本件出願は2004年9月3日に出願された、米国仮特許出願第60/606,941号(参考として本明細書に含まれる)からの優先権の利益を主張する。
連邦後援研究に関する記述
本発明は補助金PO1 AI053725、RO1 AI38339及びAI50847のもとに、国立健康協会により授与された米国政府支持によりなされた。米国政府は本発明に或る種の権利を有する。
NKT細胞は抗菌応答、坑腫瘍免疫性及びトレランスと自己免疫の間のバランスの調節を含む、幾つかの免疫機能に重要な役割を果たす。それらは典型的には保存された内因性リガンドの自己反応性認識と関連する自然記憶表現型を発現する。
NKT細胞についての幾つかの天然アゴニスト及び合成アゴニストが報告されていた。in vitro及びin vivoのNKT細胞活性化を研究するのに使用されるプロトタイプの化合物は海洋海綿アゲラス・マウリチアヌス(Agelas mauritianus)から最初に単離されたα-ガラクトシルセラミド(αGalCer)である、KRN7000である(Kawanoら, Proc. Natl. Acad. Sci. 278, 1626-29 (1997); またTaniguchiらの米国特許第6,531,453号を参照のこと)。先の研究はまたCD1d分子のリソソームのトラフィッキングについての要件(Chiu, YHら, Nat. Immunol. 3, 55-60 (2002))、並びにリソソームのプロテアーゼ(Honey, Kら, Nat. Immunol. 3, 1069-74 (2002))及びスフィンゴ脂質アクチベータータンパク質、又はサポシン(Zhou, Dら, Science 303, 523-27 (2004); Kang SJら, Nat. Immunol. 5, 175-81 (2004); Winau Fら, Nat. Immunol. 5, 169-74 (2004))の役割を証明していた。しかしながら、NKT細胞受容体の天然リガンドは既に同定されていなかった。
それ故、第一の局面において、本発明はNKT細胞をNKT細胞からのサイトカインの分泌を誘発し、NKT細胞の増殖を刺激し、又はNKT細胞の細胞表面マーカーの発現をアップレギュレートするのに充分な量のiGb3と接触させる工程を含むNKT細胞の活性化方法を提供する。
別の局面において、本発明はiGb3をNKT細胞からのサイトカインの分泌を誘発し、NKT細胞の増殖を刺激し、又はNKT細胞の細胞表面受容体の発現をアップレギュレートするのに充分な量で対象に投与する工程を含む対象のNKT細胞の活性化方法を提供する。
それ故、一実施態様において、本発明はNKT細胞をiGb3と接触させることによるNKT細胞の活性化方法を提供する。iGb3の構造は下記の化学式により表される。
本発明の方法に従ってNKT細胞を活性化するために、NKT細胞が先にリストされた観察できる効果のいずれかを誘発するのに充分な量でiGb3と接触させられる。in vivo及びex vivoのNKT細胞活性化が、本明細書で以下に説明されるように、また意図されている。
“サイトカイン”は、その用語が本明細書及び当業界で使用されるように、細胞間連絡の媒介物質として作用する細胞外シグナリングタンパク質又はペプチドである。“サイトカイン”という用語はあらゆるこのようなシグナリング分子を含み、リンホカイン、インターロイキン、腫瘍壊死因子、顆粒球-マクロファージコロニー活性化因子及びインターフェロンが挙げられるが、これらに限定されない。
