れ、遊走している樹状細胞が流入領域リンパ節の皮質傍T細胞リッチ領域への正確な経路をたどることを可能にする(Banchereau adn Steinman 1998 Nature 392:245-252)。常在しているそれらの組織から局所リンパ節へと遊走するように誘導されると、成熟な抗原保有樹状細胞は、リンパ球の循環プールに存在する抗原特異的ナイーブT細胞に遭遇するように配置される。

Med. 183:485-497)。影響を受けやすいマウス系統では、Th1細胞は、抗原に対して寛容である一方で、Th2細胞は、Il−4を分泌するように誘導される。他の系統では、抗原特異的非応答性が、両方のCD4+Tヘルパー細胞サブセットで誘導される。低用量の可溶性タンパク質により誘導され、低ゾーン寛容性と称されるこのタイプの応答(In Immunobiology, 4th edition 1999 Janeway, C.A., Travers, P., Walport, M.,
and J.D. Capra, eds. Garland Publishing, New York, N.Y. p.37)は、非防御免疫性を生じることができ、慢性感染と関連することが多い。さらに、マウスの皮膚中の毛包の密度はヒトの皮膚中の毛包の密度より約10倍高いことから、毛包を介して侵入する抗原に依存する局所免疫化手法は、マウスの場合よりもヒトの場合の方が、有効性が低い可能性が高い。
胞およびB細胞が発達していることが明らかになった。
腫瘍細胞E.G7−OVAは、ニワトリオボアルブミン(OVA)の完全遺伝子でトランスフェクトされたC57BL/6 EL4胸腺腫である。トランスフェクトした細胞系は、OVAタンパク質を発現し、OVAタンパク質はまた、E.G7−OVA細胞によりSIINFEKLを有する8つのアミノ酸ペプチド、OVA257-264へと自然にプロセシングされる。このペプチドは、Kb:SIINFEKLのような、MHCクラスI分子Kbとの複合体で原形質膜上に発現されるようになり、ここでこのペプチドが、in vitroおよびin vivoの両方で、CD8+細胞障害性T細胞(CTL)の腫瘍関連ペプチド抗原として機能することが実証された(Celluzzi et al. 1996 J. Exp. Med. 183:283)。FITCを用いた陽性結果に基づいて、本発明者等は、十分に特性化された合成SIINFEK腫瘍関連ペプチドの局所投与により、E.G7−OVA腫瘍に対する免疫性を誘導する試みを、類似のプロトコルに従って行った。
6日までに減少した。したがって、濃DMSO中の腫瘍特異的ペプチドSIINFEKLは、表皮ランゲルハンス細胞上でMHCクラスI分子Kbに組み込まれるのに十分な濃度で角質層を浸透し、遊走誘導物質フタル酸ジブチルの存在下において、流入領域リンパ節においてCD8+CTLに効率的に提示される。
本発明による抗原または1または複数のエピトープは、小ペプチド、タンパク質、または非ペプチド免疫原性化合物であり得る。抗原が、非経口または他の非局所投与経路により哺乳動物に導入される場合、抗原の大きさは開示する免疫化方法について制限を加えない。同様に、抗原が、感染性細胞(ウイルス、細菌、寄生虫等)により哺乳動物に導入される場合、抗原の特徴は制限を加えない。同様に、免疫化が、抗原性ペプチドまたはタンパク質をコードする核酸の移入を包含する場合、抗原の特徴自体は送達経路を制限しない。本発明の一形態によれば、抗原は、内因性であってもよく、例えば腫瘍抗原であり得る。
MHC分子と優先的に結合するペプチドは、最終的に約13〜17アミノ酸残基長である。実際に、MHC分子と直接相互作用することが可能な幾つかの抗原ペプチドが既知であり、標準的な技法により合成することができる。
誘導様式、続いておこるMHCおよびMHC様依存性経路による抗原提示は、別個にあるいは組み合わせて、開示する本発明の特徴を例示する。各種供給ソースからの抗原の選択に制限が課せられないという事実は、抗原特異的免疫性または多くの抗原に対する寛容性を誘導する基本的な方法の使用と一貫している。したがって、特定の抗原およびその供給ソースの選択は、作用に依存し、免疫学分野の当業者の範囲内である。
