NIK−SIVA結合
NIKのSIVAへの結合およびその同時発現の効果を、結合、同時発現および免疫沈降アッセイを用いて試験した。
抗HIS抗体は、シグマから購入した。抗mycモノクローナル抗体(クローン−9E10)は、myc−ペプチドアフィニティカラムにてマウス腹水から精製した。
HEK293T細胞を、10%ウシ胎児血清、100U/mlペニシリンおよび100μg/mlストレプトマイシンを加えたダルベッコ改変イーグル最少必須培地中で培養した。
N末端をmycタグ(EQKLISEEDL、配列番号:5)に融合させたNIK発現用ベクター(配列番号:1)を、Michael Kracht博士(ドイツ)から頂いた。ヒトSIVA1(配列番号:6)およびSIVA2(配列番号:7)のcDNAを、ESTからPCR増幅し、pcDNA3.1-HISベクター(インビトロジェン)うちにクローン化した。pEGFPプラスミドはクロンテックから購入した。マウスaly変異(G860R)(Shinkura et al., 1999; NM_016896)に相当する変異を有するヒトNIKを、部位特異的変異誘発キット(ストラタジーン)により、(センス)5'CCAAGCTATTTCAATCGTGTGAAAGTCCAAATAC(配列番号:8)および(アンチセンス)5'GTATTTGGACTTTCACACGATTGAAATAGCTTGG(配列番号:9)を用いて作製した。
pcNIKベクター由来のBamHI/XhoI消化NIK挿入断片を、Gal4 DNA結合性ドメインベクターpGBKT7(クロンテック)のBamHI/SalI認識部位内にサブクローン化した。形質転換済みヒト骨髄ライブラリー(HL4053AH,クロンテック)を、製造業者の使用説明書(酵母プロトコルハンドブック, クロンテック− www.clontech.com)に従い、ベイトとしてpGBKT−NIKを用いたツーハイブリッドスクリーニングに供した。陽性クローンを、四連選択およびβ−ガラクトシダーゼ活性アッセイにより同定した。SIVAクローンのNIKおよびNIK624〜947への結合を再確認し、NIKのTRAF2への結合は、酵母SFY526レポーター系統(クロンテック)ならびにpGBKT7およびpGADT7ベクターを用いたβ-ガラクトシダーゼ発現アッセイにより評価した。
トランスフェクトされたタンパク質の免疫共沈降のため、HEK293T細胞を90mm径プレート上に播種し(1.5×106細胞/プレート)、1日後に、リン酸カルシウム沈降法(Sambrook et al., 1989)を用い、全量10μgのDNAを用いて、10%FBSを含むDMEM培地10ml中でトランスフェクトした。コトランスフェクションでは、被験タンパク質をコードするプラスミドの1:1混合物を使用した。トランスフェクション の24時間後、細胞をリン酸緩衝生理食塩水(PBS)で1回リンスし、1mlの溶解バッファー(10 mM Tris−HCl(pH 7.6)、250 mM NaCl、1%NP−40、1 mM EDTA、1 mM PMSF)(これは、1×完全プロテアーゼ抑制因子カクテル(ロッシュ モレキュラー バイオケミカルズ)を含む)中で溶解した。予備精製したライセートを2時間4℃でプロテインG-セファロースビーズ(アマシャム バイオサイエンス)にあらかじめ吸着させた抗mycまたは抗HIS抗体2μgとともにインキュベートした。次いで、ビーズを溶解バッファーでリンスし、SDS−PAGEに供し、タンパク質をニトロセルロース膜に移し、表示した抗体でプローブ標識した。抗体を、ホースラディッシュペルオキシダーゼ(HRP)標識二次抗体により、高感度化学発光(ECL)ウエスタンブロテッィング検出システム(アマシャム)を製造業者の使用説明書にしたがって用いて可視化した。
NIK媒介NF−κB活性化を、レポーター遺伝子アッセイにより測定した。HEK293T細胞(1.5×105/ウェル)を、6ウェルプレート上に播種し、1日後に、リン酸カルシウム沈降法(Sambrook et al., 1989)によりトランスフェクトした。コトランスフェクションでは、被験タンパク質をコードするプラスミドの1:1混合物を使用した。全DNA濃度を2μg/ウェルに維持するため、pcDNA3(インビトロジェン)「空」ベクターを添加した。トランスフェクションの24時間後、細胞を回収し、溶解し、レポーター遺伝子活性をルシフェラーゼアッセイシステム(プロメガ)を用いて測定した。