NKT細胞により分泌されるサイトカインは細胞媒介炎症反応をダウンレギュレートもしくは加減し、又はその他の免疫抑制特性もしくは免疫調節特性を示し得る。免疫抑制サイトカイン又は免疫調節サイトカインの例として、IL-10、IL-4、及びIL-12、IL-13並びにGM-CSFが挙げられるが、これらに限定されない。また、NKT細胞により分泌されるサイトカインは炎症反応の増幅に関係し得る。炎症性サイトカインとして、IFN-γ、IL-2、IL-1、IL-6、IL-8、TNF、及びTGF-βが挙げられるが、これらに限定されない。サイトカインに対する宿主応答は主として多要因的であり、それ故、先にリストされた特別なサイトカインが細胞の状況に応じて、炎症反応又は免疫調節応答を誘発し得ることが理解される。更に、上記サイトカインのいずれかの組み合わせが活性化後にNKT細胞により分泌されるかもしれない。
分泌されたサイトカインのレベルの検出方法及び測定方法は当業界で公知であり、例えば、ELISA、ウェスタンブロッティング、FACS等を含む。
NKT細胞増殖はまたiGb3との接触により活性化後に誘発されるかもしれない。増殖は通常の方法、例えば、3H-チミジンアッセイもしくはBrdUとり込みアッセイ又はトリパンブルー染色によりin vitroで好適に測定される。
細胞表面マーカーのアップレギュレーションはまたNKT細胞の活性化後に好適に観察される。例えば、CD69受容体、CD25受容体、CD40L受容体及びIL-12受容体がNKT細胞の活性化後にアップレギュレートされる。免疫学的方法、例えば、FACSだけでなく、当業界で普通に使用されるその他の方法が細胞表面マーカーのアップレギュレーションを検出するのに使用し得る。
本方法において、NKT細胞の活性化は典型的にはNKT細胞のTCRをiGb3と接触させることにより開始される。iGb3は抗原提示細胞、例えば、樹状細胞の表面のCD1d分子により提示し得るが、直接の刺激、即ち、“遊離”iGb3とのTCRの接触がまた意図されている。iGb3は精製形態で提供されてもよく、又は合成であってもよい。本明細書に使用される“精製”は天然源から分離された化合物を表すが、特別な程度の純度は必要とされない。本明細書に使用される“合成”は化学合成方法に従って生成され、又は酵素もしくは基質の作用により生成された化合物を表す。例えば、iGb3はラクトシルセラミドに対するiGb3シンターゼの作用により生成されてもよく、又はiGb4(これは下記の化学構造により表される)に対するβ-ヘキソサミニダーゼの作用により生成されてもよい。
iGb3はゴルジ装置中でiGb4の合成中に、またリソソーム中でiGb4の分解中に一時的中間体として発生される(図3Aを参照のこと)。本発明者らはリソソームのiGb3がCD1d制限NKT細胞の抗原の源として利用できることを発見した。それ故、本方法はiGb4を“iGb3前駆体”として抗原提示細胞に提供することを含んでもよく、この場合、それがリソソーム中でiGb3に分解され、CD1d分子と会合され、NKT細胞への提示のために原形質膜にシャトルされる。
理論により束縛されるべきではないが、リソソームのiGb3は型I糖尿病及び癌で異常調節されたかもしれないと仮定され、そこではNKT細胞が夫々、Th2サイトカイン及びTh1サイトカインにより媒介される保護機能を与える。更に、外因性ではなく内因性のリガンドがサルモネラによる感染中にNKT細胞による保護IFN-γ放出を誘発するので、iGb3は感染中にNKT細胞を同様に活性化し得る。それ故、本発明は対象内のNKT細胞集団を活性化し、又はNKT細胞集団をex vivoで活性化し、活性化されたNKT細胞集団を対象に逆に再導入することを意図している。対象は哺乳類、例えば、ヒト又はマウスであることが好適である。