皮膚の外側層は、身体に外来物質が進入するのを阻止するように設計される。外側層は、ケラチノサイト細胞膜およびケラチンフィブリルの高密度層から形成されるため、角質層は、500Daを超える分子量を有する分子、特に親水性の分子のほとんどにとって非浸透性である。したがって、経皮投与される薬剤はほんの数種に過ぎない。かかる分子は通常、親油性の小分子である。ペプチドの経皮送達は、ペプチドが親水性かつ一般に高分子量であるために、好ましくない。数多くの研究が、ペプチドを、角質層を通過させることは非常に困難であると立証している(概説に関して、Steinstrasser and Merkle 1995 Pharm. Acta. Helv. 70:3-24)。ペプチド浸透を高め得る幾つかのアプローチの中には、親油性ベシクル、低周波超音波、エレクトロポレーション、イオン導入、および表皮内送達が包含される。これらを以下で考察する。
アセトン、アゾン、およびジメチルスルホキシド(DMSO)のような親油性溶媒は、角質を浸透することが知られている。上述のように、本発明者等は、DMSO中に溶解させたSIINFEKLのような小ペプチドが角質層を通過し、表皮の低層に侵入することを見出した。したがって、抗原浸透エンハンサに関する本発明の開示は、DMSO、ならびに対称または非対称スルフィドおよびスルホキシドのような様々な親油性溶媒(ここで、アルキル基は、1〜16個の炭素原子を含有する)、ならびにリポソームを包含する。
Mitragotri等は、インスリン(6,000Da)、IFN(17,000Da)、およびエリスロポイエチン(48,000Da)のような巨大タンパク質分子でさえも、低周波超音波を用いて角質層を通過させることができると報告した(Mitragotri et al. 1995 Science 269:850-853、参照により本明細書に援用される)。超音波は、非熱的な空洞化現象(cavitational)効果により皮膚上で作動し、角質層で拡大および接触する微小気泡を創出し、浸透性の一時的な増加をもたらす。空洞化現象は、超音波の周波数が低いほど増加し、診断画像化に使用される周波数(2〜12Mhz)よりも低周波の音波が最適である。
Vanbever等は、小(267Da)分子メトプロロールの約10%が、5回の単一450Vパルス後の4時間の間に皮膚を通って輸送され得ると開示した(Vanbever et al. 1994 Pharm. Res. 11:1000-1003、参照により本明細書に援用される)。エレクトロポレーションは、細胞にDNAを入れる目的で、細胞膜を浸透処理するのに広範に使用されている。ペプチドの経皮輸送は、高強度の電場パルスへの皮膚の限局性暴露により行われてもよく、脂質二重層に一過性の水性孔を形成し得る。
Bodde等は、1時間当たり約0.4%だけのバソプレシン(1,084Da)が角質層を通過することを見出した(Bodde et al. 1990 Biochem. Soc. Trans. 17:943-945、参照により本明細書に援用される)。イオン導入は、低強度の電流に皮膚を暴露することを含む。極性分子は、電流に応答して移動し、輸送は、表皮を通過して真皮へと突出する腺または毛包を介して行われると考えられる。この方法は、本発明に包含される。
鋭利な器具を用いれば確実に、角質層に浸透させることができる。したがって、伝統的な勾配付き針およびシリンジを用いた表皮内または真皮内注射が可能である。同様に、尖叉試験を行うのに使用される器具のような先分かれの器具を用いて、表皮に抗原性作用物質を導入することも可能である。抗原はまた、針のない注射システムで使用される高圧液体または気体により、表皮へと噴射させることもできる。かかる侵襲性の物理的な方法は、浸透の問題を解決し、さらにはランゲルハンス細胞に付随する遊走性シグナルを提供し得る。
