発生過程にある大脳皮質においては、VZ(ventral zone)領域の神経幹細胞は、まず等分裂した後に神経幹細胞と神経前記細胞に不等分裂をし、神経幹細胞の維持に働く。インテグリンβ1と相互作用する細胞内タンパク質であるILK(integrin-linked kinase)は転写抑制因子Snail/Slugの発現を亢進することにより、Eカドヘリンとβカテニンの転写の発現を抑制することが知られているが、Eカドヘリンとβカテニンを高発現しているVZ領域の細胞が移動できるようになるのは、このメカニズムを利用していることが考えられる(Campos L.S., Bioessays 27, 698-707, 2005)。一方、FGF2の存在下でβカテニンを過剰発現させると、神経前駆細胞を増殖させ続けること、またFGF2の非存在下でβカテニンを過剰発現させると、神経前駆細胞を神経分化の方向に向かわせることが知られているが、神経前駆細胞にFGFを投与すると、インテグリンβ1の発現が亢進することから、βカテニンの効果にFGF2が関与するのは、部分的に、細胞間、あるいは細胞−ECM間の微小環境によると考えられる。これらの結果から、VZにおいてタンパク質相互作用のネットワークが機能し、インテグリンβ1シグナル伝達系は他のシグナル伝達系とのクロストークを通じて、神経幹細胞を維持するか、それとも分化したり移動したりするか、の選択を行なうと考えられている(Campos L.S., Bioessays 27, 698-707, 2005)。
VZ領域の神経幹細胞の等分裂と不等分裂は、特定の分子を不等配分することにより、細胞の生存を調節することにより調節しうる。発生過程では、基底膜がPCD(programmed cell death)を調節していることが知られており、基底膜の近くや他の生存因子(例えば、増殖因子)が存在している環境にある細胞だけが生存できるため、適当なマトリックスとの接触を失った細胞では、細胞死が引き起こされる。実際、インテグリンシグナル伝達系がなくなると、PCDが誘導されたり(Zhang Z., Proc. Natl. Acad. Sci. USA 92, 6161-6165, 1995; Gilmore A.P. et al., J. Cell Biol. 149, 431-446, 2000; Gary D.S. et al., J. Neurochem 84, 878-890, 2003)、インテグリンβ1の構造変化がアポトーシスを引き起こしたりする(Prince J.M. et al., Dev. Dyn. 223, 497-516, 2002)。また、フリーのインテグリンは、カスパーゼ−8を膜に誘導して活性化し、インテグリンの結合はインテグリン−カスパーゼの相互作用を阻害し、細胞の生存を促進する(Stupack D.G. et al., J. Cell Biol. 155, 459-470, 2004)。さらに、α3β1インテグリンが、MAPKシグナル伝達系を活性化することにより、細胞の生存を促進することが知られている(Manohar A. et al., J. Cell Sci. 117, 4043-4054, 2004)。これらの結果から、in vivoでも、神経幹細胞または神経前駆細胞の生存は、インテグリンβ1を高発現しているVZにおいて、インテグリンβ1によって維持されていると考えられている(Campos L.S., Bioessays 27, 698-707, 2005)。
最近、神経前駆細胞においてインテグリンβ1の発現レベルを下げると、ネスチン陽性細胞が減少することが明らかになった(Leone D. et al. J. Cell Sci. 118, 2005)。ネスチンは、神経幹細胞あるいは神経前駆細胞の未分化状態のマーカーであり、この実験結果は、インテグリンβ1が神経幹細胞及び神経前駆細胞の未分化状態の維持に必要であることを示す。
神経幹細胞に対するガレクチン−1の結合に糖が関与しているかどうかを調べるため、ビオチン化C2Sガレクチン−1(C2S-GAL-1-bio)(産総研より提供)を用いて、ラクトースの存在下で組織染色を行った。このC2S-GAL-1は、ガレクチン−1の2番目のシステインをセリンに置換した変異体(Hirabayashi & Kasai, J Biol Chem 266, 23648-23653 1991)で、正常な炭化水素結合能を有することが既に示されている(Purkrabkova et al. Biol Cell 95, 535-545, 2003)。
SVZにおいては、SVZアストロサイトの一部は神経幹細胞として機能し、中間分化段階のTA細胞(transit amplifying cells)を経て、さらに細胞増殖の後、NB(神経芽細胞)へと分化することが知られている。この神経幹細胞は、比較的ゆっくり増殖することが知られているため、長期的にBrdUを取り込ませることにより、神経幹細胞を同定することができる。
これらの結果から、神経幹細胞または神経前駆細胞において、インテグリンβ1はガレクチン−1の受容体であり、ガレクチン−1がインテグリンβ1に結合することにより、インテグリンシグナル伝達系を活性化することが明らかになった。
Claims (4)
- 神経幹細胞または神経前駆細胞におけるインテグリンβ1を検出するための検出剤であって、
ガレクチン−1を含有することを特徴とする検出剤。 - 前記ガレクチン−1が、C2Sガレクチン−1であることを特徴とする請求項1に記載の検出剤。
- 神経幹細胞または神経前駆細胞におけるインテグリンβ1(ヒト個体内のインテグリンβ1を除く)を検出するための検出方法であって、
ガレクチン−1を前記インテグリンβ1に結合させる工程と、
前記インテグリンβ1に結合した前記ガレクチン−1を検出する工程と、
を含む検出方法。 - 前記ガレクチン−1が、C2Sガレクチン−1であることを特徴とする請求項3に記載の検出方法。