NIKをベイトとして用いたヒト骨髄ツーハイブリッドライブラリーのスクリーニングにより、NIKは、SIVAと称するタンパク質(これは、主にTおよびBリンパ球で発現されるTNF/NGFファミリーのレセプターであるCD27と会合していることが以前に示された(Prasad et al., 1997))のC末端部分に特異的に結合することがわかった。TRAFのNIKへの結合のように(Malinin et al., 1997)、本発明の研究では、SIVAのC末端部分はNIKのC末端部分に結合すること、およびこの結合はタンパク質全体でなくそれぞれの部分を結合アッセイに用いた場合、より強くなることが示された(図1a)。これは、おそらく、NIKのN末端部分が自身のC末端に結合し、したがって、この部分が他のタンパク質と結合することを阻止する傾向性によるものである(Xiao and Sun, 2000)。
CD27はリンパ球においてIκBとNF−κB2/p100の両方のプロセシングを誘導し、一方で、Ramosリンパ芽球細胞内でのNIK合成の停止は、CD27誘導NF−κB活性化を阻止する
CD70(CD27リガンド)が、リンパ球においてIκBとNF−κB2/p100の両方のプロセッシングを誘導する能力を、p100、p52、RelB、IκBαおよびp65に指向される抗体を用いたウエスタンブロット解析により測定した。誘導されるプロセッシングに対してこれらの細胞内のNIKの存在が与える効果を、NIK抑制細胞における同分子のウエスタンブロット解析により評価した。
CD70(GenBank受託番号Y13636)を、関連発現構築物でのヒト胚性腎(HEK)293T細胞の大規模トランスフェクションにより産生させた(下記参照)。MG132は、カルビオケムから購入した。Ficoll−Paqueは、アマシャム バイオサイエンスから購入した。G418は、ライフ テクノロジーから購入した。フィトヘムアグルチニン(PHA)はシグマから購入した。すべての試験において、トランスフェクト細胞の条件培地を1:4の希釈率で用いた。
抗p52抗体は、アップステート バイオテクノロジーから購入し、p65、RelB、CD27およびラミン A/Cに対する抗体は、サンタ クルス バイオテクノロジーから、抗IκBαはトランスダクション ラボラトリーから購入し、抗mycモノクローナル抗体は、実施例1に記載のようにして精製した。抗NIKモノクローナル抗体NIK−81は、N末端に結合するためのシステインを含むNIKキナーゼドメイン(RLGRGSFGEVHRMEDK、配列番号:10)内の配列に相当するKLH結合ペプチドでマウスを免疫化することによって生成させた。抗NIKモノクローナル抗体を、アフィゲル(バイオラッド)にて、BSA結合ペプチドを結合させたアフィニティカラムで精製した。
ヒト末梢血単核球(PBMC)を、軟膜試料から、450×gでのFicoll−Paque勾配遠心分離により単離した。細胞を、前処理なしでCD70での刺激に供するか、または、1μg/mlのPHAにより48時間活性化した後、PHAなしで12時間放置した。PBMC、ならびにバーキットリンパ腫起源のヒトBリンパ芽球株の細胞、Ramos細胞(Benjamin et al., 1982)、Raji細胞(Pulvertaft, 1964)を、10%ウシ胎児血清、100U/mlペニシリンおよび100μg/mlストレプトマイシンを加えたRPMI培地中で培養した。HEK293T細胞は、実施例の項目の実施例1に記載のようにして培養した。
mCD70の細胞外ドメインのcDNAを、ESTからPCR増幅し、修飾ロイシンジッパーおよびFLAGタグ(Fanslow et al., 1994)と融合させた状態でpcDNA3(インビトロジェン)うちにクローン化した。完全長NIKに相当するcDNAを、Gal4 DNA結合性ドメインベクターpGBKT7(クロンテック)うちにクローン化した。配列をNIK siRNAに非相補的となるよう改変したNIKを、(センス)5'GAGGGTCTGGAATACCTACATTCCCGCAGGATTCTGCATGGG(配列番号:11)および(アンチセンス)5'CCCATGCAGAATCCTGCGGGAATGTAGGTATTCCAGACCCTC(配列番号:12)をプライマーとして用いて作製した。
