Description
本発明は、細胞表面に発現されるTLR4/MD−2レセプターを認識するモノクローナル抗体を提供する。上記抗体はLPS誘導IL−8産生をブロックすることができる。幾つかの場合においては、本発明のモノクローナル抗体は、MD−2と複合体化していない場合のTLR4も認識する(例えば、可溶性TLR4蛋白質、細胞表面で発現されたTLR4)。例示的なモノクローナル抗体が18H10、16G7、15C1および7E3を含む。

(本明細書中においては「活性化合物」ともいわれる)の抗体または可溶性キメラポリペプチド、およびその誘導体、フラグメント、アナログおよびホモログは投与に適した薬学的組成物に配合することができる。そのような組成物は、典型的には、抗体または可溶性キメラポリペプチドおよび薬学的に受容可能なキャリアを含む。本明細書中で用いるように、用語「薬学的に受容可能なキャリア」は、薬学的組成物に適合する、いずれかのおよび全ての溶媒、分散媒体、コーディング、抗菌および抗真菌剤、等張および吸収遅延剤などを含むことを意図する。適当なキャリアは、ここに引用して援用する、当該分野における標準的な参照テキストである、Remington’s Pharmaceutical Sciencesの最近の改訂版に記載されている。そのようなキャリアまたは希釈剤の好ましい例は、限定されるものではないが、水、セーライン、リンゲル液、デキストロース溶液、および5%ヒト血清アルブミンを含む。リポソーム、および不揮発性油のような非水性ビークルも用いることができる。医薬上活性な物質のためのそのような媒体および剤の使用は当該分野でよく知られている。いずれかの慣用的な媒体または剤が活性化合物に適合しないのを除き、組成物におけるその使用が考えられる。補助的活性化合物も組成物に配合することもできる。
本発明は、モジュレーター、すなわち、MD−2アクセサリー蛋白質へのTLR4の結合を変調し、またはそうでなければそれに干渉する候補またはテスト化合物または剤(例えば、ペプチド、ペプチドミメティックス、小分子または他の薬物)、またはTLR4および/またはTLR4/MD−2複合体のシグナル伝達機能を変調する、またはそうでなければそれに干渉する候補またはテスト化合物または剤を同定するための(ここでは「スクリーニングアッセイ」ともいわれる)方法を提供する。また、異常なLPS−シグナル伝達に関連する障害を治療するのに有用な化合物を同定する方法も提供される。本発明は、ここに記載するスクリーニングアッセイにおいて同定される化合物も含む。
本発明の抗体は、適当なアッセイ、例えば、慣用的なタイプのイムノアッセイによって検出することができる。例えば、サンドイッチアッセイを行うことができ、そこでは、TLR4/MD−2複合体またはTLR4蛋白質またはそのフラグメントを固相に付着させる。インキュベーションを、試料中の抗体が固体プレート上の固定化されたポリペプチドに結合するのを可能とするのに十分な時間維持する。この最初のインキュベーションの後、固相を試料から分離する。固相を洗浄して、未結合物質、および試料にやはり存在するかもしれない非特異的蛋白質のような干渉物質を除去する。固定化されたポリペプチドに結合した注目する抗体(例えば、モノクローナル抗体18H10、16G7、15C1および/または7E3)を含有する固相を、引き続いて、第二の標識された抗体、またはビオチンまたはアビジンのようなカップリング剤に結合した抗体と共にインキュベートする。この第二の抗体はもう1つの抗−TRL4/MD−2複合体抗体、もう1つの抗−TLR4抗体またはもう1つの抗体であってよい。抗体についての標識は当該分野で良く知られており、放射性核種、酵素(例えば、マレイン酸レヒドロロゲナーゼ、ホースラディッシュペルオキシダーゼ、グルコースオキシダーゼ、カタラーゼ)フルオル(フルオレセインイソチアシアネート、ローダミン、フィコシアニン、フルオレスカルミン)、ビオチン等を含む。標識された抗体を固体と共にインキュベートし、固相に結合した標識を測定する。これらのおよび他のイムノアッセイは当業者によって容易に実施できる。
(A.安定なTLR4/MD−2トランスフェクタントの創製)
