Description
プリオン病または伝達性海綿状脳症(TSE)は、これらが感染物質の性質についての従来の考えに異議を唱えるものであるのみならず、食物と血液の安全性に対しもたらす潜在的に重大な公衆衛生上の脅威ともなることから、科学界の関心をとらえてきた。さらに、近年、新しい変異型疾病であるvCJDの流行により、動物とヒトとの間における病気感染の可能性が劇的に強調されるようになってきた。本発明は、動物およびヒトにおける伝達性海綿状脳症{Transmissible Spongiform Encephalopathies(TSE)}疾患の診断に対する合成ペプチドの使用に関するものである。
伝達性海綿状脳症(TSE)には、ヒトと動物との両方に感染する、一群の、急速に進行する、致命的な神経変性疾患が含まれる。TSEは、壊滅的な痴呆、筋間代性痙攣を伴う錐体路徴候および錐体外路系徴候、多病巣性の海綿状変化、アストログリオーシス、アミロイドプラーク、ニューロンの消失および炎症反応の不在等の臨床的および神経病理学的特徴を有し、通常、長い潜伏期間によって特徴づけられる。
プルジナーの「タンパク質のみ」仮説によると、プリオン病を引き起こす感染性の病原体はタンパク質である。プリオンの主要成分は、PrPSc(疾病に特有の「スクレーピー」イソ型)と呼ばれる折返しの異常なPrPであるようである。PrPScは、その正常な対応物(PrPC)から、立体構造の変化により発生するものと考えられている(Cohen,1998年)。この変換のメカニズムに関して曖昧さがまだ残っているが、PrPScの存在下、基質として働く正常なPrPCが、立体構造の変化を受け、自己触媒的プロセスによりPrPScになり、その結果、PrPSc凝集体とアミロイドロッドとを形成し、これにより細胞死を引き起こすことが広く受け入れられている(Hope,1986年、Horwich,1997年)。PrPCからPrPScへの構造変化は、タンパク質のβ−シート構造の量の著しい増大と、おそらく、円二色法と赤外分光法によって示されるα−らせん量の僅かな減少が関与する、決定的な立体構造変化によって裏付けられることがほとんどである。(Pan,1993年,Caughey,1991年)。PrPCからPrPScへの変換の効率は、2つのPrP配座異性体間における配列相同性の程度に依存するようである。このユニークなメカニズムのため、プリオン病は、同一の種の間で容易に伝達でき、PrPの配列相同性が十分である場合には、種から種に伝達することができる(Raymond,2000年)。したがって、提唱されたプリオン病の伝達機構が正しければ、動物とヒトとの間におけるこれらの脳障害の伝達によって、伝染病が引き起こされる可能性がある。
プリオン病は感染性を有する。動物のプリオン病の伝達率は、ヒツジからヒツジへのスクレーピー(Cuille,1936年)およびヤギへの種を超えたスクレーピー(Pattison,1957年)ならびにヒトからチンパンジーへのクルおよびCJD(Gajdusek,1966年、Gibbs,1968年)等の、感染した動物から健康な動物への脳ホモジネートの接種によって実験的に確立されてから久しい。スクレーピーのマウスへの感染の成功は、重要なブレークスルーであった(Chandler,1961年)。これにより、実験モデルが提供され、TSE研究が大いに促進された。ウシにおける最近のBSEおよびヒトの新しい変異型CJD(vCJD)の原因は、BSEの場合には、動物への飼料として与えられたスクレーピーヒツジの死体の混合物を食べた結果であり(Brown,1997年)、vCJDの場合は、BSEに感染したウシからの牛肉を食べた結果である(Bruce,1997年)と考えられた。vCJDとBSEとの間の関係は、BSE順応性を有するマカク(ヒトに最も近いモデル)およびBSEおよびvCJDを引き起こす物質を接種した同系交配のマウスの研究から得られた神経病理的証拠によりさらに支持されている(Lasmezas,1996年)。北米におけるミュールジカおよびヘラジカへのCWDの流行への具体的懸念としては、CWDが、BSEのように、狩猟を通してこの病気に曝露されまたは、感染した肉を取り扱ったり食べたりする可能性のあるヒトに感染され得るかどうかということである。これまで、人食いの風習による死者の脳組織の摂取によるクルの流行ならびに、ホルモン、組織移植および汚染された医療器具の使用によるCJDの医原性の感染等の、ヒトにおける悲劇的な偶然によるプリオン病の感染が報告されてきている。