ヘアピンsiRNAを、pSUPERベクターを用い、既述(Brummelkamp et al., 2002)のようにして発現させた。簡単には、二本鎖オリゴヌクレオチドを、ヒトNIKオープンリーディングフレーム(ヌクレオチド1513〜1531)内の9塩基対スペーサー領域(ttcaagaga 配列番号:15)により連結された領域に相当するフォワード配列およびリバース配列:センス鎖5' gatccccTACCTCCACTCACGAAGGAttcaagagaTCCTTCGTGAGTGGAGGTAtttttggaaa(配列番号:13) ;アンチセンス鎖5'agcttttccaaaaaTACCTCCACTCACGAAGGAtctcttgaaTCCTTCGTGAGTGGAGGTAggg(配列番号:14)を含むように設計した。2つのオリゴヌクレオチドをアニーリングし、pSUPERのBglIIおよびHindIII(Brummelkamp et al., 2002)部位内にクローン化し、H1 RNAプロモーター(Brummelkamp et al., 2002)の制御下で発現させた。コトランスフェクトNIKに対して5倍過剰までのこのpSUPER−NIKで、上記のようにして一過性トランスフェクションを行なった。
トランスフェクション、免疫ブロテッィングおよび免疫沈降は、実施例の項目の実施例1に記載のようにして行なった。
対照およびNIK欠損Ramos細胞のPKC活性を、CD70刺激の(0、15および30分間)後に、Signatect Protein Kinase C アッセイシステム(プロメガ)を用いて測定した。PKCの酵素活性は、リン脂質の非存在下でのアッセイ時に得られた値をその存在下で得られた値から差し引くことにより測定した。
NIKがSIVAに結合する能力は、NIKがCD27の細胞機能に、ある役割を果たし得ることを示唆した。したがって、代替NF−κB活性化経路(これには、NIK機能が関与している)に対するCD27の効果を調べた。
CD27による種々のNF−κB形態の活性化におけるNIKの役割を調べるため、Ramos細胞におけるNIK合成を、NIK合成を阻止することができるヘアピン低分子干渉性RNA(siRNA)を発現するレンチウイルス系ベクターをこれらの細胞に感染させることによって停止させた。ウエスタン解析により、一過性発現様式および安定的発現様式の両方を確認し、siRNAベクターは、有効にNIKの合成を停止させた(図2f、上および中央パネル)。既報のNF−κB代替経路におけるNIKの関与から予測されるように、CD70でのNIK(MINUS)Ramos細胞の処理では、p52またはRelBのその核への移行は誘導されなかった。これらの細胞においてCD70により誘導されたp100の移行もまた、有意に遅滞された(図2d、右パネル)。また、NIK欠損により、CD70は、IκBα分解も核p65移行(ともに標準経路の表示)も誘導することができなかった(図2e、右パネル)。
標準および代替NF−κB経路に対するNIK抑制の効果
CD40、BLyS、TNF、タプシガルジンまたはPMAが、標準および代替NF−κB経路を誘導する能力を、p100、p52、RelB、IκBαおよびp65に指向される抗体を用いたウエスタンブロット解析により測定した。これらの細胞内にNIKが存在する効果を、NIK抑制細胞における同分子のウエスタンブロット解析により評価した。
hCD154(CD40L)およびhBLyS/BAFFの産生は、関連発現構築物でのヒト胚性腎(HEK)293T細胞の大規模トランスフェクションにより行った(下記参照)。すべての試験において、トランスフェクト細胞の条件培地は、実施例2に記載のとおりとした。G. Adolf博士(Boehringer Institute, Vienna,オーストリア)より頂いたTNFを細胞に50 ng/mlの濃度で適用した。タプシガルジン、4β−ホルボール−12−ミリステート−13−アセテート(PMA)およびフィトヘムアグルチニン(PHA)は、シグマから購入した。
