【発明の属する技術分野】
本発明は、アミノ糖を分岐側鎖に有する分岐シクロデキストリン、その製造法およびその利用に関する。詳しくは、カルボン酸結合型シクロデキストリンとアミノ糖を酸アミド結合させたアミノ糖を分岐側鎖に持つシクロデキストリン誘導体とその製造方法並びにこのシクロデキストリン誘導体を用いた標的指向性薬物キャリヤー等としての利用である。
本発明により得られるシクロデキストリン誘導体は医薬品工業、化学工業の分野で広く利用される。

【従来の技術】
シクロデキストリン(以下、CDと略記することがある。)は、澱粉類に微生物酵素を作用させて得られる環状オリゴ糖で、グルコース分子がα−1,4結合で連なっており、グルコースが6,7,8個からなるα−,β−,γ−CDが良く知られている。
CDは、環の外部は親水性を示し、内部は疎水性を示す、きわめて特異な物質である。CDは、この空洞内に酸やアミンのような有極性イオン物質から無極性の脂肪族あるいは芳香族の炭化水素や希ガスまで、様々な物質を取り込んで包接複合体を形成することができる。
この包接機能を利用して、CDは環内に包接された物質の安定性の向上、物性の改変、異臭・異味除去、さらには難溶性物質の可溶化などを目的に食品、化粧品、医薬品、繊維、建材などの幅広い分野で使用されている。

さらに、CDの有用性が認識されるに従って、CDの溶解度の向上、あるいは人や動物に対する安全性を高めることを目的として、酵素反応によりCDに糖の側鎖を結合させた分岐CDや、CD環を構成するグルコースの水酸基を有機合成により、メチル化、エチル化、ヒドロキシプロピル化などを行った置換CDなどのCD誘導体が開発されている。

なかでも、糖質酸化菌を用いて合成するカルボン酸結合型シクロデキストリン、例えばグルクロニルグルコシル−β−CD(以下、GUG−β−CDと略記することがある。) は、イオン交換樹脂を用いた精製法により、容易に高純度の製品を得ることができるとともに、安全性に優れており、医薬品向けのCDとして最適である(特開平07-076594 号、同10-070993 号、同10-210996 号)。

一方、自然界に存在する糖質で、細胞表層の糖鎖は、細胞や組織認識に深く関わっている。N−アセチルグルコサミン(以下、GlcNAcと略記することがある。)、N−アセチルガラクトサミン(以下、GalNAcと略記することがある。)、N−アセチルマンノサミン(以下、ManNAcと略記することがある。)がそのような糖鎖の構成成分である。

しかしながら、従来の合成されたCDやマルトシルCD、GUG−β−CDにはGlcNAcなどのアミノ糖を結合したものはなく、組織親和性の面から標的指向性薬物キャリヤーとしては不完全であった。

本発明者らは先にタチナタマメなどを由来とするN−アセチルヘキソサミニダーゼ、または卵白リゾチームの酵素の縮合反応、または糖転移反応を利用して、β−CDおよびマルトシル−β−CDをはじめとした各種CD類にGlcNAcやGalNAcを結合させることに成功している(特開平08-325304 号、同10-25305号)。

【発明が解決しようとする課題】
しかしながら、これらの方法は、使用する酵素が入手が困難なもので、高価であるため、コスト高になるほか、これらの合成方法は加水分解酵素の逆反応や転移反応を利用するため、その収率は一般的に低かった。
これらの課題を解決するため、本発明者らはカルボン酸結合型シクロデキストリンとアミノ糖を酸アミド結合することを特徴とするアミノ糖を分岐側鎖にもつ新規分岐シクロデキストリンを合成し、これらの分岐シクロデキストリン誘導体を用いた標的指向性薬物キャリヤーとしての利用方法を開発した。

本発明の目的は、カルボン酸結合型シクロデキストリンとアミノ糖を酸アミド結合させた新規分岐シクロデキストリンとその製造法並びにこれら化合物の標的指向性薬物キャリヤーとしての利用を提供することである。