プリオン検出の感度と信頼性の高い検査法の発展は、疾病の調査、リスクアセスメントならびに、将来の治療法と組み合わされた時には疾病の防止およびに根絶の絶対的必須条件である。現在、プリオン病の診断には、3つの基本的な評価フォーマットがある。(1)動物の伝染力のバイオアッセイ:これは、齧歯類の実験的なスクレーピーにおける、伝染性プリオンの測定に関する最も感度の高い方法であり、通常、患畜の脳ホモジネートのレシピアント動物への脳内注射によって達成される。しかしながら、種々異なる動物種におけるプリオン病の伝染力の定量的測定には、「種バリア」による限界があり、病理学、潜伏期間および、PrPScの分子的特徴が異なる、はっきりと区別できるプリオン「株」が存在する。したがって、この時間のかかる高価な死後診断法は、大部分は、齧歯類で種々異なるプリオン株を識別するための研究道具として使われており、伝染性の脳物質の較正のための参照としての役割を果たしている。(2)現在のPrP免疫学的検定法:これらは、全てのプリオン病中唯一知られた品質保証付き分子であるプロテアーゼ耐性PrPScの検出に基づく方法である。免疫化学的方法(免疫組織化学およびウェスタンブロッティング法)によるPrPScの検出は、長年、動物およびヒトにおけるプリオン病の最も正確な診断を提供してきた(Schaller,1999年、Biffiger,2002年)。これらは、死後診断のために広く使用されている。この目的のために、PrPの種々の領域に対して、米国特許第4806627号明細書および欧州特許第0861900号明細書に記載され、広く使用されている3F4、6H4等多くのモノクローナル抗体およびポリクローナル抗体が育てられてきた。しかしながら、PrPC(しばしば、ずっと大量に存在する)とPrPScとを区別することができるとされる方法は非常に少なかった。1997年にコース(Korth)によって、そして、2003年にパラミチオチス(Paramithiotis)によって報告されたモノクローナル抗体は、両方共IgMであり、これらの抗体による診断評価法は開発されなかった。この結果、現在の免疫化学的方法は、ほとんどすべて、PrPScの検出の前に、通常、プロテイナーゼK(PK)消化等の非特異性のプロテオリシスによって、PrPCを減少させるか除去するステップが必要である。このような前処理によって、PrPScの最初の60〜70残基から、PrPresと呼ばれる、PK耐性を有するPrP27kDa−30kDaのコアが生じる。ついで、N−末端を切断したPrPScすなわちPrPres(病理学的サンプルだけに存在する)を検出するのに、このタンパク質の残りのC末端領域を認識する抗PrP抗体を使用することができる。組織切片には、免疫組織化学的染色のために、通常、PrPC関連のバックグラウンドを減少するための酸加水分解による前処理も行われる。それゆえ、PrPresは、これらの免疫学的検定の前駆体PrPScの代用品である。種々の免疫学的検定フォーマットの間で、ウェスタンブロッティングは、ジ−、モノ−および非グリコシル化PrPバンドのマイグレーションに基づくPrPの詳細な分子パターンを明らかにするという長所がある。この方法は、ヒトおよび動物のプリオン病における異なった脳PrPresのサブタイプを識別するためにも、広く使われてきた。現在では、より高いサンプル処理能力、より高い感度およびよりよい品質のために、ウエスタンブロット法以外の他の評価フォーマット{従来のELISA、解離促進ランタニド−蛍光免疫検定法(DELFIA,Barnard,2000年および米国特許公開第20020137114A1号明細書に記載された方法)および、ELISAと蛍光検出法と組み合わせた立体構造に依存する免疫学的検定法(CDI)(Safar,1998年、米国特許公開第20010001061A1号明細書、米国特許公開第20020001817A1号明細書)等}が開発されてきた。しかしながら、フォーマットに関係なく、PrPScは、必須の処理であるPK消化後にのみPrPCと選別し得るが、これでは、異種の生体サンプルについて最適化を図るのは困難であろうと思われる。