Description
精神及び神経学的病状は、このような病状に関連する兆候及び原因が多様であるため、効果的に治療することが極めて困難であるとされる。現在の薬剤療法は、一つの薬理学的効果に対して高度に選択的であるか、又は多様な兆候を緩和する試行の故に広範囲にわたって非選択的であるかのいずれかに重点的に取り組んでいる。高度な薬理学的選択性に重点をおいた療法では、多様な原因を伴う疾患に対する有益性が限定され、いくつかのケースでは兆候が悪化するおそれがあることも分かっている。例えば、統合失調症用のドーパミン作動性レセプターの選択的拮抗作用により、いくつかの負の兆候並びに遅発性ジスキネジーが悪化する。
他方、広範囲にわたる非選択的な薬剤療法により、多くの兆候を緩和することができるが、しばしばより多くの副作用が出る。例えば、現在の抗精神剤は、認知機能及び他の副作用の悪化、例えば起立性低血圧症、口渇、視力障害、便秘及び運動不全に関連した、アドレナリン作動性、コリン作動性、及びヒスタミン感作性のレセプターアンタゴニスト活性を有する。薬剤療法の選択にかかわらず、精神及び神経学的病状に使用される現在の製薬用化合物によっては、十分な治療はなされず、認知衰退等の兆候が残る。よって、このような病状を治療するのに使用される改善された製薬用化合物が必要とされている。
我々は、ここに記載するテトラヒドロインドロンアリールピペラジン化合物の組み合わせが、以下に記載するようなニューロパシー及び精神疾患を含む、痛み、嘔吐、神経変性に制限されない、多くの多様な精神及び神経学的病状を治療するのに有用であることを発見した。
ここで使用される場合、以下の用語は、明確に言及されない限り、以下の意味を有する。
「アルキル」なる用語は、その全てが置換されていてもよい、直鎖状鎖、分枝状鎖及び環状基を含む、飽和した脂肪族基を称する。好ましいアルキル基は1〜10の炭素原子を有する。適切なアルキル基には、メチル、エチル等が含まれ、置換されていてもよい。「ヘテロアルキル」なる用語は、炭素含有の直鎖状鎖、分枝状鎖及び環状基で、その全てが置換されていてもよく、少なくとも一のO、N又はSヘテロ原子を有するものを称する。「アルコキシ」なる用語は、エーテル、--O-アルキルを称し、ここでアルキルは上述にて定義したものである。
本発明の化合物は、一般的な概略構造{A}-L-{B}を有するもので、ここで、以下に示すように、Aはテトラヒドロインドロン誘導体であり、Lはヒドロカルビル鎖であり、Bはアリールピペラジン誘導体である。
本発明の化合物において、テトラヒドロインドロン誘導体(「THI」)は、次の式(I):
(a)Xは、CH又はNであり;
(b)R1は、水素、アルキル、アラルキル、ヘテロアラルキル、アルケニル、アラルケニル、ヘテロアラルケニル、アリール又はヘテロアリールであり;
(c)R2は、水素、アルキル、アラルキル、アリール又はヘテロアリールであり;
(d)R2’は、R2がメチルでなければ水素であり、このケースにおいて、R2’はまたメチルであり;
(e)L及びR3は以下に記載するものである]
の構造を有する。
(1)R2は、水素、アルキル、アラルキル、ヘテロアラルキル、アリール又はヘテロアリールであり;
(2)R2’は、R2がメチルでなければ水素であり、このケースにおいて、R2’はまたメチルであり;
(3)L及びR3は以下に記載するものである]
のテトラヒドロインドロン部分である。
好ましい特定の一実施態様において、R2及びR2’は双方とも水素である。この実施態様において、THI部分は未置換のテトラヒドロインドロン部分である。
上述したR3基はアリールピペラジン部分であり、本発明の化合物においては、次の式(III):
(i)R4は、水素、アルキル、ハロ、ヒドロキシ、アルコキシ、シアノ、ニトロ、ペルフルオロアルキル、ペルフルオロアルコキシ、又はヒドロキシメチルであり;
(ii)R5は、水素、アルキル、ハロ、アルコキシ、シアノ、ニトロ、ペルフルオロアルキル、ペルフルオロアルコキシ、アミノ、アミノカルボニル、アミノスルホニル、又はヒドロキシメチルであり;
(iii)R6は、アルキル、ハロ、アルコキシ、ペルフルオロアルキル、ペルフルオロアルコキシ、又はニトロであり;
(iv)R4及びR5は互いに共同する場合に、5又は6員環を形成可能で、一又は複数のヘテロ原子を含有可能であり;また