【課題を解決するための手段】
このような事情に鑑み、本発明者らはGUG−β−CDをはじめとしてカルボン酸結合型シクロデキストリンとグルコサミン(GlcN)、ガラクトサミン(GalN)、マンノサミン(ManN)などのをアミノ糖をDCC−HONSu法による縮合反応によって、アミノ糖を分岐側鎖にもつ新規分岐シクロデキストリン誘導体の合成に成功した。さらに、光学バイオセンサーを利用して、この化合物の標的指向性薬物キャリヤーとしての有用性を検討し、本発明を完成させた。

【発明の実施の形態】
本発明に関わる新規シクロデキストリン誘導体の合成材料としては、GUG−β−CDをはじめとしたカルボン酸結合型シクロデキストリンおよびGlcN、GalN、ManNなどのアミノ糖である。

本発明に関わる新規シクロデキストリン誘導体は、一般式(1)〜(3)の構造式で表され、アミノ糖がカルボン酸とアミド結合した構造を有するシクロデキストリン類である。
当該シクロデキストリン誘導体のうち、代表的な化合物は前記一般式(4)〜(6)の構造式で表される。

上記の一般式から明らかなように、本発明に関わるシクロデキストリン誘導体としては、シクロデキストリン誘導体に少なくともGlcN、GalN、ManNが結合した分岐側鎖を当該シクロデキストリン誘導体当たり1分子含んでいればよく、CDとしては、いわゆる複分岐CDも使用でき、また分岐側鎖の鎖長についても制限はない。

次に、本発明に関わるシクロデキストリン誘導体の製造法について説明する。
このシクロデキストリン誘導体は、カルボン酸結合型シクロデキストリンとアミノ糖を縮合反応させることによって得られる。
このときに用いる縮合剤としては、N−エチル−N’−3−ジメチルアミノプロピルカルボジイミド(EDC)またはジシクロヘキシルカルボジイミド(DDC)が挙げられ、EDCが特に好ましい。
この反応を行うにあたり、コハク酸イミド(HONSu)を反応液に加えてもよい。加える場合は、EDC:HONSu=10:1〜1:1が好ましいが、これに限定されない。DDCの場合も同様である。

本発明の方法では、基本的にGUG−β−CDをはじめとしたカルボン酸結合型シクロデキストリンとアミノ糖がある程度溶解する溶液中で実施すればよく、通常は水溶液として反応を行うが、水以外の有機溶媒、例えばDMFなどの有機溶媒や水と該有機溶媒との混合液を使用することができる。

次に、上記方法で生成したシクロデキストリン誘導体は、必要に応じて反応液をイオン交換樹脂、溶媒沈殿法、高速液体クロマトグラフィーなどで分画、分取して精製することが可能である。
本発明において、反応液から未反応のアミノ糖を除去するためのイオン交換樹脂としては、CM−Sephadexなどのカチオン交換樹脂が用いられ、カルボン酸結合型シクロデキストリンを除去するためのイオン交換樹脂としては、ダイヤイオンPA−406、DEAE−Sephadexなどのアニオン交換樹脂が用いられる。しかし、これら未反応の原料を除く方法は、イオン交換樹脂を用いる方法に限るものではなく、電気透析などを用いても良い。

ここで、シクロデキストリン誘導体の具体的な取得方法について述べると、上記の方法により得られた反応液から、未反応の原料をイオン交換樹脂を用いて除去したのち、アセトン再沈、高速液体クロマトグラフィーを用いて目的とするシクロデキストリン誘導体を分画することができる。
生成物が、目的とするシクロデキストリン誘導体であることは、FAB−MSにて分子量を測定し、 1H−NMRおよび13C−NMRにより構造解析を行って確認することができる。
その結果、実施例で得られた化合物は、一般式(4)〜(6)の構造式で表されるものであることを確認した。

シクロデキストリンは、難溶性薬物を包接、可溶化、安定化することができるので、薬物単独で投与する場合に比べて、バイオアベイラビリティーが改善されることが報告されている。本発明に関わるシクロデキストリン誘導体は、包接能に加えてアミノ糖を分岐側鎖に持つことから、その組織親和性は標的指向性を有する。

本発明のシクロデキストリン誘導体のリガンドおよびゲスト物質との相互作用の解析は、FAST社製の光学バイオセンサーlAsysを用いて測定することができる。すなわち、光学バイオセンサーのキュベット表面に固定化したリガンド(レクチン、薬剤)を調製し、これに当該シクロデキストリン誘導体を加え、分子間相互作用によって変動する相互差用量(レスポンス)を観察するものである。