(3)ヒツジのスクレーピーの臨床前診断のための第三眼瞼リンパ系組織の免疫組織化学処理を含むサンプル処理(O’Rourke,2000年、米国特許第6165784号明細書、米国特許第6261790号明細書)、脳および他の末しょう組織ホモジネートからのPrPScを濃縮するタングストリン酸ナトリウムによる化学的処理(Wadsworth,2001年)および、患畜および患者の尿中のプリオンタンパク質の新しいイソ型の検出(Shaked,2001b年、国際公開第0233420A2号パンフレット)のためのその他の方法も報告されている。キャピラリー電気泳動法およびフーリエ変換赤外分光法等の他の検出システムも報告されている。これらの方法は、まだその開発の初期段階にあり、技術的に複雑である。細胞プリオンを除去し、ついで、無差別に抗プリオン抗体認識を行うことによる、病原性プリオンの従来の識別法に加えて、PrPScをPrPCから区別できる、プラスミノーゲンおよびフィブリノーゲン等の他の試薬が見出された。得られた証拠から、プリオン一次配列または個々のPrPSc分子の特定の三次構造よりは、むしろ、種々の種のPrPScに共通する性質が、プラスミノーゲンとの結合の原因であり得ることが示唆された(Fischer,2000年、Maissen,2001年)。病原性プリオンタンパク質の捕捉と検出のためのプラスミノーゲンおよびその他の血清/血漿タンパクの使用への応用は、国際公開第0200713号パンフレット特許および米国特許公開第20010053533A1号明細書に記載されている(Aguzzi,2001年)。
PrPScベースの生前評価に共通の課題は、PrPScが末しょう組織または体液中に存在するかということである。技術的な困難性のため、PrPScの存在または、体液中におけるそれに関連した伝染については、実験データがほとんどなく、この課題には議論の余地がある。スクレーピーのハムスターモデルでは、血液中に低レベルの伝染力を検出し得る。罹患したハムスターの血液に由来するリンパ球リッチな軟膜における伝染力は、血漿に由来するものよりも大きいが、全血液接種物と比べると、相対的に小さな割合を占めるだけである。血液中に存在しているこの感染因子の分子的定義は明白でない。散発性のCJDペイシェントにおけるリスク因子と可能な感染源とを調査した結果、病気と、食物、輸血または他の血液産物の受容との間に有意の相関性は見出されなかった。初期のレポートでは、マウスへの脳内接種の後に、CJDペイシェントから得た血液における伝染力の存在の可能性が示されたが(Manuelidis,1985年、Tateishi,1985年)、古典的なヒトのプリオン病では、vCJDの場合を除いて、最も高い伝染力の量すなわちPrPres量は、常に中枢神経系(CNS)で見つかっており、末しょう組織での発見には一貫性がない。vCJDペイシェントの、扁桃、脾臓およびリンパ節等の周辺リンパ細網組織で容易に検出できるPrPScの存在により、リンパ細網組織に大量に存在するPrPresが、循環系と相互作用し、その結果、ごく僅かのPrPScがvCJDペイシェントの血液中に存在し、血液感染を引き起こすかも知れないと言う深刻な不安が生じた。血液中における他のTSEの伝染力は、種々の実験動物でも証明されてきている。伝染力研究のための血液の大部分は、TSEに順応した齧歯類(たとえばマウスおよびハムスター)から得られていた。マウスに順応したBSE、マウスに順応したvCJDが、脳内感染および静脈内感染を通して確立されてきた。唯一の例外モデルはヒツジで行われた研究であった。この実験では、BSE脳溶解物を接種した他のヒツジから取った全血液を輸血されたヒツジが、BSEの徴候を現した(Houston,2000年、Hunter,2002年)。しかしながら、これらの実験結果は、まだ十分検討される必要がある。そのような血液中の感染因子の検出が、PrPScとTSE疾患との関係をよりよく理解することの助けになることが期待されている。