対象中のNKT細胞集団の活性化方法はiGb3又はiGb3前駆体を対象に投与することを含む。本発明の幾つかの方法による対象への投与は最初にiGb3又はiGb3前駆体を医薬上許される担体及び/又は賦形剤とともに製剤化して所望の用量等を得ることを含んでもよい。治療化合物に適した製剤は当業界で知られている。投与はとりわけ、腹腔内、静脈内、筋肉内、皮下、経皮、経口、鼻咽頭又は経粘膜の吸収を含む、あらゆる好適な方法により行なわれてもよい。好適には、化合物がNKT細胞集団を活性化するのに有効な量で投与され、その結果、治療効果、例えば、坑腫瘍効果又は坑糖尿病効果が対象で得られる。
いずれかの所定の場合に投与される特別な用量は、当業者により公知であるように、投与される特別な化合物(例えば、iGb3又はiGb3前駆体)、治療すべき疾患、対象の状態、及び薬物の活性又は対象の応答を変更し得るその他の関連する医療因子に従って調節されることが理解されるであろう。例えば、特別な患者に特別な用量は年齢、体重、全般の健康の状態、ダイエット、投与の時期及び様式、排泄の速度、及び組み合わせて使用される薬物並びに治療が適用される特別な疾患の重度に依存する。所定の患者のための用量は通常の考慮を使用して、例えば、iGb3(又はiGb3前駆体)及び既知の薬剤の異なる活性の通例の比較、例えば、適当な通常の薬理学的プロトコルにより決められる。
対象についての最大用量は望ましくない、又は許容できない副作用を生じない最高用量である。個々の治療養生法に関する変数の数が大きく、用量のかなりの範囲が予想される。本発明のiGb3(又はiGb3前駆体)の用量が治療前の症候と較べて症候を少なくとも50%軽減すると予想される。
以下の実施例は本発明の更なる理解を助けるために示される。使用される特別な物質及び条件は本発明の更なる例示であることが意図され、特許請求の範囲の妥当な範囲を限定するものではない。
下記の物質及び方法を実施例2-5に記載される実験に使用した。
マウス ジャクソン・ラブズ(バー・ハーバー、MA)からのβ2M-/-、CD1d-/-及びCD1-TD“ノックイン”マウス(これらはテール欠失されたCD1d分子を有する)、αGa1A-/-マウスはC57BL/6バックグラウンド中であった。Hexb-/-マウス、GM2-/-マウス及びGM3-/-マウスは129/Svバックグラウンド中であった。全ての場合に、異型接合交配から得られた同腹兄弟をPCRにより遺伝子型を決め、比較分析に使用した。全てのマウスを協会の動物ケアー及び使用委員会ガイドラインに従ってシカゴ大学で特別の無病原体環境中で飼育した。
リンパ球調製及びフローサイトメトリー リンパ球調製、CD1d-αGalCerテトラマー、及びフローサイトメトリー染色を通常のプロトコル、例えば、Zhou Dら, Science 303, 523-27 (2004)(参考として本明細書にそのまま含まれる)に記載されたものに従って行なった。
CD1d制限T細胞応答 抗原提示細胞は5x105の細胞/ウェルで培養されたマウス脾臓細胞、GMC-SF及びIL4の存在下で生成され、10ng/mlのTNF-αで一夜活性化され、5x104の細胞/ウェルで培養されたマウス骨髄由来樹状細胞、及び2.5x105の細胞/ウェルで培養されたヒトPBMC又はGM-CSF/IL-4培養PBMC由来樹状細胞であった。NKTハイブリドーマを5x104の細胞/ウェルで培養し、ヒトNKT系を2.5x105の細胞/ウェルで培養した。培養上澄み中に放出されたサイトカインをヒトIL-4及びIFN-yについて通常のELISA(ファーミンゲン-ベクトン・ディキンソン、CA)により、またマウスハイブリドーマについて指示薬CTLL IL2バイオアッセイにより測定した。NKTハイブリドーマDN32.D3(Vα14+)、TCBII(Vα14-)、TBA7(VαI4-)及びラット好塩基球白血病RBL.CD1dトランスフェクタント系をPark, S-Hら, J. Immunol. 160 3128-34 (1998)(参考として本明細書にそのまま含まれる)に記載されたように使用した。