安定なTLR4/MD−2トランスフェクタントをCHO−K1およびHEK293細胞において創製した。CHO−K1細胞では、N−末端c−mycエピトープタグをコードするヒトTLR4 cDNAをpCDNA3.1(−)hygro(Invitrogen)にクローン化し、C−末端c−MycおよびプロテインCエピトープタグをコードするヒトMD−2 cDNAをpCDNA3(Invitrogen)にクローン化した。製造業者のガイドラインに従って、Fugene6TM試薬(Roche)を用いて双方の構築体をCHO細胞に共トランスフェクトした。抗生物質耐性細胞を、(共にInvitrogenからの)500μg/mLのG418および250μg/mLのヒグロマイソインBを含有する培養基にて選択した。
組換え可溶性MD−2を創製するために、選択および精製目的でのC−末端FLAGおよび6×HISタグを持つ上記蛋白質をコードするcDNAをpFASTBAC1にクローン化し、引き続いて、相同組換えによって、バクミドDNAに挿入した。ウイルスストックの創製に続いて、Sf9細胞をスーパー感染させた。48時間後、NiNTAアフィニティーマトリックス(Qiagen)を用いて、組換え蛋白質を感染した細胞上清から精製した。
ここに引用してその全体を引用するBuell et al., Blood 92:3521−3528(1998)に以前記載されたように、日0、7および28において、8週齢メスBALB/cマウス(IFFA CREDO)をRIBIアジュバント(Sigma)中の106CHO細胞/mLの皮下注射(s.c.)で免疫化した。
マウスを日0および14に採血した。TLR4/MD−2トランスフェクトされた293細胞についてのFACS分析によって、TLR4/MD−2特異的抗体力価を血清中にてアッセイした。細胞を1:250、1:2500および1:25000希釈にてマウス血清と共にインキュベートし、洗浄し、APC−コンジュゲーテッドヤギ抗−マウスIgG(H+L)抗体(Molecular Probes)と共にインキュベートし、FL−4チャネルにおいてFACScalibur(Becton Dickenson)で分析した。
特異的TLR4/MD−2血清抗体を有するマウスを、融合から3ないし4日先立って、キメラTLR4/MD−2融合蛋白質で皮下(s.c.)「過剰ブースター注射」した。排出リンパ節をマウスミエローマ細胞系P3−X63−Ag8.653との融合のためのB細胞の源として得た。B細胞抽出および細胞融合は、ここに引用してその全体を援用する、Buell et al.,Blood 92:3521−3528(1998)に以前記載されたように行った。細胞を104ミエローマ細胞/ウェルの近似的濃度で平板培養し、HAT(Sigma)を補足した培養基中で10ないし14日間増殖させた。
生きたハイブリドーマ細胞を含有するウェルからの上清を、FACS分析によって、TLR4/MD−2特異性につき、モックトランスフェクトされた細胞 vs TLR4/MD−2トランスフェクトされたミエローマ細胞に対してスクリーニングした。次いで、細胞を上清およびヤギ−抗マウスIgG(H+L)抗体(Molecular Probes)と共にインキュベートした。細胞をFL−4チャネルにおいてFACScaliburで分析した。
10%FBSを含有する2mL培養基中で2.5×105細胞/ウェルの密度にて、HEK293細胞を6ウェルプレートで平板培養した。平板培養から16時間後、製造業者のガイドラインに従って、FugeneTM試薬(Roche)を用い、細胞を0.75μgの適当なベクターでトランスフェクトした。トランスフェクションから48時間後、細胞を(図4に示すように)適当なモノクローナル抗体およびAPC−カップルドヤギ抗−マウスIgG(H+L)抗体(Molecular Probes)で染色し、FL−4チャネルにおいてFACScaliburを用いて分析した。
C末端FLAGおよびヒスチジンエピトープタグを持つ組換え可溶性MD−2を、ELISAプレート(Nunc Maxisorp)上にて、50μLのPBS中の5μg/mLの濃度で被覆した。ウェルを200μLのPBS 2%BSAでブロックし、引き続いて、PBS 1%BSA中の示された濃度にて適当なMAbでインキュベートした。PBS 0.05%Tween 20での3回の洗浄工程に続き、1:5000 希釈の50μLのHRPコンジュゲーテッドヤギ抗−マウスIgG(H+L)をウェルに加えた。更なる洗浄工程に続いて、結合をTMB基質を用いて明らかにした、プレートを650nmの波長で読んだ。