(v)R5及びR6は互いに共同する場合に、5又は6員環を形成可能で、一又は複数のヘテロ原子を含有可能である]
の構造を有する。
(i)R4は、水素、ハロ、又はアルコキシであり;
(ii)R5は、水素、アルキル、ハロ、アルコキシ、又はペルフルオロアルキルであり;
(iii)R6は、アルキル、ハロ、アルコキシ、又はペルフルオロアルキルであり;
(iv)R4及びR5は互いに共同する場合に、ナフタレン環を形成し;また
(v)R5及びR6は互いに共同する場合に、メチレンジオキシ又はエチレンジオキシ基のいずれかとすることができる。
本発明の組成物に使用されるリンカー部分(L)は、式-(CH2)m- の直鎖状鎖アルキル基とすることができ、ここでmは1〜6、好ましくは3、4又は5の整数である。また、リンカーは、次の式(IV):
(i)nは、0、1又は2であり;
(ii)R7及びR8は、水素、メチル又はエチルであり;
(iii)R9及びR9'は、双方とも水素、メチル又はエチルであり;
(iv)nが1であり、R7又はR8がメチル又はエチルであるならば、R9及びR9'は水素であり;
(v)nが1であり、R7及びR8が水素であるならば、R9及びR9'はメチル又はエチルであり;また
(vi)nが2であるならば、R9及びR9'は水素であり、R7及びR8の一方又は双方はメチル又はエチルである]
のアルキル置換されたヒドロカルビル部分であってもよい。
さらに、式(IV)のリンカーは、このようなリンカーを介してアリールピペラジン部分(さらに様々に置換することができる)に結合したTHI部分(さらに様々に置換することができる)を含む、ここで記載したもの以外の化合物と共に使用することもできる。このようなTHI部分及び他の化合物は、例えば米国特許第6770638号(その内容を参照としてここに取り込む)に記載されている。
本発明の好ましい化合物は、中枢神経系(CSN)への浸透が所望されている場合、生物学的利用能を高めるために、約1〜約4のlogPを有する。当業者であれば、このガイドラインを使用することで、本発明の化合物の生物学的利用能及びCNS浸透性を確定するために、特定のTHI部分に適したアリールピペラジン部分を選択することができる。例えば、特定の疎水性置換基を有する疎水性度の高いTHI部分を選択するならば、より親水性のアリールピペラジン部分を使用することができる。
ここに記載したテトラヒドロインドロン誘導体を合成するため、一般的に分子を完全にするアリールピペラジン部分に結合するリンカーを用い、テトラヒドロインドロン部分を置換する。この経路は:(1)ハロゲンで終結する脂肪族リンカーに結合した、適切に置換されたテトラヒドロインドロン部分を合成し、ついでアリールピペラジンとハロゲン中間生成物を反応させて、最終生成物を産出させ;又は(2)ハロゲンで終結する脂肪族リンカーに結合した、適切に置換されたアリールピペラジン部分を合成し、ついでテトラヒドロインドロンとハロゲン中間生成物を反応させて、最終生成物を産出させる工程のいずれかを含む。
本発明の例示的実施態様を合成するための次の代表的方法は、単に実施例とすることを意図したものである。医学及び/又は有機化学の当業者であれば、他の出発物質、中間生成物及び反応条件でも可能であることを理解するであろう。さらに、これらの化合物の種々の塩及びエステルも容易に作成され、これらの塩及びエステルが親化合物と同様又は等価な生物学的活性を有し得ることも理解するであろう。一般的に、これらの塩は、アニオン性対イオンとして、ハロゲン化物又は有機酸を有する。しかしながら、他のアニオンを使用することもでき、本発明の範疇に入ると考えられる。
1-{2-[4-(3,4-ジクロロフェニル)ピペラジン-1-イル]エチル}-1,5,6,7-テトラヒドロインドール-4-オンの合成
工程1:1-(2-クロロエチル)-4-(3,4-ジクロロフェニル)ピペラジンの調製
DMSO(5mL)に、(3,4-ジクロロフェニル)ピペラジン(500mg)とパウダー状の水酸化ナトリウム(87mg)が入った混合物を、2-ブロモ-1-クロロエタン(387mg)で処理し、周囲温度で16時間攪拌した。反応体を氷冷水(15mL)に注ぎ、0.5時間攪拌した。固形物が形成され、水をデカントすることにより分離させた。水相をジクロロメタン(5mL)で抽出した。固形物をジクロロメタン(5mL)で溶解させ、組み合わせた有機物を硫酸ナトリウムにて乾燥させ、濾過し、真空下で溶媒を除去した。フラッシュクロマトグラフィー(ジクロロメタン:メタノール 1:0〜10:1)により、表題の化合物として油が産出した(230mg)。