表皮において抗原とランゲルハンス細胞とを効率的に接触させることは免疫応答を誘導するのに必要であり得る一方で、それだけではワクチン接種手順に所望の強力な抗原特異的免疫性を誘導するのに十分ではない。表皮からリンパ節へランゲルハンス細胞が遊走する現象は長い年月の間知られているが、in vivoでのランゲルハンス細胞の遊走および成熟を制御する複雑な調節経路は完全には理解されていない。さらに、ランゲルハンス細胞の遊走に関して一般的に多数の誤認が存在するようである。例えば、自発的および/または連続的なランゲルハンス細胞の遊走が、抗原特異的免疫性の誘導に十分であると考えている研究者もいる(例えば、Matsuno et al. 1990 J. Exp. Med. 183:1865を参照)。この分野には、接触感作用抗原が、それ単独でランゲルハンス細胞遊走を誘導することが可能であり、したがって別個の誘導刺激は任意の抗原に必要でない場合があると提案する研究者もいる(例えば、Udey 1997 Clin. Exp. Immunol. 107:6を参照)。したがって、ランゲルハンス細胞の遊走を研究するために、アセトンおよびフタル酸ジブチル中のFITCを用いた科学者等は、遊走を誘導するのは抗原(FITC)であると示唆している(例えば、Wang et al. 1996 Immunol. 88:284を参照)。

25mg/mlの用量範囲内で投与され得る。皮膚の局所投与は、0.01〜1ml/cm2、好ましくは約0.05〜0.5ml/m2の塗布容量を含み得る。明らかに、処置される表面積が大きいほど、流入領域リンパ節に遊走するように誘導されるランゲルハンス細胞の数は多くなる。投与経路は、皮膚、膣内、直腸、気道および肺へのエーロゾル送達、ならびにGI管への投与を含む、任意の表皮および/または粘膜部位から選択され得る。ランゲルハンス細胞の遊走の化学的誘導物質が適用可能である場合、化学的誘導物質は、ニートで、あるいは約1:1000〜約9:1(誘導物質対溶媒)の比の範囲で、好ましくは約1:100〜約7:3の比の範囲で、アセトンのような有機溶媒と混合して付与されてもよい。
特異的接着分子の膜発現または機能の減少をもたらすその能力により同定する方法である。
様々なワクチン配合物および送達方法の免疫学的潜在能力を評価するために、細胞膜上で発現される特異的ペプチド:MHC複合体の数を定量的に決定することができる抗原系を使用した。今日では、生細胞上で発現され、かつT細胞に認識される抗原エピトープを定量化する唯一の直接的な方法は、個々のペプチド:MHC複合体に特異的なモノクローナル抗体(mAbs)を使用する方法である。ニワトリオボアルブミン(OVA)は、免疫学的研究のプロトタイプの実験抗原として長い年月の間使用されている40kDaのタンパク質である。OVA257-264ペプチドであるSIINFEKL(963Da)はプロセシングされて、MHCクラスI分子Kbのペプチド結合溝に提示される。mAbである25−D1.16は、SIINFEKL:Kb複合体に対して特異性を有する(Porgador et al. 1997 Immunity 6:715-726)。
図9は、樹状細胞遊走および成熟の誘導を最大限にするための、超音波、続いてフタル酸ジブチルおよびアセトンの局所塗布により皮膚へペプチド抗原(SIINFEKL)を送達する効果を示す。DMSO中のSIINFEKL(240μg)、またはDMSO単独を、C57BL/6マウスの剃毛した腹部皮膚に塗布して、ヒドロゲルで被覆して、そこに直径13mmの超音波トランスデユーサを挿入した。20kHzおよび1.6W/cm2にて、2秒毎に1秒パルスで6分間、総超音波用量288J/cm2で超音波を印加した。2時間後、アセトンとフタル酸ジブチルの50:50v/v溶液を同じ区域に局所塗布した。2日後、皮膚排出鼠径リンパ節を取り出し、免疫蛍光フリーサイトメトリーにより細胞を分析した。
1998 Nature 392:245-252)、および表皮から遊出するように誘導されたランゲルハンス細胞は、リンパ節への侵入に関して機能的に成熟しており、それらに収められた抗原に対してナイーブT細胞を活性化することが可能であるということ(Udey 1997 Clin. Exo. Immunol. 107 (Suppl. 1)6-8)は、免疫学の文献で十分に確立されている。