試料分離のために、前記パラグラフGに記載されたように、Fugene 6TMトランスフェクション試薬を用い、HEK293細胞を適当なプラスミド構築体でトランスフェクトした。トランスフェクションから48時間後、細胞を収集し、遠心によって透明化した。細胞を、引き続いて、ビオチニル化18H10(10μg/mL)と共にインキュベートし、COMPLETETMプロテアーゼ阻害剤(Roche)を含有する20mMトリスpH7.4、150mM NaCl、1%NP40中で溶解させた。
モノクローナル抗体をまずプロテインGアフィニティークロマトグラフィーを用いてハイブリドーマ細胞上清から精製した。
ヒト全血をRPMI(Sigma)中に1:4希釈し、所望の最終濃度の2倍の適当なモノクローナル抗体を含む96ウェルプレートにおいて100μL/ウェルで37℃にて30分間平板培養した。所望の最終濃度の2倍の用量のLPS(K12LD25, Sigma)を、引き続いて、5mg/mLのHSAを含有する100μLのRPMIに加え、37℃にて6時間インキュベートした。次いで、プレートを2000rpmにおいて5分間遠心し、各ウェルからの上清を保持した。前記したように、モノクローナル抗体の対801EおよびM802B(Endogen)を用い、IL−8の濃度をサンドイッチELISAによって測定した。
107ハイブリドーマ細胞を収穫し、PBSで1回洗浄し、その後、1mLのTrizolTM試薬(Invitrogen)に再懸濁した。引き続いて、製造業者のガイドラインに従って全RNAを抽出した。18H10ハイブリドーマの3つの独立したサブクローンからのVHおよびVLをコードするcDNAを、製造業者のガイドラインに従ってマウスScFvモジュール(Amersham Biosciences)を用いるRT−PCRによって創製した。増幅した産物をpGEM−Tイージーベクター(Promega Corp.)にクローン化し、T7およびSP6プライマーを用いて配列決定した。
表面TLR4/MD−2を発現するCHO細胞で免疫化したマウスを、特異的血清力価についてモニターした。TLR4/MD−2に対する応答を示すものを、組換えTLR4/MD−2キメラ蛋白質「過剰ブースター注射」した。この戦略は、非特異的CHO細胞抗原に対する応答を最小化し、同時に、過剰ブースター注射に際してTLR4/MD−2特異的応答を最大化しつつ、免疫系が最初に立体配座TLR4/MD−2複合体に暴露されたことを確認するために選択した。B細胞/ミエローマ融合に由来するハイブリドーマの上清FACSによるスクリーニングを、モックトランスフェクトされた vs TLR4/MD−2トランスフェクトされたCHO細胞で行った。ここでは18H10といわれる1つの特定のクローンからのモノクローナル抗体は、TLR4/MD−2トランスフェクトされたCHO細胞に対する特異的結合を示した(図1)。この抗体は、マウスIgイソタイピングCBAキット(Beckton Dickenson)を用いるFACSによって測定して、IgG2b κイソタイプを有することが判明した。
LPSは、TLR4/MD−2複合体でトランスフェクトされたHEK293細胞においてIL−8の産生を誘導する能力を有することが知られている。このIL−8誘導を阻害する18H10の能力は、LPS投与に先立って抗体と共に細胞を予め30分間インキュベートすることによって分析した。図2は、18H10が、1μg/mL未満の濃度でさえHEK293細胞に対するLPSの効果を阻害したことを示す。
ヒト全血においてLPS誘導IL−8産生を阻害する18H10の能力をテストした。18H10中和活性は、0.5ないし10μg/mLのモノクローナル抗体の濃度の範囲を用い、3つの異なるドナーからの血液でテストした。図3は、イソタイプがマッチした対照と比較して、全ての3つのドナーにおけるLPSによって誘導されたIL−8のレベルを18H10が有意に低下させたことを示す。18H10は、(e−biosciencesから購入した)以前に記載されたα−TLR4ブロッキングモノクローナル抗体よりも優れていることが判明した。これらの結果は、トランスフェクトされたHEK293細胞上の18H10によって認識される中和エピトープもまた全血中の細胞の表面に暴露され、および1μg/mL未満の濃度においてさえ、18H10は全血中のLPSの活性を阻害するのに十分優れていることを示す。