p65、RelB、p52、p100、IκBαに対する抗体の供給源は、実施例の項目の実施例2に記載している。抗c−Relは、サンタ クルス バイオテクノロジーから購入した。抗βアクチンおよび抗FLAGは、シグマから購入した。抗ホスホIκBαは、セル シグナリング テクノロジーから購入した。
Ramos(Benjamin et al., 1982)細胞は、実施例の項目の実施例2に記載のようにして培養した。
hCD154(CD40L)(ATCCクローン79814)およびhBLyS/BAFF(Resgenクローン631119)の細胞外ドメインのcDNAを、ESTからPCR増幅し、修飾ロイシンジッパーおよびFLAGタグ(Fanslow et al., 1994)と融合させた状態でpcDNA3(インビトロジェン)うちにクローン化した。NIK抑制は、実施例の項目の実施例2に記載のようにして生成させた。
トランスフェクション、免疫ブロテッィングおよび免疫沈降は、実施例の項目の実施例1に記載のようにして行なった。リン酸化IκBαを、プロテオソーム阻害剤MG132(25μM)で前処理(2時間)した後、検出した。
IκBα分解に対するNIKのリン酸化活性化-ループに対する抗体の効果
NIKのリン酸化活性化-ループ(αp−NIK)に対する抗体を、Ramos細胞およびBJAB細胞内に導入し、NIK活性を即座に除去した。CD70およびCD40LおよびTNFによるIκBα分解の誘導に対するこれらの抗体の効果を測定した。
CD70は、実施例の項目の実施例2に記載のようにして生成した。CD40Lは、実施例の項目の実施例3に記載のようにして生成した。TNFは、実施例の項目の実施例3に記載のようにして得た。[γ32P]ATPは、アマシャム バイオサイエンスから購入した。
IκBα抗体は、実施例の項目の実施例2に記載している。抗mycモノクローナル抗体は、実施例の項目の実施例1に記載のようにして精製した。リン酸化NIK活性化ループ(α−pNIK)に対するモノクローナル抗体は、Thr559がリン酸化されたNIK活性化ループに相当するKLH結合ペプチドでマウスを免疫化することによって生成した。抗NIKモノクローナル抗体NIK−81は、実施例の項目の実施例2に記載のようにして生成した。両方の抗NIKモノクローナル抗体を、その対応するペプチドが結合されたアフィニティカラムにて精製した。
Ramos細胞およびHEK293T細胞は、実施例の項目の実施例1に記載している。BJAB細胞(Clements et al., 1975)は、10%ウシ胎児血清、100U/mlペニシリンおよび100μg/mlストレプトマイシンを加えたRPMI培地中で培養した。
CD70の細胞外ドメインのcDNAを、実施例の項目の実施例2に記載のようにしてクローン化した。hCD154(CD40L)およびhBLyS/BAFFは、実施例の項目の実施例3に記載のようにしてクローン化した。
トランスフェクション、免疫ブロテッィングおよび免疫沈降は、実施例の項目の実施例1および実施例2に記載のようにして行なった。
Pro−ject Protein Transfection Reagent Kit(ピアース)を製造業者の使用説明書にしたがって用い、血清無含有培地中で、抗体を細胞内にトランスフェクトした。抗体トランスフェクション後、リガンドを、通常(血清含有)培地(10%FBS含有RPMI 1640)中に3〜4時間適用した。
FITCタグ化免疫グロブリン取込みの評価を、トランスフェクションの0、1、4および8時間後に、蛍光顕微鏡検査により行なった。
標準経路の活性化に対するCD70の効果
TNFおよびCD70がIKKシグナルソームを活性化する能力を比較した。標準経路の機序に対するその効果を、対照およびNIK欠損細胞において調べた。該細胞内にNIKが存在する効果を、NIK抑制細胞において同分子のウエスタンブロット解析により評価した。
CD70は、実施例の項目の実施例2に記載してあり、TNFは、実施例の項目の実施例3に記載している。