の交差反応性を表す(Porgador et al. 1997 Immunity 6:715-726)。それにも関わらず、超音波媒介性のSIINFEKLへの局所暴露後にリンパ節へ遊走するように誘導されたランゲルハンス細胞(図9Bおよび表1)は、このmAbと反応性を有する細胞数(13,216個)の5倍に相当した。これらの結果は、樹状細胞の遊走および成熟の化学的誘導と組み合わせた抗原送達のこの局所方法が、流入領域リンパ節中に多数の抗原保有樹状細胞を生じさせるという量的証拠を提供する。
図10は、A)ハンクス平衡塩溶液(HBSS)20μl、B)HBSS中のニワトリオボアルブミン(OVA)の0.05m溶液20μl、またはC)HBSS中のOVA257-264ペプチド(SIINFEKL)の0.5mM溶液20μlを、マウスの剃毛した腹部皮膚における2つの対側性部位間に均等に分割して注射した場合の効果を示す。マウスの腹部皮膚における細胞外液腔は、約10μlを超えて含有することができないようであり、そこで、1部位に付き、OVAタンパク質200μg、またはSIINFEKLペプチド4.8μgのみを注射した。しかしながら、これらの注射が皮膚に限定されたかどうかは明らかでない。注射の2時間後に、フタル酸ジブチルおよびアセトンの局所塗布を腹部皮膚に行った。2日後に流入領域リンパ節に遊走した抗原保有樹状細胞を上述した手順により分析し、各ドットプロットの右上象限に示す。
日後に流入領域リンパ節に見られる抗原保有樹状細胞数がかなり増加(それぞれ、31倍および44倍)した。これは、注射した抗原のモル用量が、皮膚に塗布され、超音波により角質層を通して送達される抗原のたった2%である場合に注目すべきと思われる。この手順により、表皮から遊出するように誘導された未熟樹状細胞は、リンパ節へ至る途中で注射された抗原を捕捉することが可能になるようであった。しかしながら、この実験計画は、たとえあるとしても、遊走誘導物質の役割に取り組むものではなかった。
図11は、所望の結果を達成する際の樹状細胞の遊走/成熟の効果的な誘導物質を塗布する役割を示す。SIINFEKLペプチド抗原(総量HBSS150μl中144μg)を、マウスの剃毛した腹部皮膚中およびその皮膚を通して、2つの対側性部位で、分割用量で注射した。この2時間後に、A)さならる処置を施さない、またはB)フタル酸ジブチルおよびアセトンの局所塗布を、腹部皮膚に施した。2日後に流入領域リンパ節に遊走した抗原保有樹状細胞を、各ドットプロットの右上象限に示す。リンパ節1つ当たりの抗原保有移入樹状細胞の絶対数を表3に示す。
なく、かつ末梢樹状細胞の遊走を誘導することができないサブユニットワクチンが、臨床的に有用であるのに十分な免疫原性を欠如する場合がある理由を実証する。
上述のように、成熟抗原保有樹状細胞がリンパ節中でナイーブT細胞と遭遇し、ナイーブT細胞を活性化するかどうかは、節に侵入する抗原保有樹状細胞数だけでなく、抗原保有樹状細胞の膜上で発現されるMHC:ペプチド複合体の密度にも依存する可能性が高い。この手順により生成される移入樹状細胞上のペプチド:MHC複合体の密度を推定するために、本発明者等は、同一の染色条件およびフローサイトメトリー設定を用いて、ペプチド:MHC特異的mAbである25−D1.16により検出される様々な細胞集団の蛍光強度を比較した。結果を表4に示したが、表4は、in vitroまたはin vivoでの様々な処置後の樹状細胞の原形質膜上で発現される特異的ペプチド:MHC複合体の数を提供する。
b:mAb25−D1.16で染色したE.G7−OVA細胞から、25−D1.16の非存在下での同じ細胞集団の非特異的染色を減じた中央蛍光強度;あるいはin vitroでのランゲルハンス細胞の場合では、SIINFEKLとともにインキュベートし、かつ25−D1.16で染色した細胞の中央蛍光強度から、SIINFEKLの非存在下でインキュベートし、かつ25−D1.16で染色した細胞の中央蛍光強度を減じた中央蛍光強度、またはin vivoでの移入ランゲルハンス細胞の場合では、同じ試料中の陰性ランゲルハンス細胞の中央蛍光強度を減じた中央蛍光強度。