18H10モノクローナル抗体の特異性を測定するために、18H10は(以前クローン化された)TLR4/MD−2複合体のウサギオルソログを認識しないという事実が利用された。N−末端FLAGTMエピトープタグを持つヒトまたはウサギTLR4、およびC−末端c−MycおよびプロテインCエピトープタグを持つヒトまたはウサギいずれかのMD−2についてのcDNAを以下の組合せにてHEK293細胞においてトランスフェクトした:(1)ヒトTLR4およびMD−2;(2)ウサギTLR4およびウサギMD−2;(3)ヒトTLR4およびウサギMD−2;(4)ウサギTLR4およびヒトMD−2。図4は抗体染色に続いてのこれらの細胞のFACS分析を示し、これは、ウサギTLR4/MD−2複合体あるいはウサギTLR4およびヒトMD−2の組合せではなく、ヒトTLR4/MD−2複合体、およびヒトTLR4およびウサギMD−2の組合せを発現する細胞を18H10が認識したことを明らかとした。これらの結果は、18H10によって認識されるエピトープはヒトMD−2に位置することを示す(図4)。
18H10ハイブリドーマクローンからのVHおよびVL配列は、RT−PCRによって全RNAから増幅した。配列分析を図6Aないし6Fに示す。
hTLR4/MD−2複合体についてのクローン化された18H10 VHおよびVLの特異性を示すために、キメラ18H10 MAbを用いて、FACS分析をhTLR4/MD−2トランスフェクトされたCHO細胞で行った(図6)。APC−標識ヤギ−抗−IgG(H+L)二次抗体を用い、示された濃度におけるMAbの特異的結合を検出した。無関係なイソタイプがマッチしたIgG1 MAbを対照として用いた。
LPSについてのクローン化された18H10 VHおよびVLの中和能力を示すために、hTLR4/MD−2トランスフェクトされたHEK293細胞のLPS依存性IL−8誘導を阻害する18H10の能力を(前記したように)テストした。図7は、元の18H10マウスMAbのそれと非常に同様に、キメラ18H10がHEK293細胞に対するLPSの効果を阻害したことを示す。

表面TLR4/MD−2を発現するCHO細胞で免疫化したマウスを特異的血清力価についてモニターした。TLR4/MD−2に対する応答を示すものは、HEK293 TLR4/MD−2トランスフェクタントで「過剰ブースター注射」した。この戦略は、TLR4/MD−2−特異的応答を同時に最大化しつつ、非−特異的CHO細胞抗原に対する応答を最小化するために選択した。B細胞/ミエローマ融合から得られたハイブリドーマからの上清のFACSによるスクリーニングを、モックトランスフェクトされたvs TLR4/MD−2トランスフェクトされたCHO細胞で行った。ここでは16G7といわれる特異的クローンからのモノクローナル抗体は、TLR4/MD−2トランスフェクトされたCHO細胞に対する特異的結合を示した(図9)。16G7は、マウスIgGイソタイピングCBAキット(Beckton Dikenson)を用いるFACSによって測定して、IgG1 κイソタイプを有することが判明した。
LPSは、TLR4/MD−2複合体でトランスフェクトされたHEK293細胞においてIL−8産生を誘導する能力を有することが知られている。このIL−8誘導を阻害する16G7の能力は、LPS投与に先立って30分間細胞を各抗体と共にプレインインキュベーティングすることによって分析した。図10は、サブ−μグラム/mL濃度においてさえ、HEK293細胞に対するLPSの効果を16G7が阻害したことを示す。
ヒト全血においてLPS誘導IL−8産生を阻害する16G7の能力をテストした。0.5ないし5μg/mLの範囲のモノクローナル抗体の濃度を用いて、16G7中和活性を3つの異なるドナーからの血液でテストした。図11は、イソタイプがマッチした対照と比較して、全ての3つのドナーにおけるLPSによって誘導されたIL−8のレベルを16G7が有意に低下させたことを示す。16G7は以前記載された(e−biosciencesからの)α−TLR4ブロッキングモノクローナル抗体よりも優れていることが判明した。(Shimazu et al., J.Exp.Med.189:1777−1782(1999)参照)。幾つかの場合において、16G7は、やはり実験に含めたα−CD14ブロッキングモノクローナル抗体と同程度に優れていることが判明した。(Kirkland et al.,J.Biol.Chem.268:24818−24823(1993)参照)。これらの結果は、トランスフェクトされたHEK293細胞についての16G7によって認識される中和エピトープもまた全血中の細胞上の表面に暴露され、16G7は、1μg/mL未満の濃度においてさえ、全血においてLPSの活性を阻害するのに十分優れていることを示す。