ヒトポリクローナルVα24Vβ11 NKT系をin vitroで健康なヒトPBLの反復αGalCer刺激により誘導し、PHA及びIL2再刺激により管理し、2種の異なるサブクローン、CD4及びDNを実験に使用した。グリフォニア・シンプリシフォリア(Griffonia Simplicifolia)イソロイシンB4(IB4)はベクター・ラボラトリィズからのものであり、また坑ヒトCD1d mAb 51をS. Porcelli博士から得た。合成糖脂質による刺激のために、APCを6時間にわたって種々の濃度の脂質(DMSO中の原液から)でパルスし、洗浄し、18-24時間にわたってNKT細胞ハイブリドーマ又は細胞系とともにインキュベートした。

坑FLAG M2アガロースビーズ(シグマ)による免疫精製のためのN末端FLAGペプチドを用いて、Sf9昆虫細胞発現系を使用して、組換え酵素を生成した。FLAG標識iGb3を、図6Aに示されたように、ウェスタンブロットにより検出した。
iGb3を合成するために、精製組換えiGb3シンターゼを100 mM トリス緩衝液(pH 7.4)中の2 mM UDP-ガラクトース(シグマ)、0.2%トリトンX-100、ラクトシルセラミド(マトレヤ)200μg及び20 mM MnCl2の混合物1mlに37℃で一夜添加した。糖脂質を逆相C18カラムクロマトグラフィーにより精製し、10%から100%までのメタノールから無勾配溶離により溶離した。反応生成物を、図6Bに示されたように、HPTLCにより分析した。糖スフィンゴ脂質サンプルをCDCl3-CD30D 1:1の反復添加、音波処理、及び乾燥窒素下の蒸発により重水素交換し、次いでDMSO-d6/2%D2O(これは化学シフト基準として0.03%テトラメチルシランを含む)0.5mLに溶解した。図6Cに示された、1-D 1H-NMRスペクトルを、弛緩遅延中の前飽和パルスによる残留HODの抑制でもって、バリアン・イノバ500 MHzスペクトロメーターで35℃で得た。生合成生成物に関するスペクトルデータを実際に同じ条件下で得られた関連糖スフィンゴ脂質標準物質のデータだけでなく、既に公表されたデータとの比較により解読した。
Hexb-/-及びHexb-/-同腹兄弟の胸腺及び脾臓からのリンパ球をCD1d-αGalCerテトラマー及び坑CD44で染色した。変異体及び野生型マウスの胸腺及び脾臓中のリンパ球の絶対数は同様であった。図1Aに示されるように、リソソームの糖スフィンゴ脂質分解酵素β-ヘキソサミニダーゼbサブユニット欠乏の、Hexb-/-は、Vα14NKT細胞中でひどい減少を示した。胸腺及び脾臓の両方におけるCD1d-αGalCerテトラマー染色は対照同腹兄弟の5%に減少され、CD1d発現の欠損のマウス、例えば、β2-ミクログロブリン-/-(β2M-/-)マウス又はCD1d-/-マウスのバックグラウンドレベルに近かった(示されていない)。対照的に、図1Bに示されるように、古典的、ナイーブ及びメモリーCD4 T細胞及びCD8 T細胞だけでなく、B細胞、γδT細胞及びNK細胞(示されていない)の発生が、保存された。
CD1d表面発現は、図2Aに示されるように、変化されなかったが、Hexb-/-細胞はVα14NKT細胞ハイブリドーマDN32.D3から応答を誘発できなかった(CD1d発現を欠いているβ-2M-/-胸腺細胞が負の対照として利用できた)。対照的に、非Vα14, CD1d自己反応性NKTハイブリドーマ、例えば、TCB11の応答が保存され、Vα14NKT細胞の推定のリソソームのリガンドの生成の選択的欠損を示唆した。
