Description
より高いリポソーム濃度を有するエルゴステロールPCリポソームは良い耐用性があり慢性及び急性毒性の両方に関して安全であることが見いだされている。調合剤のml中アンフォテリシンB15mgまでを有するこれらのリポソーム化製剤は副作用の関係するあらゆる点滴なしに投与されることができる。これらの製剤のLD50はコレステロール−PCリポソーマルアンフォテリシンBによって報告されたものより高い。

Claims (8)
- 感染症治療に使用するためのステロール濃縮混合層状アンフォテリシン(Amphotericin)挿入リポソームを水溶液に懸濁して準備した懸濁液中のステロール濃縮混合層状アンフォテリシン(Amphotericin)挿入リポソームであって、
前記水溶液が、緩衝液を含まない0.9%の塩化ナトリウム水溶液であり、
前記懸濁液中におけるアンフォテリシンB(Amphotericin B)の濃度が1〜15mg/mlであるステロール濃縮混合層状アンフォテリシン(Amphotericin)挿入リポソーム。 - 前記ステロールは、エルゴステロールであるか、コレステロールとエルゴステロールである請求項1に記載のステロール濃縮混合層状アンフォテリシン挿入リポソーム。
- 前記ステロールは前記リポソームに含まれるリン脂質と結合している請求項1に記載のステロール濃縮混合層状アンフォテリシン挿入リポソーム。
- 前記リポソームに含まれるリン脂質はホスファチジルコリンである請求項1に記載のステロール濃縮混合層状アンフォテリシン挿入リポソーム。
- 前記エルゴステロールは前記リポソームの総脂質の50モル%以下を構成する請求項2に記載のステロール濃縮混合層状アンフォテリシン挿入リポソーム。
- 前記リポソームに含まれるリン脂質はホスファチジルコリンであり、
前記ホスファチジルコリン、前記エルゴステロール及び前記アンフォテリシンBのモル比率は5:2:1である請求項2に記載のステロール濃縮混合層状アンフォテリシン挿入リポソーム。 - 前記感染症はリーシュマニアを含む細胞内原虫寄生虫によるものである請求項1に記載のステロール濃縮混合層状アンフォテリシン挿入リポソーム。
- 細胞質の半減期を増加するために及びよりよい生体分布のために製造中及び投与前に超音波処理が行われる請求項1に記載のステロール濃縮混合層状アンフォテリシン挿入リポソーム。
