(℃/cm)から、最高の高密度ポリエチレン(熱伝導度:12.4×10-4cal/s/cm2/(℃/cm)までの範囲にあり、これは石英ガラスの33.1×10-4cal/s/cm2/(℃/cm)、普通磁器の30〜41×10-4cal/s/cm2/(℃/cm)、銅の0.941cal/s/cm2/(℃/cm)に比べると著しく小さい。
銅の熱伝導度は、銀(熱伝導度:0.998)に次いで熱伝導度が大きい素材であるために、金属材料の電気伝導度の基準になっている。
従って、実用性では銅が望ましいが、耐薬品性等、目的に応じて他の金属も本発明の温度感作実験用微量試験管の素材としては応用が可能である。この性質はまた微量溶液中の電気伝導度の測定を行う場合等にもチューブとサンプル中に針状の端子を用いることで測定も容易である。
その結果、本発明によれば、微量な牛乳試料を用いて、実際の牛乳生産工場で用いられているものに近い加熱冷却温度の条件で牛乳の加熱殺菌試験が再現できるようになった。
従来からエッペンドルフチューブの名称に代表される蓋付きのポリエチレンやポリプロピレンなどのポリオレフィン製の小型試験管が試料の加熱等の実験に広く用いられている。
しかしながら、これらはポリメラーゼ連鎖反応(PCR)法のように熱感作における温度と時間管理の精密さを求められる場合には、適切ではなかった。
即ち、この方法では、試料の温度を一定まで上昇、また冷却により低下させる際に著しく時間を要し、鋭敏な温度コントロールは困難である。市乳製造過程の加熱殺菌温度カーブは再現困難である。
これは、1986年に開発されたポリメラーゼ連鎖反応(PCR)法用に用いられている小型蓋付チューブで、一般用途の小型プラスチックチューブと類似した形状をするが、加熱による内圧に絶えうる、蓋の密閉度と容器の厚さを薄くすることで、熱伝導性を改善したものである。
しかしながら、材質はポリエチレンやポリプロピレンなどのポリオレフィン製である。
このものは、通常のプラスチックチューブに比べ、試料の温度応答性は改善されているが、容器の壁が薄いことと、PCRの目的に限定された製品であるため、0.2〜0.5mlの容量の製品のみである。また、市乳製造過程の加熱殺菌温度カーブは再現困難である。
サンプルの加熱には大小のガラス製試験管も用いられてきたが、微量サンプルによる実験の温度管理は困難である。
このものは、プラスチックよりも素材の熱伝導性が高いため、試料の温度応答性は良いが微量な試料を用いた実験には不都合である。
このように従来法では上記の小型容器に試料を入れ、種々の方法により加熱し、微生物の殺菌効果が調べられてきたが、用いられる試験容器の熱伝導性が低いため、急速な加熱や急速な冷却が困難であった。
このスケールダウンした連続殺菌装置は、流体の連続加熱殺菌試験には望ましいが、加熱条件の微調整や実験的に病原体を接種しての殺菌効果の評価を繰り返し行うには困難が伴う。また、装置が非常に高価である。
本発明の温度感作実験用金属製微量試験管にプラスチックキャップを装着し、さらにデジタル温度計用のプローブを付けることで、温度加熱の条件検討や鋭敏な温度上昇と下降を試料に与えることが容易である。この温度加熱には、通常の加熱乃至冷却用のアルミブロックを備えた温度調節装置、乃至は、ウォーターバスや氷、ドライアイスなども利用可能である。
具体的には、例えば低温長時間法(63℃30分間)、高温短時間法(72℃15秒間)
などの条件にて牛乳の加熱殺菌試験が行われる。
図1に示す形状を有する温度感作実験用金属製微量試験管(銅製)を用いて、牛乳の72℃加熱冷却試験(バッチ法)を行った。
具体的には、以下のようにして行った。
まず、図1に示す形状を有する温度感作実験用金属製微量試験管(銅製)に、試料(牛乳)を0.5ml入れた。
これを精密温度制御ヒートブロックに挿入し、デジタル温度レコーダー(TR−71U)を用いて、4℃から72℃へと温度を上昇させ、次いで72度から再度4℃へと冷却する操作を行った。
このときの加熱と冷却操作の温度パターンを図2に示す。
図2によれば、72℃への上昇に40秒間、冷却に40秒間要したことが分かる。
実施例1において、図2に示す形状を有する温度感作実験用金属製微量試験管(銅製)の代わりに、ポリエチレン製のチューブを用いたこと以外は、実施例1と同様にして行った。
このときの加熱と冷却操作の温度パターンを図3に示す。
図3によれば、72℃への上昇に240秒間、冷却に260秒間要したことが分かる。
なお、図4は、実施例1の温度上昇と下降条件に温度72℃、保持時間15秒の条件を加えて、実際の工場における牛乳の72℃、15秒殺菌温度条件ができることを示した実験結果を示すグラフである。

- 牛乳についての加熱殺菌条件をシミュレーションする加熱殺菌評価試験を行うにあたり、熱伝導度が0.708cal/s/cm 2 (℃/cm)以上の金属からなる微量試験管を用いることを特徴とする、牛乳についての加熱殺菌条件をシミュレーションする加熱殺菌評価試験方法。