c:細胞1個当たりのSIINFEKL:Kb複合体の中央値は、同じ試薬および同じフローサイトメトリー設定を用いて、E.G7−OVA細胞上で検出される中央比蛍光強度から算出した。E.G7−OVA細胞上で発現されるSIINFEKL:Kb複合体の報告された数に基づいて、1蛍光強度単位=20SIINFEKL:Kb複合体。

Biotech. 18:1273-1278)に組み込まれたDNAの局所塗布(しかし、これらに限定されない)のような実験用ワクチン配合物の免疫原性および送達方法を高めることもできる。
表5は、リンパ節に続いて出現する樹状細胞上の細胞結合抗原の量に対する、樹状細胞の遊走/成熟の誘導物質の種々の組成物の効果を示す。使用する抗原はFITCであり、手順は図1で示した実験に使用した手順と同一とした。すなわち、5mg/mlのFITCを、アセトンと遊走誘導用の試験作用物質の50:50溶液中に溶解した。この実験で使用した試験作用物質は、フマル酸ジブチル(作用物質#1)、マレイン酸ジイソオクチル(作用物質#2)、およびD−酒石酸ジブチル(作用物質#3)とした。上記に詳述するように、FITC+およびFITC−樹状細胞の存在に関して、鼠径リンパ節を48時間後に検査した。

長または中止を達成または増大することができる。
図13のデータは、リンパ節への樹状細胞の遊走を有効に誘導した後に、同じリンパ節への非抗原保有樹状細胞の流入が長期にわたって続くことを実証する。この効果はまた、表1、表2および表3、ならびに米国特許出願第09/176,044号に示されるデータからも明らかである。樹状細胞の遊走および成熟の誘導の直後の流入領域リンパ節の細胞密度の増加(図3を参照)は、抗原特異的細胞分裂だけが原因であるとするとその効果が生じるのが迅速すぎると考えられるため、抗原特異的免疫系細胞の増大の結果というよりはむしろ、主に末梢血白血球の流入(Tedla et al. 1998 J. Immunol. 161:5663-5672)の結果である可能性がある。
表6に示すデータは、樹状細胞の遊走および成熟の効果的な誘導を、塗布後の初期の14日目程度に同一部位に塗布を繰り返すことにより、応答を完全に回復することができることを実証する。使用する抗原は、FITCであり、それを、フタル酸ジブチルとアセトンの50:50溶液中で、5mg/mlで、C57BL/6マウスの剃毛した腹部皮膚に局所塗布した。塗布はそれぞれ同一であり、各群のマウスの半分に第1の塗布のみ(なし)を施し、半分は、第1の投与後に指定間隔(14日または21日)で第2の塗布を施した。上記に詳述したように、各処置の48時間後に、FITC+樹状細胞に関して、鼠径リンパ節細胞を、フローサイトメトリーで検査した。
単一投与は、適切な時間間隔で、樹状細胞の遊走および成熟を繰り返し誘導することで達成することができる。
Claims (7)
- 抗原、またはその一もしくは複数のエピトープ、および親油性分子を含む抗原の免疫原性を高めるための医薬であって、
抗原、またはその一もしくは複数のエピトープは、注射により投与され、親油性分子は、局所塗布により投与され、
親油性分子は、フタル酸ジブチル、D−酒石酸ジブチル、N,N−ジエチル−トルアミド、フマル酸ジブチル、フマル酸ジ(2−エチルヘキシル)、マレイン酸ジイソオクチル、マレイン酸ジエチルヘキシル、フマル酸ジイソオクチル、安息香酸、マレイン酸ビフェニル、フタル酸ジオクチル、マレイン酸ジブチル、マレイン酸ジオクチル、コハク酸ジブチル、コハク酸ジオクチル、フタル酸ジノニル、フタル酸ジイソノニル、フタル酸ジメチル、フタル酸ジエチル、フタル酸ジプロピル、フタル酸ジフェニル、フタル酸ジベンジルブチル、およびフタル酸ジエチルメチルからなる群から選択される、医薬。 - 前記親油性分子は油/水分配係数が1より大きい、請求項1に記載の医薬。
