高濃度のシトルリン及びアルギニンが、リンゴ酸脱水酵素(MDH )及び乳酸脱水素酵素(LDH )の活性に及ぼす阻害の程度を調べることで、シトルリンの安全性をアルギニンと比較した。アルギニンのイオン対としては塩化物イオンを用い、その塩化物をイオンの影響を評価するために塩化カリウムの検定も行った。
シトルリンとヒドロキシルラジカルとの反応速度を解析し、活性酸素スカベンジャーとしてのシトルリンの機能の評価を行った。ヒドロキシルラジカルは、アスコルビン酸−過酸化水素系により活性させ、サリチル酸の水酸化をラジカル検出の指標として用いた。反応液組成は、40mM K-Pi バッファー、pH7.4, 0.26mMascorbate, 0.15mM FeEDTA, 0.6mM H2O2, 2mM salicylateと種々の適合溶質を含み、全量を400 μL とした。25℃で90分間反応させた後、サリチル酸とヒドロキシルラジカルとの反応産物である2,3-Dihydroxy-benzoic acidを誘導体化して発色させ、510nm の吸光度により定量した。その結果、シトルリンとヒドロキシルラジカルとの二次反応速度定数は、競争反応理論により算出した。基準反応速度定数として、マンニトールの文献値1.7x109M-1 s-1を用いた。
シトルリンが、ヒドロキシルラジカルから酵素や核酸等の生体分子を保護する作用を示し得るかどうかにつき検討を行った。即ち、アスコルビン酸−過酸化水素系により発生したヒドロキシルラジカルが、ピルビン酸キナーゼ(PK)の酵素活性に及ぼす影響を検討した。失活反応組成液は、100mM Tris-HClバッファーpH7.4, 2.5U ブタPK(オリエンタル社), 0.2mM ascorbate, 0.15mM FeEDTA, 0.6mM H2O2と種々の濃度のシトルリンを含む様に調製し、全量を250 μL とした。失活反応は25℃でH2O2を添加することにより開始し、一定時間毎に10μL を分取して酵素の残存活性を測定した。活性測定反応液組成は、80mM Tris-HCl バッファーpH7.4, 7.5mM MgCl2, 75mM KCl, 3.75mM ADP, 0.15mM NADH, 10UブタLDH, 0.8mM PEP及び10μL 失活処理済PK酵素を含む様に調製した。反応は、25℃で基質を添加することにより開始し、NADHの増減に伴う340nm の吸光度変化によりその初速度を測定した。シトルリンが、ピルビン酸キナーゼのヒドロキシルラジカルによる失活に及ぼす影響を図5に示す。図5において、○はシトルリン無添加、●はシトルリン200mM 添加、▲はシトルリン400mM 添加、□は活性酸素なしを、それぞれ示す。

- 200mMから600mMの濃度のシトルリンを有効成分として含有することを特徴とする、抗酸化食品添加物。