「タンパク質のみ」または「プリオンのみ」説を受け入れるか受け入れないかに拘わらず、プリオン病の病因に貢献するかもしれないプリオン以外の剤または分子を探索するために継続的な努力が払われている。この探索は、答えを得ていない多くの疑問が推進力になっている。たとえば、合成された、いかなる汚染もないプリオンタンパク質は病気を引き起こさない。正常なPrPCの病原性PrPScイソ型への変換を誘発するメカニズムは知られていない。もう一つの未解決の疑問は、動物とヒトとで観察される(病気の潜伏期間、糖鎖形成レベルおよび障害パターンによって定義される)種々のプリオン病の表現型に関連する。単一のPRNP遺伝子型について同系交配のマウスの同型接合体の連続継代の後、全てのスクレーピー株が、その元の疾病プロフィールを保持していた。これらの観察により、研究者は、種々の表現型の株が同一の細胞プリオン前駆体の異なる立体構造のイソ型によって支配されたものであるのか、あるいは、遺伝し得る株の表現型を決定するもう一つの要因があるのかどうかについて疑問を持つようになった。実際、無細胞反応で造られた試験管内の変換モデルPrPresは、動物中で新しいTSE伝染力を構成する様子を見せなかった(Caughey,2003年)。これらの疑問のため、Prusinerが提案した「Xタンパク質」と呼ばれる未知の要素があると多くの人々が考えるようになってきているが、いまだ発見には至っていない。
ヘパリンや硫酸ヘパリン等のグリカン(GAG)は、PrPScアミロイド凝集体と関連していた。この関連性に対する親和性は、GAG::PrPCよりGAG::PrPScでずっと高いので、グリカン結合領域として特徴づけられるペプチドを使用して、選択的にPrPScを捕捉しPrPCを捕捉しないようにすることが可能である。この理由から、ヘパリンまたは硫酸ヘパリン結合領域と呼ばれるペプチド:(1)カルボキシ末端フィブロネクチンのWQPPRARI(Woods,1993年、Hines,1994年)、(2)アミロイド・タンパク質前駆体のNWCKRGRKQCKTH(Small,1994年)、(3)N−末端線維芽細胞成長因子(FGF)−1のNYKKPKL(Lou,1996年)および(4)ラミニンα1鎖のC−末端G−領域のKDFLSIELVRGRVK(吉田,1999年)が合成された。
Kringle領域は、プラスミノーゲンをPrPScに選択的に結合させることに関与していた。Kringle領域が、正に荷電したアミノ酸(たとえばArgおよびLys)が関与するヘパリン/硫酸ヘパリン結合活性(水野,1994年)を有することが知られていた(Soeda,1989年)。別の出版物では、プリオン配列中に保存された二つのトリペプチド「YYR」またはむしろ、提案された三つの非連続の「YYX」が、PrPScと相互作用するが、PrPCとは相互に作用しないことが見出された(Paramithiotis,2003年、国際公開第0078344A1号パンフレット)。興味深いことに、ヒトのプラスミノーゲン配列中には、4つのTyr−Arg−Gly配列が、5つのKringle領域中4つ(Kringle領域1中のY(92)R(93)G(94),Kringle領域3中のY(264)R(265)G(266),Kringle領域4中のY(366)R(367)G(368)およびKringle領域5中のY(470)R(471)G(472))に存在していた。プラスミノーゲンには、K(19)K(20), R(61)K(62), K(77)K(78), R(153)Y(154), K(211)K(212), K(233)R(234), K(311)R(312), K(377)K(378), K(433)K(434), K(473)R(474), R(530)K(531), K(556)K(557), Y(614)K(615), R(644)K(645), R(712)Y(713), K(752)Y(753)等の16のTyr−LysまたはArg−TyrまたはLys−TyrまたはArg−LysまたはLys−ArgまたはLys−Lys配列が存在していた。さらに、数個の離れたTyr、ArgまたはLys残基が、Kringleループに形成されたジスルフィドブリッジによって、相互に近づけられていた(図1)。これらの観察に基づいて、恐らく、PrPSc複合体が関連するグリカンとの相互作用を介して、プラスミノーゲンKringle領域中とプリオン配列中のアミノ酸(Tyr,ArgおよびLys)が、PrPScへの選択的結合に寄与するものと考えられた。この理由から、2つの「濃縮されたKringle」ペプチド(YRGYRGYRGYRGおよびYRGRYGYKGKYGYRG)が合成された。