Vα14NKT細胞による認識の前にαGalCerへのリソソームのプロセシングを必要とするαGalCerのジグリコシル化誘導体のパネルを使用して(Prigozyet TIら, Science 291, 664-7 (2001)(参考として本明細書にそのまま含まれる)に記載されたように)、Hexb-/-細胞のリソソームの機能を更に探査した。図2Bに示されるように、相当する酵素の既知の基質特異性と合致して、Hexb-/-細胞によるGalNAcβ1,4GalαCerの提示の選択的欠損があり、一方、対照的に、α-ガラクトシダーゼA(αGalA)-/-細胞はGalα1,4GalαCer及びGalα1,2GalαCerについて選択的欠損を示した。これらの結果はこれらの突然変異と関連する抗原提示欠損の特異的性質を実証する。予想されるように、CD1-TD“ノック-イン”細胞は、細胞質エンドソームのターゲッティングモチーフのトランケーションを有するCD1dの損なわれたリソソームの循環のために、これらの複合体糖脂質の全ての提示に著しく欠損していた。
化学合成したグロボトリヘキソシルセラミド(Gb3、Galα1,4Galβ1,4Glcβ1,1Cer)及びイソグロボトリヘキソシルセラミド(iGb3、Galα1,3Galβ1,4Glcβ1,1Cer)を抗原提示細胞の存在下でNKT細胞を刺激するそれらの能力について試験した。図3-4に示されるように、iGb3単独はヒトVα24 NKT細胞及びマウスVα14 NKT細胞の両方の強力な刺激物質であった。αGalCerのように、iGb3は新鮮なPBMCの4日培養物中でヒトVα14NTK細胞を選択的に刺激し、増大した(図3B)。照射PBMCにより提示された合成iGb3はポリクローナルヒトNKT系(図3C、左右のパネル)だけでなく、2/2クローン化CD4系及びDN系(示されていない)による強力なTh1(IFNγ)及びTh2(IL4)サイトカイン分泌を刺激した。ネコの腸から精製された天然iGb3を含む、その他の源に由来するiGB3、及びラクトシルセラミド及びUDP-ガラクトースに対するiGb3シンターゼの作用によりin vitroで生成されたiGb3は、αGalCerに匹敵するレベルでTh1サイトカイン及びTh2サイトカインを誘発して、同様に刺激性であった(図3C、右のパネル)。CD1d発現骨髄由来樹状細胞により提示されたiGb3はmVα14 NTK細胞ハイブリドーマDN32.D3(図4A)だけでなく、5/5のその他の個々のmVα14ハイブリドーマ(示されていない)を刺激し、それは3/3の非Vα14ハイブリドーマ(示されていない)を刺激できなかった。
図4A、右のパネルに示されるように、骨髄由来樹状細胞により提示されたiGb4はマウス及びヒトについて刺激性であり(示されていない)、一方、LacCer(これはまたGM3の分解により生成し得る)は刺激性ではなかった。また図4Aに示されるように、Hexb-/-細胞はiGb3を提示したが、iGb4を提示できず、リソソーム区画へのCD1dトラフィッキングが一部サポシンの必須の機能のためにこれらの抗原を提示するのに必須であった(図4B)。こうして、リガンド認識はイソグロボ系列の少なくとも三つのサッカリド残基を必要とする。
無細胞アッセイでは、iGb3及びiGb4の両方がCD1dに前ローディングされたGT1bを置換するのにサポシンBを必要とした(図4C、左のパネル)。更に、CD1d/iGb3複合体がVα14NKT細胞を直接刺激した(図4C、右のパネル)。対照的に、CD1d/iGb4のみが非常に弱い応答を誘発した。一緒になって、これらの結果はiGb3がVα14NKT細胞の直接のリガンドであること及びin vivoで、iGb4の遠位のサッカリド残基が原形質膜におけるTCR認識の前にリソソーム中のβ-ヘキソサミニダーゼの作用により除去される必要があったことを実証する。
