【従来の技術】
う蝕(虫歯)の原因となる歯垢は、ストレプトコッカス・ミュータンスを中心とする口腔内細菌により形成される。ストレプトコッカス・ミュータンスはデキストラン合成酵素であるグルコシルトランスフェラーゼを菌体外に産生し、ショ糖を基質として粘着性多糖である不溶性グルカンを合成する。この不溶性グルカン中で口腔内細菌が繁殖して歯垢が形成される。また、ストレプトコッカス・ミュータンス等の口腔内細菌は種々の糖を利用して産生する酸が、エナメル質表面を脱灰し、う蝕を進行させる。従ってストレプトコッカス・ミュータンスによる歯垢を形成させないことが、虫歯予防にとってはきわめて重要なポイントとなる。近年、様々な抗う蝕剤が提案されているものの、う蝕の予防は充分ではなく、実際に利用しうる効果的な抗う蝕剤は未だ見出されていないのが現状である。

歯垢を形成させない方法として、ストレプトコッカス・ミュータンスをの増殖を抑制するための殺菌剤、グルコシルトランスフェラーゼによる不溶性グルカン合成の阻害剤、形成された不溶性グルカンを分解する酵素等の様々な抗う蝕剤が研究されている。中でも、天然物由来物質は、比較的安全性が高いため研究対象として注目されている。
例えば、特解昭58−121218号公報には、抗う蝕剤が開示されており、具体例としてタイソウ、ウイキョウ、芍薬、黄連、オウゴン、センブリ等が挙げられている。しかしながら、その効果は、期待されているものとはほど遠く、実際に利用しうる効果的な抗う蝕剤は未だ見出されていないのが現状である。

本発明の目的は、ストレプトコッカス・ミュータンスによる歯垢形成を阻害することのできる優れた抗う蝕剤を提供し、さらに、歯垢形成を予防することのできる口腔用組成物を提供することである。

【課題を解決するための手段】
本発明者は、上記の目的を達成するために鋭意研究を重ね、種々の植物抽出液をスクリーニングして、ストレプトコッカス・ミュータンスによる歯垢形成を効果的に阻害する天然由来の抽出物を探索した。その結果、ユッカの抽出物がこれにたいして強力な阻害作用を有してう蝕の予防に有用であることを見出し、本発明を完成するに至った。
従って本発明は、ユッカ抽出物を含有することを特徴とする抗う蝕剤である。
本発明はさらに、上記抗う蝕剤を含有していることを特徴とする口腔用組成物である。

【発明の実施の形態】
以下、本発明を詳細に説明する。
原料のユッカ(Yucca)としては、Yucca arizonica、Yucca brevifolia、Yucca schidigera、Yucca elata、Yucca intermdia、Yucca recurvifolia Salisb、Yucca smalliana Fernald、Yucca aloifolia Linn、Yucca gloriosa Linn、Yucca mohavensis、Yucca peninsularis、Yucca schottii、Yucca whipplei等があり、これらの一種を用いてもよいし、二種以上を用いてもよい。
ユッカは地上茎、地上部(種子を含む)とも抽出原料として使用することができる。原料は適宜適当な大きさに粉砕してから、抽出溶媒として水、親水性有機溶媒又はこれらの混合物で抽出処理する。水と親水性有機溶媒との混合物を使用する場合、水の含有量は全体の70容量%程度までが適当である。抽出用の親水性有機溶媒としては、メタノール、エタノールなどの低級アルコール、グリセリン、1,3−ブチレングリコール、プロピレングリコールなどの多価アルコール、アセトン等が好ましい。抽出溶媒の使用量は、重量で抽出原料の3〜10倍量程度が適当である。

本発明の口腔用組成物は、練歯磨、潤製歯磨、液体歯磨等の歯磨剤、洗口液、マウスウォッシュ、口中清涼剤、うがい用錠剤、義歯用洗浄剤、チューインガム等の形態とすることができる。これらは、上記抗う蝕剤を含有してなるものであるが、その特徴に応じ他の成分を本発明の効果を損ねない範囲で配合し、通常の方法で調製することができる。

<歯垢付着抑制実験>
ストレプトコッカス・ミュータンスの試験管壁への付着抑制効果を下記の方法で評価した。結果を表1に示す。
ストレプトコッカス・ミュータンス菌6715株(gタイプ)をブレインハートインフュージョン液体培地(BHI培地)で37℃,24時間培養して得られた菌液0.1mlと5%ショ糖添加BHI培地4.5mlおよびユッカ抽出物あるいは滅菌精製水0.5mlを試験管内で混合し検液とし、30°の角度に傾け37℃、18時間培養した。なお、上記検液において、ユッカ抽出物濃度を0.2〜5.0%で変動させた。

<Plate adhesion suppression experiment>
The effect of inhibiting the adhesion of Streptococcus mutans to the test tube wall was evaluated by the following method. The results are shown in Table 1.
Streptococcus mutans 6715 strain (g type) cultured in brain heart infusion liquid medium (BHI medium) at 37 ° C. for 24 hours, 0.1 ml of bacterial solution and 4.5 ml of BHI medium supplemented with 5% sucrose Then, 0.5 ml of yucca extract or sterilized purified water was mixed in a test tube to prepare a test solution, which was incubated at 37 ° C. for 18 hours at an angle of 30 °.