核酸は、PrPSc凝集体と関連するもう一つの分子カテゴリーである。PrPScを捕捉するのに、抗DNA抗体が有効に使用されてきた。ヒストンは、核DNAに結合することが知られている一群のタンパク質である。したがって、PrPScの捕捉能力を評価するために、(1)H2B(21−41)のAQKKDGKKRKRSRKESYSIYV、(2)H3(1−21)のARTKQTARKSTGGKAPRKQLAおよび、(3)H4(2−24)のSGRGKGGKGLGKGGAKRHRKVLRの3つのペプチドが合成された。
〔発明の概要〕
正常およびスクレーピーのハムスターの脳溶解物を、全脳組織ホモジネートを10%(w/v)含んだPBS(リン酸緩衝生理食塩水)として、ボルチモア研究および教育財団(Baltimore Research and Education Foundation)から得た。さらに、この溶解物に、5%のナトリウムデオキシコレートと5%のIgpal CA−630(NP−40と同等)とをPBSに含ませてなる10Xの洗剤ホモジネート緩衝液(1/10体積量)を加え、4℃で1時間培養し、ついで、1,000gで10分間で遠心沈降処理した。得られた上澄みを集め、評価に使用した。
9つのペプチドを、ResGen(現在は、Invitrogen社の一部門)で合成し、あるいは、Upstate Group社から購入した。それぞれのペプチドのビオチンラベル付けを、アミノヘキサノイルスペーサー(K(Lc))を持つC末端リジンで、または、アミノヘキサノイルスペーサー(AMCAP)を介してN末端で、行った。

ビオチン化されたペプチドはPBS中に1mg/mLで溶解し、使用まで−20℃で保存した。
0.5mLのDynabeads(R) M−280 ストレプトアビジン(米国,ニューヨーク,Dynal Biotech社,Cat.#112.06)をPBSで二回洗浄し、磁石(Dynal Magnetic Particle Concentrator社,MPC)でこのビーズを緩衝液から単離した。100μgのペプチドと1mLのPBSとをこの洗浄ビーズに添加した。回転付きの培養を37℃で1〜2時間行った。MPCで緩衝液からビーズを単離し、1mLのPBS(0.1%BSA)で二回洗浄した。洗浄中、室温で5分間回転を行った。ついで、ペプチドを接合したビーズを、1mMビオチンの、0.1%のBSAを含む0.2Mのトリス−塩酸,pH8.0の液中で、37℃で2時間、ブロックした。ついで、ビーズを1mLのPBS(0.1%のBSA)により2回洗浄し、1mLのPBS(0.1%のBSA、1%のTween 20)で一回洗浄し、毎回、室温で10分間培養した。その後、ビーズを、1mLのPBS(0.1%BSA)で一回洗浄し、ついで、1mLのPBS(0.05%のアジ化ナトリウム)中に4℃で保存した。
脳溶解物のPK消化の条件:脳ホモジネートを、非イオン性洗剤を使用しまたは使用しないで、PBS緩衝液中に懸濁した。全ホモジネートタンパク質の濃度は、僅か2.5mg/mLであった。PK(米国,インディアナ州,Roche Diagnostics社,Cat.#1373196)を添加して、50μg/mLの最終濃度にした。37℃で0.5〜1時間掛けて培養を行った。Pefabloc SC(米国,インディアナ州,Roche Diagnostics社,Cat.#1585916)を添加して消化を停止し、4mMの最終濃度にした。
ペプチド接合磁力ビーズを使用し、免疫沈降によって脳ホモジネートからPrPScを捕捉した。このIP手順は、次のプロトコルからなっていた:50μLのペプチド接合ビーズを、全部で1mLのIP緩衝液(3%のTween 20と3%のIgpal CA−630を含むPBS)中、PK処理したまたはPK処理していない脳ホモジネートと混合し、25℃下、2.5時間、回転付きで培養する。→MPCデバイスを使用してビーズを分離し、IP洗浄用緩衝液(2%のTween 20と2%のIgpal CA−630を含むPBS)でボルテックス(vortexing)することにより、30秒間ずつ3回ビーズを洗浄する。→ビーズをNuPAGEサンプル緩衝液と共に、10〜15分間加熱して、捕捉されたPrPScを溶出する。