Table 3 shows the results of TNSALP values obtained using monoclonal antibodies 3-29-3R and 1A5-7. These results were compared with the total ALP activity value (JSCC compliant value).

実施例1
ELISA 法による ALP 6-IgA 値とALP 6-IgG 値、及び Disk 電気泳動による ALP6 測定の感度等の比較
Disk 電気泳動で ALP 6 が 陽性だったヒト血清 2検体 80-21, 81-23 を用い、ELISA 法による ALP 6-IgA 値と ALP 6-IgG 値を求めた。次いで、それらの測定法と Disk 電気泳動による ALP 6 測定法の感度等を比較した。以下に詳述する。
ELISA 法は、一次抗体として 3-29-3R プレートを用い、免疫グロブリン結合物質である二次抗体として、1/200 希釈 HRP 標識抗ヒト IgA モノクローナル抗体及び IgG モノクローナル抗体(ザイメッド社)を用いた。まず、抗体 3-29-3R プレートを、0.05% Tween 20 を含む 100 mMトリス緩衝液(pH 7.5)200μL で 1 回洗浄した。次いで、これらのプレートに、ALP 6 陽性検体を1倍、10倍、20倍、40倍、80倍、希釈し、各検体 50 μL と100 mM トリス緩衝液(pH 7.5)50μL を加えて室温、1 時間反応させた。一次抗体反応終了後、そのプレートを、0.05% Tween 20 を含む 100 mMトリス緩衝液(pH 7.5)200μL で 3 回洗浄し、100μl の二次標識抗体 HRP 標識抗ヒト IgA モノクローナル抗体又は IgG モノクローナル抗体(ザイメッド社)を加えて更に 室温、1 時間反応させた。1 時間後に洗浄を行い、標識 HRP に対する基質溶液(OPD 溶液)を反応させ、一定時間後に発色停止試薬 1 N 硫酸 100μL を添加してその発色値を 492 nm にて比色し ELISA 法による ALP 6-IgA 値及び ALP 6-IgG 値を求めた。
一方、Disk 電気泳動は ALP アイソザイム検出キット「アルフォー」(常光)を用いた。検体として ELISA 法で使用したものと同一の ALP 6 陽性患者検体を1倍、10倍、20倍、40倍、80倍、希釈し、各 25μL 使用した。操作方法は ALPアイソザイム検出キット「アルフォー」(常光)の能書に従って実施した。
Example 1
Comparison of ALP 6-IgA and ALP 6-IgG values by ELISA and sensitivity of ALP6 measurement by disk electrophoresis
ALP 6-IgA and ALP 6-IgG values were determined by ELISA using two human sera 80-21 and 81-23 that were positive for ALP 6 by disk electrophoresis. Next, the sensitivity of the ALP 6 measurement method by disk electrophoresis was compared with those measurement methods. This will be described in detail below.
In the ELISA method, a 3-29-3R plate was used as a primary antibody, and a 1 / 200-diluted HRP-labeled anti-human IgA monoclonal antibody and IgG monoclonal antibody (Zymed) were used as secondary antibodies that are immunoglobulin binding substances. First, the antibody 3-29-3R plate was washed once with 200 μL of 100 mM Tris buffer (pH 7.5) containing 0.05% Tween 20. Next, ALP 6 positive specimens are diluted 1-fold, 10-fold, 20-fold, 40-fold, 80-fold to these plates, and 50 μL of each sample and 50 μL of 100 mM Tris buffer (pH 7.5) are added at room temperature. The reaction was performed for 1 hour. After the primary antibody reaction, the plate was washed three times with 200 μL of 100 mM Tris buffer (pH 7.5) containing 0.05% Tween 20, and 100 μl of secondary labeled antibody HRP-labeled anti-human IgA monoclonal antibody or IgG monoclonal antibody ( Zymed) was added, and the mixture was further reacted at room temperature for 1 hour. After washing for 1 hour, react with the substrate solution (OPD solution) for labeled HRP. After a certain time, add 100 μL of color stop reagent 1 N sulfuric acid and colorize the color value at 492 nm. -IgA and ALP 6-IgG values were determined.
On the other hand, Disk electrophoresis used ALP isozyme detection kit “Alfour” (Joko). The same ALP 6 positive patient specimens used in ELISA as specimens were diluted 1-fold, 10-fold, 20-fold, 40-fold, 80-fold, and 25 μL each was used. The operating method was carried out according to the ALP isozyme detection kit “Alfour” (Joko) 's document.
電気泳動の結果を図1に、ELISA の結果を図2に示す。結果として、電気泳動ではサンプルとして検体原液を用いたもののみ ALP 6 が確認されたが、ELISA 法では検体が 40〜80倍希釈されたところまで数値が確認された。電気泳動と比較すると ELISA 法では、ALP 6-IgA が 40〜80倍の感度で検出されることが判明し、良好な希釈直線性が認められた。一方、 ALP 6-IgG はこれにおよばなかった。このことから、ELISA 法ではこれまで電気泳動法では検出できなかった微量の ALP 6-IgA の検出と定量ができることが判明した。 The result of electrophoresis is shown in FIG. 1, and the result of ELISA is shown in FIG. 実施例2
乗法値法による ALP 6-IgA 測定による虚血性腸疾患の診断
一次抗体として抗体 3-29-3R を用い、二次抗体としてHRP 標識抗 IgA モノクローナル抗体及び IgG モノクローナル抗体を用いて、ELISA 法により検体中の ALP 6-IgA 値及び ALP 6-IgG 値、また、乗法値による検体中の ALP 6-IgA 値及び ALP 6-IgG 値を測定して、虚血性腸疾患の診断に適用できるかどうかを検討した。検体としては、インフォームド・コンセントを行った健常人ボランティア検体 69 検体及び虚血性腸疾患患者血清 98 検体を用いた。ELISA 法による ALP 6-IgA 値及び ALP 6-IgG 値は、実施例1と同様な方法により求めた。また、TNSALP 値をモノクローナル抗体 3-29-3R を用いて参考例2の記載の方法により測定した。乗法値による ALP 6-IgA 値又は ALP 6-IgG 値は、「ELISA 法による ALP 6-IgA 値又はALP 6-IgG 値」と「TNSALP 値」との乗法値を計算して求めた。
ELISA 法による ALP 6-IgA 値及び ALP 6-IgG 値の平均値の結果を表4に示す。
Example 2