16G7モノクローナル抗体の特異性を測定するために、16G7が(以前クローン化された)TLR4/MD−2複合体のウサギオルソログを認識しないという事実を利用した。N−末端FLAGTMエピトープタグを持つウサギまたはヒトいずれかのTLR4、およびC−末端c−MycおよびプロテインCエピトープタグを持つMD−2についてのcDNAを、以下の組合せにおいてHEK293細胞においてトランスフェクトした:(1)ウサギTLR4およびウサギMD−2;(2)ヒトTLR4およびヒトMD−2;(3)ウサギTLR4およびヒトMD−2;(4)ヒトTLR4およびウサギMD−2.図12は抗体染色に続いてのこれらの細胞のFACS分析を示し、これは、16G7が、ウサギTLR4/MD−2複合体、またはウサギTLR4およびヒトMD−2の組合せではなく、ヒトTLR4/MD−2複合体、およびヒトTLR4およびウサギMD−2の組合せを発現する細胞を認識したことを明らかとした。これらの結果は、16G7によって認識されるエピトープがヒトTLR4に位置することを示す(図12)。
16G7ハイブリドーマクローンからのVHおよびVL配列を、RT−PCRによって全RNAから増幅した。配列分析は図13Aないし13Fに示す。(http://imgt.cines.frに見出すことができるInternational Immunogenetics Information Systemを用いる)16G7 VHおよびVLヌクレオチド配列と既知のマウスVHおよびVL配列の整列により、16G7 VH配列はIgHV1サブファミリーに最も密接に似ており、他方、16G7 VLはIgKV1サブファミリーに属することを明らかとする。
(A.安定なTLR4/MD−2トランスフェクタントの創製)

15C1モノクローナル抗体の特異性を測定するために、15C1は(以前クローン化された)TLR4/MD−2複合体のウサギオルソログを認識しないという事実を利用した。N−末端FLAGTMエピトープタグを持つウサギまたはヒトいずれかのTLR4、およびC−末端c−MycおよびプロテインCエピトープタグを持つMD−2についてのcDNAを、以下の組合せにてHEK293細胞においてトランスフェクトした:(1)モックベクター(2)ヒトTLR4単独(3)ヒトTLR4およびヒトMD−2(4)ウサギTLR4およびウサギMD−2;(5)ヒトTLR4およびウサギMD−2;(6)ウサギTLR4およびヒトMD−2。図17は抗体染色に続いてのこれらの細胞のFACSの抗体を示し、これは、15C1が、ウサギTLR4/MD−2複合体、またはウサギTLR4およびヒトMD−2ではなく、TLR4単独、ヒトTLR4/MD−2複合体、およびヒトTLR4およびウサギMD−2の組合せを発現する細胞を認識したことを明らかとした。これらは、15C1によって認識されたエピトープがヒトTLR4に位置することを示す(図17)。
15C1ハイブリドーマクローンからのVHおよびVL配列をRT−PCRによって全RNAから増幅した。配列分析は図18Aないし18Fに示す。
hTLR4/MD−2複合体についてのクローン化された15C1 VHおよびVLの特異性を示すために、キメラ15C1 MAbを用いるhTLR4/MD−2トランスフェクトされたCHO細胞についてのFACS分析を行った(図19)。示された濃度におけるMAbの特異的結合は、APC−標識ヤギ−抗−ヒトIgG(H+L)二次抗体を用いて検出した。無関係なイソタイプがマッチしたIgG1 MAbを対照として用いた。
LPSについてのクローン化された15C1 VHおよびVLの中和能力を示すために、hTLR4/MD−2トランスフェクトされたHEK293細胞のLPS依存性IL−8誘導を阻害する15C1の能力を(前記したように)テストした。図20は、キメラ15C1が、15C1 MAbのそれと非常に同様にHEK293細胞に対するLPSの効果を阻害したことを示す。
(A.安定なTLR4/MD−2トランスフェクタントの創製)