IP捕捉物からの溶出サンプルを、4〜12%のNuPAGE(R)Bis−Tris Gel(米国,カリフォルニア州,Invitrogen社,Cat.#NP0302)にロードして、200Vで、45分間、非還元電気泳動に供した。免疫ブロット手順を以下のように実行した:ゲル中の分離タンパク質を、30V、60分間で、0.2μmのPVDF膜(Invitrogen,Cat#LC2002)に移動させる。→この膜組織を、BlockerTM Casein含有TBS(0.05%のTween 20)(米国,イリノイ州,Pierce Chemical社,Cat.#37532)で、振盪付きで、25℃下1時間かけるかまたは4℃で一昼夜かけてブロックする。→一次抗体として、1:3,000希釈の3F4(米国,マサチューセッツ州,Signet社,Cat.#9620−02)または、1:5,000希釈の6H4(スイス,Prionics社,Cat.#01〜011)を使用してPrPScを検出する。25℃下、振盪付きで1時間かけて、10%のブロッカーTMカゼイン含有TBST緩衝液(25mMのTri−Cl,0.2MのNaCl,0.2%のTween20,pH8.0)に一次抗体を希釈した液で膜組織を培養する。→振盪付きで3回×5分間、TBST緩衝液で洗浄する。→50%のブロッカーTMカゼイン含有TBST緩衝液中1:10,000〜1:30,000に希釈した、ホースラディッシュペルオキシダーゼ接合ヤギポリクローナル抗マウスIgG(H+L)(米国,ペンシルベニア州,Jackson ImmunoResearch Laboratories社,Cat.#115−035−003)で、膜組織を、25℃下、振盪付きで1時間かけて培養する。→振盪付きで6回×5分間、TBST緩衝液で洗浄する。→ECL化学ルミネセンス基質(米国、ニュージャージー州,Amersham Biosciences社,Cat.#RPN2109)または、SuperSignal West Dura化学ルミネセンス基質(Pierce社)を膜組織に添加し、5分間現像する。→Bio Max MR−2フィルム(米国、ニューヨーク州,Kodak社)またはChemiDoc Gel Documentation System(米国,カリフォルニア州,Bio−Rad社)へ曝露して像を得る。

ラベル付けされた、プリオンに対し特異性を有する抗体と
を含んでなるキット。
Claims (7)
- (配列ID番号1)WQPPRARIからなる群から選ばれたペプチドを含むPrPScを検出するための試薬。
- PrPScの検出方法であって、
最初に、サンプルを請求項1に記載のペプチドと接触させ、
ついで、PrPScを検出する、
ことを含む検出方法。 - PrPScを検出するための免疫学的検定法であって、
請求項1に記載のペプチドを結合させた固体担体を供給し、
当該固体担体をサンプルと接触させ、
当該担体を洗浄して結合していないサンプルを除去し、
当該固体担体をプリオンに対し特異性を有する抗体に接触させ、
PrPScが当該固体担体に結合しているかどうかを決定する検出ステップを実行する
ことを含む免疫学的検定法。 - PrPScの検出のためのキットであって、
請求項1に記載のペプチドを結合させた固体担体と、
ラベル付けされた、プリオンに対し特異性を有する抗体と
を含んでなるキット。 - PrP Sc の検出方法であって、
サンプルをプリオンに対し特異性を有する抗体に接触させ、ついで、ラベル付けされた、(配列ID番号1)WQPPRARIからなる群から選ばれたペプチドに接触させ、PrP S を検出することを含む、方法。 - PrP Sc を検出するための免疫学的検定法であって、
プリオンに対し特異性を有する抗体を結合させた固体担体を供給し、
当該固体担体をサンプルと接触させ、
当該担体を洗浄して結合していないサンプルを除去し、
当該固体担体を、ラベル付けされた、(配列ID番号1)WQPPRARIからなる群から選ばれたペプチドに接触させ、
PrP Sc が当該固体担体に結合しているかどうかを決定する検出ステップを実行する
ことを含む免疫学的検定法。 - PrP Sc の検出のためのキットであって、
プリオンに対し特異性を有する抗体を結合させた固体担体と、
