Description
実施例1においては、様々なpHで水素発酵を行った。本微生物としては、PEH9株を用いた。また、比較の対照として、サーモアナエロバクテリウム・サーモサッカロリチカムであってPEH9株とは異なる菌株であるPEH8株とATCC7956株とを用いた。培地としては、1.0%(w/v)のキシロース、0.4%(w/v)の酵母エキス(極東製薬)、0.25%(w/v)のMOPS、0.091%(w/v)のK2HPO4、0.03%(w/v)のNaH2PO4、0.02%(w/v)のMgCl2、0.01%(w/v)のCaCl2、0.03%(w/v)の(NH4)2SO4、0.002%(w/v)のFeSO4、0.04%(w/v)のL−システイン、0.11%(w/v)のレザズリンを含む水溶液を用いた。
実施例2においては、pH5.0〜5.2の範囲における水素の生産性を1Lスケールで確認した。本微生物としては、PEH9株を用いた。培地としては、1.0%(w/v)のキシロース、2%(w/v)のペプトン、0.5%(w/v)の酵母エキス、1.0%(w/v)の麦芽エキス、0.3%(w/v)のK2HPO4、0.1%(w/v)のMgSO4、0.02%(w/v)のL−システイン、0.2%(w/v)のL−リンゴ酸、0.003%(w/v)のオレイン酸、0.1%(w/v)のTween80を含む水溶液を用いた。そして、容積が1Lの発酵槽(BMJ−01、エイブル株式会社製)中で500mLの培地にPEH9株を接種し、嫌気条件下、希釈率0.3〜0.4にて連続培養を実施した。
実施例3においては、pH5.8〜6.0の範囲における水素の生産性を1Lスケールで確認した。pHの範囲が異なる点を除き、上述の実施例2と同様の条件で水素発酵を行った。
実施例4においては、様々な温度で水素発酵を行った。本微生物としては、PEH9株を用いた。培養液としては、0.5%(w/v)のキシロース、1.6%(w/v)のトリプトン、1.0%(w/v)の酵母エキス、0.4%(w/v)の塩化ナトリウムを含む水溶液を用いた。
実施例5においては、本微生物の溶解性について確認した。本微生物としては、PEH9株を用いた。また、対照として、排水処理に使用されているメタン菌(メタン発酵微生物群)を用いた。培養液としては、1Lの精製水に、10.00gのキシロース、4.00gの酵母エキス、2.50gのMOPS、0.91gのK2HPO4、0.30gのNaH2PO4、0.20gのMgCl2・6H20、0.10gのCaCl2・2H20、0.30gの(NH4)2SO4、0.02gのFeSO4・7H2O、0.40gのL−システイン、1.10mgのレザズリンを溶解したGBG培地を用いた。
実施例6においては、様々な糖類を単一の糖源として添加して水素発酵を行った。本微生物としては、PEH9株を用いた。培地としては、0.5%(w/v)のキシロース、1.6%(w/v)のトリプトン、1.0%(w/v)の酵母エキス、0.4%(w/v)の塩化ナトリウムを含み、グルコース、キシロース、アラビノース、マルトース、スクロース、セロビオース、可溶性デンプンのうちいずれか1種類を2%(w/v)添加した7種類の水溶液を用いた。
Claims (5)
- 受託番号FERM BP−10793のサーモアナエロバクテリウム・サーモサッカロリチカム菌株であることを特徴とする微生物。
- 請求項1に記載の微生物の変異株であって、pHが4.5以上、6.5以下の範囲で培養した場合に、糖1gから270mL以上の水素を生産することを特徴とする微生物。
- pHが5.0以上、6.0以下の範囲で培養した場合に、糖1gから270mL以上の水素を生産することを特徴とする請求項2に記載の微生物。
- 請求項1に記載の微生物の変異株であって、糖源としてキシロースを用いて、pHが4.5以上、6.5以下の範囲で培養した場合に、キシロースに対して2.1以上のモル比で水素を生産することを特徴とする微生物。
- 糖源としてキシロースを用いて、pHが4.8以上、5.5以下の範囲で培養した場合に、キシロースに対して2.1以上のモル比で水素を生産することを特徴とする請求項4に記載の微生物。