安定なTLR4/MD−2トランスフェクタントは、実施例9、サブセクションAにて前記したようにCHO−K1およびHEK293細胞において創製した。
組換え可溶性MD−2は、実施例15、サブセクションBにて前記したように創製した。
8週齢雌BALB/cマウス(IFFA CREDO)を実施例1、サブセクションCにて前記したように免疫化した。
マウス血清力価測定は実施例1、サブセクションDにて前記したように行った。
B細胞抽出および細胞融合を、実施例9、サブセクションDにて前記したように行い、分析した。
ハイブリドーマのスクリーニングは実施例1、サブセクションFにて前記したように行った。
7E3モノクローナル抗体の特異性は実施例1、サブセクションGにて前記したように測定した。
モノクローナル抗体はプロテインGアフィニティークロマトグラフィーを用いてハイブリドーマ細胞上清からまず精製した。
細胞アッセイIIは実施例1、サブセクションKにて前記したように行った。
107ハイブリドーマ細胞を収穫し、PBSで1回洗浄し、その後、1mLのTrizolTM試薬(Invitrogen)に再懸濁した。引き続いて、製造業者のガイドラインに従って、全RNAを抽出した。製造業者のガイドラインに従って、7E3クローンからのVHおよびVLをコードするcDNAを、マウスScFvモジュール(Amersham Biosciences)でのRT−PCRによって創製した。増幅された産物をpGEM−Tイージーベクター(Promega Corp.)にクローン化し、T7およびSP6プライマーを用いて配列決定した。
表面TLR4/MD−2を発現するCHO細胞で免疫化したマウスを、特異的血清力価についてモニターした。TLR4/MD−2に対する応答を示すものを、HEK293 TLR4/MD−2トランスフェクタント「過剰ブースター注射:」した。この戦略は、TLR4/MD−2−特異的応答を同時に最大化しつつ、非特異的CHO細胞抗原に対する応答を最小化するために選択された。B細胞/ミエローマ融合から得られたハイブリドーマからの上清をFACSによるスクリーニングを、モックトランスフェクトされたvs TLR4/MD−2トランスフェクトされたCHO細胞で行った。7E3とここではいわれる特異的クローンからのモノクローナル抗体は、TLR4/MD−2トランスフェクトされたCHO細胞への特異的結合を示した(図21)。7E3は、マウスIgイソタイピングCBAキット(Beckton Dickenson)を用いるFACSによって測定されるように、IgG1 κイソタイプを有することが判明した。
LPSは、TLR4/MD−2複合体でトランスフェクトされたHEK293細胞においてIL−8産生を誘導する能力を有することが知られている。このIL−8誘導を阻害する7E3の能力は、LPS投与に先立って30分間、細胞を各抗体と共にプレインキュベートすることによって分析した。図22は、サブ−マイクログラム/mLの濃度においてさえ、7E3がHEK293細胞に対するLPSの効果を阻害したことを示す。
ヒト全血におけるLPS誘導IL−8産生を阻害する7E3の能力をテストした。7E3中和活性は、0.5ないし5μg/mLの範囲のモノクローナル抗体濃度を用いて3つの異なるドナーからの血液でテストした。図23は、7E3が、イソタイプがマッチした対照と比較して、全ての3つのドナーにおけるLPSによって誘導されたIL−8のレベルを有意に低下したことを示す。7E3は(e−biosciencesから購入された)以前記載されたα−TLR4ブロッキングモノクローナル抗体よりも優れていることが判明した。(Shimazu et al., J.Exp.Med.189:1777−1782(1999)参照)。幾つかの場合において7E3は、やはり実験に含めたα−CD14ブロッキングモノクローナル抗体と同程度優れていることが判明した。(Kirkland et al.,J.Biol.Chem.268:24818−24823(1993)参照)。これらの結果は、トランスフェクトされたHEK293細胞上の7E3によって認識される中和エピトープもまた全血中の細胞の表面に暴露され、および7E3が、1μg/mL未満の濃度においてさえ、全血におけるLPSの活性を阻害するのに十分優れていることを示す。