抗mycは、実施例の項目の実施例1に記載のようにして精製した。抗IκBαおよび抗NIK(NIK−81)は、実施例の項目の実施例2に記載している。抗ホスホIκBαは、実施例の項目の実施例3に記載している。リン酸化NIK活性化ループ(α−pNIK)に対する抗体は、実施例の項目の実施例4に記載している。抗FLAG M2-ビーズは、シグマから、IKKα(M280 & H744)は、サンタ クルス バイオテクノロジーから、抗IKKβおよび抗IKKγは、BD―ファーミンゲンから購入した。
Ramos細胞およびPBMC細胞は、実施例の項目の実施例2に記載している。
GST−IκBαは、シグナル ファーマシューティカルより頂いた。NIK抑制ベクターは、実施例の項目の実施例2に記載している。
トランスフェクション、免疫ブロテッィングおよび免疫沈降は、実施例の項目の実施例2および実施例3に記載のようにして行った。休止PBMCとRamos細胞由来のCD70/CD27リガンド-レセプター複合体の免疫沈降では、細胞ライセートを、前述のキナーゼ試験について記載したようにして調製し、4時間、4℃で、25μlの50%M2−FLAGアガロースビーズ/ml ライセートとともにインキュベートした。TNF−レセプター複合体を、記載(Zhang et al., 2000)のようにして沈降させた。細胞ライセート由来のシグナルソームは、IKK1に対する2種類の異なる抗体の1:1混合物を用いて免疫沈降させた(M−280およびH−744, サンタ クルス)。25μlの50%プロテインA−セファロースビーズ/ml ライセートに吸着させた抗IKKα抗体10μgを用い、免疫沈降を2時間4℃で継続させた。
レセプター複合体および細胞質内シグナルソームのインビトロIKKキナーゼ活性を、細菌で発現させたGST−IκBα(1−54)(Uhlik et al., 1998 and Dejardin et al., 2002))を基質として用い、既報(Uhlik et al., 1998 and Dejardin et al., 2002)のようにして評価した。つまり、2〜4×108Ramos細胞を、30分間4℃で、溶解バッファー(20 mM HEPES pH 7.6、250 mM NaCl、0.5% NP−40、20 mM β−グリセロホスフェート、1 mM EDTA、20 mM p−ニトロフェニルホスフェート、0.1 mM バナジン酸ナトリウム、2 mM フッ化ナトリウム、1 mM DTT、1 mM PMSFおよび1×完全プロテアーゼ抑制因子カクテル)中で撹拌することにより溶解した。細胞残屑を10,000×gでの遠心分離により除去し、ライセートを、プロテインA/Gビーズ(これには、ウサギ/マウス免疫前血清が吸着されている)で予備精製し、次いで、2時間4℃で免疫沈降に供した。免疫沈降物を、溶解バッファーで4回、およびキナーゼバッファー(20 mM HEPES pH 7.6、20 mM MgCl2、20 mMβ−グリセロホスフェート、1 mM EDTA、2 mM p−ニトロフェニルホスフェートおよび2 mM DTT)で2回洗浄した。キナーゼ反応は、20μlのビーズに結合した免疫沈降タンパク質を、1μgのGST−IκBα(1−54)および5μci[γ32P]ATPを含有するキナーゼバッファー(40μl)中、30℃で30分間インキュベートすることにより進行させた。NIKのキナーゼ活性を、同じ条件下でトランスフェクトHEK293T細胞内で過剰発現され、抗myc抗体を用いて免疫沈降させた、mycタグ化NIKを用いて評価した。キナーゼ試験は、α−pNIKまたは対照としてのIgGの存在下で、免疫沈降物を抗体と1時間、4℃でプレインキュベートした後に行なった。キナーゼ反応の試料は、SDS−PAGEにて分離し、ニトロセルロース膜に移し、オートラジオグラフィーにより可視化し、示されたタンパク質をウエスタンブロット解析に供した。
標準経路における重要な事象は、IKKシグナルソームのIκBキナーゼ活性に対する刺激である。これらの2つのリガンドによる標準経路活性化の機序の違いを調べるため、TNFおよびCD70がIKKシグナルソームを活性化する能力を比較した。
TNFおよびCD70によるNF−κB活性化を開始する機序の推測的モデル
