Description
動物の内分泌系で運ばれるステロイドホルモンは、一般に標的細胞内で核内受容体と結合し、標的遺伝子の発現制御を行うことが知られている。核内受容体は大きな遺伝子ファミリーを形成しており、保存されたドメイン構造(A/B, C, D, and E)を持っている。C ドメインはDNAへの結合に、E ドメインはステロイドホルモン(リガンド)との結合に関わっており、E ドメインに存在するAF-2 (Activation Function 2) はリガンド依存的に転写活性化を起こすのに必要である。核内受容体へのリガンドの結合により、AF-2とp160 ファミリー転写コアクチベーターとの相互作用が促進され、リガンド依存的に標的遺伝子の転写が活性化される。エストロゲン受容体は核内受容体ファミリーに属しており、エストロゲンとの結合により活性化され、標的遺伝子の転写を活性化する。
例えば、酵母を利用して、エストロゲン受容体のリガンド結合部位とコアクチベーターとの間のリガンド依存的な相互作用を利用したツーハイブリッド法を用いたエストロゲン様物質検出法が報告されている(非特許文献1)。しかし、この酵母ツーハイブリッド法は無菌操作が必要であり、抽出に用いる有機溶媒自体による環境の二次汚染が懸念される。
また、無菌操作の要らない植物(シロイヌナズナ)に、DNA結合部位、転写活性化部位及び核内受容体を組み合わせたキメラ転写因子を導入したシステムも考案されている(非特許文献2)。これは仕組みがシンプルであり好ましいが、バイオセンサーとしての感度の低いことが欠点である。
更に、リガンド依存性転写因子、コアクチベータ及びレポーター遺伝子を導入した形質転換体により、エストロゲンを検知する方法も提案されている(特許文献2)。
しかるに、本発明の方法においては、核内受容体とDNA結合部位を備えたエフェクター1に加え、コアクチベーターを改変して転写活性化ドメインを連結したエフェクター2を新たに導入することにより、本来エフェクター1の産物が相互作用するコアクチベーターとのリガンド依存的な相互作用(低濃度のリガンドの存在下で起こる現象)を利用してリガンドを検出できるようになり、センサーとしての感度を格段に向上させることができることを見出し、本発明を完成させるに至った。
また本発明は、この形質転換植物をエストロゲン様物質と接触させ、この植物における前記レポータ遺伝子の発現を検出することから成るエストロゲン様物質の検出方法である。
「プロモータ」としては、35Sプロモーターのような植物ウィルス由来のもの、ノパリンシンターゼプロモータのような植物関連細菌由来のもの、その他植物細胞内で機能する天然あるいは合成されたプロモーター等が挙げられる。
「核局在化シグナル」としては、SV40ウィルスのT抗原の核移行シグナルのように直接核への輸送に関わる蛋白質に認識されるポリペプチド配列、及び核移行シグナルを有する蛋白質を介して間接的に輸送されるのに必要なポリペプチド配列等が挙げられる。
「エストロゲン受容体のリガンド結合ポリペプチドをコードする領域」とは、真核生物由来のエストロゲンに結合活性を有するポリペプチド配列を含む領域である。
「標的DNAへの結合ポリペプチドをコードする領域とエストロゲン受容体のリガンド結合ポリペプチドをコードする領域とからなるキメラ遺伝子」は、これらを含むポリペプチド領域をコードするポリヌクレオチドを連結して形成される。
「プロモータ」及び「核局在化シグナル」は上記エフェクター1配列で定義した範囲から選択でき、上記エフェクター1配列で用いたものと同じであっても異なってもよい。
「転写コアクチベータの核内受容体相互作用領域」とは、NIDと呼ばれるポリペプチド領域で LXXLL(L=leusine) モチーフを持っているポリペプチド領域である。
「転写活性化領域」とは、塩基性アミノ酸を直鎖状に多く含むポリペプチド領域、グルタミンを直鎖状に多く含むポリペプチド領域などのように、基本的転写因子・転写装置と相互作用して転写を活性化するポリペプチド領域である。
「転写コアクチベータの核内受容体相互作用領域と転写活性化領域をコードするポリヌクレオチド領域とからなるキメラ遺伝子」はこれらを含むポリペプチド領域をコードするポリヌクレオチドを連結して形成される。
「レポーター遺伝子」としては、蛍光蛋白質、測定可能な活性を有する酵素、活性検出可能な酵素以外の蛋白質をコードするポリヌクレオチド領域などが挙げられる。
このレポーター遺伝子は標的DNAの下流に位置する。
本発明のように植物を検出に用いるとさまざまな利点がある。植物の場合、根から直接環境中の化学物質を吸収できるので、化学物質を試料から抽出精製する必要がなく、また、無菌操作が必要ない。さらに、種子は長期間、安定に保存ができ、保管のための特別な装置もいらないので、他のバイオアッセイ法と比べてセンサーの保存にコストがかからない。また、例えば、1シロイヌナズナ個体から約2000粒の種子が得られるため、大量生産による測定の低コスト化が可能である。