DMSO(2mL)に、1,5,6,7-テトラヒドロインドール-4-オン(107mg)が入った溶液に、パウダー状の水酸化ナトリウム(33mg)を添加し、混合物を周囲温度で0.5時間攪拌した。ついで、工程1の1-(2-クロロエチル)-4-(3,4-ジクロロフェニル)ピペラジン(220mg)を、溶液としてDMSO(2mL)に添加し、得られた混合物を周囲温度で24時間攪拌し、約60℃で約2時間加熱し、その後、薄層クロマトグラフィー(TLC)(酢酸エチル:ジクロロメタン 1:1)により、反応が完了したことが示された。反応物を氷冷水(15mL)に注ぎ、0.5時間攪拌した。固形物が形成され、水をデカントすることにより分離させた。水相をジクロロメタン(10mL)で抽出した。固形物をジクロロメタン(5mL)で溶解させ、組み合わせた有機物を硫酸ナトリウムにて乾燥させ、真空下で溶媒を除去したところ、表題の化合物として油が得られた(250mg)。
工程2の化合物(250mg)を、必要ならば加熱して酢酸エチル(5mL)に溶解させ、アセトン(0.5mL)にシュウ酸(57mg)が入った溶液を、攪拌しつつ添加した。直ちに沈殿物が形成され、混合物を室温で0.5時間攪拌した。真空濾過及び酢酸エチルを用いた洗浄により、乾燥時にオフホワイトのパウダーが付与された(220mg)。
同じ工程3の手順を、エチル及びプロピルリンカーにおいても使用する。
1-{4-[4-(3,4-ジクロロフェニル)ピペラジン-1-イル]ブチル}-1,5,6,7-テトラヒドロインドール-4-オンの合成
工程1:1-(4-クロロブチル)-1,5,6,7-テトラヒドロインドール-4-オンの合成
DMSO(100mL)に、1,5,6,7-テトラヒドロインドール-4-オン(10.0g)が入った溶液に、パウダー状の水酸化ナトリウム(3.26g)を添加し、混合物を周囲温度で0.25時間攪拌した。ついで、1-ブロモ-4-クロロブタン(9.38mL)を添加し、得られた混合物を周囲温度で7時間攪拌し、その後、TLC(酢酸エチル:ジクロロメタン 1:1)により、反応が完了したことが示された。反応物を氷冷水(250mL)に注ぎ、0.5時間攪拌した。油が分離し、分液漏斗で分離した。水相をジクロロメタン(50mL)で抽出した。油をジクロロメタン(25mL)に溶解させ、組み合わせた有機物を硫酸ナトリウムにて乾燥させ、濾過し、真空下で溶媒を除去した。フラッシュクロマトグラフィー(酢酸エチル:ヘキサン 1:1〜2:1)により、表題の化合物として油が産出した(6.0g)。
アセトニトリル(10mL)にヨウ化ナトリウム(438mg)と工程1の1-(4-クロロブチル)-1,5,6,7-テトラヒドロインドール-4-オン(600mg)の混合物が入ったものを、6時間還流して加熱した。ついで、(3,4-ジクロロフェニル)ピペラジン(581mg)と炭酸カリウム(367mg)を添加し、16時間還流し続けた。TLC(酢酸エチル:ジクロロメタン 1:1)により、反応が完了したことが示された。反応物を氷冷水(50mL)に注ぎ、0.5時間攪拌した。油が分離し、混合物から分離した。油をジクロロメタン(15mL)に溶解させ、水及び塩水で洗浄し、ついで硫酸ナトリウム上で乾燥させ、濾過し、真空下で溶媒を除去したところ、油として表題の化合物が産出した(970mg)。
シュウ酸塩の調製を、上述した同様の方法により行った。
同じ工程3手順を、全てのブチルリンカーにおいても使用する。
精神的病状
精神及び神経学的病状は、治療的有効量の本化合物及び/又は製薬的組成物を投与することにより治療することができる。これらの化合物は抗精神病化合物として使用され、精神疾患、例えばうつ病、不安神経症、特に心的外傷後ストレス症候群、統合失調症、統合失調性感情障害、双極性障害、性的不全、躁鬱、睡眠障害、拒食症、過食症、躁鬱病、強迫障害、妄想性分娩後うつ病、分娩後精神病、生理前症候群(pre-menstrual syndrome)、薬物乱用関連精神病、及びそれらの併発症を治療するために投与することができる。また、本化合物は、認知機能を高め、パーキンソン病、ハンチントン病、アルツハイマー病、加齢に関連した認知症、及びソマン及びサラン等の毒性化学剤への暴露等の、認知力低下を伴う神経再生疾患(neuroregenerative disorders)を治療するために使用することができる。