Hexb-/-マウスにおけるVα14NKT細胞発生のブロック及びHexb-/-胸腺細胞がVα14NKTハイブリドーマを刺激することができないことはiGb3単独がmVα14NKT細胞及びhVα24NKT細胞の主な天然リガンドであり得ることを示唆した。iGb3中で見られるような末端Galα1,3Galに高度に特異性のレクチンである、グリフォニア・シンプリシフォリアイソレクチンB4(IB4)を使用してこれを試験した。図5Aに示されるように、IB4はαGalCerではなく、外因的に添加されたiGb3によりhVα24NKT細胞刺激を損なった。対照的に、坑CD1d mAbは両方の糖脂質による刺激をブロックした。これらの結果はIB4の特異性と合致し、サッカリド残基をTCR又はレクチンによる認識に露出されたCD1分子の溝の外部に置く現在の構造モデルから予想し得るように、それがたとえCD1dに結合された場合でさえもiGb3の末端サッカリドを認識することを示した。
IB4のこの性質を末端Galα1,3Gal残基がmVα14 NKT細胞及びhVα24 NKT細胞の自然刺激に有意に寄与するか否かを試験するのに利用した。図5Bに示されるように、IB4はマウスCD1dを発現するRBL細胞によるDN32.D3の自然自己反応性刺激を損ない、一方、予想されるように、対照非Vα14CD1d自己反応性ハイブリドーマ、例えば、TBA7及びTCB11の刺激は影響されなかった。更に、図5Cに示されるように、IB4はまたヒトVα24NKT系によるCD1d発現PBMC由来樹状細胞の自然認識をブロックしたが、外因的に添加されたαGalCerの認識をブロックすることができなかった。重要なことに、ヒトにおけるiGb3認識のIB4遮断はマウスよりも約1000倍少ないレクチンを一貫して必要とした(夫々、0.2ng/ml vs 0.2μg/ml)。これはおそらくヒトではなく、マウスが糖タンパク質について発現される非常に多量の付加的なIB4リガンド、Galα1,3Galエピトープを発現するからである。こうして、IB4ブロッキング実験はmVα14 NKT細胞及びhVα24 NKT細胞の両方の天然リガンドとしてのiGb3の顕著な役割の独立の確認を与えた。
図7に示された、化合物2をiGb3の生成で出発物質として使用した。また図7に示された、合成スキーム1を化合物2をドナー3とカップリングし、続いてベンジル保護基を除去することにより開始した。残っているアルコールをアシル化して化合物4を生成した。アノマーアルコールを放出し、トリサッカリドをセラミド5にカップリングした。化合物6を精製し、続いて脱保護してiGb3を良好な収率で得た。スキーム1で使用した試薬は以下のとおりであった(括弧中に収率):a)AgOTf、4ÅMS、CH2Cl2(61%);b)H2、Pd/C(10%)、EtOAc、EtOH(61%);c)Ac2O、Et3N、DMAP(95%);d)TFA、CH2Cl2(99%);e1)CCl3CN、K2CO3 e2)5、BF3-OEt、MS AW300、CH2Cl2(45%);f)NaOMe、MeOH(86%)。

Claims (30)
- NKT細胞と、NKT細胞からのサイトカインの分泌を誘発し、NKT細胞の増殖を刺激し、又はNKT細胞の細胞表面マーカーの発現をアップレギュレートするのに充分な量のiGb3とを、インビトロ又はヒトを除く対象のインビボで接触させることを特徴とするNKT細胞の活性化方法。
- iGb3が精製されているか又は合成である、請求項1記載の方法。
- NKT細胞の接触がNKT細胞のT細胞受容体を接触させることを含む、請求項1記載の方法。
- iGb3をCD1d分子を含む抗原提示細胞によりNKT細胞に提示する、請求項1記載の方法。
- 抗原提示細胞が樹状細胞である、請求項4記載の方法。
- iGb3前駆体を抗原提示細胞に与えてiGb3を生成する、請求項4記載の方法。
- iGb3前駆体がiGb4である、請求項6記載の方法。