7E3モノクローナル抗体の特異性を示すために、7E3は(以前クローン化された)TLR4/MD−2複合体のウサギオルソログを認識しないという事実を利用した。N−末端FLAGTMエピトープタグを持つウサギまたはヒトいずれかのTLR4、およびC−末端c−MycおよびプロテインCエピトープタグを持つMD−2についてのcDNAを以下の組合せにてHEK293細胞においてトランスフェクトした:(1)モックベクター(2)ヒトTLR4単独(3)ヒトTLR4およびヒトMD−2(4)ウサギTLR4およびウサギMD−2;(5)ヒトTLR4およびウサギMD−2;(6)ウサギTLR4およびヒトMD−2。図24は抗体染色に続いてのこれらの細胞のFACS分析を示し、これは、7E3が、ウサギTLR4/MD−2複合体、またはウサギTLR4およびヒトMD−2の組合せではなく、ヒトTLR4/MD−2複合体、およびヒトTLR4およびウサギMD−2の組合せを発現する細胞を認識することを明らかとした。これらの結果は、7E3によって認識されたエピトープがヒトTLR4に位置するが、MD−2の存在はMAb結合で必須でないことを示す(図24)。
7E3ハイブリドーマクローンからのVHおよびVL配列を、RT−PCRによって全RNAから増幅した。配列分析は図25Aないし25Fに示す。
hTLR4/MD−2複合体についてのクローン化された7E3 VHおよびVLの特異性を示すために、キメラ7E3 MAbを用いるhTLR4/MD−2トランスフェクトされたCHO細胞についてのFACS分析を行った(図26)。示された濃度におけるMAbの特異的結合は、APC−標識ヤギ−抗−ヒトIgG(H+L)二次抗体を用いて検出した。無関係なイソタイプがマッチしたIgG1 MAbを対照として用いた。
LPSについてのクローン化された7E3 VHおよびVLの中和能力を示すために、hTLR4/MD−2トランスフェクトされたHEK293細胞のLPS依存性IL−8誘導を阻害する7E3の能力を前記したようにテストした。図27は、キメラ7E3がHEK293細胞に対するLPSの効果を阻害したことを示す。
グリシンセリン(GGGGS3)リンカーを介してMD−2に連結されたTLR4の細胞外部分をPCRを用いて組み立てた。検出および精製目的で、FLAGおよび6×HISタグをMD−2のC−末端に含めた(図28)。
実施例1のcDNAカセットをバキュロウイルス発現ベクターpFASTBAC1(Invitrogen)にクローン化し、引き続いて、相同組換えによってバクミドDNAに挿入した。ウイルスストックの創製に続いて、Sf9細胞をスーパー感染させ、TLR4/MD−2融合蛋白質の発現を、ウエスタンブロッティングによって、感染から48時間および72時間後に細胞溶解物において分析した(図29)。
融合蛋白質を精製するために、Sf9細胞をスーパー感染から48時間後に収穫し、5容量/グラム細胞の濃度のCOMPLETETMプロテアーゼ阻害剤(Roche)を含む20mMトリスpH7.4、150mM NaCl、1%NP40に溶解させた。4℃における15時間のインキュベーションに続く、溶解物を遠心(4000rpm)および濾過(0.22μm)によって清澄化し、抗−FLAG M2 MAbアフィニティーマトリックス(Sigma)を通過させた。20mMトリス(pH7.4)、150mM NaCl、1%NP40および20mMトリス(pH7.4)、150mM NaClで順次洗浄することによって、未結合蛋白質をマトリックスから除去した。結合蛋白質を100mMグリシン(pH2.75)でカラムから溶出させ、0.5mLの画分に収集した。50μLの1Mトリス(pH9)の添加を通じて、画分を迅速に中性pHとした。蛋白質の含有量は(ペルオキシダーゼコンジュゲーテッド抗−FLAG M2)でのウエスタンブロッティング、クーマシーブリリアンントブルー染色によって分析した(図30)。
リポ多糖(LPS)(15ng/mL)を種々の濃度の本発明による精製されたキメラTLR4/MD−2と共にプレインキュベートし、引き続いて、TLR4/MD−2トランスフェクトされたHEK293細胞と共にインキュベートした。図31は、処理から24時間後における細胞培養基でのIL−8の産生を示すグラフである。
本発明をその詳細な記載に従って記載してきたが、これまでの記載は例示的なものであって、添付の請求の範囲の範囲によって規定される本発明の範囲を限定するものではない。他の態様、利点、および修飾は請求の範囲の範囲内のものである。
Claims (10)