痛みは、これらの化合物及び/又は組成物を、それを必要とする患者に有効量投与することにより、特にこれらの組成物を鎮痛用量投与することにより、本発明の化合物及び製薬用組成物で、効果的に治療することができる。本化合物で治療することができる様々な種類の痛みは、急性の痛み、慢性の痛み、侵害受容性疼痛(すなわち、無傷の神経終末を介した痛覚伝達に関連した痛み)、及び神経障害性の痛み(神経系機能不全に起因し、異痛、痛覚過敏、錯感覚及び異常錯感覚に関連した、火傷感(burning)、撃たれたような痛み(shooting)及びヒリヒリする痛み(tingling pain)により特徴付けられる)である。急性の痛みを伴う病状には、頭痛、関節炎、単純筋挫傷(simple muscle strain)及び月経困難症が含まれる。傷害受容性疼痛には、例えば術後疼痛(post-operative pain)、群発性頭痛、歯痛、手術後疼痛(surgical pain)、火傷による痛み、分娩後の痛み、アンギナ痛、尿生殖路痛、膀胱炎、関節炎に関連した痛み、AIDS、慢性の背痛、内臓痛、胃食道逆流、消化性潰瘍、感染性胃炎、炎症性大腸炎、片頭痛、緊張性頭痛、線維筋痛、神経根圧迫、例えば坐骨神経痛、三叉神経痛、中枢性疼痛、骨損傷痛、労働及び配達中の痛み、筋挫傷、アルコール依存症、疱疹性神経痛、幻肢痛、及び月経困難症痛が含まれる。病状に関連した神経障害性の痛みには、慢性の腰の痛み、関節炎に関連した痛み、癌に関連した痛み、疱疹性神経痛、幻肢痛、中枢性疼痛、オピオイド耐性神経障害痛(opioid resistant neuropathic pain)、骨損傷痛、及び労働及び配達中の痛みが含まれる。疼痛誘発性精神疾患、例えば不安神経症、うつ病及び/又は激しい情緒変化、並びに痛みに関連した嘔吐反応からの救済、及びその治療も、本化合物及び組成物により行うことができる。
本出願の化合物及び製薬用組成物は、それを必要とする患者に有効量のそれらの化合物及び/又は組成物を投与することにより、動作誘発性-及び毒素誘発性嘔吐を双方を緩和するのにも有用である。乗り物酔い、並びに化学療法剤、例えばシスプラチン、ダカルバジン、シクロホスファミド、5-フルオロウラシル、ドキソルビシン及びパクリタキソール、又は毒剤、例えばソマン又はサリンの投与に関連した嘔吐を治療することができる。本化合物は、5-HT1Aレセプターアゴニスト活性を有しているため、乗り物酔い及び化学誘発性の吐き気の双方に有用であると考えられている。多様な刺激に起因する嘔吐をブロックするのに有効な方法又は薬剤は、現在までに知られていない。例えば、乗り物酔いをブロックするのに有効であることが知られている薬剤は、化学的に誘発される嘔吐に対して有効であるとはされていない。
よって、本発明は、嘔吐を治療するための、製薬用組成物における本化合物の使用を含む。さらに、本発明は、嘔吐を治療する薬品を製造するための、これらの化合物の使用を含む。
本発明の化合物及び/又は製薬用組成物のさらなる用途は、中枢及び末梢神経系における神経新生、ニューロン再生(neuronal regeneration)又は軸索樹状突起間の複雑性を刺激することにある。これは、本発明の化合物を有効量、それを必要とする被験者に投与する工程を介して達成される。このような神経再生効果は、本化合物の5-HT1Aレセプターアゴニスト活性の結果であると考えられる。治療可能な神経変性の病状は遺伝的、自然発生的又は医原性であり、制限されるものではないが、脳卒中、脊髄損傷筋萎縮性側索硬化症、周生期低酸素症、眼の損傷及び網膜症、視神経変性、難聴、下肢静止不能症候群、湾岸戦争症候群及びトゥレット症候群が含まれる。現在、神経変性病ALSの治療に使用されている薬剤療法の例は、5-HT1Aアゴニストキサリプロデン(xaliproden)である。
よって本発明は、神経変性又は損傷を治療するための、製薬用組成物における本化合物の使用を含む。さらに、本発明は、神経変性又は損傷を治療する薬品を製造するための、これらの化合物の使用を含む。