当該シクロデキストリン誘導体の標的指向性と、包接能を利用して標的指向性薬物キャリヤーとして使用することができる。薬物などの化合物を、このシクロデキストリン誘導体と包接させる方法としては、水溶液中で当該シクロデキストリン誘導体(ホスト) と薬物(ゲスト) を包接させ、凍結乾燥、噴霧乾燥などで乾燥物を得るような通常の包接物の調製方法を適用することができる。

以下に、実施例により本発明を具体的に説明するが、本発明はこれらに限定されるものではない。
【実施例】
実施例1
GUG−β−CD(ナトリウム塩) は特開平07-76594号の実施例4に記載の方法従って合成した。
次に、GUG−β−CDを蒸留水に溶解し、EDCとHONSuを加え、0℃で1時間攪拌した後、マンノサミンを加え、0℃で2時間攪拌した。次いで、室温下にて24時間攪拌し、反応させた。
反応終了後、イオン交換樹脂アンバーライトMB3を用いてバッチ法にて、未反応物および塩を除去し、濃縮後、アセトンを加えて再沈させることによって、マンノサミニドグルクロニルグルコシル−β−CD(ManN−GUG−β−CD)を収率84%で得た。

さらに、分取用高速液体クロマトグラフィーにて精製し、当該シクロデキストリン誘導体を得た。ODSカラムを用いたHPLCクロマトグラムを図1に示した。なお、HPLC分析条件は以下の通りである。

単離した当該シクロデキストリン誘導体はFAB−MS(JEOL−LA500)にて分子量を測定したところ、分子量1634であることがわかった。FAB−MSスペクトラムを図2に示した。
さらに、 1H−NMR(JEOL SX−102A)分析を行った。当該シクロデキストリ誘導体の 1H−NMRスペクトラムを図3に示した。図から明らかなように、5.0〜5.1ppmのアノメリックC1プロトンと糖鎖、シクロデキストリンを含めたC1プロトン以外のノンアノメリックプロトンの3.2〜4.0ppmの積分値の比は1:6となっており、これは計算値の“6”とよく一致した。また、これ以外のピーク面積は相対的に少ないことから、当該シクロデキストリン誘導体、つまり一般式(6)で示されるManN−GUG−β−CDであることを確認できた。

実施例2
GUG−β−CD、EDCおよびHONSuを蒸留水に溶解し、0℃で1時間攪拌した後、グルコサミンを加え、0℃で2時間攪拌した。次いで、室温下にて24時間反応させた。
反応終了後、イオン交換樹脂アンバーライトMB3を用いてバッチ法にて、未反応物および塩を除去し、濃縮後、アセトンを加えて再沈させることによって、グルコサミニドグルクロニルグルコシル−β−CD(GlcN−GUG−β−CD)を収率94%で得た。
さらに、分取用高速液体クロマトグラフィーにて精製し、当該シクロデキストリン誘導体を得た。

さらに、分取用高速液体クロマトグラフィーにて精製し、当該シクロデキストリン誘導体を得た。

本発明により、アミノ糖を分岐側鎖に有するシクロデキストリン誘導体とその製造法が提供される。当該シクロデキストリン誘導体を用いて抗がん剤、抗炎症剤などの医薬品化合物を包接させることによって、医薬品化合物の生体内動態、生体内安定性および生体刺激性を改善し、作用時間を遅延し、さらにはバイオアベイラビリティーを改善することが期待できる。特に、当該シクロデキストリン誘導体を、組織親和性を高めた標的指向性薬物キャリヤーとして利用することが期待できる。

Example 1
GUG-β-CD (sodium salt) was synthesized according to the method described in Example 4 of JP-A-07-76594.
Next, GUG-β-CD was dissolved in distilled water, EDC and HONSu were added and stirred at 0 ° C. for 1 hour, then mannosamine was added and stirred at 0 ° C. for 2 hours. Subsequently, it was made to react by stirring for 24 hours at room temperature.

When the molecular weight of the isolated cyclodextrin derivative was measured by FAB-MS (JEOL-LA500), it was found to have a molecular weight of 1634. The FAB-MS spectrum is shown in FIG.
Further, 1 H-NMR (JEOL SX-102A) analysis was performed. The 1 H-NMR spectrum of the cyclodextri derivative is shown in FIG.