- ヒトToll様レセプター4(TLR4)/ヒトMD−2複合体に免疫特異的に結合する単離されたモノクローナル抗体であって、ここで該モノクローナル抗体は、該モノクローナル抗体を10μg/mlの濃度において、ヒトTLR4/MD−2をトランスフェクトされたHEK293細胞を含む溶液に加えた場合に、該溶液におけるリポ多糖(LPS)誘導IL−8産生を、該モノクローナル抗体の非存在下でのLPS誘導IL−8産生のレベルと比較して少なくとも50%阻害し、該抗体は、以下:
(i)配列番号3のアミノ酸配列を含む可変重鎖相補性決定領域1(VH CDR1)、配列番号4のアミノ酸配列を含む可変重鎖相補性決定領域2(VH CDR2)、配列番号5のアミノ酸配列を含む可変重鎖相補性決定領域3(VH CDR3)を含む重鎖、および
配列番号8のアミノ酸配列を含む可変軽鎖相補性決定領域1(VL CDR1)、配列番号9のアミノ酸配列を含む可変軽鎖相補性決定領域2(VL CDR2)、配列番号10のアミノ酸配列を含む可変軽鎖相補性決定領域3(VL CDR3)を含む軽鎖;
(ii)配列番号13のアミノ酸配列を含むVH CDR1、配列番号14のアミノ酸配列を含むVH CDR2、配列番号15のアミノ酸配列を含むVH CDR3を含む重鎖、および
配列番号18のアミノ酸配列を含むVL CDR1、配列番号19のアミノ酸配列を含むVL CDR2、配列番号20のアミノ酸配列を含むVL CDR3を含む軽鎖;
(iii)配列番号23のアミノ酸配列を含むVH CDR1、配列番号24のアミノ酸配列を含むVH CDR2、配列番号25のアミノ酸配列を含むVH CDR3を含む重鎖、および
配列番号28のアミノ酸配列を含むVL CDR1、配列番号29のアミノ酸配列を含むVL CDR2、配列番号30のアミノ酸配列を含むVL CDR3を含む軽鎖;ならびに
(iv)配列番号33のアミノ酸配列を含むVH CDR1、配列番号34のアミノ酸配列を含むVH CDR2、配列番号35のアミノ酸配列を含むVH CDR3を含む重鎖、および
配列番号38のアミノ酸配列を含むVL CDR1、配列番号39のアミノ酸配列を含むVL CDR2、配列番号40のアミノ酸配列を含むVL CDR3を含む軽鎖
からなる群より選択される重鎖および軽鎖の組み合わせを含む、抗体。 - ヒトMD−2蛋白質がヒトTLR4に複合体化していない場合、前記抗体が該ヒトMD−2蛋白質に免疫特異的に結合しない、請求項1に記載の抗体。
- 前記抗体がヒト化抗体である、請求項1に記載の抗体。
- 前記抗体が、以下:
(i)配列番号2のアミノ酸配列を含む重鎖可変領域と配列番号7のアミノ酸配列を含む軽鎖可変領域、
(ii)配列番号12のアミノ酸配列を含む重鎖可変領域と配列番号17のアミノ酸配列を含む軽鎖可変領域、
(iii)配列番号22のアミノ酸配列を含む重鎖可変領域と配列番号27のアミノ酸配列を含む軽鎖可変領域、および
(iv)配列番号32のアミノ酸配列を含む重鎖可変領域と配列番号37のアミノ酸配列を含む軽鎖可変領域
からなる群より選択される重鎖可変アミノ酸配列と軽鎖可変アミノ酸配列の組み合わせを含む、請求項1に記載の抗体。 - 異常なTLR4シグナル伝達に関連する病理の症状をそのような軽減が所望される被験体において軽減するための組成物であって、該組成物は、請求項1に記載の抗体を含み、該組成物は、該被験体において該病理の症状を軽減するのに十分な量にて投与されることを特徴とし、該病理は、敗血症、ベンチレーター誘導肺損傷、急性炎症、慢性炎症、自己免疫疾患および内因性可溶性ストレス因子によって誘導される障害からなる群より選択され、ここで、該内因性可溶性ストレス因子によって誘導される障害は、変形性関節症または慢性関節リウマチであり、そして、該内因性可溶性ストレス因子が、Hsp60、フィブロネクチン、ヘパラン硫酸、ヒアルロナン、gp96、β−ディフェンシン−2および界面活性剤プロテインAからなる群より選択される、組成物。
- 前記被験体がヒトである、請求項5に記載の組成物。
- 前記異常なTLR4シグナル伝達に関連する病理の症状を軽減するのに十分な前記組成物の量が、LPS誘導IL−8産生を低下させるのに十分な量である、請求項5に記載の組成物。
- 前記慢性炎症が、アレルギー状態、または喘息に関連する、請求項5に記載の組成物。
- 前記病理が、炎症性腸障害またはアテローム性動脈硬化症である、請求項5に記載の組成物。
- 請求項1に記載の抗体とキャリアとを含む、薬学的組成物。