本発明のさらなる態様は、本発明の化合物及び製薬用組成物の有効量を被験者に投与する工程を含む、被験者の中枢又は末梢神経系における神経保護/神経再生に関連したメカニズムに係る神経機能を刺激する方法にある。このシリーズにおける化合物は、化学オルガノ-リン神経ガスへの暴露に対し、神経保護するのに有用である。これらの化合物は、生物防御における化学品暴露の結果として、多様な毒性治療に相乗作用を有することが予期される、多様な脳内物質を標的としていると考えられる。
よって、本発明は神経保護のための、製薬用組成物における本化合物の使用を含む。さらに、本発明は、神経保護をする薬品を製造するための、これらの化合物の使用を含む。
関与する個々の被験者に特定の必要性に応じて、本発明の化合物は、本発明の教示に基づき、治療的に有効な濃度で提供される種々の用量で投与することができる。選択される化合物の活性度、被験者の生理学的特徴、被験者の病気又は病状の程度又は種類、及び投与方法等の要因により、選択される化合物の有効量が決定されるであろう。特定の被験者の治療に最適な用量を決定するため、一般的に、最初の用量は変更される。本化合物は、経口投与、局所投与、経皮投与、腹腔内注射、又は血流への直接的な静脈注射を含む、多様な経路を使用することにより投与することができる。また有効量の化合物は、所望するならば、脳への直接的な注入又は脳脊髄液への注入を介して投与することもできる。
ヒト被験者における、本発明の特定の化合物と製薬用組成物の治療効果及び適切な用量を測定するために、動物モデルを使用することができる。例示的な動物モデルを以下に説明する。
本出願の化合物の以下の代表的なセットを、種々の動物モデルでテストした:

1-{4-[4-(3,4-ジメチルフェニル)ピペラジン-1-イル]ブチル}-1,5,6,7-テトラヒドロインドール-4-オン
テール-フリックは、種々の化合物への暴露後の、鎮痛レベルを定める又はモニターするために使用される(D'Amour及びSmith, 1941;Harris及びPierson, 1964)。尾部に光線を集束させ、テール-フリックへの潜時を評価することにより、マウス、ラット又はサルをテストするために使用することができる。このテストは、弱い又は強い鎮痛性をスクリーニングするのに有用であることが証明されている(Deweyら, 1969)。上述にて列挙された代表的な化合物は、尾部における集束光線により誘発される激しい痛みの軽減に効果的であった。鎮痛効果は、テストされた4つの化合物の3つで測定された。
ヒト鎮痛活性と良好な相関関係にある、異なるクラスの鎮痛化合物の鎮痛活性を検出及び比較するために許容されているスタンダードは、マウスにおける、ライジング誘発性の酢酸を防止することである。R. Kosterら, 鎮痛スクリーニングのための酢酸(Acetic acid for analgesic screening)Fed. Proc. 18:412(1959)。
種々の用量の本化合物、及び他の鎮痛剤又はビヒクルと組み合わせたものを、指定された観察期間の5分前に、標準的な挑戦用量の酢酸と共に腹腔内に注射する。酢酸は0.55%溶液として調製され、体重当たり0.1ml/10グラムの容量で注射する。スコアリング目的のため、「ライジング」は、酢酸の投与後、約5分で始まる観察期間中、全身にわたる腹部の伸長又は収縮により示される。上述にて列挙された代表的な化合物は、酢酸より誘発される弱い痛みの軽減に効果的であった。
熱痛に対する薬剤誘発性鎮痛テストとして、ホットプレートを使用する。このテストの実施において、マウス又はラットを加熱したプレート上に置く。動物が、加熱したプレートに対して疼痛反応を示すまでの潜時を測定する。疼痛反応には、後足をなめることに関連してプレートから後足を引っ込めること、又は他の侵害防御機構行為が含まれる。薬剤誘発性の遅発性潜時は、鎮静反応の指標となる。上述にて列挙された代表的な化合物は、ホットプレートテストにおける疼痛反応に対する潜時を増加させるのに効果的であった。
ラット又はマウスの後足の腹側、又は足底、表面に、ホルマリン溶液を皮下注射することにより、処置された足に、急性疼痛反応を誘発させることができる。反応には、後足をなめることに関連して、床から後足を引っ込めることが含まれる。動物が床から後足を引っ込めたとされる時間、及び動物が向きを変えて後足をなめた回数が、疼痛の指標となる。鎮痛化合物により、引っ込めた時間及び足をなめた回数が低減した。このシリーズの化合物は、足底ホルマリン注射により誘発される行為を低減させるのに効果的であった。