以下、実施例にて本発明を例証するが本発明を限定することを意図するものではない。
植物内で強力に下流の遺伝子の転写を制御するプロモーターであるカリフラワーモザイクウィルス(CaMV ) 35Sプロモーターにより、hERα LBDとLexA DBDを含むキメラエストロゲン受容体、及びTIF2 NIDとVP16 ADを含むキメラ転写コアクチベーターが形質転換シロイヌナズナ個体内で常時過剰に発現している。この2つのキメラタンパク質は、SV40ウィルス由来T抗原の核移行シグナルを含んでおり、常時核内に局在している。核内において、キメラエストロゲン受容体はレポーター遺伝子であるGUS遺伝子の上流に位置するLexA タンパク質の標的配列(OLexA )に結合している。エストロゲンがキメラエストロゲン受容体のhERα LBD部分と結合することにより、hERα LBDの立体構造の変化が起こり、キメラ転写コアクチベーターのTIF2 NID部分との相互作用が起こる。この相互作用によりTIF 2 NIDに融合した単純ヘルペスウィルス由来VP16 ADの転写活性化シグナルが植物の転写装置に伝わり、エストロゲン依存的にGUS遺伝子の転写及び翻訳が引き起こされる。形質転換植物におけるGUSタンパク質の酵素活性を測定することでエストロゲンを検出することができる。
キメラエストロゲン受容体をコードするエフェクター1遺伝子はカリフラワーモザイクウィルスの35Sプロモーター(P35S)の下流に、SV40ウィルスのT-抗原の核局在シグナル(NLS)・LexA DNA結合性ドメイン(LexA DBD)・ヒトエストロゲン受容体αのリガンド結合ドメイン(hERα LBD)を連結したポリペプチドをコードする遺伝子及びアグロバクテリウムのノパリン合成酵素遺伝子のターミネーター(TNOS)を連結した。
エフェクター1遺伝子を構築するために、P35SからNLSまでをコードする断片(P35S-NLS)、LexA DBDをコードする断片、hERα LBDをコードする断片、TNOSの断片をPCR反応によりそれぞれ増幅し調製した。
P35S-NLS 断片は 35S-NLS-GFP(静岡県立大学食品栄養科学部、静岡市谷田52番1号、丹羽康夫氏より分与していただいた。Curr Biol, 6: 325-30 (1996))を鋳型、5'-GGAAGCTTGCATGCTGCAGG-3'(配列番号1)及び5'-GGGCTAGCGACCTTTCTCTTCTTCTT-3'(配列番号2)をプライマーとしてPCR反応を行い、5' 側にHindIII部位、3' 側にNheI部位が来るように調製した。
hERα LBD 断片は、hER(GenBank: NM_0000125)の281〜323番目の43アミノ酸をコードする N 末端側断片と、324〜595番目の272アミノ酸と終止コドンをコードするC末端側断片に分けて増幅した。
両断片ともヒト卵巣 cDNA ライブラリーを鋳型とし、N末端側断片は5'-GGGGCCGGCCGTCTGCTGGAGACATGAGA-3'(配列番号7)及び5'-GGGCTCAGCATCCAACAAGGCACTGAC-3'(配列番号8)をプライマーとしてPCR反応を行い、5' 側にFseI部位、3' 側にBpu1102I部位が来るように調製した。
C 末端側断片は内部のHindIII部位を塩基置換により破壊し、5'-GGGCTGAGCCCCCCATACTCTATTCCGAGTATGATCCTACCAGACCCTTCAGTGAGGCTT-3'(配列番号9)及び5'-GGGCGGCCGCTCAGACTGTGGCAGGGAA-3'(配列番号10)をプライマーとしてPCR反応を行い、5' 側にBpu1102I部位、3' 側にNotI部位が来るように調製した。
上記の4つの断片をpBI221 (BD Biosciences Clontech, Palo Alto, CA, USA)のHindIII・EcoRI部位に挿入することによりエフェクター 1プラスミドを作製した。
エフェクター2遺伝子を構築するために、P35SからNLSまでをコードする断片、hTIF2 NIDをコードする断片、VP16 ADをコードする断片をPCRによりそれぞれ増幅し調製した。
hTIF2 NID断片はヒト肝臓cDNAライブラリーを鋳型とし、5'-GGGGATCCGAGAGAGCTGACGGGCAG-3'(配列番号15)及び5'-GGTCATGAAGTGCTCTGTGAAATTCG-3'(配列番号16)をプライマーとしてPCR反応を行い、hTIF2の624番目から869番目までの246アミノ酸をコードする領域を、5' 側にBamHI部位、3' 側にBspHI部位が来るように調製した。