The ALP 6-IgA value or ALP 6-IgG value based on the multiplicative value was obtained by calculating the multiplicative value between the “ALP 6-IgA value or ALP 6-IgG value based on the ELISA method” and the “TNSALP value”.
Table 4 shows the results of the average values of ALP 6-IgA value and ALP 6-IgG value by ELISA method.
このALP 6-IgA 値で、健常人と虚血性腸疾患患者検体で平均値の差の検定を行ったところ、 t 検定における p 値 0.0006 で 2 つのグループには有意差がみとめられた。この結果により、2次抗体に HRP 標識抗ヒト IgA モノクローナル抗体を用いると虚血性腸疾患をより特異的に診断できることが判明した(陽性率 51%)。一方、ALP 6−IgG 値では、正常者と患者で、平均値が、ほとんど違いはなかった。
以上の結果から本発明の反応系例(一次抗体:ヒト TNSALP 認識モノクローナル抗体、二次標識抗体:HRP 標識抗ヒト IgA モノクローナル抗体)を用いて ALP 6-IgA を精密測定することが可能となり、しかも、それにより虚血性腸疾患診断の指標となりうることが判明した。

From the above results, it is possible to precisely measure ALP 6-IgA using the reaction system example of the present invention (primary antibody: human TNSALP-recognizing monoclonal antibody, secondary labeled antibody: HRP-labeled anti-human IgA monoclonal antibody). Thus, it has been found that it can be an index for diagnosis of ischemic bowel disease.

ii)上記 i)とは独立に、請求項6から8のいずれかの方法により検体中の IgA 結合性アルカリ性ホスファターゼ6のレベルを求め、
iii)上記 i)で得られるヒト臓器非特異型アルカリ性ホスファターゼ活性値と上記 ii)で得られる IgA 結合性アルカリ性ホスファターゼ6のレベル値との乗法値を求め、その乗法値から検体中の IgA 結合性アルカリ性ホスファターゼ6の量を求める、