オス又はメスのジャコウネズミ(S. murinus)(30〜80g)を、0700〜1730時間、ライトを用いた人工照明下、24±1℃に温度制御された室内で保持する。人工湿度を50±5%に維持する。動物は自由に水及びペレット状の猫のえさにアクセス可能である(例えば、Feline Diet 5003, PMI(登録商標) Feeds, St. Louis, USA)。
実験の日、嘔吐行為を評価するため、清浄な観察用チャンバー(約21×14×13cm)に動物を移した。化合物又はそれらそれぞれのビヒクルを皮下注射する前に、30分適応させる。化学治療嘔吐剤を静脈内投与し、続いてテスト用化合物を投与する。ついで、動物を60分観察する。嘔吐の発現は、胃腸管から固体状又は液状の物質を経口排出する(すなわち嘔吐)ことを伴う、又は物質の通過(すなわち吐気動作)を伴わない、周期的な腹部収縮により特徴付けられる。吐気及び/又は嘔吐の発現は、観察チャンバー内において、動物がその位置を変えた場合、又は吐気及び/又は嘔吐の間隔が2秒を超えた場合に、分類されると考えられる。このシリーズの化合物は、化学治療誘発性嘔吐をブロックするのに効果的である。
運動暴露による嘔吐をテストするため、少なくとも5分の順応期間後に、動物を往復式振揺器(例えば、Taitec, Double Shaker R-30, Taiyo Scientific Industrial Co Ltd.)上の透明なケージに入れる。テスト前の所定の時点で化合物を投与する。10分間、1Hzの頻度の4cm(左側に2cm、右側に2cm)移動する水平方向の動きに、動物をかける。10分暴露を使用すると、誤った負の結果を得るおそれが低減する。遅延型反応が生じるケースにおいては、運動暴露の終了後少なくとも5分間、観察し続けるが、以前の研究では、運動の停止後に嘔吐が発現することは、非常にまれであると示されている。このシリーズの化合物は、動作誘発性嘔吐をブロックするのに効果的である。
非競合NMDAレセプターアンタゴニストフェンシクリジン(PCP)は、齧歯動物の音響驚愕反応のプレパルス抑制(PPI)を低減する。プレパルス抑制におけるPCP-誘発性欠損を改善する化合物は、統合失調症の治療にも有用である。このテストにおいて、オスのC-57マウスを、グループ毎に8匹の動物を有する5つの投薬グループに割り当て、ビヒクル又はPCP(5mg/kg)の腹腔内(IP)投与の20分前に、ビヒクル又はテスト用化合物を経口(PO)又は皮下(SC)投与する。PCP投与の10分後、マウスをハミルトン-キンダー驚愕チャンバーに置き、評価する。バックグラウンド白色雑音(65db)を用いた5分間の順応期間後、マウスを5つの異なるタイプの試験に暴露する。試験は外見上ランダムな順序で、試験間の間隔をランダムに5〜25秒とし、10回の調査を行う。試験は次の通りである:刺激のみの試験(120dbの白色雑音、50msの刺激);2つの異なるプレパルス+パルスの試験、ここでは120dbパルスに120ms先行した65dbバックグラウンドを超えた20ms 5db、又は10dbの刺激;120dbパルスなしの10dbプレパルス;及び刺激なしの試験、ここではバックグラウンドノイズのみが付与される。PCPにより生じるプレパルス抑制の崩壊逆転は、抗精神病活性を有する化合物の臨床的前兆である。
条件回避反応(CAR、能動的回避)モデルは、抗精神病活性をテストするものである。逃避崩壊(錐体外路運動機能)のない回避崩壊(潜時の増加)は、抗精神病活性を有する化合物の臨床的前兆である。動物(マウス)の訓練は、試験間の間隔が可変である20の試験からなる(80%回避基準まで訓練)。約1分の順応期間後、照明及び90dbトーンの音響を付与する。5秒以内の反応(暗い区画へ横断)で試験は終了し、試験を回避反応(CAR)として記録する。マウスが5秒以内に反応しないならば、足にショック(0.8mA)を与え、ショック中の反応(暗いチャンバーに移動)を、逃避反応として記録する。ショックを回避するために、動物は、合図が出たらチャンバーの明るい側から暗い側に移動する(回避)、又はショックが与えられれば移動する(逃避)ことを学ぶ。テストセッションの20分前に、ビヒクル又はテスト用化合物を皮下投与する。これは、認知及び注意の崩壊をチェックするものである。