TNFによるNF−κBのNIK非依存的活性化における開始事象と、CD70によるNIK依存的活性化における開始事象とを比較すると、標準経路の活性化におけるNIKの関与が、特定の誘導因子の効果に限定されることが本発明者らにより示される。

Claims (15)
- 免疫障害の治療のための医薬の製造における、NIK−SIVA複合体形成を減少させることができる薬剤の使用であって、該薬剤が、(1)配列番号:3の座標123〜175記載のアミノ酸配列領域または配列番号:4の座標58〜110記載のアミノ酸配列領域に結合することができる抗体、(2)低分子干渉性RNA分子、または(3)リボザイムである使用。
- 前記免疫障害が、多発性骨髄腫(MM)、後天性免疫不全症候群(AIDs)、シェーグレン症候群(SS)、B細胞慢性リンパ性白血病(B−CLL)、全身性エリテマトーデス、炎症性大腸疾患、全身性炎症性反応症候群(SIRS)、多臓器不全症候群(MODS)および急性呼吸窮迫症候群(ARDS)からなる群より選択される請求項1記載の使用。
- 低分子干渉性RNA分子が配列番号:15のものである請求項1または2記載の使用。
- 免疫障害の治療のための医薬の製造における、細胞のNIK依存性CD27調節を減少させることができる薬剤の使用であって、該薬剤が、配列番号:3の座標123〜175記載のアミノ酸配列領域または配列番号:4の座標58〜110記載のアミノ酸配列領域に結合することができる抗体または配列番号:15の低分子干渉性RNA分子である使用。
- 前記免疫障害が、多発性骨髄腫(MM)、後天性免疫不全症候群(AIDs)、シェーグレン症候群(SS)、B細胞慢性リンパ性白血病(B−CLL)、全身性エリテマトーデス、炎症性大腸疾患、全身性炎症性反応症候群(SIRS)、多臓器不全症候群(MODS)および急性呼吸窮迫症候群(ARDS)からなる群より選択される請求項4記載の使用。
- 供される細胞内でNIK発現を特異的に下方調節することができる核酸配列を含む単離ポリヌクレオチドであって、配列番号:15の低分子干渉性RNA分子であるポリヌクレオチド。
- 請求項6記載の単離ポリヌクレオチドを含有する核酸構築物。
- 請求項7記載の核酸構築物を含む細胞。
- 配列番号:3の座標123〜175記載のアミノ酸配列領域に特異的に結合することができる抗体または抗体断片。
- 配列番号:4の座標58〜110記載のアミノ酸配列領域に特異的に結合することができる抗体または抗体断片。
- 細胞を、該細胞内でNIK依存性標準経路および代替経路を誘導することができるTNF/NGFレセプターファミリーのリガンドと接触させること、前記接触前、接触後、または接触中に細胞を個々の被験分子とともにインキュベートすること、細胞内のIκB分解、IκBαリン酸化およびp65移行から選択される標準経路活性化を示すパラメータを検出すること、および前記リガンドによって誘導される標準経路の誘導をモジュレートすることができる単一の分子/分子群を選択することを含む、NIKの活性を減少させることができる分子のインビトロスクリーニング方法であって、該TNF/NGFのリガンドが、CD70、CD40LおよびBlys/BAFFから選択される方法。
- 細胞がリンパ芽球型である請求項11記載の方法。
- リンパ芽球細胞を、該細胞内でNIKおよび標準経路を活性化することができるTNF/NGFレセプターファミリーのリガンドと接触させること、前記接触前、接触後、または接触中に細胞を個々の被験分子とともにインキュベートすること、IκB分解、IκBαリン酸化およびp65移行から選択される標準経路活性化を示すパラメータを検出すること、および前記リガンドによって誘導されるが、NIK非依存的に標準経路を誘導することができる他のどのリガンドによっても誘導されない標準経路の誘導をモジュレートすることができる単一の分子/分子群を選択することを含むNIK活性を減少させることができる分子のスクリーニング方法であって、該TNF/NGFのリガンドが、CD70、CD40LおよびBlys/BAFFから選択される方法。
- 細胞が、Ramos細胞、Raji細胞およびBJAB細胞から選択される請求項12または13記載の方法。
- NIK非依存的形態で標準経路を誘導することができるリガンドがTNFである請求項13記載の方法。