上記の3つの断片をpBI221のHindIII-SacI部位に連続して挿入することにより、エフェクター 2プラスミドを作製した。
LexAの標的配列は、5'-GATCCATACTGTATGAGCATACAGTATACTGTATGAGCATACAGTA-3'(配列番号19)と5'-GATCTACTGTATGCTCATACAGTATACTGTATGCTCATACAGTATG-3'(配列番号20)をアニーリングさせ、BamHIとBglIIの制限酵素部位にはさまれたLexAの標的配列のダイマー断片を調製した。
これを4コピー縦列につなぎ、LexAの標的配列のオクタマー断片を、5' 側にBamHI部位、3' 側にBglII部位が来るように調製した。
HindIII-BamHI リンカーは、一本鎖オリゴDNA 5'-AGCTTGGACTAGAGCTTG-3'(配列番号23)と5'-GATCCAAGCTCTAGTCCA-3'(配列番号24)を二本鎖にアニーリングさせ、調製した。上記の3つの断片をpBI221(BD Biosciences Clontech, Palo Alto, CA, USA)のHindIII-SmaI部位に連続して挿入することにより、レポータープラスミドを作製した。
この断片を pGEM-T vector(Promega Corp., Madison, WI, USA)に挿入することにより、S/M IIプラスミドを作製した。
ツーエフェクタープラスミドをエレクトロポアレーション法でアグロバクテリウム C58 株に導入後、このアグロバクテリウムをシロイヌナズナに感染させることにより、形質転換を行った。形質転換処理をしたシロイヌナズナから得られた種子(T1 種子)を 1 % sucrose, 0.8 % 植物用寒天、50μg/ml カナマイシン、100μg/ml クラフォランを含む MS 寒天培地に播種し、導入遺伝子を持つ形質転換植物体の選抜を行った。カナマイシン耐性を持つT1個体からT2種子得た。また、T2個体からT3種子を得た。
T3の種子を17β-エストラジオール(WAKO Pure Chemical Inds.)を含む MS 寒天培地に播種した。1週間生育させた形質転換個体を用いて、GUS遺伝子の発現の様子をGUS活性染色法及びGUS活性測定法により調べた。
GUS染色法は、植物体全体を用いて行われ、GUSタンパク質が基質であるX-Glucを分解して得られるインディゴブルーの発色が、0.05 nM以上のエストロゲン濃度で形質転換個体の根の部分に観察された。GUS染色が観察された部分は直接寒天培地に接触していた部位であり、培地から植物体内にエストロゲンが拡散によって浸透した結果であると思われる。
T3 種子を1 nMの17β-エストラジオールを含むMS 寒天培地に播種し、3〜14日間生育させた。生育させた形質転換個体を回収し、GUS遺伝子の発現の様子をGUS活性測定法により調べた。その結果を図5に示す。
GUS遺伝子の活性は曝露日数を経るごとに減少していく様子が見られた。
形質転換後2世代目(T2)種子を異なる濃度のエストロゲン様物質であるジエチルスチルベステロール(DES)(ICN Biomedicals Inc. (CT, USA))、p-n-ノニルフェノール(NP)(WAKO Pure Chemical Inds.)、ビスフェノール A (BPA)(WAKO Pure Chemical Inds.)、ゲニステイン(EXTRASYNTHESE S. A. (Genay, France))を含むMS 寒天培地に播種し、1週間生育させた。形質転換個体における、GUS遺伝子の発現の様子をGUS活性測定法により調べた。その結果を図6(A)〜(D)に示す。

Claims (2)
- エフェクター1配列、エフェクター2配列、標的DNA及びレポーター遺伝子を導入することにより形質転換された植物であって、該エフェクター1配列がプロモーター、核局在化シグナル及び標的DNAへの結合ポリペプチドをコードする領域とエストロゲン受容体のリガンド結合ポリペプチドをコードする領域とからなるキメラ遺伝子を有し、該エフェクター2配列がプロモータ、核局在化シグナル及び転写コアクチベータの核内受容体相互作用領域と転写活性化領域をコードするポリヌクレオチド領域とからなるキメラ遺伝子を有し、該レポーター遺伝子が該標的DNAの下流に位置することを特徴とする形質転換植物。
- 請求項1に記載の植物をエストロゲン様物質と接触させ、該植物における前記レポータ遺伝子の発現を検出することから成るエストロゲン様物質の検出方法。