また、統合失調症及び/又は他の精神病の他の臨床的に許容可能なモデルも、精神病の治療に使用される特定の化合物の効果又は用量を測定するために使用することができ、次のもの:アンフェタミン-、コカイン-、PCP-、又はMK-801-誘発性高歩行運動(Hyperlocomotion);カタレプシー;及びMK-801-誘発性歩行運動及び落下が含まれる。
ハミルトン-キンダー驚愕チャンバーを、セッションのコンディショニング、及び作製、及び驚愕反応の記録に使用する。伝統的なコンディショニング手順が使用されて、驚愕反応の相乗作用が生じる。簡単に言えば、最初の2日、ラットを、ショックグリッドが取り付けられている暗い驚愕チャンバーにおいた。5分の順応期間後、各ラットは、光(15ワット)の付与5秒前に1mAの電気ショック(500ms)を受け、該光は、ショックの持続期間中維持されている。各コンディショニングセッションにおいて、光とショックを10回付与し、ラットにテスト用化合物の水溶液を強制補給し、驚愕テストセッションを実施した。刺激に対する慣れの当初の高速段階の影響を最小にするために、セッションの開始時に、10回の連続した音響驚愕刺激(110dB、光と対になってはいない)のブロックを付与した。これに、ノイズ単独、又は光の前にノイズといった、20回の交互試験が続いた。当初の試験ブロックを除き、各試験タイプ(ノイズ単独対光+ノイズ)の驚愕反応振幅を、全テストセッションにおける各ラットについて平均化した。ノイズ単独及び光+ノイズの間の差異として、データが付与された。相乗驚愕を低減又はブロックする化合物は、抗不安活性を有すると考えられる。
ハミルトン-キンダー高架式十字迷路は、Heltonらの設計に基づいており、本来は、Lister(1987)によりマウス用として認定されていた。迷路は2つのオープンアーム(例えば、30×5×0.25cm)と2つのエンクローズドアーム(30×5×15cm)を有し、プレキシグラスから作製することができる。各迷路アームの床はコルゲート状になっており、所定のテクスチャーが付与される。アームは中心プラットフォームから伸長し、互いに90度の角度になっている。迷路を床の上45cmの高さに上げ、赤光を照射する。個々の赤外線フォトセルを迷路の各アームに沿って取り付け、クローズド、オーブン、又はノーズポーク活性をモニターする。マウスを中心プラットホームに個々に置き、クローズドアーム、オープンアーム、及びノーズポーク(迷路のクローズドアームからオープンアーム内のみに突き出した頭部)の数の計測を記録し、5分のテスト期間中、迷路の様々な区画で過ごした時間及びアーム進入の測定として使用する。本化合物の投与により、抗不安活性を表すオープンアーム活性を増加させることができる。
不安神経症の他の臨床的に許容可能なモデルも、不安神経症の治療に使用される、特定の化合物の効果又は用量を測定するために使用することができ、光/暗闇調査(Light/Dark Exploration)及び母系分離発声テスト(Maternal Separation Vocalization Tests)を含む。
尾懸垂テストは、不動性を尾部で動物を懸垂することにより誘発するもので、「行動絶望」強制水泳テストの変形である(Steruら, 1985)。チャンバーは、天井の中心にフック(4cm)が取り付けられた、17cmW×25cmH×15cmDのものであった。マウスの行動を観察可能なように、前壁はない。尾部の先端から15mmに配した粘着テープにより、マウスをフックに吊した。確実に全不動性の観察がなされるように、動物を、その胃が治験者の方に向くように位置させた。動物が受け身で吊され、少なくとも2秒静止している時間の合計として、不動性をスコア化した。観察の全期間は6分とした。上述にて列挙した代表的な化合物の投与は、テストにおける不動性の低減に効果的であった。
不安神経症の他の臨床的に許容可能なモデルも、不安神経症の治療に使用される、特定の化合物の効果及び用量を測定するために使用することができ、強制水泳テスト及びDRLテストが含まれる。
音響驚愕は感覚運動反応性のためのテストであり、薬剤誘発性副作用の可能性をプロファイルするのに使用することもできる。音響驚愕は、120dBの刺激に応じて、プレートに変換された最大力(N)として測定される。ハミルトン-キンダー驚愕チャンバーを使用し、驚愕刺激を付与し、反応性を測定して記録した。20回の連続した音響刺激のブロックを動物に付与し、反応性を平均化した。反応において薬剤誘発性が増加すると、その用量に対して、不安惹起の可能性といった有害事象のおそれがあることを示す。反応において薬剤誘発性が低下すると、その用量に対して、鎮静作用等の有害事象のおそれがあることを示す。上述にて列挙した代表的な化合物は、テストされた有効用量での音響驚愕反応において、有意な増加又は低減を誘発しない。有効用量よりも高いと、このシリーズの化合物は、音響驚愕における用量依存性の低減を誘発する。
当該技術で公知の神経学的病状の他の臨床的に許容可能なモデルも、本出願の特定の化合物及び製薬用組成物の治療効果及び適切な用量を測定するために使用することができる。これらには、とりわけ、次のテストが含まれる。
学習記憶モデル:不安相乗驚愕の獲得;受動的回避、シャトル又はステップダウン;能動的回避の獲得;モリス水迷路;及び健忘性逆転。
注意欠陥モデル:ADHD若年動物モデル(PPI及び活性);ADD加齢動物モデル;及び能動的回避モデルの獲得。
神経保護(Neuroprotectuon)/てんかん:フェンチレンテトラゾール-、ストリキニーネ-、ビキュキュリン(Bicuculine)-、ピクロトキシン-誘発性けいれん、サリン及びソマン及びMES/ECS。
パーキンソン病:アポモルフィン-、アンフェタミン-誘発性回転(6-OHDA);及びレセルピン-誘発性低体温/低歩行運動(Hypolocomotion)。
薬剤依存及び禁断:ニコチン−、ベンゾジアゼピン-、及びエタノール-誘発性禁断。
本発明の他の側面は:(1)上述した本発明の化合物(その塩及びエステルを含む)を有効量;及び(2)製薬的に許容可能な賦形剤を含有する製薬用組成物にある。
担体を含む製薬的に許容可能な賦形剤は、当該技術で一般的に知られているものから選択することができ、制限されるものではないが、投与経路に応じて、不活性な固体状希釈剤、水溶液、又は無毒の有機溶媒が含まれる。所望するならば、これらの製薬用組成物は、防腐剤及び安定剤等、及び制限されるものではないが、湿潤剤又は乳化剤、pH緩衝剤、ヒト血清アルブミン、酸化防止剤、防腐剤、静菌剤、デキストロース、スクロース、トレハロース、マルトース、レシチン、グリシン、ソルビン酸、プロピレングリコール、ポリエチレングリコール、硫酸プロタミン、塩化ナトリウム、又は塩化カリウム、鉱物性油、植物性油、及びそれらを組み合わせたものからなる群から選択される製薬的に許容可能な賦形剤等の物質を、さらに含有可能である。当業者であれば、他の担体も使用可能であることを、高く評価するであろう。

1-{4-[4-(3,4-ジクロロフェニル)ピペラジン-1-イル]ブチル}-1,5,6,7-テトラヒドロインドール-4-オン;
1-{4-[4-(4-フルオロフェニル)ピペラジン-1-イル]ブチル}-1,5,6,7-テトラヒドロインドール-4-オン;
1-{2-[4-(3,4-ジフルオロフェニル)ピペラジン-1-イル]エチル}-1,5,6,7-テトラヒドロインドール-4-オン;及び
1-{4-[4-(3,4-ジメチルフェニル)ピペラジン-1-イル]ブチル}-1,5,6,7-テトラヒドロインドール-4-オン
から選択される、請求項1に記載の化合物又は製薬的に許容可能な塩。 - 製薬的に許容可能な賦形剤と、請求項1に記載の化合物又は製薬的に許容可能な塩とを含有する製薬用組成物。
- 請求項1に記載の化合物又は製薬的に許容可能な塩の治療量を含む、痛み又はうつ病の治療のための医薬。
- 治療量が、静脈内注入、経口、局所、腹腔内、膀胱内、経皮、鼻腔内、直腸、膣、筋肉内、皮内、皮下、及び髄腔内経路からなる群から選択される投与経路により投与される、請求項4に記載の医薬。
- 治療量が、0.0001mg/kg〜60mg/kgの範囲である、請求項4に記載の医薬。
- 治療される病状がうつ病である、請求項4に記載の医薬。
- 治療される病状が痛みである、請求項4に記載の医薬。
- 痛み又はうつ病を治療する薬品の製造のための、請求項1に記載の化合物又は製薬的に許容可能な塩の使用。
